Verfahren zur Herstellung von 3-Alkylflavanonen

R2 Wasserstoff, Alkyl mit 1 - 6 C-Atomen oder Z¯(( H2)n¯s
Z Dialkylamino mit 2 - 6 C-Atomen, Pyrrolidino, Piperidino oder Morpholino und n2oder3 bedeuten sowie deren Ester, Säureadditions- und Estersalze und quartäre Ammoniumderivate wertvolle pharmakologische Eigenschaften besitzen. Sie können daher als Arzneimittel verwendet werden.
Insbesondere zeigen sie eine cholesterinspiegelsenkende Wirkung ohne jedoch wie bekannte cholesterinspiegelsenkende Mittel, z.B. 22,25-Diaza-cholesterin, Triparanol, Dehydroepiandrosteron-3-diäthylaminoäthyläther, eine unphysiologische Anreicherung von Desmosterin oder 7-Dehydrocholesterin in den Sterinen des Serums oder der Leber zu bewirken.
Ferner zeigen die neuen 3-Alkylflavanone östrogene und Antifertilitätswirkungen.
Im einzelnen führte beispielsweise eine orale Gabe von jeweils 50 mg/kg der angegebenen Verbindungen zu folgenden prozentualen Senkungen im Cholesterinspiegel von Ratten (Methodik vgl. Counsel et al., J. med. pharm.
Chem., 5, 720, 1224 (1962): 3 -Methyl-6-hydroxy-flavanon-6-sulfat, Na-Salz (A) 86% 3 -Methyl-6-hydroxy-flavanon 42% 3-n-Propyl-6-hydroxy-flavanon 37% 3-n-Propyl-6-hydroxy-flavanon-6-nicotinat cis-trans-Gemisch (B) 36%
Dabei sind die Verbindungen gut verträglich. Ihre Toxizitäten sind ausserordentlich niedrig. Beispielsweise liegt die DL55 der Verbindung A oberhalb 3,2 g/kg, diejenige der Verbindung B oberhalb 6,4 g/kg (oral an Ratten bestimmt).
Die Verbindungen der Formel I können ferner als Zwischenprodukte zur Herstellung weiterer Arzneimittel verwendet werden.
Gegenstand der Erfindung ist im Verfahren zur Herstellung von 3-Alkylflavanonen der Formel I, das darin besteht, dass man ein Keton der Formel II

und dass man gegebenenfalls in dem erhaltenen Produkt nach an sich bekannten Methoden eine funktionell abgewandelte Hydroxygruppe durch Behandeln mit hydrolysierenden oder hydrogenolysierenden Mitteln in Freiheit setzt und/oder eine freie Hydroxygruppe durch Behandeln mit veresternden oder alkylierenden Mitteln gegebenenfalls mehrstufig verestert oder alkyliert und/oder dass man gegebenen falls Verbindungen der Formel I durch Behandeln mit Säuren bzw. Alkylierungsmitteln in ihre physiologisch verträglichen Säureadditions- bzw. Estersalze bzw. quartären Ammoniumverbindungen überführt.
Als mögliche Alkylgruppen in den Resten R1 und R2 seien beispielsweise genannt: Methyl, Äthyl, Propyl, Isopropyl, n-Butyl, sek.-Butyl, Isobutyl, tert.-Butyl, n-Amyl, Isoamyl, n-Hexyl, Isohexyl.
Als Z-(CHt)11-Gruppen im Rest R2 sind vor allem zu erwähnen: 2-Dimethylaminoäthyl, 2-Diäthylamino äthyl, 3-Dimethylaminopropyl, 3-Diäthylaminopropyl, 2- Pyrrolidinoäthyl, 2-Piperidinoäthyl, 2-Morpholinoäthyl, 3-Pyrrolidinopropyl, 3 -Piperidinopropyl, 3-Morpholinopropyl.
Als Ester solcher Verbindungen der Formel I, in denen R = H ist, kommen vor allem die niederen Acylate in Frage, in denen die Acylgruppe 1 - 6 C-Atome enthält.
Im einzelnen sind typische Ester die Formiate, Acetate, Propionate, Butyrate, Isobutyrate, Valerianate, Isovalerianate, Trimethylacetate, Capronate, Isocapronate, ferner z.B. die Nicotinate, Isonicotinate, Diäthylaminoacetate und deren Säureadditionssalze, vornehmlich deren Hydrochloride. Besonders wichtig sind die Schwefelsäureund Phosphorsäureester und deren physiologisch verträgliche Metall-, insbesondere Alkalimetall- (z.B. Natrium-) und Ammoniumsalze, da in ihnen wasserlösliche und somit therapeutisch besonders gut applizierbare Derivate der Verbindungen der Formel I vorliegen.
Der Ausdruck Estersalze soll im Rahmen der vorliegenden Anmeldung die Säureadditionssalze basisch substituierter Ester und die Metall- und Ammoniumsalze saurer Ester einschliessen.
Als Verbindungen der Formel II kommen vor allem die Chalkone (X = -CR1 = CH-) in Frage.
Die Verbindungen der Formel II können vor allem durch Einwirkung von basischen oder sauren Katalysatoren zu den Flavanderivaten der Formel I cyclisiert werden. Vorzugsweise verwendet man als Katalysatoren Alkalien wie Natrium- oder Kaliumhydroxid, Natriumamid, Natriumhydrid, basisch reagierende Salze wie Natriumoder Kaliumacetat, Natrium- oder Kaliumcarbonat, Pufferlösungen, beispielsweise solche aus Citronensäure und Dinatriumphosphat oder aus Natrium- oder Kaliumdihydrogenphosphat und Borax oder aus Borsäure, Natriumhydroxid und Kaliumchlorid, organische Basen wie Piperidin, Pyridin, Tetramethylguanidin, Benzyltrimethylammoniumhydroxid, Mineralsäuren wie Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Polyphosphorsäure; organische Sulfonsäuren wie Toluolsulfonsäure oder Camphersulfonsäure.
Die Cyclisierung kann in Gegenwart eines zusätzlichen inerten Lösungsmittels wie Methanol, Äthanol, Dioxan, Tetrahydrofuran, Essigester, Essigsäure, Tetralin, Benzol, Toluol, vorgenommen werden, gegebenenfalls auch in Gemischen dieser Lösungsmittel untereinander oder mit Wasser. Es ist auch möglich, einen Überschuss des Cyclisierungsmittels als Lösungsmittel zu verwenden.
Die Cyclisierung findet bei Raumtemperatur statt und kann durch Erwärmen, gegebenenfalls bis zum Siedepunkt des verwendeten Lösungsmittels, beschleunigt werden. Die Reaktionszeit beträgt einige Minuten bis einige Tage.
Die Chalkone werden vorzugsweise erhalten durch Kondensation eines in 5-Stellung substituierten 2-Hydroxyphenyl-alkyl-ketons der Formel m

worin R1 und R2 die angegebene Bedeutung haben und Hydroxygruppen auch in funktionell abgewandelter Form vorliegen können, mit Benzaldehyd oder auch aus einem p-substituierten Phenol und einem Zimtsäurederivat in Gegenwart von Aluminiumchlorid.
Es ist nicht notwendig, das als Ausgangsprodukt zu verwendende Chalkon zu isolieren, sondern man kann auch das Reaktionsgemisch aus dem Keton III und Benzaldehyd direkt mit dem Cyclisierungsmittel behandeln.
Eine besonders bevorzugte Ausführungsform der Erfindung besteht darin, dass man ein Gemisch aus dem Keton III und Benzaldehyd mit Basen wie NaOH, KOH oder Piperidin behandelt. Die Base dient dabei sowohl als Kondensationsmittel bei der Chalkonbildung wie auch als Cyclisierungsmittel. Die Reaktion kann mit oder ohne Anwesenheit eines zusätzlichen inerten Lösungsmittels erfolgen. Bevorzugte Lösungsmittel sind niedere Alkohole wie Methanol, Äthanol, Isopropanol oder tert.-Butanol.
Die Umsetzung wird zweckmässig durch mehrstündiges Erwärmen zu Ende geführt.
Typische Ketone der Formel III sind 2,5-Dihydroxyphenyl-äthylketon (-propiophenon), -propylketon (-butyrophenon), -n-butylketon, -isobutylketon, -n-amylketon, -isoamylketon, -n-hexylketon, -isohexylketon, -n-heptylketon und-isoheptylketon, 2-Hydroxy-5-methoxyphenyl -äthylketon, -propylketon, -n-butylketon und -isobutylketon, 2-Hydroxy-5-äthoxyphenyl-äthylketon, -propylketon, -n-butylketon und -isobutylketon, 2-Hydroxy-5-n -propoxyphenyl-äthylketon, -propylketon, n-butylketon und -isobutylketon, 2-Hydroxy-5-isopropoxyphenyl-äthylketon, -propylketon, -n-butylketon und -isobutylketon, 2 -Hydroxy-5 - isobutoxyphenyl - äthylketon, -propylketon, -n-butylketon und -isobutylketon, 2-Hydroxy-5 -isoamyl- oxyphenyl-äthylketon, propylketon, -n-butylketon und -isobutylketon.
Bei den vorstehend beschriebenen Umsetzungen der Verbindungen der Formeln II bzw. m ist es möglich, dass phenolische Hydroxygruppen in funktionell abgewandelter Form vorliegen. Unter den Bedingungen der Kondensation können solche funktionell abgewandelten Hydroxygruppen in Freiheit gesetzt werden. So kann man Verbindungen, in denen Hydroxygruppen als Tetrahydropyranyläther geschützt vorliegen, in saurem oder alkalischem Medium cyclisieren; im Falle einer alkalischen Cyclisierung kann die Hydroxygruppe durch nachfolgendes kurzes Kochen mit Säure in Freiheit gesetzt werden.
Verbindungen mit als Ester geschützter Hydroxygruppe können ebenfalls in saurem oder alkalischem Medium kondensiert werden, wobei die Estergruppe verseift werden kann.
Ferner sind Äthergruppen, wie Benzyläther oder Methyläther, als Schutzgruppen geeignet. Die Spaltung solcher Äther kann beispielsweise erfolgen, wenn man als Cyclisierungsmittel Bromwasserstoffsäure unter solchen Bedingungen verwendet, unter denen bekanntermassen eine Spaltung von Phenoläther erfolgt.
Sofern in dem erhaltenen Cyclisierungsprodukt noch funktionell abgewandelte Hydroxygruppen vorhanden sind, werden sie nach an sich bekannten Methoden durch Behandeln mit hydrolysierenden oder hydrogenolysierenden Mitteln in Freiheit gesetzt. So ist es beispielsweise möglich, eine veresterte Hydroxygruppe durch Behandeln mit basischen oder sauren Mitteln zu hydrolysieren.
Als Basen kommen vornehmlich wässriges, wässrig-alkoholisches oder alkoholisches Natrium- oder Kaliumhydroxid, als Säuren vor allem Salzsäure und Schwefelsäure in Betracht. Benzyläther können durch Hydrogenolyse in Gegenwart von Edelmetall-Katalysatoren, z.B. Palladium-Kohle, gespalten werden, wobei die zugrundeliegende Hydroxygruppe in Freiheit gesetzt wird.
Es ist ferner möglich, eine freie Hydroxygruppe zu verestem oder zu alkylieren.
Eine Veresterung von Hydroxygruppen kann z.B.
durch Erhitzen mit einem Anhydrid oder Halogenid einer Carbonsäure, die vorzugsweise 1 - 6 C-Atomen besitzt, erfolgen. Typische Säuren sind Essig-, Propion-, Butter-, Isobutter-, Valerian-, Isovalerian-, Capron-, Nicotin- oder Isonicotinsäure. Die Veresterung erfolgt vorteilhaft in Gegenwart einer Base wie Pyridin oder eines Alkalisalzes der enstprechenden Säure oder auch einer geringen Menge Mineralsäure wie Schwefelsäure oder Salzsäure.
Zur Herstellung der Schwefelsäure- und Phosphorsäureester der Verbindungen der Formel I (R2 = H) setzt man diese mit Schwefelsäure, Phosphorsäure oder einem zur Veresterung geeigneten Derivat dieser Säuren um, wobei man nach an sich aus der Literatur bekannten Methoden arbeitet.
Es ist auch möglich, die Reaktion mit einem Schwefelsäure- bzw. Phosphorsäurederivat, in welchem eine bzw. zwei Hydroxygruppen blockiert sind, durchzuführen, und in den so erhaltenen Estern die vorhandenen Schutzgruppen anschliessend hydrolytisch oder hydrogenolytisch zu entfernen. Schliesslich kann man die erhaltenen Schwefelsäure- bzw. Phosphorsäureester durch Behandeln mit Basen in ihre physiologisch verträglichen Metall- bzw. Ammoniumsalze überführen.
Eine Alkylierung kann beispielsweise durch Umsetzung mit Alkylhalogeniden, -sulfaten oder niederen Alkylestern erfolgen, deren Alkylgruppe 1 - 6 C-Atome besitzt. Zur Herstellung der Diakyiaminoalkyläther verwendet man Dialkylaminoalkylhalogenide, -sulfate oder niedere Dialkylaminoalkylester, deren Dialkylaminoalkylgruppe 4 - 8 C-Atomen besitzt. In der Regel arbeitet man in Gegenwart von Alkali wie Natrium- oder Kaliumhydroxid oder -carbonat, wobei auch eines der üblichen inerten Lösungsmittel zugegen sein kann.
Dementsprechend können die Ausgangsverbindungen umgesetzt werden mit Methyljodid, Dimethylsulfat, Äthyl-, Propyl-, Isopropyl-, Butyl-, Isobutyl-, Amyl-, Isoamylhalogeniden, 2-Dimethylaminoäthyl-, 2-Diäthylaminoäthyl-, 2- (Methyl- äthylamino)-äthyl-, 2-Pyrrolidinoäthyl-, 2-piperidino äthyl-, 2-Morpholinoäthyl-, 3 -Dimethylaminopropyl-, 3 -Diäthylaminopropyl-, 3-Pyrrolidinopropyl-, 3-Piperidinopropyl- oder 3-Morpholinopropylhalogeniden oder auch mit den entsprechenden Alkoholen. Als Halogenide sind die Chloride, Bromide und Jodide geeignet. Die Verätherungen können beispielsweise nach den Methoden einer Williamson-Synthese erfolgen, wobei man von den entsprechenden Alkaliphenolaten ausgeht.
Es ist aber auch möglich, die freien Phenole mit den entsprechenden Alkoholen bzw. substituierten Aminoalkoholen in Gegenwart saurer Katalysatoren wie Schwefelsäure, Phosphorsäure, p-Toluolsulfonsäure, umzusetzen.
Ferner ist es möglich, basische Verbindungen der Formel I durch Behandeln mit Säuren in ihre physiologisch verträglichen Säureadditionssalze zu überführen. Für diese Umsetzung kommen in Frage organische und anorganische Säuren, z.B. aliphatische, alicyclische, araliphatische, aromatische oder heterocyclische ein- oder mehrbasige Carbon- oder Sulfonsäuren, wie Ameisensäure, Essigsäure, Propionsäure, Pivalinsäure, Diäthylessigsäure, Oxalsäure, Malonsäure, Bemsteinsäure, Pimelinsäure, Fumarsäure, Maleinsäure, Milchsäure, Weinsäure, Äpfelsäure, Aminocarbonsäuren, Sulfaminsäure, Benzoesäure, Salicylsäure, Phenylpropionsäure, Citronensäure, Gluconsäure, Ascorbinsäure, Isonicotinsäure, Methansulfonsäure, Naphthalin-mono- und -disulfonsäuren, Schwefelsäure, Salpetersäure, Halogenwasserstoffsäuren, wie Chlorwasserstoffsäure oder Bromwasserstoffsäure,
oder Phosphorsäuren, wie Orthophosphorsäure.
Eine Umwandlung von basischen Flavanen der Formel I in ihre physiologisch verträglichen quartären Ammoniumderivate gelingt durch Behandeln mit Alkylierungsmitteln, wie Methyljodid, Dimethylsulfat, Äthylbromid, Äthyljodid.
Vorzugsweise können nach der Erfindung Verbindungen der Formeln (sowie gegebenenfalls ihre Ester, Säureadditionssalze und quartären Ammoniumderivate) erhalten werden:

worin RG H oder R7-(CHo)n und R7 Dimethylamino, Di äthylamino, Pyrrolidino, Piperidino oder Morpholino bedeuten.
Die neuen Verbindungen können im Gemisch mit üblichen Arzneimittelträgern in der Human- oder Veterinärmedizin eingesetzt werden. Als Trägersubstanzen kommen solche organischen oder anorganischen Stoffe in Frage, die für die parenterale, enterale oder topikale Applikation geeignet sind und die mit den neuen Verbindungen nicht in Reaktion treten, wie beispielsweise Wasser, pflanzliche Öle, Polyäthylenglykole, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, Vaseline, Cholesterin usw. Zur parenteralen Applikation dienen insbesondere Lösungen, vorzugsweise ölige oder wässrige Lösungen, sowie Suspensionen oder Emulsionen.
Für die enterale Applikation können ferner Tabletten oder Dra genes, für die topikale Anwendung Salben oder Cremes, die gegebenenfalls sterilisiert oder mit Hilfsstoffen, wie Konservierungs-, Stabilisierungs- oder Netzmitteln oder
Salzen zur Beeinflussung des osmotischen Druckes oder mit Puffersubstanzen versetzt sind, angewendet werden.
Die neuen 3-Alkylflavanone werden vorzugsweise in einer Dosierung von 1 bis 500 mg pro Dosierungseinheit verabfolgt.
Beispiel I a) 1 g Benzaldehyd und 1,6 g 2,5-Dihydroxy-propiophenon werden in 8 ml trockenem Piperidin und 14 ml absolutem Äthanol 15 Stunden gekocht. Man rührt in Eiswasser ein, säuert mit Salzsäure an und extrahiert mit
Chloroform. Die organische Phase wird mit Wasser gewaschen, über Natriumsulfat getrocknet und zur Trockne eingedampft. Aus Benzol kristallisiert 3 -Methyl-6-hy- droxy-flavanon, F. 174 - 1760.
Analog erhält man aus den entsprechenden 2,5-Dihydroxyphenyl-alkyl -ketonen: 3-Äthyl-6-hydroxy-flavanon, F. 147-1490; 3-n-Propyl-6-hydroxy-flavanon, F. 145-1460: 3 -Isopropyl-6-hydroxy-flavanon ; 3-n-Butyl- 6-hydroxyflavanon; 3-Isobutyl-6-hydroxy-flavanon; 3 -n-Amyl-6-hydroxy-flavanon; 3 -Isoamyl-6-hydroxy-flavanon ; 3 -n-Hexyl-6-hydroxy-flavanon; 3 4sohexyl. 6-hydroxy-flavanon.
b) Eine Lösung von 1 g 3-Äthyl-6-hydroxy-flavanon in 20 ml Aceton wird mit einer ätherischen Lösung von 5 g Diäthylaminoäthylchlorid und 1 g wassertreiem Kaliumcarbonat versetzt, 20 Stunden gekocht, dann in Wasser eingerührt und mit Chloroform extrahiert. Der Extrakt wird mit Wasser gewaschen, über Natriumsulfat getrocknet und zur Trockne eingedampft, der Rückstand in Äthanol gelöst und die Lösung mit in Äthanol gelöster Oxalsäure versetzt. Nach einigem Stehen kristallisiert das Oxalat des 3 -Äthyl-6.(2.diäthylaminoäthoxy) -flavanons aus, F. 134-1350.
Analog erhält man 3-Methyl-6-(2-diäthylamino äthoxy)-flavanon, Hydrochlorid, F. 168-1700.
c) Eine Lösung von 1 g 3-Methyl-6-hydroxy-flavanon in 6 ml Pyridin wird bei 900 mit 1,1 g Amidosulfonsäure versetzt und 3 Stunden bei 900 gerührt. Man kühlt ab, wäscht mit 30 ml Äther, mischt die Pyridinphase mit 17 ml 12 0iger Natronlauge und 12 Pyridin, trennt ab, wäscht erneut mit Äther und dampft ein. Aus Äthanol kristallisiert das Natriumsalz des 3-Methyl-6-hydroxy-flavanon-6-schwefelsäureesters, F. 178-1800.
d) 1 g 3-n-Propyl-6-hydroxy-flavanon wird in einem Gemisch von 5 ml trockenem Pyridin und 5 ml Nicotinsäurechlorid 30 Minuten auf dem Dampfbad erhitzt. Man giesst in Wasser, saugt ab, trocknet, löst in wenig Äthanol und fällt mit ätherischer Salzsäure das Hydrochlorid des Nicotinsäureesters des 3-n-Propyl-6-hydroxy-flavanons. F. 168-1700. Die freie Base wird aus dem Hydrochlorid mit Natronlauge erhalten und durch fraktionierte Kristallisation aus Methanol in cis- (F. 131-1330) und trans-3-n-Propyl-6-hydroxy-flavanon.6.nicotinat (F. 90 920) getrennt.
e) 1 g 3-Methyl-6-hydroxy-flavanon wird in 5 ml Pyridin und 5 ml Acetanhydrid 5 Stunden auf 500 erhitzt.
Nach dem Abkühlen gibt man Wasser und Chloroform hinzu, trennt ab, wäscht die Chloroformschicht mehrfach mit Wasser und destilliert das Chloroform ab.
Man erhält 3 -Methyl-6-acetoxy-fiavanon, F. 127-1290 (Methanol).
Analog erhält man 3-Äthyl-6-acetoxy-flavanon, F.
107-1090 (Methanol).
f) Zu einer Lösung von 20 ml Phosphoroxychlorid in
210 ml absolutem Pyridin werden unter Rühren bei 50 in
15 Minuten 7,5 g 3-Methyl-6-hydroxy-flavanon gelöst und in 100 ml Pyridin getropft. Man lässt über Nacht bei Raumtemperatur stehen, giesst auf ein Gemisch von Eis und konzentrierter Salzsäure und erhitzt 90 Minuten im Dampfbad. Nach dem Abkühlen extrahiert man mit Äthylacetat, wäscht die Extrakte mit verdünnter Salzsäure, trocknet über Natriumsulfat, filtriert, destilliert das Lösungsmittel ab und erhält 3-Methyl-6-hydroxy-flavanon-6-orthophosphat.
Beispiel 2 a) 1 g o-Benzyliden-2-hydroxy-5-(2-diäthylamino- äthoxy)-propiophenon, erhalten durch Kondensation von 2.Hydroxy.5.(2.diäthylaminoäthoxy).propiophenon mit Benzaldehyd in wässerig-methanolischer Natronlauge bei Raumtemperatur, wird in 50 ml Äthanol in Gegenwart von 1 g p-Toluolsulfonsäure 2 Stunden bei 1300 im Bom- benrohr erhitzt. Man kühlt ab, gibt verdünnte Natronlauge und Chloroform hinzu, trennt ab, trocknet die Chloroformschicht über Natriumsulfat und dampft zur Trockne ein. Der Rückstand wird in Äthanol gelöst und mit ätherischer Salzsäure versetzt.
Man erhält 3 .Methyl.6.(2.diäthylaminoäthoxy) -flava- non-hydrochlorid, F. 168-1700.
b) Man schüttelt 0,5 g des wie vorstehend erhaltenen Hydrochlorids mit 2 ml verdünnter Natronlauge und 10 ml Äther, trennt die Schichten, trocknet die Ätherphase über Magnesiumsulfat und versetzt mit einem Überschuss Methyljodid. Das nach 24stündigem Stehen ausgefallene 3-Methyl-6-(2-diäthylaminoäthoxy)-flavanon-methojodid wird abfiltriert.
Beispiel 3
1 g 2-Hydroxy-5-benzyloxy-propiophenon und 0,5 g Benzaldehyd werden in 10 ml Äthanol gelöst, mit 5 g 50%iger Kalilauge versetzt und 5 Minuten geschüttelt.
Man versetzt mit Wasser, saugt ab, wäscht sorgfältig mit Wasser und erhält 3-Methyl-6-benzyloxy-flavanon, das ohne weitere Aufreinigung in 40 ml mit Chlorwasserstoff gesättigtem Ätehylacetat in 5%iger Palladiumkohle bei 350 bis zu Aufnahme der berechneten Menge Wasserstoff hydrogenolysiert wird. Man filtriert den Katalysator ab, entfernt das Lösungsmittel und kristallisiert das erhaltene 3-Methyl-6-hydroxy-flavanon aus Benzol um. F.
174-1760.
Beispiel 4
Man setzt 2 g 2-Hydroxy-5-(tetrahydropyranyl-2-oxy) -propiophenon analog Beispiel 4 mit Benzaldehyd um und kocht das erhaltene rohe 3-Methyl-6-(tetrahydro pyranyl.2.oxy).flavanon 2 Stunden mit 5%iger wässerig äthanolischer Salzsäure. Das Reaktionsgemisch wird in Wasser eingerührt und wie in Beispiel 1 a) aufgearbeitet.
Man erhält 3-Methyl-6-hydroxy-flavanon, F. 174-1760.

Claims (1)
- **WARNUNG** Anfang CLMS Feld konnte Ende DESC uberlappen **. die gegebenenfalls sterilisiert oder mit Hilfsstoffen, wie Konservierungs-, Stabilisierungs- oder Netzmitteln oder Salzen zur Beeinflussung des osmotischen Druckes oder mit Puffersubstanzen versetzt sind, angewendet werden.Die neuen 3-Alkylflavanone werden vorzugsweise in einer Dosierung von 1 bis 500 mg pro Dosierungseinheit verabfolgt.Beispiel I a) 1 g Benzaldehyd und 1,6 g 2,5-Dihydroxy-propiophenon werden in 8 ml trockenem Piperidin und 14 ml absolutem Äthanol 15 Stunden gekocht. Der Extrakt wird mit Wasser gewaschen, über Natriumsulfat getrocknet und zur Trockne eingedampft, der Rückstand in Äthanol gelöst und die Lösung mit in Äthanol gelöster Oxalsäure versetzt. Nach einigem Stehen kristallisiert das Oxalat des 3 -Äthyl-6.(2.diäthylaminoäthoxy) -flavanons aus, F. 134-1350.Analog erhält man 3-Methyl-6-(2-diäthylamino äthoxy)-flavanon, Hydrochlorid, F. 168-1700.c) Eine Lösung von 1 g 3-Methyl-6-hydroxy-flavanon in 6 ml Pyridin wird bei 900 mit 1,1 g Amidosulfonsäure versetzt und 3 Stunden bei 900 gerührt. Man kühlt ab, wäscht mit 30 ml Äther, mischt die Pyridinphase mit 17 ml 12 0iger Natronlauge und 12 Pyridin, trennt ab, wäscht erneut mit Äther und dampft ein. Aus Äthanol kristallisiert das Natriumsalz des 3-Methyl-6-hydroxy-flavanon-6-schwefelsäureesters, F. 178-1800.d) 1 g 3-n-Propyl-6-hydroxy-flavanon wird in einem Gemisch von 5 ml trockenem Pyridin und 5 ml Nicotinsäurechlorid 30 Minuten auf dem Dampfbad erhitzt. Man giesst in Wasser, saugt ab, trocknet, löst in wenig Äthanol und fällt mit ätherischer Salzsäure das Hydrochlorid des Nicotinsäureesters des 3-n-Propyl-6-hydroxy-flavanons. F. 168-1700. Die freie Base wird aus dem Hydrochlorid mit Natronlauge erhalten und durch fraktionierte Kristallisation aus Methanol in cis- (F. 131-1330) und trans-3-n-Propyl-6-hydroxy-flavanon.6.nicotinat (F. 90 920) getrennt.e) 1 g 3-Methyl-6-hydroxy-flavanon wird in 5 ml Pyridin und 5 ml Acetanhydrid 5 Stunden auf 500 erhitzt.Nach dem Abkühlen gibt man Wasser und Chloroform hinzu, trennt ab, wäscht die Chloroformschicht mehrfach mit Wasser und destilliert das Chloroform ab.Man erhält 3 -Methyl-6-acetoxy-fiavanon, F. 127-1290 (Methanol).Analog erhält man 3-Äthyl-6-acetoxy-flavanon, F.107-1090 (Methanol).f) Zu einer Lösung von 20 ml Phosphoroxychlorid in 210 ml absolutem Pyridin werden unter Rühren bei 50 in 15 Minuten 7,5 g 3-Methyl-6-hydroxy-flavanon gelöst und in 100 ml Pyridin getropft. Man lässt über Nacht bei Raumtemperatur stehen, giesst auf ein Gemisch von Eis und konzentrierter Salzsäure und erhitzt 90 Minuten im Dampfbad. Nach dem Abkühlen extrahiert man mit Äthylacetat, wäscht die Extrakte mit verdünnter Salzsäure, trocknet über Natriumsulfat, filtriert, destilliert das Lösungsmittel ab und erhält 3-Methyl-6-hydroxy-flavanon-6-orthophosphat.Beispiel 2 a) 1 g o-Benzyliden-2-hydroxy-5-(2-diäthylamino- äthoxy)-propiophenon, erhalten durch Kondensation von 2.Hydroxy.5.(2.diäthylaminoäthoxy).propiophenon mit Benzaldehyd in wässerig-methanolischer Natronlauge bei Raumtemperatur, wird in 50 ml Äthanol in Gegenwart von 1 g p-Toluolsulfonsäure 2 Stunden bei 1300 im Bom- benrohr erhitzt. Man kühlt ab, gibt verdünnte Natronlauge und Chloroform hinzu, trennt ab, trocknet die Chloroformschicht über Natriumsulfat und dampft zur Trockne ein. Der Rückstand wird in Äthanol gelöst und mit ätherischer Salzsäure versetzt.Man erhält 3 .Methyl.6.(2.diäthylaminoäthoxy) -flava- non-hydrochlorid, F. 168-1700.b) Man schüttelt 0,5 g des wie vorstehend erhaltenen Hydrochlorids mit 2 ml verdünnter Natronlauge und 10 ml Äther, trennt die Schichten, trocknet die Ätherphase über Magnesiumsulfat und versetzt mit einem Überschuss Methyljodid. Das nach 24stündigem Stehen ausgefallene 3-Methyl-6-(2-diäthylaminoäthoxy)-flavanon-methojodid wird abfiltriert.Beispiel 3 1 g 2-Hydroxy-5-benzyloxy-propiophenon und 0,5 g Benzaldehyd werden in 10 ml Äthanol gelöst, mit 5 g 50%iger Kalilauge versetzt und 5 Minuten geschüttelt.Man versetzt mit Wasser, saugt ab, wäscht sorgfältig mit Wasser und erhält 3-Methyl-6-benzyloxy-flavanon, das ohne weitere Aufreinigung in 40 ml mit Chlorwasserstoff gesättigtem Ätehylacetat in 5%iger Palladiumkohle bei 350 bis zu Aufnahme der berechneten Menge Wasserstoff hydrogenolysiert wird. Man filtriert den Katalysator ab, entfernt das Lösungsmittel und kristallisiert das erhaltene 3-Methyl-6-hydroxy-flavanon aus Benzol um. F.174-1760.Beispiel 4 Man setzt 2 g 2-Hydroxy-5-(tetrahydropyranyl-2-oxy) -propiophenon analog Beispiel 4 mit Benzaldehyd um und kocht das erhaltene rohe 3-Methyl-6-(tetrahydro pyranyl.2.oxy).flavanon 2 Stunden mit 5%iger wässerig äthanolischer Salzsäure. Das Reaktionsgemisch wird in Wasser eingerührt und wie in Beispiel 1 a) aufgearbeitet.Man erhält 3-Methyl-6-hydroxy-flavanon, F. 174-1760.PATENTANSPRÜCHE I. Verfahren zur Herstellung von 3-Alkyl-flavanonen der Formel I EMI4.1worin R1 Alkyl mit 1 - 6 C-Atomen, R2 H, Alkyl mit 1 - 6 C-Atomen oder Z(c:!)n- Z Dialkylamino mit 2 - 6 C-Atomen, Pyrrolidino, Pi peridino oder Morpholino und n 2 oder 3 bedeuten, dadurch gekennzeichnet, dass man ein Keton der For melII EMI5.1 worin EMI5.2 A OH oder sauerstoffhaltig funktionell abgewandeltes OH, X A oder Hal und Hal C1, Br, J bedeuten, R1 und R2 die angegebene Bedeutung haben, R2 aber ausserdem noch Acyl bedeuten kann, mit cyclisierenden Mitteln behandelt, und eine gegebenenfalls vorhandene Acylgruppe abspaltet.II. Verwendung der nach Patentanspruch I hergestellten tertiären Amine zur Herstellung ihrer quaternären Ammoniumsalze, dadurch gekennzeichnet, dass diese Amine mit quaternisierenden Mitteln behandelt werden.UNTERANSPRÜCHE 1. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man in dem erhaltenen Produkt eine funktionell abgewandelte Hydroxygruppe durch Behandein mit hydrolysierenden oder hydrogenolysierenden Mitteln in Freiheit setzt.2. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man in dem erhaltenen Produkt eine freie Hydroxygruppe durch Behandeln mit veresternden Mitteln verestert.3. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man in dem erhaltenen Produkt eine freie Hydroxygruppe durch Behandeln mit alkylierenden Mitteln alkyliert.4. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man erhaltene Verbindungen durch Behandeln mit Säuren in ihre physiologisch verträglichen Säureadditionssalze überführt.5. Verfahren nach Unteranspruch 2, dadurch gekennzeichnet, dass man erhaltene saure Ester durch Behandeln mit Basen in ihre physiologisch verträglichen Estersalze überführt.
