Verwendung von Salicylsäure-o-hydroxyphenylamiden zur Bekämpfung von schädlichen Mikroorganismen ausserhalb der Textilindustrie
Gegenstand der Erfindung ist die Verwendung von Salicylsäure-o-hydroxyphenylamiden der Formel

worin X ein Wasserstoffatom oder ein Halogenatom und Y eine Trifluormethylgruppe, eine Alkylgruppe mit höchstens 12 Kohlenstoffatomen, einen Cycloalkylrest, einen Phenylrest oder einen Aralkylrest bedeuten, wobei eine Alkylgruppe Y mindestens 8 Kohlenstoffatome enthält, wenn X für Wasserstoff steht, zur Bekämpfung von schädlichen Mikroorganismen ausserhalb der Textilindustrie.
Diese Salicylsäure-o-hydroxyphenylamide der Formel (1) - (nachfolgend z. T. auch einfache Salicylamide genannt) - entsprechen somit einer der Formeln

Hier bedeuten X1 ein Halogenatom, vorzugsweise ein Chlor- oder Bromatom, Y1 eine Trifluormethylgruppe, eine Alkylgruppe mit höchstens 12 Kohlenstoffatomen, einen Cycloalkylrest, einen Phenylrest oder einen Aralkylrest und Y2 eine Trifluormethylgruppe, eine Alkylgruppe mit 8 bis 12 Kohlenstoffatomen, einen Cycloalkylrest, einen Phenylrest oder einen Aralkylrest. Ist Y eine Alkylgruppe, so kann diese unverzweigt oder verzweigt sein.
Als Beispiele seien n-Octyl-, n-Dodecylund 1,1,3,3-Tetramethylbutylgruppen, und im Falle, dass der Salicylsäurerest zwei Halogenatome enthält, Methyl-, iSXthyl-, n-Propyl-, Isopropyl- und Tertiärbutylgruppen genannt. Als Cycloalkylreste kommen in erster Linie Cyclohexylreste und als Aralkylreste z. B. Benzylreste oder insbesondere l,l-Dimethyl-1-phenylmethylreste (Cumylreste) in Betracht.

hergestellt werden, wobei X und Y die angegebene Bedeutung haben. Man arbeitet vorteilhaft in einem inerten organischen Lösungsmittel und unter Zusatz eines säurebildenden Mittels, z. B. von wässriger Natriumhydroxydlösung.
Mit den Salicylamiden der Formeln (1) bis (3) können Mittel zur Bekämpfung schädlicher Mikroorganismen in üblicher, an sich bekannter Weise hergestellt und angewendet werden. Besonders wertvoll an den neuen Mitteln ist das breite antibakterielle Wirkungsspektrum, das sich sowohl auf grampositive als auch gramnegative Bakterien erstreckt. Hierbei ist in anwendungstechnischer Hinsicht die Geruchlosigkeit und Farblosigkeit der Salicylamide von besonderem Wert. Die vorliegende Erfindung umfasst somit ebenfalls deren Einsatz in der Schädlingsbekämpfung ganz allgemein.
Die Verwendung ist auf sehr breiter Basis möglich, insbesondere zum Schutze von organischen Substraten ausserhalb der Textilindustrie gegen den Befall durch zerstörende und pathogene (auch phytopathogene) Mikroorganismen. Die neuen Salicylamide eignen sich demnach sowohl als Konservierungsmittel wie auch als Desinfektionsmittel für technische Produkte aller Art, im Pflanzenschutz, in der Landwirtschaft.
Unter den technischen Produkten, welche mit Hilfe der Salicylamide konserviert werden können, seien die folgenden als Beispiele herausgegriffen:
Leime, Bindemittel, Anstrichmittel, Farb- bzw.
Druckpasten und ähnliche Zubereitungen auf der Basis von organischen und anorganischen Farbstoffen bzw.
Pigmenten, auch solche, welche als Beimischungen Casein oder andere organische Verbindungen enthalten.
Auch Wand- und Deckenanstriche, z. B. solche, die ein eiweisshaltiges Farbbindemittel enthalten, werden durch einen Zusatz der neuen Verbindungen vor dem Befall mit Schädlingen geschützt. Die Verwendung zum Holzschutz ist gleichfalls möglich.
Auch in der Zellstoff- und Papierindustrie können die neuen Salicylamide als Konservierungsmittel eingesetzt werden, u. a. zur Verhütung der bekannten, durch Mikroorganismen hervorgerufenen Schleimbildung in den zur Papiergewinnung verwendeten Apparaturen.
Ferner gelangt man durch Kombination der Verbindungen der Formeln (1) bis (3) mit wasch- bzw. oberflächenaktiven Stoffen zu Wasch- und Reinigungsmitteln mit ausgezeichneter antibakterieller bzw. antimykotischer Wirkung. Die Salicylamide können z. B. in Seifen eingearbeitet, mit seifenfreien, wasch- bzw. oberflächenaktiven Stoffen oder mit Gemischen aus Seifen und seifenfreien waschaktiven Stoffen kombiniert werden, wobei in diesen Kombinationen ihre antimikrobielle Wirksamkeit in vollem Umfang erhalten bleibt.
Reinigungsmittel, welche eine Verbindung der Formel (1) enthalten, können in Industrie und Haushalt eingesetzt werden, ebenso im Lebensmittelgewerbe, z. B.
Molkereien, Brauereien, Schlachthöfen. Auch als Bestandteil von Zubereitungen, welche dem Zwecke der Reinigung bzw. Desinfektion in Spitälern und in der medizinischen Praxis dienen, können die Salicylamide verwendet werden.
Die Wirkung kann auch in konservierenden und desinfizierenden Ausrüstungen von Kunststoffen wie z. B. Polyvinylchlorid ausgenützt werden. Bei Verwendung von Weichmachern ist es vorteilhaft, die Salicylamide dem Kunststoff im Weichmacher gelöst bzw.
dispergiert zuzusetzen. Zweckmässig ist für eine möglichst gleichmässige Verteilung im Kunststoff Sorge zu tragen. Die Kunststoffe mit antimikrobiellen Eigenschaften können für Gebrauchsgegenstände aller Art, bei denen eine Wirksamkeit gegen verschiedenste Keime, wie z. B. Bakterien und Pilze, erwünscht ist, Verwendung finden, so z. B. in Fussmatten, Badezimmervorhängen, Sitzgelegenheiten, Trittrosten in Schwimmbädern, Wandbespannungen. Durch Einverleibung in Wachs- und Bohnermassen erhält man Fussboden- und Möbelpflegemittel mit desinfizierender Wirkung.
Die Anwendungsformen können den üblichen Formulierungen von Schädlingsbekämpfungsmitteln entsprechen, beispielsweise können Mittel, die eine Verbindung der Formel (1) enthalten, gegebenenfalls auch noch Zusätze wie Trägerstoffe, Lösungsmittel, Verdünnungsmittel, Dispergier-, Netz- oder Haftmittel usw.
sowie andere Schädlingsbekämpfungsmittel enthalten.
Schliesslich können in solchen Mitteln zur Bekämpfung schädlicher Mikroorganismen auch mehrere Verbindungen der Formeln (1) bis (3) gleichzeitig vorhanden sein.
Die in den nachfolgenden Beispielen angegebenen Teile sind Gewichtsteile und die Prozente Gewichtsprozente, sofern nichts anderes angegeben ist.
Herstellungsvorschriften
22,6 Teile 3,5-Dichlorsalicylsäurechlorid werden in 100 Volumteilen Aceton gelöst. Die Lösung wird auf 50 C abgekühlt und bei dieser Temperatur eine Lösung von 19,1 Teilen 2-Amino-4-cyclohexyl-phenol in einem Gemisch von 100 Volumteilen Aceton und 100 Volumteilen Dioxan zugegeben. Bei 5 bis 10;OC wird das Reaktionsgemisch innerhalb von 10 Minuten tropfenweise mit 50 Volumteilen 2n-Natronlauge versetzt und hierauf das Ganze auf Eiswasser gegossen. Die Verbindung der Formel

scheidet sich zuerst als viskoses Ö1 ab, erstarrt jedoch nach einigem Nachrühren zu einer kristallinen Masse und wird dann abfiltriert, mit Wasser gewaschen und getrocknet. Die Ausbeute beträgt ungefähr 34 Teile; Schmelzpunkt 187-1910 C.
Nach dem Umkristallisieren aus Chlorbenzol fallen ungefähr 23 Teile der reinen Verbindung an, die bei 198,5-199,50 C schmilzt.
In ähnlicher Weise lassen sich die in der nachstehenden Tabelle aufgeführten Verbindungen der Formel (1) herstellen, wobei X und Y die angegebene Bedeutung haben.

Die Bestimmung der MIC (minimal inhibitory concentration) erfolgt nach einer an Standard-Normen angelehnten Prüfung, die eine Annäherung an absolute minimale Hemmwerte eines Wirkstoffes erlaubt.
Eine lSige und eine 0,3 % ige Lösung der Wirkstoffe in Dimethylsulfoxyd wird in Röhrchen mit steriler Brain Heart Infusion Broth (Bakterien) bzw. Bierwürzelösung (Pilze) gegeben und mit den Lösungen Verdünnungsreihen mit fortlaufender Verdünnung auf das Zehnfache angesetzt. Durch Kombination der beiden Reihen erhält man folgende kontinuierliche Verdünnungsreihe:
1000, 300, 100, 30, 10, 3 ppm usw.
Die Lösungen werden mit dem Bakterium Staphylococcus aureus bzw. den Pilzen Aspergillus niger und Rhizopus nigricans beimpft. Anschliessend wird bei Staphylococcus aureus während 48 Stunden bei 370 C (Bakteriostase) und bei den Pilzen während 72 Stunden bei 30 C bebrütet (Fungistase).
Nach den genannten Zeiten ergeben sich die minimalen Hemmwerte (ppm) der nachstehenden Tabelle.

Die Verbindungen der Formel (1) werden in geeigneten Formulierungen (z. B. als Lösungen in Dimethylsulfoxyd) bestimmter Konzentration mit warmem Brain Heart Infusion-Agar (Bakterien) resp. Mycophil-Agar (Pilze) vermischt.
Die flüssigen Mischungen werden auf eine erstarrte, keilförmige Grundagarschicht gegossen und ebenfalls erstarren gelassen.
Mit einer Pasteurpipette trägt man nun die Testorganismen senkrecht zum Gradienten linienförmig auf.
Nach einer Bebrütung von 24 Stunden bei 370 C (Bakterien) resp. 72 Stunden bei 300 C (Pilze) wird die Länge der auf dem Impfstrich gewachsenen Keime gemessen und in ppm Wirkstoff ausgedrückt.

Beispiel 1
Zur Herstellung einer antimikrobiellen Stückseife werden 1,2 g der Verbindung der Formel (28) folgen der Mischung zugesetzt:
120 g Grundseife in Schuppenform
0,12 g Dinatriumsalz der Sithylendiamin- tetraessigsäure (Dihydrat)
0,24 g Titandioxyd.
Die durch Walzen erhaltenen Seifenspäne werden mit einem Schnellrührer pulverisiert und anschliessend zu Seifenstücken gepresst.
Mit der antimikrobiellen Seife stellt man eine 5 % ige und eine 1,5 % ige Lösung in sterilem Leitungswasser her. Je 1 ml dieser Lösungen wird zu 4 ml steriler Brain Heart Infusion Broth gegeben. Durch fortlaufendes Verdünnen auf das jeweils Zehnfache werden zwei Reihen erhalten, die durch Kombination folgende kontinuierliche Verdünnungsreihen ergeben:
100, 30, 10, 3, 1 ... ppm Wirksubstanz.
Die Lösungen werden mit den Bakterien Staphylo coccus aureus bzw. Escherichia coli beimpft und
24 Stunden bei 37 C bebrütet. Nach dieser Zeit wird mit der Pipette den Lösungen 0,05 ml entnommen und über Brain Heart Infusion Schräg-Agar laufen gelassen.
Lösungen (Bakteriostase) sowie Agar-Röhrchen (Bakterizidie) werden dann weitere 24 Stunden bei 370C bebrütet.
Bei den Lösungen und den Schrägagarröhrchen wird nun die minimale Hemm- bzw. Abtötungskonzentration (ppm) bestimmt:

Beispiel 2
Auf einem Zweiwalzenstuhl wird folgende Mischung bei 1500 C während 20 Minuten ausgewalzt:
100,00 Teile Polyvinylchlorid
45,25 Teile Di-2-äthylhexylphthalat
1,5 Teile Barium-/Cadmium-laurat
0,25 Teile Stearinsäure
7,75 Teile einer Lösung von 1,55 Teilen der
Verbindung der Formel (37) in
6,25 Teilen Di-2-äthylhexylphthalat.
Der Walzenabstand wird so eingestellt, dass 1 mm dicke Felle entstehen, die anschliessend während 80 Minuten bei 165 bis 1700 C mit 14,00 kg/cm2 verpresst werden.
Zur Prüfung der Wirkung gegen Bakterien werden Rondellen zu 10 mm Durchmesser aus dem Weich Polyvinylchlorid ausgestanzt und auf Brain Heart Infusion-Agar-Platten gelegt, die mit Staphylococcus aureus vorbeimpft sind. Die Platten werden hierauf 24 Stunden bei 370 C bebrütet.
Beurteilt wird einerseits die um die Rondellen auftretende Hemmzone (HZ in mm) und anderseits das mikroskopisch feststellbare Wachstum (W in So) unter bzw. auf dem Weich-Polyvinylchlorid:

Claims (1)
- PATENTANSPRUCHVerwendung von Salicylsäure - o - hydroxyphenyl- amiden der Formel EMI5.1 worin X ein Wasserstoffatom oder ein Halogenatom und Y eine Trifluormethylgruppe, eine Alkylgruppe mit höchstens 12 Kohlenstoffatomen, einen Cycloalkylrest, einen Phenylrest oder einen Aralkylrest bedeuten, wobei eine Alkylgruppe Y mindestens 8 Kohlenstoffatome enthält, wenn X für Wasserstoff steht, zur Bekämpfung schädlicher Mikroorganismen ausserhalb der Textilindustrie.UNTERANSPRÜCHE 1. Verwendung von Salicylsäure-o-hydroxyphenylamiden der Formel EMI6.1 worin X1 ein Halogenatom und Y1 eine Trifluormethylgruppe, eine Alkylgruppe mit höchstens 12 Kohlenstoffatomen, einen Cycloalkylrest, einen Phenylrest oder einen Aralkylrest bedeuten, gemäss Patentanspruch.2. Verwendung von Salicylsäure-o-hydroxyphenylamiden der Formel EMI6.2 worin Y2 eine Trifluormethylgruppe, eine Alkylgruppe mit 8 bis 12 Kohlenstoffatomen, einen Cycloalkylrest, einen Phenylrest oder einen Aralkylrest bedeutet, gemäss Patentanspruch.3. Verwendung von Salicylsäure-o-hydroxyphenylamiden der Formel EMI6.3 worin X und Y die im Patentanspruch angegebene Bedeutung haben, gemäss Patentanspruch.4. Verwendung von Salicylsäure-o-hydroxyphenylamiden gemäss Patentanspruch, Unteranspruch 1 oder 3, die im Salicylsäurerest zwei Chlor- oder Bromatome enthalten, gemäss Patentanspruch.
