Description
Verfahren zur Darstellung von m-Dimethogy-m-oay-p-diogy-fuchson. Im Hauptpatent konnte gezeigt werden, dass man 3,3',3"-Trirnetboxy-4',4"-dioxy-fuch- son in reiner kristallisierter Form darstellen kann; in analoger Weise lässt sich auch das m- Dimethoxy-m-oxy-p-dioxy-fuchson gewinnen.
Erfindungsgemäss wird dieses Produkt dadurch hergestellt, dass man das 3,3'-Di- methoxy-3"-4,4',4''-tetraoxy-tri.phenylmetharr einer Oxydation unterwirft. Man kann hierzu derart verfahren, dass man die Oxydation unmittelbar nach der Entstehung des Leuco- triphenylmethans ausführt, zum Beispiel in dem man die Darstellung des Triphenyl- methan-derivates in Gegenwart eines Oxy dationsmittels ausführt. Für die erfindungs gemässe Oxydation sind organische Peroxyde, wie Benzoylperoxyd, besonders geeignet, und es ist von Vorteil, Essigester oder ähn liche Ester ' als Lösungsmittel bei der Oxy dation zu verwenden.
Das Produkt des Ver fahrens neigt zur Bildung von Additionspro dukten; es kann zutun Beispiel mit Säuren, Basen, zahlreichen Metallsalzen, Bisulfiten und hydroxylhaltigen Stoffen Additionsver bindungen bilden, aus welchen das Fuchson leicht regenerierbar ist. Die Absonderung des Verfahrensproduktes aus dem Oxydations gemisch erfolgt am besten über seine Addi tionsprodukte mit Halogenwasserstoffsäuren.
Das Produkt wirkt antiseptisch und bat sich besonders bei bestimmten Formen der Tuberkulose als sehr wirksam erwiesen. <I>Beispiele:</I> 1. 32 g Benzoylperoxyd werden in 400 ems Essigester gelöst und 44 g 3,3'-Dimetboxy- 3"-4,4'-4"-tetraoxy-triphenylmetharr zugege ben. Man sättigt unter Eiskühlung mit Salz säure und lässt mehrere Tage unter häufigem Schütteln stehen. Das Chlorhydrat des m- Oxy-m-dimetboxy-p-dioxy-fuchsorrs kristalli siert in metallisch glänzenden Kristallen in einer Ausbeute von 25-30 g aus.
Zersetzungs punkt bei<B>1800,</B> Es löst sich in verdünntem Alkohol mit tiefroter Farbe auf, welche durch Alkalien in Blau umschlägt. Es bildet mit Aluminiumhydroxyd, Zinkoxyd, Bleiazetat etc. dunkelblaue Salze.
Das Fuchson kann aber nicht nur als Chlorhydrat, sondern auch als freie Base iso liert werden. Zu diesem Zwecke wird das aus dem Gemisch abgeschiedene Chlorhydrat mit wässeriger Natriumazetatlösung digeriert, abgenutscht, mit Wasser gewaschen und ge trocknet. Das kristallinische Fuchson schmilzt bei etwa 230 0.
2. 5 g Guajacol und 2,8 g Protocatechu- aldehyd werden in 20 ems 11 % Salzsäuregas enthaltendem Essigester gelöst und 7,5 g Ben- zoylperoxyd (aktiver Sauerstoffgehalt 5,2 %) zugegeben. Man sättigt unter Rühren und Kühlen mit Salzsäuregas. Das Benzoylper- oxyd geht nach längerer Zeit in Lösung; jetzt verdünnt man mit 60 cm-' Essigester.
Nach Stehen scheidet sich das m-Dimethoxy- m-oxy-p-dioxy-fuchson als Chlorhydrat in dunklen, metallisch glänzenden Kristallen aus. Das Produkt ist mit dem im Beispiel 1 be schriebenen Produkt identisch.
Zur Darstellung der Natriumbisulfit-Ad- ditionsverbindung des Fuchsons wird das freie Fuchson mit der berechneten Menge Natrium- bisulfit in wässeriger Lösung bis zur Auf lösung erwärmt, filtriert und im Vakuum vom Lösungsmittel befreit. Diese Additions verbindung löst sich mässig im kalten Wasser und die Lösung gibt mit Säuren eine tiefrote Färbung.
Das freie Fuchson wird aus dieser Additionsverbindungabgesehiedeu,indern man zum Beispiel die Additionsverbindung in ver- dünnter-Salzsäui#eaufschwemnit,dur-ch Kochen die Schwefelsäure verjagt und durch Zugabe von Natriumacetatlösung die Salzsäure ent fernt. Das in Wasser schwer lösliche Produkt wird durch Filtrieren und Auswaschen mit Wasser und Trocknen gewonnen. Das Pro dukt des Verfahrens ist eine neue Verbindung.

The process product is best separated from the oxidation mixture via its addi tion products with hydrohalic acids.

The free Fuchson is separated from this addition compound, for example by flooding the addition compound in dilute hydrochloric acid, by boiling the sulfuric acid and removing the hydrochloric acid by adding sodium acetate solution. The product, which is sparingly soluble in water, is obtained by filtering and washing with water and drying. The product of the process is a new compound.
