Description
Gegenstand der Erfindung ist daher ein Verfahren gemäss Oberbegriff des Patentanspruchs 1.
Als Halogene kommen allgemein Fluor, Chlor oder Brom in Betracht, wobei Chlor und Brom bevorzugt sind. Von den Alkyl- bzw. Alkoxygruppen werden diejenigen mit 1 oder 2 Kohlenstoffatomen bevorzugt.
Bei den ausgeschlossenen Verbindungen handelt es sich um solche Verbindungen, die sich bei pharmakologischen Untersuchungen als unwirksam erwiesen haben.
Die neuen Verbindungen werden mit Hilfe des im kennzeichnenden Teil des Patentanspruchs 1 definierten Verfahrens hergestellt.
Als Schutzgruppe X kommen insbesondere Acylreste, die in üblicher Weise hydrolytisch abgespalten werden können, in Betracht.
Die Ausgangsverbindungen der Formel II lassen sich nach literaturbekannten, für die Synthese von Indolderivaten entwickelten, Verfahren herstellen. Als besonders geeignet sind die Methoden nach Fischer, nach Madelung sowie nach Bischler zu nennen.
Als Oxydationsmittel kommen insbesondere Kaliumpermanganat, Mangandioxyd und Chromtrioxyd in Betracht, wobei die letztgenannte Verbindung bevorzugt in Form ihres Pyridinkomplexes eingesetzt wird. Daneben sind auch andere Metall- oder Nichtmetallverbindungen höherer Oxydationsstufen verwendbar.
Die erfindungsgemäss erhältlichen Verbindungen zeichnen sich durch eine gute blutzuckersenkende Wirkung aus und können in üblicher Weise zu für die enterale, insbesondere orale, oder parenterale Applikation geeigneten pharmazeutischen Zubereitungen verarbeitet werden. Geeignete Applikationsformen sind beispielsweise Tabletten, Kapseln, Dragées, Sirupe, Lösungen, Suspensionen, Tropfen und Suppositorien. Zur Konfektionierung können die üblichen Hilfsstoffe, wie feste und flüssige Trägerstoffe, Gleitmittel sowie gegebenenfalls Geschmacks- und Süssstoffe, verwendet werden. Geeignete feste Trägerstoffe sind z. B. Stärke, Lactose, Mannit, Methyl-cellulose, Talkum, hochdisperse Kieselsäure, höhermolekulare Fettsäuren, wie Stearinsäure, Agar-Agar, Calciumphosphat, Magnesiumstearat, tierische und pflanzliche Fette sowie feste hochmolekulare Polymere, wie Polyäthylenglykole.
Die Aldehyde der Formel I können auch in Form ihrer physiologisch verträglichen Acetale für die Herstellung pharmazeutischer Zubereitungen eingesetzt werden.
Das nachfolgende Beispiel dient zur Erläuterung der Erfindung.
Beispiel
5-Methoxy-4-methylindol-2-carbaldehyd Variante I
Zu einer Lösung von 10 g 5-Methoxy-4-methyl-2-hydroxy-methylindol in 600 ml abs. Methylenchlorid gibt man 25 g Mangandioxyd (gefällt, aktiv, zur Synthese, «Merck») und rührt 10 Stunden bei Raumtemperatur. Danach und nach weiteren 10 Stunden gibt man jedesmal 5 g Mangandioxyd zu. Nach insgesamt 30 Stunden saugt man ab und engt die Lösung zur Trockne ein. Die Rohausbeute beträgt 8,8 g (= 89% d.Th.), Fp. 185 bis 187° C. Zum Entfernen einer braunen Verunreinigung chromatographiert man über eine kurze Säule mit Kieselgel 60 (Merck), als Elutionsmittel dient Heptan : Methyl-äthylketon = 4:1. Nach dem Verdampfen des Lösungsmittels kristallisiert man aus Toluol um; der Schmelzpunkt beträgt dann 187 bis 189° C.
Variante II
In einen 500-ml-Dreihalskolben mit Rührer und einem in die Flüssigkeit eintauchenden Thermometer gibt man zu 100 ml Pyridin bei 15 bis 20° C Innentemperatur allmählich 10 g Chromtrioxyd, dann wird noch eine Stunde nachgerührt. Zu dieser Suspension des gelben Chromtrioxyd-Pyridin-Komplexes fügt man eine Lösung von 8 g 5-Methoxy-4-methyl-2-hydroxy-methylindol (Fp.: 133 bis 135° C; aus Toluol) in 72 ml Pyridin, rührt noch zwei Stunden bei Raumtemperatur und lässt über Nacht stehen. Dann giesst man die Lösung auf das Gemisch von 200 ml konzentrierter Salzsäure und 500 g Eis,
saugt das dadurch ausgefällte Produkt ab, wäscht mit Wasser nach und kristallisiert (nach dem Trocknen) aus Toluol um. Die Ausbeute beträgt 3,7 g (= 47% d. Th.), der Schmelzpunkt liegt bei 187 bis 189° C.
In analoger Weise erhält man die folgenden Verbindungen:
a) aus 4-Methyl-2-hydroxymethyl-indol (Fp.: 68 bis 70° C; aus Tetrahydrofuran)
4-Methylindol-2-carbaldehyd, Fp.: 147 bis 149° C (aus Toluol);

4,5-Dimethylindol-2-carbaldehyd, Fp.: 206 bis 207° C (aus Toluol);
d) aus 5-Chlor-4-methyl-2-hydroxymethyl-indol (Fp.: 108° C; aus Toluol)
5-Chlor-4-methylindol-2-carbaldehyd, Fp.: 194 bis 196° C (aus Toluol);
e) aus 5-Äthoxy-4-methyl-2-hydroxymethyl-indol (Fp.: 152 bis 154° C; aus Toluol)
5-Äthoxy-4-methylindol-2-carbaldehyd, Fp. :199bis201°C (aus Toluol);
5 f) aus 5-Brom-2-hydroxymethyl-indol (Fp.: 112 bis 116° C; aus Toluol)
5-Bromindol-2-carbaldehyd, Fp.: 213bis215°C (aus Äthanol).
Die als Ausgangsverbindungen verwendeten 2-Hydroxy-lo methyl-indol-Derivate werden aus den entsprechenden Indol-2-carbonsäureestern durch Reduktion mit einem Überschuss an Lithiumaluminiumhydrid in abs. Äther hergestellt.
Claims (2)
- 615 4212PATENTANSPRÜCHE 1. Verfahren zur Herstellung neuer Indolderivate derFormel IworinAlkyl mit 1 bis 5 Kohlenstoffatomen oder Wasserstoff undR2 Wasserstoff, Halogen, Alkyl mit 1 bis 5 Kohlenstoffatomen oder Alkoxy mit 1 bis 5 Kohlenstoffatomen bedeuten, mit der Massgabe, dass Rx nur dann für Wasserstoff stehen kann, wenn R2 Halogen oder Äthyl bedeutet, dadurch gekennzeichnet, dass man eine Verbindung der Formel II(II)RCHm OHXworinX Wasserstoff oder eine Schutzgruppe bedeutet, durch Behandeln mit einem Oxydationsmittel oxydiert und eine gegebenenfalls vorhandene Schutzgruppe abspaltet.
- 2. Verfahren nach Patentanspruch 1, dadurch gekennzeichnet, dass man als Oxydationsmittel Kaliumpermanganat, Mangandioxyd oder Chromtrioxyd verwendet.In der Literatur ist bereits eine Anzahl verschiedenartig substituierter Indol-2-carbaldehyde beschrieben, jedoch ist bisher noch nichts über die blutzuckersenkende Wirksamkeit solcher Verbindungen bekannt.Es wurde nun gefunden, dass eine Gruppe bisher unbekannter, in 4- und/oder 5-Stellung substituierter, Indol-2-carbaldehyde eine überraschende blutzuckersenkende Wirkung aufweisen.
