Description
Verfahren zur Herstellung von stabilen Vitamin-A-Estern Bekanntlich ist Vitamin A gegenüber Sauerstoff äusserst empfindlich. So zersetzt es sich z. B. unter dem Einfluss von Luftsauerstoff beim Stehenlassen bereits innerhalb von wenigen Stunden. Auch die bisher meist verwendeten Ester von Vitamin A, wie das Acetat und das Palmitat, weisen eine ähnlich hohe Unbeständigkeit auf. Durch Zugabe von Antioxydantien, z. B. von a-Tocopherol, gelingt es zwar, die Stabilität des Vitamins A zu verbessern, doch ist der durch derartige Massnahmen erreichbare Effekt für viele Zwecke noch völlig ungenügend. Es wurde nun überraschend gefunden, dass Acylaminogruppen tragende Carbonsäureester des Vitamins A, selbst ohne Zusatz von Antioxydantien, eine erstaunlich hohe Stabilität besitzen. Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung von stabilen Vitamin Estern, welches dadurch gekennzeichnet ist, dass man Vitamin A oder in der OH-Gruppe abgewandelte Derivate davon mit Acylaminoresten tragenden aliphatischen, cycloaliphatischen, araliphatischen oder aromatischen Carbonsäuren oder in der -Carboxyl- gruppe abgewandelten Derivaten oder Azlactonen davon umsetzt. Der Acylaminosubstituent kann sich an beliebiger Stelle des betreffenden Säurerestes befinden. Beispielsweise können als Vertreter von stabilenVitamin A-Estern die entsprechenden Ester der nachstehend aufgeführten Säuren erwähnt werden: a-Aminosäuren, wie Serin, Methionin, a-Alanin, fl-Phenyl-alanin, Glycin; ss-Aminosäuren, wie ss-Alanin; y-Aminosäuren, wie y-Amulo-buttersäure; araliphatische Aminosäuren, wie pAmino zimtsäure, und aromatische Aminosäuren, wie p-Amino-benzoe säure und p-Amino-salicylsäure. Die Aminoreste dieser Carbonsäuren können sowohl durch einbasische als auch durch zweibasische Säurereste substituiert sein. Zweckmässig weist der Acykest die Bedeutung von Acetyl, Benzoyl oder Phthalyl auf. Die Acylreste können auch Substituenten, wie z. B. Halogenatome, Alkyl-, Nitro-, Amino-, Alkoxy- oder Hydroxylgruppen, tragen. Insbesondere haben sich die Ester des Vitamins A mit den nachstehend aufgeführten Säuren als äusserst beständig erwiesen: Hippursäure, p-Nitro-hippursäure, p-Chlor-hippursäure, Phthalylglycin, Benzoylmethionin, Acetylleucin, ss-Benzoyl-alanin und p-Acetan:inobenzoesäure. Als besonders geeignete reaktionsfähige Säurederivate der genannten Carbonsäuren haben sich die Säurehalogenide, insbesondere das Säurechlorid und die Säureanhydride, erwiesen. Falls die zur Veresterung verwendeten Carbonsäuren einen Nitrosubsti- tuenten aufweisen, kann dieser nachträglich z. B. durch katalytische Hydrierung zur Aminogruppe reduziert werden. Werden die gewünschten stabilen Ester durch Umesterung gewonnen, so verwendet man hierbei zweckmässig einen niederen Carbonsäureester des Vitamins A, z. B. Vitamin-A-Acetat, und erwärmt ihn mit einem niederalkoholischen Ester einer acylamino-substituierten Carbonsäure. Die Umesterung wird mit Vorteil in Gegenwart von Umesterungskatalysatoren; z. B. Alkalimetalihydroxyden oder Alkalimetallalkoholaten, unter kontinuierlicher Entfernung des als Nebenprodukt gebildeten niederalkoholischen Esters der niederen Carbonsäure durchgeführt. Falls zur Umesterung ein Ester einer nitrosubstituierten Carbonsäure verwendet wird, können die Niere gruppen, wenn erwünscht, nachträglich zu Aminogruppen reduziert werden. Die genannten stabilen Vitamin-A-Ester können, wie gesagt, auch durch Umsetzung von Azlactonen, z. B. von Azlactonen der Formel EMI2.1 oder der Formel EMI2.2 worin R und R"Wasserstoff oder gegebenenfalls Sauerstoff, Schwefel oder Stickstoff enthaltende Kohlenwasserstoffreste und R' gegebenenfalls Sauerstoff, Schwefel oder Stickstoff enthaltende Kohlen wasserstoftreste darstellen, mit Vitamin A erhalten werden. Beispiel 1 4 g Hippursäurechlorid werden in 50 ml absolutem Pyridin gelöst und mit 5,8 g Vitamin-A-Alkohol versetzt. Man lässt 5-6 Stunden stehen, giesst in 500 mI Wasser und schüttelt mit Essigester aus. Die Essigesterlösung wird nacheinander mit 3n Salzsäure, 2n Sodalösung und Wasser gewaschen, mit Natriumsulfat getrocknet, im Vakuum auf ein Volumen von etwa 30 ml eingedampft und mit 150 ml absolutem Äther verdünnt. Nach dem Abkühlen auf - 150 wird abgenutscht und der Rückstand aus 40 ml Acetonitril umiristallisiert. Der so erhaltene Hippursäureester des Vitamins A kristallisiert in verfilzten, gelblichen Nädelchen und schmilzt bei 123-124 unter Zersetzung. Beispiel 2 Unter Verwendung von 4,6 g p-Nitro-hippursäurechlorid anstelle des Hippursäurechlorides erhält man nach den Angaben in Beispiel 1 den p-Nitro- hippursäureester des Vitamins A, welcher nach dem Umkristallisieren aus Methanol bei 110-111 schmilzt. Beispiel 3 Unter Verwendung von 4,6 g p-Chlor-hippursäurechlorid anstelIe des Hippursäurechlorides erhält man nach den Angaben in Beispiel 1 den p-Chlorhippursäureester des Vitamins A, welcher nach dem Umkristallisieren aus Acetonitril bei 106-108 schmilzt. Beispiel 4 3,2 g 2- Phenyl - oxazolon - (5) (Azlacton der Hippursäure) werden zusammen mit 5,7 g Vitamin A-Alkohol in 50 mi Benzol gelöst und über Nacht bei Zimmertemperatur stehengelassen. Man wäscht die Benzollösung nacheinander mit 3n Salzsäure, 2n Sodalösung und Wasser, trocknet sie mit Natriumsulfat und engt im Vakuum auf etwa 10 ml ein. Nun verdünnt man mit 50 ml absolutem Äther und kühlt auf - 150 ab. Der beim Abnutschen erhaltene Niederschlag wird aus Acetonitril umkristallisiert. Der so erhaltene Hippursäureester des Vitamins A schmilzt bei 123-124 .
Claims (1)
- PATENTANSPRUCH Verfahren zur Herstellung von stabilen Estern von Vitamin A, dadurch gekennzeichnet, dass man Vitamin A oder in der OH4ruppe abgewandelte Derivate davon mit Acylaminoreste tragenden aliphatischen, cycloaliphatischen, araliphatischen oder aromatischen Carbonsäuren oder in der Carboxygruppe abgewandelten Derivaten oder Azlactonen solcher Säuren umsetzt.UNTERANSPRÜCHE 1. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man als reaktionsfähige Derivate der genannten Carbonsäuren entsprechende Carbonsäurehalogenide verwendet.2. Verfahren nach Unteranspruch 1, dadurch gekennzeichnet, dass man Hippursäurechlorid, p-Nitrohippursäurechlorid, p-Chlor-hippursäurechlorid oder Phthalyl-glycylchlorid verwendet.3. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man einen niederen Carbonsäureester des Vitamins A durch Behandlung mit einem niederalkoholischen Ester der den Acylaminorest tragenden Carbonsäure umestert.4. Verfahren nach Unteranspruch 3, dadurch gekennzeichnet, dass man die Umesterung in Gegenwart eines Umesterungskatalysators, wie eines Alkali metalihydroxydes oder eines Alkalimetallalkoholates, vornimmt.5. Verfahren nach den Unteransprüchen 3 und 4, dadurch gekennzeichnet, dass der als Nebenprodukt gebildete niederalkoholische Ester der niederen Carbonsäure kontinuierlich entfernt wird.6. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man Vitamin A mit einem Azlacton der Formel EMI2.3 oder der Formel EMI2.4 worin R und R" Wasserstoff oder gegebenenfalls Sauerstoff, Schwefel oder Stickstoff enthaltende Kohlenwasserstoffreste und R' einen gegebenenfalls Sauerstoff, Schwefel oder Stickstoff enthaltenden 5 Kohlenwasserstoffrest darstellen, umsetzt.7. Verfahren nach Unteranspruch 6, dadurch ge kennzeichnet, dass man als Azlacton 2-Phenyl-oxazo von(5) verwendet.
