<Desc/Clms Page number 3>

IMPROVEMENTS IN THE PREPARATION OF NEW STEROID SUBSTANCES
The present invention relates to improvements in the preparation of new sterol substances: of the following general formula:
EMI1.1
in which R denotes an optionally esterified hydroxy group and X denotes chlorine or bromine.
Although the Applicant does not wish to limit itself by theoretical considerations, it assumes that the substituent occupying position 11 in the [alpha] configuration On the other hand, there is uncertainty as regards the configuration of the 7- group. hydroxy, but the Applicant believes that this group also has the [alpha] configuration, as shown above.
The hydrogen atom in position 5 can have either the configuration or the configuration / 3 depending on the starting material.
The new compounds are valuable for the production of oxygenated sterols in position 11 and, in particular, for research in the
<Desc / Clms Page number 2>
synthesis of cortisone and related substances, in particular for the following reasons:
EMI2.1
It is known that 3 / -aeétcy 7: 11-dikéto-: 9-oxrdoergostene, of the following formula:
EMI2.2
constitutes a substance useful in the synthesis of cortisone and its preparation from 3 -acetoxy-7 E: 11 [alpha] -dihydroxyergosta-8 (9): 22-dene has been described by Heusser et al. (Helv. Chim. Acta, 1951, 34, 2106).
The yield, however, was only 30-35%.
It has now been found that the compounds of the following general formula:
EMI2.3
in which R denotes an optionally esterified hydroxy group and X denotes chlorine or bream, are more suitable for use in the synthesis of cortisone and are more easily isolated, while generally obtainable in better yields than the substances of formula II.
It has further been found that the compounds of formula III can be prepared in a better yield than, if a method analogous to that described by Heusser et al. (Loc. Çit) is applied, using a method in two stages with intermediate formation of the new compounds of formula I. These new compounds therefore offer two advantages, in that the dibrominated compounds 22:23 are more advantageous than the compounds # 22 and constitute useful intermediates in a process of synthesis, which is novel in comparison to previously described methods for obtaining
EMI2.4
tention of 7: ll-diketoz8: 9-okyào-steroids.
The present invention, which relates to the preparation of the new substances of formula I, as well as these substances in themselves, constitutes
<Desc / Clms Page number 3>
identifies the first of these two stages. The second stage of the process is described in Belgian patent application No. 401,157 filed on July 19, 1952.
It was found that the oxidation of the 8: 9 double bond in 3-hydroxy (or esterified hydroxy) -22: 23-dibromo-7: 11 [alpha] -dihydro- xyergost - 8 (9) -ene can be carried out using an organic peracid, such as perbenzoic acid, monoperphthalic acid, peracetic acid or performic acid, preferably using from 1 to 1.5 mol. peracid per mol. of the starting material.
The reaction can be carried out in an inert organic solvent, such as, for example, chloroform, ether, acetic acid, dioxane or benzene, either alone or in a mixture of several such sol- vants. It is not necessary that the starting material be completely soluble in the solvent or mixture of solvents chosen, since because of the greater solubility of epoxides the desired compound goes into solution as the reaction proceeds. Temperatures between -20 and + 60 C will generally be appropriate. The product of the reaction can be isolated in the usual manner.
The invention therefore encompasses, as novel compounds, the substances of formula I given above, in which R and Y have the meanings indicated.
More specifically, the invention encompasses the novel compounds of general formula I, in which R denotes either a hydroxyl group or the acetic or benzoic acid ester thereof and X denotes bromine.
The constants of some of the new compounds according to the invention will be given below.
EMI3.1
22: -dibromo- -7: 11 -trihydrox: v-8: 9-0x: vdoergos- tane has the following constants: melting point: 238-239 C /
EMI3.2

232 C
EMI3.5
Cd D = + 190
It is obvious that the foregoing constants have been determined for the purest material Applicant has been able to obtain and may undergo variations depending on the purity of a particular product.
In the preparation of the 3-hydroxy compounds of general formula I, that is to say the compounds, in which R denotes a hydroxy group, it is
<Desc / Clms Page number 4>
may be more advantageous, from the point of view of the isolation of the product, to prepare one of the 3-esters described in the present specification, by the oxidation technique, then to obtain the 3-hydroxy compound from this ester by hydrolysis.
To facilitate a better understanding of the invention, the following examples illustrate the manner in which it can be implemented.
