La présente invention concerne des compositions pharma- ceutiques contenant l'amine 1-phénylcyclohexylique et ses sels d'addition acide.
Elle a pour objet des compositions pharmaceutiques qui, administrées'à un animal vivant ou à une personne, exercent un effet sédatif sur le système nerveux central. L'agent actif de ces compositions est l'amine 1-phénycyclohexylique. Ce constituant peut ' être la base libre de l'aminé 1-phénylcyclohexylique ou un de ses sels non toxique d'addition acide. Un "sel non toxique d'addition acide" de l'amine 1-phénylcyclohexylique, du type suivant la présente invention, comprend l'un ou l'autre sel d'addition acide, qui n'est pratiquement pas plus toxique qu'un poids équivalent de

l'amine 1-phénylcyclohexylique elle-même. Parmi ces sels, on citera, par exemple, le chlorhydrate, le bromhydrate, le sulfate, le sulfamate, l'acétate, le phosphate, le succinate, le tartrate, le p-toluène-sulfonate, le benzène-sulfonate, etc.
Les compositions de la présente invention sont celles dans lesquelles le composant actifprécité est combiné avec un support ou diluant solide ou li- quide. Le mélange obtenu est mis sous l'une ou l'autre forme phar- maceutique habituelle, c'est-à-dire en tablettes, en capsules remplies de liquide et de produit sec, en solutions aqueuses ou non aqueuses pour l'administration par voie buccale ou parentérale, en dragées, en suppositoires pour l'administration par voie rectale et en inhalations, en gelées et en pulvérisations pour l'admistra- tion par voie nasale, etc. Les compositions peuvent et, dans de nombreux cas doivent,contenir des agents préservatifs et odorants appropriés.
Parmi les supports ou diluants qui peuvent être utilisés lors de la préparation des produits de la présente invention, on citera des sucres tels que la lactose et la sucrose, des dérivés cellulosiques tels que la carboxy-méthyl-cellulose de sodium, l'éthyl-cellulose, la méthyl-cellulose et le phtalate d'acétate de cellulose; la gélatine; le talc ; stéarate de magnésium; des huiles végétales telles que l'huile d'arachide, l'huile de coton, l'huile de sésame, l'huile d'olive, l'huile de mais et l'huile de théobrome ; la vaseline officinale liquide, le polyéthylène- glycol; la glycérine; le sorbitol; le propylèneglycol; l'éthanol; l'agar; les sels purgatifs aqueux et isotoniques.
Lorsqu'on prépare les compositions de la présente in- vention, il faut appliquer les procédés et précautions pharmaceuti- ques habituels. Les compositions à administrer par voie parentérale doivent .être stérilisées et, à cet effet, on utilise des ingrédients stérilisés et on effectue la préparation dans des conditions aseptiques ou en stérilisant la composition finale suivant l'un des procédés habituels tels que la filtration de Seitz. On fait passer 50 g de chlorhydrate d'amine 1-phényl- cyclohexylique par un tamis en acier inoxydable à mailles N 60, puis on les mélange intimement avec 180 g de sucre de lait et 215 g de sucrose contenant 3% d'amidon. On granule le mélange avec 40 g de pâte d'amidon? obtenue à partir d'une partie d'amidon et de sept parties d'eau. On sèche les produits granulés et on les fait passer par un tamis n 20. On mélange 17,5 g de talc, 35 g d'amidon de maïs et 1 g de stéarate de magnésium, puis on comprime le mélange en tablettes concaves de 1/4 pouce (6,35 mm)'. Rendement : environ 5. 000 tablettes, contenant chacune 10 mg de chlorhydrate d'aminé 1-phénylcyclohexylique.

EXEMPLE 2.-
On mélange 100 g de p-toluène-sulfonate d'amine 1-phényl- cyclohexylique avec 1275 g de lactose et 125 g de stéarate de magnésium. On verse le mélange obtenu dans des capsules de gélatine dure d'une contenance de 150 mg chacune. Rendement 10.000 capsules, contenant chacune 10 mg de p-toluène-sulfonate d'amine 1-phénylcyclohexylique.
EXEMPLE 3. -
On mélange 100 g de chlorhydrate d'amine 1-phénylcyclo- hexylique avec 5370 g d'amidon et 530 g de stéarate de magnésium.
On verse le mélange obtenu dans des capsules de gélatine à enveloppe dure d'une contenance de 150 mg chacune. Rendement 40.000 capsules contenant chacune 2,5 mg de chlorhydrate d'amine 1-phénylcyclo- hexylique.
EXEMPLE 4.-
On dissout 200 g d'amine 1-phénylcyclohexylique dans 4800 g d'huile de coton et on utilise la solution obtenue pour prépa- rer des capsules de gélatine molles, contenant chacune 20 mg d'amine 1-phénylcyclohexylique. Rendement : 10.000 capsules.
EXEMPLE 5.-
On dissout 80 g de chlorhydrate d'amine 1-phénylcyclo- hexylique dans 4 litres d'eau exempte de pyrogène, pour injection.
On ajoute un tampon de citrate pour porter le pH à 5,5 et on dilue la solution jusqu'à un volume de 8 litres. On verse la solution obtenue dans des ampoules contenant 1,2 ml dans des conditions stériles, de sorte que le médecin peut retirer un ml contenant 10 mg - de chlorhydrate d'amine 1-phénylcyclohexylique.
EXEMPLE 6.-
On dissout 100 g d'amine 1-phénylcyclohexylique dans 10 litres d'huile d'arachide contenant 10 g d'alcool benzylique.
On verse, dans des conditions stériles, des portions de 1,2 ml de la solution obtenue dans des capsules, de sorte que l'on peut retirer un ml contenant 10 mg d'aminé 1-phénylcyclohexylique.

On mélange 10 g de chlorhydrate d'amine 1-phénylcyclo- hexylique avec 1200 g d'huile de théobrome et on transforme le mélan ge obtenu en suppositoires, contenant chacun 25 mg de chlorhydrate d'amine 1-phénylcyclohexylique. Rendement : environ 400 supposi- toires.
EXEMPLE 8.-
On dissout 6 g d'amine 1-phénylcyclohexylique et 1,5ml d'huile d'orange, dans 400 ml de glycérine. On ajoute 300 ml d'eau à la solution,puis on.dissout, dans le mélange obtenu, 100 g de sucrose et 100 g de sorbitol. On filtre la solution pour la clarifier et on..dilue le produit de filtration à un volume d'un litre, avec de l'eau. On verse la solution obtenue dans des fioles contenant chacune 15 ml. Chaque ml de cette solution contient 6 mg d'amine 1 -phénylcyclohexylique.
EXEMPLE 9.-
On dissout 1,25 g de chlorhydrate,d'amine l-phénylcyclo- hexylique dans 400 ml d'eau et on dissout 200 g de sucrose dans la solution obtenue. On dissout 100 g de sorbitol dans la solution et on ajoute 1,5 ml d'huile d'orange et 1 g de benzoate de sodium. On filtre la solution, on dilue le produit de filtration à un litre, avec de l'eau et on verse la solution diluée dans des fioles contenant 20 ml. Chaque ml de la solution ainsi obtenue contient 1,25 mg de chlorhydrate d'amine 1 -phénylcyclohexylique.
On peut préparer, de plusieurs manières, l'amine 1-phényl- cyclohexylique et ses sels d'addition acide, utilisés dans la prépara- tion des compositions de la présente invention. un groupe amido primaire, un radical carboxylique, un ester de carboxylate ou un groupe nitrile. L'exemple suivant illustre un des procédés permettant d'effectuer cette transforraation.
.On ajoute 51 g d'acide carboxylique de cyclohexane
1-phénylique dans 500 ml de benzène, à 150 ml d'acide sulfurique concentré et on agite vigoureusement le mélange obtenu, à 40 C.
On ajoute lentement une solution de 12 g d'acide hydrazoique dans

On rend la couche aqueuse fortement basique en ajoutant un excès d'une,.solution aqueuse d'hydroxyde de sodium et on extrait l'amine libérée à l'éther. On lave le produit d'extraction d'éther à l'eau; on le sèche au carbonate de potassium et on distille l'éther de la solution séchée. On distille le résidu sous pression réduite, pour obtenir l'amine 1-phénylcyclohexylique désirée, sous forme pure.
. Point d'ébullition: environ 84-9 C à 1 mm.
On prépare le chlorhydrate d'amine 1-phénylcyclohexyli- que en dissolvant la base libre dans de l'éther et en traitant la solution avec un excès d'acide chlorhydrique isopropanolique.
On recueille le chlorhydrate précipité et on le purifie par re- cristallisation dans un mélange d'isopropanol et d'éther. Point de fusion :247-8 C. On peut préparer le bromhydrate d'amine
1-phénylcyclohexylique de la même manière, en utilisant une. solution alcoolique d'acide bromhydrique au lieu de l'acide chlor- hydrique isopropanolique.
On peut préparer l'acétate d'amine 1-phénylcyclohexylique en traitant la base libre avec de l'acide acétique glacial et en précipitant le sel par addition d'éther. On purifie le sel par recristallisation dans un mélange d'isopropanol et d'éther. Point de fusion: 144-5 C.
On peut préparer le sulfate d'amine 1-phénylcyclohexyli- que en ajoutant la base libre à une solution alcoolique contenant un équivalent d'acide sulfurique. On précipite le sel par addi-
