Abstract
Una molécula mutante de CTLA4 que se enlaza CD80 y/ o CD86, y que comprende un dominio extracelular de CTLA4, de modo que el dominio extracelular, (a) una alanina en la posicion +29 está sustituida con un aminoácido seleccionado del grupo que consiste de tirosina, leucina, triptofano, y treonina, y (b) una leucina en la posicion +104 está situada con un ácido glutámico. Dicha molécula mutante de CTLA4 comprende además una secuencia de aminoácidos que altera la solubilidad, la afinidad o la valencia de la molécula mutante de CTLA4 soluble, en donde la secuencia de aminoácidos puede comprender una region constante de inmunoglobulina humana, y puede comprender además una secuencia de aminoácidos que permite la secrecion de la molécula mutante de CTLA4 soluble. Dicha secuencia de aminoácidos puede comprender un péptido de senal de la oncostatina M. Molécula de ácido nucleico que comprende una secuencia de nucleotidos que codifica para una secuencia de aminoácidos correspondientes a la molécula mutante de CTLA4 soluble. Vector que comprende la secuencia de nucleotidos de la molécula de ácido nucleico y sistema de vector huésped que comprende el vector en una célula huésped adecuada, la cual puede ser una célula eucariotica. Método para producir una proteína mutante de CTLA4 soluble que comprende el desarrollo del sistema vector huésped para producir la proteína mutante de CTLA4 en la célula huésped, y recuperar la proteína así producida, y método para regular la interaccion de una célula T con una célula positiva a CD80 y/o CD86. Uso de la molécula mutante de CTLA4 para preparar un medicamento farmacéutico y dicho medicamento farmacéutico para tratar una enfermedad del sistema inmunologico que comprende un portador farmacéuticamente aceptable y la molécula mutante de CTLA4 soluble.
