Abstract

método para seleção de células eucarióticas expressando uma proteína heteróloga. a presente invenção refere-se a um método para selecionar pelo menos uma célula hospedeira eucariótica expressando um produto de interesse, compreendendo: (a) proporcionar uma pluralidade de células hospedeiras eucarióticas, em que a viabilidade de célula das ditas células hospedeiras depende da captação de folato, em que as ditas células hospedeiras incluem pelo menos (i) um polinucleotídeo estrangeiro que codifica um produto de interesse e (ii) um polinucleotídeo estrangeiro que codifica uma enzima da dhfr; (b) cultivar a dita pluralidade de células hospedeiras eucarióticas em um meio de cultura seletivo, compreendendo pelos menos um inibidor de dhfr e folato em uma concentração limitante, e (c) selecionar pelo menos uma célula hospedeira eucariótica expressando o produto de interesse. também é fornecido um método para expressar um produto de interesse que é baseado em células hospedeiras selecionadas por meio do dito método e um meio de cultura de célula.
