Abstract

 イズモリング(第1図)の希少糖戦略の中で、多種類の希少アルドースに作用し、多種類の希少ケトースを生産するために最も効率のよいイソメラーゼを得ることによって、多種類の希少糖を生産する反応系を確立すること。 以下の(a)又は(b)のタンパク質をコードするDNA。Pseudomonas stutzerii由来のL-ラムノースイソメラーゼである上記のDNA。配列番号2に示されるアミノ酸配列からなるタンパク質。上記のタンパク質を発現することができる発現系を含んでいる宿主細胞を培地に培養し、得られる培養物からL-ラムノースイソメラーゼ活性を有する組換えタンパク質を採取することを特徴とする組換えタンパク質の製造方法。第1図を希少糖生産に利用する方法であって、目的とする希少糖の、単糖の全体像中の位置を把握し、上記タンパク質を作用させるその最適な生産経路を設計することを特徴とする方法。

Description

新しい触媒機能を有する L -ラムノースィソメラーゼの遺伝子配列おょぴ その用途 技術分野 明
本発明は、 Pseudomonas stutzeri の生産する L -ラムノースイソメラ 一ゼをコ一ドする遺伝子配列、 さらにこれまで見いだされていなかった 新しい糖異性化反応を触媒する触媒機能を書明らかにしたものであり、 該 新規遺伝子配列、 該新規触媒機能、 ならびに、 該新規触媒機能の希少糖 生産および生理活性探索への利用に関する。
本発明において、 利用するィズモリング (Izumoring) 連携図は、 ィ ズモリング C 6 (第 5図、 商願 20 0 3— 1 6 3 0) の中でのつながり と、 ィズモリング C 5 (第 6図、 商願 2 00 3— 1 6 3 1 ) の中でのつ ながり と、 ィズモリング C 4の中でのつながり と、 C 4、 C 5、 C 6が 全てつながっている第 1図で示されるィズモリング全体図であり、 出願 前未公開のものである。
また、 いラムノ^ "スイソメラーゼは各種の微生物から単離され、 それ をコードする遺伝子の配列も報告されている。 本発明は、 土壌より分離 したバクテリア (Pseudomonas stutzeri) の L-ラムノースイソメラーゼ をコードする遺伝子配列を決定したところ、 これまで報告されている遺 伝子配列との相同性のないもので、 遺伝子的にも、 蛋白質的に.新規なも のであることが明らかとなったものである。
この配列を利用することで、 遺伝子操作を利用して酵素を大量生産し それを用いる希少糖や、 その他各種遺伝子工学的手法を用いた用途に利 用できるものである。 前から開始されていた。 その両者がドッキングした形で、 香川大学農学 部で生産された希少糖 (単糖) を用いて生理活性を探求する研究が、 1 9 9 9年から地域先導研究として開始され、 さまざまな生理活性を有す ることが発見されてきている。 単糖類は還元基 (カルボニル基) の状態によりアルドース (カルボ二 ル基としてアルデヒ ド基を持つ糖) 、 ケトース (カルボュル基としてケ トン基を持つ糖) 、 糖アルコール (別名 : ポリオール、 カルボ二ル基を 持たない糖) に大別される。 単糖類には 「希少糖」 といわれるものがあ る。 希少糖とは、 国際希少糖学会の定義によれば自然界に希にしか存在 しない糖と定義されており、 その種類によっては、 有機化学的合成方法 における収量も少ないものも多い。 このため、 希少糖について未知の性 質のものも多く、 ァロースを含めたアルドへキソース (アルドース) 希 少糖においても未知の性質が多いというのが現状である。 糖の応用研究に関し、 従来、 糖と癌との関係については、 例えば、 特 許文献 1に記載されているように、 癌の予防に有効である多糖類が知ら れている。 また、 オリゴ糖が整腸作用を持つことを利用して便秘を解消 し大腸癌などになりにくい効果をもつことや、 最近ではァガリスクなど の多糖体が癌抑制効果を持つことなどの報告、 糖鎖と癌転移関連の報告 もある。 さらに、 特許文献 2には、 D—ァロースの誘導体を有効成分と する抗腫瘍剤は開示されている。 一方、 糖類の活性酸素に対する性質を 利用したものでは、 例えば、 特許文献 3に記載されているように、 活性 酸素を抑制する性質を有する多糖類を含有させた活性酸素産生抑制剤は 知られている。 2
さらにまた、 Pseudomonas stuizeriの生産する L -ラムノースィソメラ ーゼがこれまで見いだされていなかった新しい糖異性化反応を触媒する 触媒機能をもつことを明らかにしたものである。
L -ラムノースイソメラーゼは、 希少糖 D-ァロースを D-プシコースから 生産する時に有用な酵素である。 本発明は遺伝子工学的な手法を用いて 純粋な酵素を生産し検討した結果、 これまで認めることのできなかった 新しい異性化反応を触媒する能力を見いだしたもので、 各種の希少糖の 生産に利用できるものである。 背景技術
従来の未利用資源、 特にバイオマス (例えば木材などの廃物) の有効 利用は、 それをブドウ糖へ加水分解しそれをアルコールへと変換するこ とが大きな目標であった。 しかしアルコールへ変換しても付加価値が低 いため実用化は無理であった。 また、 多糖 (未利用資源に無尽蔵に存在 する) を原料として、 中途半端に分解すると、 オリゴ糖ができる。 これ も機能性のある付加価値のあるものとして用途が開発されている。
希少糖の生理活性に着目し、 細胞を用いる実験によりその裏付けをす ることは本発明者らによってはじめられた。 2 1世紀は生命科学の世紀 とも言われており、 現在、 国際的に D N A研究、 タンパク質研究が進め られている。 ポス トゲノム研究における糖と言えば糖鎖研究が中心であ るが、 本発明者らの属する香川医科大学 (現 香川大学医学部) 、 香川 大学農学部では、 単糖に着目し、 単糖に生理活性はないか等その応用研 究を進めている。 その背景としては、 香川大学の農学部の方で希少糖の 生産に関する網羅的な研究が長年積み重ねられてきて、 近年になり一部 の希少糖の大量生産技術が確立されたことが挙げられる。 香川医科大学 (現 香川大学医学部) においても糖に生理活性を探求する研究が数年 単糖類の中で、 プシコースは、 還元基としてケトン基を持つ六炭糖で ある。 このプシコースには光学異性体として D体と L体とが有ることが知 られている。 ここで、 D -プシコースは既知物質であるが自然界に希に しか存在しないので、 国際希少糖学会の定義によれば 「希少糖」 と定義 されているが、 この D—プシコースは、 近年、 ェピメラーゼの出現 (例 えば、 特許文献 4参照) により高価ではあるが、 比較的入手が容易とな つた。 そして、 この公報に従えば、 調製された D—プシコースは、 甘味 料、 醱酵用炭素源、 試薬、 化粧品 · 医薬品の原料, 中間体などとして有 効に利用できることが示唆されている。 この公報によれば、 この甘味料 としては、 飲食物、 飼料、 歯磨き、 内服薬など経口摂取物の甘味付け嗜 好性向上に利用できる旨用途の方向性が記載されているにすぎない。 D ープシコースの光学異性体である L -プシコースについては、 可食配合 物として利用可能であることが、 例えば、 特許文献 5で詳細に開示はさ れている。
一方、 プシコースの試薬 .医薬品等の中間原料としての応用例は、 次 に示される。 例えば、 非特許文献 1によれば、 D—プシコースを原料と したヒダントイン誘導体の合成例が報告されている。 また、 非特許文献 2によれば、 D-フラク トフラノシルヌクレオシドの合成例が開示されて いる。 いずれの先行技術にも D -プシコースが医薬品等の原料 ' 中間体と して利用できることが報告されているにすぎない。 また、 特許文献 6には、 構造中に六炭糖を有するコウジ酸配糖体はメ ラニン生成抑制作用が優れているとともに、 安定性が高く、 かつ水に対 する溶解性が高く、 美白外用剤の有効成分として適していると記載され ているにすぎない。 また、 特許文献 7には、 プシコースは皮膚バリアー 機能の回復を促進して、 皮膚の表皮機能の低下による表皮増殖異常等を 防止するために有用であることが記載され、 保湿剤として有用性が記载 されているにすぎない。 また、 D-タガトースを含むいくつかの糖類を有 効成分とする過血糖付随疾患の予防および肥満防止用保健食が特許文献 8で公開されているが、 希少糖そのものの性能は記述されていない。 ま た、 特許文献 9には、 ケトへキソースの一つである D—ソルボースを含 む、 ァラビノース、 リポース、 グルコースを主要構成糖とする複合多糖 類について、 抗高脂血症用剤としての用途が公表されているにすぎない。
しかし、 「単糖」 に着目し、 希少糖の応用研究を進めるためにも、 ま た、 新規用途が完成された場合は一層、 希少糖の大量生産技術の確立が 必要となる。 一方、 Pseudomonas stutzerii LL172の生産する L ラムノースィソメラ ーゼは、 非特許文献 3で発表された以下の物理化学的性質を有する公知 酵素である。
(ィ) 作用
L -ラムノースから L-ラムニュロースへの異性化反応ならびに L-ラム二 ュロースから L -ラムノースへの異性化を触媒する酵素である。 D-アロー スと D-プシコースの間の異性化にも作用することが既知であり (非特許 文献 3 ) 、 D-プシコースから D -ァロースを生産することができる酵素で ある。 異性化酵素はもつとも高い活性を示す基質を元に命名されるため、 L-ラムノースィソメラーゼと同一で命名された酵素は、 大腸菌おょぴ枯 草菌から単離され、 それをコードする遺伝子の配列が報告されている。
(口) 基質特異性
L -ラムノースおょぴ L-ラムニュロースを基質とする。 のみならず、 L ーリキソースおょぴ L一キシルロース、 L一マンノースおよび Lーフラ ク トース、 D—リポースおょぴ D—リブロース、 D—ァロースおよび D ープシコースを基質とする。
(ハ) 作用 p Hおよび至適 p H
作用 p Hは 7. 0〜1 0. 0であり、 至適 p Hは 9. 0である。
(二) pH安定性

各種希少糖の生産法は、 希少糖生産戦略ィズモリング (第 1図で示さ れる生産過程と分子構造 (D型、 L型) により、 炭素数の異なる単糖全て をつないだ連携図) によって生産できる設計図が、 本発明者らにより完 成している。 その戦略の中のアルドースとケトース間を触媒するィソメ ラーゼは、 希少アルドースおよび希少ケトースの生産に重要である。 一 般にアルドースイソメラーゼは基質特異性が比較的広い。 すなわち、 例 えば D—キシロースイソメラーゼは D—キシロースと D—キシルロース 間の異性化を触媒するが、 この反応のみならず、 D—グルコースと D— ブラク トース間の異性化をも触媒する。 基質特異性が広いといっても、 基質となるアルドースは 3〜4種が通常である。 D—ァラビノースイソ メラーゼは、 D—ァラビノース、 L一ガラク トースおよび Lーフコース 等に作用する等のように、 構造が比較的類似したものに作用するのであ る。
そのため、 第 1図のィズモリングの種々の希少アルドース、 希少ケト ースの生産には、 その構造を考慮した検討を行うことで目的とする希少 糖を生産することが重要な検討課題となっている。 本発明は、 第 1図のィズモリング (ィズモリング全体図) を希少糖生 産に利用することを目的とする。
本発明は、 多糖 (未利用資源に無尽蔵に存在する) を原料として、 ブ ドウ糖等単糖へ変換するところまでは従来法と同じであるが、 それから 先が酵母によるアルコール発酵ではなく、 希少糖という付加価値の高い ものへの最適な生産経路を設計し、 希少糖大量生産技術を確立すること を目的とする。
本発明は、 Pseudomonas stutzeri の生産する L-ラムノースイソメラー ゼをコードする遺伝子配列を提供すること、 各種遺伝子工学的手法によ りこれまで見いだされていなかった新しい糖異性化反応を触媒する触媒 機能を発見すること、 ならびに、 新規触媒機能を希少糖生産および生理 活性探索へ利用することを目的とする。
本発明は、 新規かつ有用な L-ラムノースイソメラーゼ遺伝子の配列を 提供し、 遺伝子操作、 さらに明らかにした新規触媒機能を利用した希少 糖の生産や各種遺伝子工学的手法、 あるいは該新規触媒機能を用いた用 途に利用できるようにすることを目的とする。
本発明は、 Pseudomonas stutzeri の生産する L-ラムノースイソメラー ゼをコードする遺伝子配列を、 さらに明らかにした新規触媒機能を、 ィ ズモリング全体図を用いて、 希少糖生産に利用すること、 さらに、 希少 糖の生理活性探索に寄与することを目的とする。
さらに、 本発明は、 ィズモリング (第 1図) の希少糖戦略の中で、 多 種類の希少アルドースに作用し、 多種類の希少ケトースを生産するため に最も効率のよいィソメラーゼを得ることによって、 多種類の希少糖を 生産する反応系を確立することを目的とする。 本発明は Pseudomonas stutzerii の生産する L -ラムノースイソメラー ゼをコ一ドする遺伝子配列を明らかにしたものである。 L -ラムノースィ ソメラーゼは各種の微生物から単離され、 それをコードする遺伝子の配 列も報告されている。 しかし、 これらの起源由来の L -ラムノースイソメ ラーゼが D -プシコースに反応して D-ァロースを作るという報告はない。 本発明は、 土壌より分離したバクテリア (Pseudomonas stutzerii LL1 72) の L-ラムノースイソメラーゼをコ一ドする遺伝子配列を決定したと ころ、 これまで報告されている遺伝子配列との相同性のないもので、 遺 伝子的にも、 蛋白質的に新規なものであることが明らかとなったもので ある。 この配列を利用することで、 遺伝子操作を利用して酵素を大量生 産しそれを用いる希少糖や、 その他各種遺伝子工学的手法を用いた用途 に利用できるものである。
L-ラムノースから L -ラム-ュロースへの異性化反応ならびに L -ラム二 ュロースから L-ラムノースへの異性化反応を触媒する Pseudomonas stut zeri i 由来の L-ラムノースイソメラーゼをコードする DNAと、 該 DNAを用 いる組換え DNA技術によるポリべプチドの製造方法を提供するこどによ り解決する。 さらに、 本発明者らは研究をすすめ、 Pseudomonas stutzeriの生産す る L-ラムノースイソメラーゼがこれまで見いだされていなかった新しい 糖異性化反応を触媒する触媒機能をもつことを明らかにした。 本発明に おいては、 ィズモリング (第 1図) の中の、 異性化反応を触媒する酵素 を ラムノースィソメラーゼのもつ新たに発見された触媒能力を利用し て各種希少糖を生産するものである。
これまでは個別の異性化反応を、 個別の異なるイソメラーゼを用いて 反応していたものを、 L -ラムノースイソメラーゼの非常に広い基質特異 性を利用することで、 一つの酵素を用いて、 多種類の希少糖を生産しよ うとするものである。
すなわち、 本発明は、 L-ラムノースイソメラーゼが多くの異性化反応 を触媒することにより、 これまで不可能であった希少糖の生産を一つの 酵素を有効に利用して効率的に各種の希少糖の生産が可能になった。 すなわち、 本発明は、 以下の DNAを要旨とする。
(1) 以下の(a)または(b)のタンパク質をコードする DNA。
( a )配列番号 2に示されるァミノ酸配列からなるタンパク質
(b)配列番号 2に示されるアミノ酸配列において、 1若しくは数個 のアミノ酸が欠失、 置換若しくは付加されたアミノ酸配列からなり、 か つ L-ラムノースイソメラーゼ活性を有するタンパク質
(2) 配列番号 1に示される塩基配列若しくはその相補的配列または これらの配列の一部若しくは全部を含む配列からなる DNA。
(3) 上記(2)の DNAとストリンジェントな条件下でハイプリダイズし、 かつ、 いラムノースイソメラーゼ活性を有するタンパク質をコードする
DNA。
{ 4 ) Pseudomonas stutzerii 由来の L—ラムノースィソメラーゼである 上記(1)、 (2)または(3)の1) 。
(5) 上記の L-ラムノースイソメラーゼは、 以下の物理化学的性質を 有する酵素である上記(4)の D 。
(ィ) 作用
第 7図, 第 8図, 第 9図に太い黒線で示される異性化反応を触媒する。
(口) 作用 p Hおよび至適 p H
作用: Hは 7. 0〜: L 0. 0であり、 至適 p Hは 9. 0である。
(ハ) p H安定性 - 種々の p Hで 4°C、 1時間保持した場合、 ; H 6. 0〜 1 1. 0の範 囲で安定である。
(二) 作用温度および至適温度
作用温度は 40〜6 5°Cであり、 至適温度は 6 0°Cである。
(ホ) 温度安定性
40°C、 1 0分では安定しており、 5 0°C、 1 0分でも 9 0 %以上残 存している。
(へ) キレート剤の影響
キレート剤である EDTA、 EGTAを活性測定時に共存させても、 ほとんど活性は阻害されない。
(ト) 金属イオンの影響
1 mMのコバルトイオンにより約 3 0 %阻害される。
(チ) SD S— PAGE法による分子量
約 4 3, 00 0である。 また、 本発明は、 以下のタンパク質を要旨とする。
( 6 ) 配列番号 2に示されるァミノ酸配列からなるタンパク質。
(7) 配列番号 2に示されるアミノ酸配列において、 1若しくは数個 のアミノ酸が欠失、 置換若しくは付加されたアミノ酸配列からなり、 か つ L-ラムノースィソメラーゼ活性を有するタンパク質。
(8) L -ラムノースイソメラーゼ活性は、 以下の物理化学的性質によ つて特定されるものである上記(6)または(7)のタンパク質。
(ィ) 作用
第 7図, 第 8図, 第 9図に太い黒線で示される異性化反応を触媒する。
(口) 作用 p Hおよび至適 p H
作用 p Hは 7. 0〜 1 0. 0であり、 至適 p Hは 9. 0である。
(ハ) p H安定性 種々の p Hで 4°C、 1時間保持した場合、 : pH 6. 0〜1 1. 0の範 囲で安定である。
(二) 作用温度および至適温度
作用温度は 4 0〜6 5°Cであり、 至適温度は 60°Cである。
(ホ) 温度安定性
40°C、 1 0分では安定しており、 5 0°C、 1 0分でも 9 0 %以上残 存している。 '

(10) 上記(1)ないし(5)のいずれか記載の DNAを含む組換えべクター。 また、 本発明は、 下記の(11)の宿主細胞を要旨とする。
(11) 上記(6)、 (7)または(8)のタンパク質を発現することができ る発現系を含んでなる宿主細胞。 また、 本発明は、 下記の(12)の組換えタンパク質の製造方法を要旨と する。
(12) 上記(11)の宿主細胞を培地に培養し、 得られる培養物から L -ラ ムノースイソメラーゼ活性を有する組換えタンパク質を採取することを 特徴とする組換えタンパク質の製造方法。 さらにまた、 本発明は、 下記の(13)のィズモリング (I zumoring) 連 携図を希少糖生産に利用する方法を要旨とする。
( 13) 第 1図で示される生産過程と分子構造 (D型、 L型) により、 炭 素数の異なる単糖全てをつないだ連携図を希少糖生産に利用する方法で あって、 目的とする希少糖の、 単糖の全体像中の位置を把握し、 上記 ( 6 )、 (7 )、 (8 )または(9 )のタンパク質を作用させるその最適な生産 経路を設計す'ることを特徴とする方法。
(14) 希少糖生産が希少糖大量生産である上記(13)の方法。
( 15) 希少糖生産が未利用資源からの希少糖生産である上記(13)また は(14)の方法。
( 16) 目的とする希少糖が、 生理活性が判明した希少糖である上記(1 3)、 (14)または(15)の方法。 発明の効果
L -ラムノースイソメラーゼを遺伝子工学的手法によって、 大量に生産 することが可能となり、 本酵素を用いた D -ァロースを含む各種の希少糖 の大量生産法を確立できる。
本発明は、 最も安価に大量に入手できる原料は D—グルコース (ブド ゥ糖) である。 この D—グルコースはほとんどすべての未利用植物に大 量に存在する糖であり、 これを有 に利用して目的とする希少糖への最 適な生産経路を設計するツールを提供することができる。
また、 本発明は、 希少糖の生産分野ばかりではなく、 希少糖の持つ生 理活性を探索する研究においても有効なツールを提供することができる。 図面の簡単な説明

Pseudomonas stutzeri i に属す 齒株 「Pseudomonas stutzerii LL17 2」 は、 上記文献に記載された公知菌であり、 香川大学農学部生物資源 食糧化学科 何森健研究室に保存されている。 このたび国際出願をする に際し、 本菌株を日本国独立行政法人産業技術総合研究所 特許生物寄 託センター (日本国茨城県つくば巿東 1— 1一 1 中央第 6 ) に 2 0 0 4年 1月 6日に国際寄託している (IPOD FERM BP- 08593) 。 なお、 本菌 株は LL172aとも表示することがあるが、 LL172と LL172aは同一菌株であ る。
L -ラムノースイソメラーゼは、 L-ラムノースかち L-ラムュュロースへ の異性化反応ならびに L -ラムニュロースから L -ラムノースへの異性化を 触媒する酵素である。 Pseudomonas stutzeri i LL172の生産する L-ラムノ ースイソメラーゼは、 D -ァロースと D-プシコースの間の異性化にも作用 するので、 D-プシコースから D -ァロースを生産することができる酵素で ある。 ただし、 D-プシコースから D-ァロースを生産するためには、 Pseu domonas stutzeri i LL172由来の酵素が必要である。
Pseudomonas stutzeri i LL172由来の L-ラムノースイソメラーゼをコ一 ドする遺伝子配列は、 これまで報告されている L-ラムノースイソメラー ゼの遺伝子配列との相同性のないもので、 遺伝子的にも、 蛋白質的に新 規なものであることが明らかとなったものである。 本発明でいう L-ラムノースイソメラーゼは、 Pseudomonas stutzeri i L L172由来の L -ラムノースィソメラーゼであって、 配列番号 1に記載され るアミノ酸配列、 またはそのァミノ酸配列の中の 1個以上のァミノ酸が 他のァミノ酸で置換され、 欠失され、 ' 1個以上のァミノ酸が付加されて なるアミノ酸配列を有する。 本発明でいう遺伝子 (DNA) は、 上記の L- ラムノースィソメラーゼをコードする塩基配列を有する。 すなわち、 本発明の対象となるタンパク質と しては、 配列番号 2に示 されるアミノ酸配列からなるタンパク質 (L-ラムノースィソメラーゼ) や、 該配列番号 2に示されるアミノ酸配列において、 1若しくは数個の アミノ酸が欠失、 置換若しくは付加されたアミノ酸配列からなり、 かつ L -ラムノースイソメラーゼ活性を有するタンパク質を挙げることができ る。
また、 上記 L-ラムノースイソメラーゼ活性と しては、 好ましくは L -ラ ムノースから L-ラム-ュロースへの異性化反応ならびに L-ラム-ュロー スから L-ラムノースへの異性化を触媒する酵素活性を挙げることができ る。 また、 D-ァロースと D-プシコースの間の異性化を触媒する酵素活性 を挙げることができる。 D -アロースを D -プシコースから生産できる活性 は、 Pseudomonas stutzerii LL172由来の L-ラムノースィソメ fーゼ以外 には報告されていない。
さらに、 Pseudomonas stutzerii LL172の生産する L-ラムノースィソメ ラーゼは、 以下の物理化学的性質を有する酵素であること明らかにした。
(ィ) 作用
L -ラムノースから L-ラムュュロースへの異性化反応ならびに L -ラム二 ュロースから L-ラムノースへの異性化を触媒する。 D-ァロースと D-プシ コースの間の異性化にも作用し、 D-プシコースから D-ァロースを生産す ることができる酵素である。 以上が既知の主たる作用である。 さらに本 発明者らにより この度明らかとなった L—ラムノースイソメラーゼの新 規触媒反応を含めた全ての異性化反応は、 ィズモリングの第 7図, 第 8 図, 第 9図に示される。 基質特異性は表 1 , 2, 3参照。
L -ラムノースおよび L-ラム-ュロースを基質とする。 のみならず、 L —リキソースおよび L—キシノレロース、 L一マンノースおよび Lーフラ ク トース、 D—リポースおよび D—リブロース、 D—ァロースおよび D ープシコースを基質とする。 以上が既知の主たる基質特異性である。 第 7図, 第 8図, 第 9図より L—ラムノースイソメラーゼが単糖の多くを 基質とすることが理解できる。
すなわち、 活性の大小はあるものの、 L—ラムノースイソメラーゼが 触媒することが確認された異性化反応は第 7図中太い黒線で示したもの である。 一方、 異性化反応が確認できなかったものは、 太い点線で示し た 4種であることが一目瞭然に理解できる。
また、 第 9図、 第 1 0図に示すように、 これも活性の大小はあるもの の、 ペントースおよびテトロースにおける全異性化活性を持つことを示 している。
(ハ) 作用 p Hおよび至適 pH
作用 p Hは 7. 0〜1 0. 0であり、 至適 p Hは 9. 0である。
(二) pH安定性
種々の p Hで 4°C、 1時間保持した場合、 p H6. 0〜: L 1. 0の範 囲で安定である。
(ホ) 作用温度および至適温度
作用温度は 40〜 6 5°Cであり、 至適温度は 60°Cである。
(へ) 温度安定性
40°C、 1 0分では安定しており、 5 0°C、 10分でも 90%以上残 存している。

約 4 3, 0 0 0である。 本発明の対象となる DNAとしては、 配列番号 2に示されるアミノ酸配 列からなるタンパク質や、 配列番号 2に示されるアミノ酸配列において、 1若しくは数個のアミノ酸が欠失、 置換若しくは付加されたアミノ酸配 列からなり、 かつ L-ラムノースィソメラーゼ活性を有するタンパク質を コードする DNAや、 配列番号 1に示される塩基配列またはその相補的配 列並びにこれらの配列の一部または全部を含む DNAや、 かかる DNAとスト リンジェントな条件下でハイブリダイズし、 かつ ラムノースィソメラ ーゼ活性を有するタンパク質をコードする DNAを好ましいものとして例 示することができる。 これら DNAは、 その DNA配列情報等に基づき、 遺伝子ライプラリーなど から公知の方法により調製することができる。 また、 配列番号 1に示さ れる塩基配列またはその相補的配列並びにこれらの配列の一部または全 部をプローブとして、 各種細胞由来の DNAライブラリーに対してストリ ンジェントな条件下でハイプリダイゼーションを行ない、 該プローブに ハイプリダイズする DNAを単離することにより、 L-ラムノースィソメラ ーゼ活性を有するタンパク質をコードする DNAを得ることもできる。 か かる DNAを取得するためのハイブリダィゼーシヨンの条件としては、 例 えば、 42°Cでのハイブリダィゼーシヨン、 及び 1 X SSC、 0. 1 ° /。の SDSを含 む緩衝液による 42°Cでの洗浄処理を挙げることができ、 65°Cでのハイブ リダィゼーシヨ ン、 及び 0. 1 X SSC, 0. 1 %の SDSを含む緩衝液による 65°C での洗浄処理をより好ましく挙げることができる。 なお、 ハイブリダィ ゼーションのストリンジエンシーに影響を与える要素としては、 上記温 度条件以外に種々の要素があり、 種々の要素を適宜組み合わせて、 上記 例示したハイブリダイゼーショ ンのス ト リンジエンシーと同等のス ト リ ンジエンシーを実現することが可能である。 本発明の融合タンパク質としては、 上記本発明のタンパク質と翻訳コ ドンタンパク質とが結合しているものであればどのようなものでもよく、 翻訳コ ドンタンパク質としては、 従来知られている翻訳コドンタンパク 質であれば特に制限されるものではない。 かかる融合タンパク質は、 常 法により作製することができ、 当該分野の研究用試薬としても有用であ る。 本発明はまた、 上記本発明のタンパク質を発現することができる発現 系を含んでなる宿主細胞に関する。 かかる本発明のタンパク質をコード する遺伝子の宿主細胞への導入は、 Davisら (BASIC METHODS IN MOLECUL AR BIOLOGY, 1986) 及び Sambrookら (MOLECULAR CLONING : A LABORATORY MANUAL, 2nd Ed., Cold SpringHarbor Laboratory Press, Cold Spring Harbor, N. Y. , 1989) などの多くの標準的な実験室マニュアルに記載さ れる方法により行うことができる。 そして、 宿主細胞としては、 大腸菌、 ス ト レプトミセス、 枯草菌、 ス ト レプトコッカス、 スタフイロコッカス 等の細菌原核細胞等を挙げることができる。 また、 発現系としては、 上記本発明のタンパク質を宿主細胞内で発現 させることができる発現系であればどのようなものでもよく、 染色体、 ェピソーム及びウィルスに由来する発現系、 例えば、 細菌プラスミ ド由 来、 酵母プラスミ ド由来、 SV40のようなパポバウィルス、 ワクシニアゥ ィルス、 アデノ ウイルス、 鶏痘ウィルス、 仮性狂犬病ウィルス、 レ トロ ウィルス由来のベクター、 パクテリオファージ由来、 トランスポゾン由 来及びこれらの組合せに由来するベクター、 例えば、 コスミ ドゃファー ジミ ドのようなプラスミ ドとバタテリオファージの遺伝的要素に由来す るものを挙げることができる。 この発現系は発現を起こさせるだけでな く発現を調節する制御配列を含んでいてもよい。 上記発現系を含んでなる宿主細胞を培養して得られる本発明のタンパ ク質は、 D-ァロースの生産に用いることができる。 また、 かかる本発明 のタンパク質を細胞培養物から回収し精製するには、 硫酸アンモ-ゥム またはエタノール沈殿、 酸抽出、 ァユオンまたはカチオン交換クロマト グラフィー、 ホスホセルロースクロマトグラフィー、 疎水性相互作用ク 口マトグラフィー、 ァフィ二ティ一クロマトグラフィー、 ハイ ドロキシ アパタイ トクロマトグラフィーおよびレクチンクロマトグラフィーを含 めた公知の方法、 好ましくは、 高速液体クロマトグラフィーが用いられ る。 希少糖とは、 自然界に希にしか存在しない単糖 (アルドース、 ケトー スおよぴ糖アルコール) と定義づけることができるが、 この定義は糖の 構造や性質による定義ではないため、 あいまいである。 すなわち、 一定 量以下の存在量を希少糖というなどの量の定義はなされていないためで ある。 しかし、 一般に自然界に多量に存在するアルドースとしては D -グ ノレコース、 D-ガラク トース、 D-マンノース、 D-リボース、 D -キシロース T/JP2004/000131

L-ァラビノースの 6種類あり、 それ以外のアルドースは希少糖と定義さ れる。 ケトースとしては、 D-フラク トースが存在しており、 他のケトー スは希少糖といえる。 また糖アルコールは単糖を還元してできるが、 自 然界には D—ソルビトールが比較的多いがそれ以外のものは量的には少 ないので、 これらも希少糖といえる ώ
希少糖は、 これまで入手自体が困難であつたが、 自然界に多量に存在 する単糖から希少糖を生産する方法が開発されつつあり、 その技術を利 用して製造することができる。 以下、 ィズモリング (Izumoring) 連携図について説明する。
第 1図で示される生産過程と分子構造 (D型、 L型) により、 炭素数 4 から 6の単糖全てをつないだ連携図がィズモリング (I zumoring) の全 体図である。 すなわち、 第 1図から理解できることは、 単糖は、 炭素数 4、 5、 6全てがつながつているということである。 全体図は、 ィズモ リング C 6の中でのつながり と、 ィズモリング C 5の中でのつながり と、 ィズモリング C 4の中でのつながり と、 C 4、 C 5、 C 6が全てつなが つていることである。 この考え方は重要である。 炭素数を減少させるに は主に発酵法を用いる。 炭素数の異なる単糖全てをつなぐという大きな 連携図であることも特徴である。 また、 利用価値がないということも理 解することができる。 炭素数が 6つの単糖 (へキソース) のィズモリングは、 第 1図の下段 および第 5図に示すように、 炭素数が 6つの単糖 (へキソース) は全部 で 3 4種類あり、 アルドースが 1 6種類、 ケトースが 8種類、 糖アルコ ールが 1 0種類ある。 これらの糖は、 酸化還元酵素の反応、 アルドース 異性化酵素の反応、 アルドース還元酵素の反応で変換できることは、 本 発明者らの研究を含めた研究で知られている。 しかしながら、 これまで の研究では上のグループ、 真ん中のグループ、 下のグループは酵素反応 でつながっていなかった。 つまり、 上のグループに属している D —グル コース (ブドウ糖) や D—フラク トースは自然界に多量に存在する糖で あり安価であるが、 これらから希少糖を合成することができなかった。 ところが、 本発明者らの研究の過程で、 これを結ぶ酵素が発見された。 それはガラクチトールから D-タガトースを合成する酵素を持つ菌の培養 液中に、 全く予期しなかった D —ソルポースが発見されたことに端を発 する。 その原因を調べた結果、 この菌が D —タガトース 3ェピメラーゼ ( D T E ) という酵素を産生していることを発見した。 第 1図の下段お ょぴ第 6図に示すように、 この D T Eはこれまで切れていた D —タガト ースと D —ソルポースの間をつなぐ酵素であることがわかる。
そしてさらに驚くことに、 この D T Eは全てのケトースの 3位をェピ 化する酵素であり、 これまで合成接続できなかった D—フラク トースと D—プシコース、 L—ソルポースと Lータガトース、 D —タガ トースと D—ソルボース、 Lープシコースと L—フラク トース、 に作用するとい う非常に幅広い基質特異性を有するユニークな酵素であることが分かつ た。 この D T Eの発見によって、 すべての単糖がリング状につながり、 単糖の知識の構造化が完成し、 ィズモリング (I zumoring) と名付けた。
この第 5図をよく見てみると、 左側に L型、 右側に D型、 真ん中に D L型があり、 しかもリ ングの中央 (星印) を中心として L型と D型が点 対称になっていることもわかる。 例えば、 D—グルコースと L 一ダルコ ースは、 中央の点を基準として点対称になっている。 しかもィズモリン グ (Izumoring). の価値は、 全ての単糖の生産の設計図にもなつている ことである。 先の例で、 D —グルコースを出発点として L —グルコース を生産しようと思えば、 D —グルコースを異性化→ェピ化→還元→酸化 →ェピ化→異性化すると L—グルコースが作れることを示している。
炭素数が 6つの単糖 (へキソース) のィズモリ ング (I zumoring) を 使って、 自然界に多量に存在する糖と微量にしか存在しない希少糖との 関係が示されている。 D—グルコース、 D—フラク トース、 D—マンノ —スと、 牛乳中の乳糖から生産できる D—ガラク トースは、 自然界に多 く存在し、 それ以外のものは微量にしか存在しない希少糖と分類される。 D T Eの発見によって、 D—グルコースから D—フラク トース、 D—プ シコースを製造し、 さらに D—ァロース、 ァリ トール、 D—タリ トール を製造することができるようになった。
炭素数が 6つの単糖 (へキソース) のィズモリ ング (I zumoring) の 意義をまとめると、 生産過程と分子構造 (D型、 L型) により、 すべて の単糖が構造的に整理され (知識の構造化) 、 単糖の全体像が把握でき ること、 研究の効果的、 効率的なアプローチが選択できること、 最適な 生産経路が設計できること、 欠落部分について予見できること、 が挙げ られる。 炭素数が 5つの単糖 (ペントース) のィズモリングは、 第 1図の中段 および第 6図に示すように、 炭素数 6のィズモリングよりも小さいリン グである。 しかし、 C 6のィズモリングと同じようにアルドース 8個、 ケトース 4個および糖アルコール 4個全てを含むことに変わりは無く、 全てが酵素反応で結ばれる。 異なる点は、 酸化還元反応、 異性化反応の みでリング状に全てが連結できることである。 一方、 D T Eを用いるこ とによって、 さらに効率のよい生産経路が設計できることがわかる。
炭素数 5のィズモリ ングの特徴は、 特に第 6図から明らかなように、 炭素数 6のィズモリングが点対象に全単糖が配置されているのに対し、 左右が対象に配置されていることが大きな特徴である。 これら全ペント ースは、 酵素反応により連結されていることから、 炭素数 6のィズモリ ングの場合と全く同様に、 すべてのペントースが構造的に整理され (知 識の構造化) 、 全体像が把握できること、 研究の効果的、 効率的なアブ ローチが選択できること、 最適な生産経路が設計できること、 欠落部分 について予見できる意義を持っている。 炭素数が 4つの単糖 (テトロース) のィズモリングは、 第 1図の上段 に示すように、 テトロースの構造上の特性のため、 リングが完成しない という特徴がある。 炭素数 5のィズモリング上部半分の構造を持ってい る。 このリングの場合も、 炭素数 5, 6の場合と同様の酸化還元および 異性化反応によって連結されている。 D T Eが炭素数 4のケトースに反 応しないため、 ケトース間の反応は現在のところ存在しない。 しかし、 新規のェピメラーゼの存在が予測され、 この研究は現在研究途上である。 全体の配置は、 炭素数 5と同様に左右対称であり、 アルドース 4個、 ケトース 2個および糖アルコール 3個全てを含んでいる。 すなわち炭素 数 5, 6のィズモリングと同様の意義が存在する。 ィズモリング C 6の D _グルコースは、 ィズモリング C 5の D—ァラ ビトールぉよぴィズモリング C 4のエリスリ トールとつながつている。 この線は、 発酵法によって D—グルコースから D—ァラビトールおょぴ エリスリ トールを生産できることを示している。 すなわち、 ィズモリン グ C 6, ィズモリング C 5およびィズモリング C 4は連結されている。 この連結は、 炭素数の減少という主に発酵法による反応であり、 この D ーァラビトールおよびエリスリ トールへの転換反応の二つ以外の発酵法 によるィズモリング C 6とィズモリング C 5, C 4との連結は可能であ る。 例えば D—グルコースから D—リポースの生産も可能である。 このように、 3つのィズモリングにより全ての炭素数 4, 5, 6の単 糖 (アルドース、 ケトース、 糖アルコール) が連結されたことで、 それ ぞれの単糖が全単糖の中でその存在場所を明確に確認できる。
最も有名なキシリ トールは、 未利用資源の木質から生産できる D—キ シロースを還元することで容易に生産できることを明確に確認できる。
もしも特定の単糖が生物反応によって多量に得られた場合には、 それ を原料とした新たな単糖への変換の可能性が容易に見いだすことが可能 である。 すなわち、 この全体像から全ての単糖の原料としての位置を確 実につかむことができるため、 有用な利用法を設計することができる。 特に廃棄物や副産物から単糖が得られた場合の利用方法を容易に推定で きるのである。 本発明は、 ィズモリング (第 1図) の希少糖戦略の中で、 上記のとお り、 多種類の希少アルドースに作用し、 多種類の希少ケトースを生産す るために最も効率のよいイソメラーゼを得ることができ、 それによつて 多種類の希少糖を生産する反応を確立することが可能となった。 すなわ ち、 さらに明らかとなった新規触媒機能を、 ィズモリング全体図を用い て、 希少糖生産に利用すること、 さらに、 希少糖の生理活性探索に寄与 することが可能となった。
ィズモリングの第 7図, 第 8図, 第 9図から明らかなように、 ィズモ リングを用いて異性化反応を整理することが全体を理解する手段として 如何に有効であるかを示している。 さらに、 L一ラムノースイソメラー ゼが単糖の多くを基質とすることが理解できる。
すなわち、 活性の大小はあるものの、 L—ラムノースイソメラーゼが 触媒することが確認された異性化反応は第 7図中太い線で示したもので ある。 一方、 異性化反応が確認できなかったものは、 太い点線で示した 4種であることが一目瞭然に理解できる。
また、 第 8図、 第 9図に示すように、 これも活性の大小はあるものの、 ペントースおよびテトロースにおける全異性化活性を持つことを示して いる。
このようにィズモリングを利用することで、 L—ラムノースイソメラ ーゼの触媒する反応を明確に示すことができると同時に、 反応が確認で きないものを明確に認識することが可能である。
新しく確認された各種の異性化活性はアルドースからの反応と、 ケト ースからの反応の結果が若干異なるなど詳細な反応機構を検討すること 必要があるが、 第 7図, 第 8図, 第 9図が示すように非常に多くの希少 糖の生産に利用できる可能性を持つことが明らかとなった。 本願発明の詳細を実施例で説明する。 本願発明はこれら実施例によつ て何ら限定されるものではない。
実施例 1
L -ラムノースを大量に生産する方法の一つとして、 遺伝子工学的手法 での増産が考えられる。 そこで従来の方法で本酵素をコードする遺伝子 をクローン化し遺伝子配列およびアミノ酸配列を決定した。 その結果が 以下のとおりであった。
[配列決定]
Pseudomonas stutzeri i LL172 由来 L-ラムノースイソメラーゼ遺伝子 は、 配列表 1およぴ第 2図に示すとおり、 0RF1, 290-bpからなり 430アミ ノ酸をコードする新規の L -ラ ノースイソメラーゼ遺伝子である。 配列 表 2のアミノ酸配列からの計算分子量は 46, 946とオーセンティックの酵 4 000131

素の分子量約 43, 000よりやや大きいものであった。
本遺伝子を大腸菌で組換え発現させると本酵素を活性発現し分子量も 約 43, 000と一致した。 大腸菌に入れた実験の場合の酵素発現の結果は以下のとおりである。 従って配列表 1および第 2図に示した遺伝子配列は、 L-ラムノースィ ソメラーゼのものであると確認された。
[本遺伝子配列の特徴]
L -ラムノースイソメラーゼの遺伝子はすでに大腸菌と枯草菌で構造が 角军析されて!/ヽるカ S、 Pseudomonas stutzerii LL172 菌由来の L一ラムノ ースイソメラーゼとのアミノ酸配列の相同性は第 3図に示すとおり 20 % 以下と低く、 触媒部位も一致しないので同一の酵素ではないと断定した。 すなわちこれまで発表されている L -ラムノースィソメラーゼとは全く 新しい遺伝子配列をもつ酵素であった。
アミノ酸配列の相同性をデータベースで用いて解析すると、 第 4図に 示すとおり、 未同定の推定ィソメラーゼと 40%程度の高い相 性を示す 力^ これらの菌の遺伝子はゲノムプロジェク トによりシークェンスされ た結果であり、 酵素としては同定されていない。
以上の結果から、 本菌由来の L -ラムノースイソメラーゼ遺伝子は、 新 規の酵素をコードする遺伝子であると断定することができた。
[用途]
遺伝子配列が明確になったことで、 この遺伝子配列を利用した分子生 物学的手法による各種の実験が可能となる。
例えば、 .この遺伝子を大腸菌に形質転換し、 大量に生産することが可 能である。 その他この遺伝子にさらに何か新たな遺伝子を結合させるな どして、 新しい性質を持つ酵素を生産することが可能となる。 実施例 2
< L -ラムノースイソメラーゼをコ一ドする DNA >
1 L-ラムノースイソメラーゼの精製と部分アミノ酸配列の決定
Pseudomonas stutzeri i LL172 をトリブティックソィブロス培地で 3 0 °C 2日間培養しポリエチレンダリコール分画、 陰イオン交換クロマト グラフィ一にて精製後電気泳動で分子量と純度を確認する。 分子量は約 42, 000に単一のバンドとして得られる。 臭化シアンを用いて酵素を部分 分解し N末端および 4箇所の部分アミノ酸配列を決定した。
2 プローブの合成と染色体マッピング
上記の培地で培養後、 定法に従い CTABを用いて染色体 DNAを抽出する。 部分アミノ酸配列を元にミックスプライマーを合成し組み合わせを変え て 2回の PCRにより特異的に増幅される PCR産物を得てプローブに用いる。 プローブを用いてサザンハイブリダイゼーションを行い L -ラムノースィ ソメラーゼ遺伝子の染色体上の位置を決定した。
3 ゲノムライブラリーのスクリーニング
染色体マッピングにより制限酵素 Apalと Sac Iで消化した約 4. 6kbの断 片に遺伝子が含まれていることが分かったのでクローユングベクター pB luescriptI ISK+に連結しゲノムライブラリを構築してプローブを用いて スクリ一ユングした。 4 L -ラムノースイソメラーゼ遺伝子の解析

り ORF 1, 290 - bpからなり 430アミノ酸 (配列表 2 ) をコードする新規の L -ラムノースイソメラーゼ遺伝子である。 アミノ酸配列からの計算分子 量は 46, 946と C末端側に修飾を受ける元菌の酵素の分子量約 42, 000より やや大きい。 L-ラムノースイソメラーゼの遺伝子はすでに大腸菌と枯草 菌で構造が解析されているが、 本菌由来の ラムノースイソメラーゼと のアミノ酸配列の相同性は第 3図に示すとおり 20 %以下と低く、 触媒部 位も一致しないので同一の酵素ではないと断定した。 また、 大腸菌の ラムノースイソメラーゼと放線菌のキシロースィソメラーゼで保存され ている異性化酵素のコンセンサスアミノ酸残基 9箇所のうち 5箇所は保 存されているが Mn結合や基質結合部位は保存されていない。 アミノ酸配 列の相同性をデータベースで用いて解析すると第 4図に示すとおり未同 定の推定ィソメラーゼと 40%程度の高い相同性を示すが、 これらの菌の 遺伝子はゲノムプロジェク トによりシークェンスされた結果であり、 酵 素としては同定されていない。
以上の結果から本菌由来のいラムノースイソメラーゼ遺伝子は新規の 酵素をコードする遺伝子であると断定した。
5 組換え L-ラムノースイソメラーゼの活性発現
L -ラムノースイソメラーゼの翻訳開始コ ドンと高発現ベクター pQESO の翻訳開始コ ドンを一致させるようプライマーを設計し PCRで増幅させ た L-ラムノースィソメラーゼ遺伝子を PQE60に組み込んで大腸菌 JM109を 形質転換し組換え大腸菌を作成した。 組換え大腸菌は通常培地で 37°C— 晚培養すると L-ラムノースイソメラーゼを活性発現し N末端ァミノ酸配 列、 分子量、 および、 酵素学的諸性質は元菌由来の酵素と一致、 酵素生 産量は 10倍以上上昇し高発現が可能となった。 実施例 3
D—グルコースから希少糖である D—リキソースの生産を行った。 酵 母 Candida famata R28を用いて D—グルコースから 5 0 %の収率で D— ァラビトールを生産した。 この反応は発酵法で行った。 生産した D—ァ ラビトーノレを酢酸菌 Acetobacter acet i I F O 3 2 8 1によってほぼ 1 0 0 %の収率で D _キシルロースへ変換した。 これを L—リボースィ ソメラーゼを用いて、 D—リキソースへと異性化することができた。 生 産物はイオン交換クロマトグラフィ一等により精製結晶化し、 D—リキ ソースであることを機器分析によつて確認した。
すなわち、 D—グルコースを原料として発酵法により、 炭素数の一つ 少ない D—リキソールへ変換し、 酸化反応、 異性化反応によって希少糖 D—リキソースを生産することが可能である。 (Journal of Bioscience and Bioengineering, Vol. 88, 676, 1999 ) 実施例 4
D—グルコースを原料として発酵法で作られるエリスリ トールから希 少糖 L—エリスロースの生産を行った。 エリスリ トールを Gluconobacte r frateurii IF0 3254 を用いて L—エリスルロースに酸化した。 この反 応はほぼ 1 0 0 %の収率で得られた。 反応液から L一エリスルロースを 分離し、 これを原料として L—リポースイソメラーゼを作用させること で、 希少糖 L—エリスロースを生産できた。 1 0 gのエリスリ トールか ら 1 . 7 gの L—エリスロースを生産することができた。 ( Journal of Bioscience and Bioengineering Vol. 92, 237, 2001 )
すなわち、 D—グルコースを出発物質として、 炭素数の 2少ないエリ スリ トールを発酵法で生産し、 それを原料として用いることで、 酸化反 応と異性化反応によって希少糖 L—エリスロースを生産することが可能 である 実施例 5
実施例 2の Pseudomonas stutzerii LL172由来の L-ラムノースィソメラ ーゼをコードする遺伝子に C末端に 6個の H i sを連結するように設計 し、 それを大腸菌に形質転換した。 この H i sを連結したいラムノース イソメラーゼを大量に発現させ、 ニッケル N T Aカラムを用いた親和力 クロマトグラフィ一法によって、 純粋な酵素を大量に生産しそれぞれ固 定化して用いることで新しい機能を発揮させることが可能となった。 表 1にはアルドース、 表 2にはケトースを基質として実験を行'い、 それぞ れの反応時間後の反応液中の糖組成を H P L Cによって分析した実験結 果を整理して示した。 すなわち、 表 1は L -ラムノースイソメラーゼの反 応を基質として各種アルドースを用いて行った時の、 反応後の生産物組 成を示している。 表 2は L -ラムノースィソメラーゼの反応を基質として 各種ケトースを用いて行った時の、 反応後の生産物組成を示している。

実施例 7
《実施例 5および 6で使用した微生物の詳細な説明》
C末端にヒスタグを連結した ラムノースイソメラーゼ遺伝子を導入 した大腸菌 J M 1 0 9を用いた。
《培地組成および培養条件》
ポリぺプトン 3 . 5 %、 酵母エキス 2 . 0 %、 N a C 1 0 . 5 % ( p H 7 . 0 ) の培地に大腸菌 J M 1 0 9を接種し、 2 8 °C 1 2時間培養後 終濃度 1 mMの 1 P T G (イソプロピル一 β—D—チォガラタ トビラノ シド) を添加した。 その後 4時間培養を続け、 遠心分離により菌体を集 菌した。
《酵素の抽出、 酵素の精製および酵素の精製と固定化》
菌体を 0 . 0 5 Μリン酸ナトリ ウム緩衝液 ρ Η 7 . 0で 2回洗浄した。 洗浄菌体をアルミナ粉末とともに磨碎したのち遠心分離を行い、 不溶性 物質をアルミナ粉末とともに除去し粗酵素液をえた。 その粗酵素液を N i一 NTAカラムを用いてァフィユティークロマト法によって酵素を精 製し純粋な酵素を得た。 酵素を超純水に対して透析した後、 凍結乾燥し て粉末の純粋な酵素をえた。 その酵素 20 m gをキトパール樹脂 1 gに 吸着させることで固定化酵素を調製した。
《各種基質を用いた酵素反応条件》
上記固定化酵素 3 g、 0. 05Mグリシン N a OH緩衝液 (p H 9. 0) 3. 0 m L , 1 M Mn C 1 2 3. 0 /z Lおよび各種基質 6 0 m g (終濃度 20mgZmL) の酵素反応液組成で、 4 2°Cで反応を行った。
産業上の利用可能性
1. L -ラムノースイソメラーゼを遺伝子工学的手法によって、 大量に 生産することが可能となり、 本酵素を用いた D-ァロースを含む各種の希 少糖の大量生産法を確立できる。
L -ラムノースイソメラーゼは各種の微生物から単離され、 それをコー ドする遺伝子の配列も報告されている。 本発明は、 土壌より分離したバ クテリア (Pseudomonas stutzeri) の L-ラムノースイソメラーゼをコ一 ドする遺伝子配列を決定したところ、 これまで報告されている遺伝子配 列との相同性のないもので、 遺伝子的にも、 蛋白質的に新規なものであ ることが明らかとなったものである。
この配列を利用することで、 遺伝子操作を利用した希少糖に生産や各 種遺伝子工学的手法を用いた用途に利用できるものである。
2. 従来の未利用資源、 特に植物性バイオマス (例えば木材や各種 未利用植物資源等) の有効利用は、 それをプドウ糖へ加水分解しそれを アルコールへと変換することが大きな目標であった。 しかしアルコール へ変換しても付加価値が低いため実用化は無理であった。 本発明の特徴 は、 プドウ糖等単糖へ変換するところまでは従来法と同じであるが、 そ れから先が酵母によるアルコール発酵ではなく、 各種生物反応による希 少糖への変換である。 これによつて、 アルコールという付加価値の低い ものから、 希少糖という付加価値の高いものを生産することを可能にす るプロセスを提供することができる。
3 . 多糖 (未利用植物性資源に無尽蔵に存在する) を原料として、 部 分分解する方法等により、 オリゴ糖が生産できる。 これも機能性のある 付加価値のあるものとして用途が開発されている。 しかし、 単糖という 最小単位にまで分解するともう新しい展開はないと考えられていた。 そ れを打破したのが、 ひとつの単糖 (希少糖) から新しい単糖 (希少糖) へと次々に変換することを目標とした生産戦略ができたことが大きな意 義と考えている。 多糖を分解すると単糖になる、 それを原料として次々 に単糖 (希少糖) を生産するという発想の新しさである。 多糖は、 上流 の原料であり、 それを分解して単糖として原料とするということである。 これは、 原料がたとえ、 木 (セルロース) であろうと、 でんぷんであろ うと、 何であろうと、 どんなに異なった多糖であろうと単糖まで分解す れば同じものとなるということがその基本的戦略である。
4 . 第 1図から理解できることは、 単糖は、 炭素数 4、 5、 6全てが つながつているということである。 ィズモリング C 6の中でのつながり と、 C 4、 C 5、 C 6が全てつながっていることである。 この考え方は 重要である。 炭素数を減少させるには主に発酵法を用いる。 炭素数の異 なる単糖全てをつなぐという大きな連携図であることも特徴である。 ど のような廃棄物あるいは、 糖質副産物が得られてもこの図からその利用 法を考察することができるのである。 また、 利用価値がないということ も理解することができるのである。
5 . 単糖に関しての研究計画の中で、 第 1図のように全ての炭素数の 異なるものを包括してと らえる考え方は存在しなかった (第 1 0図参 照) 。 個別の反応は、 それぞれの目的によって行われてきた。 個別の目 的で進めてきた研究が総合的に関係づけられることで、 相互の技術をつ なぎ合わせる方向が見いだせる。 たとえば、 廃棄物あるいは副産物とし て邪魔者として扱われてきたものが、 単糖であるかぎり、 その全てにつ いて価値判断が可能となる。 そして何の原料になるかを直ぐに判断でき る。 このように、 単糖全体を図 'システムとしてとらえる新しい技術思 想を提示することができる。
6 . 本発明のこの技術思想は、 単糖を見直す、 単糖の価値を評価する 方法につながってゆく。 単糖という一般には 「自然界では最も単純な有 機物」 という概念を、 単糖全体を考慮に入れることで、 複雑でしかも可 能性が大きく広がる有機物であることを直感できるシステムにつながる。
7 . 「単糖はこれだけしかない」 、 「単糖はこれが全てである」 とい うことを認識できることの重要性がある。 逆の見方からすると、 これだ け全部を研究することで単糖全体を知ることが可能であるという研究計 画を明確にできることを示している。 限界を知ることは、 可能性を知る ことになるのである。
8 . 本発明は、 希少糖の生産分野ばかりではなく、 希少糖の持つ生理 活性を探索する研究においても有効性を発揮する。 例えば、 ある希少糖 に生理活性が判明したとき、 第 1図で示される連携図の存在位置を確認 する。 そして構造の近い希少糖に関しての生理活性との比較、 あるいは、 構造的に鏡像関係にある希少糖の生理活性を検討することで、 生理活性 の機構を分子の構造から類推する助けになるであろう。 また、 これまで ランダムに試行錯誤に研究していた生理活性の研究を、 ィズモリングの P T/JP2004/000131

全体像を把握することを基盤として、 計画的に進めることに優位に利用 できることが期待される。
9 . 本発明は希少糖の生産戦略としての有用性および、 その用途、 特 に生理活性の研究においても有用性を発揮する。 これは、 従来の構造の みからの単糖の羅列的分類と個別的認識法から、 酵素反応による個々の 単糖の連結という生産面での体系化が可能となったこと。 さらに、 希少 糖の生理機能を解析し、 ィズモリング上に性質を集積することにより、 これまで単純な羅列的理解から、 単糖全体を、 「単糖の構造」 、 「単糖 の生産法」 、 および 「単糖の生理機能」 を包括的に理解することに大ぃ に利用できると期待される。
1 0 . 本発明において明らかとなった L一ラムノースィソメラーゼの 新規触媒反応を含めて全ての異性化反応を、 ィズモリングの第 7図, 第 8図, 第 9図に示した。 図から明らかなように、.ィズモリングを用いて 異性化反応を整理することが全体を理解する手段として如何に有効であ るかを示している。 さらに、 L一ラムノースイソメラーゼが単糖の多く を基質とすることが理解できる。
すなわち、 活性の大小はあるものの、 L一ラムノースイソメラーゼが 触媒することが確認された異性化反応は第 7図中太い線で示したもので ある。 一方、 異性化反応が確認できなかったものは、 太い点線で示した 4種であることがー目瞭然に理解できる。
また、 第 8図、 第 9図に示すように、 これも活性の大小はあるものの、 ペントースおよぴテトロースにおける全異性化活性を持つことを示して いる。
1 1 . このようにィズモリングを利用することで、 L—ラムノースィ ソメラーゼの触媒する反応を明確に示すことができると同時に、 反応が 確認できないものを明確に認識することが可能である。 1 2 . 新しく確認された各種の異性化活性はアルドースからの反応と、 ケトースからの反応の結果が若干異なるなど詳細な反応機構を検討する こと必要があるが、 第 7図, 第 8図, 第 9図が示すように非常に多くの 希少糖の生産に利用できる可能性を持つことが明らかとなった。

Claims

40 請 求 の 範 囲
1. 以下の(a)または(b)のタンパク質をコードする D 。
(a )配列番号 2に示されるアミノ酸配列からなるタンパク質。
5 (b)配列番号 2に示されるアミノ酸配列において、 1若しくは数個の アミノ酸が欠失、 置換若しくは付加されたアミノ酸配列からなり、 かつ、 L -ラムノースィソメラーゼ活性を有するタンパク質。
2. 配列番号 1に示される塩基配列若しくはその相補的配列またはこ れらの配列の一部若しくは全部を含む配列からなる DNA。
10 3. 請求項 2記載の DNAとス トリンジェントな条件下でハイブリダィ ズし、 かつ L -ラムノースィソメラーゼ活性を有するタンパク質をコード する DNA。
4. Pseudomonas stutzerii 由来の L-ラムノースイソメラー" gである 請求項 1、 2または 3の DNA。
15 5. 上記の L -ラムノースイソメラーゼは、 以下の物理化学的性質を有 する酵素である請求項 4の DNA。
(ィ) 作用
第 7図, 第 8図, 第 9図に太い黒線で示される異性化反応を触媒する。
(口) 作用 p Hおよび至適 p H
20 作用 p Hは 7. 0〜 1 0. 0であり、 至適 p Hは 9. 0である。
(ハ) p H安定性
" 種々の p Hで 4°C、 1時間保持した場合、 p H6. 0〜: L 1. 0の範 囲で安定である。
(二) 作用温度および至適温度
25 作用温度は 40〜 6 5°Cであり、 至適温度は 6 0°Cである。
(ホ) 温度安定性 40°C、 1 0分では安定しており、 5 0°C、 10分でも 90%以上残 存している。
(へ) キレート剤の影響
キレート剤である ED TA、 E GT Aを活性測定時に共存させても、 ほとんど活性は阻害されない。
(ト) 金属イオンの影響
1 mMのコバルトイオンにより約 30 %阻害される。
(チ) SD S— PAGE法による分子量
約 43, 000である。
6. 配列番号 2に示されるアミノ酸配列からなるタンパク質。
7. 配列番号 2に示されるアミノ酸配列において、 1若しくは数個の アミノ酸が欠失、 置換若しくは付加されたアミノ酸配列からなり、 かつ L -ラムノースィソメラーゼ活性を有するタンパク質。
8. L -ラムノースイソメラーゼ活性は、 以下の物理化学的性質によつ て特定されるものである請求項 6.または 7のタンパク質。
(ィ) 作用
第 7図, 第 8図, 第 9図に太い黒線で示される異性化反応を触媒する。
(口) 作用 p Hおよび至適 p H
作用 p Hは 7. 0〜: L 0. 0であり、 至適 pHは 9. 0である。
(ハ) pH安定性
種々の p Hで 4°C、 1時間保持した場合、 pH6. 0〜 1 1. 0の範 囲で安定である。
(ュ) 作用温度および至適温度
作用温度は 40〜6 5°Cであり、 至適温度は 60°Cである。
(ホ) 温度安定性
40°C、 1 0分では安定しており、 5 0°C、 1 0分でも 90%以上残 存している。
(へ) キレート剤の影響
キレート剤である E D T A、 E G T Aを活性測定時に共存させても、 ほとんど活性は阻害されない。 ,
(ト) 金属イオンの影響
1 mMのコバルトイオンにより約 3 0 %阻害される。
(チ) S D S— P A G E法による分子量
約 4 3 , 0 0 0である。
9 . 請求項 6、 7または 8記載のタンパク質と、 翻訳開始コ ドンタン パク質とを結合させた融合タンパク質。
10. 請求項 1ないし 5のいずれか記載の DNAを含む組換えベクター。
11. 請求項 6、 7または 8記載のタンパク質を発現することができる 発現系を含んでいる宿主細胞。
12. 請求項 11の発現系を含んでなる宿主細胞を培地に培養し、 得られ る培養物から ラムノースィソメラーゼ活性を有する組換えタンパク質 を採取することを特徴とする組換えタンパク質の製造方法。
13. 第 1図で示される生産過程と分子構造 (D型、 L型) により、 炭素 数の異なる単糖全てをつないだ連携図を希少糖生産に利用する方法であ つて、 目的とする希少糖の、 単糖の全体像中の位置を把握し、 請求項 6、 7、 8または 9のタンパク質を作用させるその最適な生産経路を設計す ることを特徴とする方法。
14. 希少糖生産が希少糖大量生産である請求項 13の方法。
15. 希少糖生産が未利用資源からの希少糖生産である請求項 13または 14の方法。
16. 目的とする希少糖が、 生理活性が判明した希少糖である請求項 I3、 14または 15の方法。
