On peut effectuer l'hydrogénation dans des conditions neutres ou acides. Les catalyseurs que l'on emploie de préférence, sont les catalyseurs au palladium bien que l'on puisse également utiliser des catalyseurs au platine et au nickel de Raney. On obtient les meilleurs résultats en utilisant une quantité relativement importante de catalyseur. Comme solvants, on emploie, de préférence, des al- cools aliphatiques inférieurs. Les pressions d'hydrogène ne sont pas critiques et il n'est pas nécessaire d'employer des pressions d'hy- drogène très élevées. Habituellement, une pression de quelques atmos. phères est suffisante. La température n'est pas critique non plus et elle peut varier de 0 à 75 C.
Pour plus de facilité, on utilise habituellement la température ambiante (environ 20-25 C).

Lorsqu'on applique le procédé décrit ci-dessus, on obtient habituellement les produits sous forme de base libre.
On peut préparer les sels d'addition acide à partir de la base libre, par réaction avec l'acide organique ou inorganique correspondant.
Les amines 1-phénylcyclohexyliques mono-N-substituées suivant la présente invention et leurs sels non toxiques d'addition acide, possèdent des propriétés médicales utiles. Un "sel non toxique d'addition acide" d'amines 1-phénylcyclohexyliaues mono-N- substituées,du type suivant la présente invention, comprend l'un ou l'autre sel d'addition acide, qui n'est pratiquement pas plus toxique qu'un poids équivalent de l'aminé 1-phénylcyclohexylique mono-N-substituée elle-même. Parmi ces sels, on citera., par exemple, le chlorhydrate, le bromhydrate, le sulfate, le sulfamate, l'acétate, le phosphate, le succinate, le tartrate, le p-toluène-sulfonate, le benzène-sulfonate et autres. Administrés à u n animal vivant ou à une personne, ces produits exercent un effet sédatif sur le système nerveux central.
En variant la quantité de produit administré, on peut obtenir différents degrés d'effet sédatif sur le système nerveux central, ces degrés se manifestant eux-mêmes par des effets allant de l'euphorie à la dépression et à l'état catalepti.que. Les produits peuvent être utilisés pour le traitement de l'hyper-excitabilité chez les animaux et chez l'homme- Lorsqu'il s'agit d'animaux, ces. produits ont une grande valeur au point de vue pratique, car ils permettent de traiter., d'enfermer et/ou de transporter des animaux, qui. en d'autres conditions, risqueraient de blesser les personnes et eux-mêmes également. On peut utiliser ces produits pour calmer l'hyper-excitabilité chez l'homme.
Lorsqu'il s'agit d'hyper- excitabilité se manifestant par une agressivité, ces produits pré- sentent. une grande valeur d'ordre pratique si le patient doit être enfermé, car ils permettent de procéder à cette opération sans danger pour le malade ni pour les personnes chargées de le soigner. On peut également utiliser les produits delà présente invention comme anes,thésiques, en chirurgie animale et humaine, car ils peuvent être employés pour obtenir un état cataleptique. A cet effet, ils peuvent

être utilisés seuls ou avec d'autres agents anesthésiques.
Lorsqu'ils sont utilisés avec d'autres agents anesthésiques, en particulier ceux du type acide barbiturique, ils exercent l'effet des autres agents anesthésiques, permettant ainsi d'employer des quantités beaucoup plus faibles et réduisant, par conséquent, les effets se- condaires inopportuns'qui peuvent se produire, comme par exemple la dépression respiratoire.
Le dosage des produits varie évidemment suivant le mode d'administration,le sujetà traiter et l'effet désiré. On peut obtenir un état cataleptique chez les chiens, en administrant 3 à 10 mg/kg par voie buccale ou 2 à 10 mg/kg par injectionintra- musculaire; chez les chats, en administrant 1 à 5 mg/kg par voie buccale; chez les singes, en administrant 5 à 20 mg/kg par voie intrapéritonale ou 0,65 à 5 mg/kg par injection intra- veineuse et,chez l'homme adulte, en administrant 0,1 mg à 1,5 mg/kg par injection intramusculaire et 0,1 à 1 mg/kg par injection intraveineuse. On peut obtenir un effet euphorique chez l'homme, en administrant,par voie buccale, 0,75 à 10 mg.
Lorsqu'on utilise les produits pour augmenter l'effet des agents anesthésiques, la dose par voie buccale chez les adultes, est comprise entre 5 et 50 mg.
Pour pratiquer une opération chirurgicale chez les chiens, il faut, à cet effet, employer une dose de 3 à 10 mg/kg par injection intra- musculaire. Chez l'homme, des doses intraveineuses augmentant de 0,5 à 1 mg., jusqu'à ce qu'on obtienne une dose totale d'environ 1 mg/kg bloquent, environ en une heure, d'une manière relativement complète, tous les types d'énergie vitale, sans que le patient perde complètement connaissance et sans affaiblir fortement les réflexes normaux de la circulation et de la respiration, de sorte que l'on peut pratiquer d'importantes opérations sur n'importe quelle partie d corps et dans les cavités abdominales ou thoraciques. Les produits peuvent être également utilisés pour atténuer les douleurs de l'ac- couchement.

La présente invention est illustrée par l'exemple suivant.
EXEMPLE.-
On dissout 26 g de formiate d'aminé N-méthyl-N-benzyle
1-phénylcyclohexylique dans 200 ml de méthanol et l'on ajoute 10 g de catalyseur au palladium sur du charbon de bois à 20%. On soumet le mélange à l'hydrogénation sous une pression d'hydrogène de quatre atmosphères, à la température ambiante, jusqu'à ce qu'un équi- valent soit absorbé. On élimine le catalyseur. par filtration et on distille le méthanol du produit de filtration. On dilue le résidu à l'eau,on alcalinise le mélange et on l'extrait à l'éther.
On lave le produit d'extraction d'éther, on le sèche et on distille 1"éther. La distillation sous vide du résidu donne l'aminé 1- phénylcyclohexylique désirée.
On peut préparer le'chlorhydrate d'aminé 1-phénylcyclo- hexylique en ajoutant une solution éthérée de la base libre à un excès d'acide chlorhydrique isopropanolique. On recueille le sel brut et on Je purifie par recristallisation dans un mélange d'isopro- .panol et d'éther. Point de fusion l85-186 C.

On chauffe, au bain de vapeur, 26,6 g d'amine N-benzyl- 1-phénylcyclohexylique, 23 g d'acide formique et 10 g de formal- déhyde aqueux à 37%, jusqu'à ce qu'il n'y ait plus de dégagement d'anhydride carbonique (3 heures). On évapore le mélange réaction- nel sous vide, pour éliminer l'excès de formaldéhyde et d'acide formique et pour obtenir le formiate d'amine N-méthyl-N-benzyl-1- phénylcyclohexylique désiré. Ce sel est suffisamment pur pour être utilisé dans le procédé ci-dessus. On peut éventuellement .transformer le sel en base libre et utiliser la base libre dans le procédé ci-dessus.
Dans ce cas,le résidu obtenu par évaporation du produit de filtration méthanolique, peut être soumis à distil- lation sous vide, pour obtenir le produit désiré.
Si l'on utilise l'amine N-éthyl-N-benzyl-1-phénylcyclo- hexylique dans le procédé ci-dessus et si l'on effectue la modi- fication, dont question dans le paragraphe précédent., on obtient l'éthylamine 1-phénylcyclohexylique. Point d'ébullition: 114-6 C à 5 mm. Le chlorhydrate d'éthylamine l-phénylcyclohexylique préparé comme décrit ci-dessus, a un point de fusion de 236-7 C.
