Description

  

  Procédé de     préparation    d'esters     d'estradiol       La présente invention a pour objet un procédé  de préparation     d'hexahydrobenzoates        d'estradiol    qui  sont des composés nouveaux.  



  On a pu     constater    que les     3-hexahydrobenzoate,          3,17-dihexahydrobenzoate    et     17-hexahydrobenzoate          d'estradiol    sont des esters nouveaux doués de pro  priétés remarquables.

   Ce sont des     estrogènes    injec  tables à longue durée d'action.     Le    test d'Allen et       Doisy    sur la rate     ovariectomisée    prouve que     ces     esters nouveaux     entraînent    des manifestations     d'estrus     beaucoup plus prolongées que celles observées avec  les     estrogènes    habituellement utilisés.

   C'est     ainsi    que,  par exemple, le     17-hexahydrobenzoate        d'estradiol     provoque une période moyenne     d'estrus    de 11 jours  alors que le     3-benzoate        d'estradiol    n'agit que pen  dant 3 jours à la dose de 10 y.  



       Les    produits préparés selon l'invention trouvent  leur application aussi bien en médecine humaine  qu'en médecine vétérinaire.  



  Selon l'invention, on obtient les nouveaux esters       d'e.stradiol    en faisant réagir, en présence d'un agent  de condensation, un halogénure     d'hexahydrobenzoyle     ou l'anhydride correspondant sur     l'estradiol    ou un  de ses     monoesters.     



  Les     3-hexahydrobenzoate    et     3,17-dihexahydro-          benzoate        d'estradiol,    par exemple, peuvent être obte  nus en faisant réagir en présence d'un agent de  condensation, un     halogénure        d'hexahydrobenzoyle,     tel que le chlorure ou le bromure, sur une solution       d'estradiol    dans un solvant approprié et en isolant  le mono- ou le     diester    formé. Au     lieu    d'un     halo-          génure        d'hexahydrobenzoyle,    on peut également uti  liser l'anhydride correspondant.  



  Pour la     monoestérification    en 3, on opère de  préférence selon     Schotten        Baumann,    le solvant étant  par exemple l'acétone et la base, un hydroxyde alca-    lin, tel que l'hydroxyde de sodium en solution  aqueuse.  



  La     diacylation    en 3-17 peut être     réalisée    en pré  sence d'une base tertiaire telle que la     pyridine,    la       2-méthyl-5-éthyl-pyridine    ou la     diméthylaniline.     Après la réaction, on reprend de préférence par un  solvant non miscible à l'eau     comme    l'éther, le  benzène, etc., lave avec un agent acide pour éli  miner l'excès de base tertiaire puis avec un agent  alcalin pour enlever l'excès d'acide et l'acide qui n'a  pas réagi et finalement à l'eau, le solvant étant dis  tillé après avoir séché la solution ;

   puis on     recris-          tallise    le     diester    dans un solvant approprié.  



  Le     17-hexahydrobenzoate        d'estradiol    peut être  préparé à     partir    du     diester    3-17 par hydrolyse sélec  tive de ce dernier par une base appropriée aqueuse  telle que la soude     ou    un carbonate alcalin, à une  température     variant    entre la température ambiante  et le reflux, dans un solvant du     3,17-dihexahydro-          benzoate,    miscible à l'eau, tel qu'un     alcool    alipha  tique     inférieur,    de préférence l'éthanol ou le métha  nol.  



  L'obtention directe du     17-hexahydrobenzoate          d'estradiol    sans isolement du     3,17-dihexahydro-          benzoate    intermédiaire, peut s'effectuer par action de  l'anhydride     hexahydrobenzoïque    sur     l'estradiol    dans  un solvant approprié non     miscible    à l'eau, tel que  le     chloroforme,    en présence d'acide     para-toluène-          sulfonique,    puis lavage à l'eau, concentration, trai  tement à     l'ammoniaque    et extraction.  



  On peut aussi traiter un     3-acyl-estradiol,    par  exemple le     3-benzoyl-estradiol,    par un     halogénure          d'hexahydrobenzoyle    ou par l'anhydride     hexahydro-          benzoïque    comme indiqué plus haut et hydrolyser  sélectivement le     3,17-diester        formé,    à condition bien  entendu que le groupement     acyloxy    en 3 soit plus  facilement saponifiable que le groupement hexahydro-           benzoyloxy    en 17.

   Les     3-acyl-17-hexahydrobenzoyl-          estradiols    qui constituent des produits intermédiaires  de     cette    forme d'exécution peuvent également être  doués de propriétés     estrogènes    retard intéressantes.  



  Les nouveaux esters peuvent être employés en  solution dans les huiles végétales neutres, addition  nées ou non d'un glycol, ou en solution dans d'autres  solvants     injectables.    Les concentrations, variables, ne  sont limitées que par les solubilisés. On a pu ainsi  préparer des solutions contenant 10 mg de     3,17-di-          hexahydrobenzoate    et 20 mg de 3- et de     17-mono-          hexahydrobenzoate        d'estradiol    par ce d'huile d'olive.

    Il est parfois souhaitable d'ajouter aux solutions  huileuses du     monostéarate    ou du     2-éthyl-hexa-noate          d'aluminium    pour obtenir un gel     thixotropique.    Les  nouveaux esters de     l'estradiol    qu'on obtient selon la  présente invention peuvent aussi être administrés sous  forme de suspensions aqueuses     microcristallines    ou  d'émulsions dans l'eau ou de solutions dans un tiers  solvant en présence d'un agent émulsionnant neutre  et non irritant. Ces préparations sont réalisées par  des procédés connus.  



  Les exemples suivants montrent comment on peut  mettre en     aeuvre    l'invention.    Exemple 1       3-hexahydrobenzoate        d'estradiol     <I>â partir de</I>     l'estradiol     On dissout 7,

  5 g     d'estradiol    dans 50<I>ce</I> d'acé  tone et ajoute à la température ambiante 26     cm3          d'une        solution        acétonique    à     20        %        de        chlorure        d'hexa-          hydrobenzoyle    et 66     cm3    de soude normale.

   Après  une demi-heure de repos, on ajoute 20     cm3    de soude       normale,        puis        10        ce        de        solution        acétonique    à     20        %     de     chlorure        d'hexahydrobenzoyle.    On répète cette  opération après une nouvelle demi-heure de repos  en ajoutant cette fois 10 ce de soude normale et  10 ce de solution     acétonique    de chlorure. On verse  le mélange réactionnel dans 200 ce d'eau, essore et  sèche sous vide.

   On obtient ainsi 8,75 g, soit un     ren-          dement        de        81        %,        de        3-hexahydrobenzoate        d'estradiol     qu'on     recristallise    en éthanol.

   Le produit qui est  nouveau fond à     91-92,)C    sur bloc,     [a120    =     -I-        53o          1o          (c        =    1     %        dans        le        chloroforme).        Il        se        présente        sous     forme de plaquettes incolores solubles dans l'éther,  l'acétone et le chloroforme, peu solubles dans l'alcool  à froid, insolubles dans l'eau.

Le  résidu est repris dans 400 ce d'acétone chaude d'où  il cristallise par refroidissement. On obtient ainsi  25,9 g soit un rendement de 95 '0 /0, de     3,17-dihexa-          hydrobenzoate        d'estradiol.    On le recristallise dans  l'acétone.

   Le produit qui n'a pas encore été décrit       fond    à     182-183        C        sur        bloc,        [a]20        =        -f-        331>        (c        =        0,5        %     dans le chloroforme). Il se présente sous forme de  prismes allongés incolores solubles dans le chloro  forme, peu solubles à froid dans l'alcool, l'éther et       l'acétone,    insolubles dans l'eau.

   On obtient ainsi 6,6 g,       soit        un        rendement        de        71%        d'une        résine        qui        est     cristallisée dans le benzène par adjonction d'éther de  pétrole.

   Le produit qui est nouveau fond à 1740 C       sur        bloc,        [a]        D        =        -h        450             1 0     (c        =    1     %        dans        le        chlo-          roforme)    et     fournit    dans le méthanol un     solvat     F = 100  C.

   Il se présente sous forme de petits cris  taux incolores solubles dans l'éther, l'acétone, le  benzène et le chloroforme, peu soluble dans le  méthanol froid, insoluble dans l'eau. La cristallisa  tion dans le méthanol aqueux     fournit    une autre forme  cristalline F = 154-155 C.

Le résidu est cristallisé en benzène-éther de  pétrole et fournit le     17-hexahydrobenzoate        d'estradiol          avec        un        rendement        de        601%.     



  Exemple 4       3-benzoate-17-hexahydrobenzoate        d'estradiol     <I>à partir du</I>     3-benzoate        d'estradiol     On refroidit, à     011C,    une solution de 3,2g de       3-benzoate        d'estradiol    d'ans 32     cm3    de     pyridine    an  hydre puis ajoute 2,7     cm3    de chlorure     d'hexahydro-          benzoyle    et abandonne 16 heures à la température  ambiante.

   Après addition de 30     cm3    d'eau, on extrait  par 90     cm3    de chloroforme.     Les    extraits     chlorofor-          miques    sont lavés à l'acide chlorhydrique N, au bicar  bonate de sodium puis à l'eau, séchés sur sulfate de  magnésium et évaporés à sec sous vide.

   On dissout  le résidu dans l'acétone à chaud et obtient, après  refroidissement de la solution, filtration et essorage,  3,5 g (86 0/0) d'aiguilles incolores, F =     206     C,  [a] 20 = +     301,      1 (c =1 0/0, chloroforme).     Le    pro  duit qui est nouveau est insoluble dans l'eau, l'alcool,  soluble dans 100 volumes d'acétone à chaud et très  soluble dans le chloroforme.

   Thus, for example, estradiol 17-hexahydrobenzoate causes an average estrus period of 11 days while estradiol 3-benzoate only acts for 3 days at a dose of 10 µ.



       The products prepared according to the invention find their application both in human medicine and in veterinary medicine.



  According to the invention, the novel esters of estradiol are obtained by reacting, in the presence of a condensing agent, a hexahydrobenzoyl halide or the corresponding anhydride with estradiol or one of its monoesters.



  Estradiol 3-hexahydrobenzoate and 3,17-dihexahydro-benzoate, for example, can be obtained by reacting in the presence of a condensing agent, a hexahydrobenzoyl halide, such as chloride or bromide, on a solution of estradiol in a suitable solvent and isolating the mono- or diester formed. Instead of a hexahydrobenzoyl halide, the corresponding anhydride can also be used.

  The direct production of estradiol 17-hexahydrobenzoate without isolation of the 3,17-dihexahydro-benzoate intermediate, can be carried out by the action of hexahydrobenzoic anhydride on estradiol in a suitable solvent immiscible with water, such as than chloroform, in the presence of para-toluenesulfonic acid, followed by washing with water, concentration, treatment with ammonia and extraction.



  It is also possible to treat a 3-acyl-estradiol, for example 3-benzoyl-estradiol, with a hexahydrobenzoyl halide or with hexahydro-benzoic anhydride as indicated above and selectively hydrolyze the 3,17-diester formed, at condition of course that the acyloxy group in 3 is more easily saponifiable than the hexahydro-benzoyloxy group in 17.

   The 3-acyl-17-hexahydrobenzoyl estradiols which constitute intermediates of this embodiment can also be endowed with valuable estrogenic depot properties.



  The new esters can be used in solution in neutral vegetable oils, with or without the addition of a glycol, or in solution in other injectable solvents. The variable concentrations are limited only by the solubilized. It was thus possible to prepare solutions containing 10 mg of 3,17-di-hexahydrobenzoate and 20 mg of estradiol 3- and 17-monohexahydrobenzoate with this olive oil.

    It is sometimes desirable to add aluminum monostearate or 2-ethyl-hexanoate to oily solutions to obtain a thixotropic gel. The novel esters of estradiol which is obtained according to the present invention can also be administered in the form of microcrystalline aqueous suspensions or of emulsions in water or of solutions in a third solvent in the presence of a neutral emulsifying agent and not irritant. These preparations are made by known methods.



  The following examples show how the invention can be implemented. Example 1 Estradiol 3-hexahydrobenzoate <I> from </I> estradiol 7,

  5 g of estradiol in 50 <I> cc </I> of acetone and add at room temperature 26 cm3 of a 20% acetone solution of hexa-hydrobenzoyl chloride and 66 cm3 of normal sodium hydroxide.

The reaction mixture is poured into 200 cc of water, filtered off and dried under vacuum.

   In this way 8.75 g, or a yield of 81%, of estradiol 3-hexahydrobenzoate is obtained, which is recrystallized from ethanol.

   The product which is new melts at 91-92 ° C on block, [a120 = -I-53o 10 (c = 1% in chloroform). It is in the form of colorless platelets soluble in ether, acetone and chloroform, sparingly soluble in cold alcohol, insoluble in water.
