<Desc/Clms Page number 2>

"Process for the manufacture of secondary and tertiary aliphato-aromatic amines." -------------------------------------------------- ----------------
It is known that dioxyphenylethylamine, analogously to adrenaline, causes as a result of peripheral excitation of the sympathetic nervous system, a narrowing of the viscera and consequently an increase in blood pressure. On the other hand, this compound does not have analgesic effects, nor does adrenaline or the substituted derivatives of dioxyphenylethylamine resulting from the introduction of an ethyl group mu propyl.
It has surprisingly been found that compounds which contain, bonded to nitrogen, in addition to a dioxyphenylalkyl or -alkylenic group, ue, a phenylalkyl- or alkoyléniq group, ue having the basic formula:
EMI1.1
(OH) -C6H3 R1-N-R2-C6H5
R3 in which R1 and R2 symbolize straight chain or bifurcated alkyl or alkylene radicals and R3 hydrogen or a straight chain alkyl or alkylenic radical, have analgesic properties provided the following requirements are met:
<Desc / Clms Page number 2>
1 Apart from the two hydroxyl groups at the nuals one of the benzene rings is connected, this same ring should not be linked to any other hydroxyl group and the second benzene ring should be coupled to at most one free hydroxyl group.
2 The sum of the carbon atoms contained in R1 and R must be at least 4 in the case of compounds containing only two hydroxyl groups coupled to one of the benzene rings and at least 6 in the case of compounds containing an additional hydroxyl group attached to the other benzene ring,
The position of the two hydroxyl groups coupled to one of the benzene rings is irrelevant, both with respect to the ring flowing with respect to each other. The same is true of the position of the hydroxyl group bonded to the other benzene ring.
The carbon atoms present in R1 and R2 can be distributed indifferently on R1 and R2. It has been found that the analgesic action of the new substances is all the stronger the greater the total number of carbon atoms contained in R1 and R2.
Apart from the two OH groups at one of the benzene rings and possibly another OH group at the other benzene group, the two benzene groups may contain other substitution groups, such as d radicals. Hydrocarbons, for example straight chain or bifurcated alkyl or alkyl radicals, or alkyl or dioxyalkylene groups, which can influence the analgesic effect in many ways over the other actions of these compounds.
According to the invention, the manufacture of the new compound is carried out either by releasing, by a process known per se, the products of substitutions of the hydroxyl groups in compounds which differ from the substances which it is desired to obtain, only by the causes at least one of the hydroxyl groups to be converted into an ether-salt or an ether, either by introducing a phenylalkyl or phenylalkylenic radical into a primary or secondary monophenylalkyl or monophenylalkylenidical amine, the benzene ring of one of these components containing two hydroxyl groups and the benzene ring of the second component possibly containing a hydroxyl group.
The release of hydroxyl groups transformed into an ether or ether-salt can take place in a known manner, for example by heating the starting substances, either with dilute or concentrated mineral acids such as hydrobromic acid or hydriodic acid, or with aluminiup halides, such as @
