L'ester @p-hydroxyaminopropionique (ester (le la. 7E,-phényl-sérine) peut être employé sous forme (le base libre ou sous forme d'un sel.
Quand on utilise un sel, on ajoute au mélange réactionnel un équivalent d'une substance ba sique telle que l'ammoniaque aqueuse, le bi- earbonate ou l'hydroxyde de sodium. pour libérer in situ l'ester y-hydroxyaminopropio- nique sous forme de base. On peut utiliser comme milieu réactionnel une grande variété de solvants tels que l'eau, les alcools aliphati ques inférieurs soit aqueux, soit anhydres, le chloroforme, l'éther, etc.
La réduction de la y)-4-carboalcoxy-42- oxazoline en ip-4-hydroxyméthyl-42-oxazoline correspondante (étape B) peut être effectuée soit catalytiquement à l'aide d'hydrogène ga zeux en présence d'un catalyseur métallique d'hydrogénation, soit au moyen de l'hydrure de lithium et d'aluminium (LiAIH4). Cette dernière méthode de réduction est exécutée dans un solvant organique pratiquement an hydre et exempt de groupes hydroxyle, tel que l'éther, le benzène, le dioxane, le toluène, le tétrachlorure de carbone ou le tétrahydro- furane. Après l'achèvement de la réaction,
on décompose l'hydrure d'aluminium et de lithium en excès, par adjonction d'eau et on isole la vp-4-hydroxyméthyl-d2-oxazoline du mélange réactionnel. La quantité du LiAlH4 employée pour la réduction peut varier dans des limites assez larges. Il est toutefois préférable d'em ployer une quantité légèrement plus grande que la quantité théoriquement nécessaire.
La transformation de la. V-4-hydroxy- méthyl-42-oxazoline en yr4-acyloxyméthyl-d2- oxazoline (étape C) peut être effectuée en traitant la yp-4-hydroxyméthyl-J2-oxazoline à l'aide d'un halogénure ou d'un anhydride d'un acide carboxylique.
De préférence, on effectue cette réaction dans des conditions pratique ment anhydres en utilisant un excès d'agent d'acylation et un catalyseur basique tel que la pyridine, la triméthylamine, la diméthyl- aniline, la N-étbylpipéridine ou une amine tertiaire analogue. L'acylation peut aussi se faire dans un milieu réactionnel aqueux en employant un catalyseur basique tel qu'un hydroxyde ou un carbonate d'un métal alca lin, ou un des catalyseurs organiques basi ques susmentionnés.
La nitration du noyau phénylique en posi. tion 5 de la y-4-acyloxyméthyl-d2-oxazoline (étape D) se fait de préférence en traitant ce composé avec de l'acide nitrique à 100 0/0, ou avec de l'acide nitrique fumant ou avec un mélange d'acide nitrique concentré et d'acide sulfurique concentré. On maintient la tempé rature pendant la réaction au-dessous de 25 C et, de préférence, entre -10 et -I-10 C.
On préfère utiliser de l'acide nitrique à. 100 % ou de l'acide nitrique fumant, étant donné qu'on obtient avec ces agents des rendements phis élevés en y-2-méthyl-5-p-nitrophényl- oxazoline que par l'utilisation d'un mélange d'acide nitrique et d'acide sulfurique.
L'hydrolyse de la yr2-méthyl-4-a.cyloxy- méthyl-5-p-nitrophény 1- 42 - oxazoline en le composé y-4-hydroxy méthy le correspondant qui constitue la première étape de l'hydrolyse partielle en deux phases (étape D) est effec tuée à l'aide d'un agent hydrolytique alcalin à une température inférieure à 35 C.
On obtient les meilleurs résultats en utilisant un milieu réactionnel constitué par ou contenant tin solvant organique miscible avec, l'eau, dans le but d'augmenter la solubilité de l'oxazoline. Quelques-uns des solvants particulièrement appropriés dans ce but sont les suivants: mé thanol, éthanol, isopropanol, dioxane, acétone, méthyléthylcétone et des mélanges aqueux de ces solvants. Comme agents hydrolytiques alcalins, on peut utiliser les hydroxydes et les carbonates des métaux alcalins ainsi que l'hydroxyde d'ammonium et les hydroxyde des métaux alcalino-terreux.
Ces substances alcalines peuvent être employées telles quelles ou peuvent être engendrées dans le milieu réactionnel par action du solvant sur un agent hy drolytique alcalin. On peut, par exemple, ajouter de l'éthylate de sodium à un milieu réactionnel alcoolique et aqueux pour obtenir de l'hydroxyde de sodium et de l'éthanol par réaction de l'éthylate de sodium avec l'eau présente dans l'alcool aqueux.
L'étape F de l'hydrolyse peut s'effectuer par traitement de la V-2-méthyl-4-hydroxy- méthy l-5-p-nitrophényl-J''-oxazoline avec un acide minéral dilué et par neutralisation du sel du 1-p-nitrophényl-l-aeétoxy-\?-aminopro- pane-3-ol ainsi obtenu. Après la neutralisa tion dudit sel, la base libre se transforme spontanément en le @p-1-p-nitrophényl-2-acét- amidopropane-1,3-diol désiré. La durée pré férée du traitement est environ 10 à. 1.5 mi nutes pendant. lesquelles le mélange réaction nel est de préférence maintenu à une tempé rature au-dessous de 35 C.
Si on fait durer le chauffage trop longtemps, une hydrolyse complète en p-1-p-nit.rophényl-2-aniinopropane- 1,3-diol a lieu, ce qu'il faut éviter. La tempé rature préférée pour l'hydrolyse partielle est comprise entre 10 et 30 C. Si on le. désire, on peut encore utiliser un solvant or-anique miscible à l'eau pour augmenter la solubilité de l'oxazoline à hydrolyser.
Le procédé selon l'invention est décrit plus en détail dans l'exemple suivant.: Exerit.ple: 20,9 g de dl-y-phénylsérinate d'éthyle (ester éthylique de l'acide tE,-a-amino-l- hydroxy-p'-phénylpropioniqtte) et. 11,3 g de chlorhydrate d'acétimido-éthyl-éther sont agi tés dans 50 em3 de chloroforme sec pendant. une demi-heure à 25 C. On filtre le mélange réactionnel pour éliminer le chlorure d'am monium insoluble et on distille le chloroforme du filtrat dans le vide.
L'huile résiduelle ainsi obtenue est la dl-zp-2-méthyl-5-phényl-4- carbéthoxy-J2-oxazoline.
On dissout 11,7 g de cette substance dans 500 ems d'éther sec et on ajoute cette solu- tion à, une solution de 1 g de LiAlH4 dans 1250 cm3 d'éther sec pendant une période de 3 heures environ. On agite le mélange réac tionnel pendant environ 1 heure, puis on dé compose l'excès du LiAlH.I par l'addition soi gneuse de 100 em3 d'eau froide. On évapore le mélange réactionnel à siccité dans le vide et on extrait le résidu complètement avec de l'éthanol absolu froid.
L'évaporation de l'al cool donne la. dl-zp-2-méthy 1- 5 - phényl - 4- liydroxyméthyl-d2-oxazoline recherchée qui fond, après recristallisation dans de l'acétate d'éthyle, à 114 C. Au lieu d'évaporer à sic cité le mélange réactionnel comme décrit ci dessus, on peut. distiller l'éther dans le vide et extraire l'hydroxyméthyl-oxazoline recher- eliée du résidu aqueux à l'aide d'acétate (l'éthyle.
On laisse au repos pendant. une nuit, à la température ambiante, un mélange composé (le 15 g de cette dl-yp-2-méthyl-5-phényl-4- liyclroxyméthvl-42-oxazoline, de 120 em3 de pyridine sèche et de 20 cm3 d'anhydride acé tique. On traite le mélange réactionnel avec de l'eau froide et on reprend l'huile se sépa rant dans de l'acétate d'éthyle. On sèche la solution d'acétate d'éthyle, on distille ce der nier et on purifie le produit cristallin résiduel par recristallisation dans un mélange métha nol - eau; point de fusion: 160 C.
Le produit ainsi obtenu est la dl-.q,-2-méthyl-5-phényl-4- acétoxyméthyl-d2-oxazoline.
On ajoute sous agitation 12 4, de ladite acétoxyméthyl-oxazoline en petites portions à 50 em3 d'acide nitrique à 10011/o en mainte nant la température en dessous de 10 C et de préférence entre 0 et 5 C. Après que toute l'oxazoline a été ajoutée, on brasse le mé lange réactionnel pendant une demi-heure et on le verse ensuite sur de la glace.
La dl-ip-2- méthy 1- 5 - p -nitrophényl-4-acétoxyméthyl-42- oxazoline brute et insoluble ainsi obtenue est recueillie et ensuite purifiée par recristallisa- tion dans de l'éthanol ou dans de l'acétate d'éthyle.
On dissout à 0 C 10 g de ce produit dans une solution composée de 185 ems d'une solu- tion 0,2 N d'hydroxyde de sodium et de 185 em3 d'acétone. Le mélange réactionnel est laissé au repos pendant une heure, puis on le neutralise exactement par l'addition d'acide chlorhydrique. On élimine l'acétone par distil lation dans le vide et on recueille la dl-V-2- méthyl-5-p-nitrophényl - 4 - hydroxyméthyl - 42- oxazoline cristalline du résidu. Après recris- tallisation dans de l'alcool, ce produit fond à. 157-158 C.
Si on le désire, on peut aussi effectuer cette transformation en utilisant de l'ammo niaque alcoolique. Après achèvement de la réaction, on élimine l'alcool par distillation, on lave le résidu à l'eau et on purifie le pro duit insoluble par recristallisation dans de l'éthanol; point de fusion: 157-158 C.
On ajoute 6 g de cette dl-xp-2-méthyl-5-p- nitrophényl-4-hydroxyméthyl-42-oxazoline à 50 cm- d'acide chlorhydrique dilué (conte nant 2,2 ems d'acide chlorhydrique concentrée et. on agite le mélange réactionnel pendant 1.5 minutes à la température ambiante. On évapore le mélange réactionnel à siccité dans le vide et on obtient. ainsi le chlorhydrate du dl-y-1-p-nitrophényl-1-acétoxy-2-aminopro- panol-3 du point de fusion 164-165 C après recristallisation dans le méthanol. On dissout ce chlorhydrate dans de l'eau et on alcalinise la solution à froid à l'aide d'hydroxyde d'am monium.
On recueille le dl-y-1-p-nitrophényl- 2-acétamidopropanediol-1,3 qui se sépare de la solution et on le purifie par recristallisa tion dans de l'acétate d'éthyle; point, de fusion: 166,15-1670 5 C. On peut obtenir des quantités additionnelles de ce produit par extraction des eaux mères avec le dichlorure d'éthylène.
Le même produit final peut. aussi être obtenu en ajoutant la dl-y-2-méthyl-5-p-nitro- phényl-4-hydroxyméthyl-d2-oxazoline à de l'eau, en faisant bouillir le mélange pendant environ une heure et en refroidissant la solu tion et recueillant le produit.
Lorsqu'on veut effectuer l'étape d'hydro lyse en une seule opération, on peut obtenir le produit final à partir de la dl-y-2-méthyl- 5-p-nitrophényl-4-a.cétoxyméthyl - d2 - oxazolin e par une hydrolyse acide partielle telle que décrite ci-dessus. Le dl-y-1-p-nitrophényl-2- acétamido-propanediol-1,3 finalement obtenu fond à 166<B>0</B> 5-167<B>0</B> 5 C.

The amount of LiAlH4 employed for the reduction can vary within fairly wide limits. However, it is preferable to use a slightly larger amount than the theoretically necessary amount.
The transformation of the. V-4-hydroxy-methyl-42-oxazoline to yr4-acyloxymethyl-d2-oxazoline (step C) can be performed by treating the yp-4-hydroxymethyl-J2-oxazoline with a halide or a anhydride of a carboxylic acid.
Preferably, this reaction is carried out under substantially anhydrous conditions using an excess of acylating agent and a basic catalyst such as pyridine, trimethylamine, dimethylaniline, N-ethylpiperidine or a similar tertiary amine. The acylation can also be carried out in an aqueous reaction medium using a basic catalyst such as a hydroxide or a carbonate of an alkaline metal, or one of the basic organic catalysts mentioned above.
Nitration of the phenyl ring in posi. The reaction of y-4-acyloxymethyl-d2-oxazoline (step D) is preferably carried out by treating this compound with 100% nitric acid, or with fuming nitric acid or with a mixture of Concentrated nitric acid and concentrated sulfuric acid. The temperature during the reaction is maintained below 25 ° C. and preferably between -10 and -I-10 C.
It is preferred to use nitric acid at. 100% or fuming nitric acid, since phis high yields of y-2-methyl-5-p-nitrophenyl-oxazoline are obtained with these agents only by the use of a mixture of nitric acid and sulfuric acid.
The hydrolysis of yr2-methyl-4-a.cyloxy-methyl-5-p-nitrophény 1- 42 - oxazoline to the corresponding y-4-hydroxy methyl compound which constitutes the first step of the partial hydrolysis in two phases (step D) is carried out using an alkaline hydrolytic agent at a temperature below 35 C.
The best results are obtained by using a reaction medium consisting of or containing an organic solvent miscible with water, with the aim of increasing the solubility of the oxazoline. Some of the solvents particularly suitable for this purpose are as follows: methanol, ethanol, isopropanol, dioxane, acetone, methyl ethyl ketone and aqueous mixtures of these solvents. As alkaline hydrolytic agents, there may be used the hydroxides and carbonates of the alkali metals as well as the ammonium hydroxide and the hydroxides of the alkaline earth metals.
These alkaline substances can be used as such or can be generated in the reaction medium by the action of the solvent on an alkaline hydrolytic agent. One can, for example, add sodium ethoxide to an alcoholic and aqueous reaction medium to obtain sodium hydroxide and ethanol by reacting sodium ethoxide with the water present in the alcohol. aqueous.
Step F of the hydrolysis can be carried out by treating the V-2-methyl-4-hydroxy-methyl-5-p-nitrophenyl-J '' - oxazoline with a dilute mineral acid and by neutralizing the salt. 1-p-nitrophenyl-1-aeetoxy - \? - aminopropan-3-ol thus obtained. After neutralization of said salt, the free base spontaneously transforms into the desired p-1-p-nitrophenyl-2-acet-amidopropane-1,3-diol. The preferred duration of treatment is about 10 to. 1.5 minutes during. which the reaction mixture is preferably maintained at a temperature below 35 C.
If heating is continued too long, complete hydrolysis to p-1-p-nit.rophenyl-2-aniinopropane-1,3-diol takes place, which should be avoided. The preferred temperature for partial hydrolysis is between 10 and 30 C. If so. If desired, a water-miscible gold-anic solvent can also be used to increase the solubility of the oxazoline to be hydrolyzed.
The process according to the invention is described in more detail in the following example: Exerit.ple: 20.9 g of ethyl dl-y-phenylserinate (ethyl ester of tE, -a-amino-l - hydroxy-p'-phenylpropioniqtte) and. 11.3 g of acetimido-ethyl-ether hydrochloride are stirred in 50 em3 of dry chloroform for. half an hour at 25 ° C. The reaction mixture is filtered to remove insoluble ammonium chloride and the chloroform from the filtrate is distilled off in vacuo.
The residual oil thus obtained is dl-zp-2-methyl-5-phenyl-4-carbethoxy-J2-oxazoline.
11.7 g of this substance are dissolved in 500 ems of dry ether and this solution is added to a solution of 1 g of LiAlH4 in 1250 cm3 of dry ether over a period of about 3 hours. The reaction mixture is stirred for about 1 hour, then the excess LiAlH.I is decomposed by the addition of 100 em3 of cold water. The reaction mixture is evaporated to dryness in vacuo and the residue is extracted completely with cold absolute ethanol.
Evaporation of the al cool gives the. dl-zp-2-methyl 1- 5 - phenyl - 4-hydroxymethyl-d2-oxazoline desired which melts, after recrystallization from ethyl acetate, at 114 C.
