Abstract

본 발명은 속단 및 백수오 추출물을 유효성분으로 포함하는 운동능력 및/또는 근육 형성 증진용 조성물에 대한 것이다. 본 발명에 따른 조성물 섭취시 운동수행 능력이 증진되며, 근육의 발달을 증진하는 데 유용하다.

속단 및 백수오 추출물을 유효성분으로 포함하는 운동능력 및 근육 증진용 조성물
본 발명은 속단 및 백수오 추출물을 유효성분으로 포함하는 운동능력 및/또는 근육 발달 증진용 조성물에 대한 것이다.
일반적으로 근육은 지속적으로 운동을 하지 않을 경우 노화에 의해 근육기능이 저하되고 근육량 및 근육신경연접(neuromuscular junction, motor unit)의 감소가 일어나 쉽게 피로를 느끼며 무기력해져서 생활의 활력이 감소되고 삶의 질이 급격히 떨어진다(J. Appl. Physiol. 2003, 95, p1717-1727). 이를 방지하기 위하여 저항력운동(resistance training)과 같은 운동을 지속적으로 실시하고(대한민국공개특허 10-2009-0089815) 이와 더불어 적절한 식이요법을 함께하는 것이 권장되고 있다. 이와 같이, 삶의 질을 향상시키기 위해서는 규칙적인 운동이 필요하고, 운동 선수 뿐만 아니라 일반인들도 일상 생활에서 보다 많은 에너지 및 지구력을 필요로 하게 되었다. 하지만 바쁜 생활에 쫓기는 현대인의 경우 대부분 운동 보다는 건강보조제(dietary supplement)에 의존하려는 경향이 있다. 따라서 육체적 운동수행능력을 향상시키기 위한 보조제, 기능성 식품, 식품 조성물 등의 연구가 오랫동안 지속되어 왔으며 실제로 스테로이드, 카페인 등의 화합물을 복용하면 운동 능력을 증가되는 것으로 알려져 있다. 그러나 이러한 약물은 치명적인 부작용을 동반할 수 있으므로 그 사용이 극히 제한된다.

본 발명은 상기의 문제점을 해결하고 상기의 필요성에 의하여 안출된 것으로서 본 발명의 목적은 신규한 운동능력 향상용 조성물을 제공하는 것이다.
본 발명의 다른 목적은 근육 발달 증진용 조성물을 제공하는 것이다.
상기의 목적을 달성하기 위하여 본 발명은 속단 추출물 및 백수오 추출물을 유효성분으로 포함하는 운동능력 증진용 식품 조성물을 제공한다.
본 발명의 일 구현예에 있어서, 상기 속단 및 백수오 추출물은 물 또는 탄소수 1 내지 6개의 알코올 및 이들의 혼합용매로 이루어진 군에서 선택된 어느 하나의 용매로 추출한 것이 바람직하나 이에 한정되지 아니한다.
본 발명의 바람직한 실시예에 있어서 속단 추출물과 백수오 추출물의 혼합비율은 1:0.1 내지 1:10의 중량비로 혼합되는 것이 바람직하고 1:1의 중량비로 혼합되는 것이 더욱 바람직하나 이에 한정되지 아니한다.
또 본 발명은 속단 추출물 및 백수오 추출물을 유효성분으로 포함하는 근육 발달 증진용 식품 조성물을 제공한다.
또 본 발명은 속단 추출물 및 백수오 추출물을 유효성분으로 포함하는 운동능력 증진용 약학 조성물을 제공한다.
또 본 발명은 속단 추출물 및 백수오 추출물을 유효성분으로 포함하는 근육 발달 증진용 약학 조성물을 제공한다.
이하 본 발명을 설명한다.
본 발명자는 운동능력을 향상시키며 근육발달을 증진하는 천연물질을 탐색하는 연구를 수행한 결과, 속단 추출물과 백수오 추출물의 혼합물을 섭취하는 경우 운동 능력을 현저히 향상시키며, 우수한 근육 발달 증진 효과를 나타내는 것을 확인하여 본 발명을 완성하였다.
본 발명에 의한 운동능력이나 근육 발달 증진을 위한 조성물은 그 제형이 특별히 제한되지 않으나, 예를 들어 약학 조성물 또는 건강식품 조성물일 수 있다.
본 발명에 의한 조성물이 약학 조성물인 경우 통상의 방법에 따라 경구용 제형, 외용제, 좌제 및 멸균 주사용액 등의 형태로 제형화하여 사용될 수 있다.
또한, 필요에 따라서 상기 조성물에 하기의 첨가제의 1종 또는 2종 이상을 첨가 배합할 수 있다. 상기 첨가제로는, 예를 들어 그레이프프루트, 사과, 오렌지, 레몬, 파인애플, 바나나, 배 등의 각종 과즙(농축 과즙, 분말 과즙 등이어도 좋다); 비타민류(팔미트산 레티놀, 리보플라빈, 피리독신,시아노코발아민 (cyanocobalamine), 아스코르빈산 나트륨, 니코틴산 아미드, 판토텐산 칼슘, 엽산, 비오틴, 콜레칼시페롤(cholecalciferol), 중주석산콜린, 토코페롤 또는 β-카로틴 등의 수용성 및 지용성 비타민류); 향미료(레몬플레이버, 오렌지플레이버, 딸기 플레이버, 그레이프프루트플레이버 또는 바닐라 에센스 등); 아미노산, 핵산 및 그들의 염류(글루탐산, 글루탐산나트륨, 글리신, 알라닌, 아스파라긴산, 아스파라긴산 나트륨, 이노신산 등); 식물 섬유(폴리덱스트로오스,펙틴, 크산탄 고무, 글루코만난 또는 알긴산 등); 또는 미네랄류(염화나트륨, 초산나트륨, 황산마그네슘, 염화칼륨, 염화마그네슘, 탄산마그네슘, 염화칼슘, 인산 2칼륨, 인산 1나트륨, 글리세로인산칼슘, 구연산제1철나트륨, 구연산철암모늄, 구연산철, 황산망간, 황산구리, 요오드화나트륨, 솔빈산칼륨, 아연, 망간, 구리, 요오드 또는 코발트 등) 등이 포함될 수 있다.
상기 조성물에는 방부제, 안정화제, 수화제 또는 유화 촉진제, 삼투압 조절을 위한 염 및/또는 완충제 등의 약제학적 보조제 및 기타 치료적으로 유용한 물질을 추가로 함유할 수 있으며, 통상적인 방법에 따라 다양한 경구용 또는 비경구용 투여 형태로 제형화할 수 있다.
경구용 제형으로는 예를 들면, 정제, 환제, 경질 및 연질 캅셀제, 액제, 현탁제, 유화제, 시럽제, 과립제 등이 있는데, 이들 제형은 유효성분 이외에 희석제(예: 락토오스, 덱스트로오스, 수크로오스, 만니톨, 솔비톨, 셀룰로오스 및 글리신), 활택제(예: 실리카, 탈크, 스테아르산 및 그의 마그네슘 또는 칼슘염 및 폴리에틸렌글리콜)를 함유하고 있다. 정제는 마그네슘 알루미늄 실리케이트, 전분 페이스트, 젤라틴, 트라가칸스, 메틸셀룰로오스, 나트륨 카복시메틸셀룰로오스 및 폴리비닐피롤리딘과 같은 결합제를 더 함유할 수 있으며, 경우에 따라 전분, 한천, 알긴산 또는 그의 나트륨 염과 같은 붕해제, 흡수제, 착색제, 향미제, 및 감미제 등의 약제학적 첨가제를 함유할 수 있다. 정제는 통상적인 혼합, 과립화 또는 코팅 방법에 의해 제조될 수 있다. 또한, 비경구 투여용 제형의 대표적인 것은 주사용 제형으로 등장성 수용액 또는 현탁액이 바람직하다.
또한 본 발명에 의한 조성물이 건강식품 조성물이 경우에는, 예를 들어, 각종 식품류, 음료, 껌, 차, 비타민 복합제 또는 건강 기능성 식품류 등으로 제형화될 수 있다. 본 발명에서 사용하는 백수오 및 속단은 독성 및 부작용이 거의 없으므로 예방 목적으로 장기간 복용 시에도 안심하고 사용할 수 있다.
본 발명의 건강식품 조성물은 지시된 비율로 필수 성분으로서 본 발명의 추출물을 함유하는 외에는 다른 성분에는 특별한 제한이 없으며 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상술한 천연 탄수화물의 예는 모노사카라이드, 예를 들어, 포도당, 과당 등; 디사카라이드, 예를 들어 말토오스, 수크로오스 등; 및 폴리사카라이드, 예를 들어 덱스트린, 시클로덱스트린 등과 같은 통상적인 당, 및 자일리톨, 소르비톨, 에리트리톨 등의 당알콜이다. 상술한 것 이외의 향미제로서 천연 향미제(타우마틴,스테비아 추출물(예를 들어 레바우디오시드 A, 글리시르히진등) 및 합성 향미제(사카린, 아스파르탐 등)를 유리하게 사용할 수 있다. 상기 천연 탄수화물의 비율은 본 발명의 조성물 100 ㎖당 일반적으로 약 1∼20 g, 바람직하게는 약 5∼12 g이다.
상기 외에 본 발명의 조성물은 여러 가지 영양제, 비타민, 광물(전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 향미증진제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올 또는 탄산 음료에 사용되는 탄산화제 등을 함유할 수 있다. 그밖에 본 발명의 조성물들은 천연 과일 쥬스 및 과일 쥬스 음료 및 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율은 그렇게 중요하진 않지만 본 발명의 조성물 총 중량에 대하여 0∼20 중량%의 범위에서 선택되는 것이 일반적이다.
상기 본 발명의 추출물의 투여량은 당업자의 수준 내의 범위에서 한정될 수 있으며, 본 발명에 따른 조성물의 1일 투여 용량은 투여하고자 하는 대상의 미만 진행 정도,발병 시기, 연령, 건강상태, 합병증 등의 다양한 요인에 따라 달라지지만, 성인을 기준으로 할 때 일반적으로는 상기 언급된 중량비로 조합된 조성물 1 내지 5000 mg/kg, 바람직하게는 30 내지 1000 mg/kg을 1일 1 내지 2회 분할하여 투여할 수 있으며, 상기 투여량은 어떠한 방법으로도 본 발명의 범위를 한정하는 것이 아니다.
상기 본 발명의 추출물의 함량은 특별히 제한되지 않으나, 조성물 총 중량에 대하여 10∼90 중량% 범위로 함유되는 것이 바람직하다. 이는 정제 및 연질캡슐 제조시, 분말 및 기능성 성분의 함량이 10∼60%, 하드캡슐의 제조 시, 분말 및 기능성 성분의 함량이 10∼90%일 수 있는 점을 고려하여, 본 발명의 추출물을 10∼90%로 함유하는 건강식품 조성물 또는 약학 조성물을 제공할 수 있다.
본 발명의 일 실시 양태는 상기 정의된 조성물을 1회 운동 전, 운동 중 및/또는 운동 후에 섭취할 수 있다. '휴식시간 전'이란 취침전 저녁때를 뜻하는 것이고, '운동'이란 예를 들면 체조, 걷기, 조깅, 사이클, 골프, 테니스, 수영, 마라톤 또는 3종 경기를 불문하고 통상적인 의미에서 근력을 내는 행위를 말하는 것이다. '운동 후'란 1회 운동 직후 1시간 후을 말하며, 바람직하게는 운동중에 시간 간격이 있을 경우에는 전체 운동을 마친 후를 뜻한다. 더욱이, 운동이 장시간동안 진행되는 경우, 예를 들면 마라톤 또는 3종 경기와 같은 경우에는 본 발명의 조성물을 바람직하게는 운동중에 섭취한다.
따라서 본 발명은 속단 및 백수오 추출물을 유효성분으로 포함하는 운동능력 향상 또는 근육 발달 증진용 식품 조성물을 제공한다. 본 발명은 근육 발달을 증진시켜 운동 능력을 향상시키게 하며, 근육 발달의 증진이나 운동능력 향상의 기능성을 가지는 식품 제조에 효과적이다.
도 1은 본 발명의 조성물 또는 대조군의 조성물을 섭취한 후 강제 수영 운동을 실시하여 최대 운동 능력을 측정한 결과 그래프이다.
도 2는 본 발명의 조성물 또는 대조군의 조성물을 섭취한 후 트레드밀 운동을 실시하여 최대 운동 능력을 측정한 결과 그래프이다.
도 3은 본 발명의 조성물 또는 대조군의 조성물을 섭취한 후, 비복근의 근육량을 측정한 결과 그래프이다.
이하, 본 발명을 실시예에 의해 상세히 설명한다.단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예에 한정되는 것은 아니다.
<실시예 1>
백수오 및 속단 추출물의 제조
건조된 백수오와 속단 각 100 g을 길이 2-5 mm 정도의 크기로 분쇄한 후 물 2 L를 혼합하고 10시간동안 교반시키면서 가열 추출하였다. 추출 용액을 섭씨 70도까지 냉각하고, 백수오와 속단 중량 대비 3%의 α-아밀라제를 추출액에 첨가하고 섭씨 70도에서 6시간동안 반응시켰다. 추출액의 온도를 섭씨 95도로 15분간 가열한 후 곧 바로 급냉시켜 α-아밀라제의 활성을 정지시켰다. 15,000 rpm으로 20분간 원심분리하여 상등액을 분리한 후, 활성탄소 20 g 을 넣고 상온에서 30분간 교반하였다. 상온에서 여과하고 투과물은 농축하여 사용하였다.
<실시예 2>
<2-1> 실험 동물
4주령 ICR계 수컷 생쥐를 분양받아 1주간 적응시킨 후, 실험 동물을 각 5마리씩 실험군을 다음과 같이 분류하였다. 대조군, 운동군, 실험군의 3개의 그룹으로 설정하였으며, 실험군과 운동군은 실시예 2-2 및 실시예 2-3에서 기술한 운동을 매일 실시하였다. 실험군은 속단, 및 백수오 혼합추출물을 3 mg/kg을 1일 1회 총 2주간 경구투여 하였으며, 대조군과 운동군은 대조물질로 증류수를 투여하였다.
<2-2> 강제 수영 실험
플라스틱 수조 (지름 15 cm x 높이 22 cm) 에 섭씨 25도의 물을 채우고 수조에서 수영하게 하였다. 꼬리에는 체중의 5%에 해당하는 추를 매달았으며, 매일 20분씩 2주동안 수영하도록 하였다.
2주 후, 실험 물질 투여의 종료와 함께 강제 수영 실험을 실시하여 운동 능력을 측정, 비교하였다. 생쥐가 탈진할 때까지의 수영시간을 측정하였으며, 생쥐의 코가 수면 아래로 가라앉은 후 5초이상 수면으로 올라오지 못할 때를 탈진으로 판정하였다.
그 결과 도 1에서 보는 바와 같이, 대조물질을 섭취한 운동군은 운동을 하지 않은 대조군에 비하여 수영 시간이 늘어남을 확인하였으나 통계적으로 유의하지는 않았다. 이에 반하여 실험군은 대조군에 비하여 통계적으로 유의하게 운동시간이 늘어남을 확인하였다. 속단, 백수오 혼합추출물 (실험군)은 운동을 수행한 운동군보다도 통계적으로 유의하게 수영 시간이 향상됨을 확인하였다.
이로써 본 발명의 조성물이 운동능력을 향상시키는 효능이 뛰어난 것을 확인하였다.
<2-3> 트레드밀 실험 (treadmill test)
실험군 및 운동군은 처음 1주일 동안은 매일 15 m/min 의 속도로 20분동안 트레드밀 운동하였으며, 두번째 주에는 20 m/min의 속도로 30분동안 매일 트레드밀 운동을 실시하였다. 트레드밀 후방에는 1A 의 약한 전류를 흘려 생쥐가 지속적으로 운동을 할 수 있도록 하였다.
2주 후, 실험 물질 투여의 종료와 함께 최대 운동 능력을 측정하였다. 20 m/min 의 속도로 생쥐가 탈진할 때까지 운동한 시간을 측정하였으며, 트레드밀에서 뛰지 못하고 전극으로 밀려나와 10초 이상 머무르며 트레드밀로 들어가지 못할 때를 탈진으로 판정하였다.
그 결과 도 2에서 보는 바와 같이, 대조물질을 섭취한 운동군은 운동을 하지 않은 대조군에 비하여 달리기 시간이 늘어남을 확인하였으나 통계적으로 유의하지는 않았다. 이에 반하여 실험군은 대조군에 비하여 통계적으로 유의하게 운동시간이 늘어남을 확인하였다. 속단, 백수오 혼합추출물은 동일한 운동을 수행한 운동군 보다도 통계적으로 유의하게 달리기 시간이 향상됨을 확인하였다.
이로써 본 발명의 조성물이 운동능력을 향상시키는 효능이 뛰어난 것을 확인하였다.
<2-4> 비복근 (gastrocnemius) 근육량 측정
2주간의 실험 물질 섭취 기간이 끝나고 생쥐들을 희생시켜 비복근을 분리하여 무게를 측정하였다.
그 결과 도 3에서 보는 바와 같이, 대조물질을 섭취한 운동군은 운동을 하지 않은 대조군에 비하여 비복근의 양이 늘어남을 확인하였으나 통계적으로 유의하지는 않았다. 이에 반하여 실험군은 대조군에 비하여 통계적으로 유의하게 비복근 근육량이 늘어남을 확인하였다. 속단, 백수오 혼합추출물은 운동을 수행한 운동군 보다도 통계적으로 유의하게 비복근량이 향상됨을 확인하였다.
이로써 본 발명의 조성물이 근육 발달을 증진시키는 효능이 뛰어난 것을 확인하였다.

Claims (10)

  1. 속단 추출물 및 백수오 추출물을 유효성분으로 포함하는 운동능력 증진용 식품 조성물.
  2. 제1항에 있어서, 상기 속단 및 백수오 추출물은 물 또는 탄소수 1 내지 6개의 알코올 및 이들의 혼합용매로 이루어진 군에서 선택된 어느 하나의 용매로 추출한 것을 특징으로 하는 식품조성물.
  3. 제1항에 있어서, 상기 속단 추출물 및 백수오 추출물의 혼합비율은 1:1 중량비인 것을 특징으로 하는 식품 조성물.
  4. 속단 추출물 및 백수오 추출물을 유효성분으로 포함하는 근육 발달 증진용 식품 조성물.
  5. 제4항에 있어서, 상기 속단 및 백수오 추출물은 물 또는 탄소수 1 내지 6개의 알코올 및 이들의 혼합용매로 이루어진 군에서 선택된 어느 하나의 용매로 추출한 것을 특징으로 하는 식품조성물.
  6. 속단 추출물 및 백수오 추출물을 유효성분으로 포함하는 운동능력 증진용 약학 조성물.
  7. 제6항에 있어서, 상기 속단 및 백수오 추출물은 물 또는 탄소수 1 내지 6개의 알코올 및 이들의 혼합용매로 이루어진 군에서 선택된 어느 하나의 용매로 추출한 것을 특징으로 하는 약학조성물.
  8. 제6항에 있어서, 상기 속단 추출물 및 백수오 추출물의 혼합비율은 1:1 중량비인 것을 특징으로 하는 약학 조성물.
  9. 속단 추출물 및 백수오 추출물을 유효성분으로 포함하는 근육 발달 증진용 약학 조성물.
  10. 제9항에 있어서, 상기 속단 및 백수오 추출물은 물 또는 탄소수 1 내지 6개의 알코올 및 이들의 혼합용매로 이루어진 군에서 선택된 어느 하나의 용매로 추출한 것을 특징으로 하는 약학조성물.
