Abstract

COMPOSIÇÃO FARMACÊUTICA, E, MÉTODO PARA A PREPARAÇÃO DE UMA COMPOSIÇÃO FARMACÊUTICA. A presente invenção diz respeito a composições farmacêuticas na forma particulada ou em formas de dosagem sólidas compreendendo sirolimus (rapamicina) e/ou derivados e/ou análogos deste. Composições da invenção apresentam urna biodisponibilidade aceitável de sirolimus e/ou um derivado e/ou um análogo deste. As composições farmacêuticas da invenção são projetadas para liberar sirolimus de urna maneira controlada, de forma tal que os níveis plasmáticos permaneçam na estreita janela terapêutica que existe para esta classe de substancias. Espera-se que um perfil de liberação lento, onde a concentração do pico foi reduzida sem perda significativa da biodisponibilidade, iuntamente com menos absorção variável, melhore a razão segurança/eficácia do medicamento. Além disso, espera-se que composições de acordo com a invenção fbrneçam um efeito do alimento reduzido significativo e uma liberação atrasada de sirolimus para reduzir o número de efeitos colaterais relacionados ao trato gastrintestinal.
