Description
Verfahren zur Herstellung eines Insulinpräparates. Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung eines Insulin präparates. Das Verfahren ist dadurch ge kennzeichnet, dass ein In8U1111SalZ und ScOm- brin in saurer Lösung miteinander in Re aktion gebracht werden, wodurch Scombrin- insulinat gebildet wird.
Bei der Durchführung der Reaktion ist es vorteilhaft, die Gewichtsmenge des Prot- amins so zu bemessen, dass sie etwa '/s bis '/io der Gewichtsmenge des Insulinsalzes beträgt.
Zu bemerken ist, dass die Reaktion, wie sich zeigte, bereits bei gewöhnlicher Tem peratur stattfindet und sehr schnell vor sich geht. Dabei wird der saure Rest des Insulin salzes abgespalten.
Beispiel: 10 Gewichtsteile Insulin und 1,2 Ge wichtsteile Scombrin werden in saurer Lö sung miteinander in Reaktion gebracht. Der Säuregrad der Lösung wird grösser gewählt, als einem pH-Wert von zirka 4 entspricht, und die Reaktion wird bei gewöhnlicher Temperatur ausgeführt. Infolge des genann ten Säuregrades der Lösung verbleibt die bei der Reaktion gebildete, neue chemische Verbindung (Scombrininsulinat) in Lösung; sie kann aber durch Erhöhung des pH-Wer- tes ausgefällt werden. Das Produkt ist ein in Wasser schwer lösliches, weisses und amorphes Pulver, welches sich beim Er wärmen bei zirka 100 bis 120 0 0 zu zer setzen anfängt.
Die gebildete chemische Verbindung Prot- amininsulinat ist im pH-Wertgebiet 7 bis 7,5 sehr schwer löslich, während im Gegensatz hierzu beide Reaktionskomponenten bei diesen pH-Werten leicht löslich sind.
Die ganze Reaktionsmasse besteht nach Beendigung der rasch verlaufenden Reaktion aus einer Lösung von Protamininsulinat. Dieses Präparat wird zweckmässig in Am pullen eingefüllt, die je einen einer gewissen Anzahl internationaler Insulineinheiten ent- sprechenden Inhalt aufnehmen. Die Lösung sollte nicht als solche, sondern erst nach Fällung durch Zusatz eines alkalisierenden Stoffes benutzt werden.
Die durch Benutzung eines solchen In sulinpräparates entstandene Wirkung in dem menschlichen Organismus wird bedeutend langsamer verlaufen und infolgedessen auch bedeutend milder sein, als es bei Verwen dung von Insulinhydrochlorid der Fall ist, und dieser langsamere Wirkungsverlauf be wirkt zugleich, dass die Wiederholung einer Einspritzung erst nach einer verhältnismässig langen Zeitperiode stattzufinden braucht.
Process for the production of an insulin preparation. The subject of the present patent is a process for the production of an insulin preparation. The method is characterized in that an In8U1111SalZ and scombrin are brought into reaction with one another in acidic solution, whereby scombrin insulinate is formed.
When carrying out the reaction, it is advantageous to measure the amount by weight of the protamine in such a way that it amounts to about ½ to ½ of the amount by weight of the insulin salt.
It should be noted that the reaction, as has been shown, already takes place at normal temperature and is very rapid. The acidic remainder of the insulin salt is split off.
Example: 10 parts by weight of insulin and 1.2 parts by weight of scombrin are brought into reaction with one another in an acidic solution. The acidity of the solution is chosen to be greater than a pH value of about 4, and the reaction is carried out at ordinary temperature. As a result of the named acidity of the solution, the new chemical compound formed during the reaction (scombrin insulinate) remains in solution; however, it can be precipitated by increasing the pH value. The product is a white and amorphous powder which is sparingly soluble in water and which begins to set at around 100 to 120 0 0 when it is heated.
The chemical compound formed, protamine insulinate, is very sparingly soluble in the pH range from 7 to 7.5, while in contrast to this, both reaction components are easily soluble at these pH values.
After the rapid reaction has ended, the entire reaction mass consists of a solution of protamine insulinate. This preparation is expediently filled into ampoules which each hold a content corresponding to a certain number of international insulin units. The solution should not be used as such, but only after precipitation by adding an alkalizing substance.
The effect in the human organism resulting from the use of such an insulin preparation will be significantly slower and consequently also significantly milder than is the case with the use of insulin hydrochloride, and this slower course of action also means that an injection is not repeated until after needs to take place over a comparatively long period of time.
