Verfahren <SEP> zur <SEP> Herstellung <SEP> von <SEP> 2,3-ungesättigten <SEP> Chinuclidin-3-carbonsäure-alkyle\^F Chinuclidin-3-carbonsäure und Abkömm linge davon sind bis heute nicht bekanntge worden. Es wurde gefunden, dass solche Ver bindungen beständig sind und nach dem hier beschriebenen Verfahren gut hergestellt wer den können.
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von 2.,3 ungesättigten Chinuclidin-3-carbonsäurealkyl- estern; und das Verfahren ist dadurch ge kennzeichnet, dass man 3-Chinuclidone durch Anlagerung von Cyanwasserstoffsäure in 3- Oxy - 3 - cyanochinuelidine überführt, daraus durch Hydrolyse die 3-Oxy - 3 - cliinuclidin- carbonsäuren herstellt,
diese in ihre Alkyl- ester überführt und aus diesen 3-Oxy-Ver- bindimgen Wasser abspaltet. Die so erhaltenen 2,3 - ungesättigten Chinuclidin-3=carbonsäure- alkylester können gegebenenfalls zu den Chinuclidin-3-carbonsäure-alkylestern hydriert werden.
Die Synthese der neuen Chinuclidinderi- vate verläuft nach folgendem Schema:

3-Chinuclidön I wird durch Anlagerung von Blausäure in das 3-Oxy-3-cyano-chinu= elidin II übergeführt. Man geht vorteilhaft von einem Salz der Cyariwasserstoffsäure, wie dem Kalium- oder Natriumsalz, aus und setzt die Säure in schwach saurem Reaktions medium in Freiheit;
dabei verwendet man in erster Linie ein Salz des Chinuclidons wie ein halogensaures Salz in wässeriger Lösung. Die Umwandlung der Nitrile II in die Car- bonsäuren erfolgt in an sich bekannter Weise durch Hydrolyse.
Die Überführung der Car bonsäuren III in ihre Alkylester IV erfolgt in an sich bekannter Weise durch Vereste- rung, beispielsweise durch Kochen mit Alka- nol unter dem Einfluss von Säuren,
durch Behandeln mit Diazoalkanen oder durch Ein- wirkung von Alkanolen auf die entsprechen den Carbonsäiurechloride. Die Wasserabspal tung aus der 3-Oxy - chinuclidin - 3 - carbon- säüre-alkyclestern IV erfolgt vorzugsweise mit Thionylchlorid. Die Doppelbindung der entsprechenden 2,
3-ungesättigten Chinuclidin- derivate V lässt sich mit katalytisch angereg tem Wasserstoff, insbesondere in Gegenwart eines Platinkatalysators, absättigen.
Je nach Ausführungsform des Verfahrens erhält man die neuen Verbindungen in Form der freien Basen oder ihrer Salze. Aus den Salzen lassen sich in üblicher Weise die Basen gewinnen; freie Basen können nach bekannten Methoden in ihre Salze abgewandelt werden.
Dabei eignen sich zur Bildung von thera peutisch verwendbaren Salzen solche mit an organischen Säuren, wie zum Beispiel Halogen wasserstoffsäuren, Schwefelsäuren, Salpeter säure, Phosphorsäure, Rhodanwasserstoffsäure oder mit organischen Säuren, wie zum Bei spiel mit Essigsäure, Propionsäure, Oxalsäure, Malonsäure, Bernsteinsäure, Äpfelsäure, Me- thansi-lfonsäure, Äthansulfonsäure,
Oxyäthan- siilfonsäiire, Benzol- oder Toluolsulfonsäure oder mit therapeutisch wirksamen Säuren. Von den neuen Basen können durch Behand lung mit quaternisierenden Mitteln quater- näre Ammoniumsalze hergestellt werden, so zum Beispiel durch Umsetzung mit reaktions fähigen Estern niederer Alkanole, zum Bei spiel Alkylhalogeniden, Dialkyl'sulfaten oder den Alkylestern organischer Sulfonsäuren,
wie der Toluolsulfonsäure.
Die neuen Chinuclidinderivate sind thera peutisch wirksam. So zeigen sie ausgespro chene sympathicomimetliische Eigenschaften, die für eine therapeutische Wirkung auf die gestörte Darmfunktion praktisch von Bedeu- tung sind. Sie sollen als Heilmittel Verwen. dung finden und als Zwischenprodukte zur Herstellung von Desinfektionsmitteln, Textil hilfsstoffen und von Heilmitteln dienen.
Das erfindungsgemässe Verfahren wird in den nachfolgenden Beispielen beschrieben. Zwischen Gewichtsteilen und Volumteilen be steht die gleiche Beziehung wie zwischen Gramm und Kubikzentimeter. Die Tempera turen sind in Celsiusgraden angegeben.
Beispiel <I>1</I> Zu einer eisgekühlten Lösung von 1 & Ge wichtsteilen 3-Chinuclidonhydrochlorid in 22.,5 Volumteilen Wasser wird eine Lösung von 7,3 Gewichtsteilen Kaliilmcyanid in 22,5 Vo- lumteilen Wasser im Laufe von 15 Minuten zugetropft. Das alsbald sich ausscheidende kristalline Cyanhydrin wird noch eine weitere Stunde bei 0 gerührt, abfiltriert und mit wenig Eiswasser gewaschen.
Nach dem Trock nen werden 16 Gewichtsteile des farblosen 3-0xy-3-cyano-chinuclidins vom Schmelzpunkt 1'53 bis 155 erhalten. Aus Dioxan kristalli siert die Verbindung in farblosen Prismen vom Schmelzpunkt 159 unter Zersetzung. In ganz reinem Zustand schmilzt sie bei 172 bis 173 und liefert ein Pikrat, welches nach Kristallisation aus Isopropanol bei etwa 133 bis 134 unter Gasentwicklung schmilzt. Die Schmelze erstarrt wieder und schmilzt schliess lich unter Zersetzung bei 172 bis 1'73 .
9;5 Gewichtsteile 3-Oxy-3-cyano-chinuclidin werden in 130 Volumteilen konzentrierter Salzsäure gelöst und während 48 Stunden bei Raiuntemperatur stehengelassen. Dann wird im Vakuum vollständig eingedampft. Durch Kristallisation des Rückstandes aus Äthanol- Äther wird das Hydrochlorid der 3-Oxy-3- chinuclidin-carbonsäure in Form farbloser Na deln vom Schmelzpunkt 252 bis 25.5 ' (unter Zersetzung) erhalten.
Diese werden mit 130 Volumteilen bei 0 mit Chlorwasserstoff ge sättigtem absolutem Methanol versetzt und 48 Stunden unter gelegentlichem Schütteln bei etwa 2'0 stehengelassen. Durch Eindamp fen . wird das Hydrochlorid des 3-Oxy-3- carbomethoxy-chinuclidins erhalten. Zur Isolierung der freien Esterbase wird das Salz mit eiskalter, konzentrierter Kalitim- earbonatlösung versetzt und die ausgeschie dene Base mit Chloroform extrahiert.
Die Chloroformlösung wird über Pottasche ge trocknet und eingedampft, wobei 8,9 Gewichts teile 3-Oxy - 3 - carbomethoxy - chinuclidin zu rückbleiben. Dieses kristallisiert aus Chloro- form-Pentan in Rosetten vom Schmelzpunkt 122 und liefert mit Methyljodid in Aceton ein Methjodid vom Schmelzpunkt 191 bis192 . Das Pikrat der Base kristallisiert aus Äthanol in gelben Nadeln vom Schmelzpunkt 188 bis 18'9 .
Das Hydrochlorid bildet aus Aceton zu Büscheln vereinigte Prismen vom Schmelz punkt 140 bis 141 .
8,8 Gewichtsteile 3-Oxy - 3 - carbomethoxy- chinuclidin werden portionenweise in 50 Vo- lumteile eisgekühltes Thionylchlorid einge tragen und die Lösung anschliessend während 48 Stunden unter Rückfluss gekocht.
Die ent standene dunkelgelbe Lösung wird im Va- luzum vollständig eingedampft und der kri stalline Rückstand in heissem Aceton aufge nommen, mit etwas Tierkohle aufgekocht und filtriert. Beim Einengen der acetonischen Lö sung kristallisieren 7 Gewichtsteile Hydro- chlorid des ungesättigten Esters in farblosen Nadeln vom Schmelzpunkt 178 bis 179 .
<I>Beispiel 2</I> Eine Lösung von 64 Gewichtsteilen des in Beispiel 1 beschriebenen 3-Carbomethoxy- 1-azabicyclo - (2,2,2)-oct-2,enhydrochlorids in 50 Volumteilen Methanol wird in Gegenwart von 4,3 Gewichtsteilen Platin bei Raumtem- peratur und gewöhnlichem Druck hydriert, wobei die berechnete Menge Wasserstoff auf genommen wird.
Die Lösung wird filtriert und einge dampft und der Rückstand aus Aceton um- , kristallisiert, wobei @6 Gewichtsteile des ge sättigten Esters als leicht hygroskopisches Hydrochlorid vom Schmelzpunkt 167 bis 1'69 erhalten werden. Aus dem Hydrochlorid kann mittels Pikrinsäure in Äthanol das Pikrat . hergestellt werden, welches in gelben Nadeln vom Schmelzpunkt 155 bis 15'6 kristallisiert.

The present invention relates to a process for the preparation of 2nd, 3rd unsaturated quinuclidine-3-carboxylic acid alkyl esters; and the process is characterized in that 3-quinuclidones are converted into 3-oxy - 3 - cyanochinuelidines by addition of hydrocyanic acid, and the 3-oxy - 3 - cliinuclidine carboxylic acids are produced therefrom by hydrolysis,
these are converted into their alkyl esters and water is split off from these 3-oxy compounds. The 2,3-unsaturated quinuclidine-3-carboxylic acid alkyl esters obtained in this way can optionally be hydrogenated to give the quinuclidine-3-carboxylic acid alkyl esters.
The synthesis of the new quinuclidine derivatives proceeds according to the following scheme:

3-Quinuclidon I is converted into 3-oxy-3-cyano-chinu = elidin II by the addition of hydrocyanic acid. It is advantageous to start from a salt of hydrocyanic acid, such as the potassium or sodium salt, and leave the acid free in a weakly acidic reaction medium;
a quinuclidone salt such as a halogenated salt in aqueous solution is primarily used. The conversion of the nitriles II into the carboxylic acids takes place in a manner known per se by hydrolysis.
The conversion of the carboxylic acids III into their alkyl esters IV takes place in a manner known per se by esterification, for example by boiling with alkanol under the influence of acids,
by treatment with diazoalkanes or by the action of alkanols on the corresponding carboxylic acid chlorides. The elimination of water from the 3-oxy-quinuclidine-3-carboxylic acid-alkyl ester IV is preferably carried out with thionyl chloride. The double bond of the corresponding 2,
3-Unsaturated quinuclidine derivatives V can be saturated with catalytically excited hydrogen, in particular in the presence of a platinum catalyst.
Depending on the embodiment of the process, the new compounds are obtained in the form of the free bases or their salts. The bases can be obtained from the salts in the usual way; Free bases can be converted into their salts by known methods.
For the formation of therapeutically usable salts, those with organic acids, such as hydrohalic acids, sulfuric acids, nitric acid, phosphoric acid, hydrochloric acid or with organic acids, such as, for example, acetic acid, propionic acid, oxalic acid, malonic acid, succinic acid, are suitable. Malic acid, methanesulfonic acid, ethanesulfonic acid,
Oxyethane, benzene or toluene sulfonic acid or with therapeutically active acids. Quaternary ammonium salts can be produced from the new bases by treatment with quaternizing agents, for example by reaction with reactive esters of lower alkanols, for example alkyl halides, dialkyl sulfates or the alkyl esters of organic sulfonic acids,
like toluenesulfonic acid.
The new quinuclidine derivatives are therapeutically effective. They show pronounced sympatheticomimetic properties that are of practical importance for a therapeutic effect on the disturbed intestinal function. They are to be used as a remedy. and serve as intermediate products in the manufacture of disinfectants, textile auxiliaries and medicinal products.
The process according to the invention is described in the following examples. The relationship between parts by weight and parts by volume is the same as that between grams and cubic centimeters. The temperatures are given in degrees Celsius.
Example <I> 1 </I> A solution of 7.3 parts by weight of potassium cyanide in 22.5 parts by volume of water is added to an ice-cold solution of 1 part by weight of 3-quinuclidone hydrochloride in 22.5 parts by volume of water in the course of 15 minutes added dropwise. The crystalline cyanohydrin which separates out is stirred for a further hour at 0, filtered off and washed with a little ice water.
After drying, 16 parts by weight of the colorless 3-0xy-3-cyano-quinuclidine with a melting point of 1'53 to 155 are obtained. The compound crystallizes from dioxane in colorless prisms with a melting point of 159 with decomposition. In a completely pure state it melts at 172 to 173 and produces a picrate which, after crystallization from isopropanol, melts at about 133 to 134 with evolution of gas. The melt solidifies again and finally melts with decomposition at 172 to 1'73.
9; 5 parts by weight of 3-oxy-3-cyanoquinuclidine are dissolved in 130 parts by volume of concentrated hydrochloric acid and left to stand at room temperature for 48 hours. Then it is completely evaporated in vacuo. By crystallizing the residue from ethanol-ether, the hydrochloride of 3-oxy-3-quinuclidine-carboxylic acid is obtained in the form of colorless needles with a melting point of 252 to 25.5 '(with decomposition).
These are mixed with 130 parts by volume at 0 with absolute methanol saturated with hydrogen chloride and left to stand for 48 hours with occasional shaking at about 20. By evaporation. the hydrochloride of 3-oxy-3-carbomethoxy-quinuclidine is obtained. To isolate the free ester base, ice-cold, concentrated potassium carbonate solution is added to the salt and the base which has been separated out is extracted with chloroform.
The chloroform solution is dried over potash and evaporated, 8.9 parts by weight of 3-oxy-3-carbomethoxy-quinuclidine remaining. This crystallizes from chloroform pentane in rosettes with a melting point of 122 and, together with methyl iodide in acetone, yields a meth iodide with a melting point of 191 to 192. The picrate of the base crystallizes from ethanol in yellow needles with a melting point of 188 to 18.9.
From acetone, the hydrochloride forms prisms with a melting point of 140 to 141, which are combined in clusters.
8.8 parts by weight of 3-oxy-3-carbomethoxyquinuclidine are added in portions to 50 parts by volume of ice-cold thionyl chloride and the solution is then refluxed for 48 hours.
The resulting dark yellow solution is completely evaporated in a Valuzum and the crystalline residue is taken up in hot acetone, boiled with a little animal charcoal and filtered. When the acetone solution is concentrated, 7 parts by weight of the hydrochloride of the unsaturated ester crystallize in colorless needles with a melting point of 178 to 179.
<I> Example 2 </I> A solution of 64 parts by weight of the 3-carbomethoxy-1-azabicyclo- (2,2,2) -oct-2, enhydrochloride described in Example 1 in 50 parts by volume of methanol is added in the presence of 4, 3 parts by weight of platinum hydrogenated at room temperature and normal pressure, taking up the calculated amount of hydrogen.
The solution is filtered and evaporated and the residue is recrystallized from acetone, 6 parts by weight of the saturated ester being obtained as a slightly hygroscopic hydrochloride with a melting point of 167 to 1,69. The picrate can be obtained from the hydrochloride using picric acid in ethanol. be produced, which crystallizes in yellow needles with a melting point of 155 to 15'6.
