まず、この出願の発明の核酸回収チップの基本構成の一例を図 1 の実 施例を用いて説明する。 核酸回収チップ 1 0 0で、 スライドガラス等の 光学的に透明な基板 1 0 1上に、 クロムの蒸着層などの光学吸収を持つ 薄膜層 1 0 2が配置されている。 透過光で観察をする場合には、 薄膜層 1 0 2の膜厚は、 光を完全に吸収しない程度でかつむらの無い程度の薄 いものであることが望ましい。 例えば、 クロムの場合には、 膜厚 5 O A で蒸着すれば可視領域の透過光 7 0 %程度であり、 この発明の用途に用 いるのに支障ない。 その他、 T i , V , Fe, Co, Ni , Mo , W のいずれか の金属を用いてもよい。 これらの金属は、 金属層の表面にできた酸化物 層がシラノール基と共有結合することで、 基板表面に核酸プロ一ブ層 1 1 3を固定することができる。 吸光薄膜層 1 0 2の上には、 細胞 1 0 4 を培養するための容器の壁 1 0 3が積層されている。 容器の大きさや配 置は、細胞の種類に応じて適宜選択するが、たとえば、神経細胞の場合、 幅 5 0 程度以上であることが望ましい。 また、 各容器の間に細胞は 移動できないが、 軸索等が他の細胞に接合できるように経路 1 1 0を空 けておいてもよい。 また、 容器の壁 1 0 3の高さは細胞が乗り越えない 程度の高さを持つ必要がある。 たとえば神経細胞の場合には、 高さ 2 0 m以上あればよい。 特に容器の壁 1 0 3の素材がァガロースを用いた 場合には、 細胞との接着性も無く、 また、 細胞に対してのシグナル物質 でもないことから細胞にとっては無害なだけでなく、 培養実験データへ の影響が小さく最適である。 細胞 1 0 4を培養するとき、 培養液の循環 が必要な場合には、 容器の壁 1 0 3を含む細胞培養領域をすベて覆う構 造の光学的に透明な容器 1 0 6を被せて、 管 1 0 7から溶液を導入し、 管 1 0 8から廃液を回収すればよい。 このとき、 電極となる領域 1 0 2 と対になるように、 導電性を持つ領域 1 0 9を容器 1 0 6内上面に付加 する。 この領域 1 0 9の厚さは、 領域 1 0 2と同様に光透過性を有する 程度に薄く蒸着する。 ここで容器 1 0 5内の細胞の様子は対物レンズ 1 1 2を用いて連続観察することができる。 また、 対物レンズを通じて近 赤外光の集束光を基板表面に照射することで、 局所的に加熱して、 前記 核酸プローブ層 1 1 3と結合した核酸成分を選択的に熱変性させて回 収することができる。 電源モジュール 1 1 1を用いて領域 1 0 2と領域 1 0 9の間に電界を印加することができる。 ま'たこの実施例では、 領域 1 0 2が 1枚の電極として構成されているが、 各細胞容器ごとに独立し て動作する複数の電極アレイを用いてもよい。
図 2は、 この出願の発明の核酸回収チップを用いた細胞中の核酸回収 手順を例示した模式図である。 図 2 ( 1 ) は、 細胞を導入する前の基板 表面の状態を示した模式図である。 基板表面 2 0 1には核酸成分を捕獲 するために核酸プローブ 2 0 2が固定されている。 ここで、 核酸プロ一 ブ 2 0 2としては、 m R N Aを選択的に回収するために p o 1 y— Tを 中心とした配列で構成してもよいし、 特定の核酸成分を回収したい場合 は、 特定の核酸成分に相補的な配列で構成してもよいし、 あるいは、 複 数の異なる配列の核酸プローブを混合して配列させてもよい。図 2 ( 2 ) は、 前記基板表面 2 0 1上に細胞 2 0 3が配置されている様子を示した 模式図である。 細胞 2 0 3の状態を顕微鏡観察しながら特定の細胞状態 となったときに、 顕微鏡対物レンズを通じて照射した赤外線集束光によ つて細胞を破砕してもよいし、 各細胞培養容器の電極が独立している場 合は、 電極に直流あるいは交流の電場を印加して細胞を破砕してもよレ 。 あるいは一斉にすベての容器の細胞を破碎する場合は、 界面活性剤等の 薬剤を導入することで細胞を破砕してもよい。 図 2 ( 3 ) は、 細胞破砕 後の細胞内タンパク質と核酸成分との分離手順を示した模式図である。 基板表面 2 0 1と対極との間に直流電界を印加することで、 タンパク質 成分と核酸成分とを分離する。 タンパク質と核酸成分とで等電点が大き く異なっているため、 核酸成分のみを基板表面 2 0 1に引き寄せ、 基板 表面からタンパク質成分 2 0 4を遊離させて溶液を流して排除するこ とができる。 また、 細胞中の核酸成分を、 基板表面 2 0 1上に 2次元に 射影したかたちで固定することができる。 図 2 ( 4 ) は、 その後、 先ほ どとは反対の直流電界を印加して、 基板表面の核酸プローブ 2 0 5と結 合しなかった核酸成分 2 0 6を除去することができることを示してい る。 図 2 ( 5 ) は、 次に集束光加熱によって核酸プローブと結合した細 胞内核酸成分を局所的に解離させ回収する手順を示した模式図である。 対物レンズ 2 0 7によって照射した集束光によって加熱した領域 2 0 8の核酸プローブに結合した核酸試料 2 0 9のみが選択的に溶液中に 解離することから、 この領域の解離した核酸成分を含む溶液を回収する ことで細胞内の特定領域の核酸成分の選択回収が可能となる。
図 3は、 この出願の発明の核酸回収チップを用いる光学系および溶液 送液系の 1実施例としての装置構成を模式的に例示したものである。 こ の装置では細胞等の試料を核酸回収チップ 1 0 0で培養しながらその 状態変化を観察するために、 顕微鏡観察系、 培養液循環系を、 同時に核 酸回収チップ 1 0 0上の核酸を選択回収させるために集束光照射系を 持っている。 図 3からもわかるように顕微観察光学系の光路上に核酸回 収チップ 1 0 0を配置しており、 この核酸回収チップ 1 0 0に溶液を供 給する培養液供給'廃棄部が連結されている。まず、顕微観察光学系は、 以下のような構成になっている。 光源 3 0 1から照射された光は、 フィ ルタ一 3 0 2で特定の波長に調整され、 コンデンサレンズ 3 0 3によつ て集光されて、 核酸回収チップ 1 0 0に照射される。 照射された光は、 透過光として対物レンズ 3 0 5での観察に用いられる。 核酸回収チップ 1 0 0内部の透過光像は、 ミラ一 3 1 1によってフィルタ 3 1 2通過後、 カメラ 3 1 3に誘導され、 カメラの受光面に結像する。 従って、 核酸回 収チップ 1 0 0の素材は、 フィルタ 3 0 2で選択された波長の光に対し て、 光学的に透明な素材であることが望ましい。 具体的には、 ホウケィ 酸ガラス、 石英ガラス等のガラスや、 ポリスチレン等の樹脂やプラスチ ック、 あるいはシリコン基板等の固体基板および、 ァガロース等の高分 子を用いる。 また特にシリコン基板を用いる場合は波長 9 0 0 n m以上 の波長の光を観測に用いる。 また、 図 1 の吸光層 1 0 2のところで述べ たように、 光の吸収が前記特定の波長において 8 0 %未満となるような 膜厚あるいは吸収を持たない波長を選択的に用いることが望ましい。 また、 光源 3 0 8から照射された光も、 フィルダー 3 0 9で波長選択 された後に、 ダイクロイツク · ミラ一 3 1 0によって対物レンズ 3 0 5 に誘導され、 核酸回収チップ 1 0 0内部の蛍光観察の励起光として用い られる。 核酸回収チップ 1 0 0から発した蛍光は再度対物レンズ 3 0 5 によって集光され、 フィルタ一 3 1 2によって励起光をカツ トした後の 蛍光と透過光のみをカメラ 3 1 3で観察することができる。 このとき、 フィルター 3 0 2、 3 0 9、 3 1 2の組み合わせを調整することで、 透 過光のみをカメラ 3 1 3で観察したり、 あるいは蛍光のみを観察したり, 透過光像と蛍光像を同時に観察することも出来る。 光路内には、 レーザ 一光源 3 0 7で発生させたレーザー光を可動ダイクロイツク ·ミラー 3 0 6によって対物レンズ 3 0 5に導入する機構も備わっている。 このレ —ザ一は対物レンズ 3 0 5によって集束光となり、 核酸回収チップ 1 0 0を局所的に加熱することができる。 集光点を移動させる場合には、 可 動ダイクロイック · ミラ一を移動させることで、 核酸回収チップ 1 0 0 内でのレーザ一の集束位置を動かすことが可能である。 このレーザ一の 波長としては、 水の吸収を持たず、 光化学作用を持たない波長が望まし い。 たとえば N d : Y A Gレーザ一の 1 0 6 4 n mなどでは、 水、 ガラ ス、 ァガロース等に対して顕著な吸収はなく、 選択的にクロム薄膜層の みでレーザ一光吸収が起こり、 光が吸収されたクロム薄膜層の光集束点 近傍のみで局所的に発熱する。
カメラで得られた画像データは画像処理解析装置 3 1 4によって解 析され、 さまざまな解析結果を基に可動ダイクロイツク,ミラ一 3 0 6 や、 核酸回収チップ 1 0 0が載っている温調機能付可動 X Yステージ 3 0 4の位置を制御するために X-Y 方向に自在に移動させるステージ移 動用モータ一 3 1 5を駆動することができる。 これによつて細胞の形状 を認識したり、 認識後にその細胞を追跡し、 つねに画像の中心に位置さ せたり、 対物レンズとの距離を調節することで画像のピントを特定の細 胞に合わせたりすることが可能となる。 あるいは、 一定時間の周期で可 動ダイクロイツク · ミラ一 3 0 6や、 培養マイグロチャンパ 1 0 0が載 つている温調機能付ステ一—ジ 3 0 4を制御したり、 一定間隔でステージ 移動用モータ一 3 1 5を駆動することができる。
培養液供給,廃棄部について説明すると、 核酸回収チップ 1 0 0に複 数の種類の種類、 濃度の異なる培養液あるいは細胞破碎用の試薬等を細 胞の状態に応じて溶液槽 3 1 6から供給装置 3 1 7によって供給され た培養液は、 供給装置内の温度調節機構によって液温を調節され、 さら に溶存空気交換機構.によって溶存気体の成分が調整され、 流速を調節さ れながら核酸回収チップ 1 0 0に培養液等の試薬を供給する構造にな つている。 容器 1 0 0の培養液はまた P C R反応機能つき分配器 3 1 8 によって P C R反応増幅した後にキヤビラリ一電気泳動装置 3 1 9に 導入して核酸回収チップ 1 0 0から供給された試料核酸の配列を調べ たり、 あるいは廃液を廃液溜 3 2 0におくることができる。
もちろん、 この出願の発明は以上の例に限定されることなしに、 その 細部について様々な形態が可能であることは言うまでもない。 産業上の利用可能性
以上詳しく説明したとおり、 この出願の発明によって、 培養している 細胞の特定の状態での核酸成分の空間分布を解析することが可能とな る。

Claims

請求の範囲
1 . 透明基板上に電気伝導性領域が配置され、 この領域の上に核酸結 合部と細胞収容容器部が配設された細胞中の核酸回収チップであって 前記の電気伝導性領域に電位を印加する手段と、 細胞収容容器部中で細 胞が培養される手段とが具備されているとともに、 前記の電気伝導性領 域は、 特定の波長の光に対して吸収を有し、 この特定波長の光が照射さ れることで局所的に発熱し、 電気伝導性領域上に結合した核酸成分が局 所的に解離 · 回収されるようにしたことを特徴とする核酸回収チップ。
2 . 電気伝導性領域に電位を印加する手段として、 電気伝導性領域と これに対向配置された上部電気伝導性部とを有していることを特徴と する請求項 1の核酸回収チップ。
3 . 細胞収容容器部中で細胞が培養される手段として、 細胞収容容器 部を内包して、 細胞培養液の流通を可能とした箱体容器部が具備されて いることを特徴とする請求項 1または 2の核酸回収チップ。
4 . 一細胞を収容する細胞収容容器部が 1以上配設されていることを 特徴とする請求項 1ないし 3のいずれかの核酸回収チップ。
5 . 請求項 1ないし 4のいずれかの核酸回収チップを備えた核酸回収 装置であって、 核酸回収チップの電気伝導性領域に特定波長の光を照射 して局所的に加熱させる光学系とともに、 この領域に電位を印加する電 源系を有していることを特徴とする核酸回収装置。
6 . 細胞の状態を観察する観察系を有していることを特徴とする請求 項 5の核酸回収装置。
7 . 細胞の培養液の流通系を有していることを特徴とする請求項 5ま たは 6の核酸回収装置。
