  Procédé de préparation   d'une      O. déméthyl-N-désacétyl-N-méthyl-colchicine   
 On sait que 1'extraction du Colchicum autumnale par l'alcool conduit, après fractionnement, à trois fractions caractéristiques. La pre  mière,    à réaction neutre, renferme la colchicine, le colchicoside, ainsi que divers satellites (cf.

  La seconde fraction, aminée, renferme la   N-dés-    acétyl-N-méthyl-colchicine (cf. Kiselew et coll.,

   On isole dans la troisième fraction qui présente une réaction phénolique, la   2-déméthyl-    colchicine qui en est le constituant principal.



   L'invention a pour objet un procédé de préparation   d'une    O-déméthyl-N-désacétyl-N-méthyl-colchicine qui se distingue nettement des autres principes ci-dessus mentionnés, en ce qu'elle est à la fois de nature phénolique et de nature   méthyl-aminée.    Ce caractère amphotère, rencontré pour la première fois dans la série des constituants du colchique, est mis à profit dans le nouveau procédé d'extraction de la O-dé  méthyl-N-désacétyl-N-méthyl-colchicine.   



   Le procédé selon la présente invention est donc caractérisé par le fait qu'on épuise les graines de colchique préalablement pulvérisées et dégraissées, par au moins un premier solvant dissolvant tous les principes actifs, on reprend dans un second solvant organique non miscible à l'eau lesdits principes actifs ainsi extraits, on en sépare les composantes basiques qu'on fait passer de la phase organique en phase aqueuse acide sous forme d'un de leurs sels hydrosolubles, on les extrait après neutralisation par un solvant organique non miscible à   1'eau,    on sépare de ladite solution organique les composantes amphotères qu'on fait passer de la phase organique en phase aqueuse sous forme du sel qu'elles fournissent avec une base, on extrait lesdites composantes amphotères après neutralisation par un solvant organique non miscible à l'eau,

   on évapore à sec la solution organique obtenue et on cristallise le résidu de l'évaporation dans un solvant. La
O-déméthyl-N-désacétyl-N-méthyl-colchicine ainsi obtenue étant ensuite si on le désire, transformée en un de ses dérivés.



   Le solvant utilisé pour extraire du résidu obtenu après dégraissage les principes actifs de la graine de colchique, peut être l'eau, un alcool inférieur ou le mélange d'un alcool inférieur avec un carbure aliphatique inférieur halogène.



  On utilise de préférence l'alcool aqueux à 70 %.



  D'après une forme particulière d'exécution du procédé selon l'invention, on élimine l'alcool par distillation sous vide, on extrait la phase aqueuse résultante par le chloroforme et épuise la solution chloroformique par l'acide chlorhydrique N. La phase aqueuse acide qui contient tous les principes aminés du colchique, sous forme de chlorhydrates, est neutralisée par le bicarbonate de sodium puis extraite au chloroforme pour fournir une solution organique riche en   N-désacétyl-N-méthyl-colchicine,    IV (substance F) et autres substances basiques.



  Son extraction rapide et à froid par la soude   N/10,    qui n'affecte pas la N-désacétyl-N-méthyl-colchicine, conduit à une solution qu'on acidifie immédiatement par l'acide chlorhydrique puis neutralise par le bicarbonate de sodium. Cette solution ne contient que les principes amphotères du colchique. On en extrait la
O-déméthyl-N-désacétyl-N-méthyl-colchicine par le chloroforme que l'on évapore et cristallise en méthanol.

   Le produit résultant est un solvat qui perd environ 9,5  /o de son poids par chauffage à   1370    C sous vide, par suite de l'élimination du solvant de cristallisation et qui   fond à 1310 C,    [a] 2D0   logo            50      (c      =      1, 0/o,    chloroforme). Il répond à la formule   C20H2305N    et est à la fois de nature phénolique et aminée.



   La structure de la O-déméthyl-N-désacétyl  N-méthyl-colchicine,      I,    à l'exception de l'emplacement du groupement phénolique libre, est prouvée par les réactions suivantes :   -L'acétylation    fournit un   N, O-diacétate, 111,   
 qui fond à   2230    C, [a]   EO      =2290            50   
 (c = 0,5  /o, chloroforme) et répond à la
 formule centésimale   C24H2707N.   



  -L'hydrolyse acide conduit à la O-déméthyl
   N-désacétyl-N-méthyl-colchicéine,    IIa, qui
 est caractérisée sous forme de son triben
   zoate, llb, C40H33O8N    F.   2320      C,    [a]   2D0 =   
   2720          50 (c = 0,5 o/o chloroforme).



   La   méthylation    de la   0-déméthyl-N-dés-      acétyl-N-méthyl-colchicine,      l,    par le diazométhane fournit la   N-désacétyl-N-méthyl-colchi-      cine, IV,    F.   183-1850    C, identique à un échantillon authentique de   démécolcine    obtenu par extraction du colchique suivant les procédés connus et d'où l'on passe facilement à la   N-mé-    thyl-colchicine,   V,    F.   2270    C, par   acétylation.   



   La O-déméthyl-N-désacétyl-N-méthyl-colchicine trouve son application dans le domaine biologique et industriel de la colchicine, notamment dans les modifications de la mitose et la création de polyploides soit par épandage de solutions aqueuses ou de suspensions des produits sur des sols cultivés, soit par traitement préalable des semences avec les produits purs ou dilués dans un solvant ou sur supports.



   Exemple   
Extraction de la O-déméthyl-N-désacétyl-N-mé-   
 thyl-colchicine.



   On dégraisse 150 kg de semences   pulvéri-    risées de colchique par 500 litres d'éther de pétrole puis les épuise à froid par 700 litres d'alcool à   70 ID/o.    L'alcool est éliminé par distillation sous vide et la solution aqueuse résultante extraite par 5 fois 35 litres de chloroforme. On réunit les extraits chloroformiques et les agite avec 3 fois 10 litres d'acide chlorhydrique N. On sépare la phase aqueuse acide, la neutralise par une solution saturée de bicarbonate de sodium et la traite à nouveau par 4 fois 2 litres de chloroforme. On extrait à froid la solution chloroformique par 4 fois 250 cm3 d'eau de soude   N/10    puis par 500 cm3 d'eau. Les solutions aqueuses basiques réunies sont acidifiées par l'acide chlorhydrique puis neutralisées par le bicarbonate de sodium.

   On extrait une dernière fois par 5 fois 200 cm3 de chloroforme, sèche la solution chloroformique sur sulfate de magnésium et l'évapore à sec sous vide. On obtient ainsi 5,9 g d'un résidu qui, trituré dans un peu de méthanol fournit 2,8 g de solvat cristallisé de O-déméthyl-N  désacétyl-N-méthyl-colchicine.    Le produit qui

 est nouveau fond à   131o C,    [a]   2D =104  + 50    (c = 1    /o, chloroforme). Après recristallisation    en méthanol,   F = 131 C    [a]   20D=-190##5#    (c = 1%, chloroforme).

 à   137  C    sous vide : perte 9,5  /o).

 La   O-déméthyl-N-désacétyl-N-méthyl-col-    chicine ainsi obtenue peut être transformée en un de ces dérivés, par exemple, de la manière suivante :
 A. Transformation en   diacétate de la O-dé-       méthyl-N-désacétyl-N-méthyl-colchicine   
 On traite pendant seize heures, à   200    C, 400 mg de O-déméthyl-N-désacétyl-N-méthylcolchicine obtenue selon l'exemple   susmen-      tionné    par 8 cm3 d'anhydride acétique en présence de 8 cm3 de pyridine, verse le mélange réactionnel dans de l'eau, extrait au chloroforme, lave la solution chloroformique successivement à l'acide chlorhydrique N, à la soude N, puis à l'eau. On sèche sur sulfate de magnésium et évapore à sec sous vide.

   Le résidu est cristallisé en acétate d'éthyle pour fournir 400 mg de diacétate de O-déméthyl-N-dés  acétyl-N-méthyl-colchicine.    Le produit qui n'a pas encore été décrit fond à 223 C, [a]   ri      = - 229##5#    (c = 0,5    /o,    chloroforme).



  On le sèche pour l'analyse pendant deux heures à   100     C.

 B. Hydrolyse de la   O-déméthyl-N-désacé-       tyl-N-methyl-colchicine et preparation   
 de la   tribenzoyl-O-demethyl-N-desace-       tyl-N-methyl-colchiceine.   



   On chauffe au bain-marie pendant deux heures 580 mg de O-déméthyl-N-désacétyl-Nméthyl-colchicine en solution dans 52   cm3    d'eau contenant 6 cm3 d'acide chlorhydrique 2 N, neutralise le mélange réactionnel par une solution saturée de bicarbonate de sodium et 1'extrait par le mélange chloroforme-méthanol (2 : 1). La solution obtenue est lavée par 20 cm3 d'eau, séchée sur sulfate de magnésium et évaporée à sec sous vide. Le résidu   résul-    tant est traité à froid par 2   cm3    de chlorure de benzoyle en présence de 2 cm3 de pyridine.

   On ajoute 30   cm3    d'eau, reprend par le chloroforme et obtient après lavages à l'acide chlorhydrique N, à la soude N, puis à l'eau, séchage sur sulfate de magnésium et évaporation à sec, 600 mg d'un résidu qu'on cristallise en acétate d'éthyle. On obtient ainsi 260 mg de tribenzoyl  
O-deméthyl-N-desacétyl-N-méthyl-colchicéine.   



  Le produit qui est nouveau fond à   231-2320 C,      [a] 21) =-2720 50    (c = 0, 5%, chloroforme).



  On sèche pour l'analyse deux heures à   100  C.   



  Analyse: C40H33O8N = 655,7

   On dissout 50 mg de O-déméthyl-N-dés  acétyl-N-méthyl-colchicine    dans 5 cm3 du mélange méthanol-chloroforme (1 : 4) et traite par 15 cm3 d'une solution   toluénique    à   5 O/o    de   diazométhane.    On abandonne seize heures à la température ambiante, détruit l'excès de diazométhane par adjonction de quelques gouttes d'acide acétique et lave par 5 cm3 de soude N.



  La phase organique restante est lavée à l'eau, sé chée sur sulfate de magnésium et évaporée à sec sous vide. Le résidu cristallise dans l'acétate d'éthyle et fournit 30 mg de   N-désacétyl-N-mé-    thyl-colchicine brute, F.   177-180  C,    qui sont chromatographiés sur 2 g d'alumine neutre. Le résidu provenant de   l'éluat    chloroformique fournit, après cristallisation en acétate d'éthyle, 10 mg de   N-désacétyl-N-méthyl-colchicine,   
F.   183-185o    C, identique à un échantillon authentique de   N-désacétyl-N-méthyl-colchicine    obtenu par extraction du colchique suivant les procédés connus.



   D. Préparation de la   N-méthyl-colchicine   
 par   acetylation    de la   N-désacétyl-N-       méthylcolchicine   
 On dissout 6 mg du produit de   méthylation    de la O-déméthyl-N-désacétyl-N-méthyl-colchicine préparée selon le paragraphe C, dans 0,5 cm3 de pyridine, ajoute 0,5   cm d'anhydride    acétique et abandonne le mélange une nuit à 16  C. On verse dans 5   cm3    d'eau, extrait au chloroforme, lave à l'acide chlorhydrique, au bicarbonate de sodium et à 1'eau et évapore à sec sous vide après séchage sur sulfate de ma  gnésium.    Le résidu cristallise après adjonction d'une goutte d'acétate d'éthyle.

   On essore, lave à l'acétate d'éthyle et obtient ainsi 1,5 mg de   N-méthyl-colchicine,    F.   2250    C.



   Ce produit est identique à la   N-méthyl-    colchicine préparée à partir d'un échantillon authentique de N-désacétyl-N-méthyl-colchicine en suivant la même technique.

   The subject of the invention is a process for the preparation of an O-demethyl-N-deacetyl-N-methyl-colchicine which is clearly distinguished from the other principles mentioned above, in that it is both phenolic in nature and methyl-amine in nature. This amphoteric character, encountered for the first time in the series of constituents of colchicum, is taken advantage of in the new process for extracting O-de-methyl-N-deacetyl-N-methyl-colchicine.



   The process according to the present invention is therefore characterized by the fact that the colchicum seeds pulverized and degreased beforehand are exhausted with at least a first solvent dissolving all the active principles, the residue is taken up in a second organic solvent immiscible with water. said active ingredients thus extracted, the basic components are separated therefrom which are passed from the organic phase to the acidic aqueous phase in the form of one of their water-soluble salts, they are extracted after neutralization with an organic solvent immiscible with water , the amphoteric components are separated from said organic solution which are passed from the organic phase to the aqueous phase in the form of the salt which they provide with a base, said amphoteric components are extracted after neutralization with an organic solvent immiscible with water,

   the organic solution obtained is evaporated to dryness and the residue from the evaporation is crystallized from a solvent. The
O-demethyl-N-desacetyl-N-methyl-colchicine thus obtained being then, if desired, transformed into one of its derivatives.



   The solvent used to extract the active principles of colchicum seed from the residue obtained after degreasing can be water, a lower alcohol or the mixture of a lower alcohol with a halogenated lower aliphatic carbide.



  Preferably 70% aqueous alcohol is used.



  According to a particular embodiment of the process according to the invention, the alcohol is removed by vacuum distillation, the resulting aqueous phase is extracted with chloroform and the chloroform solution is exhausted with hydrochloric acid N. The aqueous phase acid which contains all the amino principles of colchicum, in the form of hydrochlorides, is neutralized with sodium bicarbonate and then extracted with chloroform to provide an organic solution rich in N-deacetyl-N-methyl-colchicine, IV (substance F) and others basic substances.

   150 kg of pulverized colchicum seeds are degreased with 500 liters of petroleum ether and then cold exhausted with 700 liters of 70 ID / o alcohol. The alcohol is removed by vacuum distillation and the resulting aqueous solution extracted with 5 times 35 liters of chloroform. The chloroform extracts are combined and stirred with 3 times 10 liters of N hydrochloric acid. The acidic aqueous phase is separated, neutralized with a saturated solution of sodium bicarbonate and again treated with 4 times 2 liters of chloroform. The chloroform solution is cold extracted with 4 times 250 cm3 of N / 10 sodium hydroxide water then with 500 cm3 of water. The combined basic aqueous solutions are acidified with hydrochloric acid and then neutralized with sodium bicarbonate.

   Extracted one last time with 5 times 200 cm3 of chloroform, the chloroform solution is dried over magnesium sulfate and evaporated to dryness under vacuum. In this way 5.9 g of a residue is obtained which, triturated in a little methanol, gives 2.8 g of crystallized solvate of O-demethyl-N deacetyl-N-methyl-colchicine. The product that

 B. Hydrolysis of O-demethyl-N-desacetyl-N-methyl-colchicine and preparation
 tribenzoyl-O-demethyl-N-desacetyl-N-methyl-colchiceine.



   Heated in a water bath for two hours 580 mg of O-demethyl-N-deacetyl-N-methyl-colchicine dissolved in 52 cm3 of water containing 6 cm3 of 2N hydrochloric acid, neutralized the reaction mixture with a saturated solution of sodium bicarbonate and the extract with a chloroform-methanol mixture (2: 1). O-demethyl-N-desacetyl-N-methyl-colchiceine.



  The product, which is new, melts at 231-2320 C, [a] 21) = -2720 50 (c = 0.5%, chloroform).



  Dry for analysis two hours at 100 ° C.

   D. Preparation of N-methyl-colchicine
 by acetylation of N-desacetyl-N-methylcolchicine
 6 mg of the methylation product of O-demethyl-N-deacetyl-N-methyl-colchicine prepared according to paragraph C is dissolved in 0.5 cm3 of pyridine, added 0.5 cm of acetic anhydride and the mixture is abandoned. overnight at 16 ° C.
