   La présente invention concerne un procédé de prépara- tion d'amine 1-phénylcyclohexylique et de ses sels d'addition acide. 



   Suivant la présente invention, on prépare l'amine 1-phénylcyclohexylique en hydrogénant catalytiquement l'amine N-benzyl-l-phénylcyclohexylique, pour éliminer le groupe benzyli- que. On peut effectuer   l'hydrogénation   dans des conditions neutres ou acides. Les catalyseurs que l'on utilise de préférence sont les catalyseurs au palladiu, bien que l'on puisse également em- ployer les catalyseurs au platine et au nickel de Raney. On obtient les meilleurs résultats en   utilisanttune   quantité relati- vement importante de catalyseur. Comme solvants, il est préfé-

 rable d'employer les alcools aliphatiques inférieurs. Les pres- sions d'hydrogène ne sont pas critiques et il n'est pas néces- saire d'employer des pressions très élevées.

   Dans la plupart des cas, une pression de quelques atmosphères suffit. La tempé- rature n'est pas critique non plus et elle peut varier entre 0 et 75 C. Pour la facilité, on utilise habituellement la tempé- rature ambiante (environ 20 à 25 C). 



   Lorsqu'on applique le procédé décrit ci-dessus, on obtient souventl'aminé l-phénylcyclohexylique sous forme de base libre. On peut obtenir les sels d'addition acide de l'amine   1-phénylcyclohexylique   à partir de la base libre, par traitement avec un acide. Parmi les nombreux sels qui peuvent être prépa- rés de cette manière, on citera, par exemple, les sels d'acides minéraux tels que le chlorhydrate, le bromhydrate, le sulfate et le phosphate;

   des sels d'acides organiques tels que le succi- nate,le benzoate, l'acétate, le p-toluène-sulfonate et des sels avec d'autres acides tels que le   sulfamate.   Lorsque, sui- vant le procédé décrit ci-dessus, on obtient un sel d'addition acide d'amine   1-phénylcyclohexylique,   on peut obtenir la base libre en traitant les sels d'addition acide avec un réactif alca- lin tel qu'une solution d'hydroxyde de sodium, une solution de bicarbonate de sodium, une solution de carbonate de potassium,etc.. 



   L'aminé l-phénylcyclohexylique et ses sels non toxi- ques d'addition acide possèdent des propriétés médicales utiles. 



  Un "sel non toxique d'addition acide" d'amine   1-phénylcyclohexy-   lique, du type suivant la présente invention, comprend l'un ou l'autre sel d'addition acide qui n'est pratiquement pas plus toxique qu'un poids équivalent de l'amine   1-phénylcyclohexylique   elle-même. Parmi ces sels, on citera, par exemple, le chlorhy- drate, le   bromhydrate,   le sulfate, le sulfamate, l'acétate, le phosphate, le succinate, le tartrate, le p-toluène-sulfonate, le benzène-sulfonate, etc.. Administrés à un animal vivant ou à une personne., ces produits exercent un effet sédatif sur le système

Les produits peuvent être utilisés pour le traitement de l'hyper-excitabilité chez les animaux et chez l'homme. 



  Lorsqu'il s'agit d'animaux, ces produits ont une grande valeur au point .de vue pratique, car ils permettent de traiter,   d'en-   fermer et/ou de transporter des animaux., qui, en d'autres con- ditions, risqueraient de blesser les personnes et eux-mêmes également.  en administrant 10 à 30 mg/kg. par injection intra-musculaire ou 2 à 15   mg/kg   par injection intra-veineuse et, chez l'homme adulte, en administrant 0,5 à 3 mg/kg par injection intra- musculaire et 0,5 à 3   mg/kg   par injection intra-veineuse. On peut obtenir un effet euphorique chez l'homme, en administrant, par voiebuccale,' 1,25 à 15 mg. Lorsqu'on utilise les produits pour augmenter l'effet des agents anesthésiques, la dose par voie buccale chez   les adultes   est comprise entre 5 et 50 mg. 



  Pour pratiquer une opération chirurgicale chez les chiens, il faut, à cet effet, employer une dose de 3 à 10 mg/kg, par injec- tion intra-muschlaire. 



   On peut appliquer et administrer les produits sous l'une ou l'autre forme pharmaceutique habituelle, c'est-à-dire en tablettes, en capsules remplies de liquide ou de produit sec, en solutions aqueuses ou non aqueuses pour l'administration par voie buccale ou parentérale, en dragées, en suppositoires pour l'administration par voie rectale et en inhalations, en gelées et en pulvérisations pour l'administration par voie nasale, etc. 



   La présente invention est illustrée par l'exemple suivant : EXEMPLE. - 
On soumet à' l'hydrogénation 96 gr d'amine N-benzyl-1-   phénylcy clohexylique   dans 300 ml. de méthanol contenant un équivalent d'acide chlorhydrique (31 ml. d'acide chlorhydrique concentré), sous quatre atmosphères d'hydrogène, en présence de 25 gr de catalyseur au palladium sur du charbon de bois à 20%, jusqu'à ce qu'un équivalent d'hydrogène soit absorbé. On élimine le catalyseur par filtration et on évapore le méthanol du produit de filtration. On dilue le résidu à l'eau; on alcalinise le mélange avec une solution d'hydroxyde de sodium et on l'ex- trait à l'éther.

   On sèche les produits d'extraction d'éther, on   distille$l'éther     et'-l'on   distille le résidu sous vide pour

 obtenir l'amine l-phénylcyclohexlique désirée. Point d'ébulli- tion : 92-5 C à 0,8 mm. 



   On prépare .le chlorhydrate d'amine l-phénylcyclohexyli- que en dissolvant la base libre dans de l'éther et en traitant' la solution avec un excès d'acide chlorhydrique isopropanolique. 



  On   recualle   le chlorhydrate précipité et on le purifie par recristallisation dans un mélange d'isopropanol et   d'éther.   



  Point de fusion :   247-8 C.   On peut préparer le bromhydrate d'amine   l-phénylcyclohe:xylique   de la même manière, en utilisant une solu- d'acide tion alcoolique/bromhydrique au lieu d'acide chlorhydrique iso- propanolique. 



   On peut préparer l'acétate d'aminé l-phénylcyclohexyli- que en traitant la base libre avec de l'acide acétique glacial et en précipitant le sel par addition d'éther. On purifie le sel par recristallisation dans un mélange d'isopropanol et d'éther. Point de fusion: 144-5 C. 



   Le produit de départ utilisé dans le procédé ci-dessus, c'est-à-dire l'amine   N-benzyl-1-phénylcyclohexylique,   peut être préparé de la manière suivante : 
On reflue un mélange se composant de 214 gr de benzyl- amine, de 200 gr de cyclohexanone et de 300 ml. de toluène, jusqu'à ce qu'il n'y ait plus écoulement d'eau. Tout en agitant., on ajoute la solution chaude à une solution de lithium phényli- que, préparée à partir de   472   gr. de bromobenzène et de   44   gr. de lithium dans 200 ml. d'éther. On agite le mélange réactionnel pendant deux heures, puis on le décompose en ajoutant prudem- ment 500 ml. d'eau. On élimine la couche d'éther, on la lave à l'eau et on la sèche.

   On distille l'éther et on distille le résidu sous vide, pour obtenir   l'aminé   N-benzyl-1-phényl- cyclohexylique désirée. Point d'ébullition: 145-50 C à 0,2 mm.

  The precipitated hydrochloride is collected and purified by recrystallization from a mixture of isopropanol and ether.



  Melting point: 247-8 C. The 1-phenylcyclohe: xyl amine hydrobromide can be prepared in the same manner, using an alcoholic / hydrobromic acid solution instead of isopropanolic hydrochloric acid.



   1-Phenylcyclohexyl amine acetate can be prepared by treating the free base with glacial acetic acid and precipitating the salt by adding ether. The salt is purified by recrystallization from a mixture of isopropanol and ether. Melting point: 144-5 C.



   The starting material used in the above process, i.e. the N-benzyl-1-phenylcyclohexyl amine, can be prepared as follows:
