2,6,11,15-Tetramethyl-hexadecahexaen (2,4,6,10,12,14)-in- (8)-dial- ( 1,16) [C2o-in-dialdehyd] oder
2,6,11,15-Tetramethyl-hexadecaheptaen (2,4,6,8,10,12,14)-dial- (l, 16) [C2o-en-dialdehyd] mit einem I-Carbalkoxy-3-methyl-pentadien- (1, 3) yliden- (5)-triarylphosphin in einem inerten Lösungs- mittel kondensiert, aus dem gebildeten Addukt Triarylphosphinoxyd, gewünschtenfalls durch Erwär- men, abspaltet und im erhaltenen Ester die allenfalls vorhandene Dreifachbindung partiell hydriert und den Polyencarbonsäureester gewünschtenfalls isomerisiert.
Die genannten Ausgangsverbindungen können z. B. wie folgt hergestellt werden : Dehydro-, 6-apo-2-carotinal und ss-Apo-2-carotinal
Man kondensiert o-Ameisensäureäthylester mit Propenyläthyläther in Gegenwart von Bortrifluorid ätherat zu l, 1, 3,3-Tetraäthoxy-2-methylpropan.
Durch saure Hydrolyse gewinnt man den freien Methylmalondialdehyd in wässeriger Lösung und veräthert ihn durch azeotrope Destillation mit Benzol und Isopropylalkohol in Gegenwart von p-Toluolsulfosäure als Katalysator zum 3-Alkoxy-2-methyl-propen (2)-al- (1). Das 3-Alkoxy-2-methyl-propen- (2)-al- (l) einerseits und ss-Cl9-Aldehyd anderseits werden mittels einer metallorganischen Reaktion mit Acetylen kondensiert, das gebildete Acetylendiol mit Säure behandelt, das erhaltene Dehydro-ss-apo-4-carotinal durch aufeinanderfolgende Acetalisierung, Kondensation mit einem Vinyläther und Behandlung mit Säure in Dehydro--apo-3-carotinal übergeführt, diese Verbindung durch aufeinanderfolgende Acetali sierung,
Kondensation mit einem Propenyläther und Behandlung mit Säure in Dehydro-ss-apo-2-carotinal übergeführt. Durch Partialhydrierung an der Dreifachbindung, z. B. mittels Lindlar-Katalysator, erhält man das ss-Apo-2-carotinal.
I-Carbalkoxy-3-methyl-pentadien- (1, 3)-yliden (5)-triarylphosphin
Es wird eine Lösung von Lithiumacetylid in flüssigem Ammoniak hergestellt, eine Lösung von ss-Acetyl-acrylsäurealkylester in Äther zugegeben und das Reaktionsgemisch 4 Stunden unter Rückfluss gehalten. Man verdünnt mit Äther, lässt unter Durchleiten von Stickstoff das Ammoniak verdampfen, hydrolysiert mit gesättigter Ammoniumchloridlösung und extrahiert die wässerige Phase mit Essigester. Die organischen Auszüge werden getrocknet und unter Vakuum eingeengt. Die gewonnene sirupöse Substanz wird in Essigester gelöst und in Gegenwart von Lindlar-Katalysator unter Normalbedingungen zum 1-Carbalkoxy-3-methyl-3-hydroxy-pentadien- (1, 4) hydriert.

Der erhaltene Ester wird in einem inerten Lösungsmittel mit einem Halogenierungsmittel behandelt und in das l-Carbalkoxy-3-methyl-5-halogen- pentadien- (1, 3) übergeführt. Zum Beispiel führt die Behandlung des Methylesters mit Phosphortribromid in absolutem Ather zum 1-Carbomethoxy-3-methyl- 5-brom-pentadien- (1, 3), n29 = 1,5385. Die erhaltene Halogenverbindung wird nacheinander in einem inerten Lösungsmittel mit einem Triarylphosphin kondensiert und mit einem halogenwasserstoffbinden- den Mittel, z. B. einem Natriumalkoholat, behandelt.
So erhält man aus dem 1-Carbomethoxy-3-methyl- 5-brom-pentadien- (1, 3) durch Kondensation mit Triphenylphosphin in Essigester das entsprechende Phosphoniumbromid, dessen Methylenchloridlösung beim Versetzen mit der äquivalenten Menge einer alkoholischen Natriummethylatlösung das 1-Carbo- methoxy-3-methyl-pentadien- (1,3)-yliden- (5)-triphenylphosphin liefert. Die Isolierung der Phosphine ist nicht erforderlich ; die die Phosphine enthaltenden Lösungen oder Suspensionen können für die erste Stufe des erfindungsgemässen Verfahrens direkt verwendet werden.
Zur Umsetzung der genannten Ausgangsstoffe verwendet man zweckmässig die äquivalente Menge der Komponenten oder einen tÇberschuss der Phosphinverbindung und arbeitet, wie gesagt, in einem inerten Lösungsmittel, wie Ather, Petroläther, Benzol, Methylenchlorid, Dioxan oder Tetrahydrofuran. Vorteilhaft vermischt man die Komponenten bei Raumtemperatur, wobei man zweckmäRig die überstehende Luft durch ein inertes Gas, wie z. B. Stickstoff, ersetzt. Die Reaktion erfolgt spontan bei der Zugabe einer Lösung des Aldehydes zu einer Lösung oder Suspension der Phosphinverbindung. Je nach Wahl der Lösungsmittel bleiben die gebildeten Kondensa tionsprodukte in Lösung oder sie fallen aus.
Verwendet man Methylenchlorid als Lösungsmittel, so sind sowohl die Ausgangsprodukte als auch die entstehenden Kondensationsprodukte weitgehend in Lösung. Die Kondensationsprodukte zerfallen bei Raumtemperatur allmählich in die Polyenmono-bzw. Polyendicarbonsäureester und das entsprechende Triarylphosphinoxyd. Durch Erwärmen wird diese Reaktion beschleunigt. Die Zersetzung gelingt z. B. leicht durch mehrstündizes Kochen der Methylenchloridlösung unter Rückfluss.
Falls die erhaltenen Ester eine Dreifachbindung enthalten, werden sie der Partialhydrierung unterworfen und können ausserdem isomerisiert werden.
Dies kann in an sich bekannter Weise geschehen, z. B. durch Hydrierung einer Suspension des eine Dreifachbindung aufweisenden Esters in einem inerten Lösungsmittel, wie Essigester, Toluol, Petroläther usw., mit einem bleivergifteten Palladiumkatalysator.
Die anfallenden cis-Verbindungen können gewünsch- tenfalls nach an sich bekannten Methoden, vorzugsweise durch Erwärmen, zu den entsprechenden alltrans-Verbindungen isomerisiert werden. Diese cisbzw. all-trans-Verbindungen sind wenig toxisch. Sie zeichnen sich durch eine besonders intensive Rotfärbung aus und können in der Lebensmittelindustrie sowie in der Kosmetik verwendet werden. Zum Beispiel sind die ausgehend von Dehydro--apo-2- carotinal erhältlichen cis-Monocarbonsäureester wertvolle Färbezusätze für die Herstellung von Lippenstiftmassen mit neuen Farbnuancen.
Beispiel 1
Zu einer Lösung von 24 g Triphenylphosphin in 120 ml Essigester gibt man 19,5 g 1-Carbomethoxy- 3-methyl-5-brom-pentadien- (1, 3) und lässt 24 Stunden bei Zimmertemperatur stehen. Das Phosphoniumbromid fällt in Form eines Kristallkuchens an und kann nach Abgiessen der überstehenden Lösung direkt weiterverwendet werden. Man löst in 150 ml Methylenchlorid und versetzt unter Durchleiten von Stickstoff mit 31 ml 2n methanolischer Natriummethylatlösung. Man rührt 1 Stunde bei Zimmertemperatur und setzt dem Reaktionsgemisch rasch eine Lösung von 6 g C20-en-dialdehyd in 100 ml Methylenchlorid zu, rührt 1 Stunde bei Zimmertemperatur und kocht hierauf 5 Stunden unter Rück- fluss.
Nach dem Erkalten wäscht man die tiefrote Reaktionslösung mit 200 ml Wasser, trocknet und filtriert durch neutrales Aluminiumoxyd (Aktivitäts- stufe 5). Man wäscht mit Methylenchloridlösung nach, versetzt das klare Filtrat mit 150 ml Alkohol und destilliert Methylenchlorid bis zur beginnenden Kristallisation ab. Man lässt 2 Stunden in einem Eisbad stehen und nutscht hierauf den gebildeten Kristallbrei ab. Man wäscht mit kaltem Alkohol, mit tiefsiedendem Petroläther und trocknet im Vakuum bei 30-40 C. Man erhält so 5 g
1,26-Dicarbomethoxy-3,7,11,16,20,24-hexa methyl-hexacosa-tridecaen (1,3,5,7,9,11,13,15,17,19,21,23,25) vom Schmelzpunkt 199, C ; U. V.-Maxima 450,478, 513 mti ; EI% 2425, 3675,3380.
Durch Einengen der Mutterlauge und Chromatographieren des Rückstandes an neutralem, schwach aktivem Aluminiumoxyd in BenzoljMethylenchlorid erhält man noch eine weitere Menge C ; 4-Tridecaen-dicarbonsäure vom Schmelzpunkt 185-190 C.
Beispiel 2
Das gemäss den Angaben von Beispiel 1 aus 19,5 g 1-Carbomethoxy-3-methyl-5-brom-pentadien- (1,3) und 24 g Triphenylphosphin gewonnene Tri phenylphosphoniumbromid wird mittels Natriummethylatlösung in das entsprechende Ylid übergeführt und dieses mit 6 g C20-in-dialdehyd umgesetzt.
Reaktion und Aufarbeitung werden analog Beispiel 1 durchgeführt. Man erhält 5 g einer ersten Fraktion von 1, 26-Dicarbomethoxy-3,7,11,16,20,24 hexamethyl-hexacosa-dodecaen (1, 3,5,7,9,11,15,17,19,21,23,25)-in- (13) in Form feiner, braunroter Kristalle vom Schmelzpunkt 178-180 C ; U. V.-Maxima bei 457 und 484 m, c4 ; Ei m = 2740 und 2460. Aus den Mutterlaugen können auf gleiche Weise wie in Beispiel 1 weitere 2 g gewonnen werden.
Zur Partialhydrierung löst man 5,5 g 1, 26-Dicarbomethoxy-3,7,11,16,20,24 hexamethyl-hexacosa-dodecaen (1, 3,5,7,9,11,15,17,19,21,23,25)-in- (13) in 100 ml Essigester und hydriert mit 1 g Lindlar Katalysator (mit Hilfe von Blei desaktivierter Palladiumkatalysator in Gegenwart von Chinolin) bis zur Aufnahme von 250-270 ml Wasserstoff ; nach dem Abfiltrieren des Katalysators und Eindampfen der Lösung erhält man eine teils kristalline, teils amorphe Masse, die im U. V. folgende Absorptionen zeigt : bei 340 m, u, Etc% = 574 (cispeak) ; bei 518 mu, E I% m = 2610 (Hauptmaximum).
Durch mehrstündiges Kochen in Petroläther unter N2 verschwindet der Cis-peak und nach Umkristallisieren aus Methanol/Methylenchlorid erhält man 1, 26-Dicarbomethoxy-3,7,11,16,20,24 hexamethyl-hexacosa-tridecaen (1,3,5,7,9,11,13,15,17,19,21,23,25), das in jeder Beziehung mit dem Produkt aus Beispiel 1 identisch ist.
Beispiel 3
10 g 1-Carbomethoxy-3-methyl-5-brom-pentadien (1,3) werden wie in Beispiel 1 beschrieben mit 12 g Triphenylphosphin umgesetzt und anschliessend mit Natriummethylatlösung in das entsprechende Ylid übergeführt. Dieses wird mit 6 g ss-Apo-2-carotinal kondensiert. Reaktion und Aufarbeitung werden analog Beispiel 1 durchgeführt. Auf diese Weise erhält man 4 g 22- [2, 6,6-Trimethyl-cyclohexen- (l)-yl]-l-carbo methoxy-3,7,11,16,20-pentamethyl-docosa undecaen- (1, 3,5,7,9,11,13,15,17,19,21) in Form feiner, schwärzlichroter Kristalle. U. V. Maxima 493 m, und 527 m ; E = 2890 und 2260. Aus den Mutterlaugen können weitere Mengen des Farbstoffes isoliert werden.
Beispiel 4
10 g 1-Carbomethoxy-3-methyl-5-brom-pentadien- (1, 3) werden wie in Beispiel 1 beschrieben mit 12 g Triphenylphosphin umgesetzt und anschliessend mit Natriummethylatlösung in das entsprechende Ylid übergeführt. Dieses wird mit 6 g Dehydro--apo-2- carotinal kondensiert. Reaktion und Aufarbeitung werden analog Beispiel 1 durchgeführt. Auf diese Weise erhält man 5 g
22- [2, 6,6-Trimethyl-cyclohexen-(l)-yl]-l-carbo methoxy-3,7,11,16,20-pentamethyl-docosa decaen- (1, 3,5,7,9,11,15,17,19,21)-in- (13) in Form feiner, bräunlicher Kristalle vom Schmelzpunkt 191-192 C. U. V.-Maxima bei 464 und 494 m, ; ElC% = 2430 und 1840.
1 g des erhaltenen C37-in-Säuremonomethylesters wird in 20 ml Essigester suspendiert und mit 0,5 g Lindlar-Katalysator in Gegenwart von 0,02 ml Chinolin hydriert, bis die Hydrierung nach Aufnahme von etwa 75 ml Wasserstoff praktisch zum Stillstand gekommen ist. Man filtriert vom Katalysator ab, wäscht den Katalysator mit Methylenchlorid aus und dampft die tiefrot gefärbte Lösung unter 35 C im Teilvakuum ein. Der Rückstand zeigt im U. V.-Licht Absorptionsmaxima bei 380 m, (cispeak), 490,521 ; Ei = 735,1320,938.
Zur Isomerisierung versetzt man den erhaltenen C37-Cis-monosäuremethylester mit 20 ml Petroläther (Siedepunkt 80-110 C) und kocht während 1 Stunde unter Rückfluss. Man lässt über Nacht bei 20 C stehen und nutscht hierauf die gebildeten Kristalle ab. Man erhält auf diese Weise 0,6 g
22- [2, 6,6-Trimethyl-cyclohexen- (l)-yl]-l-carbo methoxy-3,7,11,16,20-pentamethyl-docosa undecaen- (1, 3,5,7,9,11,13,15,17,19,21) vom Schmelzpunkt 171-172 C ; U. V.-Maxima bei 493,526 und eine Schulter bei 470 m, ; El°/= = 2890 und 2260.
Claims (1)
- PATENTANSPRUCH I Verfahren zur Herstellung von Polyencarbon- säureestern der Formel EMI3.1 worin Ri einen 1-Carbalkoxy-3-methyl-buten- (1, 3)-yl (4)-rest bedeutet und R2 die Bedeutung von R hat oder der 2,6,6-Trimethyl-cyclohexen-(l)-yl-(l)-rest ist, dadurch gekennzeichnet, dal3 man 17- [2, 6,6-Trimethyl-cyclohexen-(l)-yl-(l)] 2,6,11,15-tetramethyl-heptadecaheptaen (2,4,6,10,12,14,16)-in- (8)-al- (l) ; 17- [2, 6,6-Trimethyl-cyclohexen- (l)-yl- (l)] 2,6,11,15-tetramethyl-heptadecaoctaen (2,4,6,8,10,12,14,16)-al- (l) ;2,6,11,15-Tetramethyl-hexadecahexaen (2,4,6,10,12,14)-in- (8)-dial- (1,16) oder 2,6,11,15-Tetramethyl-hexadecaheptaen (2,4,6,8,10,12,14)-dial- (1,16) mit einem 1-Carbalkoxy-3-methyl-pentadien-(l, 3) yliden- (5)-triarylphosphin in einem inerten Lösungs- mittel kondensiert, aus dem gebildeten Addukt Triarylphosphinoxyd abspaltet und im erhaltenen Ester die allenfalls vorhandene Dreifachbindung partiell hydriert.UNTERANSPRUCHE 1. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man die Kondensation in Methylenchlorid als Lösungsmittel durchführt und die Lösung des erhaltenen Reaktionsproduktes erwärmt.2. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man die Abspaltung von Triarylphosphinoxyd durch Erwärmung erwirkt.3. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man die erhaltenen Ester isomerisiert.PATENTANSPRUCH II Verwendung der nach dem Verfahren gemäss Patentanspruch I hergestellten Polyencarbonsäureester zur Färbung von Lebensmitteln oder kosmetischen Zubereitungen.
