PERFECTIONNEMENTS AUX COMPOSES DE PYRIMIDINE.
La présente invention concerne une nouvelle série de pyrido- (3,2-d)-pyrimidines présentant un intérêt comme composés intermédiaires pour la préparation de médicaments et comme agents d'inhibition du développement de microorganismes.
La synthèse de ces composés comprend la préparation initiale

d'une 2y4'dihydroxypyrdo-(3t2-d)-pyrimidine en faisant réagir un acide 3-aminopicolinique avec de l'urée

et la transformation du produit en 2,4-dichloropyrido-(3,2-d)-pyrimidine correspondante.. Les substances désirées sont formées à partir de 2,4-dichloropyrido-(3,2-d)-pyrimidines par réaction avec des agents de la classe des aminés.
Par traitement par la thiourée on obtient la 2,4-dimercap- topyrido-(3,2-d)-pyrimidineo Cette dernière est un intermédiaire convenable pour la préparation de dérivés portant différents substituants fonc- tionnels en positions 2 et 4, de facon analogue à celle décrite dans les demandes de brevets américains n 0 2610393, 2610394 et 26103950

Préparation de 2,4dihvdroxvnvrdo=(3,2-d)-pvrimidine On chauffe 40 g d'acide 3-aminapucolinique dans un récipient avec 80 g d'urée. Le mélange qui fond à 110 est maintenu à 160 jusqu'à ce que la masse fondue limpide devienne pâteuse. La température est ensuite graduellement portée à 180 et le chauffage interrompu après 5 minuteso
La matière solide refroidie est dissoute dans 400 cm3 d'hydroxyde de sodium 2 N chaud et on laisse reposer la solution à la température ambiante pendant 1 heure puis on filtre.
Le filtrat est chauffé au bain de vapeur tandis qu'on le sature d'un courant d'anhydride carboniqueo La solution refroidie fournit 21 g d'un composé fondant à 360 . Une petite quantité ést recristallisée de l'acide acétique glacial aux fins d'analyse.
EXEMPLE 20-

On ajoute 10 g de 2l4-dihydroxypyrido-(3,2-d)-pyrimidine à 150 cm3d'oxychlorure de phosphore et 50 g de pentachlorure de phosphore.
La solution est chauffée à reflux pendant 2 heures et l'excès d'oxychlç- rure de phosphore chassé par distillation sous vide, puis on ajoute le résidu à 200 g de glace pilée. La solution aqueuse froide est extraite au chloroforme et l'extrait au chloroforme est lavé à l'eau et séché sur le sulfate de magnésium. L'évaporation du chloroforme laisse 5,1 g d'un produit légèrement jaune fondant à 170-175 . Une petite quantité du produit est recristallisée de "Skellysolve C" et donne des plaques blanches fondant à 177 .
EXEMPLE 3.-

On ajoute 2,5 g de 2,4-dichloropyrido-(3.2-d)-pyrimidîne à 20 cm3 d'ammoniac en solution alcoolique (alcool saturé d'ammoniac an- hydre à 0 C), on place cette solution dans une bombe et on la chauffe à 155 -160 pendant 18 heures. La solution refroidie est ajoutée à un volume étal d'eau et la solution rendue fortement basique par3de l'hydroxyde de sodium. Le produit filtré est recristallisé de 20 cm d'éthanol à 50% ce qui fournit de fines aiguilles blanches fondant à 317-319 .

La solution refroidie fournit 1,3 g d'aiguilles brunes fondant à 168-1700. Une seconde recristallisation de l'éthanol n'élève pas le point de fusion.
EXEMPLE 5. -

On dissout 6 g de thiourée dans 200 cm3d'alcool absolu et on refroidit la solution à la température ordinaire puis on ajoute 6,0 g de

2,±-dichloropyrido-(3,2-d)-pyrimidine brute et on chauffe la solution à reflux au bain de vapeur pendant 5 heures. Le mélange de réaction est ensuite refroidi et filtré et le précipité dissous dans de l'hydroxyde de sodium 2 N froid et précipité par de l'acide acétique dilué. Le précipité filtré est lavé et séché et fournit 4,3 g de poudre jaune-vert amorphe.
On purifie 2 g de ce produit par extraction au soxhlet avec de l'alcool absolu et on obtient 1,6 g de cristaux jaune-orange se décomposant à 335-340 .
EXEMPLE 6-

2-mercapto-4-aminopyrido-(3.2-d)-pyrimidine.
On dissout 2,3 g de 2,4-dimercaptopyrido-(3,2-d)-pyrimidine brute dans 100 cm3d'hydroxyde d'ammonium concentré et on chauffe légèrement la solution au bain de vapeur. Après 1/2 heure environ on voit appas- raitre un précipité vert clair. On laisse évaporer la solution jusqu'à 25 en! puis on ajoute à nouveau 100 cm3 d'hydroxyde d'ammonium concentré et on chauffe la solution au bain de vapeur. On répète cette opération trois fois puis on filtre la solution à chaud et on lave le précipité à l'hydroxyde d'ammonium concentré froid et on sèche. Le rendement atteint 1,8 g de petites aiguilles jaune-orange se décomposant à 345 .

The present invention relates to a new series of pyrido- (3,2-d) -pyrimidines of interest as intermediate compounds for the preparation of medicaments and as agents for inhibiting the growth of microorganisms.

Preparation of 2,4dihvdroxvnvrdo = (3,2-d) -pvrimidine 40 g of 3-aminapucolinic acid are heated in a container with 80 g of urea. The mixture which melts at 110 is maintained at 160 until the clear melt becomes pasty. The temperature is then gradually raised to 180 and the heating is interrupted after 5 minutes.
The cooled solid is dissolved in 400 cm3 of hot 2N sodium hydroxide and the solution is allowed to stand at room temperature for 1 hour and then filtered.
The filtrate is heated in a steam bath while saturated with a stream of carbon dioxide. The cooled solution provides 21 g of a compound, melting at 360 °. A small amount is recrystallized from glacial acetic acid for analysis.

10 g of 2l4-dihydroxypyrido- (3,2-d) -pyrimidine are added to 150 cm3 of phosphorus oxychloride and 50 g of phosphorus pentachloride.
The solution is heated under reflux for 2 hours and the excess of phosphorus oxychloride removed by vacuum distillation, then the residue is added to 200 g of crushed ice. The cold aqueous solution is extracted with chloroform and the chloroform extract is washed with water and dried over magnesium sulfate. Evaporation of the chloroform leaves 5.1 g of a slightly yellow product, melting at 170-175. A small amount of the product is recrystallized from "Skellysolve C" to give white plaques melting at 177.
EXAMPLE 3.-

2.5 g of 2,4-dichloropyrido- (3.2-d) -pyrimidîne are added to 20 cm3 of ammonia in alcoholic solution (alcohol saturated with anhydrous ammonia at 0 C), this solution is placed in a bomb and heated at 155-160 for 18 hours. The cooled solution is added to a standard volume of water and the solution made strongly basic by sodium hydroxide. The filtered product is recrystallized from 20 cm3 of 50% ethanol which gives fine white needles melting at 317-319.

The cooled solution provides 1.3 g of brown needles, melting at 168-1700. A second recrystallization of ethanol does not raise the melting point.

The reaction mixture is then cooled and filtered and the precipitate dissolved in cold 2N sodium hydroxide and precipitated with dilute acetic acid. The filtered precipitate is washed and dried to give 4.3 g of amorphous yellow-green powder.
2 g of this product are purified by Soxhlet extraction with absolute alcohol to give 1.6 g of yellow-orange crystals which decompose at 335-340.

2.3 g of crude 2,4-dimercaptopyrido- (3,2-d) -pyrimidine are dissolved in 100 cc of concentrated ammonium hydroxide and the solution is heated slightly in a steam bath. After about 1/2 hour, a light green precipitate appears. The solution is allowed to evaporate to 25%! then again 100 cm3 of concentrated ammonium hydroxide are added and the solution is heated in a steam bath. This operation is repeated three times then the solution is filtered hot and the precipitate is washed with cold concentrated ammonium hydroxide and dried. The yield reaches 1.8g of small yellow-orange needles decomposing to 345.
