Verfahren zur Herstellung von 2-Hydroxy-1,2,3,4,6,7-hexahydro-11bH-benzo[a]chinolizinen Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von 2-Hydroxy-1,2,3,4,6,7-hexa- hydro-11bH-benzo[a]chinolizinen, welches dadurch gekennzeichnet ist, dass man ein 1-(,l-Phenyl-äthyl)- 2-oxo-4-alkylendioxy-piperidin,
in welchem die 5-Stel- lung des Piperidinkerns durch einen Alkyl- oder Aral- kylrest substituiert und der Phenylkern in mindestens einer der Stellungen 2, 3 und 4 durch eine Alkoxy- gruppe oder in zwei der Stellungen 2, 3 und 4 durch eine Alkylendioxygruppe substituiert ist, in Gegen wart von Phosphoroxychlorid cyclisiert,
sodann die Alkylendioxygruppe im erhaltenen Cyclisierungspro- dukt in 2-Stellung mit Säure hydrolytisch abspaltet, das entstandene substituierte 2-Hydroxy-3,4,6,7-tetra- hydro-benzofa]chinoliziniumsalz mit Alkali behandelt und das erhaltene substituierte 2-Hydroxy-6,7-di- hydro-4H-benzo[a]chinolizin mittels Wasserstoff in Gegenwart eines Palladiumkatalysators hydriert.
Die als Ausgangsstoffe verwendeten substituierten 1- (ss - Phenyl- äthyl)- 2 - oxo-4-alkylendioxy-piperidine können nach der von BAN in Pharm. Bull. (Japan), 3, 53 (1955), beschriebenen Methode hergestellt wer den. Einige dieser Ausgangsstoffe, wie z. B. 1-[ss-(3,4 Dimethoxy-phenyl)-äthyl]-2-oxo-4-äthylendioxy-5-(n- butyl)-piperidin und 1-[ss-(3,4-Dimethoxy-phenyl)- äthyl]-2-oxo-4-äthylendioxy-5-isobutyl-piperidin, stel len neue Verbindungen dar.
Der Substituent in der 5-Stellung des Piperidin- kerns der Ausgangspiperidine ist, wie gesagt, ent weder ein Alkylrest, vorzugsweise ein niederer Alkyl- rest, z.
B. Methyl, Äthyl, Propyl, Isopropyl, n-Butyl oder Isobutyl, oder ein Aralkylrest, beispielsweise Benzyl oder Phenäthyl. Der Phenylkern der Aus- gangspiperidine ist in einer oder mehreren der Stel lungen 2, 3 und 4 vorzugsweise durch niederes Alkoxy, wie z.
B. Methoxy, Äthoxy, n-Propoxy, Iso- propoxy, oder in zwei der Stellungen 2, 3 und 4 vor- zugsweise durch eine niedere Alkylendioxygruppe, wie z.
B. Äthylendioxy, substituiert. Die Alkylendioxy- gruppe in der 4-Stellung des Piperidinkerns der Aus- gangspiperidine ist vorzugsweise eine Äthylendioxy- gruppe. Der Ringschluss des substituierten Ausgangspiperi- dins wird zweckmässigerweise nach der Methode von Bischler-Napieralski durchgeführt,
indem das substi tuierte Piperidin in einem wasserfreien inerten Lö sungsmittel, z. B. in trockenem Toluol, bei erhöhter Temperatur, z. B. bei der Temperatur eines sieden den Wasserbades, mit Phosphoroxychlorid behandelt wird. Das Cyclisierungsprodukt kann ohne weitere Reinigung für die nächste Stufe verwendet werden.
Die Behandlung des erhaltenen Cyclisierungspro- duktes mit Säure wird zweckmässigerweise mittels einer verdünnten Mineralsäure, z. B. verdünnter wäss riger Salzsäure, bei erhöhter Temperatur, vorzugs weise bei ,etwa 50 bis 60 , durchgeführt. Wenn für diese Behandlung Salzsäure verwendet wird, so erhält man die Chinolizinium-Verbindung in Form ihres Chlorids. Dieses Salz kann in entsprechende Chino- liziniumsalze mit anderen Anionen, z.
B. durch Be handlung des Chlorids mit Kaliumjodid in das ent sprechende Jodid, oder durch Ionenaustausch unter Verwendung von Ionenaustauschharzen, übergeführt werden.
Das bei der zweiten Stufe erhaltene substituierte 2-Hydroxy-3,4,6,7-tetrahydro-benzo[a]chinolizinium- salz wird, wie gesagt, mit Alkali behandelt, um die ent sprechend substituierte 2-Hydroxy-6,7-dihydro-4H- benzo[a]chinolizinbase zu bilden. Die Behandlung mit Alkali erfolgt zweckmässigerweise mittels eines verdünnten wässrigen Alkahmetallhydroxyds, z. B. verdünntem wässrigem Natriumhydroxyd, vorzugs weise etwa bei Raumtemperatur.
Als Katalysator für die Hydrierung der erhaltenen substituierten 2-Hydroxy-6,7-dihydro-4H-benzo[a]- chinolizinbase wird zweckmässig palladisierte Kohle verwendet, wobei diese Hydrierung in einem niederen alkoholischen Lösungsmittel, wie z. B. Methanol, bei Raumtemperatur oder bei erhöhter Temperatur, z. B. bei etwa 70 , durchgeführt werden kann. Die erfin dungsgemäss erhaltenen substituierten 2-Hydroxy-1,2, 3,4,6,7-hexahydro-11bH-benzo[a]chinolizinbasen kön nen durch Behandlung mit einer Säure, z. B. mit Chlor-, Brom- oder Jodwasserstoff, in entsprechende Additionssalze übergeführt werden.
Die nach dem erfindungsgemässen Verfahren er haltenen Verbindungen und ihre Säureadditionssalze können als Heilmittel oder als Zwischenprodukte für die Herstellung von Heilmitteln verwendet werden.
In den folgenden Ausführungsbeispielen sind alle Temperaturen in Celsiusgraden angegeben.
<I>Beispiel 1</I> 7,9 g kristallines 1-[ss-(3,4-Dimethoxy-phenyl)- äthyl]-2-oxo-4-äthylendioxy-5-äthyl-piperidin (Smp. 65-67 ), 25 ml trockenes Toluol und 20 ml Phosphor- oxychlorid werden auf einem Dampfbad während 21/1 Stunden erhitzt. Die Lösung wird unter vermin dertem Druck zur Trockne eingedampft, und der Rückstand wird mit 30 ml 2n Salzsäure behandelt und während einiger Minuten auf 50-60 erwärmt. Die Lösung wird filtriert, abgekühlt und mit einer wässrigen Lösung von Kaliumjodid behandelt.
Das in Form eines Öles ausgeschiedene 2-Hydroxy-3 äthyl-9,10-dimethoxy-3,4,6,7-tetrahydro-benzo[a]- chinoliziniumjodid wird mit Chloroform ausgezogen. Nach Entfernung des Chloroforms wird das Jodid aus einem Gemisch von Methanol und Diäthyläther in Form gelber Prismen kristallisiert (4,85 g); Smp. 171 bis 172,5 (Zers.), 240-24l (Zers.).
Das so erhaltene Jodid wird in Methanol gelöst und mit wässriger Natriumhydroxydlösung behandelt. Das erhaltene 2 - Hydroxy- 3 - äthyl- 9,10-dimethoxy- 6,7-dihydro-4H-benzo[a]chinolizin wird mit Benzol extrahiert. Durch Eindampfen des Extraktes erhält man die Base in Form eines gelben, festen Stoffes, der aus Essigsäureäthylester in Form gelber Prismen vom Smp. 168-170 kristallisiert. Diese Base wird in 15 ml Methanol gelöst und mittels palladisierter Kohle bei etwa 70 katalytisch hydriert.
Die Wasser stoffaufnahme erfolgt ziemlich rasch und erreicht 2 Mol. Nach dem Filtrieren des Hydrierungsgemisches wird das Methanol unter vermindertem Druck ent fernt und das erhaltene 2-Hydroxy-3-äthyl-1,2,3,4, 6,7-hexahydro-11bH-benzo[a]chinolizin aus einem Gemisch von Essigsäureäthylester und Petroläther in Form von Nadeln vom Smp. 155-157 kristallisiert. Mit Bromwasserstoffsäure setzt sich die Base zum Hydrobromid vom Smp. 231,5-233 um.
<I>Beispiel 2</I> 7,9 g kristallines 1-[ss-(3,4-Dimethoxy-phenyl)- äthyl]-2-oxo-4-äthylendioxy-5-äthyl-piperidin (Smp. 65-67 ), 25 ml trockenes Toluol und 20 ml Phos- phoroxychlorid werden auf einem Dampfbad während 21/4 Stunden erhitzt. Das Reaktionsgemisch wird fil triert, worauf das Filtrat unter vermindertem Druck zur Trockne eingedampft und der Rückstand mit 30 ml 2n Salzsäure behandelt und während einiger Minuten auf 50-60 erwärmt wird.
Die filtrierte ab gekühlte Lösung wird dann mit wässrigem Natrium hydroxyd behandelt. Dann wird das basische Material mit Benzol extrahiert und der Benzolextrakt einge dampft. Man erhält dabei 2-Hydroxy-3-äthyl-9,10- dimethoxy-6,7-dihydro-4H-benzo-[a]chinolizin, das aus Essigsäureäthylester in Form gelber Prismen vom Smp. 169-171,5 kristallisiert.
Die Base wird in 15 ml Methanol gelöst und mit tels palladisierterKohle bei 20 katalytisch hydriert. Die W asserstoffaufnahme erfolgt ziemlich rasch und erreicht 2 Mol. Nach dem Filtrieren des Hydrierungsgemisches wird das Methanol unter vermindertem Druck ent fernt und das erhaltene 2-Hydroxy-3-äthyl-1,2,3,4,6,7- hexahydro-11bH-benzo[a]chinolizin aus einem Ge misch von Essigsäureäthylester und Petroläther in Form von Nadeln vom Smp. 155-157 kristallisiert.
Mit Bromwasserstoffsäure setzt sich diese Base zu einem Hydrobromid vom Smp. 231,5-233 um. <I>Beispiel 3</I> 4,37 g 1-[/3-(3,4-Dimethoxy-phenyl)-äthyl]-2-oxo- 4-äthylendioxy-5-(n-butyl)-piperidin werden in 20 ml trockenem Toluol und 10 ml Phosphoroxychlorid ge löst. Die Lösung wird während 3 Stunden auf einem siedenden Wasserbad erhitzt. Dann werden das To luol und das Phosphoroxychlorid im Vakuum ent fernt. Der Rückstand wird in heisser 2n Salzsäure ge löst.
Die wässrige, saure Lösung wird filtriert, mittels 2n Natriumhydroxydlösung alkalisch gestellt und mit Benzol extrahiert. Durch Abdampfen des Benzols er hält man 2-Hydroxy-3-(n-butyl)-9,10-dimethoxy-6,7- dihydro-4H-benzo[a]chinolizin in Form eines gelben Stoffes, der aus Essigsäureäthylester in Form von Nadeln (15g) vom Smp. 180-182 kristallisiert.
Die analog zu den Angaben in Beispiel 2 durch geführte Hydrierung der Base ergibt 2-Hydroxy-3-(n butyl)-9,10-dimethoxy-1,2,3,4,6,7-hexahydro-11bH- benzo[a]chinolizin vom Smp. 125-127,5 (aus Essig ester-Petroläther). Das Hydrochlorid schmilzt bei 229 bis 231 .
<I>Beispiel 4</I> 50 g [ss-(3,4-Dimethoxy-phenyl)-äthyl]-2-oxo-4- äthylendioxy-5-isobutyl-piperidin werden in 20 ml trockenem Toluol und 10 ml Phosphoroxychlorid ge löst, worauf die Lösung auf einem siedenden Wasser bad während 3 Stunden erhitzt wird. Dann werden das Toluol und das Phosphoroxychlorid im Vakuum entfernt. Der Rückstand wird in heisser 2n Salzsäure gelöst. Die wässrige, saure Lösung wird filtriert, mit tels 2n Natriumhydroxydlösung alkalisch gestellt und mit Benzol extrahiert.
Durch Entfernen des Benzols erhält man 2-Hydroxy-3-isobutyl-9,10-dimethoxy- 6,7-dihydro-4H-benzo[a]chinolizin, das sich aus einem Gemisch von Essigsäureäthylester und Petrol- äther (Siedebereich 40-60 ) in Form gelber Prismen vom Smp. 154-157 abscheidet (1,8 g).
Die analog zu den Angaben im Beispiel 2 durch geführte Hydrierung der Base ergibt 2-Hydroxy-3-iso butyl-9,10-dimethoxy-1,2,3,4,6,7-hexahydro-11bH- benzo[a]chinolizin vom Smp. 167-169 . Das Hydro- chlorid schmilzt bei 206-207 .
Process for the preparation of 2-hydroxy-1,2,3,4,6,7-hexahydro-11bH-benzo [a] quinolizines The present invention relates to a process for the preparation of 2-hydroxy-1,2,3,4, 6,7-hexa-hydro-11bH-benzo [a] quinolizines, which is characterized in that a 1 - (, l-phenyl-ethyl) - 2-oxo-4-alkylenedioxy-piperidine,
in which the 5-position of the piperidine nucleus is substituted by an alkyl or aralkyl radical and the phenyl nucleus is substituted in at least one of the positions 2, 3 and 4 by an alkoxy group or in two of the positions 2, 3 and 4 by an alkylenedioxy group is substituted, cyclized in the presence of phosphorus oxychloride,
then the alkylenedioxy group in the cyclization product obtained is hydrolytically split off with acid in the 2-position, the substituted 2-hydroxy-3,4,6,7-tetrahydrobenzofa] quinolizinium salt formed is treated with alkali and the substituted 2-hydroxy- 6,7-dihydro-4H-benzo [a] quinolizine is hydrogenated using hydrogen in the presence of a palladium catalyst.
The substituted 1- (ss-phenyl-ethyl) -2-oxo-4-alkylenedioxy-piperidines used as starting materials can be prepared by the method described by BAN in Pharm. Bull. (Japan), 3, 53 (1955) . Some of these raw materials, such as B. 1- [ss- (3,4 dimethoxyphenyl) ethyl] -2-oxo-4-ethylenedioxy-5- (n-butyl) piperidine and 1- [ss- (3,4-dimethoxyphenyl ) - ethyl] -2-oxo-4-ethylenedioxy-5-isobutyl-piperidine, represent len new compounds.
The substituent in the 5-position of the piperidine nucleus of the starting piperidines is, as I said, ent neither an alkyl radical, preferably a lower alkyl radical, e.g.
B. methyl, ethyl, propyl, isopropyl, n-butyl or isobutyl, or an aralkyl radical, such as benzyl or phenethyl. The phenyl nucleus of the starting piperidines is in one or more of the positions 2, 3 and 4 preferably by lower alkoxy, such as.
B. methoxy, ethoxy, n-propoxy, isopropoxy, or in two of the positions 2, 3 and 4 preferably by a lower alkylenedioxy group, such as.
B. ethylenedioxy substituted. The alkylenedioxy group in the 4-position of the piperidine nucleus of the starting piperidines is preferably an ethylenedioxy group. The ring closure of the substituted starting piperidine is expediently carried out according to the method of Bischler-Napieralski,
by the substituted piperidine in an anhydrous inert Lö solvent such. B. in dry toluene, at an elevated temperature, e.g. B. at the temperature of a boiling water bath, treated with phosphorus oxychloride. The cyclization product can be used for the next stage without further purification.
The treatment of the resulting cyclization product with acid is expediently carried out using a dilute mineral acid, e.g. B. dilute aqueous hydrochloric acid, at elevated temperature, preferably at, about 50 to 60 carried out. If hydrochloric acid is used for this treatment, the quinolizinium compound is obtained in the form of its chloride. This salt can be converted into corresponding quinolizinium salts with other anions, e.g.
B. by treating the chloride with potassium iodide in the ent speaking iodide, or by ion exchange using ion exchange resins, are converted.
The substituted 2-hydroxy-3,4,6,7-tetrahydro-benzo [a] quinolizinium salt obtained in the second stage is, as I said, treated with alkali to form the correspondingly substituted 2-hydroxy-6,7- to form dihydro-4H-benzo [a] quinolizine base. The treatment with alkali is conveniently carried out by means of a dilute aqueous alkali metal hydroxide, e.g. B. dilute aqueous sodium hydroxide, preferably at about room temperature.
As a catalyst for the hydrogenation of the substituted 2-hydroxy-6,7-dihydro-4H-benzo [a] - quinolizine base is conveniently used palladized carbon, this hydrogenation in a lower alcoholic solvent, such as. B. methanol, at room temperature or at an elevated temperature, e.g. B. at about 70, can be performed. The substituted 2-hydroxy-1,2, 3,4,6,7-hexahydro-11bH-benzo [a] quinolizine bases obtained according to the invention can be obtained by treatment with an acid, e.g. B. with chlorine, bromine or hydrogen iodide, be converted into corresponding addition salts.
The compounds obtained by the process according to the invention and their acid addition salts can be used as medicinal products or as intermediates for the preparation of medicinal products.
In the following working examples, all temperatures are given in degrees Celsius.
<I> Example 1 </I> 7.9 g of crystalline 1- [ss- (3,4-dimethoxyphenyl) ethyl] -2-oxo-4-ethylenedioxy-5-ethyl-piperidine (melting point 65- 67), 25 ml of dry toluene and 20 ml of phosphorus oxychloride are heated on a steam bath for 21/1 hours. The solution is evaporated to dryness under reduced pressure, and the residue is treated with 30 ml of 2N hydrochloric acid and heated to 50-60 for a few minutes. The solution is filtered, cooled and treated with an aqueous solution of potassium iodide.
The 2-hydroxy-3-ethyl-9,10-dimethoxy-3,4,6,7-tetrahydro-benzo [a] -quinolizinium iodide which has separated out in the form of an oil is extracted with chloroform. After the chloroform has been removed, the iodide is crystallized from a mixture of methanol and diethyl ether in the form of yellow prisms (4.85 g); M.p. 171 to 172.5 (dec.), 240-24l (dec.).
The iodide obtained in this way is dissolved in methanol and treated with aqueous sodium hydroxide solution. The 2-hydroxy-3-ethyl-9,10-dimethoxy-6,7-dihydro-4H-benzo [a] quinolizine obtained is extracted with benzene. Evaporation of the extract gives the base in the form of a yellow, solid substance which crystallizes from ethyl acetate in the form of yellow prisms with a melting point of 168-170. This base is dissolved in 15 ml of methanol and catalytically hydrogenated at about 70 using palladised charcoal.
The absorption of hydrogen takes place fairly quickly and reaches 2 mol. After filtering the hydrogenation mixture, the methanol is removed under reduced pressure and the 2-hydroxy-3-ethyl-1,2,3,4, 6,7-hexahydro-11bH obtained -benzo [a] quinolizine crystallized from a mixture of ethyl acetate and petroleum ether in the form of needles with a melting point of 155-157. With hydrobromic acid, the base is converted to the hydrobromide with a melting point of 231.5-233.
<I> Example 2 </I> 7.9 g of crystalline 1- [ss- (3,4-dimethoxyphenyl) -ethyl] -2-oxo-4-ethylenedioxy-5-ethyl-piperidine (m.p. 65- 67), 25 ml of dry toluene and 20 ml of phosphorus oxychloride are heated on a steam bath for 21/4 hours. The reaction mixture is filtered, whereupon the filtrate is evaporated to dryness under reduced pressure and the residue is treated with 30 ml of 2N hydrochloric acid and heated to 50-60 for a few minutes.
The filtered, cooled solution is then treated with aqueous sodium hydroxide. Then the basic material is extracted with benzene and the benzene extract is evaporated. This gives 2-hydroxy-3-ethyl-9,10-dimethoxy-6,7-dihydro-4H-benzo [a] quinolizine, which crystallizes from ethyl acetate in the form of yellow prisms with a melting point of 169-171.5.
The base is dissolved in 15 ml of methanol and catalytically hydrogenated at 20 using palladized carbon. The uptake of hydrogen takes place fairly quickly and reaches 2 mol. After the hydrogenation mixture has been filtered off, the methanol is removed under reduced pressure and the 2-hydroxy-3-ethyl-1,2,3,4,6,7-hexahydro-11bH obtained is removed -benzo [a] quinolizine crystallized from a mixture of ethyl acetate and petroleum ether in the form of needles with a melting point of 155-157.
This base reacts with hydrobromic acid to form a hydrobromide with a melting point of 231.5-233. <I> Example 3 </I> 4.37 g of 1 - [/ 3- (3,4-dimethoxyphenyl) ethyl] -2-oxo-4-ethylenedioxy-5- (n-butyl) piperidine dissolved in 20 ml of dry toluene and 10 ml of phosphorus oxychloride. The solution is heated on a boiling water bath for 3 hours. Then the toluene and the phosphorus oxychloride are removed in vacuo. The residue is dissolved in hot 2N hydrochloric acid.
The aqueous, acidic solution is filtered, made alkaline with 2N sodium hydroxide solution and extracted with benzene. By evaporating off the benzene he keeps 2-hydroxy-3- (n-butyl) -9,10-dimethoxy-6,7-dihydro-4H-benzo [a] quinolizine in the form of a yellow substance, which is obtained from ethyl acetate in the form of Needles (15g) of m.p. 180-182 crystallized.
The hydrogenation of the base carried out analogously to the information in Example 2 gives 2-hydroxy-3- (n butyl) -9,10-dimethoxy-1,2,3,4,6,7-hexahydro-11bH-benzo [a ] quinolizine of m.p. 125-127.5 (from ethyl acetate-petroleum ether). The hydrochloride melts at 229 to 231.
<I> Example 4 </I> 50 g of [ss- (3,4-dimethoxyphenyl) ethyl] -2-oxo-4-ethylenedioxy-5-isobutyl-piperidine are dissolved in 20 ml of dry toluene and 10 ml of phosphorus oxychloride ge dissolved, whereupon the solution is heated on a boiling water bath for 3 hours. Then the toluene and the phosphorus oxychloride are removed in vacuo. The residue is dissolved in hot 2N hydrochloric acid. The aqueous, acidic solution is filtered, made alkaline with 2N sodium hydroxide solution and extracted with benzene.
Removal of the benzene gives 2-hydroxy-3-isobutyl-9,10-dimethoxy-6,7-dihydro-4H-benzo [a] quinolizine, which is made from a mixture of ethyl acetate and petroleum ether (boiling range 40-60 ) in the form of yellow prisms with a melting point of 154-157 (1.8 g).
The hydrogenation of the base carried out analogously to the details in Example 2 gives 2-hydroxy-3-isobutyl-9,10-dimethoxy-1,2,3,4,6,7-hexahydro-11bH-benzo [a] quinolizine from m.p. 167-169. The hydrochloride melts at 206-207.
