Les différents procédés de préparation de tels es- ters sont connus. Ils consistent à faire réagir une substan- ce de formule (1) avec une substance de formule (2) dans les- quelles R, R1' R2 sont des restes alcoyles, x et y étant deux radicaux différents qui sont éliminés au cours de la réaction à l'exception d'un atome d'oxygène se trouvant dans l'un d'eux.

Les restes alcoyles R, R1 et R2 peuvent être en partie ou en totalité identiques ou différents; ils peuvent en parti- culier être des restes méthyle, éthyle, propyle.
Les deux radicaux X et Y qui doivent être différents dans les substances amenées à réagir l'une avec l'autre, peu- vent être soit un groupe -OH, -ONa, soit un atome d'halogène.

Parmi les esters qui peuvent tre préparés par le pro- cédé objet de l'invention, le méta-diéthylaminoéthoxybenzoate d'éthyle est spécialement intéressant vu ses propriétés. Cet ester possède en effet un pouvoir anesthésique par contact su- périeur à celui de la diéthyl-amino-2,6-diméthyl-acétalinide (lignocaïne) et de la cocaïne, mais inférieur à celui du chlorhydrate d butyloxycinchoninate de diéthylènediamine (cinchocaïne). Son pouvoir anesthésique par injection est su- périeur à celui du chlorhydrate de p-amino-benzoyl-diéthyl- aminoéthanol (procaïne) et de la diéthyl-amino-2,6-diméthyl- acétalinide, non additionnées d'adrénaline, mais inférieur à celui de la cinchocaine (sans adrénaline)
et de la lignocaïne additionnée d'adrénaline. Son pouvoir anesthésique est poten- tialisé par l'adrénaline, comme c'est le cas pour les anesthé- siques étalons. Ce nouvel ester permet aussi de préparer un anesthésique efficace sans adrénaline, ce qui est intéressant dans les cas où il existe une hypersensibilité neurovégétative (tachycardie extrasystoles syncope) à l'adrénaline.
Voici comment le procédé de l'invention peut être réa- lisé pour la préparation du métadiéthylaminoéthoxybenzoate d'éthyle
Exemple 1
On dissout, d'une part 0,46 g de sodium métallique dans 20 cc d'alcool éthylique absolu, et, d'autre part 3,32 g de m-oxybenzoate d'éthyle dans la même quantité d'alcool éthy- lique absolu. On mélange à froid les deux solutions obtenues.
On ajoute au tout 2,71 g de chloréthyldiéthylamine fraîchement distillée. Après quelques instants, la solution se trouble et devient laiteuse., On chauffe alors au bain-marie plusieurs

heures. On laisse refroidir, puis filtre le chlorure de sodium formé. Le filtrat est évaporé sous pression réduite et laisse un résidu sous forme d'une huile jaune pâle. On lave cette dernière à l'eau, puis on l'extrait à l'éther et on sèche la solution éthérée. Après élimination de l'éther on obtient le méta-diéthylaminoéthoxybenzoate d'éthyle que l'on transforme en son chlorhydrate.
Ce dernier est une poudre blanche, solu- ble dans l'eau et dans l'alcool et insoluble dans l'éther; son point de fusion est de 121 à 122 C, selon la méthode de la Pharmacopée Helvetica, et de 122 à 123 C avec l'appareil électrique "Culatti".
