본 발명은 (a) 분석대상의 생시료에 상기 응집형-형성 폴리펩타이드의 다이머형을 스파이킹(spiking)하는 단계; (b) 상기 단계 (a)의 결과물을 인큐베이션 시켜 상기 응집형-형성 폴리펩타이드의 응집형을 추가적으로 형성시키는 단계; (c) 상기 단계 (b)의 결과물에 상기 응집형-형성 폴리펩타이드의 응집형에 결합하는 결합제에 시그널발생 표지가 결합된 결합제-표지를 접촉시키는 단계; 및 (d) 상기 응집형-형성 폴리펩타이드의 응집형에 결합된 결합제-표지로부터 발생되는 시그널을 검출하는 단계를 포함하는 생시료(biosample)의 응집형-형성 폴리펩타이드의 응집형을 검출하는 방법에 관한 것이다.

Description

응집형-형성 폴리펩타이드의 응집형을 검출하는 방법
본 특허출원은 2016년 3년 16일에 대한민국 특허청에 제출된 대한민국 특허출원 제10-2016-0031534호에 대하여 우선권을 주장하며, 상기 특허출원의 개시 사항은 본 명세서에 참조로서 삽입된다.
본 발명은 생시료(biosample)의 응집형-형성 폴리펩타이드의 응집형을 검출하는 방법 또는 키트에 관한 것이다.
우선, 단백질을 구성하는 폴리펩타이드는 멀티머를 형성하여 기능적인 단백질을 이루는 경우도 있지만, 정상적인 상태에서는 모노머로 존재하다가 비정상적인 상태(예컨대, 미스폴딩형으로 전환)가 되면 멀티머를 형성하여 응집되어 질병을 유발하는 경우가 많다(Massimo Stefani, et al., J. Mol . Med. 81:678-699(2003); and Radford SE, et al., Cell. 97:291-298(1999)).
예를 들어, 단백질의 비정상적인 응집 또는 미스폴딩과 관련된 질환 또는 질병은 알츠하이머 질환, 크로이츠펠트-야콥병, 스폰지폼 뇌질환(Spongiform encephalopathies), 파킨슨 질환, 헌팅톤 질환, 근위축성 측삭경화증(Amyotrophic lateral sclerosis), 서핀 결핍증(Serpin deficiency), 폐기종(emphysema), 경변증(cirrhosis), 제 II형 당뇨병, 일차 전신성 아밀로이드증, 이차 전신성 아밀로이드증, 전두측두엽성 치매(Fronto-temporal dementias), 노인 전신성 아밀로이드증, 가족성 아밀로이드 다발신경병(familial amyloid polyneuropathy), 유전성 대뇌 아밀로이드 맥관병(hereditary cerebral amyloid angiopathy) 및 혈투석-관련 아밀로이드증을 포함 한다.
이러한 질환 또는 질병의 유무 또는 진행 정도를 측정하는데 있어, 항원의 양이 시료 속에 매우 적거나 항원의 크기가 매우 작아서 측정하기 어렵거나, 혹은 몸 속의 항원의 양과 시료 속의 항원의 양이 비례하지 않는 경우에, 예컨대, 알츠하이머 질환에 관여하는 Aβ(아밀로이드-베타)도 정상인에 비해 비정상인에서 Aβ 올리고머 레벨이 높은 것으로 알려져 있지만, 혈액시료 내 Aβ 올리고머 양의 검출이 어렵거나 혈액 시료 내에 비정형적으로 Aβ 올리고머가 존재할 때 진단이 어려울 수 있다.
또한, 측정하고자 하는 항원이 너무 작거나 양이 적어서 sandwich ELISA를 통한 질환의 진단이 쉽지 않는 경우가 있다.
이에, 본 발명자들은 환자와 정상인 간의 진단 시그널의 차이(differentiation)를 극대화한 응집형-형성 폴리펩타이드의 응집형 검출 방법의 개발 필요성을 인식하였다.
본 명세서 전체에 걸쳐 다수의 논문 및 특허문헌이 참조되고 그 인용이 표시되어 있다. 인용된 논문 및 특허문헌의 개시 내용은 그 전체로서 본 명세서에 참조로 삽입되어 본 발명이 속하는 기술 분야의 수준 및 본 발명의 내용이 보다 명확하게 설명된다.
상기 배경 하에서, 본 발명자들은 응집형-형성 폴리펩타이드의 응집형을 검출하는 신규 방법을 개발하기 위하여 폭넓은 연구를 해왔고, 그 결과, 폴리펩타이드의 응집형 형성을 억제하는 방어 시스템(clearing system)의 차이를 이용하여 환자와 정상인 간의 진단 시그널의 차이(differentiation)를 극대화한 응집형-형성 폴리펩타이드의 응집형 검출 방법을 개발하였다.
따라서, 본 발명의 목적은 생시료(biosample)의 응집형-형성 폴리펩타이드의 응집형을 검출하는 방법을 제공하는 데 있다.
본 발명의 다른 목적은 생시료(biosample)의 응집형-형성 폴리펩타이드의 응집형을 검출하기 위한 키트를 제공하는 데 있다.
본 발명의 다른 목적 및 이점은 하기의 발명의 상세한 설명, 청구범위 및 도면에 의해 보다 명확하게 된다.
본 발명의 일 양태에 따르면, 본 발명은 다음의 단계를 포함하는 생시료(biosample)의 응집형-형성 폴리펩타이드의 응집형을 검출하는 방법을 제공한다: (a) 분석대상의 생시료에 상기 응집형-형성 폴리펩타이드의 다이머형을 스파이킹(spiking)하는 단계; (b) 상기 단계 (a)의 결과물을 인큐베이션 시켜 상기 응집형-형성 폴리펩타이드의 응집형을 추가적으로 형성시키는 단계; (c) 상기 단계 (b)의 결과물에 상기 응집형-형성 폴리펩타이드의 응집형에 결합하는 결합제에 시그널발생 표지가 결합된 결합제-표지를 접촉시키는 단계; 및 (d) 상기 응집형-형성 폴리펩타이드의 응집형에 결합된 결합제-표지로부터 발생되는 시그널을 검출하는 단계, 여기서, 상기 단계 (b)의 인큐베이션은 상기 스파이킹한 응집형-형성 폴리펩타이드의 다이머형이 상기 생시료에 의해 멀티머화 할 수 있는 충분한 시간 동안 실시한다.
본 발명은 폴리펩타이드의 응집형 형성을 억제하는 방어 시스템(clearing system)의 차이를 이용하여 환자와 정상인 간의 진단 시그널의 차이(differentiation)를 극대화한 응집형-형성 폴리펩타이드의 응집형 검출 방법에 관한 것이다.
본 명세서에서 용어 “응집형-형성 폴리펩타이드”는 멀티머형(올리고머형)을 형성할 수 있거나 모노머와의 소수성 상호작용에 의한 응집형을 형성할 수 있는 폴리펩타이드를 의미한다. 특히, 하기 구조적 변화는 다양한 질환을 유발한다. 예컨대, 알츠하이머 질환, 크로이츠펠트-야콥병, 스폰지폼 뇌질환(Spongiform encephalopathies), 파킨슨 질환, 헌팅톤 질환, 근위축성 측삭경화증(Amyotrophic lateral sclerosis), 서핀 결핍증(Serpin deficiency), 폐기종(emphysema), 경변증(cirrhosis), 제 II형 당뇨병, 일차 전신성 아밀로이드증, 이차 전신성 아밀로이드증, 전두측두엽성 치매(Fronto-temporal dementias), 노인 전신성 아밀로이드증, 가족성 아밀로이드 다발신경병(familial amyloid polyneuropathy), 유전성 대뇌 아밀로이드 맥관병(hereditary cerebral amyloid angiopathy) 및 혈투석-관련 아밀로이드증을 포함한다.
일반적으로, 상기 응집형-형성 폴리펩타이드의 비-응집형은 정상이고 응집형은 질환, 특히 알츠하이머 질환, 크로이츠펠트-야콥병 또는 파킨슨 질환과 같은 신경 퇴행성 질환을 유발한다.
본 발명의 일 구현예에 따르면, 상기 스파이킹한 응집형-형성 폴리펩타이드의 다이머형의 멀티머화를 시키는 상기 생시료는 상기 응집형-형성 폴리펩타이드의 멀티머형이 관여된 질환을 갖는 인간의 생시료이고, 보다 바람직하게는 상기 생시료에 의해 멀티머화 할 수 있는 충분한 인큐베이션 시간은, 응집형-형성 폴리펩타이드의 멀티머형이 관여된 질환을 갖는 인간의 생시료를 이용하여 발생된 시그널이 정상의 인간의 생시료를 이용하여 발생된 시그널보다 1.3-20배 크게 하는데 충분한 시간이다.
이하, 생시료(biosample)의 응집형-형성 폴리펩타이드의 응집형을 검출하기 위한 본 발명의 방법을 단계별로 상세하게 설명하면 다음과 같다:
(a) 스파이킹(spiking)하는 단계
우선, 본 발명의 방법은 분석대상의 생시료에 상기 응집형-형성 폴리펩타이드의 다이머형을 스파이킹(spiking)하는 단계를 포함한다.
본 명세서에서, 용어 “생시료(biosample)”는 분석하고자 하는 유기체-유래 시료를 의미한다. 상기 생시료는 생물원의 세포, 조직, 또는 생체액, 또는 본 발명에 따라 분석될 수 있는 다른 미디엄(medium)을 의미하고, 이는 인간으로부터 채취한 시료, 동물로부터 채취한 시료, 인간 또는 동물을 위한 식품으로부터 채취한 시료를 포함한다. 바람직하게는, 분석 대상의 생시료는 혈액, 혈청, 혈장, 림프액, 우유, 소변, 대변, 눈물, 타액, 정액, 뇌 추출물(예컨대, 뇌 균질액), 척수액(SCF), 충수, 비장 및 편도 조직 추출물을 포함하는 체내 유체 시료이다. 보다 바람직하게는, 상기 생시료는 혈액이고, 가장 바람직하게는, 혈장이다.
본 발명의 다른 구현예에 따르면, 상기 응집형-형성 폴리펩타이드는 알츠하이머 질환에 관여하는 Aβ 펩타이드와 tau 단백질, 크로이츠펠트-야콥병 및 스폰지폼 뇌질환에 관여하는 프라이온, 파킨슨 질환에 관여하는 α-시누클레인, 일차 전신성 아밀로이드증에 관여하는 Ig 경사슬, 이차 전신성 아밀로이드증에 관여하는 혈청 아밀로이드 A, 전두측두엽성 치매에 관여하는 tau 단백질, 노인 전신성 아밀로이드증에 관여하는 트랜스티레틴, 가족성 아밀로이드 다발신경병에 관여하는 트랜스티레틴, 유전성 대뇌 아밀로이드 맥관병에 관여하는 시스타틴 C , 혈투석-관련 아밀로이드증에 관여하는 β2-마이크로글로블린, 헌팅톤 병에 관여하는 헌팅틴, 근위축성 측삭 경화증에 관여하는 슈퍼옥사이드 디스뮤타아제, 서핀 결핍증, 폐기종, 및 경변증에 관여하는 서핀, 및 제 II형 당뇨병에 관여하는 아밀린을 포함한다. 보다 바람직하게는, 상기 응집형-형성 폴리펩타이드는 알츠하이머 질환에 관여하는 Aβ 펩타이드 또는 tau 단백질, 또는 파킨슨 질환에 관여하는 α-시누클레인이고, 가장 바람직하게는 Aβ 펩타이드이다.
본 명세서에서 용어 “스파이킹(spiking)”은 분석대상의 생시료에 응집형-형성 폴리펩타이드의 다이머형을 첨가 또는 첨가 후 혼합하는 과정을 의미한다.
본 명세서에서 용어 “멀티머”에는 올리고머도 포함된다.
본 명세서에서 용어 “다이머”는 두 개의 모노머가 결합되어 형성되는 것이다.
본 발명에 따르면, 분석대상의 생시료에 상기 응집형-형성 폴리펩타이드의 다이머형을 스파이킹(spiking)하는 경우는, 응집형-형성 폴리펩타이드의 응집형 형성을 억제시키는 방어시스템(clearing system) 차이를 이용하여 환자와 정상인 간의 진단 시그널의 차이(differentiation)를 극대화 하고자, 즉 환자의 생시료는 방어시스템(clearing system)의 정도가 낮아 응집형-형성 폴리펩타이드의 응집형 형성이 촉진되는데 반해 정상인의 생시료에서는 방어시스템(clearing system)의 정도가 높아 응집형-형성 폴리펩타이드의 응집형 형성이 감소되어 진단 시그널의 차이(differentiation)가 극대화 된다.
본 발명의 또 다른 구현예에 따르면, 상기 응집형-형성 폴리펩타이드의 다이머형은 상기 응집형-형성 폴리펩타이드의 모노머형인 서열목록 제 1 서열의 아미노산 서열로 이루어진 Aβ 펩타이드 두 개가, 상기 서열목록 제 1 서열의 아미노산 서열로 이루어진 Aβ 펩타이드의 26번 Cys 잔기에 의해서 이황화 결합되어 이루어진다.
본 발명의 다른 구현예에 따르면, 상기 단계 (a)의 결과물에 완충액을 추가적으로 첨가한다. 보다 바람직하게는, 상기 완충액은 생시료에 대하여 3-15 배(v/v)로 첨가되고, 보다 더 바람직하게는 5-13 배(v/v)로 첨가되며, 보다 더욱 더 바람직하게는 7-11 배(v/v)로 첨가되고, 보다 더 더욱 더 바람직하게는 8-10 배(v/v)로 첨가된다.
본 발명에 이용되는 완충액은 당업계에 공지된 다양한 완충액을 이용할 수 있으나, 바람직하게는 상기 완충액은 비이온성 계면활성제-함유 인산염 완충액이다.
본 발명에 이용되는 인산염 완충액에 함유되는 비이온성 계면활성제는 당업계에 공지된 다양한 비이온성 계면활성제를 이용할 수 있고, 바람직하게는 알콕시레이티드알킬에테르, 알콕시레이티드알킬에스테르, 알킬폴리글리코사이드, 폴리글리세릴에스테르, 폴리솔베이트류 및 슈가에스테르를 포함한다. 보다 바람직하게는 Tween-20 또는 Triton X-100이 사용되며, 가장 바람직하게는 Tween-20이 사용된다.
(b) 응집형-형성 폴리펩타이드의 응집형을 추가적으로 형성시키는 단계
그 다음, 본 발명의 방법은 (b) 상기 단계 (a)의 결과물을 인큐베이션 시켜 상기 응집형-형성 폴리펩타이드의 응집형을 추가적으로 형성시키는 단계를 포함한다.
본 발명의 가장 큰 특징 중 하나는 생시료 속에 측정하고자 하는 응집형-형성 폴리펩타이드의 응집형(항원)의 양이 매우 적거나 응집형-형성 폴리펩타이드의 응집형(항원)의 크기가 매우 작아서 측정하기 어렵거나, 또는 인체 내 응집형-형성 폴리펩타이드의 응집형(항원)의 양과 생시료 속의 응집형-형성 폴리펩타이드의 응집형(항원)의 양이 비례하지 않은 경우에, 상술한 상기 응집형-형성 폴리펩타이드의 다이머형을 생시료에 스파이킹(spiking)하여 응집형-형성 폴리펩타이드의 응집형을 추가적으로 형성시킴으로써, 질병 또는 질환의 유무 또는 진행 정도를 측정할 수 있다는 것이다.
본 발명의 다른 구현예에 따르면, 상기 단계 (b)의 응집형-형성 폴리펩타이드의 응집형의 추가적인 형성은 상기 단계 (a)의 결과물을 온도 1-50℃에서 인큐베이션(incubation)시켜 실시하고, 보다 바람직하게는 온도 35-50℃에서 인큐베이션(incubation)시켜 실시하며, 보다 더 바람직하게는 온도 35-45℃에서 실시하며, 보다 더욱 더 바람직하게는 온도 35-40℃에서 실시한다.
본 발명에서, 상기 단계 (b)의 인큐베이션은 상기 스파이킹한 응집형-형성 폴리펩타이드의 다이머형이 상기 생시료에 의해 멀티머화 할 수 있는 충분한 시간 동안 실시하고, 보다 바람직하게는 상기 생시료에 의해 멀티머화 할 수 있는 충분한 인큐베이션 시간은, 응집형-형성 폴리펩타이드의 멀티머형이 관여된 질환을 갖는 인간의 생시료를 이용하여 발생된 시그널이 정상의 인간의 생시료를 이용하여 발생된 시그널보다 1.3-20배 크게 하는데 충분한 시간이다.
본 발명의 또 다른 구현예에 따르면, 인간의 생시료를 이용하여 발생된 시그널이 정상의 인간의 생시료를 이용하여 발생된 시그널보다 1.3-20배 크게 하는데 충분한 시간동안 실시하기 위한, 상기 단계 (b)의 응집형-형성 폴리펩타이드의 응집형의 추가적인 형성은 상기 단계 (a)의 결과물을 1시간 내지 12일 동안 인큐베이션(incubation)시켜 실시하고, 보다 바람직하게는 1일 내지 10일 동안 실시하며, 보다 더 바람직하게는 1일 내지 8일 동안 실시하고, 보다 더욱 더 바람직하게는 1일 내지 6일 동안 실시하며, 보다 더 더욱 더 바람직하게는 2일 내지 6일 동안 실시하고, 가장 바람직하게는 2일 내지 5일 동안 실시한다.
본 명세서에서 용어 “인큐베이션”은 분석대상의 생시료를 소정의 온도에서 일정기간 동안 스탠딩 하는 것(kept to stand) 또는 쉐이킹(shaking) 하는 것을 의미하고, 쉐이킹 하는 경우에는 바람직하게는 마일드 쉐이킹(mild shaking) 하는 것을 의미한다.
본 발명의 가장 큰 특징 중 다른 하나는 생시료를 소정의 온도에서 일정 기간 스탠딩(즉, 인큐베이션) 함으로써, 생시료에 존재하는 스파이킹 된 응집형-형성 폴리펩타이드의 다이머형과 응집형-형성 폴리펩타이드가 서로 잘 응집(aggregation)되도록 하여 환자와 정상인 간의 진단 시그널의 차이(differentiation)를 극대화하였다는 점이다.
(c) 상기 단계 (b)의 결과물에 상기 응집형-형성 폴리펩타이드의 응집형에 결합하는 결합제-표지의 접촉
그리고, 본 발명의 방법은 (c) 상기 단계 (b)의 결과물에 상기 응집형-형성 폴리펩타이드의 응집형에 결합하는 결합제에 시그널발생 표지가 결합된 결합제-표지를 접촉시키는 단계를 거친다.
본 발명에서 상기 응집형-형성 폴리펩타이드의 응집형에 결합하는 결합제는 항체, 펩타이드 앱타머, 어드넥틴(AdNectin), 어피바디(affibody, 미국 특허 제5,831,012호), 아비머(Avimer, Silverman, J. et al, Nature Biotechnology 23(12):1556(2005)) 또는 쿠니쯔 도메인(Kunitz domain, Arnoux B et al., Acta Crystallogr. D Biol . Crystallogr . 58(Pt 7):12524(2002)), 및 Nixon, AE, Current opinion in drug discovery & development 9(2):2618(2006))이다.
본 발명에서 상기 응집형-형성 폴리펩타이드의 응집형에 결합하는 결합제에 결합되는 시그널발생 표지는 화합물 표지(예컨대, 바이오틴), 효소 표지(예컨대, 알카린 포스파타아제, 페록시다아제, β-갈락토시다아제 및 β-글루코시다아제), 방사능 표지(예컨대, I125 및 C14), 형광 표지(예컨대, 플루오레세인), 발광 표지, 화학발광 표지 및 FRET(형광 공명 에너지 전달; fluorescence resonance energy transfer) 표지를 포함하나, 이에 한정되는 것은 아니다.
(d) 상기 응집형-형성 폴리펩타이드의 응집형에 결합된 결합제-표지로부터 발생되는 시그널의 검출
마지막으로, 본 발명의 방법은 (d) 상기 응집형-형성 폴리펩타이드의 응집형에 결합된 결합제-표지로부터 발생되는 시그널을 검출하는 단계를 포함한다.
상기 응집형-형성 폴리펩타이드의 응집형에 결합된 결합제-표지로부터 발생되는 시그널의 검출은 당업계에 공지된 다양한 방법으로 실시할 수 있으며, 예컨대 항원-항체 반응과 관련된 면역분석법을 이용하여 실시할 수 있다.
본 발명의 또 다른 구현예에 따르면, 상기 단계 (c) 및 (d)는 다음의 단계를 포함하는 방법으로 실시된다: (c-1) 상기 응집형을 포획(capturing)하는 상기 응집형-형성 폴리펩타이드상의 에피토프를 인식하는 포획 항체에 상기 단계 (b)의 결과물을 접촉시키는 단계; (c-2) 상기 응집형-형성 폴리펩타이드상의 에피토프를 인식하는 검출 항체에 상기 포획된 응집형을 접촉시키는 단계; 및 (c-3) 응집형-검출 항체 복합체를 검출하는 단계.
이러한 검출 방법은 두 가지 형태의 항체, 즉 포획 항체 및 검출 항체를 이용한다. 본 명세서에서 용어, “포획 항체”는 생시료에서 검출하고자 하는 응집형-형성 폴리펩타이드에 결합 할 수 있는 항체를 의미한다. 용어, “검출 항체”는 상기 포획 항체에 의해 포획된 응집형-형성 폴리펩타이드에 결합할 수 있는 항체를 의미한다. “항체”는 항원에 결합할 수 있는 면역글로불린 단백질을 의미한다. 본 명세서에 이용된 항체는 검출 하고자 하는 에피토프, 항원 또는 항원 단편에 결합할 수 있는 전체 항체뿐만 아니라 항체 단편(예컨대, F(ab’)2, Fab’, Fab, Fv)을 포함한다.
상기 검출 방법은 응집형-형성 폴리펩타이드 상의 에피토프를 특이적으로 인식하는 한 세트의 포획 항체 및 검출 항체를 이용하는 것으로서, 상기 포획 항체 및 검출 항체가 특이적으로 인식하는 상기 에피토프는 서로 동일하거나 오버래핑 되어 있다.
포획 항체 및 검출 항체에 대한 에피토프를 언급하면서 사용되는 용어, “오버래핑(overlapped with)”은 완전히 또는 부분적으로 오버래핑된 아미노산 서열을 포함하는 에피토프를 포괄한다. 예를 들어, 6E10, FF51 및 WO2 항체에 대한 에피토프는 인간 Aβ 펩타이드 서열의 각각, 아미노산 3-8, 1-4 및 4-10으로 이루어진 아미노산 서열을 가진다. 이러한 에피토프는 완전히 오버래핑된 에피토프로 설명될 수 있다.
본 발명의 다른 구현예에 따르면, 인간 Aβ 펩타이드 서열을 언급하면서 표현하는 경우, 상기 에피토프는 아미노산 3-8, 1-4 또는 4-10으로 이루어진 아미노산 서열을 가진다.
본 발명의 또 다른 구현예에 따르면, 상기 포획 항체가 인식하는 에피토프는 상기 응집형-형성 폴리펩타이드에서 반복되지 않는 서열이고, 상기 검출 항체가 인식하는 에피토프는 상기 응집형-형성 폴리펩타이드에서 반복되지 않는 서열이다. 본 발명의 검출 방법에 따르면, 포획 항체에 결합된 응집형-형성 폴리펩타이드는 검출 항체와 더 이상 결합 할 수 없고, 이는 검출 항체가 인식하는 추가적인 에피토프가 존재하지 않기 때문이다.
본 발명의 다른 구현예에 따르면, 상기 포획 항체 및 검출 항체는 서로 동일하다. 즉, 포획 항체 및 검출 항체에 특이적으로 결합되는 에피토프는 동일한 것이 바람직하다.
본 발명의 또 다른 구현예에 따르면, 상기 포획 항체는 고체 기질에 결합된다. 이러한 형태의 공지의 물질은 폴리스티렌 및 폴리프로필렌, 유리, 금속, 및 젤과 같은 탄화수소 중합체를 포함한다. 상기 고체 기질은 딥스틱, 마이크로티터 플레이트, 입자(예컨대, 비드), 친화성 컬럼 및 이뮤노블롯 멤브레인(예컨대, 폴리비닐리덴 플로라이드 멤브레인) 형태로 있을 수 있다(참조: 미국 특허 제5,143,825호, 제5,374,530호, 제4,908,305호 및 제5,498,551호).

본 발명에서, 응집형-형성 폴리펩타이드에 결합할 수 있는 항체를 융합 방법(Kohler and Milstein, European Journal of Immunology, 6:511-519(1976)), 재조합 DNA 방법(미국 특허 제4,816,56호) 또는 파지 항체 도서관(Clackson et al, Nature, 352:624-628(1991); 및 Marks et al, J. Mol . Biol., 222:58, 1-597(1991))과 같은 종래 기술에 따라 면역원으로 종전에 기재된 에피토프를 이용하여 준비할 수 있다. 상기 항체 제조를 위한 일반적인 방법은 Harlow, E. and Lane, D., Antibodies: A Laboratory Manual, Cold Spring Harbor Press, New York, 1988; Zola, H., Monoclonal Antibodies: A Manual of Techniques, CRC Press, Inc., Boca Raton, Florida, 1984; 및 Coligan, CURRENT PROTOCOLS IN IMMUNOLOGY, Wiley/Greene, NY, 1991에 기재되어 있다.
단일클론 항체 제조를 위한 하이브리도마 세포주의 준비는 불멸 세포주(immortal cell line) 및 항체를 생산하는 임파구의 융합으로 실시된다. 상기 단일클론 항체의 제조는 당업계에 공지된 기술을 이용하여 실시할 수 있다. 다클론 항체는, 상술한 항원을 적합한 동물에 주입하고, 항체를 포함하는 항혈청을 수집한 다음, 공지된 친화성 기술에 의해 항체를 분리하는 방법에 따라 제조될 수 있다.
응집형-검출 항체 복합체의 검출은 당업계의 공지된 다양한 방법으로 실시할 수 있다. 응집형-검출 항체 복합체의 형성은 생시료에 응집형의 존재를 나타낸다. 상기 단계는 종래의 방법에 따라, 예컨대, Enzyme Immunoassay, E. T. Maggio, ed., CRC Press, Boca Raton, Florida, 1980 및 Harlow and Lane, eds. Antibodies: A Laboratory Manual(1988) Cold Spring Harbor Laboratory Press, Cold Spring Harbor, N.Y에 기재된 바와 같이 다양한 검출할 수 있는 표지/기질 쌍을 이용하여, 정량적으로 또는 정성적으로 실시 할 수 있다.
상기 검출 항체를 알카린 포스파타아제로 표지하는 경우에는, 브로모클로로인돌일 포스페이트(BCIP), 니트로 블루 테트라졸리움(NBT) 및 ECF를 발색 반응을 위한 기질로서 이용할 수 있다; 호스 래디쉬 페록시다아제로 표지하는 경우, 기질로서 클로로나프톨, 아미노에틸카바졸, 디아미노벤지딘, D-루시페린, 루시게닌 (비스-N-메틸아크리딘이움 나이트레이트), 레소루핀 벤질 에테르, 루미놀, 암플렉스 레드 시약(10-아세틸-3,7-디하이드록시페녹사진), TMB(3,3,5,5-테트라메틸벤즈이딘), ECL(enhanced chemiluminescence) 및 ABTS(2,2-아진-디[3-에틸벤즈티아졸린 술포네이트]) 등이 이용된다.
이러한 방법으로 응집형-형성 폴리펩타이드의 멀티머형이 관여된 질환을 갖는 인간의 생시료를 이용하여 발생된 시그널이 정상의 인간의 생시료를 이용하여 발생된 시그널보다 1.3-20배 크게 할 수 있고, 보다 바람직하게는 1.5-10배, 보다 더 바람직하게는 1.6배-10배 크게 할 수 있다.
본 발명의 다른 양태에 따르면, 본 발명은 응집형-형성 폴리펩타이드의 다이머형을 포함하는 생시료(biosample)의 응집형-형성 폴리펩타이드의 응집형을 검출하기 위한 키트를 제공한다.
본 발명의 키트는 상술한 본 발명의 생시료(biosample)의 응집형-형성 폴리펩타이드의 응집형을 검출하는 방법을 이용하는 것으로서, 이 둘 사이에 공통된 내용은 반복 기재에 따른 명세서의 과도한 복잡성을 피하기 위하여, 그 기재를 생략한다.
본 발명의 다른 구현예에 따르면, 상기 키트는 응집형-형성 폴리펩타이드 상의 에피토프를 인식하는 포획 항체; 및 상기 포획 항체가 인식하는 상기 에피토프를 인식하는 검출항체를 추가적으로 포함한다.
본 발명의 특징 및 이점을 요약하면 다음과 같다:
(a) 본 발명은 생시료(biosample)의 응집형-형성 폴리펩타이드의 응집형을 검출하는 방법 또는 키트를 제공한다.
(b) 본 발명의 방법은 생시료 속에 측정하고자 하는 응집형-형성 폴리펩타이드의 응집형(항원)의 양이 매우 적거나 응집형-형성 폴리펩타이드의 응집형(항원)의 크기가 매우 작아서 측정하기 어렵거나, 또는 인체 내 응집형-형성 폴리펩타이드의 응집형(항원)의 양과 생시료 속의 응집형-형성 폴리펩타이드의 응집형(항원)의 양이 비례하지 않은 경우에, 폴리펩타이드의 응집형 형성을 억제하는 방어 시스템(clearing system)의 차이를 이용하여 환자와 정상인 간의 진단 시그널의 차이(differentiation)를 극대화 하였다.
(c) 본 발명은 편리하고 신속한 방식으로 실시할 수 있으며, 이는 생시료(biosample)의 응집형-형성 폴리펩타이드의 응집형을 검출하는 방법의 자동화를 가능하게 한다.
도 1 및 도 2는 본 발명의 실시예에 따라, s26C-Beta-Amyloid (1-40) Dimer를 처리한 후 3일과 4일 동안 인큐베이션된 샘플에서 MDS(multimer detection system)(6E10/FF51HRP set)을 이용한 Aβ 올리고머(oligomer) 검출 결과를 보여준다.
도 3는 본 발명의 실시예에 따라, S26C-Beta-Amyloid (1-40) Dimer를 처리한 후 0일, 1일, 2일, 3일, 4일, 5일 동안 인큐베이션된 샘플에서 MDS(multimer detection system)(6E10/FF51HRP set)을 이용한 Aβ 올리고머(oligomer) 검출 결과를 보여준다.
도 4은 본 발명의 실시예에 따라, S26C-Beta-Amyloid (1-40) Dimer 0.25 ng을 처리 후 0일, 5일 인큐베이션한 샘플과 무처리 한 후 0일, 5일 동안 인큐베이션된 샘플에서 MDS(multimer detection system)(6E10/FF51HRP set)을 이용한 Aβ 올리고머(oligomer) 검출 결과를 나타낸다.
도 5 및 6는 본 발명의 실시예에 따라, S26C-Beta-Amyloid (1-40) Dimer를 처리한 후 1일과 2일 동안 인큐베이션된 샘플에서 MDS(multimer detection system)(6E10/WO2HRP set)을 이용한 Aβ 올리고머(oligomer) 검출 결과를 나타낸다.
도 7는 본 발명의 실시예에 따라, S26C-Beta-Amyloid (1-40) Dimer를 처리 후 1일, 2일 3일 4일 동안 인큐베이션된 샘플에서 MDS(multimer detection system)(6E10/WO2HRP set)을 이용한 Aβ 올리고머(oligomer) 검출 결과를 보여준다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예
실시예 1: 실험재료의 준비
코팅버퍼(Carbonate-Bicarbonate Buffer), PBST, TBST 및 PBS는 sigma 사로부터 구입하였다. Block Ace는 Bio-rad 사로부터 구입하였다. 버퍼 A는 TBST에 Block Ace를 0.4%로 희석하여 제조하였다. 블록킹(blocking) 버퍼는 D.W에 1% Block Ace가 희석되도록 제조하였다. HBR1은 Scantibodies Laboratory 사에서 구입하였다. 6E10 항체는 Biolegend 사로부터 구입하였다. WO2-HRP 항체는 Absolute Antibody사로부터 구입하였다. FF51-HRP는 ㈜ The H lab에서 구입하였다. WO2-HRP 항체는 Absolute Antibody사로부터 구입하였다. 재조합(recombinant) Aβ1-42는 Biolegend 사로부터 구입하였다. 재조합 S26C-Beta-Amyloid (1-40) Dimer 는 JPT사에서 구입하였다. 혈장(plasma) 샘플은 분당 서울대병원 및 중앙대 병원에서 제공받았다. ECL 용액은 Rockland 사로부터 구입하였다. 플레이트는 Nunc 사로부터 구입하였다. 6E10, FF51 및 WO2항체에 대한 에피토프는 인간 Aβ 펩타이드 서열의 각각, 아미노산 3-8, 1-4 및 4-10으로 이루어진 아미노산 서열을 가진다. S26C-Beta-Amyloid (1-40) Dimer의 시퀀스는 DAEFRHDSGYEVHHQKLVFFAEDVGCNKGAIIGLMVGGVV이며 각 단량체의 26번 Cystein 잔기에 의해서 이황화 결합한 Dimer 형태를 이루고 있다.
실시예 2: 6E10 플레이트 제조
6E10 항체(항 Aβ 단백질, Biolegend) 30 μg을 코팅 버퍼(sigma) 10 ml에 희석하고 플레이트(Nunc)에 100 μl를 각 웰에 분주 후 4℃ 냉장고에 하루 동안 반응 시켰다. 상기 플레이트를 PBS에 3번 세척하고 D.W에 1% Block Ace가 녹여진 블록킹 버퍼 240 μl를 분주 후 실온에서 2시간 반응 시켰다. 상기 플레이트는 PBS로 3번 세척하고 실온에 30분간 건조 시킨 뒤 사용하였다.
실시예 3: 대조군 준비
양성 대조군(positive control)은 재조합 Aβ1-42(rec. Aβ)(1 μg/ml) 10 μl에 PBST 990 μl를 첨가하여 100 μl를 사용하였다. 음성 대조군(negative control)은 PBS 100 μl를 사용하였다.
실시예 4: 샘플 준비
샘플은 2 샘플을 기준으로 하여 준비하였다. 얼려진 혈장 샘플을 37℃ 히트 블록(heat block)에 15분간 녹인 후 30초간 볼텍싱(vortexing) 후 사용 하였다. S26C-Beta-Amyloid (1-40) Dimer 0.25 ng이 스파이킹(spiking)된 샘플은, 혈장 20 μl에 HBR1(0.123 mg/ml) 8.08 μl와 PBST 180 μl, S26C-Beta-Amyloid (1-40) Dimer(0.25 ng/10 μl) 20 μl를 혼합하여 전체 228.08 μl가 되도록 준비하였다. 또한, 재조합 펩타이드를 스파이킹하지 않은 샘플은 혈장 20 μl에 HBR1(0.123 mg/ml) 8.08 μl와 PBST 200 μl를 혼합하여 전체 228.08 μl가 되도록 준비하였다.
실시예 5: 인큐베이션(incubation)
상기 실시예 4에서 S26C-Beta-Amyloid (1-40) Dimer가 처리되어 준비된 샘플은 37℃ 인큐베이터에서 각 0일, 1일, 2일, 3일, 4일, 5일간 인큐베이션 시켰다(6E10/FF51HRP set). 또한 상기 실시예 4에서 S26C-Beta-Amyloid (1-40) Dimer가 무 처리되어 준비된 샘플은 37℃ 인큐베이터에서 0일, 5일간 인큐베이션 시켰다(6E10/FF51HRP set). 그리고, 상기 실시예 4에서 S26C-Beta-Amyloid (1-40) Dimer가 처리되어 준비된 샘플은 37℃ 인큐베이터에서 각 1일, 2일, 3일, 4일간 인큐베이션 시켰다(6E10/WO2HRP set).
실시예 6: 6E10 / FF51HRP set: S26C -Beta- Amyloid (1-40) Dimer를 처리한 후 3일과 4일 동안 인큐베이션된 샘플에서 MDS(multimer detection system)을 이용한 Aβ 올리고머(oligomer) 검출
6E10 코팅 플레이트(3 μg/ml)에 양성 대조군 및, 음성 대조군에 S26C-Beta-Amyloid (1-40) Dimer가 0.25 ng 처리되어 3일과 4일 동안 인큐베이션된 샘플을 각각 100 μl씩 분주 한 후 실온에서 1시간 반응 시켰다. 상기 플레이트를 TBST로 3번 세척하고 FF51-HRP 항체를 버퍼 A에 0.5 μg/ml이 되도록 제조 후 100 ul씩 분주하였다. 상기 플레이트를 TBST로 3번 세척 후 ECL 용액을 100 μl 분주 하였다. ECL과 반응된 플레이트는 루미노미터 기기(perkinelmer)에 삽입하고 발광(luminescent) 시그널을 측정하였다. 그 결과는 아래 도 1, 2와 같다.
도 1 및 2는 S26C-Beta-Amyloid (1-40) Dimer를 첨가 후 인큐베이션 시간에 따라 AD 샘플과 Non AD 샘플간의 시그날의 변화를 보여준다. 3과 4일 인큐베이션된 각 조건에서 AD와 Non AD간에 차이를 보여준다.
도 1 및 2를 통해서 보건대, Non AD 환자 샘플과 비교시 AD 환자의 샘플에서의 Aβ 올리고머의 시그널이 높게 나오는 것은, AD환자 샘플에서 Aβ 올리고머 형성을 억제시키는 방어 시스템이 Non AD환자에서 보다 덜 활성화되는 것으로 판단되어진다.
실시예 7: 6E10 / FF51HRP set: S26C -Beta- Amyloid (1-40) Dimer를 처리한 후 0일, 1일, 2일, 3일, 4일, 5일 동안 인큐베이션된 샘플에서 MDS(multimer detection system)을 이용한 Aβ 올리고머(oligomer) 검출
6E10 코팅 플레이트(3 μg/ml)에 양성 대조군 및, 음성 대조군에 S26C-Beta-Amyloid (1-40) Dimer가 0.25 ng 처리되어 0일, 1일, 2일, 3일, 4일, 5일 동안 인큐베이션된 샘플을 각각 100 μl씩 분주 한 후 실온에서 1시간 반응 시켰다. 상기 플레이트를 TBST로 3번 세척하고 FF51-HRP 항체를 버퍼 A에 0.5 μg/ml이 되도록 제조 후 100 ul씩 분주하였다. 상기 플레이트를 TBST로 3번 세척 후 ECL 용액을 100 μl 분주 하였다. ECL과 반응된 플레이트는 루미노미터 기기(perkinelmer)에 삽입하고 발광(luminescent) 시그널을 측정하였다. 그 결과는 아래 도 3와 같다.
도 3는 S26C-Beta-Amyloid (1-40) Dimer를 첨가 후 인큐베이션 시간에 따라 AD 샘플과 Non AD 샘플간의 시그날의 변화를 확인한 그래프로 인큐베이션 시간이 증가함에 따라 AD샘플의 시그날이 Non AD샘플의 시그날에 비해 1.15배, 1.34배, 1.64배, 2.14배, 3.01배, 3.35배로 증가되는 것을 보여 준다.
도 3을 통해서 보건대, Non AD 환자 샘플과 비교시 AD 환자의 샘플에서의 Aβ 올리고머의 시그널이 높게 나오는 것은, AD환자 샘플에서 Aβ 올리고머 형성을 억제시키는 방어 시스템이 Non AD환자에서 보다 덜 활성화되는 것으로 판단되어진다.
실시예 8: 6E10 / FF51HRP set: S26C -Beta- Amyloid (1-40) Dimer 0.25 ng을 처리 후 0일, 5일 인큐베이션한 샘플과 무처리 한 후 0일, 5일 동안 인큐베이션된 샘플에서 MDS(multimer detection system)을 이용한 Aβ 올리고머(oligomer) 검출
6E10 코팅 플레이트(3 μg/ml)에 양성 대조군 및, 음성 대조군에 S26C-Beta-Amyloid (1-40) Dimer가 0.25 ng 처리된 후 0일, 5일 처리 되지 않은 각 0일, 5일 동안 인큐베이션된 샘플을 각각 100 μl씩 분주 한 후 27℃ incubator에서 정치 상태로 1시간 반응 시켰다. 상기 플레이트를 TBST로 3번 세척하고 FF51-HRP 항체를 버퍼 A에 0.5 μg/ml이 되도록 제조 후 100 ul씩 분주하였다. 상기 플레이트를 TBST로 3번 세척 후 ECL 용액을 100 μl 분주 하였다. ECL과 반응된 플레이트는 루미노미터 기기(perkinelmer)에 삽입하고 발광(luminescent) 시그널을 측정하였다. 그 결과는 아래 도 4과 같다.
도 4은 S26C-Beta-Amyloid (1-40) Dimer를 첨가 하지 않고 0일과 5일, 첨가한 후 0일과 5일간 인큐베이션된 샘플에서 Aβ 올리고머를 측정한 데이터로서, S26C-Beta-Amyloid (1-40) Dimer를 스파이킹 하지 않았을 때와 스파이킹 하였을 때, 시간에 따라 AD 샘플의 시그날이 Non AD 샘플보다 시그날이 증가되는 것을 보여주었다.
S26C-Beta-Amyloid (1-40) Dimer를 스파이킹 하지 않고 0일과 5일간 인큐베이션한 샘플의 경우는 AD의 시그날이 Non AD의 시그널보다 각각 1.09배, 1.97배의 차이를 보여 주었으며 0일에서 5일 간 Aβ oligomer의 변화량은 1.8배 증가 하였다. 반면, S26C-Beta-Amyloid (1-40) Dimer를 0.25 ng 스파이킹한 후 0일과 5일간 인큐베이션한 샘플의 경우는 AD의 시그날이 Non AD의 시그널보다 각각 1.1배, 3.47배의 차이를 보여 주었으며 0일에서 5일 간 Aβ oligomer의 변화량은 3.15배 증가되는 것을 보여 준다.
도 4을 통해서 보건대, Non AD 환자 샘플과 비교시 AD 환자의 샘플에서의 Aβ 올리고머의 시그널이 높게 나오는 것은, AD환자 샘플에서 Aβ 올리고머 형성을 억제시키는 방어 시스템이 Non AD환자에서 보다 덜 활성화되는 것으로 판단되어진다.
실시예 9: 6E10 / WO2HRP set: S26C -Beta- Amyloid (1-40) Dimer를 처리한 후 1일과 2일 동안 인큐베이션된 샘플에서 MDS(multimer detection system)을 이용한 Aβ 올리고머(oligomer) 검출
6E10 코팅 플레이트(3 μg/ml)에 양성 대조군 및, 음성 대조군 에 S26C-Beta-Amyloid (1-40) Dimer가 0.25 ng 처리되어 1일과 2일 동안 인큐베이션된 샘플을 각각 100 μl씩 분주 한 후 실온에서 1시간 반응 시켰다. 상기 플레이트를 TBST로 3번 세척하고 WO2-HRP 항체를 버퍼 A에 0.25 μg/ml이 되도록 제조 후 100 ul씩 분주하였다. 상기 플레이트를 TBST로 3번 세척 후 ECL 용액을 100 μl 분주 하였다. ECL과 반응된 플레이트는 루미노미터 기기(perkinelmer)에 삽입하고 발광(luminescent) 시그널을 측정하였다. 그 결과는 아래 도 5, 6와 같다.
도 5 및 6는 S26C-Beta-Amyloid (1-40) Dimer를 첨가 후 인큐베이션 시간에 따라 AD 샘플과 Non AD 샘플간의 시그날의 변화를 보여준다. 1과 2일 인큐베이션된 각 조건에서 AD와 Non AD간에 차이를 보여준다.
도 5 및 6를 통해서 보건대, Non AD 환자 샘플과 비교시 AD 환자의 샘플에서의 Aβ 올리고머의 시그널이 높게 나오는 것은, AD환자 샘플에서 Aβ 올리고머 형성을 억제시키는 방어 시스템이 Non AD환자에서 보다 덜 활성화되는 것으로 판단되어진다.
실시예 10: 6E10 / WO2HRP set: S26C -Beta- Amyloid (1-40) Dimer를 처리 후 1일, 2일 3일 4일 동안 인큐베이션된 샘플에서 MDS(multimer detection system)을 이용한 Aβ 올리고머(oligomer) 검출
6E10 코팅 플레이트(3 μg/ml)에 양성 대조군 및, 음성 대조군에 S26C-Beta-Amyloid (1-40) Dimer가 0.25 ng 처리되어 1일, 2일, 3일, 4일 동안 인큐베이션된 샘플을 각각 100 μl씩 분주 한 후 실온에서 1시간 반응 시켰다. 상기 플레이트를 TBST로 3번 세척하고 WO2-HRP 항체를 버퍼 A에 0.25 μg/ml이 되도록 제조 후 100 ul씩 분주하였다. 상기 플레이트를 TBST로 3번 세척 후 ECL 용액을 100 μl 분주 하였다. ECL과 반응된 플레이트는 루미노미터 기기(perkinelmer)에 삽입하고 발광(luminescent) 시그널을 측정하였다. 그 결과는 아래 도 7와 같다.
도 7는 S26C-Beta-Amyloid (1-40) Dimer를 첨가 후 인큐베이션 시간에 따라 AD 샘플과 Non AD 샘플간의 시그날의 변화를 확인한 그래프이다. 그 결과로 인큐베이션 1일차에서는 AD샘플의 평균 시그널이 Non AD의 평균 시그날보다 1.19배 높았으며 전체적으로 샘플 시그널이 상승하였다. 반면, 2일차, 3일차, 4일차에서는 전체적으로 높아졌던 샘플의 시그널 값이 떨어지면서 AD샘플이 Non AD샘플 보다 1.69배, 1.50배, 1.41배 차이를 보이는 것을 확인 하였다.
도 7를 통해서 보건대, Non AD 환자 샘플과 비교시 AD 환자의 샘플에서의 Aβ 올리고머의 시그널이 높게 나오는 것은, AD환자 샘플에서 Aβ 올리고머 형성을 억제시키는 방어 시스템이 Non AD환자에서 보다 덜 활성화되는 것으로 판단되어진다.
이상으로 본 발명의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적인 기술은 단지 바람직한 구현예일 뿐이며, 이에 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.

Claims (31)

  1. 다음의 단계를 포함하는 생시료(biosample)의 응집형-형성 폴리펩타이드의 응집형을 검출하는 방법:
    (a) 분석대상의 생시료에 상기 응집형-형성 폴리펩타이드의 다이머형을 스파이킹(spiking)하는 단계;
    (b) 상기 단계 (a)의 결과물을 인큐베이션 시켜 상기 응집형-형성 폴리펩타이드의 응집형을 추가적으로 형성시키는 단계;
    (c) 상기 단계 (b)의 결과물에 상기 응집형-형성 폴리펩타이드의 응집형에 결합하는 결합제에 시그널발생 표지가 결합된 결합제-표지를 접촉시키는 단계; 및
    (d) 상기 응집형-형성 폴리펩타이드의 응집형에 결합된 결합제-표지로부터 발생되는 시그널을 검출하는 단계,
    여기서, 상기 단계 (b)의 인큐베이션은 상기 스파이킹한 응집형-형성 폴리펩타이드의 다이머형이 상기 생시료에 의해 멀티머화 할 수 있는 충분한 시간 동안 실시한다.
  2. 제 1 항에 있어서, 상기 스파이킹한 응집형-형성 폴리펩타이드의 다이머형의 멀티머화를 시키는 상기 생시료는 상기 응집형-형성 폴리펩타이드의 멀티머형이 관여된 질환을 갖는 인간의 생시료인 것을 특징으로 하는 방법.
  3. 제 2 항에 있어서, 상기 생시료에 의해 멀티머화 할 수 있는 충분한 인큐베이션 시간은, 응집형-형성 폴리펩타이드의 멀티머형이 관여된 질환을 갖는 인간의 생시료를 이용하여 발생된 시그널이 정상의 인간의 생시료를 이용하여 발생된 시그널보다 1.3-20배 크게 하는데 충분한 시간인 것을 특징으로 하는 방법.
  4. 제 1 항에 있어서, 상기 생시료(biosample)는 혈액인 것을 특징으로 하는 방법.
  5. 제 4 항에 있어서, 상기 혈액 시료는 혈장(plasma)인 것을 특징으로 하는 방법.
  6. 제 1 항에 있어서, 상기 응집형-형성 폴리펩타이드는 Aβ 펩타이드, tau 단백질, 프라이온, α-시누클레인, Ig 경사슬, 혈청 아밀로이드 A, 트랜스티레틴, 시스타틴 C, β2-마이크로글로블린, 헌팅틴, 슈퍼옥사이드 디스뮤타아제, 서핀 및 아밀린으로 구성된 군으로부터 선택된 것을 특징으로 하는 방법.
  7. 제 6 항에 있어서, 상기 응집형-형성 폴리펩타이드는 Aβ 펩타이드 인 것을 특징으로 하는 방법.
  8. 제 1 항에 있어서, 상기 응집형-형성 폴리펩타이드의 다이머형은 서열목록 제 1 서열의 아미노산 서열로 이루어진 Aβ 펩타이드의 26번 Cys 잔기에 의해서 이황화 결합되어 이루어진 다이머형인 것을 특징으로 하는 방법.
  9. 제 1 항에 있어서, 상기 단계 (a)의 결과물에 완충액을 추가적으로 첨가하는 것을 특징으로 하는 방법.
  10. 제 9 항에 있어서, 상기 완충액은 생시료에 대하여 3-15 배(v/v)로 첨가되는 것을 특징으로 하는 방법.
  11. 제 10 항에 있어서, 상기 완충액은 비이온성 계면활성제-함유 인산염 완충액인 것을 특징으로 하는 방법.
  12. 제 1 항에 있어서, 상기 단계 (b)의 응집형-형성 폴리펩타이드의 응집형의 추가적인 형성은 상기 단계 (a)의 결과물을 온도 1-50℃에서 인큐베이션(incubation)시켜 실시하는 것을 특징으로 하는 방법.
  13. 제 1 항에 있어서, 상기 단계 (b)의 응집형-형성 폴리펩타이드의 응집형의 추가적인 형성은 상기 단계 (a)의 결과물을 1시간 내지 12일 동안 인큐베이션(incubation)시켜 실시하는 것을 특징으로 하는 방법.
  14. 제 1 항에 있어서, 상기 단계 (c) 및 (d)는 다음의 단계를 포함하는 방법으로 실시되는 것을 특징으로 하는 방법:
    (c-1) 상기 응집형을 포획(capturing)하는 상기 응집형-형성 폴리펩타이드상의 에피토프를 인식하는 포획 항체에 상기 단계 (b)의 결과물을 접촉시키는 단계;
    (c-2) 상기 응집형-형성 폴리펩타이드상의 에피토프를 인식하는 검출 항체에 상기 포획된 응집형을 접촉시키는 단계; 및
    (c-3) 응집형-검출 항체 복합체를 검출하는 단계.
  15. 제 14 항에 있어서, 상기 검출 항체는 상기 단계 (c-1)의 에피토프와 동일한 또는 오버래핑된 에피토프를 인식하는 검출 항체인 것을 특징으로 하는 방법.
  16. 제 14 항에 있어서, 상기 포획 항체는 고체 기질에 결합된 것을 특징으로 하는 방법.
  17. 제 14 항에 있어서, 상기 검출 항체는 검출 가능한 신호를 생성시키는 표지를 가지는 것을 특징으로 하는 방법.
  18. 제 17 항에 있어서, 상기 검출 항체에 결합된 표지는 화합물 표지, 효소 표지, 방사능 표지, 형광 표지, 발광 표지, 화학발광 표지 및 FRET 표지인 것을 특징으로 하는 방법.
  19. 응집형-형성 폴리펩타이드의 다이머형을 포함하는 생시료(biosample)의 응집형-형성 폴리펩타이드의 응집형을 검출하기 위한 키트.
  20. 제 19 항에 있어서, 상기 생시료(biosample)는 혈액인 것을 특징으로 하는 키트.
  21. 제 20 항에 있어서, 상기 혈액 시료는 혈장(plasma)인 것을 특징으로 하는 키트.
  22. 제 19 항에 있어서, 상기 응집형-형성 폴리펩타이드는 Aβ 펩타이드, tau 단백질, 프라이온, α-시누클레인, Ig 경사슬, 혈청 아밀로이드 A, 트랜스티레틴, 시스타틴 C, β2-마이크로글로블린, 헌팅틴, 슈퍼옥사이드 디스뮤타아제, 서핀 및 아밀린으로 구성된 군으로부터 선택된 것을 특징으로 하는 키트.
  23. 제 22 항에 있어서, 상기 응집형-형성 폴리펩타이드는 Aβ 펩타이드 인 것을 특징으로 하는 키트.
  24. 제 19 항에 있어서, 상기 응집형-형성 폴리펩타이드의 다이머형은 서열목록 제 1 서열의 아미노산 서열로 이루어진 Aβ 펩타이드의 26번 Cys 잔기에 의해서 이황화 결합되어 이루어진 다이머형인 것을 특징으로 하는 키트.
  25. 제 19 항에 있어서, 상기 키트는 완충액을 추가적으로 포함하는 것을 특징으로 하는 키트.
  26. 제 25 항에 있어서, 상기 완충액은 비이온성 계면활성제-함유 인산염 완충액인 것을 특징으로 하는 키트.
  27. 제 19 항에 있어서, 상기 키트는 응집형-형성 폴리펩타이드 상의 에피토프를 인식하는 포획 항체; 및 상기 포획 항체가 인식하는 상기 에피토프를 인식하는 검출항체를 추가적으로 포함하는 것을 특징으로 하는 키트.
  28. 제 27 항에 있어서, 상기 검출 항체는 상기 포획 항체가 인식하는 상기 에피토프와 동일한 또는 오버래핑된 에피토프를 인식하는 검출항체인 것을 특징으로 하는 키트.
  29. 제 27 항에 있어서, 상기 포획 항체는 고체 기질에 결합된 것을 특징으로 하는 키트.
  30. 제 27 항에 있어서, 상기 검출 항체는 검출 가능한 신호를 생성시키는 표지를 가지는 것을 특징으로 하는 키트.
  31. 제 30 항에 있어서, 상기 검출 항체에 결합된 표지는 화합물 표지, 효소 표지, 방사능 표지, 형광 표지, 발광 표지, 화학발광 표지 및 FRET 표지인 것을 특징으로 하는 키트.
