Abstract

본 발명은 신규 구조의 원핵세포 내에서 유전자 발현을 감소시키는 맞춤형 합성 sRNA, 그 제법 및 그 용도에 관한 것으로, 보다 상세하게는 MicC, SgrS 및 MicF 중 어느 하나의 sRNA 유래의 Hfq 결합 부위(Hfq binding site) 및 표적 유전자 mRNA와 상보적 결합을 형성하는 영역을 포함하는 합성 sRNA, 그 제법 및 그 용도에 관한 것이다. 본 발명에 따른 합성 sRNA는 표적 유전자의 mRNA와의 결합력을 조절함으로써 억제 정도를 조절할 수 있는 장점이 있으며, 유전자 발현을 조절하는 합성 sRNA를 통해 기존의 유전자 결실을 통한 과정 없이 효과적으로 제작, 목적 유전자 발현을 감소시킬 수 있어 재조합 미생물 제조에 유용하며, 다양한 균주에서 빠르게 적용할 수 있어 균주별 대사 능력 측정 및 최적 균주 선정에 매우 적합하다. 아울러, 합성 sRNA를 통한 미생물 대사 흐름 조작에 의해 수득된 본 발명에 따른 tyrosine 혹은 cadaverine을 고효율로 생산하는 재조합 미생물은 의약품 및 산업용제 미생물로 유용하다. 즉, 본 발명에 따른 sRNA를 이용하여 대사산물의 고효율 생산을 위한 발현 억제 타겟 유전자 선별을 용이하게 할 수 있다. 이에 다양한 대사 산물의 효율적 생산을 위한 재조합 균주의 제작 및 효율적 생산방법 확립에 사용할 수 있는바 매우 유용하다.
