본 발명은 친수성 설파살라진 및 히알루론산을 포함하는 안과 염증 질환 치료용 조성물 및 이의 제조 방법에 관한 것이다. 또한, 본 발명은 친수성 설파살라진 및 히알루론산의 치료학적 유효량을 개체에 투여하는 단계를 포함하는, 안과 염증 질환의 치료 방법에 관한 것이다.
【배경기술】
눈은 생활에 필요한 대부분의 정보를 받아들이는 중요한 감각기관이다. 눈은 외막, 중막, 내막 및 굴절매질로 구성되는데, 외막은 까만 눈동자를 덮고 있는 앞표면인 각막 또는 결막과 그 뒤에 이어진 공막으로 되어 있고, 중막은 홍채와 모양체, 그리고 맥락막으로 구성되며, 내막은 망막으로 이루어진다, 수정체와 유리체 및 안방수는 굴절매질에 해당된다. 눈의 기능적 장애 또는 상실은 생활의 질을 떨어뜨리는 큰 요인의 하나가 되며, 노화, 질환 및 시력에 좋지 않은 영향을 줄 수 있는 기타 요인 때문에 눈의 건강을 유지하는 것이 중요해지고 있다.
안구 염증 질환은 충혈, 부기, 감염, 자극 또는 외상과 관련된 질환을 의미한다. 안구의 구조 중, 전안부 또는 외안부는 안구의 앞 부분 및 근접 조직을 .포함하고 있으며 , 환경에 지속적으로 노출되기 때문에 특히 염증 질환이 발생할 가능성이 높다.
외안부 및 전안부의 염증성 질환은, 각막염, 결막염, 각결막염, 안구 건조 증후군, 안검염, 공막염 상공막염, 홍채염, 흥채모양체염, 포도막염 , 또는 수술 후 염증을 포함한다. 이 중에서, 각결막염은 세균, 바이러스, 알레르기, 또는 환경적 요인에 의해 각막과 결막에 발생하는 염증 질환으로서, 가장 일반적으로 발병되는 안질환이며, 보통은 잘 치유되나 경우에 따라서는 치명적이어서 조직손상에 의한 실명까지 초래할 수 있다. 이러한 안구 염증성 질환의 치료 방법으로 현재로서는 항히스타민제, 글루코코르티코이드 등의 스테로이드제 또는 알레르기 예방, 차단제 등을 사용하고 있다.
그러나, 안구 염증 질환을 치료하기 위한 항생제의 사용은 감염된 세균에 대한 살균력은 있으나 각막 내 염증세포에 대한 활성억제와 감소에 대한 효과는 기대하기 어려운 문제가 있다. 또한, 스테로이드성 항염증 약물의 경우 장기간 사용시 또는 장기간 사용 후 사용 중단 시, 급성 부신 기능 부전으로 인해 전신 쇠약감, 발열, 근육통, 관절통 및 식욕감퇴 등의 증상이 나타나며, 세균, 바이러스 등에 대한 감염의 위험성이 증가하고, 체중이 늘고 체형이 변하며, 불면증이 잘 동반되는 문제가 있다. 특히, 글루코코르티코이드의 점안제 또는 연고는 환자의 자의적인 판단에 의해 장기적으로 사용할 경우 안압 상승으로 인한 녹내장, 백내장 또는 세균성 감염 등의 부작용을 유발하기 쉽다.
이에, 과거 몇 년 동안 안구 염증 질환의 치료 및 예방에 관한 기술이 많이 진보되었지만, 여전히 안구 염증 질환을 예방 및 치료하는 향상된 방법 및 조성물이 요구되고 있는 실정이다.
본 발명에서는 안구 염증 질환의 치료를 위하여 사용되는 항생제 또는 스테로이드성 항염증 점안제의 문제점인 각막상피세포의 손상 회복력 약화의 문제를 개선하고자, 세포의 재생이 중요한 각막상피의 손상회복에 대한 치료제로서 설파살라진 -히알루론산 복합 점안제를 개발하였다.
이와 관련하여, 한국등록특허 제 0490286호는 설파살라진 또는 그 수용성 염을 활성성분으로 하는, 녹내장에 의한 망막의 허혈, 당뇨병성 망막증에 의한 망막 손상, 포도막염에 의한 망막 신경의 퇴화 등을 치료할 수 있는 망막손상 치료용 안약 조성물을 제공하고 있다.
또한, 한국등록특허 제 0894042호는 설파살라진 -히알루론산 흔합물을 이용하여 후발성 백내장을 억제할 수 있는 조성물을 제공하고 있다.
그러나, 상기 특허문헌들을 비롯한 선행문헌의 어디에도, 친수성 설파살라진 및 히알루론산 복합제를 안과 염증 질환, 특히 외안부 및 전안부의 염증성 질환의 치료에 적용하는 기술을 알려진 바가 없다. 【발명의 상세한 설명】
【기술적 과제】
본 발명자들은, 각막 염증 동물 모델을 이용하여 친수성 설파살라진 및 히알루론산 복합 점안제의 염증 억제, 각막 부종 억제 효과 및 안구 건조 치료 효과를 확인하여 본 발명을 완성하였다.
【기술적 해결방법】
본 발명의 하나의 목적은, 친수성 설파살라진 및 히알루론산을 포함하는 안과 염증 질환 치료용 점안제 조성물을 제공하는 것이다.
또한, 본 발명은 친수성 설파살라진 및 히알루론산의 치료학적 유효량을 개체에 투여하는 단계를 포함하는, 안과 염증 질환의 치료 방법을 제공하는 것이다.
본 발명의 또 하나의 목적은, 상기 친수성 설파살라진 및 히알루론산을 포함하는 안과 염증 질환의 치료용 점안제 조성물의 제조 방법을 제공하는 것이다.
【유리한 효과】
본 발명의 친수성 설파살라진 및 히알루론산 복합 점안제는 안과 염증 질환 치료제로서, 종래의 항생제 또는 스테로이드성 약물 기반의 치료제로 인한 부작용의 발생을 낮추는 비스테로이드성 치료제로 안전하게 사용될 수 있다.

하나의 양태로서, 본 발명은 친수성 설파살라진 및 히알루론산을 포함하는, 안과 염증 질환 치료용 점안제 조성물에 관한 것이다.
또 하나의 양태로서, 본 발명은 친수성 설파살라진 및 히알루론산의 치료학적 유효량을 개체에 투여하는 단계를 포함하는, 안과 염증 질환 치료 방법에 관한 것이다.
본 발명에서 설파살라진 (Sulfasalazine) 은, 설파피리딘 (Sulfapyridine)과 5—아미노살리실산 (5-aminosal icylic acid, 5-ASA)이 아조 결합 (azo bond)하여 형성된 화합물을 말한다.
상기 설파살라진은 수용액에 대하여 난용성이어서, 약제로 사용되기 위해서는 친수성 설파살라진으로 가용화할 필요가 있다. 이러한 친수성 설파살라진은 설파살라진에 염산, 염화나트륨 또는 염화칼륨 등을 첨가하여 산 부가염 또는 알칼리 부가염의 형태로 얻어 질 수 있고, 보다 바람직하게는 설파살라진을 페그화 (PEGylation) 하여 얻어질 수 있다.
바람직하게, 본 발명의 친수성 설파살라진은 페그화된 설파살라진을 포함할 수 있다. 상기 페그화된 설파살라진은 설파살라진을 폴리에틸렌글라이콜을 포함하는 균형염용액에 첨가하여 얻어 질 수 있다. 이러한 균형염용액은 다양한 분자량 및 농도의 폴리에틸렌글라이콜을 포함할 수 있는데, 예를 들어 , 300 내지 500, 바람직하게는 380 내지 420의 중량평균분자량을 갖는 폴리에틸렌글라이콜을 0.1%에서 5% (v/v), 바람직하게는 1% (v/v) 이상의 농도로 포함할 수 있다.

한편ᅳ 본 발명에서 히알루론산 (hyaluronic acid)은 결합조직 내의 세포 간질에 널리 분포되어 생리 조건 하에서 점착성 및 탄력성 용액을 형성함으로써, 흥채, 망막 등의 조직, 혈관 내피세포, 상피세포 등을 기계적으로 보호하며 완층역할을 제공하는 것으로 알려진, 세포외 매트릭스의 주요 성분이다.
본 발명에서 상기 히알루론산은 물성, 형태,크기 등에 제한이 없으며 무균 처리된 것이 바람직하나, 보다 바람직하게는, 1.0 X 106내지 4.0 X 106 g/몰의 중량평균분자량을 갖는 히알루론산을 사용할 수 있다.
또한, 상기 히알루론산은 약제학적으로 허용가능한 염의 형태로 사용될 수 있는데, 상기 약제학적으로 허용가능한 염은 나트륨염, 칼륨염 , 칼슘염 등을 포함하며, 바람직하게는 나트륨염을 포함한다.
바람직하게, 본 발명에서는, 상기 히알루론산의 농도가 0.01 내지
0.5 %(w/v) , 보다 바람직하게는 0.05 내지 0.2 %(\v/v)인 것이 바람직하다. 상기 범위 미만으로 사용될 경우 점탄성이 너무 낮아 습윤 및 윤활 작용에 부적절한 문제가 발생할 수 있고, 상기 범위를 초과하는 경우 과다한 고점탄성으로 인해 침투성에 부적절한 문제가 발생하여 점안제로서의 적합한 유동성을 확보하기 어려운 문제가 있다.
본 발명자들은 한국등록특허 0894042호에서, 설파살라진—히알루론산 흔합물을 이용하여 후발성 백내장을 억제할 수 있는 조성물을 제공한 바 있다. 상기 선행 기술에서는, 후발성 백내장 수술시에 설파살라진 -히알루론산 흔합물을 주입하여 안구 내의 볼륨을 유지 시켜야 하기 때문에, 2.0 %(w/v)의 고농도의 히알루론산이 흔합된 고점탄성 조성물로 제조할 필요가 있었다.
이에 반하여, 본 발명에서는 설파살라진 -히알루론산 흔합물이 안과 염증 질환의 치료에 효능을 가진다는 것을 새로이 발견하였으며, 안과 염증 질환 치료를 위한 점안제 조성물로 제공하기 위하여 히알루론산의 농도를 0.01 내지 0.5 %(w/v) 로 낮추어 히알루론산이 완전히 용해된 조성물로 제제화한 특징이 있다.
본 발명에서 친수성 설파살라진 및 히알루론산 흔합물은, 안과 염증 질환의 치료, 개선 및 /또는 완화를 위하여 사용될 수 있다.
본 발명의 점안제는, 약학적으로 허용 가능한 첨가제를 더욱 포함할 수 있다. 상기 약학적으로 허용가능한 첨가제란, 생물체를 상당히 자극하지 않고 투여 유효성분의 생물학적 활성 및 특성을 저해하지 않는 담체 또는 희석제를 말한다. 상기 약학적으로 허용가능한 첨가제의 예로는 등장화제, 완층제, 안정계, H조절제 등이 있다.
본 .발명에 따른 점안제는 액제의 제형인 것이 바람직하므로, 수용액을 포함하는 담체를 더욱 포함할 수 있다.
상기 수용액을 포함하는 담체는 증류수, 인산완층액 (Phosphate Buffered Saline), 균형염용액 (Balanced Salt Solution) 및 생리식염수 (Saline)로 이루어진 군에서 선택된 1종 이상의 약학적으로 허용되는 담체를 포함할 수'있다. 이 때 사용되는 담체의 함량은 제조하고자 하는 점안액의 총 용량에 필요한 양에 맞추어 사용량을 조절할 수 있다.
또한, 본 발명의 점안제는, 약학적으로 허용되는 염을 더 포함할 수 있다. 상기 약학적으로 허용되는 염의 구체적인 예로는 염산, 염화나트륨, 염화칼륨 또는 이들의 흔합물을 예시할 수 있다.

본 발명에서 안과 염증 질환은 안구에 관련된 염증 질환을 모두 포함할 수 있다. 상기 염증이란, 안구의 층혈, 부기, 감염, 자극 또는 외상과 관련된 증상을 의미할 수 있다.
일예로, 본 발명의 안과 염증 질환은, 외안부 및 전안부의 염증성 질환을 포함할 수 있다.
예를 들어, 상기 외안부 및 전안부의 염증성 질환은, 각막염, 결막염, 각결막염 , 안구 건조 증후군, 안검염, 공막염 , 상공막염 , 흥채염, 흥채모양체염, 포도막염, 또는 수술 후 염증을 포함할 수 있다.
다른 일예로, 본 발명의 안과 염증 질환은, 각막, 결막 또는 각결막에 관련된 염증 질환을 포함할 수 있으며, 예를 들어, 다음과 같은 질환을 예시할 수 있다:
1) 안구건조 관련 염증 질환 : 건조성 각결막염 또는 이와 관련된 안 염증, 눈물 생성이 억제된 환자에 있어 눈물 부족으로 인한 안 염증 또는 안구건조증과 관련된 각결막 상피 장해
2)비감염성 각결막염 : 외부 손상에 의하여 발생하는 각결막염 (예를 들어, 직접 각막에 손상이 입었을 경우, 산 또는 염에 의한 손상에 의한 경우, 화상에 의한 손상에 의한 경우, 또는 콘택트 렌즈 착용에 의한 경우 등), 알레르기성 각결막염 (예를 들어, 고초열성 결막염 춘계 각결막염 , 아토피성 각결막염, 플릭텐성 각결막염, 접촉성 안검염에 의한 결막염 등), 심한 부종과 혈관을 동반하는 충혈이 있는 급만성 비감염성 결막염, 전안부의 비감염성 염증 질환 (예를 들어, 전포도막염, 상공막염, 공막염), 또는 안과 수술 (예를 들어, 사시절개술, 백내장, 녹내장 수술) 후의 각결막염, 또는
3) 감염성 각결막염 : 바이러스성 각결막염, 세균성 각결막염, 진균성 각결막염, 클라미디아성 각결막염, 리케치아 각결막염, 또는 기생층성 각결막염.
또 하나의 양태로서, 본 발명은 상기 친수성 설파살라진 및 히알루론산을 포함하는 각결막염 치료용 점안제 조성물의 제조 방법에 관한 것이다.
바람직한 양태로서 , 본 발명은
(a) 설파살라진을 친수화 시키는 단계; 및
(b) 상기 친수화된 설파살라진에 히알루론산을 첨가 및 흔합하는 단계를 포함하는, 각결막염 치료용 점안제 조성물의 제조 방법에 관한 것이다.
상기 (a) 단계는 설파살라진을 친수화 시키는 단계로, 난용성 화합물을 가용화 시키기 위하여 통상적으로 사용되는 것으로 알려진 방법 및 장치를 별다른 제한 없이 사용할 수 있으나, 바람직하게는 설파살라진을 페그화 (PEGylation)시키는 단계를 포함할 수 있다.
구체적으로, 설파살라진을 페그화 시키는 단계는 설파살라진을 폴리에틸렌글라이콜을 포함하는 균형염용액에 첨가하는 단계를 포함할 수 있다. 상기 균형염용액에 포함되는 폴리에틸렌글라이콜은 설파살라진을 페그화하여 친수화 시킬 수 있다. 이와 같이, 설파살라진을 페그화하여 친수화 시키면, 화학적 구조의 변경 없이 수용성의 액체로 사용할 수 있고, 점안제로 용이하게 제작할 수 있다. 상기 설파살라진 및 폴리에틸렌글라이콜에 관한 구체적인 내용은 상술한 내용과 같다.
바람직하게, 상기 (a) 단계는 친수화된 설파살라진의 농도가 0.005 내지 0.1 %(w/v)가 되도록 조절하는 단계를 포함할 수 있다.
상기 (b) 단계는 상기 친수화된 설파살라진에 히알루론산을 첨가 및 흔합하는 단계로, 친수화된 설파살라진이 포함된 균형염용액에 적절한 양의 히알루론산을 첨가하여 농도를 조절할 수 있으며 , 상기 히알루론산의 물성, 형태, 크기 등에는 제한이 없으며, 무균 처리된 것이 바람직하다.
바람직하게, 상기 (b) 단계는 히알루론산의 농도가 0.01 에서 0.5 (w/v) 가 되도록 조절하는 단계를 포함할 수 있다.
또한, 친수화된 설파살라진에 히알루론산을 흔합하는 단계는, 친수화된 설파살라진 및 히알루론산을 20 내지 35°C 에서 6 시간 내지 24 시간 흔합하는 것이 바람직하다. 상기 흔합시의 온도 범위 미만인 경우에는, 점안제의 성분들이 균질하게 흔합되지 않을 수 있고, 상기 온도 범위 초과인 경에는 제제의 안정적인 조제가 쉽지 않기 때문에 바람직하지 않다. 또한, 상기 흔합 시간 범위 미만인 경우에는, 점안제의 성분들이 균질하게 흔합되지 않을 수 있고, 상기 시간을 초과하는 경우에는 히알루론산의 점도에 변화가 생길 수 있다.
한편, 상기 각결막염 치료용 점안제 조성물의 제조 방법에서는, 약학적으로 허용되는 첨가제 , 담체 또는 염을 첨가하는 단계를 더 포함할 수 있다. 이러한 약제학적으로 허용되는 담체 또는 염은 상기 각결막염 치료용 점안제 조성물의 제조하는 과정에서 별 다른 제한 없이 첨가될 수 있으나, 상기 친수화된 설파살라진 및 히알루론산이 완전히 흔합된 이후에 첨가하는 것이 바람직하다.
【발명의 실시를 위한 형태】
이하, 본 발명을 실시예에 의해 상세히 설명한다. 단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명이 하기 실시예에 의해 한정되는 것은 아니다. 실시예 1. 설파살라진 -히알루론산 복합 점안제의 제조
설파살라진 (Sigma Aldrich사)을 30%(w/v)의 폴리에틸렌글라이콜 (분자량 400, Sigma Aldrich사)이 흔합된 균형염용액 (pH 7.4) 10 ml 에 5mg/ml 의 농도로 가한 다음, 교반기에서 6시간 동한 흔합하여 페그화 (Pegylation)하여 가용화여 가용화된 설파살라진을 제조하였다.
다음으로, 10ml의 균형염용액에 0.1¾(w/v)로 히알루론산 가루 (Sigma Aldrich사)를 흔합하고, 설파살라진이 0.05%(0.5mg/ml )의 농도가 되도톡 상기 페그화된 설파살라진 포함 용액을 히알루론산과 흔합하였다. 설파살라진의 농도는 페그화단계에서 조절하였다. 모든 용액이 균질화 되도록 상온에서 6시간 교반하였다. 균질화되어 투명한 맑은 용액이 되면 점안제로 사용하였다 (도 1). 실시예 2. 설파살라진 -히알루론산 복합 점안제의 농도별 각막상피세포 독성 확인
각막상피세포에 대한 세포독성을 관찰하기 위하여, 사람의 각막상피세포주 (Human corneal epithelial cell line, HCET)를 배양하였고, 배양된 세포에 대한 독성은 CCK assay (Cell Counting Kit-8, Dojindo molecular technologies, INC)를 이용하여 확인하였다. 보다 상세하게, 본 발명에서 사용된 각막상피세포는 시미안 바이러스
40(Simian Virus 40)를 감염시켜 만든 세포인 HCET 세포를 사용하였다. 본 세포는 일본 오사카대학교 의과대학의 Kaoru Araki-Sasaki 교수로부터 분양 받았다. 각막상피세포는 5% FBSCfetal bovine serum), 5 mg/mL 인술린, 0.1 mg/mL 콜레라톡신, 10 ng/mL hEGF( human epidermal growth factor) 및 0.5% DMS0(dimethyl sulfoxide)를 첨가한 세포배양액 (MEM:Ham F12 = 1:1)을 사용하여 5% C02 가 유지되는 37 °C의 항온 항습기에서 배양하였다. 본 실시예에서는 각막상피세포 (HCET)는 6-웰 플레이트에 각 웰당 2 X 104 (eel ls/well)로 배양하였다.
위의 조건으로 배양된 HCET 세포의 배양 24시간 후에, 설파살라진 -히알루론산 복합 점안제를 설파살라진 농도만 다르게 하여 (0.005~5.0 mg/ml) 혈청이 없는 배양액에 배양액대 점안제를 5:1로 처리하였다. 대조군은 혈청이 포함되지 않은 배양액올 사용하였다. 약물 처리 12시간 후에 CCK assay를 하여 세포 독성을 확인하였다.
그 결과, 설파살라진 히알루론산 복합 점안제는 설파살라진의 농도를
5mg/ml (0.5%, w/v) 까지 올렸을 때, 70% 이상의 세포 생존율을 보였다 (도 2). 실시예 3. 설파살라진 -히알루론산 복합 점안제의 각결막 염증 억쎄 S^:
동물은 흰쥐 (Sprague daw ley, 300g, male)를 사용하였고 NaOH를 이용한 화학약물에 의한 각막손상 모델 (alkali burn)을 이용하였다. 3mm paper disk에 0.1M의 NaOH용액 (50ul)를 적시고, 각막에 30초간 덮어 각막손상을 유도하였다. 각막에서 염증반웅은 헤마톡실린 에오신 염색과 F4/80항원에 대한 면역 염색을 수행하여 확인하였다. 각막 부종에 대한 분석은 각막 상피세포층 (corneal epithelium)과 각막 기질층 (corneal stroma)의 비로 분석하였다.
각막 손상 후, 실시예 1에서 제조한 가용화한 설파살라진, 및 설파살라진 -히알루론산 점안제를 각각 하루에 두 번 투여하였다. 대조군은 히알루론산 0.1%를 투여하였다.
각막 손상 후, 3일 후에 안구를 적출하고 파라핀 블럭을 만들어 조직절편을 만든 다음, 헤마톡실린 에오신 염색과 마크로파아지 마커인 F4/80 항원으로 면역염색을 시행하여 관찰한 결과, 대조군 (control)에서는 각막의 부종과 염증세포의 각막 침투가 관찰되었으나, 설파살라진 -히알루론산 복합 점안제를 투여한 군에서는 부종 및 염증세포의 수가 줄어들어 있었다. 또한, 설파살라진 -히알루론산 복합 점안제는 히알루론산과 흔합되지 않은 같은 농도의 설파살라진 용액보다 염증억제 및 부종억제에 효과가 우수하였다 (도 3).
정량적인 결과를 보면, 정상의 각막상피세포 (corneal epithelium)와 기질 층 (stroma)의 비는 4.6土 1이었으나 각막염증 유도 후에는 10±2.5로 증가하였다. 설파살라진 -히알루론산 복합 점안제를 투여한 실험군의 경우 0.05mg/m 1(0.005%) 투여군에서는 7.6±1.8로, 0. lmg/ml (0.01%) 투여군에서는
5.6土 1.4로 관찰되어, 각막 부종이 억제되었음을 확인할 수 있었다. 또한, 0.1mg/ml(0.01%)의 설파살라진 용액을 투여한 실험군에서는 7.1±2.1로 관찰되어, 결과적으로 설파살라진 -히알루론산 복합 점안제가 히알루론산과 흔합되지 않은 같은 농도의 설파살라진 용액보다 각막 부종 억제 효과가 우수하였다 (도 4).
염증세포에 대한 분석은 각막에 존재하는 마크로파지 (F4/80 positive eel Is)의 수로 실시하였으며, 결과는 각막염증 유도 후에는 31.5±4.9개로 관찰되었고, 히알루론산이 없는 설파살라진 용액 (0.1mg/ml(0.01%))을 투여한 군에서는 21.3±5.5개로, 설파살라진 -히알루론산 복합 점안제 투여군의 경우 0. lmg/ml (0.01%) 투여군에서 16.2±3.3 개의 마크로파지가 관찰되었다 (도 5). 실시예 4. 설파살라진 -히알루론산 복합 점안제의 건조성 각 결막염에서 염증 억제 효과
건조성 각결막염 (Keratoconjunctivitis sicca)에 대한 실험을 위하여 건성안 (dry eye) 동물모델을 제작하였다. 동물은 휜쥐 (sd rat)를 사용하였고, 건조성 각결막염의 유도는 1% 황산 아트로핀 (1% atropine sulfate)과 0.1¾) 벤잘코니움 클로라이드 (0.1% benzalkonium chloride)를 하루에 두 번씩 2주간 점안하여 유도하였다.
설파살라진 -히알루론산 복합제 (0.1mg/ml)의 투여는 1% 아트로핀 (1% atropine sulfate)과 0.1% 벤잘코니움 클로라이드 (0.1% benzalkonium chloride) 투여 후 일주일부터 일주일간 하루에 두 번 점안 투여하였다. 비교약물로 0.1% 히알루론산과 0.05¾>사이클로스포린 점안제를 사용하였다. 건조성 각결막염 동물모델에서 각막상피 손상의 확인은 2% 플루오르세인 용액으로 각막상피가 손상된 부위를 염색하여 확인하였고 (도 6), 각막의 염증은 각막올 냉동 절삭 (cryosection)하여 헤마톡실린과 에오신 염색을 실시하여 확인하였다 (도 8).
건조성 각결막염 (건성안)이 유도된 각막에서 건조로 인한 각막 상피세포의 상태를 플루오르세인 염색을 시행하여 건조로 인한 각막상피세포의 손상이 있음을 확인하였다 (도 6). 플루오르세인 염색약은 건강한 정상 각막상피세포는 염색되지 않고 손상되었거나 각막상피가 없는 각막 부위가 염색되는 특징을 가지고 있다 (노란색으로 염색된 영역). 플루오르세인 염색 후 염색된 면적은 전체 각막 면적에 대해 %로 나타내었다. 、대조군 (control, 약물 비처리군)에서는 76土 6.5%로 각막표면의 대부분이 염색이 되었으나, 히알루론산 (0.1%) 54 ± 7.4%, 사이클로스포린 (0.05%) 29 ±4.2%,설파살라진 -히알루론산 복합제 (0.01%)에서는 37 ± 5.7¾>로 각막표면의 형광염색이 현저하게 줄어든 것을 확인할 수 있었다. (도 6 7). 이것으로 제조된 설파살라진 -히알루론산 복합제 (0.01%)가 각막상피세포의 손상을 억제하며, 보호효과가 있는 것으로 관찰되었다.
건조성 각결막염 (건성안)에서 각막내의 염증세포를 확인하기 위하여 헤마톡실린-에오신 염색을 시행한 결과, 건성안의 각막에서 등글고 다핵 (polynuclear)을 가진 세포들 (polynucleocytes)이 관찰되었다 (도 8). 건조성 각결막염 모델에서 염증세포의 각막 침입 억제효과는 대조군 (control, 약물 비처리군)에 대하여 비율로 나타내었다. 염증 억제 효과는 히알루론산 (0.1¾ 0.76土 0.1 사이클로스포린 (0.05%) 0.35 ± 0.07, 설파살라진 -히알루론산 복합제 (0.01%)에서는 0.48土 0.09로 관찰되었다 (도 9). 위의 동블실험 결과로 설파살라진 -히알루론산 복합제 (0.01%)가 건조성 각결막염에 대해 염증억제 효과가 있음을 확인하였다. 위의 실시예의 결과는 설파살라진 -히알루론산 복합 점안제는각막염증의 완화에 효과가 있으며, 설파살라진 단독 투여보다 효과가 크다는 것을 의미한다.
위의 결과로 비추어 볼 때, 본 발명의 설파살라진—히알루론산 복합 점안제는 안구, 특히 전안부 또는 외안부의 염증 치료에 효과적으로 사용될 수 있다.

건조성 각결막염 또는 이와 관련된 안 염증, 눈물 부족으로 인한 안 염증, 또는 안구건조증과 관련된 각결막 상피 장해를 포함하는 안구건조 관련 염증 질환;
외부 손상에 의하여 발생하는 각결막염, 알레르기성 각결막염, 부종과 혈관을 동반하는 층혈이 있는 급만성 비감염성 결막염, 전안부의 비감염성 염증 질환, 또는 안과 수술 후의 각결막염을 포함하는 비감염성 각결막염; 또는
바이러스성 각결막염, 세균성 각결막염, 진균성 각결막염, 클라미디아성 각결막염, 리케치아 각결막염, 또는 기생층성 각결막염을 포함하는 감염성 각결막염인 조성물.
