Verfahren zur Herstellung der Verbindung III aus Verbindung IV durch intramolekulare Cyclisierung.
Die Verbindung der Formel IV wird cyciisiert durch Erhitzen in einem Lösungsmittel in Gegenwart eines gewöhnlich für die Dieckmann-Reaktion verwendeten basischen Katalysators, beispielsweise metallisches Natrium oder Kalium, Natriumhydrid oder Natriumäthoxid, wobei die intramolekulare Cyclisierung der Verbindung unter Bildung der Verbindung III vor sich geht. Diese Reaktion erfolgt mit besseren Ergebnissen, wenn eine katalytische Menge von Methanol oder Äthanol oder eines anderen Niederaikohois zugesetzt wird.
Als Lösungsmittel zur Verwendung in dieser Reaktion eignen sich aromatische Kohlenwasserstoffe, wie Benzol oder Toluol, oder Äther, wie Dioxan, Tetrahydrofuran, 1 ,2-Di- methoxyäthanol oder Diäthylenglykoldimetl1ylätheT. Die Er hitzungstemperatur ist nicht besonders eingeschränkt, es werden aber gewöhnlich Temperaturen im Bereich von 60 - SO0C verwendet.
Verfahren zur Herstellung der Verbindung II aus Verbindung III.
Beispielsweise wird die Verbindung der Formel III und ein konventionelles Dehydrierungsmittel, wie 2,3 -Dichlor- -S,6dicyano- 1 4-benzochinon (DDQ), Tetrachlor-1,4-benzo- chinon (Chloranil), Tetracyanoäthylen, Palladium-Kohlenstoff (Pd-C), N-Bromtsuccinimid (NBS), Mangan-dioxid oder Selen-dioxid während einer kurzen Zeitspanne, bei einer Temperatur in der Nähe des Siedepunktes des verwendeten Lösungsmittels, erhitzt. Oder es wird auch die Verbindung der Formel III in obigem Lösungsmittel ohne Verwendung eines Dehydrierungsmittels in der Wärme behandelt, oder die Verbindung III kann auch direkt auf eine Temperatur oberhalb ihres Schmelzpunktes erhitzt werden, wobei sich die Verbindung II bildet.
Das bevorzugte Lösungsmittel ist ein inertes Lösungsmittel, beispielsweise ein aromatischer Kohlenwasserstoff, wie Benzol, Toluol oder Xylol, ein Ather, wie Dioxan, oder ein Alkohol, wie Methanol oder Äthanol. Die Erhitzungstemperatur beträgt etwa 60 - 2600C.
Verfahren zur Herstellung der Verbindung I aus Verbindung II' durch Hydrolyse.
Von den Verbindungen II wird die Verbindung II', in welcher R2 ein Cyano oder ein Niederalkoxycarbonyl ist, mittels üblicher Methoden hydrolysiert, unter Bildung der Verbindung T. Diese Reaktion kann so erfolgen, dass man die Verbindung II' mit einer Mineralsäure erhitzt und dann das Reaktionsprodukt in Wasser schüttet; oder durch einfaches Erhitzen auf 70 - 1200C oder durch Erwärmen dieser Verbindung in einem Lösungsmittel, wie Wasser oder Wasser enthaltendem heehsiedendem Äther, wie Dioxan oder 1 ,2-Di- methoxyäthan oder Wasser enthaltendem Alkohol, wie Äthanol, Propanol oder Äthylenglykol, in Gegenwart einer organischen Säure oder Mineralsäure.
Auch kann diese Reaktion so durchgeführt werden, dass man die Verbindung II' zusammen mit Wasser oder einem Wasser enthaltenden Alkohol in Gegenwart einer Base erhitzt oder erwärmt.
Bei der Verbindung II', in welcher R2 ein Niederalkoxycarbonyl ist, geht die Hydrolyse unter milderen Bedingungen vor sich, als bei der Verbindung, in welcher R2 eine Cyanogruppe ist.
Beispiele von Säuren, die bei der Hydrolyse verwendet werden können, sind anorganische Säuren, wie Chlorwasserstoff-, Bromwasserstoff-, Jodwasserstoff-, Schwefel- oder Phosphorsäure und organische Säuren, wie Essig-, Oxal oder Toluolsulfonsäure.
Beispiele der Basen sind Aikalimetallhydroxide, wie Natrium- oder Bariumhydroxid, Alkalimetall-carbonate, wie Natrium- oder Kaliumcarbonat und Natriumacetat.
In der Verbindung der Formel II, in welcher R'l eine Acylgruppe ist, wird die Acylgruppe in obiger Reaktion hydrolysiert und durch ein Wasserstoffatom ersetzt.
Verwendung der Verbindung II" zur Herstellung der Verbindung I durch Oxidation.
Von den Verbindungen der Formel II werden die Verbindungen II", worin R2 ein Niederalkanoyl ist, zur Herstellung der Formel I durch Oxidation verwendet. Diese Oxidation ist ein übliches Verfahren und kann so erfolgen, dass man die Verbindung II" in einem Lösungsmittel löst oder suspendiert und ein Oxidationsmittel auf diese Verbindung einwirken lässt.
Geeignete Oxidationsmittel, welche in dieser Reaktion verwendet werden können, sind beispielsweise Permanganate, wie Kaliumpermanganat, Hypohalogenite, wie Kaliumhypo chiont, Natriumhypobromit oder Natriumhypoiodit und Salpetersäure. Bei dieser Reaktion kann auch eine indirekte Oxidationsmethode verwendet werden, in welcher man ein Gemisch von Jod und Pyridin auf die Verbindung zuerst einwirken lässt und dann das so erhaltene Produkt mit Alkali behandelt.
Die Behandlung der Verbindung II mit Brom oder Jod in einer Natriumhydroxid- oder Kaliumhydroxid-Lösung bewirkt dasselbe, wie die Oxidation mit einer hypohalogeniden Säure.
Das Lösungsmittel und die Reaktionstemperaturen sollen entsprechend den Eigenschaften des verwendeten Oxidationsmittels gewählt werden. Geeignete Lösungsmittel sind Wasser, Äther, wie Dioxan oder 1,2-Dimethoxyäthan, Ketone, wie Aceton, Pyridine, wie Pyridin oder Picolin, Halogenalkane, wie Chloroform oder Tetrachlormethan, Alkohole, wie Äthanol oder t-Butanol, oder Gemische von Wasser, mit obengenannten organischen Lösungsmitteln. Im allgemeinen schreitet die Reaktion bei Zimmertemperatur genügend oder auch unter Eiskühlung fort.
Verbindung II', worin R'1 eine Acylgruppe ist, kann oxidiert und dann hydrolysiert werden, oder unter solchen Bedingungen oxidiert werden, welche auch eine Hydrolyse induzieren.
Bei der Herstellung von Verbindung II oder I aus Verbindung IV in obigen Reaktionsreihen kann das Produkt jeder Stufe isoliert und dann der nachfolgenden Reaktion unterworfen werden; es kann aber auch das Verfahren zur Herstellung der nächsten Verbindung direkt, ohne jegliche Isolierung und Reinigung, erfolgen. Falls der Reaktionskatalysator, die Temperatur, das Lösungsmittel usw. auf geeignete Weise gewählt werden, kann die Verbindung II oder I in einer Stufe aus der Verbindung IV oder III erhalten werden.
Im Falle aber eines Verfahrens, welches lediglich die Endstufe der Herstellung der Verbindung I durch Hydrolyse der isolierten Verbindung II' umfasst, kann jener Fall, bei welchem in einer Verbindung II' der Substituent R'2 ein Niederalkoxycarbonyl ist, weggelassen werden.
Das Ausgangsmaterial der Formel IV ist eine neue Verbindung und kann leicht durch Erhitzen von Verbindungen a) und b) in Gegenwart einer Base gemäss dem folgenden Reaktionsschema erhalten werden:

Die Verbindungen der Formel 1, die aus den erfindungsgemäss hergestellten Verbindungen hergestellt werden können, können mittels üblicher Methoden isoliert und gereinigt werden. Die Verbindungen I können in freiem Zustand, als Hydrat oder in Form eines Salzes, je nach den Ausgangsmaterialien und den Reaktionsbedingungen, erhalten werden.
Überdies können die Verbindungen I in pharmazeutisch annehmbare Aminsalze oder Carbonsäuresalze, durch Behandlung mit Säure oder Alkali oder vice versa, umgewandelt werden. Die Säure kann aus einer Vielfalt von organischen und anorganischen Säuren gewählt werden; es kann beispielsweise verwendet werden Chlorwasserstoff-, Essig-, Milch-, Bernstein-, Oxal- und Methansulfonsäure.
Die klinische Dosis dieser Verbindung I hängt vom Körpergewicht, Alter und Verabreichungsweise ab, liegt aber im allgemeinen im Bereich von 100 mg bis 5 g pro Tag, vorzugsweise 200 mg - 3 g pro Tag.
Die Verbindung der Formel I kann als Heilmittel, beispielsweise in Form von pharmazeutischen Präparaten, die die Verbindung im Gemisch mit anorganischen oder organischen, festen oder flüssigen pharmazeutischen Formulierungshilfsmitteln, die sich für orale, parenterale, enterale oder lokale Verabreichungen eignen, verwendet werden. Diese Präparate können ferner andere therapeutisch wertvolle Substanzen enthalten.
Die Verbindungen der Formel I und ihre Salze haben überlegene antibakterielle Wirksamkeiten und werden in verschiedenen Formulierungen in der Human- und Veterinärmedizin verwendet.
Die antibakterielle Wirksamkeit der typischen Verbindungen der Formel I sind in folgenden Tabellen I - IV angeführt.
In Tabellen II - IV wurden die ED,, und LD50-Werte gemäss der Behrens-Kaerber'schen Methode (Arch. Exp. Path.
Pharm. 162,480 (1931)) bestimmt.
Verbindungen Nr. 1-4 in folgenden Tabellen bedeuten die folgenden Verbindungen: Verbindung Nr. 1: 8-Äthyl-5,8-dihydro-5-oxo-2-(1-piperazin- yi)-pyrido(2,3 -d)pyrimidin-6-carbonsäure, Verbindung Nr. 2: 8-Äthyl-2-(4-methyl-1-piperazinyl)-5,8-di- hydro-5-oxopyrido(2,3 -dXpyrimidin-6-carbonsäure, Verbindung Nr. 3: 8-Äthyl-2-(4-äthyl-1-piperazinyl)-5,8-di- hydro-5-oxopyrido(2,3 -d)pyrimidin-6-carbonsäure, Verbindung Nr. 4: 8-Äthyl-5,8-dihydro-5-oxo-2-(l-piperazin- yl)-pyrido(2,3-d)pyrimidin-6-carbonsäure.

Die Zahlen in der Tabelle zeigen das Verhältnis der Anzahl der Überlebenden gegenüber der verwendeten Gesamtzahl. 0/6 bedeutet, dass sämtliche 6 Mäuse verendeten und 6/6 bedeutet, dass sämtliche 6 Mäuse überlebten.
Die Verbindung war eine alkalische Lösung für intravenöse Verabreichung und in Suspension für orale Verabreichung, die 0,2% CMC enthielt, und wurde männlichen Mäusen (ddY-S) vom Gewicht von etwa 20 g verabreicht.
Nach 7 Tagen wurde der Eintritt des Todes registriert, und die LD50 -Werte wurden abgeschätzt.

Die vorliegende Erfindung wird im weiteren durch einige Beispiele näher erläutert.
Beispiel I Ä thyl-8-äthyl-2-(4-methyl-1 -piperazinyl)-S,8-dihydro- -5-oxo-pyrido(2,3-d) pyrimidin-6-carboxylat
Zu einer Lösung von 4,0 g Äthyl-4-(äthyl-2-äthoxy-car bonyläthyl)-amino-2-(4-methyl-1 -piperazinyl)-pyrimidin-5- -carboxylrat in 10 ml von 1 ,2-Dimethoxyäthan wurden 0,75 g einer 50%igen Natriumhydrid-Suspension zugesetzt, und das Gemisch wurde auf Rückfluss während 1 h erhitzt. Nach Enitfernung des Lösungsmittels wurde der resultierende Rückstand mit Essigsäure neutralisiert und mit Chloroform extrahiert. Der Extrakt wurde dann mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet, und das Lösungsmittel wurde abdestilliert.
Man erhielt ein Zwischenprodukt, welches gewünschtenfalls aus n-Hexan-aceton umkristallisiert wurde und Äthyl-8-äthyl-2-(4-methyl-l-piperazinyl)-5-oxo- -5,6,7,8-tetrahydropyrido(2,3-d)pyrimidin-6-carboxylat in reiner Form mit F von 97 - 100"C ergab.
Dieser so erhaltene Tetrahydro-ester als Zwischenprodukt wurde in Benzol gelöst, und das Gemisch wurde auf Rückfluss während 1,5 h erhitzt. Der sich nach Kühlen ab geschiedene Feststoff gesammelt und aus Dimethylformamid umkristaltisiert. Man erhielt 2,5 g des oben angeführten Produktes mit F von 146 - 1470C.
Beispiel 2 8-Äthyl-2-(4-methyl4-piperazinyl) J.d-diydro=5- -oxopyrido(2,3-d)pyrimidin-6-carbonitril
Zu einem Gemisch mit 4,3 g Äthyl-4-(äthyI-2-cyano-äthyl)- -amino-2-(4-methyi-1 -piperazinyl) -pvrimidin-5carboxylat und 10 ml Dioxan wurden 0,8 g einer 50%igen Natrium hydridSuspension bei Zimmertemperatur unter Rühren zugegeben, und das Gemisch wurde auf Rückfluss während 30 min erhitzt. Nach Entfernung des Lösungsmittels wurde der sich ergebende Rückstand mit Essigsäure neutralisiert und mit Chloroform extrahiert.
Der Extrakt wurde gemäss Beispiel 1 aufgearbeitet, und man erhielt ein Zwischenprodukt, welches gegebenenfalls aus n-Hexan-aceton umkristallisiert werden kann und 8 -Äthyl-2-(4-methyl- 1 -piperazinyl)- -5- oxo-5,6,7,8-tetrahydropyrido(2,3-d)pyrimidin-6-carbonitril in reiner Form mit F von 172 - 174 C ergab.
Das erhaltene Tetrahydro-carbonitril wurde in 100 ml trockenem Benzol gelöst, und es wurden 2,5 g Chloranil zugesetzt. Das Gemisch wurde auf Rückfluss während 30 min erhitzt. Der sich nach Kühlen abgeschiedene Feststoff wurde gesammelt und aus Methanol umkristallisiert, und man erhielt 2,9 g des Produktes mit F von 255 - 258"C.
Beispiel 3
2-(4-Acetyl-l -piperazinyl)-8-äthyl-5,8-dihydro-5- -oxopyrido(2,3-dlpyrimidin-6-carbonitril
Zu einem Gemisch, das 5,0 g Äthyl-2-(4-acetyl-1 -pipera- zinyl)4-äthyl-2-cyanoäthyl-aminopyrimidin-5-carboxylat und 50 ml Äthylenglykol-dimethyläther enthielt, wurden 2,0 g einer 50%igen Natriumhydridsuspension zugesetzt. Dann wurde das Gemisch bei 950C während 1 h erhitzt. Das Gemisch wurde zur Trockne im Vakuum eingeengt und der sich ergebende Rückstand mit Essigsäure neutralisiert und mit Chloroform extrahiert. Das Lösungsmittel wurde vom Extrakt abgedampft.
Man erhielt ein öliges Zwischenprodukt, das 2-(4-Acetyl-1 -piperazinyl)-8-äthyl-5-oxo-5,6,7,8-tetrahydro- pyrido(2,3-d)-pyrimidin-6-carbonitril. Dieses Nitril wurde ohne weitere Reinigung in 50 ml Benzol gelöst, und es wurden zu diesem 2,5 g Chloranil zugefügt. Das Gemisch wurde auf Rückfiuss während 30 min erhitzt und der sich, nach Kühlung, abgeschiedene Feststoff gesammelt, mit Benzol gewaschen und aus Methanol umkristallisiert Man erhielt auf diese Weise 3,4 g des Produktes mit F von 3170C.
Beispiel4 Äthyl-2-(4-acetyl-1 -piperazinyl)-8-äthyI-5,8-dihydro- -5.oxopyrido(2,3-d)pyrimidin-6-carboxylat
Gemäss dem im Beispiel 3 beschriebenen Verfahren und unter Verwendung von Äthyl-2-(4-acetyl-l -piperazinyl)-4- -(äthyi-2-äthoxycarbonyläthyl)-aminopynmidin-5-carboxylat als Ausgangsmaterial erhielt man das oben angegebene Produkt mit F von 208 - 2100C (Zers.).
BeispielS 6-Acetyl-8-äthyl-5,8-dEhydro-2-(4-methyl -1 -piperazinyl)-5-oxopyrido(2,3-d)pyrimidin
Unter Befolgung des im Beispiel 3 beschriebenen Verfahrens und unter Verwendung von Äthyl-2-(4-acetyl-1-pipera- zinyl)-4-(äthyl-3-oxobutyl)-aminopyrimidin-5-carboxylat gelangte man zum oben angeführten Produkt mit F von 198 - 200 C
A) 8-Äthyl-2-(4-methyl-1 -piperazinyl)-5,8-dihydro- -5-oxopyndo(2,3-d)pyrimiiiin-6-carbonsüuie
Ein Gemisch, das 3,0 g 8-Äthyl-2-(4-methyl-1-piperazin- yl)-5,8-dihydro-5-oxopyrido(2,3-d)pyrimidin-6-carbonitril, 270 ml Essigsäure und 150 ml konzentrierte Chlorwasserstoffsäure enthält, wurde bei 90"C während 25 h erhitzt.
Das Gemisch wurde dann zur Trockne im Vakuum eingeengt. Der sich ergebende Rückstand wurde durch Zugabe einer 5%igen wässrigen Lösung von Natriumhydroxid neutralisiert und mit Chloroform extrahiert. Der Extrakt wurde nach dem Trocknen eingeengt und hinterliess einen rohen Feststoff, welcher aus Äthanol umkristallisiert wurde und 2,6 g des Produktes mit F von 232 - 2330C ergab.
B) 8-Äthyl-2-(4-äthyl-J -piperazinyl)-5,8-dihydro-5- -oxopyrido(2.3-d)pyrimidin-6-carbonsäure
Das Verfahren gemäss A) wurde wiederholt unter Verwendung von 8-Äthyl-2-(4-äthyl- 1 -piperazinyl)-5,8 < 1ihydro- -5-oxopyrido(2,3 4)pvrimidin-6-carbonitnl. Man erhielt das oben angeführte Produkt mit F von 228 - 2300C.
C) 8-Äthyl-5,8-dihydro-5-oxo-2-(J -piperazinyl) -pyrido(2,3-d) pyrmdin-6-carbonsäure-hydrochlorid
Ein Gemisch, das 5,0 g 2-(4-Acetyl-1 wiperazinyE)-8-äthyl- -5,8.dihydro-5-oxopyndo(2,3-d)pyrimidin-6.carbom.tnl, 200 ml Essigsäure und 200 ml konzentrierte Chlorwasserstoffsäure enthielt, wurde bei 900C während 3 h erhitzt. Danach wurde das Gemisch zur Trockne im Vakuum eingeengt. Zum Rückstand wurde eine kleine Menge konzentrierter Chlorwasserstoffsäure zugefügt und ein sich ergebendes Hydrochlorid wurde gesammelt und mit Äthanol gewaschen. Man erhielt 4,7 g des Produktes mit F von über 300"C.
D) 8-Athyl-5,8-dihydro-2-(4-methyl-1- -piperazinyl) -5.oxopyrido(2,3-d)pyrimidin-6-carbonsäure
Zu einer Lösung von Natriumhypobromit, welche hergestellt wurde durch Zugabe von 3,8 g Brom zu einem Gemisch, das 10 ml Dioxan und 16 ml einer 15%igen wässrigen Natriumhydroxidlösung enthielt, unter Kühlung unterhalb 10 C, wurden 2,0 g 6-Acetyl-8-äthyl-5,8-dihydro-2-(4-methyl -piperazinyl)-5-oxopyrido(2,3 -d)pyrimidin unter Rühren zugefügt. Nachdem das Gemisch bei Zimmertemperatur während 2 h stehen gelassen wurde, wurde das sich als Nebenprodukt gebildete Bromoform, durch Extrahieren mit Chloro- form, entfernt.
Die alkalische, wässrige Schicht wurde dann mit Essigsäure neutralisiert, um den Feststoff abzutrennen, welcher dann gesammelt und aus verdünntem Äthanol umkristallisiert wurde und das Produkt mit F von 232 - 233 C ergab.
E) X thyl -8-äthyl -2-(4-methyl -1- piperazinyl)-5,8- -dEhydro-S-oxo pyrido(2,3-d)pyrimidin-6-carboxylat
Zu einer Lösung von 3,0 g Äthyl-8-äthyl-2-(4-methyl-1- piperazinyl)-5-oxo-5,6,7,8-tetrahydropyrido(2,3-d)pyrimidin- -6-carboxylat in 100 ml trockenem Benzol wurden 2,8 g Chloranil zugesetzt, und das Gemisch wurde bei Rückfluss während 30 min erhitzt. Nach Kühlen wurde der sich abgeschiedene Feststoff gesammelt und aus Dimethylformamid umkristallisiert. Man erhielt schliesslich 2,8 g Produkt mit F von 146- 147 C.
F) 8- thyl-2-4-methyl-1- -piperazinyl)-5,8-dihydrn- 5-oxopyrido(2,3-d)pyrimidin-6-carbonitrll
2 g von 8-ÄthyS-2-(4-methyl-1-piperazinyl)-5-oxo-5,6,7,8- -tetrahydropyrido(2,3-d)pyrimidin-6-carbonitril wurden direkt bei 2200C während 7 min erhitzt, und nach Kühlen wurde das Produkt aus Methanol umkristallisiert. Man erhielt 1,7 g des obengenannten Produktes mit F von 255 258 C.
G) 8-Athyl-5,8-dihydro-5-ono-2-(I piperazinyl)- pyrido- (2,3-d)pyrimidin-6-carbonsäure
Unter Verwendung von 5,0 g von Äthyl-4-(äthyl-2-äthoxy carbonyläthyl3-amino-2-(1 -piperazinyl)-pyrimidin-5-carboxy- lat als Ausgangsmaterial wurde als Zwischenprodukt das Äthyl-5,8-dihydro-8-äthyl-5-oxo-2-(1 -piperazinyl > -pyrido(2,3- -d)pyrimidin-6-carboxylat auf wie im Beispiel 3 beschriebene Weise präpariert. Dieses Zwischenprodukt wurde gegebenenfalls durch Umkristallisieren aus Aceton gereinigt und dies zeigte einen F von 156 - 1580C. Dieses Carboxylat wurde in 30 ml einer 7%igen wässrigen Natriumhydroxidlösung durch Erhitzen bei 90 C während 20 min gelöst.
Nach Kühlen wurde die so erhaltene Lösung mit Essigsäure neutralisiert, und man erhielt einen Niederschlag, welcher gesammelt und aus Dimethylformamid umknstallisiert wurde. Man erhielt auf diese Weise 4,3 g des Produktes mit F von 253 - 255 C.
H) 8-Äthyl-5,8dihydro-5-oxo-2-(l -piperazinyl)-pyndo- (2,3-d)pyrimicSin-6-carbonsäure
Unter Verwendung von 10,0 g Äthyl-4-(äthyl-3-oxobutyl) -amino-2-(l -piperazinyl)-pyrimidin-5-carboxylat als Ausgangsmaterial wurde auf die im Beispiel 3 beschriebene Weise 6-Acetyl-8-äthyl-5,8-dihydro-5-oxo-2-(1-piperazinyl)-py- rido(2,3-d)pyrimidin präpariert. 7,3 g dieses Zwischenproduktes wurden ohne weitere Reinigung aufgearbeitet und auf die unter F) beschriebene Weise oxidiert. Man erhielt 4,2 g des Produktes mit F von 253 - 2550C.
1) if thyl-4-(dthyl-2-äthoxyearbonyläthyl)-amino-2-(4- -nethyl-i-piperazinyl)-pyrimidin-5-carboxylat
Zu einer Lösung von 3,9 g Äthyl-4-chlor-2-(4-methyl-1- -piperazinyl)-pyrimidin-5-carboxylat in 10 ml Diäthylenglykoldimethyläther wurden 2,4 g Äthyl-3-äthylaminopropionat und 1,3 g Natriumbicarbonwat zugesetzt. Das sich ergebende Gemisch wurde bei 140 - 1500C während 2 h erhitzt. Nach Entfernen des Lösungsmittels wurde der Rückstand in Chloroform aufgenommen Die Chloroformlösung wurde dann mit Wasser gewaschen, getrocknet und eingeengt und hinterliess das ölige Produkt. Das öl wurde auf Silikagel mit Chloroform als Eluat chromatographiert und ergibt das Produkt in einer reinen Form.
Die Struktur des Produktes wurde durch dessen Infrarotabsorptionsspektrum (IR) und das magnetische Kernresonanzspektrum (NMR) bestätigt.
IR-Spektrum (flüssiger Film, cm 1730, 1700, 1678, 1350 und 1230.

Gemäss dem unter I) beschriebenen Verfahren und unter Verwendung von 1,6 g von 3-Äthylaminopropionitril statt von Äthyl-3-äthylaminopropionat erhielt man das ölige Produkt.

Of the compounds of the formula II, the compounds II ″, in which R2 is a lower alkanoyl, are used to prepare the formula I by oxidation. The compounds of the formula 1 which can be prepared from the compounds prepared according to the invention can be isolated and purified by means of customary methods. The compounds I can be obtained in the free state, as a hydrate or in the form of a salt, depending on the starting materials and the reaction conditions.

The present invention is further illustrated by a few examples.
Example I Ethyl 8-ethyl-2- (4-methyl-1-piperazinyl) -S, 8-dihydro--5-oxo-pyrido (2,3-d) pyrimidine-6-carboxylate
To a solution of 4.0 g of ethyl 4- (ethyl-2-ethoxy-carbonylethyl) -amino-2- (4-methyl-1-piperazinyl) -pyrimidine-5- carboxylate in 10 ml of 1, 2 -Dimethoxyethane was added 0.75 g of a 50% sodium hydride suspension and the mixture was heated to reflux for 1 hour. After removing the solvent, the resulting residue was neutralized with acetic acid and extracted with chloroform. The extract was then washed with water, dried over anhydrous sodium sulfate, and the solvent was distilled off.
An intermediate product was obtained which, if desired, was recrystallized from n-hexane-acetone and ethyl-8-ethyl-2- (4-methyl-1-piperazinyl) -5-oxo -5,6,7,8-tetrahydropyrido (2 , 3-d) pyrimidine-6-carboxylate in pure form with F of 97-100 ° C.
This intermediate tetrahydroester thus obtained was dissolved in benzene and the mixture was heated to reflux for 1.5 hours. The solid which separated out after cooling was collected and recrystallized from dimethylformamide. 2.5 g of the above-mentioned product with an F of 146-1470C were obtained.
Example 2 8-Ethyl-2- (4-methyl4-piperazinyl) J.d-diydro = 5-oxopyrido (2,3-d) pyrimidine-6-carbonitrile
To a mixture with 4.3 g of ethyl 4- (äthyI-2-cyano-ethyl) - -amino-2- (4-methyi-1 -piperazinyl) -pvrimidine-5carboxylate and 10 ml of dioxane were 0.8 g of a 50% sodium hydride suspension was added at room temperature with stirring, and the mixture was heated to reflux for 30 min. After removing the solvent, the resulting residue was neutralized with acetic acid and extracted with chloroform.
The extract was worked up according to Example 1, and an intermediate product was obtained which, if necessary, can be recrystallized from n-hexane-acetone and 8-ethyl-2- (4-methyl-1-piperazinyl) -5-oxo-5,6 , 7,8-tetrahydropyrido (2,3-d) pyrimidine-6-carbonitrile in pure form with F of 172-174 ° C.
The obtained tetrahydro-carbonitrile was dissolved in 100 ml of dry benzene and 2.5 g of chloranil was added. The mixture was heated to reflux for 30 minutes. The solid which separated out after cooling was collected and recrystallized from methanol, and 2.9 g of the product with an F of 255-258 ° C. were obtained.
Example 3
2- (4-Acetyl-1-piperazinyl) -8-ethyl-5,8-dihydro-5-oxopyrido (2,3-dlpyrimidine-6-carbonitrile
To a mixture containing 5.0 g of ethyl 2- (4-acetyl-1-piperazinyl) 4-ethyl-2-cyanoethyl-aminopyrimidine-5-carboxylate and 50 ml of ethylene glycol dimethyl ether, 2.0 g added to a 50% sodium hydride suspension. The mixture was then heated at 950 ° C. for 1 hour. The mixture was concentrated to dryness in vacuo, and the resulting residue was neutralized with acetic acid and extracted with chloroform. The solvent was evaporated from the extract.
An oily intermediate was obtained, the 2- (4-acetyl-1-piperazinyl) -8-ethyl-5-oxo-5,6,7,8-tetrahydropyrido (2,3-d) -pyrimidine-6- carbonitrile. This nitrile was dissolved in 50 ml of benzene without further purification, and 2.5 g of chloranil was added to it. The mixture was heated to reflux for 30 min and the solid which had separated out after cooling was collected, washed with benzene and recrystallized from methanol. 3.4 g of the product with an F of 3170 ° C. were obtained in this way.
Example 4 Ethyl 2- (4-acetyl-1-piperazinyl) -8-ethyl-5,8-dihydro--5-oxopyrido (2,3-d) pyrimidine-6-carboxylate
According to the method described in Example 3 and using ethyl 2- (4-acetyl-1-piperazinyl) -4- - (ethyi-2-ethoxycarbonylethyl) aminopynmidine-5-carboxylate as starting material, the above product was obtained F from 208-2100C (dec.).
Example S 6-Acetyl-8-ethyl-5,8-dEhydro-2- (4-methyl -1 -piperazinyl) -5-oxopyrido (2,3-d) pyrimidine
Following the procedure described in Example 3 and using ethyl 2- (4-acetyl-1-piperazinyl) -4- (ethyl-3-oxobutyl) aminopyrimidine-5-carboxylate, the above product was obtained F from 198 - 200 C.
A) 8-Ethyl-2- (4-methyl-1-piperazinyl) -5,8-dihydro--5-oxopyndo (2,3-d) pyrimylin-6-carboxylic acid
A mixture containing 3.0 g of 8-ethyl-2- (4-methyl-1-piperazin-yl) -5,8-dihydro-5-oxopyrido (2,3-d) pyrimidine-6-carbonitrile, 270 ml Acetic acid and 150 ml of concentrated hydrochloric acid was heated at 90 "C for 25 hours.
The mixture was then concentrated to dryness in vacuo. The resulting residue was neutralized by adding a 5% aqueous solution of sodium hydroxide and extracted with chloroform. After drying, the extract was concentrated and left a crude solid which was recrystallized from ethanol and gave 2.6 g of the product with an F of 232-2330C.
B) 8-Ethyl-2- (4-ethyl-J-piperazinyl) -5,8-dihydro-5-oxopyrido (2.3-d) pyrimidine-6-carboxylic acid
The process according to A) was repeated using 8-ethyl-2- (4-ethyl-1-piperazinyl) -5.8 <1ihydro--5-oxopyrido (2,3 4) pvrimidine-6-carbonitnl. The above-mentioned product with an F of 228-230 ° C. was obtained.
C) 8-Ethyl-5,8-dihydro-5-oxo-2- (J -piperazinyl) -pyrido (2,3-d) pyrmdine-6-carboxylic acid hydrochloride
A mixture containing 5.0 g of 2- (4-acetyl-1 wiperazinyE) -8-ethyl- -5,8.dihydro-5-oxopyndo (2,3-d) pyrimidine-6.carbom.tnl, 200 ml Acetic acid and 200 ml of concentrated hydrochloric acid was heated at 90 ° C. for 3 hours. The mixture was then concentrated to dryness in vacuo. A small amount of concentrated hydrochloric acid was added to the residue, and a resulting hydrochloride was collected and washed with ethanol. 4.7 g of the product with an F of over 300 ° C. were obtained.
D) 8-Ethyl-5,8-dihydro-2- (4-methyl-1-piperazinyl) -5-oxopyrido (2,3-d) pyrimidine-6-carboxylic acid
To a solution of sodium hypobromite, which was prepared by adding 3.8 g of bromine to a mixture containing 10 ml of dioxane and 16 ml of a 15% strength aqueous sodium hydroxide solution, with cooling below 10 ° C., were 2.0 g of 6-acetyl -8-ethyl-5,8-dihydro-2- (4-methylpiperazinyl) -5-oxopyrido (2,3 -d) pyrimidine was added with stirring. After the mixture was left to stand at room temperature for 2 hours, the bromoform formed as a by-product was removed by extraction with chloroform.
The alkaline aqueous layer was then neutralized with acetic acid to separate the solid, which was then collected and recrystallized from dilute ethanol to give the product with an F of 232-233C.
E) Xethyl -8-ethyl -2- (4-methyl -1-piperazinyl) -5,8- -dEhydro-S-oxo pyrido (2,3-d) pyrimidine-6-carboxylate
To a solution of 3.0 g of ethyl 8-ethyl-2- (4-methyl-1-piperazinyl) -5-oxo-5,6,7,8-tetrahydropyrido (2,3-d) pyrimidine-6 carboxylate in 100 ml of dry benzene was added to 2.8 g of chloranil and the mixture was heated at reflux for 30 minutes. After cooling, the deposited solid was collected and recrystallized from dimethylformamide. 2.8 g of product with an F of 146-147 ° C. were finally obtained.
F) 8-ethyl-2-4-methyl-1-piperazinyl) -5,8-dihydric-5-oxopyrido (2,3-d) pyrimidine-6-carbonitrII
2 g of 8-EthyS-2- (4-methyl-1-piperazinyl) -5-oxo-5,6,7,8- -tetrahydropyrido (2,3-d) pyrimidine-6-carbonitrile were directly at 220.degree Heated for 7 min and, after cooling, the product was recrystallized from methanol. 1.7 g of the abovementioned product with an F of 255 258 C. were obtained.
G) 8-Ethyl-5,8-dihydro-5-ono-2- (I piperazinyl) -pyrido- (2,3-d) pyrimidine-6-carboxylic acid
Using 5.0 g of ethyl 4- (ethyl-2-ethoxy carbonylethyl3-amino-2- (1-piperazinyl) pyrimidine-5-carboxy- lat as starting material, ethyl-5,8-dihydro was obtained as an intermediate product -8-Ethyl-5-oxo-2- (1 -piperazinyl> -pyrido (2,3- -d) pyrimidine-6-carboxylate was prepared as described in Example 3. This intermediate product was purified, if necessary, by recrystallization from acetone and this showed an F of 156-1580 ° C. This carboxylate was dissolved in 30 ml of a 7% strength aqueous sodium hydroxide solution by heating at 90 ° C. for 20 minutes.
After cooling, the solution thus obtained was neutralized with acetic acid to give a precipitate, which was collected and reprecipitated from dimethylformamide. In this way, 4.3 g of the product with an F of 253-255 C.
H) 8-Ethyl-5,8dihydro-5-oxo-2- (1-piperazinyl) -pyndo- (2,3-d) pyrimicSine-6-carboxylic acid
Using 10.0 g of ethyl 4- (ethyl-3-oxobutyl) -amino-2- (l -piperazinyl) -pyrimidine-5-carboxylate as starting material was in the manner described in Example 3 6-acetyl-8- ethyl-5,8-dihydro-5-oxo-2- (1-piperazinyl) pyrido (2,3-d) pyrimidine. 7.3 g of this intermediate product were worked up without further purification and oxidized in the manner described under F). 4.2 g of the product with an F of 253-2550C were obtained.
1) if ethyl 4- (dthyl-2-ethoxy carbonylethyl) -amino-2- (4- -ethyl-i-piperazinyl) -pyrimidine-5-carboxylate
To a solution of 3.9 g of ethyl 4-chloro-2- (4-methyl-1- -piperazinyl) -pyrimidine-5-carboxylate in 10 ml of diethylene glycol dimethyl ether were 2.4 g of ethyl 3-ethylaminopropionate and 1.3 g sodium bicarbonate added. The resulting mixture was heated at 140-1500C for 2 hours. After removing the solvent, the residue was taken up in chloroform. The chloroform solution was then washed with water, dried and concentrated, and the oily product left behind. The oil was chromatographed on silica gel with chloroform as the eluate to give the product in a pure form.
The structure of the product was confirmed by its infrared absorption spectrum (IR) and nuclear magnetic resonance spectrum (NMR).
IR spectrum (liquid film, cm 1730, 1700, 1678, 1350 and 1230.
Nuclear Magnetic Resonance Spectrum (CDCl3, ppm! 8): 8.50 (1H, singlet), 4.27 (2H, quartet, J = 7 Hz), 3.90-3.83 (4H, multiplet), 2.34 (3H, singlet), 1.33 (3H, triplet, J = 7 Hz), 1.25 (3H, triplet, J = 7 Hz), and 1.20 (3H, triplet, J = 7 Hz).
K) ethyl 4- (ethyl-2-cyzoethyl) -amino-2- (4-piperazinyl) -pyrimidine-5-carboxylate
The oily product was obtained in accordance with the process described under I) and using 1.6 g of 3-ethylaminopropionitrile instead of ethyl 3-ethylaminopropionate.
IR spectrum (liquid film, cm-1): 2240, 1698, 1568 and 1578.
Nuclear Magnetic Resonance Spectrum (CDCl3, ppm / b): 8.60 (1H, singlet), 3.71 (2H, quartet, J = 7 Hz), 3.40 (2H, quartet, J = 7 Hz), 1, 35 (3H, triplet, J = 7 Hz), and 1.24 (3H, triplet, J = 7 Hz).
