Description
Verfahren zur Herstellung neuer Aminopyrazole
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von 2-R-3-Aminopyrazolen, worin R einen unsubstituierten oder substituierten Kohlenwasserstoffrest oder einen gesättigten oder ungesättigten heterocyclischen Rest darstellt, und die in 5-Stellung durch einen Pyridyl-(3)-rest substituiert sind und ihrer Salze.
Kohlenwasserstoffreste in 2-Stellung sind z. B. gesättigte oder ungesättigte aliphatische, alicyclische, ali- cyclisch-aliphatische, araliphatische oder aromatische Kohlenwasserstoffreste, wie niedere gerade oder verzweigte Alkyl- oder Alkenylreste, z. B. Methyl-, Äthyl-, Propyl-, Isopropyl-, gerade oder verzweigte, in beliebiger Stelle verbundene Butyl-, Pentyl, Hexyloder Heptylreste, Allyl- oder Methallylreste, Cycloalkyl- oder Cycloalkenylreste, wie Cyclopentyl-, Cyclohexyl-, Cycloheptyl-, Cyclopentenyl-, Cyclohexenylreste, Cycloalkyl- oder -alkenyl-alkylreste, wie Cyclopentyl- oder Cyclohexenyl-methyl-, -äthyl- oder -propyl- reste, Aralkyl-oder Aralkenyl- wie Phenylmethyl-, äthyl-, -vinyl- oder -propylreste,
oder Aryl-, insbesondere Phenylreste. Als heterocyclische oder heterocyc- lisch-aliphatische Reste kommen vor allem einkernige in Frage, wie Pyridyl- oder Piperidyl-, z. B. N-Alkylpiperidyl-(4)-reste.
Wenn nicht anders angegeben, bedeutet in der Be schreibung der Ausdruck nieder im Zusammenhang mit organischen Gruppen oder Verbindungen solcher Gruppen oder Verbindungen, welche höchstens 7 Kohlenstoffatome enthalten.
Als Substituenten der genannten aliphatischen Reste in 2-Stellung sind besonders freie oder substituierte Oxy-, Mercapto- oder Aminogruppen zu nennen, worin die Substituenten vorzüglich aliphatischer Natur sind, z. B. niedere Alkoxy-, Alkylmercapto- oder Mono- oder Di-alkyl- oder -cycloalkyl-aminogruppen, Alkylen-, Oxaalkylen-, Azaalkylen- oder Thia-alkylenaminogruppen, wie Methyl-, Äthyl-, Propyl-, Butyl-, Pentyl oder Hexyl-oxy-oder-mercaptogruppen, Methyl-, Dimethyl-, Äthyl-, Diäthyl-, Propyl-, Dipro pyl-, N-Methyl-N-propyl-, N-Methyl-N-cyclopentyl-, Butyl-, Di-butyl-amino-gruppen, Pyrrolidino-, Pipert- dino-, Morpholino oder Piperazinogruppen, z.
B. die Piperazino-, N-Methyl-pliperazino-, N-Oxyäthyl-pipera zinogruppe.
Weitere Substituenten, die für die aliphatischen Reste in Frage kommen, sind Halogenatome, wie Chlor oder Brom.
Alicyclische ; Reste in 2-Stellung können vor allem niedere Alkylreste tragen.
Aromatische oder heterocyclische Reste in 2-Stellung mögen besonders Halogenatome oder die oben gezeigten Alkyl- oder Alkoxygruppen aufweisen, während z. B. in den alicydisch-aliphatischen, araliphati schein oder heterocyclisch-aliphatischen Resten beide Teile wie angegeben substituiert sein können.
Die neuen Verbindungen können in den Kernen noch weiter substituiert sein. Hierbei ist z. B. Substitution in 4-Stellung zu erwähnen. Als Substituenten kommen dabei z. B. in Betracht: niedere Alkylreste, gegebenenfalls durch niedere Alkyl- oder Alkoxygrup- pen oder Halogenatome substituierte Phenylreste oder Halogenatome. Niedere Alkyl- oder Alkoxyreste sind insbesondere Methyl- Äthyl, Propyl, Isopropyl oder gerade oder verzweigte, in beliebiger Stelle verbundene Bntyl-, Pentyl, Hexyl- oder Heptylreste bzw. die entsprechenden Alkoxygruppen und Halogenatome vor allem Fluor, Chlor, Brom oder das Pseudohalogen Trifluormethyl.
Die neuen Verbindungen besitzen antiinflammatorische, antipyretische und antiallergische, die Verbindungen mit einem N-Alkyl-piperidyl-4-rest in 2-Stellung auch hypotensive Wirkung und können dementsprechend als Medikamente, z. B. bei der Behandlung von entzündlichen oder allergischen Prozessen bei Mensch und Tier Verwendung finden. Sie sind auch wertvolle Zwischenprodukte für die Herstellung weiterer, insbesondere als Pharmazeutika verwendbarer Vtrbindungen.
Besonders wertvoll sind Verbindungen der Formel

und Salze davon, worin R einen niederen Alkyl-, Oxyalkyl-, Aminoalkyl-, Mono- oder Di-niederalkylamino-alkyl-, Alkylen-imino-alkyl-, Azaalkyleniminoalkyl-, Oxaalkylenamino-alkyl-oder Cycloalkylrest, z. B. einen der oben erwähnten, oder einen gegebenenfalls durch niedere Alkyl- oder Alkoxygruppen oder Halogenatome substituierten Phenylrest oder einen Pyridylrest, z. B. den Pyridiyl-2-rest, oder einen N-A1kyl-piperidylrest, wie den N-Methyl-piperidyl-(4)-rest, und R' Wasserstoff oder einen niederen Alkylrest darstellen.
Hervorzuheben sind hierbei die Verbindungen der Formel

und ihre Salze, worin R einen niederen Alkylrest, wie Methyl, Äthyl, Propyl, Butyl oder Pentyl, vor allem aber einen verzweigten oder nicht in l-Stellung gebundenen Propyl-, Butyl-, Pentyl- oder Hexylrest, einen Cycloalkylrest, wie Cyclopentyl oder Cyclohexyl, einen Phenyl-, Halogenphenyl-, Niederalkoxyphenyl- oder Niederalkylphenylrest oder einen Pyridyl- wie den Pyridyl-(2)-rest oder einen N-Alkyl-piperidylrest, wie den N-Methyl-pip eridyl- (4)-rest bedeutet.
In erster Linie ist das 2-sek.-Butyl-3-amino-5-pyri dyl-(3)-plyrazol und seine Salze zu erwähnen.
Das erfindungsgemässe Verfahren zur Herstellung der Verbindungen ist dadurch gekennzeichnet, dass man entsprechende Pyridyl-(3)-ketone, die in a-Stellung zur Carbonylgruppe mindestens ein Wasserstoffatom und eine Nitrilgruppe oder eine reaktionsfähige Iminogruppen-haltig abgewandelte Carboxylgruppe, wie eine Iminoäther- oder Amidingruppe aufweisen, z. B. 3-(a-Cyan-alkanoyl)-pyridin, oder funktionelle Oxoderivate dieser Ketone, mit 2-R-Hydrazinen, worin R die oben gegebene Bedeutung hat, umsetzt. Die Carbonylgruppe kann also auch in reaktionsfähiger, abgewandelter Form, z. B. in der Enolat- oder Ketalform vorliegen.
Die Reaktion kann in an sich bekannter Weise, vorteilhaft in Anwesenheit von Verdünnungsmitteln und gegebenenfalls bei erhöhter Temperatur und/oder in Anwesenheit von Kondensationsmitteln, z. B. starken Säuren, wie Salzsäure, Arylsulfonsäuren und ähnlichen Säuren durchgeführt werden.
Erhaltene 4-unsubstituierte Verbindungen können leicht in 4-Stellung halogeniert werden, z. B. durch Behandlung mit chlorierenden oder bromierenden Mitteln, wie vor allem elementarem Chlor oder Brom, oder Chlor oder Brom abgebenden Verbindungen.
Ein Ausgangsstoff kann auch unter den Reaktionsbedingungen gebildet oder in Form eines Salzes verwendet werden.
Die genannten Reaktionen werden z. B. in üblicher Weise in An- oder Abwesenheit von Verdünnungs-, Kondensations- und/oder katalytischen Mitteln, bei erniedrigter, gewöhnlicher oder erhöhter Temperatur, gegebenenfalls im geschlossenen Gefäss durchgeführt.
Die aus Ausgangsstoffe verwendeten, gegebenenfalls monosubstituierten, Pyridoyl-(3)-acetonitrile, sowie die entsprechenden Iminoäther und Amidine und ihre Salze können z. B. erhalten werden, wenn man Pyridin-3-carbonsäuren, gegebenenfalls in Form ihrer reaktionsfähigen funktionellen Derivate, wie Ester, Anhydride oder Halogenide in an sich bekannter Weise in gegebenenfalls monosubstituierte Pyridoyl-(3)-aceto- nitrile überführt und diese gegebenenfalls in Imino äther oder Amidine überführt. So kann man einen Pyridyl-(3)-carbonsäureester, wie einen Alkyl-, z. B. den Äthylester mit einem gegebenenfalls monosubstituierten Acetonitril oder Malonsäurederivat, z. B. einem Malonsäureester oder -nitrilester oder -dinitril umsetzen und in an sich bekannter Weise aus dem Kondensationsprodukt die Pyridoyl-(3)-acetonitrile bilden.
Man kann aber auch aus einem Pyridyl-(3)-carbonsäurehalogenid über das Diazoketon und gegebenenfalls ein a-Halogenketon zu den Pyridoyl-(3)-acetonitrilen gelangen.
Die übrigen Ausgangsstoffe sind bekannt oder lassen sich ebenfalls nach bekannten Methoden gewinnen.
Die neuen Verbindungen werden je nach den Reaktionsbedingungen und Ausgangsstoffen in freier Form oder in Form ihrer Salze erhalten. Die Salze der neuen Verbindungen können in an sich bekannter Weise in die freien Verbindungen übergeführt werden, z. B. Säureadditionssalze durch Reaktion mit einem basischen Mittel. Anderseits können gegebenenfalls erhaltene freie Basen mit anorganischen oder organischen Säuren Salze bilden. Zur Herstellung von Säureadditionssalzen werden insbesondere therapeutisch verwendbare Säuren verwendet, z. B.
Halogenwasserstoffsäuren, beispielsweise Salzsäure oder Bromwasserstoffsäure, Perchlorsäure, Salpetersäure oder Thiocyansäure, Schwefel- oder Phosphorsäuren, oder organische Säuren, wie Ameisensäure, Essigsäure, Propionsäure, Glykolsäure, Milchsäure, Brenztraubensäure, Oxalsäure, Malonsäure, Bernsteinsäure, Maleinsäure, Fumarsäure, Äpfelsäure, Weinsäure, Zitronensäure, Ascorbinsäure, Hydroxymaleinsäure, Dihydroxymaleinsäure, Benzoesäure, Phenylessigsäure, 4-Amino-benzoesäure, 4-Hydroxy-benzoesäure, Anthranilsäure, Zimtsäure, Mandelsäure, Salicylsäure, 4-Amino-salicylsäure, 2-Phenoxybenzoesäure, 2-Acetoxy-benzoesäure, Methansulfonsäure, Athansulfonsäure, Hydroxyäthansulfonsäure, Benzolsulfonsäure, p-Toluol-sulfonsäure, Naphtalinsulfonsäure oder Sulfanilsäure, oder Methionin, Tryptophan, Lysin oder Arginin. Dabei können Mono- oder Polysalze vorliegen.
Enthalten die neuen Verbindungen asymmetrische Kohlenstoffatome, so können sie in Form von Racematen oder Racematgemischen vorliegen, die sich in üblicher Weise trennen bzw. in die Antipoden zerlegen lassen.
Die neuen Verbindungen können als Heilmittel in Form von pharmazeutischen Präparaten verwendet werden, welche diese Verbindungen zusammen mit pharmazeutischen, organischen oder anorganischen, festen oder flüssigen Trägerstoffen, die für enterale, z. B. orale, oder parenterale Gabe geeignet sind, enthalten.
In den folgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1 14, 6 g Nicotinoyl-acetonitril werden mit 12, 2 g Benzylhydrazin in 150 cm3 abs. Alkohol während 10 Stunden am Rückfluss gekocht. Man dampft die alkoholische Lösung nach Reinigung mit Kohle bis zur beginnenden Kristallisation ein, gibt etwas Äther hinzu und nutscht die Kristalle ab. Man erhält so das 2-Benzyl-3amino-5-pyridyl-(3)-pyrazol der Formel

Das als Ausgangsprodukt verwendete Nicotinoylacetonitril lässt sich wie folgt herstellen:
Zu 46 g Natrium in 1,5 Liter siedendem Toluo] werden langsam 500 cm3 abs. Alkohol zugetropft.
Nach dem Lösen des Natriums wird abdestilliert bis der Siedepunkt auf 91" steigt. Man lässt dann die Reaktionslösung auf 90" abkühlen, gibt 200 g Nicotinsäure-äthylester und 120 g Acetonitril zu und kocht während 7 Stunden am Rückfluss. Nach dem Abkühlen wird mit Wasser versetzt und die Toluolschicht abgetrennt. Nach Zugabe von 252 cm3 7,04-n. Salzsäure zur wässrigen Phase fällt nach starker Kühlung das Nicotinoyl-acetonitril der Formel

7,3 g Nicotinoyl-acetonitril werden mit 6, 55 g ss-Diäthylamino-äthyl-hydrazin in 75 cm3 Äthanol während 10 Stunden am Rückfluss gekocht. Man dampft die alkoholische Reaktionslösung anschliessend zur Trockne ein, löst den Rückstand in 2-n. Salzsäure und extrahiert mit Äther. Die saure wässrige Lösung wird mit konzentrierter Natronlauge alkalisch gestellt und mit Chloroform extrahiert. Die Chloroformlösung wird getrocknet und eingedampft. Anschliessend dampft man die Reaktionslösung stark ein, und gibt etwas Äther zu, worauf sich das Monohydrochlorid von 2-(ss-Diäthylamino- äthyl)-3-amino-5-pyridyl-(3)-pyrazol vom F. 159-1610 ausscheidet.
Beispiel 6
Eine Lösung von 10,1 g N-Methyl-piperidyl-(4)-hydrazindihydrochlorid in 260 cm3 98 0/oigem Äthanol wird mit einer Lösung von 1,15 g Natrium in 20 cm3 abs. Athanol versetzt. Man filtriert das ausgeschiedene Kochsalz ab, gibt das Filtrat zu einer Lösung von 7,3 g Nicotinoyl-acetonitril in 50 cm3 abs. Alkohol und kocht 10 Stunden am Rückfluss. Anschliessend dampft man die alkoholische Reaktionslösung zur Trockne ein, löst den Rückstand in Wasser auf und schüttelt mit Seither aus. Die wässrige Lösung wird mit Natronlauge stark alkalisch gestellt und mit Chloroform extrahiert.
Man dampft die Chioroformlösung ein und kristallisiert den Rückstand aus Äthanol um. Man erhält so das 21N-Methyl-pipendyl-(4)1- 83 amino-5-pyridyl-(3 )-pyrazol der Formel

3,3 g der obigen Base gelöst in 20 cm3 Äthanol werden mit 32,1 cm3 0,8-n. alkoholischer Salzsäure versetzt, worauf sich das Dihydrochlorid von 2-[N-Methyl-piperidyl-(4)]-3-amino-5-pyridyl-(3)-pyrazol vom F. 271-273 ausscheidet.
Claims (1)
- PATENTANSPRUCH 1 Verfahren zur Herstellung von 2-R-3-Aminopyrazolen, wobei R einen unsubstituierten oder substituierten Kohlenwasserstoffrest oder einen gesättigten oder ungesättigten heterocyclischen Rest darstellt, die in 5-Stellung durch einen gegebenenfalls substituierten Pyridyl-(3)-rest substituiert sind, dadurch gekennzeichnet, dass man entsprechende Pyridyl-(3)-ketone, die in a-Stellung zur Carbonylgruppe mindestens 1 Wasserstoffatom und eine Nitrilgruppe oder eine reaktionsfähige, iminogruppen-haltig, abgewandelte Carboxylgruppe enthalten, oder funktionelle Oxoderivate dien Ketone mit 2-R-Hydrazinen umsetzt.UNTERANSPRÜCHE 1. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man Verbindungen der Formel EMI4.2 worin R einen Alkyl-, Oxyalkyl-, Aminoalkyl-, Monooder Di-alkyl-amino-alkyl-, Alkylenimino-alkyl-, Azaalkylenimino-alkyl-, Oxaalkylenamino-alkyl-oder Cycloalkylrest, mit jeweils 1-7 C-Atomen in den Alkylgruppen oder einen gegebenenfalls durch Alkyl- oder Alkoxygruppen mit 1-7 C-Atomen oder Halogenatome substituierten Phenylrest, einen Pyridylrest oder einen N-Alkyl-piperidylrest und R' Wasserstoff oder einen Alkylrest mit 1-7 C-Atomen darstellt, herstellt.2. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man Verbindungen der Formel EMI4.3 worin R einen Alkylrest mit 1-7 C-Atomen, einen Cycloalkylrest, einen Phenyl-, Halogenphenyl-, einen Alkoxyphenyl- oder Alkylphenyl-rest mit 1-7 C-Atomen, in der Alkoxy-, bzw. Alkylgruppe, einen Pyridylrest oder einen N-Alkyl-piperidylrest darstellt, herstellt.3. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man das 2-sek.-Butyl-3-amino-5-pyridyl-(3)-pyrazol der Formel herstellt. EMI4.44. Verfahren nach Patentanspruch I oder einem der Unteransprüche 1-3, dadurch gekennzeichnet, dass man erhaltene Salze in die freien Verbindungen umwandelt.5. Verfahren nach Patentanspruch I oder einem der Unteransprüche 1-3, dadurch gekennzeichnet, dass man erhaltene freie Verbindungen in ihre Salze umwandelt.PATENTANSPRUCH II Verwendung der nach dem Verfahren gemäss Patentanspruch I erhaltenen, in 4-Stellung des Pyrazolrings unsubstituierten Verbindungen zur Herstellung von Verbindungen, die in 4-Stellung des Pyrazolrings durch Halogenatome substituiert sind, dadurch gekennzeichnet, dass man erhaltene in 4-Stellung unsubstituierte Verbindungen halogeniert.
