PATENTANSPRÜCHE
1. Verfahren zur Herstellung von Harnstoff-Derivaten der allgemeinen Formel I

in der Rj eine Gruppe der allgemeinen Formel II
Het — (CH2)mZ(CH2)n- (II)
bedeutet, wobei Het einen 5- oder 6-gliedrigen, Stickstoff enthaltenden, gegebenenfalls durch einen niederen Alkylrest, eine Hydroxyl- oder Aminogruppe, ein Chlor- oder Bromatom substituierten heterocyclischen Ring darstellt, Z ein Schwefelatom oder eine Methylengruppe bedeutet, m den Wert 0, 1 oder 2 und n den Wert 2 oder 3 hat, mit der Massgabe, dass die Summe von m und n 3 oder 4 ist, R2 ein Wasserstoffatom, einen niederen Alkylrest oder eine Gruppe der allgemeinen Formel II darstellt, in der Het, m, n und Z die vorstehende Bedeutung haben, X ein Sauerstoffatom oder die Gruppe ^:NH und R3 ein Wasserstoffatom, einen niederen Alkyl-, Aryl- oder Arylalkylrest darstellt, und ihren Salzen mit Säuren, dadurch gekennzeichnet, dass man eine Verbindung der allgemeinen Formel III

worin A Niederalkyl bedeutet, mit einer Verbindung der Formel R3-X-NH2 oder R3R4N-NH2, worin R4 eine Schutzgruppe bedeutet, umsetzt und anschliessend eine allfällig vorhandene Schutzgruppe R4 aus dem Endprodukt entfernt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass X Sauerstoff und R3 Wasserstoff bedeuten.
3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass R2 Niederalkyl bedeutet.
4. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass R2 und Rt gleich sind.
5. Verfahren nach einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass R4 Het-CH2Z(CH2)2 bedeutet.
6. Verfahren nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass Z Schwefel bedeutet.
7. Verfahren nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass Het den Imidazol-, Thiazol-, Isothiazol-oder Pyridinring bedeutet, welche mit Methyl, Hydroxyl, Chlor oder Brom substituiert sein können.
8. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man N-Hydroxy-N'-methyl-N"-{2-[(4-methyl-5-imid-azolyl)methylthioJ äthyl}guanidin, herstellt.
9. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man N-Hydroxy-N',N"-bis{2-[(4-methyl-5-imidazolyl)-methylthio] äthyl}guanidin herstellt.
10. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man N-Methoxy-N'-methyI-N"-{2-[(4-methyl-5-imid-azolyl)methylthio] äthyl} guanidin herstellt.
11. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man N-Methoxy-N',N"-bis{2-[(4-methyl-5-imidazolyl)-methylthio]äthyl}guanidin herstellt.
12. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man N-Amino-N'-methyl-N"-{2-[(4-methyl-5-imid-azolyl)methylthioJ äthyl}guanidin herstellt.
13. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man N-Amino-N',N"-bis{2-[(4-methyl-5-imidazolyl)-methylthio]äthylj-guanidin herstellt.
14. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man N-Hydroxy-N'-methyl-N"-{2-[(2-thiazolyl)methyl-thio] äthyl jguanidin herstellt.
15. Verfahren nach einem der Ansprüche 1 bis 14, dadurch gekennzeichnet, dass man von einem Säureadditionssalz der Verbindung der Formel III ausgeht.
16. Anwendung des Verfahrens nach Anspruch 1 auf Ausgangsverbindungen der Formel III, wie sie erhalten werden,
indem man einen Thioharnstoff der Formel V

mit einer Verbindung der Formel AY, worin A Niederalkyl und Y Halogen bedeuten, oder mit einer Verbindung A-OH in Gegenwart einer Säure HY umsetzt.
Aus den CH-Patentschriften 529 751 und 576 478 sind Harnstoffderivate bekannt, die therapeutische Wirksamkeit besitzen. Die Aufgabe der vorliegenden Erfindung besteht darin, andere Harnstoffderivate, die heterocyclisch substituiert sind, zur Verfügung zu stellen, die ebenfalls therapeutische Wirksamkeit besitzen und insbesondere als Histamin-H2-Re-zeptor-Antagonisten brauchbar sind.
Diese Aufgabe wird durch das im Anspruch 1 definierte Verfahren zum Herstellen von Verbindungen der Formel I gelöst.
Der Ausdruck «niederer Alkylrest» bedeutet insbesondere Alkylreste mit 1 bis 4 Kohlenstoffatomen.
Spezielle Beispiele für den Rest Het sind der Imidazol-, Pyridin-, Thiazol-, Isothiazol-, Oxazol-, Isoxazol-, Triazol-und Thiadiazolring.
Die allgemeine Formel I steht beispielhaft für andere tautomere Formen dieser Verbindungen.
In bevorzugten Verbindungen der allgemeinen Formel I bedeutet X ein Sauerstoffatom, R3 ein Wasserstoffatom und R2 einen niederen Alkylrest. Bevorzugt sind ferner Verbindungen, in denen der Rest R2 die gleiche Bedeutung hat wie der Rest R,. In diesen beiden bevorzugten Verbindungsgruppen hat m vorzugsweise den Wert 1 und n den Wert 2. In weiteren bevorzugten Verbindungen der allgemeinen Formel I bedeutet Z ein Schwefelatom. Daher stellen Verbindungen der allgemeinen Formel I, in der Rt und/oder R2 die Gruppe Het-CH2S-(CH2)2- bedeutet, eine besonders wichtige Gruppe von Verbindungen der Formel I dar. Bevorzugte Beispiele für den Rest Het sind der gegebenenfalls durch ein Chlor- oder Bromatom, eine Methyl- oder Hydroxylgruppe substituierte Imidazol-, Thiazol-, Isothiazol- oder Pyridinring.
Schliesslich sind Verbindungen der allgemeinen Formel I bevorzugt, in der R3 ein Wasserstoffatom bedeutet.
Spezielle Beispiele für erfindungsgemäss hergestellte Verbindungen sind
N-Hydroxy-N'-methyl-N"-{2-[(4-methyl-5-imidazolyl)-me-thythio] -äthyl }-guanidin, N-Hydroxy-N',N"-bis- {2- [(4-me-thyl-5-imidazoIyl)-methylthio]-äthyl}-guanidin, N-Methoxy-N'-methyl-N"-{2-[(4-methyl-5-imidazolyl)-me-thylthio]-äthyl}-guanidin, N-Methoxy-N',N"-bis-{2-[(4-me-thyl-5-imidazolyl)-methylthio]-äthyl}-guanidin, N-Amino-N'--methyl-N"-{2-[(4-methyl-5-imidazolyl)-methylthio]-äthyl}-guanidin, N-Amino-N',N"-bis-{2-r(4-methyl-5-imidazolyl)--methylthio]-äthyl}-guanidin und N-Hydroxy-N'-methyl-N"--{2-[(2-thiazolyl)-methylthio]-äthyl}-guanidin.

Die Umsetzung erfolgt vorzugsweise in Gegenwart einer Base, wie Kaliumbicarbonat. Nach dem Abspalten der Schutzgruppe, beispielsweise mit Salzsäure, werden die Verbindungen der allgemeinen Formel I erhalten.
Die Thioharnstoffverbindungen der allgemeinen Formel III, in der R2 ein Wasserstoffatom oder einen niederen Alkylrest bedeutet, sind aus der GB-PS 1 338 169 bekannt. Die Verbindungen der allgemeinen Formel III, in der R2 eine Gruppe der allgemeinen Formel II darstellt, sowie Verfahren zu ihrer Herstellung sind aus der DT-OS 2 433 625 bekannt.
Beispielsweise wird durch Umsetzung eines Amins der allgemeinen Formel

liefert die Verbindungen der allgemeinen Formel III.
Wenn R4 und R2 die gleiche Bedeutung haben, können die Verbindungen der allgemeinen Formel III ohne Isolierung des Zwischenproduktes der allgemeinen Formel X durch Umsetzung von Schwefelkohlenstoff mit einem Überschuss von mindestens 2 Mol des Amins der allgemeinen Formel R1-NH2, vorzugsweise in einem Lösungsmittel, wie Äthanol, hergestellt werden.
Die Salze der Verbindungen der allgemeinen Formel I leiten sich ab von anorganischen oder organischen Säuren, beispielsweise von Salzsäure, Bromwasserstoffsäure, Jodwasserstoffsäure, Schwefelsäure oder Maleinsäure.
Viele physiologisch wirksame Verbindungen entfalten ihre biologische Aktivität im Organismus durch Wechselwirkung mit bestimmten Rezeptoren. Ein Beispiel hierfür ist Histamin, das unterschiedliche Organreaktionen bewirkt. Diejenigen biologischen Wirkungen des Histamins, die durch sogenannte «Antihistamine», wie Mepyramin, Diphenhydramin und Chlorpheniramin, blockiert werden, werden durch Histamin-Ht-Rezeptoren übertragen; vgl. Ash und Schild, Brit. J. Phar-mac. Chemother., Bd. 27 (1966), S. 427. Gewisse Histamin-wirkungen werden durch Antihistamine jedoch nicht gehemmt. Diese Wirkungen werden durch Rezeptoren übertragen, die von Black et al. als Histamin-H2-Rezeptoren bezeichnet wurden. Sie können durch eine von Black et al., Nature, Bd. 236 (1972), S. 385, beschriebene, Burimamid bezeichnete Verbindung blockiert werden. Unter Histamin-H2-Rezeptoren sind somit diejenigen Histamin-Rezeptoren zu verstehen, die nicht durch z.B. Mepyramin, jedoch durch Burimamid blockiert werden. Histamin-H2-Rezeptorenblok-ker werden als Histamin-H2-Antagonisten bezeichnet.
Die Blockade der H2-Rezeptoren ist in solchen Fällen nützlich, in denen die Histaminwirkungen durch Antihistamine nicht gehemmt werden. Histamin-H2-Antagonisten sind beispielsweise brauchbar zur Hemmung der Magensäuresekretion und als Antiphlogistica. Ferner wirken sie auf das kariovaskuläre System, beispielsweise indem sie die Wirkungen von Histamin auf den Blutdruck hemmen. In bestimmten Fällen, beispielsweise bei Entzündungen oder zur Hemmung der Wirkungen von Histamin auf den Blutdruck, sind auch Kombinationen von Histamin-Hj- und -H2-Antagonisten brauchbar.
Die Verbindungen der Formel I sind Histamin-H2-Rezep-torenblocker, d.h. sie hemmen solche Histaminwirkungen im Körper, die durch Antihistamine, wie Mepyramin, nicht, jedoch von z.B. Buriamid gehemmt werden. Beispielsweise hemmen die Verbindungen der Formel I die durch Histamin stimulierte Sekretion der Magensäure im perfundierten Magen von mit Urethan anästhetisierten Ratten bei intravenöser Verabreichung von 0,5 bis 256 |xMol/kg. Auf diese Methode wird von Ash und Schild, a.a.O., Bezug genommen. Die antagonistische Wirkung dieser Verbindungen lässt sich ferner durch ihre Fähigkeit nachweisen, auch andere Histaminwirkungen zu hemmen, die nach Ash und Schild nicht von Histamin-Hi-Rezeptoren übertragen werden, beispielsweise durch Versuche am isolierten rechten Vorhof von Meer5

schweinchen sowie am isolierten Rattenuterus. Ferner hemmen die Verbindungen auch die durch Pentagastrin oder Nahrung stimulierte Basalsekretion von Magensäure.
Die antiphlogistische Wirkung der Verbindungen wird anhand üblicher Versuche, beispielsweise nach dem Ratten-pfotentest, nachgewiesen. Hierbei wird durch ein Reizmittel ein Ödem erzeugt. Durch subkutane Injektion einer Dosis der Verbindungen wird das Pfotenvolumen vermindert. Die Hemmung der vasodilatierenden Wirkung von Histamin durch die Verbindungen der Formel I wird ebenfalls nach bekannten Methoden, beispielsweise durch Bestimmung des Blutdrucks von anästhetisierten Katzen, nachgewiesen.
Eine wirksame Dosis der Verbindungen der Formel I (etwa 1 bis 10 (iMol/kg) bewirkt eine 50prozentige Hemmung der Magensäuresekretion in anästhetisierten Ratten und eine 50prozentige Hemmung von durch Histamin induzierter Tachycardie am isolierten Vorhof von Meerschweinchen.
Die Arzneistoffe können in üblichen Darreichungsformen oral zusammen mit Trägerstoffen und/oder Verdünnungsmitteln und/oder Hilfsstoffen oder parenteral in einer einzigen Dosis oder in mehreren Dosen verteilt, verabreicht werden.
Die Beispiele erläutern die Erfindung.
Beispiel 1
N-Hydroxy-N'-methyl-N"-{2-[(4-methyl-5-imidazolyl)--methylthio]-äthylj-guanidin-dihydrochlorid
(a) In eine Lösung von Ti,2 g N-Methyl-N'-{2-[(4-me-thyl-5-imidazolyl)-methylthio]-äthyl]-thioharnstoff in 600 ml Methanol wird wasserfreies Chlorwasserstoffgas geleitet, und das Gemisch wird 5 Stunden unter Rückfluss erhitzt. Das Gemisch wird eingeengt, der verbliebene Rückstand mit Iso-propanol aufgenommen und das entstandene Gemisch erneut eingedampft. Es wird ein kristalliner Feststoff erhalten, der aus einem Gemisch von Isopropanol und Diäthyläther umkristallisiert wird. Ausbeute 96,2 g N,S-DimethyI-N'-{2-[(4--methyl-5-imidazolyl)-methylthio]-äthyl}-isothioharnstoff-di-hydrochlorid, das nach Umkristallisation aus einem Gemisch von Isopropanol und Diäthyläther bei 191 bis 192°C schmilzt.
(b) Ein Gemisch von 3,3 g der in (a) erhaltenen Verbindung, 2,1 g Hydroxylamin-hydrochlorid, 10 g Kaliumbicar-bonat und 50 ml wasserfreiem Dimethylformamid wird
4 Stunden bei 85°C kräftig gerührt. Nach dem Abkühlen werden die anorganischen Verbindungen abfiltriert, das Filtrat wird eingeengt und in einem Gemisch von 40 ml 1 n Salzsäure und 10 ml Äthanol gelöst. Nach dem Eindampfen und Digerieren des restlichen Öls mit Isopropanol wird ein Feststoff erhalten, der aus wässrigem Isopropanol umkristallisiert wird. Ausbeute 0,85 g der Titel Verbindung vom F. 218 bis 219°C.
Beispiel 2
N-Hydroxy-N,N"-bis-{2-[(4-methyl-5-imidazolyl)-methyl-thio]-äthyl}-guanidin-trihydrochlorid
(a) Eine Lösung von 34,0 g 4-Methyl-5-[(2-aminoäthyl)--thiomethyl]-imidazol und 7,6 g Schwefelkohlenstoff in 250 ml Äthanol wird 6 Stunden unter Rückfluss erhitzt.
Nach dem Eindampfen wird der Rückstand an einer mit Kieselgel gefüllten Säule mit einem Gemisch von Isopropanol und Äthylacetat als Laufmittel und sodann mit einem Gemisch von Isopropanol und Äthanol chromatographisch gereinigt. Ausbeute 18 g N,N'-Bis-{2-[(4-methyl-5-imidazo-lyl)-methylthio]-äthyl}-thioharnstoff vom F. 133 bis 135°C.
(b) Gemäss Beispiel 1 werden durch Umsetzung von 7,7 g der in (a) erhaltenen Verbindung mit einer Lösung von Chlorwasserstoff in Methanol 9,0 g S-Methyl-N,N'-bis-{2-
-[(4-methyl-5-imidazolyI)-methylthio]-äthyI}-isothioharn-stoff-trihydrochlorid erhalten, das nach Umkristallisation aus Isopropanol bei 212 bis 215°C schmilzt.
(c) Ein Gemisch von 15,2 g der in (b) erhaltenen Verbindung, 7,0 g Hydroxylamin-hydrochlorid, 16,0 g Kaliumbicar-bonat und 150 ml wasserfreiem Dimethylformamid wird 3 Stunden bei 90°C kräftig gerührt. Nach dem Abkühlen wird das Gemisch filtriert. Das Filtrat wird eingedampft. Der Rückstand wird an einer mit Kieselgel gefüllten Säule mit einem Gemisch von Chloroform und einer Lösung von Ammoniak in Methanol als Laufmittel gereinigt. Nach Behandlung mit einem Überschuss von Chlorwasserstoff in Äthanol wird der Rückstand in Isopropanol gelöst. Es kristallisiert die Titelverbindung aus. Ausbeute 2,1 g vom F. 224 bis 225°C.
Beispiel 3
N-Methoxy-N'-methyl-N"-{2-[(4-methyl-5-imidazolyl)--methylt hio]-äthyl ì-guanidìn-dihydrocldorid
Ein Gemisch von 6,6 g der in Beispiel 1 (a) erhaltenen Verbindung, 5,0 g Methoxyamin-hydrochlorid, 8,0 g Ka-liumbicarbonat und 60 ml Wasser wird 24 Stunden unter Rückfluss erhitzt. Nach dem Abkühlen wird das Gemisch mit einem Überschuss von Natriumchlorid versetzt. Die Lösung wird fünfmal mit jeweils 100 ml Chloroform extrahiert. Die Chloroformextrakte werden vereinigt und eingeengt, und der Rückstand wird an einer mit Kieselgel gefüllten Säule mit einem Gemisch von Chloroform und einer Lösung von Ammoniak in Methanol gereinigt. Nach Zugabe von überschüssigem Chlorwasserstoff in Äthanol wird der Rückstand mit Acetonitril digeriert. Ausbeute 2,5 g der Titelverbindung vom F. 182 bis 185°C.
Beispiel 4
N-Methoxy-N' ,N"-bis-{2-[(4-me thyl-5-imidazolyl)-methyl-thio]-äthyl}-guanidin-trihydrochlorid
Ein Gemisch von 5,1 g der in Beispiel 2 (b) erhaltenen Verbindung, 2,5 g Methoxyamin-hydrochlorid, 5,0 g Kalium-bicarbonat und 30 ml Wasser wird 24 Stunden unter Rückfluss erhitzt. Die entstandenen Produkte werden mit 1 n Natronlauge und n-Butanol ausgeschüttelt und die Schichten werden getrennt. Die n-Butanollösung wird mit 1 n Salzsäure extrahiert. Die vereinigten Extrakte werden eingeengt und in Isopropanol gelöst. Nach dem Abfiltrieren der anorganischen Stoffe wird als Filtrat die Titelverbindung erhalten. Ausbeute 0,6 g.

Beispiel 5
N-Amino-N'-methyl-N"-{2-[(4-methyl-5-imidazolyl)--methylthiojäthylj-guanidin-di/iydrochlorid
Ein Gemisch von 8,3 g der in Beispiel 1 (a) erhaltenen Verbindung, 4,1 g Hydrazinsulfat, 12,5 g Kaliumbicarbonat und 100 ml wasserfreiem Dimethylformamid wird 3 Stunden bei 90°C gerührt. Nach dem Abkühlen werden die anorganischen Verbindungen abfiltriert. Das Filtrat wird mit einem Überschuss von Chlorwasserstoff in Äthanol versetzt und eingedampft. Der Rückstand wird mit Isopropanol digeriert.

Der erhaltene Feststoff wird aus wässrigem Isopropanol umkristallisiert. Ausbeute 1,94 g der Titelverbindung vom F. 234 bis 235°C.
Beispiel 6
N-A mino-N' ,N"-bis- {2-[ (4-methyl-5-imidazolyl)-methylthio ]--äthyij-guanidin-trihydrochlorid
Ein Gemisch von 5,1 g der in Beispiel 2 (b) erhaltenen Verbindung, 1,6 g Hydrazinsulfat, 7,0 g Kaliumbicarbonat und 60 ml wasserfreiem Dimethylformamid wird 3 Stunden bei 70°C gerührt. Nach dem Abkühlen werden die anorganischen Verbindungen abfiltriert. Das Filtrat wird mit einem Überschuss von Chlorwasserstoff in Äthanol versetzt und sodann eingedampft. Es hinterbleibt die Titelverbindung.
Beispiel 7
N-Hydroxy-N'-methyl-N"-[2-(2-thiazolylmethylthio)-äthylJ--guanidin-dihydrochlorid
(a) Eine Lösung von 4,7 g N-Methyl-N'-[2-(2-thiazolyl-methylthio)-äthylJ-thioharnstoff in 50 ml Methanol wird mit 5 ml Methyljodid versetzt und 10 Minuten unter Rückfluss erhitzt. Die entstandenen Produkte werden eingeengt und mit einem Anionenaustauscherharz IRA-401 in der Cl~-Form und sodann mit einem Überschuss von Chlorwasserstoff in Äthanol behandelt. Die Lösung wird eingeengt. Ausbeute 5,6 g N,S-Dimethyl-N'-[2-(2-thiazolyl-methylthio)-äthyl]-iso-thioharnstoff-dihydrochlorid.
(b) Ein Gemisch von 0,46 g der in (a) erhaltenen Verbindung, 0,35 g Hydroxylamin-hydrochlorid, 1,5 g Kaliumbicarbonat und 10 ml wasserfreiem Dimethylformamid wird
4 Stunden bei 90°C gerührt und sodann mit Chloroform und Wasser ausgeschüttelt. Die Schichten werden getrennt. Die vereinigten Chloroformschichten werden eingedampft, und der Rückstand wird mit einem Überschuss von Chlorwasserstoff in Äthanol behandelt. Nach dem Einengen wird der Rückstand mit Isopropanol digeriert. Es werden 0,10 g der Titelverbindung vom F. 171 bis 174°C erhalten. NMR-Spektrum: 2HG Dimethylsulfoxid:

Beispiel 8
N-Methyl-N'-methylamino-N"-{2-[(4-methyl-5-imidazolyl)--methylthio]-äthyl}-guanidin-dihydrochlorid
Ein Gemisch von 6,7 g der in Beispiel 1 (a) erhaltenen Verbindung, 5,7 g N-Methyl-N-tert.-butoxycarbonylhydrazin--sulfat, 10,0 g Kaliumbicarbonat und 80 ml wasserfreiem Dimethylformamid wird 3 Stunden bei 90°C gerührt. Nach dem Abkühlen wird das Gemisch filtriert und das Filtrat eingeengt. Der Rückstand wird mit einer Lösung von Chlorwasserstoff in Äthanol unter Rückfluss erhitzt. Nach dem Einengen wird der Rückstand digeriert. Es wird die Titelverbindung erhalten.
Beispiel 9
N-Methyl-N'-hydroxy-N"-{3-[(4-methyl-5-imidazolyl)--methylthio]-propyl}-guanidin-dihydrochlorid
Gemäss Beispiel 1 (a) wird N-Methyl-N'-{3-[(4-methyl--5-imidazoIyI)-methylthio]-propyl}-thioharnstoff mit einer
Lösung von Chlorwasserstoff in Methanol umgesetzt. Das erhaltene N,S-Dimethyl-N'-{3- [(4-methy 1-5-imidazoly l)-me-thylthio]-propyl}-isothioharnstoff-dihydrochlorid wird gemäss Beispiel 1 (b) mit Hydroxylamin umgesetzt. Es wird die Titelverbindung erhalten.
Beispiel 10
N-Methyl-N'-hydroxy-N"-[4-(4-imidazolyl)-butyl]-guanidin--dihydrochlorid
Gemäss Beispiel 1 (a) wird N-Methyl-N'-[4-(4-imidazolyl)--butylj-thioharnstoff mit einer Lösung von Chlorwasserstoff in Methanol umgesetzt. Der erhaltene N,S-Dimethyl-N'-[4--(4-imidazolyl)-butyl]-isothioharnstoff wird gemäss Beispiel 1 (b) mit Hydroxylamin versetzt. Es wird die Titelverbindung erhalten.
Beispiel 11
N-Methyl-N'-hydroxy-N"-[4-(2-thiazolyl)-butyl]-guanidin--dihydrochlorid
N-Methyl-N'-[4-(2-thiazolyl)-butyl]-thioharnstoff wird mit 66prozentiger Jodwasserstoffsäure in das entsprechende Hy-drojodid überführt, das in Methanol gelöst wird. Die Lösung wird mit Methyljodid versetzt und 2 Stunden unter Rückfluss erhitzt. Nach dem Einengen hinterbleibt ein Öl, das zu N,S--Dimethyl-N '- [4-(2-thiazolyl)-butyl] -isothioharnstoff-dihy dro-jodid kristallisiert. Die Umsetzung dieser Verbindung mit Hydroxylamin gemäss Beispiel 1 (b) liefert die Titelverbindung.
Beispiel 12
N-Methyl-N'-hydroxy-N"-{2-[( 3-chlor-2-pyridy l)-methy 1-thio]-äthyiy-guanidin-dihydrochlorid
Gemäss Beispiel 1 (a) wird N-Methyl-N'-{2-[3-chlor-2--pyridyl)-methylthio]-äthyl}-thioharnstoff in das N,S-Dime-thyl-N'- {2- [(3-chlor-2-pyridyl)-methylthio] -äthyl}-isothio-harnstoff-dihydrochlorid überführt, das nach Umsetzung mit Hydroxylamin gemäss Beispiel 1 (b) die Titelverbindung liefert.
Beispiel 13
N-Methyl-N'-phenoxy-N"-{2-[(4-methyl-5-imidazolyl)--methylthioj-äthyiy-guanidin-dihydrochlorid
Die Umsetzung der in Beispiel 1 (a) erhaltenen Verbindung mit Phenoxyamin gemäss Beispiel 1 (b) liefert die Titelverbindung.
Beispiel 14
N-Methyl-N'-benzyloxy-N"-{2-[(4-methyl-5-imidazolyl)--methylthio]-äthyl}-guanidin-dihydrochlorid

(h) N-Hydroxy-N'-methyl-N"-{2-[2-(4-methyl-5-imidazolyl)--äthylthio]-äthyl}-guanidin-dihydrochlorid.
Beispiel 16
Durch Umsetzung der in Beispiel 1 (a) erhaltenen Verbindung mit n-Propoxyamin und n-Butoxyamin gemäss Beispiel 1 (b) werden die folgenden Verbindungen erhalten: 5 N-Methyl-N'-n-propoxy-N"-{2-[(4-methyl-5-imidazolyl)-me-thylthio] -äthyl}-guanidin-dihydrochlorid und N-Methyl-N'-n-butoxy-N"-{2-[(4-methy 1-5-imidazoly l)-me-thylthio]-äthyl}-guanidin-dihydrochlorid.

Gemäss Beispiel 1 (a) werden durch Umsetzung von
(a) S-Methyl-N-{2-[(4-methyl-5-imidazolyl)-methylthio]--äthyl}-isothioharnstoff-dihydrochlorid,
(b) S-Methyl-N-n-butyl-N'-{2-[(4-methyl-5-imidazolyl)-me-15 thylthio]-äthyl}-isothioharnstoff-dihydrochlorid und
(c) S-Methyl-N-{2-[(4-methyl-5-imidazolyl)-methylthio]--äthyl}-N'-|2-[(2-thiazolyl)-methylthio]-äthyl}-isothio-harnstoff-trihydrochlorid,
die aus den entsprechenden Thioharnstoffverbindungen nach 20 dem Verfahren von Beispiel 1 (a) hergestellt wurden, mit Hydroxylamin gemäss Beispiel 1 (b) die folgenden Verbindungen erhalten.
(a) N-Hydroxy-N'-{2-[(4-methyl-5-imidazoIyl)-methylthio]--äthyl}-guanidin-dihydrochlorid, 25 (b) N-Hydroxy-N'-n-butyl-N"-{2-[(4-methyl-5-imidazolyl)--methylthio] -äthyl}-guanidin-dihydrochlorid und (c) N-Hydroxy-N'-{2-[(4-methyl-5-imidazolyl)-methylthio]--äthyl}-N"-{2-[(2-thiazolyl)-methylthio]-äthyl}-guanidin--trihydrochlorid.
