Verfahren zur Herstellung von neuen Derivaten des p-Aminoalkylbenzolsulfonamids
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Derivate des p-Aminoallcyl-benzolsul- fonamids.

Als reaktionsfähige Cyansäurederivate eignen sich Ha logencyane, wie Chlorcyan oder Bromcyan, oder Cyansäureester, insbesondere Cyansäurephenylester. Die Umsetzung erfolgt vorzugsweise in Gegenwart eines mit Wasser mischbaren oder nichtmischbaren inerten organischen Lösungsmittels in An- oder Abwesenheit von Wasser.
Geeignete inerte organische Lösungsmittel sind beispielsweise Kohlenwasserstoffe, wie Benzol, Toluol oder Xylol, niedere Alkanole, wie Methanol oder Äthanol, ätherartige Flüssigkeiten, wie Äther, Dioxan oder Tetrahydrofuran, chlorierte Kohlenwasserstoffe, wie Methylenchlorid, niedere Ketone, wie Aceton oder Methyläthylketon, Carbonsäureester, wie Essigsäureäthylester, Carbonsäurenitrile, wie Acetonitril, oder Sulfone, wie Tetrahydro-thiophen -l ,l-dioxid. Alkalimetallhydroxide, Alkalimetallhydrogencarbonate, Alkalimetallcarbonate oder Alkalimetallphosphate, wie die entsprechenden Natriumoder Kaliumverbindungen.
Ferner können auch Calciumcarbonate sowie Calciumphosphate und Magnesiumcarbonat eingesetzt werden.
Als Ausgangsstoffe der allgemeinen Formel II eignen sich solche Verbindungen, deren Symbole m, Rl, R2, R oder R und Rr mit den Symbolen übereinstimmen, die anschliessend an Formeln I u. Die nach dem erfindungsgemässen Verfahren erhaltenen Verbindungen der allgemeinen Formel I werden anschliessend gewünschtenfalls in ihre Salze mit anorganischen sowie organischen Säuren übergeführt. Die Herstellung dieser Salze erfolgt z.B. durch Umsetzung der Verbindungen der allgemeinen Formel I mit der äquivalenten Menge einer Säure in einem geeigneten wässrigorganischen oder organischen Lösungsmittel, wie z.B. Methanol, Äthanol, Diäthyläther, Chloroform oder Methylenchlorid.
Zur Verwendung als Arzneistoffe können anstelle der freien Verbindungen der allgemeinen Formel I deren pharmazeutisch annehmbare Salze mit Säuren eingesetzt werden. Geeignete Additionssalze sind z.B. Salze mit Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Äthansulfonsäure, -Hydroxyäthansulfonsäure, Essigsäure, Milchsäure, Oxalsäure, Bernsteinsäure, Fumarsäure, Maleinsäure, Apfelsäure, Weinsäure, Citronensäure, Salicylsäure, Phenylessigsäure, Mandelsäure und Embonsäure, sowie Salze mit blutzuckersenkenden Sulfonylharnstoffen, wie z.B. p-Toluolsulfonyl-butyl-harnstoff, p-Chlorbenzolsulfonyl-propylharnstoff, p-[2-(2-Methoxy-5-chlorbenzamido) -äthyl] -phenylsulfonyl-cyclohexylharnstoff .
Die neuen Wirkstoffe werden vorzugsweise peroral verabreicht. Die täglichen Dosen bewegen sich zwischen 30 und 300 mg für erwachsene Patienten mit normalem Gewicht. Geeignete Doseneinheitsformen, wie Dragees, Tabletten, enthalten vorzugsweise 30-300 mg eines erfindungsgemässen Wirkstoffes und zwar 20 bis 80% einer Verbindung der allgemeinen Formel I.
Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen der allgemeinen Formel I und von bisher nicht beschriebenen Zwischenprodukten näher, stellen jedoch keineswegs die einzigen Ausführungsformen derselben dar. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel I
28 ml 4 N Natronlauge u. 4,3 g Äthylenimin werden auf 100 gekühlt. Hierauf wird eine Suspension von 26,2 g p-(2-Acetamidoäthyl)-Dhenylsulfochlorid in 100 ml Aceton unter Rühren und Kühlen zugegeben, so dass die Temperatur nicht über 0 steigt. Nach beendetem Zutropfen wird 30 Minuten bei 00 weitergerührt. Dann wird das Kühl bad entfernt und die Lösung des erhaltenen 1 - p - [p - (2- Acetamido-äthyl)-phenylsulfonyl]-aziridin mit
100 ml n-Propylamin versetzt. Die Temperatur steigt auf 40-500. Das Reaktionsgemisch wird 1 Stunde weitergerührt u. anschliessend am Rotationsverdampfer das über schüssige Amin abdestilliert.
Der Kristallbrei, der neben Kochsalz das gewünschte N1-[p-(2-Acetamido-äthyl)-phe nyl-sulfonyl]-N,-n-propyl-äthylendiamin enthält, wird in
56 ml 2 N Natronlauge gelöst u. unter Rühren portionenweise mit 10,6 g Bromcyan versetzt, wobei man die Temperatur nicht über 400 steigen lässt. Nach einer Stunde wird die erhaltene Substanz mit Methylenchlorid extra hiert, mit 20 ml 2 N Natronlauge u. zweimal mit 100 ml Wasser gewaschen. Nach dem Trocknen und Eindampfen der Methylenchloridlösung wird der Rückstand aus Essigester umkristallisiert und ergibt das l-[p-(2-Acetamido- -äthyl)-phenylsulfonyl]-2-imino-3- n - propyl - imidazolidin vom Smp. 138-1400.
Beispiel 2
28 ml 4 N Natronlauge und 4,3 g Äthylenimin werden auf - 100 gekühlt. Hierauf wird eine Suspension von 27,6 g p-(2-Propionamidoäthyl) -phenylsulfochlorid in 100 ml Aceton unter Rühren und Kühlen zugegeben, so dass die Temperatur nicht über 0 steigt. Nach beendetem Zutropfen wird 30 Minuten bei 00 weitergerührt.
Dann wird das Kühlbad entfernt und die Lösung des erhaltenen 1 -[p-(2-Propionamido.äthyl)-phenylsulfonyl]-azi- ridin mit 100 ml n-Propyl-amin versetzt. Die Temperatur steigt auf 40-500. Das Reaktionsgemisch wird 1 Stunde weitergerührt und anschliessend wird am Rotationsverdampfer das überschüssige Amin abdestilliert. Der Kristallbrei, der neben Kochsalz das gewünschte (N1-[p-(2- -Propionamido-äthyl)-phenylsulfonyl] -N5 - n-propyl-äthy- lendiamin enthält, wird in 56 ml 2N Natronlauge gelöst und unter Rühren portionenweise mit 10,6 g Bromcyan versetzt, wobei man die Temperatur nicht über 400 steigen lässt. Nach einer Stunde wird die erhaltene Substanz mit Methylenchlorid extrahiert, mit 20 ml 2 N Natronlauge und zweimal mit 100 ml Wasser gewaschen.
Nach dem Trocknen und Eindampfen der Methylenchloridlösung wird der Rückstand aus Essigester umkristallisiert und ergibt das l-[p-(2-Propionamido-äthyl)-phenylsulfo- nyl]-2-imino-3-n-propyl-imidazolidin vom Smp. 127-1280.
In analoger Weise erhält man aus 100 ml Propylamin und 29 g p-(2-Butyramido.äthyl)-phenylsulfochlorid das 1 -[p-(2-Butyramido-äthyl)-phenylsulfonyl]-2-imino-3-pro- pyl-imidazolidin, Smp. 114-1160.
Beispiel 3
28 ml 4 N Natronlauge und 4,3 g Äthylenimin werden auf - 100 gekühlt. Hierauf wird eine Suspension von 35,5 g p- [2-(2-Methoxybenzamid o)äthyl].phenylsulfochlo.
rid in 100 ml Aceton unter Rühren und Kühlen zugegeben, so dass die Temperatur nicht über 0 steigt. Nach beendetem Zutropfen wird 30 Minuten bei 00 weitergerührt. Dann wird das Kühlbad entfernt und die Lösung des erhaltenen [2-(2.Methoxy.benzamido.äthyl).phenyl.
sulfonyl]-aziridin mit 100 ml n-Propylamin versetzt. Die Temperatur steigt auf 40-500. Das Reaktionsgemisch wird
1 Stunde weitergerührt und anschliessend wird am Rotationsverdampfer das überschüssige Amin abdestilliert.

10,6 g Bromcyan versetzt, wobei man die Temperatur nicht über 400 steigen lässt. Nach einer Stunde wird die erhaltene Substanz mit Methylenchlorid extrahiert, mit
20 ml 2 N Natronlauge und zweimal mit 100 ml Wasser gewaschen. Nach dem Trocknen und Eindampfen der Methylenchloridlösung wird der Rückstand aus Essigester umkristallisiert und ergibt das l-[p- < 2-(2-Methoxy- .benzamido).äthyl > .phenylsulfonyl].2.imino -3- n-propyl- -imidazolidin vom Smp. 144.1470.

100 ml Aceton unter Rühren und Kühlen zugegeben, so dass die Temperatur nicht über 0 steigt. Nach beendetem Zutropfen wird 30 Minuten bei 0 weitergerührt.
Dann wird das Kühlbad entfernt und die Lösung des erhaltenen l-[p-(2-Butyramido-äthyl)-phenylsulfonyl]-aziri- din mit 100 ml t-Butyl-amin versetzt. Die Temperatur steigt auf 40-500. Das Reaktionsgemisch wird 1 Stunde weitergerührt und anschliessend wird am Rotationsverdampfer das überschüssige Amin abdestilliert. Der Kristallbrei, der neben Kochsalz das gewünschte Nl-[p-(2- -Butyramido-äthyl)-phenylsulfonyl]-N2-t-butyl - äthylendiamin enthält, wird in 56 ml 2N Natronlauge gelöst und unter Rühren portionenweise mit 10,6 g Bromcyan versetzt, wobei man die Temperatur nicht über 400 steigen lässt. Nach einer Stunde wird die erhaltene Substanz mit Methylenchlorid extrahiert, mit 20 ml 2 N Natronlauge und zweimal mit 100 ml Wasser gewaschen.

-phenylsulfonyl] -2-imino-3 -cycloheptyl-imidazolidin, Smp. 166-1670; 100 ml Cycloheptylamin und 29,0 g p-(2-Butyramido -äthyl)-phenylsulfochlorid das l-[p-(2-Butyramido-äthyl)- -phenylsulfonyl] -2-imino-3 -cycloheptyl-imid azolidin, Smp. 144,5-1450.
Beispiel 5
28 ml 4 N Natronlauge und 5,7 g Propylenimin werden auf - 100 gekühlt. Hierauf wird eine Suspension von 29,0 g p-(2 - Butyramidoäthyl) - phenylsulfochlorid in ]00 ml Aceton unter Rühren und Kühlen zugegeben, so dass die Temperatur nicht über 0 steigt. Nach beendetem Zutropfen wird 30 Minuten bei 00 weitergerührt.
Dann wird das Kühlbad entfernt und die Lösung des erhaltenen l-[p-(2-Butyramido-äthyl)-phenylsulfonyl]-2-me- thyl-aziridin mit 100 ml Cyclohexylamin versetzt. Die Temperatur steigt auf 40-500. Das Reaktionsgemisch wird
1 Stunde weitergerührt und anschliessend wird am Rotationsverdampfer das überschüssige Amin abdestilliert.
Der Kristallbrei, der neben Kochsalz das gewünschte 2 -[p-(2-Butyramido - äthyl) - phenylsulfonyl] - 1 - cyclohexylamino-propan enthält, wird in 56 ml 2 N Natronlauge gelöst und unter Rühren portionenweise mit 10,6 g Bromcyan versetzt, wobei man die Temperatur nicht über 400 steigen lässt. Nach eine Stunde wird die erhaltene Substanz mit Methylenchlorid extrahiert, mit 20 ml 2 N Natronlauge und zweimal mit 100 mol Wasser gewaschen.
Nach dem Trocknen und Eindampfen der Methylenchloridlösung wird der Rückstand aus Essigester umkristallisiert und ergibt das 1 - [p.(2.Acetamidoäthyl).phenylsulfo.
nyl] - 2 - imino -3- cyclohexyl-5-methyl - imidazolidin vom Smp. 109-1110.
Beispiel 6
28 ml 4 N Natronlauge und 5,7 g Propylenimin werden auf - 100 gekühlt. Hierauf wird eine Suspension von 26,2 g p-(9-Acetamidoäthyl)-phenylsulfochlorid in 100 ml Aceton unter Rühren und Kühlen zugegeben, so dass die Temperatur nicht über 00 steigt. Nach beendetem Zutropfen wird 30 Minuten bei 00 weitergerührt. Dann wird das Kühlbad entfernt und die Lösung des erhaltenen 1 -[p.(2.Acetamido.äthyl).phenylsulfonyl] -2-methyl-aziridin mit 100 ml n-Butylamin versetzt. Die Temperatur steigt auf 40-500. Das Reaktionsgemisch wird 1 Stunde weitergerührt und anschliessend wird am Rotationsverdampfer das überschüssige Amin abdestilliert.
Der Kristallbrei, der neben Kochsalz das gewünschte 2-[p-(2-Acetamido-äthyl)- -phenylsulfonyl]-l-n-Butylamino-propan enthält, wird in 56 ml 2 N Natronlauge gelöst und unter Rühren portionenweise mit 10,6 g Bromcyan versetzt, wobei man die Temperatur nicht über 400 steigen lässt. Nach einer Stunde wird die erhaltene Substanz mit Methylenchlorid extrahiert, mit 20 ml 2 N Natronlauge und zweimal mit 100 ml Wasser gewaschen. Nach dem Trocknen und Eindampfen der Methyleuchloridlösung wird der Rückstand aus Essigester umkristallisiert und ergibt das 1-[p-(2 -Acetamido - äthyl)- phenylsulfonyl] -2-imino-3-n-butyl-5 -methyl-imidazolidin vom Smp. 98-99%.
Claims (1)
- PATENTANSPRUCHVerfahren zur Herstellung von neuen Derivaten des p-Aminoalkyl-benzolsulfonamids der allgemeinen Formel I, EMI5.1 in welcher m 2oder3, R1 eine Alkylgruppe mit höchstens 12 Kohlenstoffato men, eine Alkenylgruppe mit 3-5 Kohlenstoffatomen, eine Cycloalkyl- oder Cycloalkenylgruppe mit höch stens 7 Kohlenstoffatomen oder eine Phenylalkylgrup.pe mit höchstens 9 Kohlenstoffatomen, R2 Wasserstoff oder eine Alkylgruppe mit höchstens 2 Kohlenstoffatomen, R3 Wasserstoff, eine Alkylgruppe oder Chloralkylgruppe mit höchstens 7 Kohlenstoffatomen, eine Alkenyl gruppe mit höchstens 5 Kohlenstoffatomen, eine Cy.cloalkyl- oder Cycloalkenylgruppe mit höchstens 8 Kohlenstoffatomen, eine Phenylgruppe oder eine Phe nylalkyl- oder Phenylalkenylgruppe von höchstens 10 Kohlenstoffatomen, wobei die als R5 bzw. in R5 vor liegende Phenylgruppe durch Halogen bis Atomnum mer 35, Trifluormethylgruppen, Alkylgruppen mit höchstens 4 Kohlenstoffatomen, Hydroxylgruppen, Alkoxy- oder Alkylthiogruppen mit höchstens 2 Koh lenstoffatomen ein- bis dreifach substituiert sein kann.und R Wasserstoff oder die Methylgruppe bedeutet, und ihrer Additionssalze mit anorganischen oder organischen Säuren, dadurch gekennzeichnet, dass man eine Verbindung der allgemeinen Formel II, EMI5.2 in welcher m, R1, R5, R und R. die unter Formel I angegebene Bedeutung haben und R5 Wasserstoff, eine Arylmethyl-, Diarylmethyl- oder eine Triarylmethylgruppe, die Methyl- oder die Allylgruppe bedeutet, mit einem reaktionsfähigen Cyansäurederivat unter Ringschluss kondensiert und gegebenenfalls die erhaltenen Verbindungen mit einer anorganischen oder organischen Säure in ein Additionssalz überführt.
