Abstract

Die Erfindung stellt Polypeptide eines eine hdm2-Protein-spezifische T-Zell Antowort vermittelnden murinen alpha / beta T-Zell Rezeptors oder funktioneller Varianten oder Teile davon oder diese kodierende Nukleinsäuren, funktionelle Varianten oder Teile davon zur Verfügung. Dises bewirken, dass hdm2-Protein-exprimierende Zellen von T-Zellen, die mit jenen Genen ausgestattet wurden, erkannt werden, Zytokine ausgeschüttet werden, und eine Z-Zell-induzierte Lyse und/oder Apoptose von Tumor-oder Leukämiezellen herbeigeführt wird.

Beschreibung
Die Erfindung betrifft Polypeptide des eine hdm2-Protein-spezifische T-Zell Antwort vermittelnden murinen α/ß T-Zell Rezeptors, diese kodierende Nukleinsäuren und deren Verwendung bei der Therapie, Diagnose und/oder Prävention von mit dem hdm2 -Protein assoziierten Erkrankungen.
Die Antigenerkennung durch T-Lymphozyten (TZL) ist entscheidend für die Erzeugung und Regulierung einer effektiven Immunantwort. Der charakteristische T-Zelllinien-Marker ist der T-Zell-Antigen-Rezeptor (TZR). Es gibt zwei definierte Typen von TZR: Einer ist ein Hete- rodimer von zwei Disulfid-verbundenen Polypeptiden (α und ß); der andere ist zwar strukturell ähnlich, besteht jedoch aus γ- und δ-Polypeptiden. Beide Rezeptoren sind mit einem Set von fünf Polypeptiden, dem CD3 -Komplex assoziiert und bilden so zusammen den T-Zell Rezeptor-Komplex (TZR-CD3-Komplex). Der α/ß-TZR ist der funktioneil bedeutendste, da er in über 95% aller T-Zellen exprimiert wird und die tragende Immunantwort vermittelt.
α/ß-T-Zellen können in zwei verschiedene sich überschneidende Populationen unterteilt werden: Eine Untergruppe, die den CD4-Marker trägt und hauptsächlich die Immunantwort unterstützt (TH) und eine Untergruppe, die den CD8-Marker trägt und im wesentlichen zytoto- xisch ist (Tc). CD8+-T-Zellen erkennen Antigene in Assoziation mit MHC-Klasse-I- Molekülen. Solche Antigene können unter anderem tumorspezifische oder tumorassoziierte Peptidantigene sein. Nach Erkennung der Peptidantigene wird die betreffende Zelle abgetötet, indem die T-Zelle die Zielzelle lysiert und/oder Apoptose dieser Zielzellen induziert oder Zytokine (z.B. IL-2, IFN-γ) freisetzt.
Unter den tumorassoziierten Peptidantigenen (TAA), die im Kontext von MHC-Klasse-I- Molekülen auf der Oberfläche von Tumorzellen präsentiert werden, sind die sogenannten „universellen" TAA von besonderem Interesse. Diese TAA leiten sich überwiegend von zellulären Proteinen ab, die in normalen Zellen schwach exprimiert und in Tumorzellen überex- primiert werden. Zu diesen Proteinen gehört unter anderem das humane Homolog des „Mouse-Double-Minute-2" Proto-Antigens (mdm2), das sogenannte „humane mdm2" oder abgekürzt „hdm2" Proto-Onkoprotein (Roth et al. 1998), das nicht nur in einer Reihe solider Tumore, sondern auch bei den hämatologischen Neoplasien (malignen hämatologischen Systemerkrankungen) AML, ALL und CLL überexprimiert wird (Zhou et al, 2000).
Oligopeptide des hdm2-Proteins können im Kontext mit MHC-Klasse-I-Molekülen auf der Zelloberfläche präsentiert werden und repräsentieren attraktive Zielstrukturen für CD8- positive T-Zellen. Das Ausmaß der T-Zell Antwort verläuft hierbei in einem definierten kinetischen Fenster (Kersh et al., 1998). Der Komplex aus Peptid-MHC und TZR-CD3 multimeri- siert, um eine effiziente Signaltransduktion zu bewirken, wobei die genaue Stöchiometrie und das Ausmaß der Oligomerisierung noch umstritten ist.
Voraussetzung für die Entwicklung immuntherapeutischer Verfahren zur Behandlung von bösartigen Tumorerkrankungen ist die Identifizierung von Onkoprotein-spezifischen T-Zell Rezeptoren. Mit solchen T-Zell Rezeptoren können unter bestimmten Voraussetzungen T- Zellen mit Antigen-Spezifität im allgemeinen und Tumorreaktivtät im besonderen versehen werden mit dem Ziel, daß die T-Zellen die Remission und die Eradikation eines bestimmten Tumors herbeifuhren. Beschriebene Methoden zur Identifizierung von hochaffinen Peptid- spezifischen TZRs beinhalten die „phage-,, oder „yeast-display" -Methodik (Holler et al., 1999) bzw. eine Tetramer-abhängige „TCR display" -Methodik (Kessels et al., 2000).
Aus Theobald et al. („Targeting p53 as a general tumor antigen", P.N.A.S. USA 92, pp. 11993-11997, 1995) ist die Herstellung von p53-spezifischen zytotoxischen T-Zellen nach der Injektion von Peptiden aus der Sequenz von p53 bekannt. Mittels dieser T-Zelllinien konnte anschließend eine Auswahl von menschlichen Tumorzellen lysiert werden. Die Isolierung von Genen spezifischer T-Zell Rezeptoren der gegen p53 gerichteten lyrischen T-Zellen wird nicht beschrieben. Die WO 97/32603 beschreibt allgemein ein Verfahren zur Herstellung von re- kombinanten T-Lymphozyten, die gegen Tumorgewebe gerichtete spezifische T-Zell Rezeptoren exprimieren. Dabei wird eine HLA-transgene Maus (in diesem Fall HLA-A2) mit tu- mor-assoziiertem Antigen immunisiert, um so die Produktion von zytotoxischen T- Lymphozyten zu bewirken, die spezifische T-Zell Rezeptoren auf ihrer Oberfläche exprimieren. Als tumorassoziierte Antigene werden Peptide verschiedener Gene, wie Her-2/neu, Ras, p53, Tyrosinase, MART, gplOO, MAGE, BAGE und MUC-1 beschrieben. Aus den Her-2/neu spezifischen T-Lymphozyten wird dann die Nukleotidsequenz, die mindestens eine variable Region der α- und ß-Kette des entsprechenden nicht menschlichen T-Zell Rezeptors enthält isoliert und in Form verschiedener molekularbiologisch modifizierter genetischer (u.a. „humanisierter") TZR-Konstrukte verwendet. So beschreibt die WO 97/32603 Fusionsproteine von variablen Regionen von TZR mit der ζ-Region von CD3 oder CD8 oder CD 16, sowie die Verwendung von flexiblen Linker der Aminosäuresequenz (GGGGS)3.
Clay et al. („Efficient transfer of a tumor antigen-reactive TCR to human peripheral blood lymphocytes confers anti-tumor reactivity"; J. Immunology, 1999, 163: 507-513) beschreiben die Isolierung von Genen der α-TZR und ß-TZR Ketten eines MART-1 spezifischen TZR und dessen Expression in humanen peripheren Blut-Lymphocyten (PBLs). Die TZR sind gegen die Aminosäuren 25-35 von MART-1 gerichtet. Weiterhin wird die Lyse von verschiedenen metastatischen Melanomzelllinien und ein retrovirales Vektorsystem unter Verwendung von IRES-Elementen beschrieben.
Darcy et al. („Redirected perforin-dependent lysis of colon carcinoma by ex vivo genetically engineered CTL" J. Immunol., 2000, 164:3705-3712) beschreiben ein immunotherapeutisches Verfahren für Colon-Karzinome unter der Verwendung von scFv anti-CEA Rezeptor transdu- zierten ZTL, Perforin und γ-IFN. Das chimäre spezifische Rezeptorkonstrukt wird über einen retroviralen Vektor in primäre Maus-T-Lymphocyten transduziert. Diese Zellen wurden in Mäuse injiziert, die vorher mit Colon-Karzinomzelllinien beimpft waren.
Weijtens et al. („A retroviral vector System ,'STITCH'; in combination with an optimized Single chain antibody chimeric receptor gene structure allows efficient gene transduction and expression in human T-lymphocytes", Gene Therapy (1998) 5,1995-1203) und Willemsen et al. „Grafting primary human T lymphocytes with cancer-specific single chain and two chain TCR" Gene Therapy, 2000 7, 1369-1377) beschreiben retrovirale Vektorsysteme zur Trans- duktion von Genen in aktivierte T-Lymphozyten. Dieses System wird verwendet, um die Expression von Antikörper-basierten chimären Rezeptoren in der Membran von T-Zellen zu bewirken. Diese T-Zellen können dann gegen spezifische Krebszellen eingesetzt werden. Die Wirksamkeit des retroviralen Vektorsystems wird anhand der spezifischen Erkennung und Zytolyse von Nierenzellkarzinomen beschrieben. Ähnlich beschreiben Eshar. et al. („Specific activation and targeting of cytotoxic lymphocytes through chimeric single chains consisting of antibody-binding domains and the γ or ζ subunits of the immunoglobulin and T-cell re- ceptors" P.N.A.S. USA, 90, pp. 720-724, 1993) die Herstellung von tumorspezifischen Lym- phozyten und ihre Verwendung in der Immuntherapie auf der Basis von Chimären, welche die variablen Regionen eines Antikörpers (scFV's) mit der konstanten Region des T-Zell Rezeptors umfassen (sogenannte „single-chain TCR" (scTCR)). Diese chimären Gene konnten auf der Oberfläche von zytolytischen T-Zell-Hybridomen exprimiert werden und bewirkten die Ausschüttung von Interleukin-2 nach Kontakt mit dem Antigen.
Ein spezifisch gegen hdm2 gerichteter TZR und dessen Verwendung wird jedoch durch die oben genannten Publikationen sowie in der übrigen Literatur weder erwähnt noch nahegelegt.
Der vorliegenden Erfindung liegt daher die Aufgabe zugrunde, die murinen Gene der gegen das hdm2-Protein gerichteten T-Zell Rezeptoren α-TZR und ß-TZR zur Verfügung zu stellen. Diese bewirken, daß hdm2-Protein-exprimierende Zellen von CD8-positiven T-Zellen erkannt werden, Zytokine ausgeschüttet werden, und eine T-Zell-induzierte Lyse und/oder Apoptose von Tumor- oder Leukämiezellen herbeigeführt wird.
Erfindungsgemäß wird diese Aufgabe durch das Polypeptid des eine hdm2-Protein- spezifische T-Zell Antwort vermittelnden murinen α/ß T-Zell Rezeptors gemäß SEQ ID Nr. 1 oder SEQ ID Nr. 2 oder funktioneller Varianten oder Teile davon oder dieses kodierende Nukleinsäuren, funktioneile Varianten oder Teile davon gelöst.
Aus einer doppelt-A2.1-transgenen Maus [huCD8α/ß x A2.1/Kb]Fι in C57BL/6 genetischer Hintergrund wurden die Gene des eine hdm2-Protein-spezifische T-Zell Antwort vermittelnden murinen α/ß T-Zell Rezeptors isoliert, die eine HLA-A2.1 restringierte und gegen das Peptid der Aminosäuren 81-88 des hdm2-Proteins gerichtete spezifische T-Zell- Antwort vermitteln. Die Polypeptide dieses TZR waren bisher unbekannt. Die Gene wurden als Wildtyp (WT) oder modifizierte Konstrukte retroviral in humane periphere Blut-Lymphocyten (PBLs) eingeschleust und die HLA-restringierte Antigen-Erkennung funktioneil durch CD8- vermittelte zyto toxische Lyse verschiedener Zeil-Linien im 51 Chrom-Freisetzungstest überprüft. Die erfindungsgemäßen Gene des hdm2-spezifischen TZR gehören nicht zu den bisher bekannten geeigneten Zielen für die Diagnose - wie beispielsweise der Indikation - und/oder der Behandlung - wie beispielsweise der Modulation - von mit hdm2-Protein in Zusammenhang stehenden Erkrankungen oder für die Identifizierung von pharmakologisch aktiven Substanzen, so daß sich aus dieser Erfindung völlig neue Therapieansätze ergeben. Ein weiterer Aspekt der Erfindung betrifft ein Fusionsprotein, umfassend das erfindungsgemäße Polypeptid oder funktionelle Varianten oder Teile davon oder dieses kodierende Nukleinsäuren, funktionelle Varianten oder Teile davon.
Das Fusionsprotein kann dadurch gekennzeichnet sein, daß es die ζ-Region von CD3 oder CD8 oder CD 16 oder Teile davon umfaßt, insbesondere die ζ-Region von humanem CD3 oder CD8 oder CD 16 oder Teile davon. Bevorzugt ist ein erfindungsgemäßes Fusionsprotein, das einen flexiblen Linker umfaßt (Whitlow et al., „An improved linker for single-chain Fv with reduced aggregation and enhanced proteolytic stability", Prot. Engin. 6(8), pp. 989-995, 1993), insbesondere einen Linker der Aminosäuresequenz (GGGGS)3. Insbesondere kann das erfindungsgemäße Fusionsprotein die ζ-Kette des CD3 -Komplexes oder ITAM-Motive der ζ- Kette oder Teile davon umfassen, insbesondere die ζ-Kette von humanem CD3 oder Teile davon. Das Fusionsprotein kann weiterhin dadurch gekennzeichnet sein, daß es CD8α oder das Lck-Bindungsmotiv von CD8α umfaßt oder Teile davon, insbesondere von humanem CD8α.
Bei dem erfindungsgemäßen Fusionsprotein kann es sich weiterhin um eine chimäre partiell oder vollständig humanisierte α und/oder ß TZR-Kette handeln, insbesondere gemäß SEQ ID Nr. 3 oder SEQ ID Nr. 4. Ein weiterer Aspekt der Erfindung betrifft ein Fusionsprotein, bei dem es sich um einen Einzelketten T-Zell Rezeptor handelt, insbesondere gemäß SEQ ID Nr. 5 oder SEQ ID Nr. 6. Das erfindungsgemäße Fusionsprotein kann aber auch dadurch gekennzeichnet sein, daß es sich um einen an andere funktionelle Komponenten gekoppelten α/ß- TZR handelt, insbesondere gemäß SEQ ID Nr. 7 oder SEQ ID Nr. 8.
Ein weiterer Gegenstand der Erfindung ist ein Verfahren zur Herstellung eines erfindungsgemäßen Fusionsproteins zur Diagnose und/oder Behandlung vom mit hdm2-Protein in Zusammenhang stehenden Erkrankungen oder zur Identifizierung von pharmakologisch aktiven Substanzen, z.B. in einer geeigneten Wirtszelle, bei dem eine erfindungsgemäße Nukleinsäure verwendet wird.
Hergestellt werden hierbei Fusionsproteine, die die oben beschriebenen erfindungsgemäßen Polypeptide enthalten, wobei die Fusionsproteine selbst bereits die Funktion eines Polypep- tids der Erfindung aufweisen oder erst nach Abspaltung des Fusionsanteils die spezifische Funktion aktiv ist. Vor allem zählen hierzu Fusionsproteine mit einem Anteil von ca. 1-200, vorzugsweise ca. 1-150, insbesondere ca. 1-100, vor allem ca. 1-50 fremden Aminosäuren. Beispiele solcher Peptidsequenzen sind prokaryotische Peptidsequenzen, die z. B. aus der Galactosidase von E. coli abgeleitet sein können. Weiterhin können auch virale Peptidsequenzen, wie zum Beispiel vom Bakteriophagen Ml 3 verwendet werden, um so Fusionsproteine für das dem Fachmann bekannte "phage display"- Verfahren zu erzeugen.
Zur Aufreinigung der erfindungsgemäßen Proteine kann ein weiteres/weiterer Polypep- tid("tag") angefügt sein. Erfindungsgemäße Protein-tags erlauben beispielsweise die hochaffine Absorption an eine Matrix, stringentes Waschen mit geeigneten Puffern, ohne den Komplex in nennenswertem Maße zu eluieren und anschließend die gezielte Elution des absorbierten Komplexes. Beispiele der dem Fachmann bekannten Protein-tags sind ein (His)6-tag, ein Myc-tag, ein FLAG-tag, ein Strep-tag, ein Strep-tag II, ein Hämagglutinin-tag, Glutathion-Transferase (GST)-tag, Intein mit einem Affinitäts-Chitin-Bindungs-tag oder Maltose-bindendes Protein (MBP)-tag. Diese Protein-tags können sich N-, C-terminal und/oder intern befinden.
Neben den aus Zellen isolierten natürlichen Polypeptiden können alle erfindungsgemäßen Polypeptide oder deren Teile unter zellfreien Bedingungen hergestellt worden sein, z. B. durch Synthese oder durch in v tro-Translation. So kann das gesamte Polypeptid oder Teile davon zum Beispiel mit Hilfe der klassischen Synthese (Merrifield-Technik) synthetisiert werden. Teile der erfindungsgemäßen Polypeptide eignen sich insbesondere zur Gewinnung von Antiseren, mit deren Hilfe geeignete Genexpressionsbanken durchsucht werden können, um so zu weiteren funktioneilen Varianten des erfindungsgemäßen Polypeptids zu gelangen.
Die Erfindung betrifft auch Polypeptide, die Derivate eines Antikörpers mit Spezifität für hdm2-81-88 Peptid, insbesondere präsentiert im Kontext von HLA-A2.1 sind.
Weiterhin umfaßt die Erfindung retro-inverse Peptide oder Pseudopeptide gemäß der Poly- peptidsequenz der SEQ ID Nr. 1 bis SEQ ID Nr. 8 oder funktionelle Varianten oder Teile davon. Diese Peptide weisen anstelle der -CO-NH-Peptidbindungen -NH-CO-Bindungen auf.
Die Aufgabe der Erfindung wird weiterhin durch eine erfindungsgemäße Nukleinsäure gelöst, die eine DNA, RNA, PNA (Peptide nucleic acid) oder p-NA (Pyranosyl nucleic acid), vor- zugsweise eine DNA, insbesondere eine doppelsträngige DNA ist mit einer Länge von mindestens 8 Nukleotiden, vorzugsweise mit mindestens 18 Nukleotiden, insbesondere mit mindestens 24 Nukleotiden ist. Die Nukleinsäure kann dadurch gekennzeichnet sein, daß die Sequenz der Nukleinsäure mindestens ein Intron und/oder eine polyA-Sequenz aufweist. Sie kann auch in Form ihrer antisense-Sequenz vorliegen.
Für die Expression des betreffenden Gens ist im allgemeinen eine doppelsträngige DNA bevorzugt, wobei der für das Polypeptid kodierende DNA-Bereich besonders bevorzugt ist. Dieser Bereich beginnt mit dem ersten in einer Kozak Konsensus Sequenz (Kozak, 1987, Nucleic. Acids Res. 15:8125-48) liegenden Start-Codon (ATG) bis zum nächsten Stop-Codon (TAG, TGA bzw. TAA), das im gleichen Leseraster zum ATG liegt. Eine weitere Verwendung der erfindungsgemäßen Nukleinsäuresequenzen ist die Konstruktion von anti-sense Oli- gonukleotiden (Zheng und Kemeny, 1995, Clin. Exp. Immunol. 100:380-2) und/oder Ribozymen (Amarzguioui, et al. 1998, Cell. Mol. Life Sei. 54:1175-202; Vaish, et al., 1998, Nucleic Acids Res. 26:5237-42; Persidis, 1997, Nat. Biotechnol. 15:921-2). Mit anti-sense Oligonukleotiden kann man die Stabilität der erfindungsgemäßen Nukleinsäure verringern und/oder die Translation der erfindungsgemäßen Nukleinsäure inhibieren. So kann beispielsweise die Expression der entsprechenden Gene in Zellen sowohl in vivo als auch in vitro verringert werden. Oligonukleotide können sich daher als Therapeutikum eignen. Diese Strategie eignet sich beispielsweise auch für Haut, epidermale und dermale Zellen, insbesondere, wenn die antisense Oligonukleotide mit Liposomen komplexiert werden (Smyth et al., 1997, J. In- vest. Dermatol. 108:523-6; White et al., 1999, J. Invest. Dermatol. 112:699-705). Für die Verwendung als Sonde oder als "antisense" Oligonukleotid ist eine einzelsträngige DNA oder RNA bevorzugt.
Neben den aus Zellen isolierten natürliche Nukleinsäuren können alle erfindungsgemäßen Nukleinsäuren oder deren Teile auch synthetisch hergestellt worden sein. Weiterhin kann zur Durchführung der Erfindung eine Nukleinsäure verwendet werden, die synthetisch hergestellt worden ist. So kann die erfindungsgemäße Nukleinsäure beispielsweise chemisch anhand der in den SEQ ID Nr. 1 bis SEQ ID Nr. 8 beschriebenen Proteinsequenzen unter Heranziehen des genetischen Codes z. B. nach der Phosphotriester-Methode synthetisiert werden (siehe z. B. Uhlmann, E. & Peyman, A. (1990) Chemical Revievs, 90, 543-584). Oligonukleotide werden in der Regel schnell durch Endo- oder Exonukleasen, insbesondere durch in der Zelle vorkommende DNasen und RNasen, abgebaut. Deshalb ist es vorteilhaft, die Nukleinsäure zu modifizieren, um sie gegen den Abbau zu stabilisieren, so daß über einen langen Zeitraum eine hohe Konzentration der Nukleinsäure in der Zelle beibehalten wird (WO 95/11910; Macadam et al., 1998, WO 98/37240; Reese et al., 1997, WO 97/29116). Typischerweise kann eine solche Stabilisierung durch die Einführung von einer oder mehrerer Internukleotid-Phosphorgruppen oder durch die Einfuhrung einer oder mehrerer Nicht- Phosphor-Internukleotide, erhalten werden.
Geeignete modifizierte Internukleotide sind in Uhlmann und Peymann (1990 Chem. Rev. 90, 544) zusammengefaßt (WO 95/11910; Macadam et al., 1998, WO 98/37240; Reese et al., 1997, WO 97/29116). Modifizierte Internukleotid-Phosphatreste und/oder Nicht- Phosphorbrücken in einer Nukleinsäure, die bei einer der erfindungsgemäßen Verwendungen eingesetzt werden können, enthalten zum Beispiel Methylphosphonat, Phosphorothioat, Phosphoramidat, Phosphorodithioat, Phophatester, während Nicht-Phosphor-Internukleotid- Analoge, beispielsweise Siloxanbrücken, Carbonatbrücken, Carboxymethylester, Acetami- datbrücken und/oder Thioetherbrücken enthalten. Es ist auch beabsichtigt, daß diese Modifizierung die Haltbarkeit einer pharmazeutischen Zusammensetzung, die bei einer der erfindungsgemäßen Verwendungen eingesetzt werden kann, verbessert.
Ein weiterer Aspekt der vorliegenden Erfindung betrifft einen Vektor, vorzugsweise in Form eines Plasmids, Shuttle Vektors, Phagemids, Cosmids, Expressionsvektors, adenoviralen Vektors, retroviralen Vektors (Miller, et al. „Improved retroviral vectors for gene transfer and expression", BioTechniques Vol. 7, No. 9, p 980, 1989) und/oder gentherapeutisch wirksamen Vektors, der eine erfindungsgemäße Nukleinsäure enthält.
So kann die erfindungsgemäße Nukleinsäure in einem Vektor vorzugsweise in einem Expressionsvektor oder gentherapeutisch wirksamen Vektor enthalten sein. Vorzugsweise enthält der gentherapeutisch wirksame Vektor T-Zell spezifische regulatorische Sequenzen, die funktioneil mit der erfindungsgemäßen Nukleinsäure verbunden sind. Die Expressionsvektoren können prokaryotische oder eukaryotische Expressionsvektoren sein. Beispiele für prokaryotische Expressionsvektoren sind für die Expression in E. coli z.B. die Vektoren pGEM oder pUC- Derivate und für eukaryotische Expressionsvektoren für die Expression in Saccharomyces cerevisiae z. B. die Vektoren p426Met25 oder p426GALl (Mumberg et al. (1994) Nucl. Acids Res., 22, 5767-5768), für die Expression in Insektenzellen z. B. Bac lovirus- Vektoren wie in EP-B1-0 127 839 oder EP-B1-0 549 721 offenbart, und für die Expression in Säugerzellen z. B. die Vektoren Rc/CMV und Rc/RSV oder SV40-Vektoren, welche alle allgemein erhältlich sind.
Im allgemeinen enthalten die Expressionsvektoren auch für die jeweilige Wirtszelle geeignete Promotoren, wie z. B. den trp-Promotor für die Expression in E. coli (siehe z. B. EP-B1-0 154 133), den Met 25, GAL 1 oder ADH2-Promotor für die Expression in Hefen (Rüssel et al. (1983), J. Biol. Chem. 258, 2674-2682; Mumberg, supra), den Baculovirus-Polyhedrin- Promotor für die Expression in Insektenzellen (siehe z. 13. EP-B1-0 127 839). Für die Expression in Säugetierzellen sind beispielsweise Promotoren geeignet, die eine konstitutive, regulierbare, gewebsspezifische, zellzyklusspezifische oder metabolischspezifische Expression in eukaryotischen Zellen erlauben. Regulierbare Elemente gemäß der vorliegenden Erfindung sind Promotoren, Aktivatorsequenzen, Enhancer, Silencer und/oder Repressorsequen- zen. Beispiel für geeignete regulierbare Elemente, die konstitutive Expression in Eukaryonten ermöglichen, sind Promotoren, die von der RNA Polymerase III erkannt werden oder virale Promotoren, CMV-Enhancer, CMV-Promotor, CMV-LTR-Hybride, SV40 Promotor oder LTR-Promotoren z. B. von MMTV (mouse mammary tumour virus; Lee et al. (1981) Nature 214, 228-232) und weitere virale Promotor- und Aktivatorsequenzen, abgeleitet aus beispielsweise HBV, HCV, HSV, HPV, EBV, HTLV oder HIV. Ein Beispiel für ein regulierbares Element, das eine regulierbare Expression in Eukaryonten ermöglicht, ist der Tetracycli- noperator in Kombination mit einem entsprechenden Repressor (Gossen M. et al. (1994) Curr. Opin. Biotechnol. 5, 516-20).
Beispiele für regulierbare Elemente, die T-Zell spezifische Expression in Eukaryonten ermöglichen, sind Promotoren oder Aktivatorsequenzen aus Promotoren oder Enhancern von solchen Genen, die für Proteine kodieren, die nur in diesen Zelltypen exprimiert werden.
Beispiele für regulierbare Elemente, die zellzyklusspezifische Expression in Eukaryonten ermöglichen, sind Promotoren folgender Gene: cdc25, Cyclin A, Cyclin E, cdc2, E2F, B-myb oder DHFR (Zwicker J. und Müller R. (1997) Trends Genet. 13, 3-6). Beispiele für regulierbare Elemente, die metabolischspezifische Expression in Eukaryonten ermöglichen, sind Promotoren, die durch Hypoxie, durch Glukosemangel, durch Phosphatkonzentration oder durch Hitzeschock reguliert werden. Der erfindungsgemäße Vektor kann zur Transfektion einer Wirtszelle verwendet werden, bei der es sich bevorzugterweise um eine T-Zelle handelt. Besonders bevorzugt ist eine Wirtszelle, die dadurch gekennzeichnet ist, daß sie auf ihrer Oberfläche ein erfindungsgemäßes Poly- peptid oder Fusionsprotein exprimiert.
Um die Einführung von erfindungsgemäßen Nukleinsäuren und damit die Expression des Polypeptids in einer eu- oder prokaryotischen Zelle durch Transfektion, Transduktion, Transformation oder Infektion zu ermöglichen, kann die Nukleinsäure als Plasmid, als Teil eines viralen oder nicht-viralen Vektors oder Partikels vorliegen. Als virale Vektoren oder Partikel eignen sich hierbei besonders: Baculoviren, Vakziniaviren, Retroviren, Adenoviren, adenoas- soziierte Viren und Herpesviren. Als nicht-virale Träger eignen sich hierbei besonders: Viro- somen, Liposomen, kationische Lipide, oder poly-Lysin konjugierte DNA.
Beispiele von gentherapeutisch wirksamen Vektoren sind Virusvektoren, beispielsweise Ade- novirusvektoren oder retrovirale Vektoren (Lindemann et al., 1997, Mol. Med. 3: 466-76; Springer et al., 1998, Mol. Ein bevorzugter Mechanismus, erfindungsgemäße Polypeptide in vivo zur Expression zu bringen, ist der virale Gen-Transfer, insbesondere mit Hilfe retroviraler Partikel. Diese werden vorzugsweise genutzt, entsprechende Zielzellen, vorzugsweise T-Lymphozyten, des Patienten ex vivo mit den für erfindungsgemäße Polypeptide kodierenden Genen oder Nukleo- tidsequenzen durch Transduktion zu versehen. Die Zielzellen können daraufhin im Sinne eines adoptiven Zelltransfers wieder in den Patienten reinfundiert werden, um mit der de novo eingefügten Spezifität tumorizide und/oder immunmodulierende Effektorfunktionen zu übernehmen. Jüngst wurden auf diesem Wege sehr gute gentherapeutische Erfolge in der Behandlung der durch Immuninkompetenz gekennzeichneten SCID-Xl- Krankheit bei Neugeborenen erzielt, in dem hämatologische Vorläuferzellen mit einem analogen intakten Transgen einer in den Kindern vorkommenden nicht-funktionellen mutierten Variante des γ- Kettengens, das für die Differenzierung in die verschiedenen Effektorzellen des adaptiven Immunsystems essentiell ist, retroviral versehen wurden (Cavazzana-Calvo et al., 2000). Weiterhin besteht die Möglichkeit den Gentransfer in vivo durchzuführen, einerseits durch präferentiell stereotaktische Injektion der infektiösen Partikel, andererseits durch direkte Applikation von Viren-produzierenden Zellen (Oldfield, et al. Hum. Gen. Ther., 1993, 4:39-69).
Die zum Transfer von Genen häufig eingesetzten viralen Vektoren sind nach heutigem Stand der Technik vorwiegend retrovirale, lentivirale, adenovirale und adeno-assoziierte virale Vektoren. Diese sind von natürlichen Viren abgeleitete zirkuläre Nukleotidsequenzen, in denen zumindest die viralen Strukturprotein-kodierenden Gene durch das zu transferierende Konstrukt ausgetauscht werden.
Retrovirale Vektorsysteme schaffen die Voraussetzung für eine langhaltende Expression des Transgens durch die stabile, aber ungerichtete Integration in das Wirtsgenom. Vektoren der jüngeren Generation besitzen keine irrelevanten und potentiell immunogenen Proteine, des weiteren gibt es keine vorbestehende Immunität des Empfängers gegenüber dem Vektor. Re- troviren enthalten ein RNA-Genom, das in eine LipidhüUe verpackt ist, die aus Teilen der Wirtszellmembran und Virusproteinen besteht. Zur Expression viraler Gene wird das RNS- Genom revers transkribiert und mit dem Enzym Integrase in die Zielzell-DNS integriert. Diese kann daraufhin von der infizierten Zelle transkribiert und translatiert werden, wodurch virale Bestandteile entstehen, die sich zu Retroviren zusammenfügen. RNS wird ausschließlich dann in die neu entstandenen Viren eingefügt. Das Genom der Retroviren besitzt drei essentielle Gene: gag, das für virale Strukturproteine, sogenannte gruppenspezifische Antigene kodiert, pol für Enzyme wie Reverse Transkriptase und Integrase und env für das Hüllprotein („envelope"), das für die Bindung des wirtsspezifischen Rezeptors verantwortlich ist. Die Produktion der replikationsinkompetenten Viren findet nach Transfektion in sogenannten Verpackungszelllinien statt, die zusätzlich mit den gag/pol-kodierenden Genen ausgestattet wurden und diese „in trans" exprimieren und somit die Ausbildung replikationsinkompetenter (d.h. gag/pol-deletierter) transgener Viruspartikel komplementieren. Eine Alternative ist die Cotransfektion der essentiellen Virusgene, wobei nur der das Transgen enthaltende Vektor das Verpackungssignal trägt.
Die Separation dieser Gene ermöglicht einerseits die beliebige Kombination des gal/pol- Leserahmens mit aus verschiedenen Stämmen gewonnenen e«v-Leserahmen, wodurch Pseu- dotypen mit verändertem Wirtstropismus entstehen, andererseits kann dadurch die Bildung replikationskompetenter Viren innerhalb von Verpackungszellen drastisch reduziert werden. Das aus „gibbon ape leukemia virus" (GALV) abgeleitete Hüllprotein, das im „stitch" bzw. „bullet" -Vektorsystem Verwendung findet, ist in der Lage humane Zellen zu transduzieren und ist in der Verpackungszelllinie PG13 mit amphotropen Wirtsbereich etabliert (Miller et al., 1991). Zusätzlich wird die Sicherheit durch selektive Deletion von nicht-essentiellen Virussequenzen zur Verhinderung einer homologen Rekombination und somit der Produktion replikationskompetenter Partikel erhöht.
Neue, nicht-virale Vektoren bestehen aus autonomen, sich selbst-integrierenden DNS- Sequenzen, den Transposonen, die durch z.B. liposomale Transfektion in die Wirtszelle ein- geschleußt werden und erstmals erfolgreich zur Expression humaner Transgene in Säugerzellen eingesetzt wurden (Yant et al., 2000).
Gentherapeutisch wirksame Vektoren lassen sich auch dadurch erhalten, daß man die erfindungsgemäße Nukleinsäure mit Liposomen komplexiert, da damit eine sehr hohe Transfekti- onseffizienz, insbesondere von Hautzellen, erreicht werden kann (Alexander und Akhurst, 1995, Hum. Mol. Genet. 4: 2279-85). Hilfsstoffe, die den Transfer von Nukleinsäuren in die Zelle erhöhen, können beispielsweise Proteine oder Peptide, die an DNA gebunden sind oder synthetische Peptid-DNA-Moleküle, die den Transport der Nukleinsäure in den Kern der Zelle ermöglichen, sein (Schwartz et al. (1999) Gene Therapy 6, 282; Branden et al. (1999) Nature Biotech. 17, 784). Hilfsstoffe umfassen auch Moleküle, die die Freisetzung von Nukleinsäuren in das Cytoplasma der Zelle ermöglichen (Planck et al. (1994) J. Biol. Chem. 269, 12918; Kichler et al. (1997) Bioconj. Chem. 8, 213) oder beispielsweise Liposomen (Uhl- mann und Peymann (1990) supra). Eine andere besonders geeignete Form von gentherapeutischen Vektoren läßt sich dadurch erhalten, daß man die erfindungsgemäße Nukleinsäure auf Goldpartikeln aufbringt und diese mit Hilfe der sogenannten "Gene Gun" in Gewebe, bevorzugt in die Haut, oder Zellen schießt (Wang et al., 1999, J. Invest. Dermatol., 112:775-81).
Für die gentherapeutische Anwendung der erfindungsgemäßen Nukleinsäure ist es auch von Vorteil, wenn der Teil der Nukleinsäure, der für das Polypeptid kodiert, ein oder mehrere nicht kodierende Sequenzen einschließlich Intronsequenzen, vorzugsweise zwischen Promotor und dem Startcodon des Polypeptids, und/oder eine polyA-Sequenz, insbesondere die natürlich vorkommende polyA-Sequenz oder eine SV40 Virus polyA-Sequenz, vor allem am 3 '- Ende des Gens enthält, da hierdurch eine Stabilisierung der mRNA erreicht werden kann (Jackson, R. J. (1993) Cell 74, 9-14 und Palmiter, R. D. et al. (1991) Proc. Natl. Acad. Sei. USA 88, 478-482).
Ein weiterer Gegenstand der vorliegenden Erfindung ist eine Wirtszelle, insbesondere eine T- Zelle, die mit einem erfindungsgemäßen Vektor oder einem anderen erfindungsgemäßen Genkonstrukt transformiert ist. Wirtszellen können sowohl prokaryotische als auch eukaryotische Zellen sein, Beispiele für prokaryotische Wirtszellen sind E. coli und für eukaryotische Zellen Saccharomyces cerevisiae oder Insektenzellen.
Ein besonders bevorzugte transformierte Wirtszelle ist eine transgene T- Vorläuferzelle oder eine Stammzelle, die dadurch gekennzeichnet ist, daß sie ein erfindungsgemäßes Genkonstrukt oder eine erfindungsgemäße Expressionskassette umfaßt. Verfahren zur Transformation oder Transduktion von Wirtszellen und/oder Stammzellen sind dem Fachmann gut bekannt und umfassen zum Beispiel Elektroporation oder Mikroinjektion. Eine besonders bevorzugte transformierte Wirtszelle ist eine patienteneigene T-Zelle, die nach der Entnahme mit einem erfindungsgemäßen Genkonstrukt transfiziert wird. Erfindungsgemäße Wirtszellen können insbesondere dadurch erhalten werden, daß dem Patienten eine oder mehrere Zellen, bevorzugterweise T-Zellen, insbesondere CD8+-T-Zellen entnommen werden, die dann ex vivo mit einem oder mehreren erfindungsgemäßen genetischen Konstrukten transfiziert oder transdu- ziert werden, um so erfindungsgemäße Wirtszellen zu erhalten. Die ex vivo generierten spezifischen T-Zellen können dann anschließend in den Patienten reimplantiert werden. Das Verfahren ähnelt somit dem bei Darcy et al. („Redirected perforin-dependent lysis of colon carcinoma by ex vivo genetically engineered CTL" J. Immunol., 2000. 164:3705-3712) beschriebenen Verfahren unter der Verwendung von scFv anti-CEA Rezeptor transduzierten ZTL, Perforin und γ-IFN.
Ein weiterer Aspekt der Erfindung betrifft ein Verfahren zur Identifizierung von hdm2- Protein-spezifischen Antigenen das dadurch gekennzeichnet ist, daß hdm2-präsentierende Tumorzellen oder Fraktionen davon mit einer erfindungsgemäßen Wirtszelle unter Bedingungen zusammengebracht werden, bei denen die Tumorzellen oder Fraktionen davon nur dann lysiert werden, wenn der Tumor das hdm2-Protein-spezifische Antigen präsentiert, für welches das exprimierte Polypeptid oder Fusionsprotein spezifisch ist. Ein weiterer Aspekt der Erfindung betrifft ein Verfahren zur Herstellung eines Antikörpers, vorzugsweise eines polyklonalen oder monoklonalen Antikörpers zur Diagnose und Monitoring von mit hdm2-Protein assoziierten Erkrankungen oder zur Identifizierung von pharma- kologisch aktiven Substanzen, bei dem ein Antikörper produzierender Organismus mit einem erfindungsgemäßen Polypeptid oder funktionelle Äquivalente davon oder Teile davon mit mindestens 6 Aminosäuren, vorzugsweise mit mindestens 8 Aminosäuren, insbesondere mit mindestens 12 Aminosäuren immunisiert oder mit einer ein erfindungsgemäßes Polypeptid kodierenden Nukleinsäure vakziniert wird.
Das Verfahren erfolgt nach dem Fachmann allgemein bekannten Methoden durch Immunisieren eines Säugetiers, beispielsweise eines Kaninchens, mit dem erfindungsgemäßen Polypeptid oder den genannten Teilen davon, gegebenenfalls in Anwesenheit von z. B. inkomplettem Freundschen Adjuvans und/oder Aluminiumhydroxidgelen (siehe z. B. Diamond, B. A. et al. (1981) The New England Journal of Medicine, 1344-1349). Die im Tier aufgrund einer immunologischen Reaktion entstandenen polyklonalen Antikörper lassen sich anschließend nach allgemein bekannten Methoden leicht aus dem Blut isolieren und z. B. über Säulenchromatographie reinigen. Monoklonale Antikörper können beispielsweise nach der bekannten Methode von Winter & Milstein (Winter, G. & Milstein, C. (1991) Nature, 349, 293-299) hergestellt werden.
Ein weiterer Gegenstand der vorliegenden Erfindung ist ein Antikörper zur Diagnose und/oder Behandlung von mit hdm2-Protein assoziierten Erkrankungen oder zur Identifizierung von pharmakologisch aktiven Substanzen, der gegen ein erfindungsgemäßes Polypeptid gerichtet ist und mit den erfindungsgemäßen Polypeptiden spezifisch reagiert, wobei die oben genannten Teile des Polypeptids entweder selbst immunogen sind oder durch Kopplung an geeignete Träger, wie z. B. bovines Serumalbumin in ihrer Immunogenität gesteigert werden können. Dieser Antikörper ist entweder polyklonal oder monoklonal, bevorzugt ist ein mono- klonaler Antikörper. Unter dem Begriff Antikörper versteht man gemäß der vorliegenden Erfindung auch gentechnisch hergestellte und gegebenenfalls modifizierte Antikörper bzw. anti- genbindende Teile davon, wie z.B. chimäre Antikörper, humanisierte Antikörper, multifunk- tionelle Antikörper, monovalente bzw. oligovalente Antikörper, bi- oder oligospezifische Antikörper, einzelsträngige Antikörper, F(ab)- oder F(ab)2-Fragmente (siehe z.B. EP-B1-0 368 684, US 4,816,567, US 4,816,397, WO 88/01649, WO 93/06213, WO 98/24884). Die erfindungsgemäßen Antikörper können zur Diagnose und/oder Behandlung von mit hdm2-Protein assoziierten Erkrankungen oder zur Identifizierung von pharmakologisch aktiven Substanzen verwendet werden.
Die vorliegende Erfindung betrifft weiterhin ein Verfahren zur Herstellung eines Arzneimittels zur Behandlung von mit hdm2 -Protein assoziierten Erkrankungen, dadurch gekennzeichnet, daß mindestens eine Nukleinsäure, mindestens ein Polypeptid, mindestens eine Wirtszelle oder mindestens ein Antikörper nach einem der vorgenannten Ansprüche zusammen mit geeigneten Zusatz- und Hilfsstoffen kombiniert wird.
Die vorliegende Erfindung betrifft weiterhin ein nach diesem Verfahren hergestelltes Arzneimittel zur Behandlung von mit hdm2 -Protein assoziierten Erkrankungen, das mindestens eine Nukleinsäure, mindestens ein Polypeptid oder mindestens einen Antikörper gemäß der vorliegenden Erfindung, gegebenenfalls zusammen mit geeigneten Zusatz- und Hilfsstoffen, enthält. Die Erfindung betrifft weiterhin die Verwendung dieses Arzneimittels zur Behandlung von mit hdm2 -Protein assoziierten Erkrankungen.
Die Therapie der von mit hdm2-Protein assoziierten Erkrankungen kann auf herkömmliche Weise, z.B. durch Infusionen oder Injektionen erfolgen, die die erfindungsgemäßen Arzneimittel enthalten. Die Verabreichung der erfindungsgemäßen Arzneimittel kann weiterhin gegebenenfalls in Form von Liposomenkomplexen bzw. Goldpartikelkomplexen erfolgen. Die Behandlung mittels der erfindungsgemäßen Arzneimittel kann aber auch über orale Dosierungsformen, wie z.B. Tabletten oder Kapseln, über die Schleimhäute, zum Beispiel der Nase oder der Mundhöhle, oder in Form von unter die Haut implantierten Dispositorien erfolgen. TTS sind zum Beispiel aus den EP 0 944 398 AI, EP 0 916 336 AI, EP 0 889 723 AI oder EP 0 852 493 AI bekannt. Die erfindungsgemäßen (Poly)peptide und deren Derivate können auch zur aktiven und/oder passiven Immunisierung von Patienten mit Erkrankungen, insbesondere Tumorerkrankungen, die mit hdm2 in Zusammenhang stehen, eingesetzt werden, um die Induktion, Erzeugung und Zunahme vom hdm-spezifischen ZTL zu erreichen und die Tumor- und Leukämiezellen der betreffenden Patienten spezifisch abzutöten. Solche Erkrankungen umfassen zum Beispiel solide Tumorerkrankungen, lymphohämatopoetische Neopla- sien, maligne hämatologische Erkrankungen, auch in Form eines multiplen Myeloms (oder Plastozytoms), eines histiozytischen Lymphoms und eines CML-Blastenschubs. Bei einer besonders bevorzugten Art der Behandlung wird dem Patienten eine oder mehrere Zellen, bevorzugterweise T-Zellen, insbesondere CD8 -T-Zellen entnommen, die dann ex vivo mit einem oder mehreren erfindungsgemäßen genetischen Konstrukten transduziert oder transfiziert werden. Die ex vivo generierten spezifischen T-Zellen können dann anschließend in den Patienten re-implantiert werden. Das Verfahren ähnelt somit dem bei Darcy et al. („Redirected perforin-dependent lysis of colon carcinoma by ex vivo genetically engineered CTL" J. Immunol., 2000, 164:3705-3712) beschriebenen immunotherapeutischen Verfahren bei Colon-Karzinomen unter der Verwendung von scFv anti-CEA Rezeptor transduzierten ZTL, Perforin und γ-IFN.
Die vorliegende Erfindung betrifft weiterhin ein Verfahren zur Herstellung eines Tests zur Auffindung funktioneller Interaktoren in Zusammenhang mit hdm2-Protein assoziierten Erkrankungen, welches dadurch gekennzeichnet ist, daß mindestens eine Nukleinsäure, mindestens ein Polypeptid oder mindestens ein Antikörper gemäß der Erfindung zusammen mit geeigneten Zusatz- und Hilfsstoffen kombiniert wird.
Unter dem Begriff "funktionelle Interaktoren" im Sinne der vorliegenden Erfindung sind alle diejenigen Moleküle, Verbindungen und/oder Zusammensetzungen und Stoffgemische zu verstehen, die mit den erfindungsgemäßen Nukleinsäuren, Polypeptiden oder Antikörpern, gegebenenfalls zusammen mit geeigneten Zusatz- und Hilfsstoffen, unter geeigneten Bedingungen in Wechselwirkung treten können. Mögliche Interaktoren sind einfache chemische organische oder anorganische Moleküle oder Verbindungen, können aber auch Peptide, Proteine oder Komplexe davon umfassen. Die funktioneilen Interaktoren können aufgrund ihrer Wechselwirkung die Funktion(en) der Nukleinsäuren, Polypeptide oder Antikörper in vivo oder in vitro beeinflussen oder auch nur an die erfindungsgemäßen Nukleinsäuren, Polypeptide oder Antikörper binden oder mit ihnen andere Wechselwirkungen kovalenter oder nicht- kovalenter Weise eingehen.
Die Erfindung umfaßt weiterhin einen erfindungsgemäß hergestellten Test zur Identifizierung funktioneller Interaktoren in Zusammenhang von mit hdm2-Protein assoziierten Erkrankungen, der mindestens eine Nukleinsäure, mindestens ein Polypeptid oder mindestens einen Antikörper gemäß der vorliegenden Erfindung, gegebenenfalls zusammen mit geeigneten Zusatz- und Hilfsstoffen, enthält. Oft kann so das pathologische Verhalten der Zellen in vitro nachgeahmt werden und es können Substanzen gesucht werden, die das normale Verhalten der Zellen wieder herstellen und die ein therapeutisches Potential besitzen. Zudem läßt sich dieses Testsystem zum Screening von Substanzen ausnutzen, die eine Interaktion zwischen dem erfindungsgemäßen Polypeptid und einem funktionellen Interaktor inhibieren.
Ein Gegenstand der vorliegenden Erfindung ist auch ein Arzneimittel zur Indikation und Therapie von mit hdm2 -Protein assoziierten Erkrankungen, das eine erfindungsgemäße Nukleinsäure oder ein erfindungsgemäßes Polypeptid und gegebenenfalls geeignete Zusatz- oder Hilfsstoffe enthält, sowie ein Verfahren zur Herstellung eines solchen Arzneimittels zur Behandlung von mit hdm2 -Protein assoziierten Erkrankungen, bei dem eine erfindungsgemäße Nukleinsäure oder ein erfindungsgemäßes Polypeptid mit einem pharmazeutisch annehmbaren Träger formuliert wird.
Als Therapeutika und/oder Prophylaktika kommen insbesondere Impfstoffe, rekombinante Partikel oder Injektionen oder Infusionslösungen in Betracht, die als Wirkstoff (a) das erfindungsgemäße TZR-Rezeptor Polypeptid und/oder seine Derivate und/oder (b) eine erfindungsgemäße Nukleinsäure enthalten und/oder (c) in vitro oder ex vivo erzeugte T- Lymphozyten, die einen spezifisch gegen hdm2 gerichteten TZR enthalten.
Für die gentherapeutische Anwendung beim Menschen ist vor allem ein Arzneimittel und/oder rekombinanter Partikel geeignet, das die erfindungsgemäße Nukleinsäure in nackter Form oder in Form eines der oben beschriebenen gentherapeutisch wirksamen Vektoren oder in mit Liposomen bzw. Goldpartikeln komplexierter Form enthält. Der pharmazeutische Träger ist beispielsweise eine physiologische Pufferlösung, vorzugsweise mit einem pH von ca. 6,0-8,0, vorzugsweise von ca. 6,8-7,8. Insbesondere von ca. 7,4 und/oder einer Osmolarität von ca. 200-400 milliosmol/Liter, vorzugsweise von ca. 290-310 milliosmol/Liter. Zusätzlich kann der pharmazeutische Träger geeignete Stabilisatoren, wie z. B. Nukleaseinhibitoren, vorzugsweise Komplexbildner wie EDTA und/oder andere dem Fachmann bekannte Hilfsstoffe enthalten.
Ein weiterer Gegenstand der Erfindung betrifft ein Verfahren zur Herstellung eines Polypep- tids zur Diagnose und/oder Behandlung von mit hdm2 -Protein in Zusammenhang stehenden Erkrankungen oder zur Identifizierung von pharmakologisch aktiven Substanzen in einer geeigneten Wirtszelle, das dadurch gekennzeichnet ist, daß eine erfindungsgemäße Nukleinsäure auf geeignete Weise exprimiert wird. Das Polypeptid wird so beispielsweise durch Expression der erfindungsgemäßen Nukleinsäure in einem geeigneten Expressionssystem, wie oben bereits beschrieben, nach dem Fachmann allgemein bekannten Methoden hergestellt. Als Wirtszellen eignen sich beispielsweise die E. coli Stämme DHS, HB101 oder BL21, der Hefestamm Saccharomyces cerevisiae, In- sektenzellinien, z. B. von Spodoptera frugiperda, oder die tierischen Zellen COS, Vero, 293, HaCaT, und HeLa, die alle allgemein erhältlich sind.
Ein erfindungsgemäßes Diagnostikum zur Therapie-Überwachung enthält das erfindungsgemäße Polypeptid bzw. die oben näher beschriebenen immunologisch wirksamen Teile davon. Das Polypeptid bzw. Teile davon, die vorzugsweise an eine Festphase, z. B. aus Nitrocellulo- se oder Nylon, gebunden sind, können beispielsweise mit der zu untersuchenden Körperflüssigkeit, z. B. Blut, in vitro in Berührung gebracht werden, um so beispielsweise mit Autoimmunantikörpern reagieren zu können. Der Antiköφer-Peptid-Komplex kann anschließend beispielsweise anhand markierter anti-Human-IgG- oder anti-Human-IgM-Antiköφer nachgewiesen werden. Bei der Markierung handelt es sich beispielsweise um ein Enzym, wie Per- oxidase, das eine Farbreaktion katalysiert. Die Anwesenheit und die Menge an anwesenden Autoimmunantiköφern kann somit über die Farbreaktion leicht und schnell nachgewiesen werden.
Ein anderes Diagnostikum zur Therapie-Überwachung enthält die erfindungsgemäßen Anti- köφer selbst. Mit Hilfe dieser Antiköφer kann beispielsweise eine Gewebeprobe leicht und schnell dahingehend untersucht werden, ob das betreffende Polypeptid in einer erhöhten Menge vorhanden ist, um dadurch einen Hinweis auf mit hdm2-Protein in Zusammenhang stehende Erkrankungen zu erhalten. In diesem Fall sind die erfindungsgemäßen Antiköφer beispielsweise mit einem Enzym, wie oben bereits beschrieben, markiert. Der spezifische Antiköφer-Peptid-Komplex kann dadurch leicht und ebenso schnell über eine enzymatische Farbreaktion nachgewiesen werden.
Ein weiteres erfindungsgemäßes Diagnostikum umfaßt eine Sonde, vorzugsweise eine DNA- Sonde, und/oder Primer. Dies eröffnet eine weitere Möglichkeit, die erfindungsgemäßen Nukleinsäuren, zum Beispiel durch die Isolierung aus einer geeigneten Genbank anhand einer geeigneten Sonde zu erhalten (siehe z. B. J. Sambrook et al., 1989, Molecular Cloning. Als Sonde eignen sich beispielsweise DNA- oder RNA-Fragmente mit einer Länge von ca. 100-1000 Nukleotiden, vorzugsweise mit einer Länge von ca. 200-500 Nukleotiden, insbesondere mit einer Länge von ca. 300-400 Nukleotiden deren Sequenz aus den Polypeptiden gemäß den SEQ ID Nr. 1 bis SEQ ID Nr. 8 des Sequenzprotokolls abgeleitet werden kann.
Alternativ können anhand der abgeleiteten Nukleinsäuresequenzen Oligonukleotide synthetisiert werden, die sich als Primer für eine Polymerase Kettenreaktion eignen. Als Primer eignen sich beispielsweise DNA-Fragmente mit einer Länge von ca. 10-100 Nukleotiden, vorzugsweise mit einer Länge von ca. 15 bis 50 Nukleotiden, insbesondere mit einer Länge von 20-30 Nukleotiden deren Sequenz aus den Polypeptiden gemäß den SEQ ID Nr. 1 bis SEQ ID Nr. 8 des Sequenzprotokolls abgeleitet werden kann.
Der Begriff "kodierende Nukleinsäure" bezieht sich auf eine DNA-Sequenz, die für ein isolierbares bioaktives erfindungsgemäßes Polypeptid oder einen Vorläufer kodiert. Das Polypeptid kann durch eine Sequenz in voller Länge oder jeden Teil der kodierenden Sequenz kodiert werden, solange die spezifische, beispielsweise enzymatische Aktivität erhalten bleibt.
Es ist bekannt, daß kleine Veränderungen in der Sequenz der erfindungsgemäßen Nukleinsäuren vorhanden sein können, zum Beispiel durch die Degenerierung des genetischen Codes, oder daß nicht translatierte Sequenzen am 5' und/oder 3 '-Ende der Nukleinsäure angehängt sein können, ohne daß dessen Aktivität wesentlich verändert wird. Diese Erfindung umfaßt deshalb auch sogenannte "funktionelle Varianten" der erfindungsgemäßen Nukleinsäuren.
Mit dem Begriff "funktionelle Varianten" sind alle DNA-Sequenzen bezeichnet, die komplementär zu einer DNA-Sequenz sind, die unter stringenten Bedingungen mit einer abgeleiteten Referenzsequenz oder Teilen davon, insbesondere der hypervariablen V(D)JC-Region, hybridisieren und eine zu dem entsprechenden erfindungsgemäßen Polypeptid ähnliche oder identische Aktivität aufweisen. Unter "stringenten Hybridisierungsbedingungen" sind solche Bedingungen zu verstehen, bei denen eine Hybridisierung bei 60°C in 2,5 x SSC-Puffer, gefolgt von mehreren Waschschritten bei 37°C in einer geringeren Pufferkonzentration erfolgt und stabil bleibt.
Unter dem Begriff "funktionelle Varianten" im Sinne der vorliegenden Erfindung versteht man Polypeptide, die funktionell mit dem erfindungsgemäßen Polypeptiden verwandt sind, d. h. Strukturmerkmale der Polypeptide aufweisen. Beispiele funktioneller Varianten sind die entsprechenden Polypeptide, die aus anderen Organismen als der Maus, also dem Menschen, bzw., vorzugsweise aus nicht-menschlichen Säugetieren wie z. B. Affen, Schweinen und Ratten stammen. Andere Beispiele funktioneller Varianten sind Polypeptide, die durch unterschiedliche Allele des Gens, in verschiedenen Individuen oder in verschiedenen Organen eines Organismus kodiert werden. Von der vorliegenden Erfindung werden insbesondere auch funktionelle TZR- Varianten erfaßt, die das identische Epitop des hdm2-Polypeptids erkennen und eine spezifische T-Zell Antwort auslösen.
Im weiteren Sinne versteht man darunter auch Polypeptide, die eine Sequenzhomologie, insbesondere eine Sequenzidentität, von ca. 70%, vorzugsweise ca. 80%, insbesondere ca. 90%, vor allem ca. 95% zu dem Polypeptid mit der Aminosäuresequenz gemäß einer der SEQ ID Nr. 1 bis SEQ ID Nr. 8 und/oder zu anhand der Peptidsequenzen abgeleiteten DNA Sequenzen aufweisen. Darunter zählen auch Additionen, Inversionen, Substitutionen, Deletionen, Insertionen oder chemische/physikalische Modifikationen und/oder Austausche oder Teile des Polypeptids im Bereich von ca. 1-60, vorzugsweise von ca. 1-30, insbesondere von ca. 1-15, vor allem von ca. 1-5 Aminosäuren. Beispielsweise kann die erste Aminosäure Methionin fehlen, ohne daß die Funktion des Polypeptids wesentlich verändert wird.
Die Erfindung soll nun weiter anhand der beigefügten Beispiele und Figuren erläutert werden, ohne durch diese eingeschränkt zu werden. Es zeigt:

In den Primersequenzen bedeuten Y = C / T; R = A / G; V = A / C / G; W = A / C / T
Figur 1 zeigt eine FACS-Analyse (siehe 1.4) mittels eines vß6-spezifischen FITC-markierten Antiköφers (Pharmingen). Es wurde sowohl die murine T-Zell-Linie als auch der daraus entstandene Klon 3 übeφriift. Als Isotyp-Kontrolle diente anti-TCR Vα2 - FITC (BD), als Positiv-Kontrolle der anti-pan ßTZR - FITC - Antiköφer (BD). Als Negativ-Kontrolle diente der Influenza-Matrixprotein-spezifische T-Zell-Klon 58.
Figur 2 zeigt eine vß6-spezifische Antiköφer-Blockade des murinen T-Zell-Klons 3 im cytotoxischen 51 Chrom-Freisetzungstest (siehe 1.4). Die Blockade wurde dosisabhängig eva- luiert. Als Negativ-Kontrolle diente zum einen ein anti-Vα3.2 - Antiköφer (BD) und zum anderen der p53 - 264-272 - spezifische T-Zell-Klon 46, der gegenüber dessen eigenes zu erkennendes p53-Peptid gegengeprüft wurde. Als Zielzelle fungierte die TAP-defiziente Zelllinie T2, die exogen mit 10"8 M Peptid beladen wurde.
Figur 3 zeigt Beispiele des Design hdm2-spezifischer Konstrukte auf DNS-Ebene (siehe 2.), wobei muV α/ß murines variables α/ß Gensegment; muC α/ß murines konstantes α/ß Gensegment; huC α/ß humanes konstantes α/ß Gensegment; TM: Gensegment für Transmembrane kodierend; Li: Gensegment für (GGGGS)3-Motiv kodierend und HuCD3ζ humanes CD3ζ-Ketten Gensegment bedeutet. Die für die Klonierung relevanten Restriktions- Schnittstellen sind angegeben.
Figur 4 zeigt dieselben Beispiele des Design hdm2-spezifischer Konstrukte auf Protein-Ebene (siehe 2.), wobei muV α/ß murine variable α/ß-Domäne; muC α/ß murine konstante α/ß- Domäne; huC α/ß humane konstante α/ß-Domäne; SP: murines Wildtyp-Signalpeptid; TM: Transmembrane; Li Linker mit (GGGGS) - Motiv und huζ: humane CD3ζ-Kette mit 3 konsekutiven IT AM - Motiven bedeutet. Die aus den bakteriellen Klonen abgeleiteten Nummern sind für die individuellen Ketten angegeben.
Figur 5 zeigt den Polylinker des Konstrukts muvα-Li-muvß-mucß (siehe 2.3). Der Polylinker ist letztlich über Kpn I und Dra III in das resultierende Konstrukt eingesetzt worden.

Figur 7 zeigt die Fusionsstelle des chimären Doppelketten-TZR in Fusion zur humanen CD3ζ-Kette muvα-hucα-huCD3ζ (siehe 2.4). Das extracytosolische, über eine Disulfidbrük- ke dimerisierende Cystein ist mit relativer Position zum Start-Methionin angegeben.
Figur 8 zeigt die Fusionsstelle des chimären Doppelketten-TZR in Fusion zur humanen CD3ζ-Kette muvß-hucß-huCD3ζ (siehe 2.4). Das extracytosolische, über eine Disulfldbrücke dimerisierende Cystein ist mit relativer Position zum Start-Methionin angegeben.
Figur 9 zeigt die "fluorescence activated cell sorting" (FACS) Analyse der unter 2.) beschriebenen Konstrukte nach retroviraler Transduktion. Dargestellt sind Beispiele durchschnittlicher Transduktions-Effizienzen für OKT3 -aktivierte PBL-Kulturen 6 Tage nach Transdukti- on. Diese liegen im Bereich von 10-25% der eingestezten CD3+-Lymphozyten. Als Negativ- Kontrolle diente ein Transfektions-/Transduktions - Ansatz mit dem leeren retroviralen Vektor pBullet.
Figuren 10 bis 12 zeigen Peptid-Titrationen (siehe 3.4.1) wobei T2-Zielzellen exogen mit dem hdm2-81-88-Peptid in einer Konzentrationsreihe von 1*10" M bis 1*10" M beladen wurden. Als Referenz fungierte der murine T-Zell-Klon 3 hdm2-81-88 (Synonym: P2) der huCD8 x A2Kb - transgenen Maus. Die gezeigten 51 Chrom-Freisetzungstests wurden mit transduzierten PBLs durchgeführt, die über vß6-gerichtete magnetische Zeil-Sortierung (Miltenyi-Biotec) auf über 90% vß6+-T-Zellen angereichert wurden. Figur 10: als Negativ-Kontrolle diente ein Konstrukt eines nicht-funktionellen Einzelketten-T-Zell-Rezeptors (nfTCR), das sich zwar vergleichbar gut auf der Oberfläche der T-Zellen exprimieren ließ und dadurch auch über magnetische Zeil-Sortierung anreicherbar war, aber keine Funktionalität im CTL-Test bewies. Figuren 13 bis 15 zeigen die Erkennung von maligne transformierten Zeil-Linien. Die gewählten Zielzellen waren HLA-A2+. Als Referenz fungierte der murine T-Zell-Klon 3 hdm2- 81-88 (Synonym: P2) der huCD8 x A2Kb - transgenen Maus. Die gezeigten 51Chrom- Freisetzungstests wurden mit transduzierten PBLs durchgeführt, die über vß6-gerichtete magnetische Zeil-Sortierung (Miltenyi-Biotec) auf über 90% vß6+-T-Zellen angereichert wurden. Figur 13: als Negativ-Kontrolle diente ein Konstrukt eines nicht-funktionellen Einzelketten-T-Zell-Rezeptors (nfTCR), das sich zwar vergleichbar gut auf der Oberfläche der T- Zellen exprimieren ließ und dadurch auch über magnetische Zeil-Sortierung anreicherbar war, aber keine Funktionalität im CTL-Test bewies. Beispiele
Es wurden die Gene der T-Zell-Rezeptoren αTZR und ßTZR aus einer doppelt-transgenen Maus huCD8 x A2Kb präpariert, die eine HLA-A2.1 restringierte und hdm2-Protein / 81-88 Peptid-spezifische T-Zell-Antwort vermitteln. Die erhaltenen Gene wurden als Wildtyp (WT) oder modifizierte Konstrukte retroviral in humane periphere Blut-Lymphozyten (PBLs) eingeschleust und die HLA-restringierte Antigen-Erkennung funktionell durch CD8-vermittelte cytotoxische Lyse verschiedener Zeil-Linien im 51 Chrom-Freisetzungstest übeφriift. 1.) Gewinnung der für die α- und ß-Ketten der T-Zell-Rezeptoren kodierenden Gene 1.1) Präparation der cytoplasmatischen RNS
Die cytoplasmatische RNS wurde isoliert, um zum einen gezielt die noch unprozessierte, Intron-haltige RNS, DNS-Bruchstücke als auch ribosomale RNS, die vorwiegend im Zellkern lokalisiert sind, abzutrennen und eine Anreicherung der gewünschten "messenger"-RNS zu erzielen. Protokoll und Materialien entsprechen weitgehend mit kleinen Modifikationen dem "Rneasy Mini Kit" der Firma QIAGEN: für eine Präparation wurden durchschnittlich 50* 106 murine Zellen des hdm2/81-88 - spezifischen T-Zell-Klons 3 aus den Kulturschalen einer "24- well-Platte" vereint und in RPMI 1640 (Biowhittaker) serumfrei gewaschen. Die Zellen wurden unter RNase-freien Bedingungen zu je 12,5* 106 Zellen pro Inkubationsröhrchen aliquo- tiert und im Zellmembranlyse-Puffer (4°C-gekühlt) für 20 min auf Eis inkubiert:

Für diesen Puffer wie alle weiteren wäßrigen Lösungen, die angesetzt werden mußten, wurde DEPC-behandeltes und doppelt-autoklaviertes deionisiertes (MilliQ von Millipore) Wasser benutzt.
Die weiteren Schritte erfolgten gemäß einem dem Fachmann geläufigen Verfahren mit Hilfe des QIAGEN-ProtokoUs unter Rnase-freien Bedingungen bei Raumtemperatur und werden kurz beschrieben:
Die milde solubilisierten und aliquotierten Zellen wurden bei 300g / 2' (Minuten) / 4°C zentri- fugiert und der Überstand abgenommen. Es wurden 600 μl eines ß-Mercaptoethanol- (MSH) haltigen (1.45 mM) "RLT"-Puffers hinzugesetzt, gut gemischt und 430 μl 96% ETOH unter Mischen hinzugesetzt. Es wurde die Lösung je Aliquot auf eine Silika-Minispin-Säule gegeben und für 15" (Sekunden) bei 10.000 Upm (Umdrehungen pro Minute) in einer Eppendorf- Tischfuge zentrifugiert. Es erfolte jeweils ein Wasch in 700 μl "RW1 " sowie in 500 μl 77%- ETOH-haltigen "RPE"-Puffer. Abschließend wurden 500 μl des identischen "RPE"-Puffers hinzugegeben und die Säule für 2' bei 14.000 Upm maximal getrocknet. Die Elution der RNS erfolgte mit 54 μl Rnase-freiem Wasser bei 10.000 Upm für 2'. Die Ausbeuten lagen bei 30 μg RNS pro 12,5* 106 murinen T-Zellen. Die RNS wurde zu 6 μg / 10 μl bei -20°C weggefro- ren.
1.2) Amplifikation des ß-Ketten-Gens mittels gekoppelter RT-PCR
Die TZR-kodierende RNS mußte spezifisch in DNS umgeschrieben und amplifiziert werden, bevor diese kloniert werden konnte. Hierzu wurde methodisch die RT-PCR herangezogen, die im gleichnamigen Kit "Titan One Tube RT-PCR-System" der Firma Röche (1888 382) implementiert ist. Es wurde das "Ein-Schritt-Protokoll" gewählt, bei dem die Reverse Transkription ohne weitere Intervention von der Polymerase-Kettenreaktion gefolgt wird. Bei den verwendeten Enzymen handelte es sich um die AMV - Reverse Transkriptase und ein Gemisch der Thermus Aquaticus / Pwo - thermostabilen Polymerase ("Expand™ High fidelity" Enzym-Mix), die in einem universalen Reaktionspuffer zum Einsatz kamen. Da die Transkriptionsrate der TZR-Gene und aufgrund der Varianz der TZR-Gene die Konservierung der für die variablen Domänen kodierenden Gensequenzen gering ist, schloß sich eine "nested PCR" (Polymerase-Kettenreaktion) an, um die Spezifität und die Signalstärke der amplifi- zierten DNS zu erhöhen. Die Variabilität des v-Gensegmentes, dessen Subfamilienzugehörig- keit a priori nicht vorhergesagt werden konnte, wurde mit einem degenerierten Primer (for_bTCR_v4; SeqlD Nr. 9) versucht zu erfassen, der einer Basensequenz für die ersten konservierten Aminosäuren nach dem Signalpeptid kodierend entsprach. Ein artifizielles Gensegment, das für ein murines TZR-Signalpeptid kodiert, sollte ursprünglich über synthetische Oligonukleotide vorgeschaltet werden, um eine Direktion des in vivo synthetisierten Proteins in das Lumen des Endoplasmatischen Retikulums zu gewährleisten. Es wurde ein innerhalb des konstanten Gensegmentes revers hybridisierender Primer (rev_bTCR_c4; SeqlD Nr. 10) gewählt, um der zusätzlichen Varianz der beiden möglichen c-Gensegmente (c- ßl oder c-ß2) der ß-Kette, die einige Basenunterschiede an deren 3'-Ende aufweisen, zu entgehen. Die Oligonukleotide der "nested PCR" besaßen neben der TZR-Komplementarität zusätzlich an dessen 5 '-Ende Sequenzen der Restriktionsschnittstellen für EcoRI bzw. Ascl. Der in Vorwärtsrichtung hybridisierende Primer (for_Εco_bTCR_v5; SeqlD Nr. 11) der nested "PCR" war in der Sequenz des v-Gensegmentes deckungsgleich zum degenerierten RT-PCR - Primer, wohingegen der zum c-Gensegment komplementäre in Rückwärtsrichtung hybridisierende Primer (rev_Asc_bTCR_c6; SeqlD Nr. 12) mehr als 120 Basen relativ zum reversen 1. Primer des TZR-Leserahmens einwärts versetzt lag, so daß es sich genau um eine "3'- located nested PCR" handelte. Hierdurch wurde die Spezifität der PCR erhöht, da in 2 unabhängigen PCR-Schritten an 2 unterschiedlichen Orten ein sequenzspezifisches Erkennungsereignis stattgefunden haben muß. Das komplette c-Gensegment wurde mittels eines reversen degenerierten Primers (rev_Asc_bTCR_c2; SeqlD Nr. 14) zum Erfassen der beiden potentiell möglichen cßl-/cß2-Gensegmente und eines in Leserichtung entsprechenden Primers (for_Eco_bTCR_c2; SeqlD Nr. 13) des cß -Gensegmentes über RT-PCR ohne nachgeschaltete "nested PCR" erfaßt und kloniert. Im Überlappungsbereich der beiden Amplifikate im cß- Bereich befand sich eine singuläre Ncol-Schnittstelle, mittels derer das trunkierte vß-/cß- und das vollständige cß-Gensegment in einer anschließenden Subklonierung zusammengefügt werden konnten.
Das Pipettierschema und das PCR-Protokoll der RT-PCR entsprach prinzipiell dem durch die Firma Röche beschriebenen Verfahren. Die anschließende "nested PCR" erfolgte analog unter Ersatz der Komponenten Rnase-Inhibitor und DTT durch autoklaviertes Wasser. Statt der 2 μl RΝA ( 0,1 -1 μg) wurde 2 μl des Amplifikats der RT-PCR eingesetzt. Die technischen Voraussetzungen des PCR-Geräts ermöglichten den simultanen Test verschiedener Hybridisie- rungstemperaturen im 2. Schritt des klassischen PCR-Zyklus in einem Temperaturgradienten von 35°C - 55°C in Intervallen für 5 ausgesuchte Temperaturen von 36°C, 39°C, 44°C, 52°C und 56°C. Für die höheren Temperaturen von 44-56°C konnten singuläre Banden der gewünschten Länge erzielt werden, dessen Intensität durch die 2. PCR verstärkt wurden und ausreichend waren, um in Vektoren inseriert zu werden.
Die Sequenzierung von 8 bakteriellen Klonen ergab einige randomisierte Basenaustausche, die auf der zu anderen Polymerasen vergleichbar hohen Fehlerrate des Expand™ - Enzym- Mixes beruhten. Die gewählten Klone trugen zudem alle die Sequenzinformation für das Si- gnalpeptid und einen kurzen Bereich des nicht-kodogenen, 5'-untranslatierten Bereiches von weniger als 100 Basen, so daß davon ausgegangen werden konnte, daß der v-Gensegment- spezifische degenerierte Primer eine ähnliche Sequenz flußaufwärts erkannte und die zusätzliche Sequenz des Wildtyp-Signalpeptids lieferte.
1.3) Amplifϊkation des α-Ketten-Gens mittels RACE-PCR
Für die Sequenzermittlung der α-Kette war die RACE-PCR die zum Ziel führende Strategie: das analog dem oben beschriebenen Ansatz konzipierte degenerierte Oligonukleotid erbrachte ausschließlich die kodogene Sequenz eines aufgrund einer Leserasterverschiebung in der CDR3-Schleife nicht-funktionellen T-Zell-Rezeptors. Die allele Exklusion ist nicht in dem selben Maße valid wie für die ß-Kette des TZR. Die RACE-Technologie ist im gleichnamigen System der Firma Röche (1 734 792) etabliert und muß mit αTZR-Gen-spezifischen Oligonu- kleotiden ergänzt werden: hierzu wird im Rahmen der "5'-RACE-PCR" die 5'-lokalisierte Varianz der kodogenen Sequenz umgangen, indem eine bekannte, kurze Adenin- Oligonukleotid-Sequenz am 3'-Ende der revers transskribierten cDNS-Sequenz mittels der terminalen Transferase generiert wird, die in Folge als Erkennungssequenz für einen in Leserichtung 5'-lokalisierten PCR-Primer fungiert. Die reversen PCR-Primer liegen innerhalb der invarianten Sequenz, die für die konstante Domäne kodiert. Für die reverse Transkription wurde der Primer rev_Asc_aTCR_cl (SeqlD Nr. 15) verwendet, der zusammen mit for_Eco_aTCR_c6 (SeqlD Nr. 18) auch zur Amplifikation via RT-PCR des Volllängen cα- Segmentes verwendet wurde. Es wurde das Konzept einer konsekutiven 2-Schritt-"nested PCR" mit Hilfe der Primeφaare "Oligo d(T)-Ankeφrimer" (Komponente des RACE-PCR- Kits) / rev_aTCR_c5 (SeqlD Nr. 16) und "PCR Ankeφrimer" (Komponente des RACE- PCR-Kits) / rev_Asc_aTCR_c4 (SeqlD Nr. 17) verfolgt, so daß auch hier die Subklonierung der vollständigen Sequenz, die für die konstante Domäne kodiert, sich anschloß. Die PCR- Primer mußten so liegen, daß im Überlappungsbereich der amplifizierten Genabschnitte die singuläre Ncol-Schnittstelle lag. Die trunkierte, das vα-Gensegment-enthaltende PCR- Amplifikat wurde über 5'Saä und 3' -Ascl in pΝEB193 (New England Biolabs) kloniert, wohingegen das konstante Gensegment über EcoRI und Ascl in pNΕB193 inseriert wurde. Die Amplifikate wurden klassisch kloniert (J. Sambrook et al., 1989, Molecular Cloning. A Labo- ratory Manual 2nd edn., Cold Spring Harbor Laboratory, Cold Spring Harbor) mittels der Reinigung über LMP-Gele und Phenol/Chloroform-Extraktion, bevor diese restriktionsenzyma- tisch verdaut und die Enden mit Hilfe des "High Pure PCR Product Purification Kit" (Röche, 1 732 668) abgetrennt wurden.
Das Mehrschrittprotokoll lehnt sich weitgehend an das kommerzielle Protokoll an unter Zuhilfenahme eines programmierbaren PCR-Gerätes: die reverse Transkription, die Transferase- Reaktion und die anschließende 2-Schritt "nested PCR" sind im PCR-Gerät "MasterCycler" der Firma Eppendorf durchgeführt worden. Im Rahmen der spezifischen α-Ketten- Amplifikation kam die Gradiententechnologie im Hybridisierungsschritt für die Temperaturen 39°C, 44°C, 52°C und 56°C zur Anwendung. Es wurde die R/w-Polymerase (Stratagene) gewählt, da zu diesem Zeitpunkt dieses als das Enzym mit der geringsten Fehlereinbaurate galt. Die 1.PCR ergab bereits eine schwache Bande der zu erwartenden Größe von 0.9 kB im ge- samten Temperaturspektrum, deren Intensitäten durch die nachfolgende PCR verstärkt werden konnten und zudem auftretende "Primer-Dimer" herausverdünnt werden konnten. In der abschließenden PCR ergab sich eine Verkürzung des Amplifikates um 70 Bp gemäß der theoretischen Lage der versetzt hybridisierenden Primer. Im Agarose-Gel zeichnete sich eine Doppelbande von 100 Bp Längendifferenz ab, die auf das Vorhandensein der funktionellen und der nicht-funktionellen α-Kette hinwies. Die Banden wurden gemeinsam im präparativen Gel ausgeschnitten, um beide Amplifikate in der anschließenden Klonierung zu erfassen. Es wurden 7 Klone gefunden, die für einen funktionellen Leserahmen kodierten und identische Sequenzen besaßen.
1.4) Überprüfung der gefundenen Subfamilienzugehörigkeiten
Die Existenz eines vß6- bzw. vαlO-Gensegmentes in dem murinen hdm2-Protein / 81-88 spezifischen T-Zell-Klon 3 wurde auf verschiedenen Wegen verifiziert:
a) RT PCR mittels eines vß6-spezifischen Primers bzw. eines vαlO-spezifischen Primers. Die RNS wurde zuvor aus dem murinen hdm2-P2 spezifischen T-Zell-Klon 3 gereinigt, indem die spezifischen T-Zellen über anti-huCD8 magnetisch markierte Antiköφer von den murinen "feeder"-Zellen getrennt wurden. Als Negativ-Kontrolle diente ein Influenza- Matrixprotein-(FluMl 58-66) spezifischer T-Zell-Klon. b) FACS-Analyse (Figur 1) mittels eines vß6-spezifischen FITC-markierten Antiköφers (Pharmingen). Es wurde sowohl die murine T-Zell-Linie als auch der daraus entstandene Klon 3 übeφrüft. Als Positiv-Kontrolle diente ein FITC-markierter Antiköφer, der den ß- Ketten-Anteil subfamilienunabhängig erkennt. c) vß6-spezifische Antiköφer-Blockade des murinen T-Zell-Klons 3 im cytotoxischen 1 Chrom-Freisetzungstest (Figur 2).
2.) Beispiele des Design hdm2-spezifischer Konstrukte (Figur 3 [SEQ ID Nrn 38-45] und 4 [SEQ ID Nrn 1-8])
Der retrovirale Vektor pBullet, ein funktionelles Derivat von pStitch (Weijtens et al., 1998) gehört zu den sogenannten Spleiß- Vektoren, in denen das Transgen im selben Sequenzen- Kontext relativ zu den Spleiß-Akkzeptor- und Donor-Elementen SA und SD kloniert werden muß wie das env-Gen, um eine maximale Expression zu gewährleisten. Hierzu wird über entsprechende Primer eine Ncol-Schnittstelle innerhalb des Startcodons "ccATGg" konzipiert. Dies veränderte für beide Wildtyp-Ketten αTZR und ßTZR die 2. Aminosäure von Lys nach Glu bzw. Asn nach Asp. Am 3'-Ende der Leserahmen wurde die singuläre BamRl- Schnittstelle gewählt. Die Restriktionsenzyme und modifizierenden Enzyme wurden von New England Biolabs (NEB) bezogen. Bei der thermostabilen Polymerase handelte es sich zumeist um die native oder klonierte R/w-DNS-Polymerase (Pyrococcus furiosus, Stratagene) oder um die R/5c-DNS-Polymerase (Pyrococcus sp., Life Technologies)
2.1) Klonierung der murinen Wildtyp-Gene αTZR (muvα-mucα; SeqlD Nrn. 1-38) und ßTZR (muvß-mucß; Seq ID Nrn. 2-39)
Für den Fall, daß die Ncol-Schnittstelle nicht intern existierte (partiell humanisierte Ketten), konnten die Gensegmente direkt über Primer amplifiziert und kloniert werden, die die Sequenzen für Ncol enthielten (for_ΝcoI_aTCR_4, SeqlD Nr. 19; for_NcoI_bTCR_4, SeqlD Nr. 21).
Da die Ncol-Schnittstelle für die Wildtyp-Ketten im Gensegment für die konstanten Domänen vorkam, wurden die kodogenen Leserahmen über Oligonukleotide (for_ΝcoI_aTCR_5b, SeqlD Nr. 20; for_NcoI_bTCR_5b, SeqlD Nr. 22) amplifiziert, die 5'-terminal derart verkürzte, stumpfendige Ncol-Schnittstellen besaßen, so daß diese nach stumpfendigem Auffüllen Ncol-geschnittener Vektor-DΝS mit Hilfe der T4-DΝS -Polymerase mit letzteren zu vollständigen Ncol-Schnittstellen komplementierten. rev_aTCR_BamHI (SeqlD Νr. 23) und rev_bTCR_BamHI_l (SeqlD Νr. 24) wurden als abschließender reverser Primer für alle murinen Doppelketten- und Einzelketten-Konstrukte benutzt.
2.2) Generierung der chimären, partiell humanisierten TZR-Gene hu αTZR (muvα- hucα; SeqlD Νr. 40) und hu ßTZR (muvß-hucß; SeqlD Νr. 41)
Für eine funktionelle Chimärisierung der murinen TZRs sollten 2 Kriterien erfüllt werden: a) die murinen konstanten Domänen sollten derart gegen die entsprechenden humanen Domänen ausgetauscht werden, so daß diese Domänen in ihrer Immunglobulin-ähnlichen Faltungsstruktur vollständig erfaßt werden. Ebenso waren die vollständigen variablen Domänen zu erfassen, um keine Beeinträchtigung der in dieser Domäne nahezu car- boxyterminal liegenden CDR3 -Schleife zu verursachen. b) Die Fusion hatte derart zu erfolgen, daß der Übergang möglichst kein Νeoantigen generierte, das zu einer ungewollten Immunreaktion führen könnte. Im Fall von b) ergaben sich im allgemeinen für beide Ketten im Vergleich humaner mit muri- ner Sequenzen folgende Eigenschaften: in einem Bereich von bis zu 8 Aminosäuren hinter dem letzten Konsensusmotiv "GxG", das die CDR3-Schleife abschließt, sind die Positionen relativ stark konserviert. Dieser Bereich wird durch eine hochkonservierte Sequenz von bis zu 11 Aminosäuren abgeschlossen. Hiernach beginnen die murinen von den humanen Sequenzen, für jede Spezies hoch konserviert, abzuweichen. Der zentrale Bereich liegt in der ausgedehnten Schleife, die die variable Domäne mit der konstanten verbindet (siehe Kriterium a)). Im Idealfall war die Chimärisierung innerhalb einer hoch konservierten Aminosäure (AS)- Triplett-Sequenz vorzunehmen.
Im Fall des αTZR bestand die hochkonservierte Triplettsequenz aus "Q-N-P" (Base 400-408 relativ zum Startcodon des murinen αTZR). An Position 407-415 schloß sich eine singuläre /wNI-Schnittstelle an, die zur Fusion genutzt werden konnte: die 3'-versetzte Lage der AlwNl bedingte, daß statt des in humanen Sequenzen konservierten Aspartats das in murinen Sequenzen konservierte Glutamat vorlag. Dieser konservative Aminosäureaustausch wurde als wenig immunogen und damit tolerabel erachtet. Der chimäre, hoch-konservierte Bereich lautete Il33-Q-N-P-E-P-A-V-Y-Q-L-R144 in dem R144 die erste humanspezifische Aminosäure darstellt. I befindet sich an der neunten Position hinter dem die CDR3 -Schleife abschließenden Konsensusmotiv "GxG".
Die Klonierungsstrategie sah vor, die LMP-Agarose-isolierten EcoRI/ /wNI-geschnittenen vα-Segmente mit den AlwNl/BamHl- verdauten cα-PCR-Amplifikaten zu ligieren, die gemischten Ligationsprodukte mit EcoRI und BamRl nachzuschneiden und das präparierte Li- gationsprodukt der potentiell korrekten Fragmentlänge in den Εxpressionsvektor pcDNA3.1(- ) (Invitrogen) zu klonieren. Ein asymmetrischer Testverdau ergab die Klone mit der gewünschten Abfolge der Ligationsedukte.
Die humanen konstanten Gensegmente cα bzw cß wurden durch klassische RT-PCR, wie sie als Bestandteil des oben beschriebenen Roche-Kits implementiert ist, mit anschließender "nested" PCR aus der humanen Leukämie-Zelllinie Jurkat isoliert.
Im Fall des humanen cα- Gensegmentes mußte mit Hilfe mutagener PCR-Primer an 2 Positionen zum einen die .4/wNI-Schnittstelle (T nach A; s. Tabelle unten) generiert werden und zum anderen die Kompatibilität zur murinen Sequenz hergestellt werden (c nach a; s. Tabelle unten), da die mittleren 3 Positionen der /wNI-Schnittstelle degeneriert sein dürfen:

Die in der murinen Sequenz natürlich vorkommende -4/wNI-Schnittstelle ist fett dargestellt, die 3 '-versetzte Schnittposition ist unterstrichen. Das degenerierte Triplett ist klein geschrieben. Die erforderlichen Basenaustausche für den humanen „Primer" zur Beibehaltung einer Maussequenz-kompatiblen /wNI-Schnittstelle sind fett dargestellt. Die 5 '-initiale Base des mutagenen Primers ist kursiv. Die resultierende chimäre AS-Sequenz ist fett dargestellt (s. Text); das hoch-konservierte AS-Triplett ist zudem unterstrichen.
Die Amplifikation des cα-Fragmentes erfolgte über eine „nested" PCR. Die Mutationen wurden in den „nested" Primer (for_hucaTCR_AlwNI_3; SeqlD Nr. 27) gelegt, wohingegen der RT-PCR - Primer (for_hucaTCR_2; SeqlD Nr. 25) noch der humanen Wildtyp-Sequenz entsprach. Der reverse „nested" Primer rev_hucaTCR_BamHI_l (SeqlD Nr. 28) besaß sowohl das Stopcodon als auch die klonierbare RαmHI-Schnittstelle, wohingegen der reverse RT-PCR-Primer (rev_hucaTCR_2; SeqlD Nr. 26) den 3 '-liegenden nicht-kodogenen Bereich erkannte.
Im Fall des ßTZR kam ebenso das "nested" PCR-Prinzip gekoppelt an eine stumpfendige Li- gation zur Anwendung (SeqlD Nrn. 29-32). Die hoch-konservierte Triplettsequenz bestand aus "E-D-L" (Base 400-408 relativ zum Startcodon des murinen ßTZR). Deckungsgleich befand sich eine das variable Gensegment umfassende und singuläre BstYl- Schnittstelle (Base 402-407). Ein für das humane konstante Domänen-Gensegment spezifischer Primer (for_hucbTCR_4; SeqlD Nr. 31) sollte über sein 5'-terminales Ende, das mit einem phos- phorylierten „T" die letzte Base der potentiell palindromen Erkennungssequenz von BstYl darstellt, das mit ÄstYI-verdaute (produziert überhängende 5 '-Enden, die mit der T4-DNS- Polymerase komplementär unter Zugabe von dNTP's aufgefüllt und nicht abgedaut werden) und stumpfendig aufgefüllte 3 '-Ende des murinen variablen Domänen-Gensegmentes komplementieren, so daß der richtige Leserahmenkontext erhalten blieb. Das vß-Fragment wurde mit EcoRI nachgeschnitten und aus einem LMP-Gel isoliert, das cß-PCR-Amplifikat wurde als zusätzlich enzymatisch phosphoryliertes (T4-Polynukleotid-Kinase, NΕB) einseitig stump- fendiges BamHl - Fragment in der Ligation eingesetzt. Die gemischten Ligationsprodukte wurden mit EcoRI / BamHl nachgeschnitten und das Fragment mit der potentiell richtigen Fragmentlänge in pcDNA3.1(-) kloniert. Ein asymmetrischer Testverdau ergab die Klone mit der gewünschten Abfolge und Orientierung der Ligationsedukte.

Der chimäre hoch-konservierte Bereich lautete E -D-L-N in dem N die erste human- spezifische Aminosäure darstellt. E133 befindet sich an der siebten Position hinter dem konservierten "GxG"-Motiv, das die CDR3-Schleife abschließt.
2.3) Generierung der Einzelketten-TZRs muvα-Li-muvß-muccß(SeqID Nr. 42) und muvß-Li-muvα-muccα(SeqID Nr. 43)
Einzelketten-TZRs bestehen aus der Fusion der variablen Domänen vα mit vß und einer angefügten konstanten Domäne cα oder cß (Eshar et al., 1993). Der Vorteil solcher Konstrukte liegt in der definierten 1 : 1 -Stöchiometrie der beiden variablen Domänen in einem Konstrukt und der definierten Reaktivität aufgrund der auszuschließenden heterogenen Paarung endogener mit exogener Doppelketten-TZRs. Die variablen Domänen wurden über einen flexiblen Polylinker eines redundanten (GGGGS)3-Motivs verbunden. Die Enden des synthetischen Oligonukleotids beherbergten singuläre Schnittstellen, mit denen das carboxyterminale Ende der einen variablen Domäne, hinter der essentiellen CDR3-Schleife gelegen, mit dem Anfang der anderen variablen Domäne, hinter der Sequenz, die für das signalpeptid kodiert, liegend verknüpft werden konnten. Kozeptionell wurden fünf Kriterien beachtet: a) der Polylinker mußte lang genug sein, um die Enden der variablen Enden zu verbinden. Hierzu wurden murine TZR-Kristallstrukturen einer MHC-H-2Kb-Spezifität zu Rate gezogen. b) der Polylinker hatte die Enden derart zu verbinden, so daß dieser zum einen die Spezifität der CDR3-Schleife nicht beeinträchtigte, zum anderen das Signalpeptid der nachgeschalteten variablen Domäne hinreichend eliminierte unter Beibehaltung ihrer funktionellen Struktur. Um dies zu bewerkstelligen, wurden in dem Polylinker Aminosäure-Sequenzen der jeweiligen variablen Domäne mit aufgenommen für den Fall, in dem die singulären Schnittstellen nicht ideal lagen. c) Der Polylinker mußte zum einen flexibel sein, was mit Hilfe einer sterisch begünstigenden Aneinanderreihung von Glycin-Resten bewerkstelligt werden sollte, zum anderen hinreichend löslich sein, um Aggregatbildung und Präzipitation zu vermeiden: die Hydroxyl- Gruppe des Serin-Restes bildet Wasserstoff-Brücken mit solvatisierenden Wasser- Molekülen aus und bildet lokale Dipole. d) um die Transkription im richtigen Leserahmen nicht zu beeinträchtigen, wurden die kodo- genen Tripletts für die Glycin-Reste variiert ("GGC" und "GGA"). e) der Polylinker mußte dieselben internen singulären Schnittstellen besitzen wie die Wildtypsequenzen, um in einem 2-Schritt-Klonierungsverfahren die Zwischenprodukte aufzuschneiden und die nahezu vollständigen Wildtypsequenzen über diese singulären Schnittstellen 3'-terminal anzufügen.
Unter diesen Voraussetzungen war es möglich zwei Orientierungen der variablen Domäne zuzulassen :
Im Fall des Konstruktes muvα-Li-muvß-mucß (SeqlD Nr. 42; Figur 5) wurde die singuläre Schnittstelle Kpnl hinter dem letzten Konsensus-Motiv "GxG" der variablen vα-Domäne und Drälll als die die Signalpeptid kodierende Sequenz abschließende Restriktionsschnittstelle in der vß-Domäne gewählt. In einem ersten Schritt wurde das hybridisierte, doppelsträngige Oligonukleotid beidseitig mit Kpnl geschnitten und zwischen die vα-kodierende und cα- kodierende Sequenz des Wildtyp-αTZR kloniert. Hiernach wurde das chimäre Zwischenprodukt mit DrάllllBamHl aufgeschnitten und die die ßTZR-kodierende Sequenz, die über einen DrallllBamHl- Verdau ihre die Signalpeptid-kodierende Sequenz verloren hatte, hineinklo- niert. Das erste Glycin des Polylinkers lag an Position 6 relativ zum aminoterminalen Gly des "GxG"-Konsensus-Motivs in vα, das letzte Serin des Polylinkers, aufgrund einer zwischengeschalteten stumpfendigen Ligation über ein Arginin verbunden, vor Position Asp22 der vß- Domäne. Im Fall des Konstruktes muvß-Li-muvα-mucα (SeqlD Nr. 43; Figur 6) wurde die singuläre Schnittstelle Sc l hinter dem letzten Konsensus-Motiv "GxG" der variablen vß-Domäne und Bsgl als die die Signalpeptid kodierende Sequenz abschließende Restriktionsschnittstelle in der vα-Domäne gewählt. In einem ersten Schritt wurde das hybridisierte, doppelsträngige Oligonukleotid beidseitig mit Scαl geschnitten und zwischen die vß-kodierende und cß- kodierende Sequenz des Wildtyp-ßTZR kloniert. Hiernach wurde das chimäre Zwischenprodukt mit BsgllBamHl aufgeschnitten und die die αTZR-kodierende Sequenz, die über einen

ihre die Signalpeptid-kodierende Sequenz verloren hatte, hineinkloniert. Das erste Glycin des Polylinkers lag an Position 6 relativ zum aminoterminalen Gly des "GxG"-Konsensus-Motivs in vß, das letzte Serin des Polylinkers vor Position Val24 der vα- Domäne.
2.4) Generierung der Doppelketten-TZRs in Fusion zur humanen CD3ζ-Kette muvα- hucα-huCD3ζ (SeqlD Nr. 44; Figur 7) und muvß-hucß-huCD3ζ (SeqlD Nr. 45; Figur 8)
Die Signaltransduktion der Doppelketten-TZRs sollte optimiert werden. Hierzu fusionierte man die jeweiligen Doppelketten-α/ßTZRs an die nahezu vollständige CD3ζ-Untereinheit des CD3 -Komplexes. Diese Fusion bewerkstelligt gemäß verschiedener Publikationen (Eshar et al., 1994) eine effiziente und CD3 -unabhängige Kopplung des TZR an die in der Kaskade flußaufwärts bzw. flußabwärts liegenden Effektoren Lck56 bzw. ZAP-70.
Es wurde eine modifizierte Ligations-PCR-basierende Technik etabliert, mit deren Hilfe man unabhängig von singulären Schnittstellen beliebige Fusionen zur Herstellung chimärer Proteine erzeugen konnte und die Spezifität der erhaltenen PCR-Amplifikate deutlich erhöht wurde im Vergleich zu einer konventionellen Ligations-PCR. Die genetische Fusion wird über einen reversen, chimären Primer (rev_huca-hu_tm_Zeta, SeqlD Nr. 36, Figur 7; rev_hucb- hu_tm_Zeta; SeqlD Nr. 37, Figur 8) definiert, der mit einem Vorwärts-Primer (T7_for; SeqlD Nr. 33) als PCR-Amplifikat den "Mega-Primer" produziert: in einem ersten, konventionellen Ligations-PCR-Schritt wird für den 5'-liegenden Fusionspartner ein "Megaprimer" generiert, der zudem als Sequenzanhängsel eine kurze für den 3 '-liegenden Fusionspartner spezifische Sequenz trägt. Um die Spezifität der PCR-Signale deutlich zu erhöhen, werden bereits in der ersten PCR Primer verwendet, die außerhalb des kodogenen, chimären Amplifi- kates an den vorgelegten „Templates" hybridisieren (T7_for; T7_rev; SeqlD Nr. 33, 34), um dann in der zweiten PCR inwärts-liegende ("nested") Primer zu verwenden. In dieser liefert der Mega-Primer zusammen mit einem auf den 3 '-liegenden Fusionspartner hybridisierenden reversen "nested" Primer (rev_hZeta_BamHI_4; SeqlD Nr. 35) ein Zwischenprodukt, das mit einem dritten, am 5'-Ende "nested" liegenden Vorwärts-Primer (for_aTCR_NcoI_5b; for_bTCR_NcoI_5b; SeqlD Nr.s 20, 22) ein an den Enden getrimmtes PCR-Amplifikat liefert.
Der Fusionspunkt wurde auf die die Disulfidbrücken-bildenden Cysteine festgelegt: hierdurch verloren die TZRs ihre cytoplasmatische Sequenz, die Transmembrane und einen kurzen Bereich extracytoplasmatisch liegender Aminosäuren bis zum dimerisierenden Cystein, die humane D -Kette verlor ebenso einen kurzen extraycytoplasmatischen Abschnitt, sollte aber exklusive ihres Cystin-bildenden Cysteins für den transmembranen und cytosolischen Bereich inkusive der drei bivalenten "ITAM"-Motive kodieren. Der reverse chimäre Primer kodiert für die 3'-terminal liegenden 21 Basen des jeweiligen partiell humanisierten TZR und für die 5'-terminal liegenden 21 Basen der humanen CD3ζ-Kette (Figur 7). Die chimären Konstrukte sind von der konstanten Domäne an humanisiert.
3.) Transduktion humaner peripherer Blut Lymphozyten (PBLs)
Zur Transduktion humaner peripherer T-Lymphozyten wurde ein funktionelles Derivat des pStitch-Systems (Weijtens et al., 1999), verwendet. Die zur Veφackung notwendigen retroviralen Gene werden über individuelle Plasmide im Wege einer Cotransfektion der Veφak- kungszellinie 293T kodiert (Soneoka et al., 1995): pHit60 kodiert für die gag-pol - Struktur- und Polymerase - Gene aus dem Moloney murinen Leukämie Virus (MoMuLV), pColt-Galv für das env - Hüllprotein des "gibbon ape leukemia virus", das imstande ist, an den Na+- /Phosphat-Synporter Pit humaner Zellen zu binden und damit letztere zu transduzieren. Die chimären Viruspartikel besitzen somit einen amphotropen Pseudotyp und können verschiedene Säugerzellen, außer Maus, transduzieren.
3.1) Transfektion der Verpackungszelllinie 293T
Die isolierten bakteriellen Klone der in das pStitch-Derivat klonierten T-Zell-Rezeptor-Gene wurden über Plasmid-Präparationen, die eine Entfernung residueller Endotoxine versprechen (Qiagen, Produkt 12362), gereinigt und zu 1 μg/μl eingestellt. Die DNS wurde über die Cal- ciumphosphat-Präzipitation transient in die Veφackungszelllinie 293 T eingeführt (GiboBRL- Life Technologies, Produkt 18306-019). Hierbei werden im Rahmen der WT- oder modifizierten Doppelketten-T-Zell-Rezeptoren αTZR und ßTZR bis zu 80 μg DNS eingesetzt: 20 μg αTZR - Konstrukt 20 μg ßTZR - Konstrukt 20 μg pColt-Galv 20 μg pHit 60
Im Fall der Einzelketten-TZRs werden 60 μg DNS eingesetzt. 293T wurde in einem modifizierten DMEM-Medium (DMEM/H) kultiviert:
DMEM, 4,5% Glukose (BioWhittaker)
10% hitzeinaktiviertes FKS
2mM Glutamin lx Penicillin/Streptomycin lx nicht-essentielle Aminosäuren
25 mM HEPES
Am Vortag der Transfektion wurden die Zellen 293T auf 0,9*106 Zellen pro T25-Flasche und Transfektionsansatz in 5ml DMEM/H ausgesät. 4 h vor Transfektion wurde das Medium gegen frisches, auf Raumtemperatur (RT) erwärmtes DMEM-H (3ml) ersetzt. Die Transfektion erfolgte laut Vorschrift des kommerziellen Protokolls (Life Technologies). 1ml des Transfek- tionsansatzes wurde zu der jeweiligen Flasche unter vorsichtigem Hinzutropfen pipettiert. Das DNS-Ca3(PO4)2-Präzipität sollte fein verteilt sich auf den adhärenten Zellen niederlegen.
Am darauffolgenden Morgen wurde das Medium gegen frisches, auf RT erwärmtes DMEM/H ausgetauscht. 6 h später erfolgte die Kokultivierung mit den aktivierten PBLs .
3.2) Transduktion der aktivierten PBLs
3.2.1) Aktivierung peripherer Blut-Lymphozyten (PBLs)
3 Tage vor der angesetzten Kokultivierung wurden fikollisierte PBLs zu 2*106 Zellen/ml in huRPMI-P jeweils in 2ml einer 24well-Platte (Zellgewebe-behandelte Oberflächen) ausgesät. Die Aktivierung erfolgte über den kreuz-vernetzenden Antiköφer OKT-3 (Orthoclone- Diagnostics) zu 20 ng/ml.
huRPMI-P:
RPMI 1640 (2mM Glutamin) ohne Phosphat (Life Tee, 11877-032 ) 10 % humanes, hitzeinaktiviertes AB-Serum (HLA-A2.1 seropositiv) 25 mMHEPES 1 x Penicillin/Streptomycin (Life Tee.)
Die Platten wurden in einem Brutschrank bei 37°C und 5% CO2 inkubiert.
3.2.2) Kokultivierung
Zur Kokultivierung wurden die aktivierten PBLs aus den jeweiligen Vertiefungen einer 24well-Platte vereinigt und gezählt. Adhärente Monozyten wurden verworfen. Die Zellen werden abzentrifugiert (1500 φm, 5 min, RT) und in einer Konzentration von 2,5* 106 Zellen/ml in frischem huRPMI-P aufgenommen und in den Brutschrank zurückgestellt. Das Medium wurde zuvor zu 400 U/ml IL-2 (Chiron) und 5 μg/ml Polybren (Sigma) eingestellt.
Jeder Transfektionsansatz wurde 6h nach Medium Wechsel nacheinander trypsiniert: hierzu wurde jede T25 mit 3ml HBSS (Life Technologies) gewaschen, mit 1 ml Trypsin-EDTA (Life Technologies) für maximal 5 Minuten inkubiert, die gelösten Zellen quantitativ aufgenommen und in 4ml vorgelegtes huRPMI-P (RT) unter Rühren getropft. Die 293 T Zellen werden mit 2500 rad bestrahlt. Diese werden abzentrifugiert (1500 φm, 5 min, RT) und in 4 ml frisches, eingestelltes huRPMI-P, mit 400U/ml IL-2 und 5 μg/ml Polybren supplementiert, resuspendiert. Zu dem Ansatz wurde 1 ml der eingestellten PBLs gegeben und der Ansatz (0,5* 106 PBLs/ml) für drei Tage im Brutschrank (37°C, 5% CO2) inkubiert.
Am Tag 3 nach Kokultivierung wurden die suspendierten PBLs abgenommen und in frischem, mit IL-2 (400 U/ml) supplementiertem Medium huRPMI-P zu 0,5* 106 Zellen/ml resuspendiert. 3 Tage später erfolgte ein neuerlicher Split in frisches Medium. In diesen 7 Tagen wurde maximal expandiert bis zum Übergang auf T75-Flaschen. Diese Zellen konnten direkt in einer immunologischen Anfärbung (FACS-Analsyse; Kap. 3.3, Figur 9) oder in einem klassischen 51 Chrom-Freisetzungstest (Kap. 3.4, Figuren 10-15) eingesetzt werden.
3.3) Beispiele der FACS-Analyse
Die unter 2.) beschriebenen Konstrukte wurden nach retroviraler Transduktion im "fluo- rescence activated cell sorting" (FACS) analysiert. Hierzu wurden 0,25* 106 Zellen sättigend mit Fluorophor-markierten Antiköφern gefärbt: die heterolog exprimierte ß-Kette wurde mit anti-vß6-FITC (BD) nachgewiesen; die Gesamtheit der T-Zellen über den Marker anti-CD3- PC5 (Coulter-Beckman) (Figur 9). Als Negativ-Kontrolle diente eine mit dem leeren pStitch-Derivat transduzierte Probe. Der Wildtyp muvα-mucα/muvß-mucß (2.1) sowie die chimären Konstrukte muvα-hucα/muvß- hucß (2.2) erzielten Transduktionseffizienzen im Bereich von 10-25%) 6 Tage nach Kokultivierung. Die Expression konnte in mehreren Donoren HLA-A2 - positiver T-Zellen reproduziert werden. Das cytosolisch exprimierte Grün-fluoreszierende Protein (eGFP, umkloniert aus eGFP-Nl der Firma Clontech) diente als externer Standard und zeigte auf, daß die vergleichbar schwache Transduktionseffizienz als Ausdruck des prozentualen Nachweises erfolgreich transduzierter T-Zellen ein methodisches Problem und kein intrinsisches Problem der TZR-Transgene ist.
3.3.1) Anreicherung vß6-positiver T-Zellen über magnetische Sortierung
Um die erfolgreich transduzierten T-Zellen anzureichern, wurden diese über magnetisch markierte vß6-Antiköφer im magnetischen Feld sortiert (laut Vorschrift Miltenyi-Biotec). Hierzu wurde ein magnetisch markierter Antiköφer verwendet, der den Ratten IgG-Anteil der vß6- spezifischen Antiköφer erkennt (Miltenyi-Biotec "Goat anti-rat IgG MicroBeads", Produkt 485-01). Es konnten Anreicherungen vß6-positiver CD3+ - T-Zellen von über 90% erreicht werden.
3.4) Cytolytische Aktivität der transduzierten T-Zellen
Die transduzierten T-Zellen wurden in einem klassischen 5 I Chrom-Freisetzungstest auf ihre zytotoxische Spezifität hin übeφriift. In diesem System wurden Zielzellen radioaktiv durch Inkoφoration von 51 Chrom markiert. Erkannten die retroviral modifizierten Effektorzellen die Zielzelle peptidspezifisch, wurden letztere durch die Effektorfunktionen der T-Zelle in die Apoptose getrieben und durch Lyse getötet. Das Ausmaß des freigesetzten Chrom-Nuklids trifft eine Aussage über die Effektivität der Zeil-Erkennung und Lyse. Die Effektivität wurde über einen weiten Bereich des Verhältnisses eingesetzter Effektor-Zellen zu Zielzellen (E:T) getestet. Als Referenz diente der murine hdm2-81-88 peptidspezifische T-Zell-Klon 3, aus dem die T-Zell-Rezeptor-Gene isoliert wurden. Als Zielzellen dienten:
T2: humane TAP-defiziente Zelllinie die exogen mit beliebigem Peptid zu beladen war. Das spezifische Peptid war hdm2-81-88, ein irrelevantes Kontroll-Peptid entstammte dem Influenza-Matrixprotein FluMl . Saos-2/6: Transfektante der humanen Saos-2 -Zelllinie, die hdm2 exprimiert und endogen prozessiert.
Saos2/143: Transfektante der humanen Saos-2-Zelllinie, die die p53-Mutante Val143Ala trägt. Die p53-Mutante reguliert die Expression von hdm2 in positiver Weise JY: EBV-transformierte LCL-B-Zelllinie UocBl 1, EU-3: pre-B-Zell-Leukämie IM-9: Plasmocytom
3.4.1) Peptid-Titration (Figuren 10-12)
Die Zelllinie T2 wurde exogen mit dem hdm2-81-88-Peptid in einer Konzentrationsreihe von 1* 10" M bis 1*10" M beladen. Es konnte sowohl für muvα-mucα/muvß-mucß (2.1) als auch muvα-hucα/muvß-hucß (2.2) eine Peptid-spezifische und Dosis-abhängige cytotoxische T- Zell- Antwort beobachtet werden. Für die beiden Konstrukte lagen bei E:T =20:1 die halbmaximalen Lysen im subnanomolaren Bereich eingesetzten Peptids. Als Negativ-Kontrolle fungierte ein nicht-funktioneller Einzel-Ketten T-Zell-Rezeptor, der zwar exprimiert und laut
3.3.1) anreicherbar war, aber keine Lysis-Aktivität aufwies. Als Positiv-Kontrolle diente der murine hdm2-81-88 peptidspezifische T-Zell-Klon 3.
3.4.2) Erkennung von maligne transformierten Zeil-Linien (Figuren 13-15)
Die oben beschribenen Zelllinien konnten Peptid-spezifisch lysiert werden. Einige der pre-B- Zell-Leukämien konnten im Fall der WT-Ketten bei hohen E:T's zu 100 % lysiert werden. Ebenso wie im Fall der Peptid-Titration zeigten die Wildtyp-Ketten (2.1) eine höhere Lysis- Effizienz im Vergleich zu den partiell humanisierten, chimären Ketten (2.2). Als Negativ- Kontrolle fungierte ein nicht-funktioneller Einzel-Ketten T-Zell-Rezeptor, der zwar exprimiert und laut 3.3.1) anreicherbar war, aber keine Lysis-Aktivität aufwies. Als Positiv- Kontrolle diente der murine hdm2-81-88 peptidspezifische T-Zell-Klon 3.

Claims

Patentansprüche
1. Polypeptid eines eine hdm2-Protein-spezifische T-Zell Antwort vermittelnden murinen α/ß T-Zell Rezeptors gemäß SEQ ID Nr. 1 oder SEQ ID Nr. 2 oder funktioneller Varianten oder Teile davon oder dieses kodierende Nukleinsäuren, funktionelle Varianten oder Teile davon.
2. Fusionsprotein, umfassend ein Polypeptid gemäß Anspruch 1 oder funktionelle Varianten oder Teile davon oder dieses kodierende Nukleinsäuren, funktionelle Varianten oder Teile davon.
3. Fusionsprotein nach Anspruch 2, dadurch gekennzeichnet, daß es die ζ-Region von CD3 und/oder CD8, CD 16 oder Teile davon umfaßt, insbesondere die ζ-Region von humanem CD3 und/oder CD8, CD 16 oder Teile davon.
4. Fusionsprotein nach Anspruch 2 oder 3, weiterhin einen flexiblen Linker umfassend, insbesondere einen Linker der Aminosäuresequenz (GGGGS)3.
5. Fusionsprotein nach Anspruch 2, dadurch gekennzeichnet, daß es sich um ein chimäres, zumindest partiell humanisiertes Fusionsprotein handelt, insbesondere gemäß SEQ ID Nr. 3 oder SEQ ID Nr. 4.
6. Fusionsprotein nach einem der Ansprüche 2 bis 5, dadurch gekennzeichnet, daß es sich um einen Einzelketten T-Zell Rezeptor handelt, insbesondere gemäß SEQ ID Nr. 5 oder SEQ ID Nr. 6.
7. Fusionsprotein nach einem der Ansprüche 2 bis 4, dadurch gekennzeichnet, daß es sich um einen Doppelketten T-Zell Rezeptor handelt, insbesondere gemäß SEQ ID Nr. 7 oder SEQ ID Nr. 8.
8. α- oder ß-Kette eines T-Zell-Rezeptors, der die Antigen-Erkennungssequenz eines für die Aminosäuren 81-88 des Proteins hdm2 oder eines Komplexes von hdm2 81-88 und HLA- A2 spezifischen Antiköφers umfaßt.
9. Polypeptid nach einem der Ansprüche 1 bis 8, dadurch gekennzeichnet, daß das Polypeptid synthetisch hergestellt worden ist.
10. Nukleinsäure nach Anspruch 1 oder 2, dadurch gekennzeichnet, daß sie eine DNA oder RNA, vorzugsweise eine DNA, insbesondere eine doppelsträngige DNA mit einer Länge von mindestens 8 Nukleotiden, vorzugsweise mit mindestens 18 Nukleotiden, insbesondere mit mindestens 24 Nukleotiden ist.
11. Nukleinsäure nach einem der Ansprüche 1, 2 oder 10, dadurch gekennzeichnet, daß die Sequenz der Nukleinsäure mindestens ein Intron und/oder eine polyA-Sequenz aufweist.
12. Nukleinsäure nach einem der Ansprüche 1, 2, 10 oder 11 in Form ihrer komplementären „antisense"-Sequenz.
13. Nukleinsäure nach einem der Ansprüche 1, 2 oder 10 bis 12, dadurch gekennzeichnet, daß die Nukleinsäure synthetisch hergestellt worden ist.
14. Vektor, vorzugsweise in Form eines Plasmids, Shuttle Vektors, Phagemids, Cosmids, Expressionsvektors, retroviralen Vektors adenoviralen Vektors oder Partikels und/oder gentherapeutisch wirksamen Vektors, umfassend eine Nukleinsäure nach einem der Ansprüche 1, 2 oder 10 bis 13.
15. Wirtszelle, transfiziert mit einem Vektor oder infiziert oder transduziert mit einem Partikel gemäß Anspruch 14.
16. Wirtszelle nach Anspruch 15, dadurch gekennzeichnet, daß es sich um eine T-Zelle oder eine T-Zell- Vorläuferzelle oder eine Stammzelle handelt.
17. Wirtszelle nach Anspruch 15 oder 16, dadurch gekennzeichnet, daß auf ihrer Oberfläche ein Polypeptid oder Fusionsprotein gemäß einem der Ansprüche 1 bis 9 exprimiert wird.
18. Verfahren zur Identifizierung von hdm2-Protein-spezifischen Antigenen, dadurch gekennzeichnet, daß hdm2 -präsentierende Tumorzellen oder Fraktionen davon mit einer Wirtszelle gemäß Anspruch 17 unter Bedingungen zusammengebracht werden, bei denen die Tumorzellen oder Fraktionen davon nur dann lysiert werden, wenn der Tumor das hdm2- Protein-spezifische Antigen präsentiert, für welches das exprimierte Polypetid oder Fusionsprotein spezifisch ist.
19. Verfahren zur Herstellung eines gegen ein Polypeptid, Fusionsprotein oder eine Nukleinsäure gemäß einem der Ansprüche 1 bis 13 gerichteten Antiköφers, vorzugsweise eines polyklonalen oder monoklonalen Antiköφers zur Diagnose, Behandlung und/oder Überwachung der Behandlung von mit hdm2-Protein assoziierten Erkrankungen und/oder zur Identifizierung von pharmakologisch aktiven Substanzen, dadurch gekennzeichnet, daß ein Antiköφer produzierender Organismus mit einem Polypeptid oder funktionelle Äquivalente davon oder Teile davon mit mindestens 6 Aminosäuren, vorzugsweise mit mindestens 8 Aminosäuren, insbesondere mit mindestens 12 Aminosäuren nach einem der Ansprüche 1 bis 9 immunisiert bzw. mit diesen kodierenden Nukleinsäuren vakziniert wird.
20. Antiköφer, hergestellt gemäß Anspruch 19, dadurch gekennzeichnet, daß er gegen ein Polypeptid nach einem der Ansprüche 1 bis 9 gerichtet ist.
21. Rekombinanter Antiköφer, dadurch gekennzeichnet, daß er die Antigen- Erkennungssequenz der α- oder ß-Kette eines für die Aminosäuren 81-88 des Proteins hdm2 oder eines Komplexes von hdm 2 81-88 und HLA-A2 spezifischen T-Zell- Rezeptors umfaßt.
22. Verfahren zur Herstellung eines Arzneimittels zur Behandlung von mit hdm2-Protein assoziierten Erkrankungen, dadurch gekennzeichnet, daß mindestens eine Nukleinsäure, mindestens ein Polypeptid, mindestens eine Wirtszelle oder mindestens ein Antiköφer nach einem der vorgenannten Ansprüche zusammen mit geeigneten Zusatz- und Hilfsstoffen kombiniert wird.
23. Arzneimittel zur Behandlung von mit hdm2-Protein assoziierten Erkrankungen, dadurch gekennzeichnet, daß es mindestens eine Nukleinsäure, mindestens ein Polypeptid, mindestens eine Wirtszelle oder mindestens einen Antiköφer nach einem der vorgenannten Ansprüche, gegebenenfalls zusammen mit geeigneten Zusatz- und Hilfsstoffen, enthält.
24. Verwendung eines Arzneimittels nach Anspruch 23 zur Behandlung von mit hdm2- Protein assoziierten Erkrankungen.
25. Verfahren zur Herstellung eines Tests zur Auffindung funktioneller Interaktoren in Zusammenhang mit hdm2 -Protein assoziierten Erkrankungen, dadurch gekennzeichnet, daß mindestens eine Nukleinsäure, mindestens ein Polypeptid oder mindestens ein Antiköφer nach einem der vorgenannten Ansprüche zusammen mit geeigneten Zusatz- und Hilfsstoffen kombiniert wird.
26. Test zur Identifizierung funktioneller Interaktoren in Zusammenhang mit hdm2 -Protein assoziierten Erkrankungen, dadurch gekennzeichnet, daß er mindestens eine Nukleinsäure, mindestens ein Polypeptid oder mindestens einen Antiköφer nach einem der vorgenannten Ansprüche, gegebenenfalls zusammen mit geeigneten Zusatz- und Hilfsstoffen, enthält.
