Verfahren zur Trennung von DL-threo-ss-Phenyl-serin in die optischen Antipoden. DL-ss-Phenyl-serin lässt sich nach den An gaben von Erlenmeyer (Liebigs Annalen der Chemie, Bd. 284, S. 41) aus Benzaldehyd und Glykokoll synthetisieren. Optisch aktives Pheny l-serin, von welchem nach Theorie zwei Antipodenpaare existieren, ist bis jetzt nicht bekannt.
Es wurde gefunden, dass das nach der ErlenmeyerschenSynthese erhaltene ss-Phenyl- serin vorwiegend, zum mindesten zu 90%, nur aus einer DL-Form, nämlich der threo-DL- Form, besteht, so dass dessen Spaltung in die optischen Isomeren vorwiegend nur zwei Anti poden liefert.
Gemäss der vorliegenden Erfindung kann DL-threo-ss-Pheny 1-serin dadurch in seine opti schen Antipoden getrennt werden, dass man die genannte Verbindung verestert, den erhal tenen Ester an der Aminogruppe acyliert, z. B. durch Einwirkung von Acetanhydrid, die erhaltene Verbindung durch Verseifung über führt in DL-threo-N-Aeyl-ss-phenyl-serin, auf letzteres eine optisch aktive Base, z. B. Chi nin, einwirken lässt, die entstandenen dia- stereomeren Salze aus einem geeigneten Lö sungsmittel, z. B.
Wasser, fraktioniert kristalli siert, das zuerst auskristallisierende Diastereo- mere zerlegt, den entstandenen N-Acyl-ss- phenyl-serin-antipoden von der Base trennt, ersteren entacyliert und den gebildeten Phenyl-serin-antipoden isoliert, das nach Aus seheidung des ersten Diastereomeren in der Mutterlauge vorhandene zweite diastereomere Salz zerlegt, im Reaktionsgemische das N- Acyl-ss-phenyl-serin von der Base trennt, ersteres isoliert,
den zweiten threo-N-Acyl-fl- phenyl-serin-antipoden durch Umkristallisie- ren aus einem geeigneten Lösungsmittel, z. B. Essigester/Äthanol 1 :1, von Begleitsubstanzen trennt, anschliessend entacyliert und den zwei ten ss-Phenyl-serin-antipoden isoliert.
Ob man bei der Durchführung dieses Verfahrens zu erst den links- oder den rechtsdrehenden Anti poden erhält, ist abhängig von der Natur der verwendeten optisch aktiven Base; durch letz tere sowie durch die Natur der bei der N- Acy lierung verwendeten Acylierungsmittel wird auch der Drehsinn der optisch aktiven Zwischenprodukte bestimmt.
Die Veresterung des DL-threo-ss-Phenyl- serins kann mit einem für Aminosäuren üblichen Veresterungsmittel durchgeführt werden. Ebenso kann die Acylierung der Aminogruppe nach einem üblichen Verfah ren erfolgen, z.
B. durch die Einwir kung von Acetanhydrid. Die Ver seifung des erhaltenen N-Acyl-phenyl-serin-esters wird man selbstverständlich in der Weise leiten, dass nur eine Hydrolyse der Estergruppe, nicht aber auch eine Spaltung der Acylamino- gruppe erfolgt;
zweckmässigerweise geschieht dies durch Behandlung mit der berechneten Menge eines wässrigen Alkalis, z. B. 10 % iger Natronlauge.
Das DL-threo-N-Acyl=ss-phenyl-serin wird, wie bereits gesagt, durch Einwirkung einer optisch aktiven Base in ein Salz übergeführt. Es können zu diesem Zwecke verschiedene optisch aktive Basen, wie sie für die Tren nung von Racematen gebräuchlich sind, Ver wendung finden; mit. Vorteil lässt man Chinin auf eine wässrige Lösung des Aminosäurederi- vates einwirken. Hierbei bilden sich diastereo inere Salze der optischen Antipoden des Amino- säurederivates, welche durch fraktionierte Kristallisation voneinander getrennt werden.
Für die fraktionierte Kristallisation wird man ein Lösungsmittel wählen, in welchem die beiden diastereomeren Salze in verschiedenem Grade löslieh sind; für die Chininsalze. z. B. eignet sieh Nasser.
Die Einzelheiten bei der Aufarbeitung der optischen Antipoden der Aminosäurederivate und Überführung dersel ben in (-)- bzw. (+)-threo-ss-Phenyl-serin richten sich nach der Art der Base bzw. des N-Acylrestes; falls Chinin als optisch aktive Base verwendet wird, kann man beispielsweise wie folgt vorgehen:
Das zuerst. auskristallisierende diastereo- mere Chininsalz, welches, wenn es am Stiek- stoff der Aminosäure acetyliert ist, einen Zer setzungspunkt 153 bis 156 aufweist und eine optische Aktivität von [a]D = -106 (c = 2 in Methanol) zeigt, wird in 50 % igem Alkohol gelöst.
Durch Zusatz der berechneten Menge wässrigen Alkalis wird das Chinin in Freiheit gesetzt; es fällt bei Konzentrierung der Lö sung, insbesondere durch Entfernung des Al kohols im Vakuum, fast vollständig aus; in Lösung verbleibende Chininreste können aus der wässrigen Lösung mit Chloroform extra hiert werden.
Die nunmehr erhaltene Lösung des Alkalisalzes des N-Acyl-ss-phenyl-serin- antipoden wird nun zweckmässigerweise auf ein geringes Volumen unter vermindertem Druck eingedampft, worauf man den threo-N- Aeyl-ss-phenyl-serin-antipoden mit Salzsäure ausfällen kann.
Wenn man als N-Ae3Tl-deri- vate die Acetylderivate hergestellt hat, erhält man auf diese Weise (+) -threo-I\T-Acet.@-1-ss- phenyl-serin,welches bei 163 bis 164 .C unter Zersetzung schmilzt und eine optische Aktivi tät von [a] 20 = + 16,3 (e = 2 in lllethanol) aufweist. Der genannte Antipode kann da durch entacyliert werden, dass man denselben mit wässriger Mineralsäure, z.
B. 3 n-Salz- säure, erwärmt; man kann aus dem Reaktions- gemisehe die Aminosäure in an sich bekannter Weise isolieren. Letztere kann beispielsweise dadurch erfolgen, dass man die entstandene Lösung eindampft, den Rückstand in wenig Wasser aufnimmt, die Lösung mit Alkohol verdünnt und daraus das (-)-threo-ss-Phenyl- serin durch Neutralisieren der Lösung, z. B.
mit Ammoniak, ausfällt. (-)-tlireo-ss-Phenyl- serin ist charakterisiert durch einen Zerset zungspunkt bei etwa<B>195</B> bis 1980C und eine optische Aktivität von [a] D = -49 (e = 2 in 6 n-HCl).
Die Aufarbeitung des diastereomeren Chi- ninsalzes kann auch mittels Salzsäure bewerk stelligt werden; es bildet sieh dann Chinin- hydrochlorid und threo-N-Acyl-ss-phenyl-serin, welch letzteres anschliessend verseift wird, wo bei man z. B. threo-ss-Plienyl-serin-hy droehlo- rid erhält.
Durch Neutralisation einer alko holischen Lösung des letzteren mit einer Base, wie Ammoniak oder Pyridin, kann man dann das (-)-thi-eo-ss-Phenyl-serin ausfällen.
Das nach Ausscheidung des schwerer lös lichen diastereomeren Chininsalzes in der Mutterlauge verbleibende zweite diastereomere Chininsalz kann in analoger Weise zerlegt werden wie die erstausgefallene Fraktion. Falls mit Alkali zerlegt werden will, empfiehlt es sich, die Lösung vor dem Zusatz des Al kalis auf etwa das halbe Volumen zu konzen trieren.
Der nach der Ausscheidung des Chi nins erhaltene threo-N-Acz-1-ss-phenyl-serin- antipode (die Acetylverbindung ist. links drehend) wird zweckmässig von ihn begleiten den Stereoisomeren (vor allem Reste der DL- Form) getrennt. Dies kann durch einmaliges Umkristallisieren aus einem geeigneten Lö sungsmittel, z.
B. einem 1 :1- Geiuiseh von Essigester und Äthanol, gesehehen. Falls zur N-Acylierung Essigsäure verwendet worden ist, erhält man auf diese Weise (-)-tlireo-N- Acetyl-ss-pheny 1-serin vom Schmelzpunkt 7.63 bis 165 C und einer optischen Aktivität. von [a] D = -17,1 (c = 2 in Methanol).
Die I:ntacylierung des so erhaltenen threo-N-Aeyl- ss-phen5#1-serin-antipoden und die Isolierung des (+ )-threo-ss-Phenyl-serins können in glei- eher Weise erfolgen, wie oben für die erste Fraktion beschrieben. Die so erhaltene Ver bindung zersetzt sich bei 195 bis 197 C und zeigt eine optische Aktivität von [a] D = + 49 (c = 2 in 6 n-HCl). Bei Verwendung anderer optisch aktiver Basen kann die Aufarbeitung der bei der Kristallisation gewonnenen Frak tionen auf ähnliche Weise durchgeführt wer den.
Die tlii-eo-ss-Phenyl-serin-antipoden sind wertvoll als Zwischenprodukte für den Auf bau von Chloramplienicol bzw. von Analogen desselben.
<I>Beispiel:</I> 100 g DL-tlireo-ss-Phenyl-serin werden in <B>100</B> em3 absolutem Äthylalkohol suspendiert, worauf ein kräftiger Strom von Salzsäuregas bis zum Sieden des Alkohols eingeleitet wird. Nun kühlt man das Reaktionsgeiniseh mit Eis und leitet bis zur Sättigung Salzsäuregas ein. Nach 24stündigem Kühlen wird die alkoholi sche Salzsäure im Vakuum auf dem Dampf bad entfernt.
Als Rückstand bleibt das Hy- drochlorid des DL-tltreo-ss-Phenyl-serin-ätliyl- esters in Form eines Kristallkuellens. Das aus Alkohol/Äther umkristallisierte Hydrochlorid schmilzt bei 140 bis 143 C. Der freie Ester kann aus der wässrigen Lösung des Hydro- chlorids durch Zugabe von Ammoniak ausge fällt werden; nach Umkristallisation aus Essig ester schmilzt die Verbindung bei 82 bis 84 C.
300g DL-threo-ss-Phenyl-serin-äthylester werden durch Erwärmen auf dem Dampfbad in 600 ein3 Eisessig gelöst. Die auf Zimmer temperatur abgekühlte Lösung wird mit 177 cm3 Essigsäureanhydrid (1,1 141o1) ver setzt. Durch Kühlen mit Eiswasser wird die Temperatur unter 40 C gehalten. Nach Be endigung der Reaktion wird die Reaktions- inisehung kühl gestellt, wobei das DL-threo-N- Acetylderivat auskristallisiert. Sodann wird abgesaugt und bis zum Verschwinden des Essigsäuregeruches mit Wasser gewaschen.
Darauf wird bei 95 C getrocknet. Ausbeute 305g. Schmelzpunkt. 172 bis 176 C (unkorr.). Die Verbindung kann aus Alkohol umkristalli siert werden, wodurch der Schmelzpunkt auf 179 bis 180 C (unkorr.) steigt.
305g DL-threo-N-Acetyl-ss-phenyl-serin- äthyfester werden mit 487 cm3 10 % iger Na- tronlauge (1141o1) versetzt und unter gelegent lichem Schütteln während 30 Minuten auf dem Dampfbad erhitzt.
Dabei löst sich der grösste Teil; vom geringen Rest wird abfil- triert. Die Lösung wird auf dem Dampfbad unter vermindertem Druck bis zur beginnen den Kristallisation des Natriumsalzes ein geengt. Das Ausgefallene wird durch gelin des Erwärmen wieder in Lösung gebracht und diese mit konzentrierter Salzsäure kongosauer gemacht. Nach 2stündigem Kühlen wird das auskristallisierte DL-threo-N-Acetyl-ss-phenyl- serin abgenutscht. Ausbeute: 250g. Schmelz punkt 142 bis 146 C (unkorr.) unter Zerset zung.
Nach Umkristallisieren aus Essigester/ Methanol steigt der Schmelzpunkt auf 148 bis 150 C (unkorr.) unter Zersetzung.
112 g DL-threo-N-Acetyl-ss-phenyl-serin werden in 500 cm3 heissem Wasser gelöst und zu einer Suspension von 162g Chinin in 2 Liter Wasser von 90 C gegeben. Nach kräf tigem Schütteln tritt Lösung ein. Vom gerin gen Rückstand wird abfiltriert und die Lösung bei Zimmertemperatur stehengelassen. Dabei kristallisiert das Chininsalz vom (+ )-threo-N- Acetyl-ss-phenyl-serin in farblosen Prismen (schwerer lösliches Salz I). Nach 48 Stunden wird abgesaugt, der Rückstand mit wenig kal tem Wasser gewaschen und getrocknet. Aus beute: 128 g.
Schmelzpunkt 154 bis 156 C (un- korr.) unter Zersetzung. [a]D = -107,5 (c = 2 in Methanol).
Das schwerer lösliche Chininsalz I wird in etwa 700 em3 50 % igem Alkohol gelöst und mit. einer Lösung von 9,4 g Ätznatron in 1.00 cm3 Wasser versetzt.
Sodann wird der grösste Teil des Alkohols durch Abdestillieren unter vermindertem Druck entfernt und vom ausgefallenen Chinin abfiltriert. Die Lösung wird zweimal mit je 150 em3 Chloroform ex trahiert, um die letzten Reste des Chinins zu entfernen, und dann unter vermindertem Druck auf ein Volumen von etwa 1.00 cm3 ein geengt.. Dann wird das optisch aktive (+ ) - threo-N-Acetyl-ss-plienyl-serin mit konzentrier ter Salzsäure ausgefällt.
Dieses Zwischenpro- dukt besitzt einen Schmelzpunkt von 163 bis 164 C (unkorr.) unter Zersetzung und einen Drehwert von [a] D = + 16,3 (c = 2 in Me- thanol)
. Durch 2stündiges Erhitzen mit etwa 150 em3 3 n-Salzsäure wird entacetyliert. Dann wird im Vakuum zur Trockne eingedampft, zweimal mit je 50 em3 Wasser versetzt. und jedesmal wieder zur Trockne eingedampft. Der Rückstand wird in möglichst wenig Wasser gelöst, mit ungefähr dem 4fachen Volumen Alkohol versetzt und mit konzentriertem Am moniak neutralisiert. Dabei kristallisiert die Aminosäure aus und kann durch Filtration isoliert werden.
Die Verbindung besitzt einen Drehwert von [a] D = -46,0 (e = 2 in 6 n- Salzsäure) und einen Zersetzungspunkt von 195 bis 198 C (unkorr.). Das (-)-threo-ss- Phenyl-serin kann von noch beigemengtem Ammoniumehlorid durch Umkristallisieren aus Wasser befreit und rein erhalten werden. Der Drehwert steigt dann auf [a]"' = -49 (e = 2 in 6 n-Salzsäure).
Die Mutterlauge, aus welcher das schwerer lösliche Salz I auskristallisiert wurde, wird unter vermindertem Druck auf etwa die Hälfte des ursprünglichen Volumens ein geengt, und eine allfällige Ausscheidung wird auf dem Wasserbad wieder gelöst. Die wäss- rige Lösung wird mit 10,5 g Ätznatron in <B>100</B> ein- Wasser versetzt. Dabei bildet sieh das Natriumsalz von threo-N-Acetyl-ss-phenyl- serin, und die Chininbase fällt aus.
Von letz terer wird abfiltriert und die Lösung zur Ent fernung der letzten Chininmengen zweimal mit je 150 em3 Chloroform extrahiert. Dann wird die Lösung unter vermindertem Druck bis auf etwa 120 em3 eingeengt. Dabei beginnt das Natriumsalz auszufallen. Sodann wird letz teres durch leichtes Erwärmen wieder gelöst, die Lösung mit konzentrierter Salzsäure kongo sauer gemacht und über Nacht gekühlt. Am andern Tag wird das auskristallisierte rohe 1,hreo-N-Acetyl-ss-phenyl-serin abgesaugt und getrocknet.
Schmelzpunkt 150 bis 151 C (un- korr.) unter Zersetzung; [a] D = -13,2 (c = 2 in Methanol). Dieses Material enthält noch etwas Kochsalz und ist optisch noch un rein. Es wird zur Reinigung aus etwa 200 cm3 einer Mischung aus Essigester/Methanol <B>1:1</B> umkristallisiert und Meist dann einen Dreh wert von [a] D = -17,1 (c = 2 in Methanol) und einen Schmelzpunkt von 163 bis 165 C (unkorr.) auf.
Aus dieser Verbindung wird in analoger Weise, wie beim (-)-threo-ss-Phe- nyl-serin beschrieben, die Aminosäure durch Entacetylieren und Neutralisieren gewonnen. [a] D = +45,2 (c = 2 in 6 n-Salzsäure); Zersetzungspunkt 195 bis 197 C. Das reine (+) -threo-ss-Phenyl-serin besitzt nach einmali gem Umkristallisieren aus Wasser einen Dreh wert von [a120 = +49 (e = 2 in 6 n-Salz- säure).

Whether you first get the left-handed or right-handed anti-pods when performing this process depends on the nature of the optically active base used; the sense of rotation of the optically active intermediates is also determined by the latter and by the nature of the acylating agents used in the N acylation.
The esterification of DL-threo-ss-phenylserine can be carried out with an esterification agent customary for amino acids. Likewise, the acylation of the amino group can be carried out according to a customary process, e.g.
B. through the action of acetic anhydride. The saponification of the N-acyl-phenyl-serine ester obtained will of course be conducted in such a way that only hydrolysis of the ester group takes place, but not also cleavage of the acylamino group;

The DL-threo-N-acyl = ss-phenyl-serine is, as already mentioned, converted into a salt by the action of an optically active base. For this purpose, various optically active bases, such as those commonly used for the separation of racemates, can be used; With. It is advantageous to allow quinine to act on an aqueous solution of the amino acid derivative. Here, diastereomeric salts of the optical antipodes of the amino acid derivative form, which are separated from one another by fractional crystallization.

The details of working up the optical antipodes of the amino acid derivatives and converting them into (-) - or (+) - threo-ss-phenyl-serine depend on the type of base or N-acyl radical; if quinine is used as the optically active base, one can proceed as follows, for example:
That first. crystallizing diastereomeric quinine salt which, when acetylated on the nitrogen of the amino acid, has a decomposition point 153 to 156 and shows an optical activity of [a] D = -106 (c = 2 in methanol), is shown in 50 % alcohol dissolved.
The quinine is set free by adding the calculated amount of aqueous alkali; it falls when the solution is concentrated, in particular by removing the alcohol in vacuo, almost completely; Quinine residues remaining in solution can be extracted from the aqueous solution with chloroform.
The solution of the alkali salt of the N-acyl-ss-phenyl-serine antipode now obtained is expediently evaporated to a small volume under reduced pressure, whereupon the threo-N-aeyl-ss-phenyl-serine antipode can be precipitated with hydrochloric acid .

The diastereomeric quinine salt can also be worked up using hydrochloric acid; It then forms quinine hydrochloride and threo-N-acyl-ss-phenyl-serine, which the latter is then saponified, where one z. B. threo-ss-plienyl-serine-hy droehlo- rid receives.
By neutralizing an alcoholic solution of the latter with a base such as ammonia or pyridine, the (-) - thi-eo-ss-phenyl-serine can then be precipitated.
The second diastereomeric quinine salt remaining in the mother liquor after the elimination of the more difficultly soluble diastereomeric quinine salt can be broken down in a manner analogous to that of the first precipitated fraction. If you want to decompose with alkali, it is advisable to concentrate the solution to about half the volume before adding the alkali.
The threo-N-Acz-1-ss-phenyl-serine antipode (the acetyl compound is anticlockwise) obtained after excretion of the chinine is expediently separated from the stereoisomers accompanying it (especially residues of the DL form). This can be achieved by recrystallizing once from a suitable solvent such.

The tlii-eo-ss-phenyl-serine antipodes are valuable as intermediates for the construction of chloramplienicol or its analogues.

The free ester can be precipitated from the aqueous solution of the hydrochloride by adding ammonia; After recrystallization from ethyl acetate, the compound melts at 82 to 84 C.
300g DL-threo-ss-phenyl-serine-ethyl ester are dissolved in 600 ein3 glacial acetic acid by heating on the steam bath. The solution, cooled to room temperature, is mixed with 177 cm3 of acetic anhydride (1.1 141o1). The temperature is kept below 40 ° C. by cooling with ice water. 112 g of DL-threo-N-acetyl-ss-phenyl-serine are dissolved in 500 cm3 of hot water and added to a suspension of 162 g of quinine in 2 liters of water at 90 ° C. After vigorous shaking, solution occurs. The small residue is filtered off and the solution is left to stand at room temperature. The quinine salt crystallizes from the (+) -threo-N-acetyl-ss-phenyl-serine in colorless prisms (less soluble salt I). After 48 hours it is filtered off with suction, the residue is washed with a little cold water and dried. From booty: 128 g.

The less soluble quinine salt I is dissolved in about 700 em3 50% alcohol and mixed with. a solution of 9.4 g caustic soda in 1.00 cm3 water.
Most of the alcohol is then removed by distilling off under reduced pressure and the quinine which has precipitated out is filtered off. The solution is extracted twice with 150 cubic meters of chloroform each time in order to remove the last remains of the quinine, and then concentrated under reduced pressure to a volume of about 1.00 cm3. The (-) - threo-ss-phenyl-serine can be freed from any added ammonium chloride by recrystallization from water and obtained in pure form. The mother liquor, from which the sparingly soluble salt I was crystallized, is concentrated under reduced pressure to about half its original volume, and any precipitate is redissolved on the water bath.
