Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Hentriakontapeptiden der Sequenz

Das Hentriakontapeptid der Sequenz Ia wird im folgenden auch als ß-Endorphin bezeichnet.
Das erfindungsgemässe Verfahren zur Herstellung der Hentriakontapeptide der Sequenz I ist dadurch gekennzeichnet, dass man eine solche Verbindung mittels der Festphasenpeptidsynthese synthetisiert.
Die Isolierung von ß-Endorphin aus natürlichen Quellen kann nach an sich bekannten Verfahren erfolgen. Bevorzugte natürliche Quellen sind Schafshypophysen, insbesondere Kamelhypophysen. So liefert z.B. saure Extraktion von Kamelhypophysen mit Aceton eine Fraktion D, aus der wiederum durch Chromatographie an Carboxymethylcellulose, wie von Li et al., Biochemistry 14, 947 (1975), beschrieben, die Komponente erhalten wird. Die Reinigung der Komponente N durch Gelfiltration und Elektrophorese nach an sich bekannten Methoden liefert das gewünschte ß-Endorphin.
Identifikation der Aminosäuren und Sequenzanalyse des aus natürlichem Material isolierten ß-Endorphins kann durch enzymatischen Abbau des aus der gereinigten Komponente N erhaltenen ß-Endorphins mit Trypsin und Subtilisin in an sich bekannter Weise erfolgen. Die Abbauprodukte wurden durch Papierchromatographie und anschliessende Hochspannungselektrophorese aufgetrennt. Während der Trypsinabbau 11 Flecken lieferte, wurden beim Subtilisinabbau 10 Flecken erhalten. Die einzelnen Flecken werden eluiert und der Aminosäuregehalt wurde mittels eines automatischen Analysators bestimmt. Die Sequenzanalyse erfolgte nach der Methode des Dansyl-Edman-Abbyus. Unter Verwendung dieser Methoden wurde ß-Endorphin als ein Hentriakontapeptid mit der oben angegebenen Sequenz erkannt.
Die Synthese des ß-Endorphins kann unter Verwendung allgemein bekannter Techniken auf dem Gebiet der Festphasensynthese von Peptiden durchgeführt werden. Gemäss einer bevorzugten Ausführungsform wird aminogeschütztes Glutamin, welches die endständige Carboxylgruppe des ß-Endor-phins liefert, mit einem in der Festphasensynthese üblicherweise verwendeten Harz, wie z.B. Benzhydrylamin-Polystyrol, das mit 1-2% Divinylbenzol vernetzt ist, verwendet. Die Aminoschutzgruppe wird dann selektiv entfernt, wobei ein je nach Art der Schutzgruppe unterschiedliches Reagens verwendet wird. In einer bevorzugten Ausführungsform wird als Aminoschutzgruppe der t-Butyloxycarbonylrest (Boc) und als selektives Spaltungsreagens 50%ige Trifluoressigsäure in Methylenchlorid verwendet.
Nach Abspaltung der Schutzgruppe wird das Glutamin-Harz mit aminogeschütztem Glycin, vorzugsweise N-Boc-gly-cin, insbesondere in Form des symmetrischen Anhydrids in an sich bekannter Weise behandelt, so dass eine Peptidbin-dung zwischen der freien Aminogruppe des Glutaminrestes und der Carboxylgruppe des Glycins hergestellt wird.
Der Zyklus der Schutzgruppenabspaltung und Kupplung mit symmetrischen Anhydriden von geschützten Aminosäuren wird dann mit den restlichen Aminosäuren in der Reihenfolge der ß-Endorphin-Sequenz wiederholt. Einige dieser Aminosäuren erfordern, ausser dem Schutz der a-Amino-gruppe, die Blockierung einer reaktiven Seitengruppe. Solche Aminosäuren und deren Schutzgruppen sind die folgenden:
Glu(OBzl), Ser(Bzl), Lys(oBr-Z), Tyr(oBr-Z)
und His(Boc)
worin oBr-Z = o-Brombenzyloxycarbonyl, Bzl = Benzyl und Boc = t-Butoxycarbonyl.
Nach Beendigung der Synthese erhält man das folgende geschützte Hentriakontapeptid, das an das Styrol-Divinyl-benzoI-Copolymerisat gebunden ist:

worin oBr-Z, Bzl und Boc wie oben definiert sind und ® das Styrol-Divinylbenzol-Copolymerisat darstellt.
Die Abspaltung des Peptids vom Harz wird durch Behandlung mit flüssigem Fluorwasserstoff unter gleichzeitiger Abspaltung aller Schutzgruppen bewerkstelligt und liefert das gewünschte ß-Endorphin.
Obgleich das ß-Endorphin die Aminosäuresequenz der Reste 61-91 des Hormons ß-Lipotropin besitzt, ist seine lipo-lytische Aktivität sehr gering im Vergleich zu der des kompletten Hormons. ß-Endorphin besitzt eine signifikante Opiat-Agonistwirkung und ist daher ein wirksames, nicht-sucht-machendes Analgetikum. Synthetisches und natürliches ß-Endorphin haben gleiche physikalische und biologische Eigenschaften.
Die Analoga des ß-Endorphins, die durch die Sequenz I definiert sind, entsprechen Teilstücken des ß-Lipotropins des Schweins und des Menschen. Sie werden zweckmässigerweise in Analogie zu der vorstehend beschriebenen ß-Endorphin-synthese hergestellt.
Die erfindungsgemässen Verbindungen können als pharmazeutische Präparate mit direkter oder verzögerter Freigabe des Wirkstoffs in Mischung mit einem für die enterale, perkutane oder parenterale Applikation geeigneten organischen oder anorganischen inerten Trägermaterial, wie z.B. Wasser, Gelatine, Gummi arabicum, Milchzucker, Stärke, Magne-siumstearat, Talk, pflanzlichen Ölen, Polyalkylenglykolen, Vaseline, usw. verwendet werden. Die pharmazeutischen Präparate können in fester Form, z.B. als Tabletten, Dragées, Suppositorien, Kapseln; in halbfester Form, z.B. als Salben; oder in flüssiger Form, z.B. als Lösungen, Suspensionen oder Emulsionen, vorliegen. Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten weitere Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Mittel zur geschmacklichen Verbesserung, Salze zur Veränderung des osmatischen Druckes oder Puffersubstanzen. Die Herstellung der pharmazeutischen Präparate kann in der jedem Fachmann geläufigen Weise erfolgen.
Bei der Anwendung pharmazeutischer Präparate mit systemischer Wirkung kann als Dosisrichtlinie etwa 5 bis 750 mg/ kg gelten.
Beispiel 1 Festphasensynthese von ß-Endorphin
Die Verknüpfung von a-Benzyl-N«-t-butyloxycarbonyl-glutamat mit dem Benzhydrylaminharz wurde mittels seines symmetrischen Anhydrids bewerkstelligt. 2,5 g Benzhydryl-aminhydrochloridharz, enthaltend 4,38 mMol freies Amin pro g, wurden mit 5 % Diisopropyläthylamin (DIEA) in CH2C12 neutralisiert und dann mit 3 Äquivalenten des symmetrischen Anhydrids in CH2C12 während 45 Minuten behandelt. Es wurden weitere 5 ml 5 % DIEA in CH2C12 zugegeben. Nach 10 Minuten wurde filtriert, dreimal mit 30 ml CH2C12 und dreimal mit 30 ml absolutem Äthanol gewaschen. Nach nochmaliger Behandlung des Harzes mit der gleichen Menge symmetrischen Anhydrids wurde es eine Stunde über P205 getrocknet. Vollständigkeit der Umsetzung wurde mit dem Gisintest geprüft. Nach Hydrolyse einer Probe in Propionsäure/12 N HCl, lieferte die Aminosäureanalyse einen Peak, der einer Menge von 0,23 mMol Gin pro g (68% Spaltung) entsprach. Die Identifizierung des mit Fluorwasserstoff gespaltenen Produktes als Glutamin erfolgte dünn-schichtchromatographisch in 1-Butanol/Essigsäure/Wasser (4:1:1, v/v) und in 1-Butanol/Pyridin/Essigsäure/Wasser (6:6:1,2:4,6, v/v).
Die Umsetzung der Boc-Aminosäuren mit N,N'-Dicyclo-hexylcarbodiimid (DCCI) wurde folgendermassen durchgeführt: 1,9 mMol der Boc-Aminosäure in 6 ml CH2C12 wurden auf 0°C abgekühlt und mit 1,6 ml 0,6 M DCCI in CH2C12 vermischt. Nach 20minütigem Rühren bei 0°C wurde der Niederschlag von Dicyclohexylharnstoff abfiltriert und mit 2,4 ml CH2C12 gewaschen. Das Filtrat wurde sofort für die Kupplungsreaktion verwendet.
Das vorstehend beschriebene Harz wurde dem folgenden Syntheseschema unterworfen:
1. dreimaliges Waschen mit je 15 ml CH2C12;
2. Entfernung der Boc-Gruppe mit 50% Trifluoressig-säure in CH2C12 während 15 Minuten;
3. Waschen zweimal mit je 15 ml CH2C12;
4. Waschen zweimal mit je 15 ml 50% Dioxan in CH2C12;
5. Waschen zweimal mit je 15 ml CH2C12;
6. Neutralisation mit 15 ml 5% DIEA in CH2C12;
7. Waschen sechsmal mit je 15 ml CH2C12;
8. Zusatz der Lösung des symmetrischen Anhydrids der Boc-Aminosäure und Schütteln während 30 Minuten;
9. Zusatz von 0,20 Äquivalenten 5 % DIEA in CH2C12 und Schütteln während 20 Minuten;
10. Waschen dreimal mit je 15 ml CH2C12 und
11. Waschen dreimal mit je 15 ml absolutem Äthanol.
Dieser Zyklus wurde jeweils mit den folgenden N-ge-schützten Aminosäuren wiederholt:
Boc-Gly, Boc-Lys(oBr-Z), Boc-Lys(oBr-Z), Boc-His(Boc), Boc-Ala, Boc-Asn, Boc-Lys(oBr-Z), Boc-Ile, Boc-Ue, Boc-Ala, Boc-Asn*, Boc-Lys(oBr-Z), Boc-Phe, Boc-Leu, Boc-Thr, Boc-Val, Boc-Leu, Boc-Pro, Boc-Thr, Boc-Gln, Boc-Ser(Bzl), Boc-Lys(oBr-Z), Boc-Glu(OBzl), Boc-Ser(Bzl), Boc-Thr, Boc-Met, Boc-Phe ,Boc-Gly, Boc Glv und Boc-Tyr(oBr-Z).
* Nach der Einführung von Asn, wurde das Peptid getrocknet (Ausbeute 4,7 g) und ein aliquoter Teil (400 mg) wurde entfernt. Das restliche Peptidharz wurde dann nach dem gleichen, oben beschriebenen Schema zur Einführung weiterer Aminosäuren behandelt, mit zwei Ausnahmen: Zur Neutralisation wurde 2mal mit 5 % DIEA in CH2C12 behandelt und in der zweiten Stufe der Anhydridkupplung wurde Trifluoräthanol bis zur einer Konzentration von 20% zugesetzt.
Von 0,6 g geschütztem und an das Harz gebundenem ß-Endorphin wurde die Boc-Schutzgruppe abgespalten. Es wurde neutralisiert und das getrocknete Harz in Gegenwart von 1,8 ml Anisol und 15 ml flüssigem Fluorwasserstoff bei 0°C eine Stunde gerührt. Fluorwasserstoff wurde durch einen Stickstoffstrom entfernt und der ölige Rückstand wurde 2mal mit 15 ml Äthylacetat gewaschen. Das Peptid wurde aus dem Harz 3mal mit 15 ml 50%iger Essigsäure extrahiert, die vereinigten Filtrate wurden unter vermindertem Druck auf ein geringes Volumen eingeengt und der Gelfiltration an Sepha-dex G-10 in 0,5 N Essigsäure unterworfen. Es wurde 1 Peak festgestellt. Lypophilisation lieferte 110 mg eines Materials, das dann an Carboxymethylcellulose chromatographiert wurde. Isolierung des Haupt-Peaks lieferte 66 mg Material, das weiter gereinigt wurde durch Verteilungschromatographie an Sephadex G-50. Der Hauptpeak wiederum lieferte 52 mg hochreines ß-Endorphin (19,6% Ausbeute bezogen auf das Ausgangsharz).
Dünnschichtchromatographie des gereinigten Materials in n-Butanol/Pyridin/Eisessig/Wasser (15:10:3:12, v/v) lieferte einen Fleck mit einem Rt Wert von 0,35. Papierelektrophorese des synthetischen ß-Endorphins lieferte einen einzigen Fleck bei pH 3,7 und 6,9 mit den Rr Werten (bezogen auf Lys) von 0,65 bzw. 0,42. Gelelektrophorese des gereinig-

ten Materials lieferte ebenfalls eine einzige Bande bei pH 4,5 während einer Stunde. Eine Probe von 1 mg wurde der Elek-trofokussierung in 5 % Polyacrylamidgel unterworfen, wobei eine Bande resultierte mit einem PI von ungefähr 10,0. Die Aminosäureanalyse nach saurer Hydrolyse lieferte die folgenden Werte:

Die Aminosäureanalyse nach vollständigem enzymatischem Abbau (zunächst mit Trypsin und Chymotrypsin, dann mit Leucinaminopeptidase) lieferte die folgenden Werte:

Biologische Aktivität von ß-Endorphin Die Opiat-Agonist-Aktivität von natürlichem ß-Endorphin wurde nach der Methode von Simon et al., Proc. Nat. Acad. Sei. 70, 1947 (1973), bestimmt unter Verwendung von Membranen aus Meerschweinchengehirnen. Die Ergebnisse sind in der folgenden Tabelle 1 zusammengefasst:

Aus den Werten dieser Tabelle geht hervor, dass das ß-Endorphin eine starke Opiat-Agonist-Aktivität aufweist und dass es, auf molarer Basis, eine 3,42mal stärkere Wirkung als Normorphin besitzt.
Die Iypolytische Aktivität von natürlichem ß-Endorphin wurde mit derjenigen des ß-Lipotropins von Schafen an Kaninchenfettzellen unter Verwendung der Methode von Li et al., Arch. Biochem. Biophys. 169, 669 (1975), verglichen. Die erhaltenen Resultate sind in der folgenden Tabelle 2 zusammengefasst:

Beispiel 2
Nach der in Beispiel 1 beschriebenen Methode wurde ein Analoges vom ß-Endorphin, das in seiner Sequenz einem Teil des Schweine-ß-Lipotropins entspricht, hergestellt, wobei in Position in 23 anstelle von Isoleucin Valin eingebaut wurde. Die Sequenz des erhaltenen Hentriakontapeptids lautet:

Beispiel 3
Nach der in Beispiel 1 beschriebenen Methode wurde ein Analoges des ß-Endorphins mit der Aminosäuresequenz
H-Tyr-Gly-Gly-Phe-Met-Thr-Ser-Glu-Lys-Ser-Gln-Thr-Pro-Leu-Val-Thr-Leu-Phe-Lys-Asn-Ala-Ile-Ile-Lys-Asn-Ala-Tyr-Lys-Lys-GIy-Glu-OH
hergestellt, wobei in Position 27 anstelle von Boc-His(Boc) Boc-Tyr(oBr-Z) eingesetzt und als C-terminale Aminosäure Glutamin in Form von Boc-GIu(Bzl) verwendet wurde. Dieses Analoge entspricht in seiner Sequenz einem Teil des menschlichen ß-Lipotropins.
Beispiel 4 Parenterale Formulierung
Eine stabilisierte Lösung von ß-Endorphin in destilliertem Wasser wurde in sterile Ampullen abgefüllt, so dass jede Ampulle 50 mg Peptid enthielt. Die Lösungen wurden dann lyophilisiert, so dass ein trockenes Pulver erhalten wurde, aus dem jederzeit durch Zusatz von isotonischer Salzlösung eine Injektionslösung rekonstituiert werden kann.

mit flüssigem Fluorwasserstoff behandelt, wodurch nicht nur das Peptid vom festen Träger abgetrennt sondern auch gleichzeitig alle Peptidschutzgruppen entfernt werden.
4. Verfahren gemäss Anspruch 1, dadurch gekennzeichnet, dass man ein Hentriakontapeptid der Sequenz
H-Tyr-GIy-Gly-Phe-Met-Thr-Ser-Glu-Lys-Ser-GIn-Thr-Pro-Leu-Val-Thr-Leu-Phe-Lys-Asn-AIa-Ile-Ile-Lys-Asn-Ala-Tyr-Lys-Lys-Gly-Glu-OH
herstellt.
