PERFECTIONNEMENTS A LA PREPARATION DE NOUVELLES SUBSTANCES
STEROIDES.

Chim. Acta, 1951, 34, 2108) et par Chamberlain(J.A.C.S. 1951, 73, 2396) . Ces compos

 <Desc/Clms Page number 2>

sés connus comportent une chaîne latérale non saturée.
 EMI2.1

 EMI2.4
 acéty10xy, propyloxy ou benzovlo, et
X = brome

IMPROVEMENTS IN THE PREPARATION OF NEW SUBSTANCES
STEROIDS.

The present invention relates to improvements - @ @ nomemts to the preparation of new steroid substances of the following general formula:
 EMI1.1
wherein R denotes an optionally esterified hydroxy group and X denotes chlorine or bromine. These compounds have been shown to be particularly valuable in research relating to the synthesis of cortisone and related substances. Thus, the above compounds can be oxidized in a series of stages, so as to form 8: 9-epoxy-7: 11-diketones, which can be dehalogenated and reduced to 11-ketone compounds.

These half compounds, by transformation of the side chain in C17 into the side chain of cortisone and by development of the 3-keto system # 4, for example by the method of Djerassi, providing cortisone. The conversion of the new compounds described in the present specification into cortisone has been or will be the subject of patent applications filed by the applicant.

Compounds similar to the new compounds described herein have been described by Heusser et al. (Helv. Chim. Acta, 1951, 34, 2108) and by Chamberlain (J.A.C.S. 1951, 73, 2396). These compos

Its known products have an unsaturated side chain.
 EMI2.1
The Applicant has, however, discovered that the bihalogenated compounds described in the present specification are more suitable in a synthesis process, in that they are isolated more easily and with a higher yield than the corresponding unsaturated compounds.

Thus, the halogenated substituents at the 22:23 positions have the effect of appreciably decreasing the solubility of the compounds, which greatly facilitates their isolation. In fact; provided that a suitable solvent is used for
 EMI2.2
their preparation, as described below, can be t: ber, advantage of a difference in solubility between the starting materials and the products obtained; - to such an extent that these latter products precipitate in a more or less pure form during their formation.

The subject of the invention is therefore, as new compounds, those which have the following general formula:
 EMI2.3
in which R and X have the meanings indicated above.

The invention relates in particular to the compounds of the general formula given above, in which the symbols R and X have the following preferred meanings:
R = hydroxy or acyloxy group, in particular
 EMI2.4
 acety10xy, propyloxy or benzovlo, and
X = bromine
For illustrative purposes, the following are physical constants of some of the novel compounds described herein.

 EMI3.1
According to another feature of the invention, the new "écmbo = ses are prepared by hydrolysis and, emX1t, in moderate conditions, of compounds of the following general formula:

 EMI3.2

 EMI3.3
in which R and X have the meanings indicated above, in solution in a mixture of solvents consisting of an organic solvent miscible with 1 - water and water, in the presence of a strong mineral acid devoid of oxidative properties.
 EMI3.4
tes under the conditions present, the normality liu 1I (above-mentioned mixture of solvents with respect to said mineral acid being between 0.1 and 2 N.-
The reaction is preferably carried out at temperatures between -20 and + 60 C and advantageously at room temperature. As mineral acid, it is preferred to use sulfuric acid.

It should be noted that too severe conditions, ie the application of high temperatures and long reaction times, can give rise to the formation of # -7-ketone compounds. The operation must therefore be done under moderate conditions and the expression "moderate conditions" indicates that conditions of temperature and time are applied which give rise to the formation of the desired diols, which can easily be determined by preliminary tests.

The Applicant prefers to adjust the relative proportions of the organic solvent miscible with water with respect to the water in the mixture of solvents, so that the starting materials are easily soluble therein, while the final products precipitate therein during of their training. The relative proportions of organic solvent relative to water, suitable for giving the results specified above, vary according to the solubility of the particular product in preparation, and according to the nature of the particular solvent employed. The appropriate proportions in each given case must be determined empirically by a preliminary test. As suitable organic solvent, there may be mentioned
 EMI3.5
 tetraqydrofuran.

The Applicant has found, for example, that by using a 20% solution of 3 f -acetoxy-22: 23-di.-bromo-9: 11-époxyertpst-7-ene in a mixture of 3 parts dibromo by volume epoxy of a 2N mineral acid and 50 parts

Although the Applicant does not wish to limit herself by theoretical considerations,

it assumes that the substituents occupying the 5 and 11 positions have the [alpha] configuration. In addition, although it is not certain of the configuration of the hydroxy group at position 7, Applicants assume

that it has the Jb configuration
