Verfahren zur Herstellung von Alkanolaminderivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Alkanolaminderivaten, die ss-adrenergische Blockierungswirkung besitzen und daher für die Behandlung oder Prophylaxe von Herzkrankheiten, z. B. Angina pectoris und Herzarrhythmien, und für die Behandlung von erhöhtem Blutdruck und Phäochromocytom beim Menschen brauchbar sind.
Erfindungsgemäss werden neue Alkanolaminderivate der Formel:

worin R1 einen Alkylrest mit bis zu 12 Kohlenstoffatomen, der gegebenenfalls durch einen oder zwei der folgenden Substituenten: Hydroxylgruppen, Alkoxygruppen mit bis zu 5 Kohlenstoffatomen, Phenyl- und Phenoxygruppen substituiert ist, wobei die Phenyl- und Phenoxygruppen ihrerseits gegebenenfalls durch ein oder mehrere Chlor- oder Bromatome oder Methyl-, Athyl-, Methoxy- oder Athoxygruppen weiter substituiert sind, oder R1 einen Cycloalkylrest mit bis zu 8 Kohlenstoffatomen oder einen Alkenylrest mit 3 bis 6 Kohlenstoffatomen, R2 einen Alkanoyl-, Cycloalkancarbonyl-, Aroyl-, Aralkanoyl-, Aralkenoyl-, Aryloxyalkanoyl- oder Arylsulfonylrest, je mit bis zu 10 Kohlenstoffatomen, oder einen Halogenalkanoyl-, Alkenoyl-, Alkylsulfonyl- oder Alkoxycarbonylrest, je mit bis zu 6 Kohlenstoffatomen,
und R8 ein Halogenatom, den Cyanorest oder einen Alkylthio-, Cycloalkyl-, Alkanoyl- oder Alkoxycarbonylrest, je mit bis zu 6 Kohlenstoffatomen, oder den Phenyl- oder Phenoxyrest, welche gegebenenfalls durch ein oder mehrere Halogenatome oder Nitrogruppen oder Alkyl- oder Alkoxygruppen mit bis zu 4 Kohlenstoffatomen substituiert sind, oder einen Alkylrest mit bis zu 4 Kohlenstoffatomen, welcher durch die Hydroxyl- oder Phenylgruppe oder eine Alkoxygruppe mit bis zu 6 Kohlenstoffatomen oder ein oder mehrere Halogenatome substituiert ist, darstellen, sowie die Säureadditionssalze derselben hergestellt. Selbstverständlich soll die obige Definition von Alkanolaminderivaten alle möglichen Stereoisomeren und Gemische derselben umfassen.
Als geeignete Bedeutung von R1, wenn es einen gegebenenfalls substituierten Alkylrest darstellt, seien z. B. die folgenden Reste erwähnt: ein Alkylrest mit insbesondere 3, 4 oder 5 Kohlenstoffatomen, der vorzugsweise am a-Kohlenstoffatom verzweigt ist, z. B.
der Isopropyl-, sek.-Butyl- oder tert.-Butylrest, wobei dieser Alkylrest gegebenenfalls durch einen oder zwei Substituenten substituiert sein kann, die aus Hydroxylgruppen, Alkoxyresten mit bis zu 5 Kohlenstoffatomen, z. B. Methoxy- und Propoxyresten, und Phenyl- und Phenoxyresten, die ihrerseits gegebenenfalls durch ein oder mehrere Chlor- oder Bromatome oder einen oder mehreren Methyl-, Äthyl-, Methoxy- oder Äthoxyreste substituiert sein können, gewählt sind. Eine spezifische Bedeutung von R1, wenn es einen gegebenenfalls substituierten Alkylrest darstellt, ist daher der Isopropyl-, sek.-Butyl-, tert.-Butyl-, 2-Hydroxy- 1,1 -dimethyläthyl- oder 1,1-Dimethyl-2-phenyläthylrest.
Als geeignete Bedeutung von Rt, wenn es einen Cycloalkylrest darstellt, sei beispielsweise der Cyclopropyl-, Cyclobutyl- oder Cyclopentylrest erwähnt.
Als geeignete Bedeutung von R1, wenn es einen Alkenylrest darstellen, sei beispielsweise der Allylrest erwähnt.
Als geeignete Bedeutung von R2 sei z. B. der Formyl-, Acetyl-, Propionyl-, Butyryl-, Valeryl-, Pivaloyl-, Pentanoyl-, Hexanoyl-, Heptanoyl-, Chloracetyl-, Cyclopropancarbonyl-, Cyclohexancarbonyl-, Acryloyl-, Crotonoyl-,. B enzoyl-, p-Methylbenzoyl-, Chlorbenzoyl-, Phenylacetyl-, Cinnamoyl-, Phenoxyacetyl-, Methansulfonyl-, Sithansulfonyl-, Benzolsulfonyl- Toluol-p-sulfonyl- oder der Athoxycarbonylrest genannt.
Als geeignete Bedeutung von R3 sei z. B. das Fluor-, Chlor-, Brom- oder Jodatom, oder der Methylthio-, Athylthio-, Cyclohexyl-, Acetyl-, Propionyl-, Methoxycarbonyl- oder Äthoxycarbonylrest, oder der Phenyl oder Phenoxyrest, der p-Tolyloxyrest, der Hydroxymethyl-, Methoxymethyl-, n-Butoxymethyl-, Trifluormethyl-, Benzyl- oder a-Phenyläthylrest oder der Cyanorest genannt.
Geeignete Säureadditionssalze der erfindungsgemäss erhältlichen Alkanolaminderivate sind z. B. von anorganischen Säuren abgeleitete Salze, beispielsweise Hydrochloride, Hydrobromide, Phosphate oder Sulfate, oder von organischen Säuren abgeleitete Salze, beispielsweise Oxalate, Lactate, Tartrate, Acetate, Salicylate, Citrate, Benzoate, fi-Naphthoate, Adipate oder 1,1 Methylen-bis-(2-hydroxy-3-naphthoate), oder von sauren synthetischen Harzen, z. B. sulfonierten Polystyrolharzen, z. B. Zeo-Karb 225 (Markenprodukt), abgeleitete Harze.
Eine besonders bevorzugte Gruppe von erfindungsgemäss erhältlichen Alkanolaminderivaten sind die Verbindungen der Formel:

worin R3 obige Bedeutung besitzt und R6 einen Alkylrest mit bis zu 6 Kohlenstoffatomen bedeuten, sowie die Säureadditionssalze derselben.
Spezifische erfindungsgemäss erhältliche Alkanolaminderivate werden im folgenden in den Beispielen 1 und 2 beschrieben. Unter diesen Derivaten werden jene bevorzugt, welche die oben zuletzt angegebene Formel haben, wobei entweder Rss den Methylrest und R3 das Fluor-, Chlor- oder Bromatom oder den Methylthiooder den Phenylrest oder R6 den Athylrest und R3 das Fluor-, Chlor-, Brom- oder Jodatom oder den Phenyl-, p-Tolyloxy-, Methoxymethyl-, Benzyl-, a-Phenyläthyl-, Cyclohexyl- oder Propionylrest oder R6 den n-Propylrest und R3 den p-Tolyloxyrest oder R6 den n-Butyloder n-Hexylrest und R3 das Bromatom bedeuten, und ihre Säureadditionssalze.
Andere bevorzugte Verbindungen entsprechen der Formel I, in welcher R3 ein Bromatom und R2 den Propionylrest bedeuten und R' den sek.-Butyl- oder den Cyclopropylrest darstellt. Unter den bevorzugten Verbindungen dieser Gruppe sind auch die Säureadditionssalze zu verstehen.
Das Verfahren gemäss der Erfindung ist dadurch gekennzeichnet, dass man ein Aminoderivat der Formel:

oder ein Säureadditionssalz davon mit einer Verbindung der Formel R1Z, nämlich einem reaktionsfähigen Ester, der von einem Alkohol der Formel R'OH, in welcher R' obige Bedeutung besitzt, abgeleitet ist, umsetzt.
Als geeignete Bedeutung von Z sei z. B. das Chlor-, Brom- oder Jodatom, der p-Toluol-sulfonyloxyrest oder ein Rest der Formel -OSO ,ORt erwähnt. Eine besonders geeignete Verbindung der Formel R'Z ist Isopropylbromid.
Die Umsetzung kann zweckmässig in Gegenwart einer Base, z. B. einer anorganischen Base, beispielsweise Natrium- oder Kaliumcarbonat, in Gegenwart eines Katalysators, beispielsweise Kaliumjodid, und in Gegenwart eines Verdünnungsmittels oder Lösungsmittels, beispielsweise Athanol oder Isopropanol, ausgeführt werden. Die Umsetzung kann zweckmässig bei erhöhter Temperatur, z. B. bei einer Temperatur zwischen 50 und 2000 C, beispielsweise bei etwa 700 C, ausgeführt werden.
Das als Ausgangsprodukt verwendete Aminoderivat (II) kann durch Umsetzung des entsprechenden Epoxyds oder Halogenhydrins mit Ammoniak hergestellt werden.
Die erfindungsgemäss erhältlichen Alkanolaminderivate können durch Umsetzung mit einer Säure in bekannter Weise in die Säureadditionssalze übergeführt werden.
Wie oben angegeben, sind die erfindungsgemäss erhältlichen Alkanolaminderivate wertvoll für die Behandlung oder Prophylaxe von Herzkrankheiten. Ferner besitzen einige der erfindungsgemäss erhältlichen Alkanolaminderivate eine selektive ss-adrenergische Blockierungswirkung. Verbindungen, welche diese selektive Wirkung zeigen, besitzen einen grösseren Grad von spezifischer Wirkung bei der Blockierung der Herz-ss Rezeptoren als der A-Rezeptoren in den peripheren Blutgefässen und Bronchenmuskeln. Daher kann bei einer solchen Verbindung eine solche Dosis gewählt werden, dass die Verbindung zwar die chronotrope Herzwirkung eines Brenzkatechinamins [z.
B. von Isoprenalin, d. h. 1-(3,4-Dihydroxyphenyl)-2-isopropylaminoäthanol] blockiert, aber die durch Isoprenalin hervorgerufene Entspannung des glatten Luftröhrenmuskels oder die periphere gefässerweiternde Wirkung des Isoprenalins nicht blockiert. Wegen dieser selektiven Wirkung kann eine dieser Verbindungen vorteilhafterweise zusammen mit einem sympathikomimetischen Bronchodilatator, z. B. Isoprenalin, Orciprenalin, Adrenalin oder Ephedrin, zur Behandlung von Asthma und anderen die Luftwege schädigenden Krankheiten verwendet werden, da die selektiv wirksame Verbindung im wesentlichen die unerwünschte stimulierende Wirkung des Bronchodilatators auf das Herz hemmt, aber die erwünschte therapeutische Wirkung des Bronchodilatators nicht behindert.
Es wird erwartet, dass die bevorzugten Verbindungen Menschen bei einer oralen Dosis zwischen 20 mg und 600 mg täglich in Einzeldosen im Abstand von 6 bis 8 Stunden oder bei einer intravenösen Dosis zwischen 1 mg und 20 mg täglich verabreicht werden. Bevorzugte orale Dosisformen sind Tabletten oder Kapseln, die zwischen 10 und 100 mg, vorzugsweise 10 oder 40 mg, des Wirkstoffes enthalten. Bevorzugte intravenöse Dosierungsformen sind sterile wässrige Lösungen der Alkanolaminderivate oder der nicht toxischen Säureadditionssalze derselben, welche Lösungen zwischen 0,05 und 1% (Gewicht/Volumen) Wirkstoff, insbesondere 0,1 % (Gewicht/Volumen) Wirkstoff enthalten.
Die Erfindung wird erläutert durch die folgenden Beispiele, in welchen die Teile gewichtsmässig angegeben sind:
Beispiel I
Ein Gemisch von 0,5 Teilen 3-Amino-1-(2-brom4-propionylaminophenoxy)-2-propanol, 1,5 Teilen Iso propylbromid, 20 Teilen Isopropanol, 1 Teil Kaliumcarbonat und 0,1 Teil Kaliumjodid wird während 12 Stunden bei einer Temperatur von 700 C gerührt. Das Gemisch wird filtriert und das Filtrat unter vermindertem Druck zur Trockne eingedampft. Dem Rückstand werden 5 Teile Wasser zugegeben und das Gemisch mit 50 Teilen Essigester extrahiert. Der Essigesterextrakt wird bis auf 1/4 des Volumens eingedampft, darauf 1 Stunde stehengelassen und das Gemisch wird filtriert.
Das Filtrat wird zur Trockne eingedampft und der Rückstand mit 10 Teilen Äther geschüttelt. Das Gemisch wird filtriert und der feste Rückstand aus Essigester kristallisiert, wobei 1-(2-Brom-4-propionylaminophe- noxy)-3-isopropylamino-2-propanol vom Smp. 1470 C erhalten wird.
Das Ausgangsprodukt kann folgendermassen hergestellt werden:
3,0 Teile 1-(2-Brom-4-propionylaminophenoxy)2,3-epoxypropan werden einer Lösung von 12 Teilen Ammoniak und 0,05 Teilen Triäthylamin in 100 Teilen Methanol zugegeben und das Gemisch wird während 12 Stunden bei Raumtemperatur stehengelassen und darauf zur Trockne eingedampft. Der Rückstand wird mit 20 Teilen Äther geschüttelt, die Mischung wird filtriert und der feste Rückstand aus Äthanol kristallisiert. Das erhaltene 3-Amino-1-(2-brom-4-propionylaminophenoxy)-2-propanol schmilzt bei 1250 C.
Beispiel 2
Das in Beispiel 1 beschriebene Verfahren wird unter Verwendung anderer Ausgangsprodukte wiederholt, wobei die in der folgenden Tabelle angeführten Verbindungen erhalten werden.

Process for the preparation of alkanolamine derivatives
The present invention relates to a process for the preparation of new alkanolamine derivatives which have β-adrenergic blocking action and are therefore used for the treatment or prophylaxis of heart diseases, e.g. B. angina pectoris and cardiac arrhythmias, and for the treatment of hypertension and pheochromocytoma in humans are useful.
According to the invention, new alkanolamine derivatives of the formula:

wherein R1 is an alkyl radical with up to 12 carbon atoms, which is optionally substituted by one or two of the following substituents: hydroxyl groups, alkoxy groups with up to 5 carbon atoms, phenyl and phenoxy groups, the phenyl and phenoxy groups in turn optionally being substituted by one or more chlorine groups or bromine atoms or methyl, ethyl, methoxy or ethoxy groups are further substituted, or R1 is a cycloalkyl radical with up to 8 carbon atoms or an alkenyl radical with 3 to 6 carbon atoms, R2 is an alkanoyl, cycloalkanecarbonyl, aroyl, aralkanoyl, aralkenoyl -, aryloxyalkanoyl or arylsulfonyl radical, each with up to 10 carbon atoms, or a haloalkanoyl, alkenoyl, alkylsulfonyl or alkoxycarbonyl radical, each with up to 6 carbon atoms,

A suitable meaning of R1 when it represents an optionally substituted alkyl radical are, for. B. the following radicals mentioned: an alkyl radical with in particular 3, 4 or 5 carbon atoms, which is preferably branched on the a-carbon atom, z. B.
the isopropyl, sec-butyl or tert-butyl radical, it being possible for this alkyl radical to be optionally substituted by one or two substituents consisting of hydroxyl groups, alkoxy radicals with up to 5 carbon atoms, e.g. B. methoxy and propoxy radicals, and phenyl and phenoxy radicals, which in turn may optionally be substituted by one or more chlorine or bromine atoms or one or more methyl, ethyl, methoxy or ethoxy radicals, are selected. A specific meaning of R1 when it represents an optionally substituted alkyl radical is therefore isopropyl, sec-butyl, tert-butyl, 2-hydroxy-1,1-dimethylethyl or 1,1-dimethyl-2 -phenylethyl radical.
A suitable meaning of Rt when it represents a cycloalkyl radical is, for example, the cyclopropyl, cyclobutyl or cyclopentyl radical.
The allyl radical may be mentioned as a suitable meaning of R1 when it represents an alkenyl radical.
A suitable meaning of R2 is e.g. B. the formyl, acetyl, propionyl, butyryl, valeryl, pivaloyl, pentanoyl, hexanoyl, heptanoyl, chloroacetyl, cyclopropane carbonyl, cyclohexane carbonyl, acryloyl, crotonoyl ,. Benzoyl, p-methylbenzoyl, chlorobenzoyl, phenylacetyl, cinnamoyl, phenoxyacetyl, methanesulfonyl, sithanesulfonyl, benzenesulfonyl, toluene-p-sulfonyl or the ethoxycarbonyl radical.
A suitable meaning of R3 is e.g. B. the fluorine, chlorine, bromine or iodine atom, or the methylthio, ethylthio, cyclohexyl, acetyl, propionyl, methoxycarbonyl or ethoxycarbonyl radical, or the phenyl or phenoxy radical, the p-tolyloxy radical, the hydroxymethyl , Methoxymethyl, n-butoxymethyl, trifluoromethyl, benzyl or a-phenylethyl radical or the cyano radical.
Suitable acid addition salts of the alkanolamine derivatives obtainable according to the invention are, for. B. salts derived from inorganic acids, such as hydrochlorides, hydrobromides, phosphates or sulfates, or salts derived from organic acids, such as oxalates, lactates, tartrates, acetates, salicylates, citrates, benzoates, fi-naphthoates, adipates or 1,1 methylene bis- (2-hydroxy-3-naphthoate), or from acidic synthetic resins, e.g. B. sulfonated polystyrene resins, e.g. B. Zeo-Karb 225 (branded product), derived resins.
A particularly preferred group of alkanolamine derivatives obtainable according to the invention are the compounds of the formula:

where R3 has the above meaning and R6 is an alkyl radical having up to 6 carbon atoms, as well as the acid addition salts thereof.
Specific alkanolamine derivatives obtainable according to the invention are described in Examples 1 and 2 below. Among these derivatives, those are preferred which have the formula given last above, where either Rss is the methyl radical and R3 is the fluorine, chlorine or bromine atom or the methylthio or phenyl radical or R6 is the ethyl radical and R3 is the fluorine, chlorine, bromine or iodine atom or the phenyl, p-tolyloxy, methoxymethyl, benzyl, a-phenylethyl, cyclohexyl or propionyl radical or R6 the n-propyl radical and R3 the p-tolyloxy radical or R6 the n-butyl or n-hexyl radical and R3 mean the bromine atom, and their acid addition salts.
Other preferred compounds correspond to the formula I, in which R3 is a bromine atom and R2 is the propionyl radical and R 'is the sec-butyl or cyclopropyl radical. The preferred compounds of this group also include the acid addition salts.
The process according to the invention is characterized in that an amino derivative of the formula:

or an acid addition salt thereof with a compound of the formula R1Z, namely a reactive ester which is derived from an alcohol of the formula R'OH, in which R 'has the above meaning.
A suitable meaning of Z is e.g. B. the chlorine, bromine or iodine atom, the p-toluene-sulfonyloxy radical or a radical of the formula -OSO, ORt mentioned. A particularly suitable compound of the formula R'Z is isopropyl bromide.
The reaction can conveniently be carried out in the presence of a base, e.g. B. an inorganic base, for example sodium or potassium carbonate, in the presence of a catalyst, for example potassium iodide, and in the presence of a diluent or solvent, for example ethanol or isopropanol. The reaction can conveniently take place at elevated temperature, e.g. B. at a temperature between 50 and 2000 C, for example at about 700 C, performed.
The amino derivative (II) used as the starting material can be prepared by reacting the corresponding epoxide or halohydrin with ammonia.
The alkanolamine derivatives obtainable according to the invention can be converted into the acid addition salts in a known manner by reaction with an acid.
As indicated above, the alkanolamine derivatives obtainable according to the invention are valuable for the treatment or prophylaxis of heart diseases. Furthermore, some of the alkanolamine derivatives obtainable according to the invention have a selective β-adrenergic blocking effect. Compounds exhibiting this selective activity have a greater degree of specific activity in blocking cardiac receptors than the A receptors in peripheral blood vessels and bronchial muscles. Therefore, such a compound may be selected at a dose such that the compound exhibits the chronotropic cardiac action of a catecholamine [e.g.
B. of isoprenaline, d. H. 1- (3,4-Dihydroxyphenyl) -2-isopropylaminoethanol], but does not block the relaxation of the tracheal smooth muscle caused by isoprenaline or the peripheral vasodilating effect of isoprenaline. Because of this selective action, one of these compounds can advantageously be used in conjunction with a sympathomimetic bronchodilator, e.g. B. isoprenaline, orciprenaline, adrenaline or ephedrine, can be used for the treatment of asthma and other diseases damaging the airways, since the selectively active compound substantially inhibits the undesirable stimulatory effect of the bronchodilator on the heart, but does not hinder the desired therapeutic effect of the bronchodilator .
It is expected that the preferred compounds will be administered to humans at an oral dose between 20 mg and 600 mg daily in single doses spaced 6 to 8 hours apart or at an intravenous dose between 1 mg and 20 mg daily. Preferred oral dosage forms are tablets or capsules which contain between 10 and 100 mg, preferably 10 or 40 mg, of the active ingredient. Preferred intravenous dosage forms are sterile aqueous solutions of the alkanolamine derivatives or the non-toxic acid addition salts thereof, which solutions contain between 0.05 and 1% (weight / volume) active ingredient, in particular 0.1% (weight / volume) active ingredient.

A mixture of 0.5 part of 3-amino-1- (2-bromo4-propionylaminophenoxy) -2-propanol, 1.5 parts of isopropyl bromide, 20 parts of isopropanol, 1 part of potassium carbonate and 0.1 part of potassium iodide is added for 12 hours stirred at a temperature of 700 C. The mixture is filtered and the filtrate is evaporated to dryness under reduced pressure. 5 parts of water are added to the residue and the mixture is extracted with 50 parts of ethyl acetate. The ethyl acetate extract is evaporated to 1/4 of the volume, then left to stand for 1 hour and the mixture is filtered.
The filtrate is evaporated to dryness and the residue is shaken with 10 parts of ether. The mixture is filtered and the solid residue is crystallized from ethyl acetate, 1- (2-bromo-4-propionylaminophenoxy) -3-isopropylamino-2-propanol with a melting point of 1470 ° C. being obtained.
The starting product can be manufactured as follows:
3.0 parts of 1- (2-bromo-4-propionylaminophenoxy) 2,3-epoxypropane are added to a solution of 12 parts of ammonia and 0.05 part of triethylamine in 100 parts of methanol and the mixture is left to stand for 12 hours at room temperature and then added evaporated to dryness. The residue is shaken with 20 parts of ether, the mixture is filtered and the solid residue is crystallized from ethanol. The 3-amino-1- (2-bromo-4-propionylaminophenoxy) -2-propanol obtained melts at 1250 C.
