Abstract

SISTEMAS E MéTODOS PARA A PRODUçãO DE PROTEìNA A presente invenção refere-se a sistemas e métodos para a produção de proteínas de interesse. A invenção emprega células animais ou vegetais geneticamente manipuladas, que tenham capacidades modificadas de dobramento ou processamento de proteínas. Em um aspecto, a invenção apresenta células geneticamente manipuladas compreendendo um ou mais cassetes de expressão recombinantes que codificam (1) uma proteína de interesse e (2) um polipeptídio que seja funcional na via de resposta de proteína não dobrada (UPR) das células. A co-expressão do polipeptídio aumenta significativamente o rendimento da proteína de interesse nas células geneticamente manipuladas. Em um exemplo, as células geneticamente manipuladas são células animais, e o polipeptídio co-expressado é um componente ou modulador de uma via UPR mediada por XBP1 ou ATF6.
