Abstract

Die vorliegende Erfindung betrifft Verfahren zur Vermehrung von Ribonukleinsäuren, die Schritte umfassen, bei denen man (a) mittels eines Einzelstrang-Primers definierter Sequenz, einer RNA-abhängigen DNA Polymerase und Desoxyribonukleosidtriphosphaten einen DNA-Einzelstrang durch reverse Transkription aus einer RNA erzeugt; (b) die RNA entfernt; (c) mittels eines Einzelstrang-Primers, der eine Box-Sequenz umfasst, einer DNA-Polymerase und Desoxyribonukleosidtriphosphaten einen DNA-Doppelstrang erzeugt; (d) den Doppelstrang in Einzelstränge auftrennt; (e) mittels eines Einzelstrang-Primers, der im 5'-Bereich die Sequenz eines Promotors und im 3'-Bereich dieselbe definierte Sequenz wie der Primer in (a) umfasst, einer DNA-Polymerase und Desoxyribonukleosidtriphosphaten DNA-Doppelstränge aus einem der in (d) entstandenen Einzelsträngen erzeugt; (f) mittels einer RNA-Polymerase und Ribonukleosidtriphosphaten eine Vielzahl von RNA Einzelsträngen erzeugt, die an beiden Enden definierte Sequenzen umfassen. Die Erfindung betrifft ferner Kits zur Vermehrung von Ribonukleinsäuren gemäss einem der obigen Verfahren, welche die folgenden Bestandteile umfassen: mindestens einen Einzelstrang-Primer, der die Sequenz eines Promotors umfasst; (b) mindestens einen Einzelstrang-Primer, der eine Box-Sequenz umfasst; (c) eine RNA-abhängige DNA-Polymerase; (d) Desoxyribonukleosidtriphosphate; (e) eine DNA-abhängige DNA-Polymerase; eine RNA-Polymerase; und (g) Ribonukleosidtriphosphate.
