Verfahren zur Darstellung von Äthiocholensäurederivaten Die ErfindJung betrifft ein Verfahren zur Herstellung von 3 - Keto - 1l a - oxy - 44 - äthio- eliol'ensäure, ausgehend von 3ss,11a-Diaeyloxy- äthioehollansäureestern.
Die Ausgangsprodukte können zum Bei spiel nach der von A. Katz, Ilelv. Chim. Acta 31., 995 (1948), beschriebenen Methode, durch katalytisehe Hydriering des Esters der 3ss,11a.- Diacyloxy - 414 - äthiocholensäure dargestellt werden, die ihrerseits aus Sarmentogenin durch Verestern, Oxydieren und Abspalten von Wasser erhalten werden kann.
Die Endprodukte dieser Synthese unter scheiden sich von gewissen bekannten ähn lichen Verbindungen mit. einem Substituenten in 11-Stellung darin, dass der 11-Substituent in a-Stellüng des Steroidskelets angeordnet. ist.
Als Ausgangsprodukte seien zum Beispiel der bekannte Methylester der 3ss,11a-Diacet- oxy-äthiocholansäure mit F. --- 180 C, das 3f,11a-Dibenzoat mit F. = 130 C, das bisher noch nicht beschrieben wurde, und das 3ss- Aeetat-7la-benzoat mit F. = 242 C erwähnt.
Die 3-Keto-lla-oxy-d4-äthiocholensäure wird erfindungsgemäss in der Weise herge stellt., dass man einen Ester einer 3ss,11a-Dieyl- oxy-äthiocholansäure partiell zu der entspre chenden 3-Oxyverbindung verseift., diese zur 3-Ketoverbindung oxydiert, durch Halogenie- rung und Dehydrohalogenierung eine Doppel bindung zwischen den C-Atomen 4 und 5 ein- führt und,
das erhaltene r'thiocholensäure- Derivat zur 3-Keto-lla-oXy-44-äthiocholen- säure verseift. In der so erhaltenen Verbin dung kann .die 11a-IIydroxylgruppe leicht acyliert werden.
Diese Synthese wird durch das Reaktions schema in der Zeichnung erläutert, wobei R1 und R.2 Acylreste und Rs einen Alkoholrest darstellen.
Die Umwandlung der Verbindung I in die Verbindung II tritt durch vorsichtiges Ver seifen, unter Anwendung milder Reaktions bedingungen ein, z. B. bei niederer Tempera tur, niederer Konzentration des Verseifungs- mittels oder kurzer Reaktionsdauer. Es kön nen hierfür die üblichen Mittel angewendet werden, z.
B. alkalisch reagierende Verbin dungen, wie Alkalihydmoxydle, Alhalicarbonate und -bicarbonate, in einem organischen Lö- sungsmittel und Säuren, z. B. Mineralsäuren., in alkoholischer Lösung.
Die Oxydation der Hydroxylgruppe in Verbindung II zur Ketogruppe kann mit den üblichen oxydierenden Mitteln durchgeführt werden, z. B. mit Chromtrioxyd in Eisessig lösung, Permanganatlösung oder N-Halogen amiden, wie z. B. N-Brom succinimid, N-Brom- aceta.mid und dergleichen.
Die Verbindung III wird vorzugsweise in Eisessiglösung bromiert und anschliessend zur Dehydrobromierung in an sich bekannter Weise [vergl. z. B. T. I4. Kritehevsky und Mitarbeiter, J. A. C. S. 73, 184, 189 (1951) mit Pyridin, Sernica.rbazid oder einem Hy- drazinderivat behandelt, wobei in dien letzten beiden Fällen nachträglich die Ketogruppe wieder freigesetzt werden muss.
Die völlige Verseifung des :ithiocholen- säure-Derivates kann zum Beispiel mit Alkali- hydroxyd@en, -carbonaten oder -bicarbonaten oder mit. Säuren, z. B. Mineralsäure in alko holischer Lösung, durchgeführt werden. Durch Acylierrurg der 11a-Hydroxylgruppe der Ver bindung VI, z. B. mit Säuren, wie niederen Fettsäuren, Säureanhydriden und Säurehalo geniden, erhält man die Verbindungen VII.
Die neuen Endprodukte dieser Synthese sind für die Darstellung von Nebennierenrin- denhormonen und ähnliehe-n Verbindungen von Bedeutung.

Die Lösung wird mit. Was.ser, dann mit. 2n-Sch-#vefelsärire, dann mit Natrium- earbonatlösung und Wasser gewasehen, schliess lieh über Natriumsulfat get.roeknet und im Vakuum eingedampft. (991 mg).

17ethydester <I>von</I> 3-Keto-1la-acetoxy-J4- dthiocholensäure <I>(V)</I> <B>71.0</B> mg des Methylesters der 4-Brom-3-keto- l la-aeetoxy-äthi@ocholan@säLLre (IV) mit F. =168 bis l.74 C werden in. 35 em3 hochgereinigtem Eisessig gelöst.
Hierzu wird eine Lösung von <B>620</B> mg Semiearbazidlhydrochlorid und .920 mg kristallisiertem Nat.riumaceta!t in 1,5 em3 Was- ;er hinzu;
efügt. Die Lösung wird 2 Stunden bei 65 C unter Stickstoffatmosphäre erwärmt, dann werden 7,8 em3 frisch destillierte Brenz- traubensäure und 2,2 g kristallisiertes Na triumacetat in 10,7 em3 Wasser hinzugefügt und die 1Misehung 1,5 Stunden lang auf 70 C erwärmt, worauf sie 15 -Minuten bei 80 C stehengela:

Der schwach gelbe Rückstand (600 mg) wird an 18g Aluminiumoxyd chromatographiert.
Die mit. Petroläther-Benzol (1 :1) und, rei nem Benzol eluierten Fraktionen ergeben 479 mg Substanz. Aus Äther und Äther-Petrol- äther kristallieieren 119,5 mg des Methylesters von 3-Keto-lla-acet.oxy-44-äthioeholensäure in Form kurzer, rosettenartiger Kristallstäbehen mit F. = 153-155 C aus.
Nach Umkristalli- sieren aus Äther-Hexan werden Plättchen oder stäbehenförmige Kristalle mit R.änderung und F. = 155-157 C erhalten.
Spezifische Drehung [a]" = -i-120,9 2 (c = 1,0622 in Chloroform).
Die Elementaranalyse ergab folgende Werte 3,991 mg der Substanz ergaben 10,382 mg C02 und 2,870 mg H20.

sung 2 Stun den lang auf dem Wasserbad gekocht, wobei man 0,-15 e.m3 Wasser hinnüügt. Nach Zugabe von 10 em3 Wasser -wird das Methanol im Vakuum entfernt und die wässerige Lösung zweimal: jeweils mit 10 em3 Chloroform aus geschüttelt.. Unter Eiskühlung wird die Mi- sehung mit. Salzsäure angesäuert, wobei die. Säure amorph ausfällt.
Nach nveimaligem Aussehütteln mit je 60 cm3 Chloroform-Äther (1 :4) wird der Extrakt dreimal mit 3 em3 Wasser gewaschen, über Natriumsulfat ge trocknet und eingedampft. Man erhält 358 mg praktisch farblose Substanz, die aus Aceton sofort auskristallisiert. Die Kristalle, welche bei 189-198 C schrne:lzen, sind schwach lös lich in Äther, Benzol, Aceton und Chloroform.
356 mg dieses rohen Produktes (VI) werden in 3 em3 absolutem Pyridin gelöst und. mit 2 em3 Essigsäureanhydrid 1.6 Stunden lang auf 40 C erwärmt.
Nach Zugabe von 1 em3 Wasser erwärmt man -die Mischung eine Stunde auf 100 C, worauf sie in üblicher Weise aufgearbeitet wird. Man erhält daraufhin eine praktisch farblose Substanz (402 mg), die in Äther ge löst -wird, worauf sofort Kristallisation ein setzt, die noch durch Zugabe von Hexan ver bessert werden kann.
Nach Umkristal'lisieren aus einer Äther-llexan-Misehung erhält man 309,5 mg prismenartiger Kristallstäbehen mit F. = 225-229 C. Die kristallisierte Säure kann einer Molekulardestillation bei 0,01 mm I:Ig und einer Temperatur des Heizbades von ungefähr 210 C unterworfen -werden, ohne dass Zersetzung eintritt. Hierbei bleibt kein Rückstand übrig.
Das Destillat kristallisiert aus Äther-Hexan in Form durchsichtiger Kri- stallstäbchen, die zwischen 145 und 175 C undurchsichtig werden Lind bei 225-229 C schmelzen, wobei sie sich gelb färben.
Eine Probe dieser Substanz -wird mit. einer ätherischen Diazomethanlösung vermischt, worauf sich der Methylester (V) bildet, der aus Äther-Hexan in Form von Plättehen mit F. = 155-158 C auskristallisiert. Spezifische Drehung [a]D = + 120,2 20 (c = 0,9397 in Aceton). Die Elementaranalyse ergab folgende Werte 4,020 mg der Substanz ergaben 10,410 mg C02 und 2,989 mg H20.
C!22,3005 (374,46) berechnet.: C = 70,56; H = 8,08 /o gefunden: C = 70,67; H = 8,32%.
