Verfahren zur Herstellung eines Gemisches von racemischem Ephedrin und racemischem Pseudoephedrin. Gegenstand der Erfindung ist ein Ver fahren zur Herstellung eines Gemisches von racemischem Ephedrin und racemischem Pseudoephedrin, das dadurch gekennzeichnet ist, dass man d-Ephedrin bei höherer Tem peratur mit Alkalialkoholat behandelt.
Die Racemisierung des d-Ephedrins er folgt zu dem Zweck der Verwertung des therapeutisch nicht verwendbaren d-Ephe- drins, das bei der Gewinnung von 1-Ephe- drin, aus,der synthetisch erhaltenen Racem.- verbindung anfällt. Aus dem erfindungs gemäss erhältlichen Racemgemisch kann wie der 1-Ephedrin durch Spaltung gewonnen werden.
Es sind in der Literatur zahlreiche Ver suche erwähnt, um optisch aktives Ephedrin zu racemisieren. So versuchte Schmidt (ver gleiche Archiv der Pharmazie 244, S. 239 bis 240) 1-Ephedrin durch Erhitzen mit der zehnfachen Menge Salzsäure zu inaktivie ren. Er erhielt dabei jedoch rechtsdrehendes Pseudo-Ephedrin (d-ps-Ephedrin). Dieser Befund wurde von Emde in der Zeitschrift Archiv der Pharmazie 244, S. 244, bestätigt. Weitere Racemisierungsversuche beschreibt Schmidt im Archiv der Pharmazie 250, S. 154 ff.
Bei der Einwirkung von Baryt bei Temperaturen bis, 210' fand nur eine Zer setzung, dagegen keine Inaktivierung von 1-Ephedrin statt. Dagegen wurde angege ben, dass d-ps-Ephedrin bei der Einwirkung von Baryt 1-Ephedrin geben soll. Allerdings konnte diese Angabe von Spaeth und Göh- ring (Monatshefte für Chemie, 41, 319 f.) nicht bestätigt werden.
Weiterhin. hat sich gezeigt, dass 1-Ephe- drin durch alkoholische Kalilauge nicht ver ändert wird. Durch Behandlung mit ver dünnter Schwefelsäure wurde es in d-ps- Ephedrin übergeführt.
1-Ephedrin und d-ps- Ephedrin ergaben mit Essigsäureanhydrid behandelt d-ps-Acetylephedrin. Durch Be handlung mit salpetriger- Säure erhält man aus 1-Ephedrin d-ps-Ephedrin. Bei keinem der vorstehend erwähnten Versuche konnte Inaktivierung beobachtet. werden.
Schliesslich sei noch auf die Versuche von Späth, optisch aktives Ephedrin bezw. ps- Ephedrin zu racemisieren, in den Berichten der Deutschen Chemischen Gesellschaft 58, S. 197 und S. 1268 hingewiesen, die eben falls erfolglos verliefen.
Die für die Racemisierung nach dem Verfahren der Erfindung geeigneten Alkali alkoholate können sowohl Natrium- als auch Kaliumverbindungen sein. Als Alkohole kommen Methyl-, Äthyl-, Propyl-, Butyl-, Amyl- und auch aromatische Alkohole, bei spielsweise Benzylalkohol, und ferner cyklo- aliphatische Alkohole, wie z. B. Cyklohega- nol, in Betracht.
Um nach erfolgter Um setzung ohne Schwierigkeiten den über schüssigen Alkohol abdestillieren zu können, werden zweckmässigerweise Alkoholate sol cher Alkohole verwendet, deren Siedepunkt nicht allzu hoch und vorzugsweise bei 15 mm Hg-Druek unterhalb 167-168 C liegt.
In diesem Fall ist das Abdestillieren des Alko hols ohne Ephedrinverlust möglich, da Ephe- drin bei 15 mm HK-Druck erst bei 167 bis <B>168'</B> siedet. Selbstverständlich können auch Mischungen von Alkalialkoholaten von ver schiedenen Alkoholen Verwendung finden.
Die für die Racemisierung verwendete Alkalialkoholatmenge kann sehr gering sein. Sie beeinflusst die Umlagerungstemperatur und die Umlagerungszeit. Die benötigten Mengen Alkalialkoholat können von 1/,o bis 2 Mol variieren. Je mehr Alkalialkoholat verwendet wird und je höher die Tempera tur ist, desto schneller geht die Umlagerung vor sieh, desto grösser ist aber auch die Ge fahr von Verlusten durch Zersetzung des Ausgangsmaterials.
Die Durchführung der Racemisierung kann im Sehmelzfluss, d. h. ohne Zusatz von Lösungsmittel, erfolgen. Vorteilhafter ist je doch die Durchführung der Raeemisierung in Lösung, da diese Arbeitsweise eine bessere Regulierung der Reaktion erlaubt. Als Lö sungsmittel kommen vor allem indifferente Kohlenwasserstoffe in Betracht, und zwar vorzugsweise solche, die einen genügend hohen, beispielsweise über 140' gelegenen Siedepunkt aufweisen.
Als derartige Kohlen- wasserstoffe seien beispielsweise erwähnt Dekahydronaphthalin, Tetrahydronaphtha- lin, Naphthalin, Cymol, hochsiedende Ben zol- und Benzin-Kohlenwasserstoffe etc. Als Lösungsmittel bei der Racemisierung sind aber auch hoch siedende Alkohole, wie z. B. Benzylalkohol etc. geeignet.
Die Temperaturgrenzen, innerhalb derer die Umlagerung durchgeführt werden kann, liegen etwa bei 135 bis 210'. Als am gün stigsten haben sich Temperaturen von 170 bis<B>190'</B> erwiesen. Beispiele: 1. 1 g Natrium wird in 20 cm' Methanol gelöst und die Lösung mit 500 em@ Deka- hydronaphthalin (Dekalin) und 60 g d- Ephedrin versetzt.
Anschliessend destilliert man 120 cm' Dekalin bei gewöhnlichem Druck ab und kocht den Kolbeninhalt vier Stunden bei einer Innentemperatur von zirka 195 . Dann wird abgekühlt und die Basen werden mit verdünnter Schwefelsäure aus der Dekalinlösung ausgeschüttelt.
Die Schwe- felsäurelösung wird stark alkalisch gemacht und die ausfallenden, racemischen Basen mit Äther aufgenommen. Wird der Ätherrück stand mit Salzsäure neutral gelöst und mit Kaliumogalatlösung versetzt, so kristallisiert das Ozalat des d-1-Ephedrins aus. Aus der Mutterlauge fällt mit Kalilauge 42% d-1 Ephedrin aus. Der Rest ist d-1-ps-Ephedrin in einer Ausbeute von<B>53%.</B>
2. 1 g Natrium wird in 20 cm' Methanol gelöst und die Lösung mit 30 g d-Ephedrin versetzt. Dann wird der Methylalkohol ab destilliert und die klare Schmelze zwei Stun den auf<B>190'</B> erhitzt. Löst man die Schmelze in Salzsäure neutral auf und trennt man, wie in Beispiel 1 über das Ozalat, so erhält man d-l-Ephedrin in einer Ausbeute von 45%, d-1-ps-Ephedrin in einer Ausbeute von 49 %.
3. 40 g metallisches Natrium wurden in 1800 cm' Amylalkohol gelöst. Anschliessend ,wurden 1000 cm3 Amylalkohol abdestilliert. Zur konzentrierten Natrium-amylatlösung wurden nun. 80 g d-Ephedrinbase hinzugege ben und das Ganze 42 Stunden gekocht. Nach dem Abkühlen wurden die entstan denen Basen mit Schwefelsäure ausgeschüt telt.
Isoliert man aus der schwefelsauren Lösung die Basen durch Fällen mit Natron lauge und Ausschütteln mit Äther und trennt die so erhaltenen Basen wie in Beispiel 1 über das Ogalat, so erhält man 59 g d-1 Ephedrin-Ogalat und 15 g d-1-ps-Ephedrin- ba,se.

The racemization of d-ephedrine is carried out for the purpose of utilizing the therapeutically unsuitable d-ephe- drine which is obtained from the synthetically obtained racemic compound when 1-ephe- drine is obtained. There are numerous attempts mentioned in the literature to racemize optically active ephedrine. Schmidt attempted to inactivate 1-ephedrine by heating it with ten times the amount of hydrochloric acid. However, he received dextrorotatory pseudo-ephedrine (d-ps-ephedrine). This finding was confirmed by Emde in the journal Archiv der Pharmazie 244, p. 244. Schmidt describes further attempts at racemization in the Archiv der Pharmazie 250, p. 154 ff.
When barite was exposed to temperatures up to '210', 1-ephedrine was only decomposed, but not inactivated. On the other hand, it was stated that d-ps-ephedrine should give 1-ephedrine when exposed to barite. However, this statement could not be confirmed by Spaeth and Göhring (monthly magazine for chemistry, 41, 319 f.).
Farther. it has been shown that 1-ephorin is not changed by alcoholic potassium hydroxide solution. It was converted into d-ps-ephedrine by treatment with dilute sulfuric acid.
1-ephedrine and d-ps-ephedrine treated with acetic anhydride gave d-ps-acetylephedrine. Treatment with nitrous acid gives 1-ephedrine d-ps-ephedrine. Inactivation was not observed in any of the experiments mentioned above. will.
Finally, the attempts by Späth to produce optically active ephedrine or To racemize ps-ephedrine, pointed out in the reports of the German Chemical Society 58, p. 197 and p. 1268, which were also unsuccessful.
The alkali metal alcoholates suitable for racemization by the process of the invention can be both sodium and potassium compounds. The alcohols are methyl, ethyl, propyl, butyl, amyl and aromatic alcohols, for example benzyl alcohol, and also cyclo-aliphatic alcohols, such as. B. Cyclohega- nol, into consideration.
In order to be able to distill off the excess alcohol without difficulty after the reaction has taken place, alcoholates of such alcohols are expediently used whose boiling point is not too high and preferably below 167-168 ° C. at 15 mm Hg.
In this case, the alcohol can be distilled off without loss of ephedrine, since ephe- drine only boils at 167 to 168 'at a pressure of 15 mm. Mixtures of alkali metal alcoholates of different alcohols can of course also be used.
The amount of alkali metal alcoholate used for the racemization can be very small. It influences the relocation temperature and the relocation time. The required amounts of alkali metal alcoholate can vary from 1 /, o to 2 mol. The more alkali alcoholate is used and the higher the tempera ture, the faster the rearrangement takes place, but the greater the risk of losses due to decomposition of the starting material.
The racemization can be carried out in the Sehmelzfluss, d. H. without the addition of solvents. However, it is more advantageous to carry out the reaction in solution, since this mode of operation allows better regulation of the reaction. In particular, inert hydrocarbons are suitable as solvents, preferably those which have a sufficiently high boiling point, for example above 140 °.
Examples of such hydrocarbons are decahydronaphthalene, tetrahydronaphthalene, naphthalene, cymene, high-boiling benzene and gasoline hydrocarbons, etc. High-boiling alcohols, such as, for example, alcohols, are also used as solvents in the racemization. B. benzyl alcohol, etc. suitable.
The temperature limits within which the rearrangement can be carried out are approximately 135 to 210 °. Temperatures of 170 to <B> 190 '</B> have proven to be the most favorable. Examples: 1. 1 g of sodium is dissolved in 20 cm 'of methanol and 500 em of decahydronaphthalene (decalin) and 60 g of d-ephedrine are added to the solution.
120 cm of decalin is then distilled off at normal pressure and the contents of the flask are boiled for four hours at an internal temperature of about 195. It is then cooled and the bases are shaken out of the decalin solution with dilute sulfuric acid.
The sulfuric acid solution is made strongly alkaline and the precipitating, racemic bases are taken up with ether. If the ether residue is dissolved neutrally with hydrochloric acid and a solution of potassium galate is added, the ozalate of d-1-ephedrine crystallizes out. 42% d-1 ephedrine is precipitated from the mother liquor with potassium hydroxide. The remainder is d-1-ps-ephedrine in <B> 53% yield. </B>
2. 1 g of sodium is dissolved in 20 cm 'of methanol and 30 g of d-ephedrine are added to the solution. Then the methyl alcohol is distilled off and the clear melt is heated to <B> 190 '</B> for two hours. If the melt is dissolved neutrally in hydrochloric acid and separated via the ozalate as in Example 1, d-l-ephedrine is obtained in a yield of 45%, d-1-ps-ephedrine in a yield of 49%.
3. 40 g of metallic sodium were dissolved in 1800 cm 'of amyl alcohol. Then 1000 cm3 of amyl alcohol were distilled off. The concentrated sodium amylate solution was now. 80 g of d-ephedrine base are added and the whole is cooked for 42 hours. After cooling, the bases formed were extracted with sulfuric acid.
