La présente invention a pour objet un procédé de préparation de nouveaux dérivés de l'oxazolidinone-2, de formule générale :

Ces nouveaux dérivés de l'oxazolidinone-2 sont préparés en faisant réagir un chlorure d'acide, éven- tuellement sous la configuration cis ou trans, racémi- que ou optiquement active de la formule générale suivante :

Br, par des radicaux comme CHS, OH, NO2, NH2, alcoxy inférieur ou méthylène dioxy,
laissant réagir ce chlorure sur un composé à caractère basique de formule générale suivante Y-AZ dans laquelle Z peut représenter un reste aminé aliphatique ou un reste hétérocyclique comme pipéri- dino, morpholino, pyrrolidino N-mÚthylpiperazino, N-pipéronyl-pipérazino ou analogues.
Il est bien entendu que si deux radicaux Ri et R3, situés sur les carbones 4 et 5 adjacents, sont différents et R2 et R4 identiques, ou si les 4 radicaux R1, R2,
R3, R4 sont tous diffÚrents, les dÚrivÚs obtenus existent sous deux formes cis et trans et font, à ce titre, partie de l'invention, ainsi que leur diastéréo-iso- mères.
II est avantageux d'opérer dans un solvant qui peut être un carbure aromatique tel que le benzène, ou un éther aliphatique ou hétérocyclique (tétra- hydrofurane) en présence d'un excès du composé Y-AZ ou d'une base tertiaire (triéthylamine ou pyridine) faisant fonction d'accepteur d'hydracide.
II est particulièrement avantageux d'ajouter à une solution du composé Y-AZ, choisi dans le solvant préféré, contenant ou non une des bases tertiaires précédemment citées, le chlorure d'acide dissous dans le même solvant. La réaction sera effectuée à une température peu élevée, comprise entre la température ambiante et une température de l'ordre de 700 C.
Les nouveaux dérivés de l'oxazolidinone-2, qui sont des bases fortes, peuvent être purifiés par des méthodes physiques, telles que cristallisation, chromatographie, ou chimiques, telles que formation de sels avec des acides minéraux ou organiques, cristallisation de ceux-ci dans un solvant approprié et dé- composition de ces derniers en milieu alcalin. Dans ces opérations, la nature de l'anion est indifférente, dans la mesure où il conduit à un sel bien défini et aisément cristallisable.
Ces bases seront, en général, conservées sous forme de sels d'addition.
Comme sels d'addition avec les acides minéraux, on peut citer, en outre, les chlorhydrate, bromhydrate, sulfate, phosphate, méthane-sulfonate et, avec les acides organiques, les acétate, propionate, maléate, fumarate, succinate, benzoate, tartrate, malate, oxalate.
Les exemples suivants illustrent l'invention. Les points de fusion indiqués ont été déterminés au bloc
Kofler. Les rendements sont donnés par rapport au chlorure d'acide de départ.
Exemple 1
(d, l)-cis (oxo-2 mÚthyl-4 phÚnyl-5 oxazolidinyl-3) carboxylate de dimethylamino-2'ethyle.
Une solution de 4, 5 g de chlorure de l'acide (d, l), cis (oxo-2 méthyl-4 phÚnyl-5 oxazolidinyl-3) carboxylique fondant à 115 dans 90 cm-3 de benzène anhydre est ajoutée, goutte à goutte, en 25 minutes et en refroidissant à 10 , à une solution de 3, 35 g de diméthylaminoéthanol dans 10 cm3 de benzène anhydre. Lorsque l'addition est terminée, on laisse le mélange sous agitation pendant 2 1/2 heures à température ordinaire. Au bout de ce temps, on traite à l'eau, décante et extrait plusieurs fois la couche benzénique avec de l'acide chlorhydrique à 7 /o. Les portions acides sont réunies ; on lave plusieurs fois à l'éther puis alcalinise à pH 9 avec du carbonate de potassium et extrait plusieurs fois à l'Úther.
Les extraits éthérés réunis sont lavés plusieurs fois à l'eau et sèches sur carbonate de potassium. Après évapo- ration sous pression réduite, à basse température, on obtient finalement 5, 5 g de base fondant à 540. Rendement : 100 /o.
Par addition d'une solution d'acide maléique à la solution de la base précédente dans l'éthanol, on obtient le maléate acide fondant à 138 . Le chlorure d'acide de départ a été préparé par action d'une solution toluénique de phosgène sur une suspension de (d, l), cis méthyl-4 phÚnyl-5 oxazolidinone-2 fondant à 146 , en présence de pyridine, dans le benzène.
De la même façon, la titulaire a préparé les composés suivants : a) (d, l) cis (oxo-2 mÚthyl-4 phÚnyl-5 oxazolidinyl-3)

carboxylate de dimethylamino-2'ethyle, dont
le fumarate acide fond à 170 , à partir du di méthylamino éthanol et du chlorure de l'acide (d, 1) (oxo-2 méthyl-4 diphÚnyl-5, 5 oxazolidinyl3) carboxylique (huileux) préparé comme suit : à une solution de 8, 2 g de méthyl-4 diphÚnyl-5, 5 oxazolidinone-2, fondant à 2320, dans 200 cm3 de tétrahydrofurane anhydre, on ajoute en une fois 1, 87 g d'une suspension de HNa à 50 ouzo dans l'huile minérale. On chauffe ensuite 1 heure à reflux.
La solution obtenue est refroidie à 200 et coulée en 15 mn dans 80 g d'une solution toluéni- que de phosgène à 20 %, à + 10 . Il se forme un précipité de chlorure de sodium : on agite une heure à l'ébullition, refroidit, filtre le sel et Úvapore sous pression réduite à basse température.
Le résidu visqueux obtenu est trituré dans l'éther ; on filtre l'insoluble constitué par du produit de départ n'ayant pas réagi et évapore le filtrat sous pression réduite à basse température. Le résidu huileux obtenu est utilisé tel quel pour la réaction d'estérification.
L'oxazolidinone de départ a été préparé par dégradation de Curtius à partir de la (diphényl- 3, 3 hydroxy-3 méthyl-2 propionyl) hydrazine, fondant à 2050, elle-même obtenue par réaction de l'hydrate d'hydrazine sur le diphÚnyl-3, 3 hydroxy-3 méthyl-2 propionate de méthyle dans l'éthanol à reflux.
(oxo-2 phenyl-S oxazolidinyl-3) carboxylate de dimethyl amino Úthyl, fumarate acide. A une solution de 12, 7 g de diméthylaminoéthanol dans 50 cm3 de benzène sec, on ajoute en 30 minutes, 13, 9 g de chlorure de l'acide (d, l) (oxo-2 phényl- 5 oxazolidinyl-3) carboxylique en solution dans 200 cm3 de benzène sec. La température intérieure croît de 20 à 30 , tandis qu'il se forme un abondant précipité. On chauffe le mélange 1 heure à 50O, puis refroidit, et traite avec 2 X 100 cm3 d'eau.
La solution benzénique est ensuite extraite plusieurs fois avec de l'acide chlorhydrique à 10 /o. On réunit les solutions acides et alcalinise avec du carbonate de potassium ; on extrait plusieurs fois à l'éther et sèche les solutions éthérées sur carbonate de potassium. Après évaporation de l'éther sous pression réduite, on obtient 15 g de base huileuse dont le fumarate acide fond à 137 .
Le chlorure d'acide de départ, non isolé, a été préparé par action d'un excès de phosgène sur une suspension toluénique de sel de sodium de la phÚnyl-5 oxazolidinone-2 fondant à 890.
(oxo-2 diphÚnyl-5, 5 oxazolidinyl-3) carboxylate de dimethylamino-ethyle, fondant à 117 . Le chlorhydrate correspondant fond à 210 .
(oxo-2 diphenyl-S, S oxazolidinyl-3) carboxylate de (mÚthyl-4' pipÚrazinyl-1') Úthyle, fondant à 200-202O. Le bis méthane sulfonate fond à 238¯ (déc.).
(d, l) (oxo-2 phÚnyl-5 Úthyl-5 oxazolidinyl-3) carboxylate de dimethylaminoethyle, dont le fumarate acide correspondant fond à 1600. k) (d, l) cis (oxo-2 mÚthyl-4 cyclohexyl-5 oxazolidi
nyl-3) carboxylate de dimÚthylaminoÚthyle, dont
le chlorhydrate fond à 165 .
Le chlorure d'acide de départ, non isole, a
été préparé à partir de la (d, l) cis (méthyl-4 cy
clohexyl-5) oxazolidinone-2 fondant à 123o, elle
même préparée par cyclisation au moyen de
phosgène, en présence de potasse du (d, l) cis
cyclohexyl-1 amino-2 propanol-1, dont le di
chlorhydrate fond à 217 .
1) (d, l) cis (oxo-2 phényl-4 méthyl-5 oxazolidinyl-
3) carboxylate de diméthylaminoéthyle, fondant
à 770. Le fumarate acide correspondant fond à
149-150 . Le chlorure d'acide de départ, non
isolé, a été préparé à partir de la (d, l) cis (mé
thyl-5 phÚnyl-4) oxazolidinone-2, fondant à
1060, elle-même préparée par cyclisation au
moyen de carbonate d'éthyle, en présence d'une
trace de méthylate de sodium, du phÚnyl-1 amino
1 propanol-2.
m) (oxo-2 pentamÚthyl¯ne-5, 5 oxazolidinyl-3) carb
oxylate de diméthylaminoéthyle, dont le chlor

fondant à 102 , elle-même préparée par action
de l'hydrazine sur l'hydroxy-3 caproate d'éthyle. p) (d, l) (oxo-2 benzyl-5 oxazolidinone-3) carboxy
late de dimÚthylaminoÚthyle, dont le fumarate
acide fond à 150 (avec déc.).
Le chlorure d'acide de départ, non isolé, a
été préparé à partir de la benzyl-5 oxazolidinone
2 fondant à 103 , elle-même préparée par dégra-
dation de Curtius de la benzyl-3 hydroxy-3 pro
pionyl hydrazine fondant à 102 , elle-même pré
parée par action de l'hydrazine sur le benzyl-3
hydroxy-3 propionate d'éthyle (Eb/2 140-145 ). q) (d, l) (methylenedioxy-3', 4' phenyl)-5 oxo-2 oxa-
zolidinyl-3 carboxylate de dimethylaminoethyle,
dont le chlorhydrate fond à 187 .
Le chlorure d'acide de départ, non isolé, a
été préparé à partir de la (méthylène-dioxy-3', 4',
phÚnyl)-5 oxazolidinone-2 fondant à 128 , elle
même préparée par cyclisation au moyen du
phosgène, en présence de potasse, du (méthylène
dioxy-3', 4' phényl)-1 amino-2 Úthanol-1 dont le
chlorhydrate fond à 196o, lui-même préparé par
réduction au moyen de AlkH4 dans le tétrahydro-
furane de la cyanhydrine du pipéronal. r) (dithienyl-5, 5 oxo-2 oxazolidinyl-3) carboxylate
de diméíhylaminoéthyle, dont le chlorhydrate
fond à 178 .
Le chlorure d'acide de départ, non isolé, a
été préparé à partir de la dithiÚnyl-5, 5 oxazolidi
none-2 fondant à 146 , elle-même préparée par
cyclisation au phosgène en présence de potasse,
du dithi?nyl-1, 1 amino-2 éthanol-1. s) (d, l) (oxo-2 phényl-5 cyclohexyl-5 oxazolidinyl
3) carboxylate de dimÚthylaminoÚthyle, dont le
chlorhydrate fond à 225 .
Le chlorure d'acide de départ, non isolé, a
été préparé à partir de la phÚnyl-5 cyclohexyl-5
oxazolidinone-2 fondant à 190 , elle-même pré
parée par dégradation de Curtius de la cyclohexyl
3 phÚnyl-3 hydroxy-3 propionyl hydrazine fondant
vers 92 .
Exemple 2
(d, l) cis (oxo-2 méthyl-4 phényl-5 oxazolidinyl-3)
carboxylate de morpholino-2'éthyle.
Une solution de 12 g de chlorure de l'acide (d, l), cis (oxo-2 méthyl-4 phÚnyl-5 oxazolidinyl-3) carboxylique dans 250 cm3 de benzène anhydre est ajoutée, goutte à goutte, à 10 , en 25 minutes, à une solution de 6, 05 g de morpholino éthanol et de 5, 05 g de triéthylamine dans 20 cm3 de benzène anhydre. Lorsque l'addition est terminée, le mélange est encore agité pendant 2 1/2 heures puis traité comme à l'exem- ple 1. On obtient finalement 10, 4 g de base fondant à 88 , dont le fumarate acide fond à 149 . Rendement : 60 /o.
Exemple 3
(d, 1) cis (oxo-9 méthyl-4 phenyl-5 oxaolidinyl-3)
Ri-dimethylaminoethyl carboxamide.
A une solution de 22, 5 g de chlorure de l'acide (d, l) cis (oxo-2 méthyl-4 ph?nyl-5 oxazolidinyl-3) carboxylique fondant à 115e, dans 250 cm3 de tétrahydrofurane anhydre, on ajoute goutte à goutte en 15 minutes, une solution de 16, 5 g de diméthylamino- éthylamine dans 50 cm3 de tétrahydrofurane. La température croît de 19 à 31¯ tandis qu'il se forme un précipité. Lorsque la coulée est terminée, on chauffe à 500 pendant 75 minutes, puis on évapore le solvant sous pression réduite. On dissout le résidu pateux dans 200 cm3 d'eau, alcalinise à pH 9 avec du carbonate de potassium et extrait plusieurs fois à l'Úther.
Les portions éthérées réunies sont lavées plusieurs fois à l'eau et séchées sur carbonate de potassium sec. Après évaporation du solvant sous pression réduite, on obtient 25 g de base huileuse brute.
Par addition d'éther chlorhydrique à une solution de la base dans l'isopropanol, on obtient finalement 25, 7 g de chlorhydrate fondant à 197 . Rendement 83, 6 /o.
Les produits obtenus par ce procédé décrit sont des produits intéressants pour l'industrie chimique : ils possèdent, en effet, des propriétés fongicides et constituent des intermédiaires de synthèse.
Ces produits sont, d'autre part, intéressants du point de vue de l'industrie pharmaceutique, car ils présentent des propriétés analgésiques, antipyrétiques et anti-inflammatoires, et certains d'entre-eux étant, également, des anticonvulsivants et des antitussifs.

It is understood that if two radicals R1 and R3, located on the adjacent carbons 4 and 5, are different and R2 and R4 identical, or if the 4 radicals R1, R2,
R3, R4 are all different, the derivatives obtained exist in two cis and trans forms and are, as such, part of the invention, as well as their diastereoisomers.
It is advantageous to operate in a solvent which may be an aromatic carbide such as benzene, or an aliphatic or heterocyclic ether (tetrahydrofuran) in the presence of an excess of the compound Y-AZ or of a tertiary base (triethylamine or pyridine) acting as a hydracid acceptor.
It is particularly advantageous to add, to a solution of the compound Y-AZ, chosen from the preferred solvent, containing or not one of the tertiary bases mentioned above, the acid chloride dissolved in the same solvent. The reaction will be carried out at a low temperature, between room temperature and a temperature of the order of 700 C.
The new derivatives of oxazolidinone-2, which are strong bases, can be purified by physical methods, such as crystallization, chromatography, or chemical methods, such as formation of salts with inorganic or organic acids, crystallization of these in an appropriate solvent and decomposition of the latter in an alkaline medium. In these operations, the nature of the anion is immaterial, insofar as it results in a well-defined and easily crystallizable salt.
These bases will, in general, be preserved in the form of addition salts.
As addition salts with mineral acids, there may be mentioned, in addition, the hydrochloride, hydrobromide, sulfate, phosphate, methane-sulfonate and, with organic acids, acetate, propionate, maleate, fumarate, succinate, benzoate, tartrate , malate, oxalate.
The following examples illustrate the invention. The indicated melting points were determined in block
Kofler. The yields are given relative to the starting acid chloride.
Example 1
(d, l) -cis (2-oxo-4-methyl-5-oxazolidinyl-3) -carboxylate of 2-dimethylamino-ethyl.
A solution of 4.5 g of (d, l), cis (2-oxo-4-methyl-5-oxazolidinyl-3) carboxylic acid chloride, melting at 115 in 90 cm-3 of anhydrous benzene, is added dropwise. dropwise over 25 minutes and cooling to 10, to a solution of 3.35 g of dimethylaminoethanol in 10 cm3 of anhydrous benzene. When the addition is complete, the mixture is left under stirring for 2 1/2 hours at room temperature. At the end of this time, the treatment is carried out with water, decanted and the benzene layer extracted several times with 7% hydrochloric acid. The acidic portions are combined; it is washed several times with ether and then basified to pH 9 with potassium carbonate and extracted several times with Úther.
The combined ethereal extracts are washed several times with water and dried over potassium carbonate. After evaporation under reduced pressure at low temperature, 5.5 g of base, melting at 540, are finally obtained. Yield: 100%.
By adding a solution of maleic acid to the solution of the previous base in ethanol, the acid maleate is obtained, melting at 138. The starting acid chloride was prepared by the action of a toluene solution of phosgene on a suspension of (d, l), cis-methyl-4-phnyl-5-oxazolidinone-2 melting at 146, in the presence of pyridine, in the benzene.

the acid fumarate melts at 170, from di methylamino ethanol and chloride of (d, 1) (2-oxo-4-diphÚnyl-5, 5-oxazolidinyl3) carboxylic acid (oily) prepared as follows: to a solution of 8, 2 g of methyl-4-diphÚnyl-5, 5 oxazolidinone-2, melting at 2320, in 200 cm3 of anhydrous tetrahydrofuran, is added at once 1.87 g of a suspension of HNa at 50 ouzo in the mineral oil. Then heated for 1 hour at reflux.
The solution obtained is cooled to 200 and poured over 15 min into 80 g of a 20% toluene solution of phosgene, at + 10. A precipitate of sodium chloride forms: it is stirred for one hour at the boil, cooled, the salt filtered and Úvaporated under reduced pressure at low temperature.
The viscous residue obtained is triturated in ether; the insoluble material consisting of unreacted starting material is filtered off and the filtrate is evaporated off under reduced pressure at low temperature. The oily residue obtained is used as it is for the esterification reaction.
The starting oxazolidinone was prepared by Curtius degradation from (3-diphenyl-3-hydroxy-2-methyl-2-propionyl) hydrazine, melting at 2050, itself obtained by reaction of hydrazine hydrate with 3, 3-diphÚnyl-3-hydroxy-2-methyl propionate in ethanol under reflux.
Dimethyl amino Úthyl (2-oxo-phenyl-S-oxazolidinyl) carboxylate, acid fumarate. To a solution of 12.7 g of dimethylaminoethanol in 50 cm3 of dry benzene, 13.9 g of (d, l) (2-oxo-phenyl-5-oxazolidinyl-3) carboxylic acid chloride is added over 30 minutes. dissolved in 200 cm3 of dry benzene. The internal temperature increases from 20 to 30, while an abundant precipitate is formed. The mixture is heated for 1 hour at 50O, then cooled, and treated with 2 × 100 cm3 of water.
The benzene solution is then extracted several times with 10% hydrochloric acid. The acid solutions are combined and basified with potassium carbonate; it is extracted several times with ether and the ethereal solutions are dried over potassium carbonate. After evaporation of the ether under reduced pressure, 15 g of oily base are obtained, the acid fumarate of which melts at 137.
The starting acid chloride, not isolated, was prepared by the action of an excess of phosgene on a toluene suspension of the sodium salt of 5-phenyloxazolidinone-2 melting at 890.
Dimethylamino-ethyl (2-oxo-diphÚnyl-5, 5-oxazolidinyl-3) carboxylate, mp 117. The corresponding hydrochloride melts at 210.
(2-oxo-diphenyl-S, S-oxazolidinyl-3) (methyl-4 'pipÚrazinyl-1') Úthyl carboxylate, melting point 200-202O. Bis methane sulfonate melts at 238¯ (dec.).
(d, l) (2-oxo-5-phÚnyl-5-oxazolidinyl-3) dimethylaminoethyl carboxylate, the corresponding acid fumarate of which melts at 1600. k) (d, l) cis (2-oxo-4-methyl-cyclohexyl-5 oxazolidi
dimÚthylaminoÚthyl-3) nylcarboxylate, of which
the hydrochloride melts at 165.
The starting acid chloride, not isolated, has
been prepared from (d, l) cis (methyl-4 cy
clohexyl-5) oxazolidinone-2 melting at 123o, it
even prepared by cyclization using
phosgene, in the presence of (d, l) cis potash
1-cyclohexyl 2-amino propanol-1, including di
hydrochloride melts at 217.

The starting acid chloride, not isolated, has
was prepared from (methylene-dioxy-3 ', 4',
phÚnyl) -5 oxazolidinone-2 melting at 128, it
even prepared by cyclization using the
phosgene, in the presence of potash, (methylene
dioxy-3 ', 4' phenyl) -1 amino-2 Úthanol-1 whose
hydrochloride melts at 196o, itself prepared by
reduction by means of AlkH4 in tetrahydro-
piperonal cyanohydrin furan. r) (dithienyl-5, 5 oxo-2 oxazolidinyl-3) carboxylate
of diméíhylaminoethyl, including hydrochloride
melts at 178.
The starting acid chloride, not isolated, has
been prepared from dithiÚnyl-5, 5 oxazolidi
none-2 melting at 146, itself prepared by
cyclization with phosgene in the presence of potash,
1,1-dithienyl-2-aminoethanol-1. s) (d, l) (2-oxo-5-phenyl-5-cyclohexyl-oxazolidinyl
3) dimÚthylaminoÚthyl carboxylate, of which the
hydrochloride melts at 225.
The starting acid chloride, not isolated, has
been prepared from 5-phÚnyl-5-cyclohexyl
oxazolidinone-2 melting at 190, itself pre
trimmed by Curtius degradation of cyclohexyl
3-phenyl-3-hydroxy-3-propionyl hydrazine fondant
around 92.
Example 2
(d, l) cis (2-oxo-4-methyl-phenyl-5-oxazolidinyl-3)
morpholino-2'ethyl carboxylate.
A solution of 12 g of (d, l), cis (2-oxo-4-methyl-5-oxazolidinyl-3) carboxylic acid chloride in 250 cm3 of anhydrous benzene is added, dropwise, at 10, in 25 minutes, to a solution of 6.05 g of morpholino ethanol and 5.05 g of triethylamine in 20 cm3 of anhydrous benzene. When the addition is complete, the mixture is further stirred for 2 1/2 hours and then treated as in Example 1. Finally, 10.4 g of base melting at 88 are obtained, the acid fumarate of which melts at 149. Yield: 60 / o.
Example 3
(d, 1) cis (9-oxo-4-methyl-5-phenyl-3-oxaolidinyl)
Ri-dimethylaminoethyl carboxamide.
To a solution of 22.5 g of (d, l) cis (2-oxo-4-methyl-5-oxazolidinyl-3) carboxylic acid chloride, melting at 115%, in 250 cm3 of anhydrous tetrahydrofuran, is added dropwise in 15 minutes, a solution of 16.5 g of dimethylamino-ethylamine in 50 cm3 of tetrahydrofuran. The temperature increases from 19 to 31¯ as a precipitate forms. When the casting is complete, the mixture is heated at 500 for 75 minutes, then the solvent is evaporated off under reduced pressure. The pasty residue is dissolved in 200 cm3 of water, basified to pH 9 with potassium carbonate and extracted several times with Úther.
The combined ethereal portions are washed several times with water and dried over dry potassium carbonate. After evaporation of the solvent under reduced pressure, 25 g of crude oily base are obtained.
By adding hydrochloric ether to a solution of the base in isopropanol, 25.7 g of hydrochloride are finally obtained, melting at 197. Yield 83.6 / o.
The products obtained by this described process are interesting products for the chemical industry: they have, in fact, fungicidal properties and constitute synthetic intermediates.
These products are, on the other hand, interesting from the point of view of the pharmaceutical industry, since they exhibit analgesic, antipyretic and anti-inflammatory properties, and some of them also being anticonvulsants and cough suppressants.
