Description
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung neuer substituierter Arylalkylamine der Formel I

R2 ein Wasserstoffatom, ein Chloratom oder die Methoxy-gruppe;
R3 ein Wasserstoffatom oder die Methoxygruppe oder zusammen mit R2 die Methylendioxy- oder Äthylendioxygruppe;
Rt und Rs die gleich oder verschieden sein können, Wasserstoffatome oder niedere Alkylgruppen;
R6 ein Wasserstoffatom oder eine niedere Alkoxygruppe;
R7 eine niedere Alkoxygruppe oder zusammen mit R6 die Methylendioxy- oder Äthylendioxygruppe;
X die Carbonyl- oder Sulfonylgruppe; und n die Zahl 2 oder 3 bedeuten, sowie von deren physiologisch verträglichen Säureadditionssalzen mit anorganischen oder organischen Säuren.
Für die bei der Definition der Reste Rx, R4 und R5 erwähnten Alkylreste kommen insbesondere die Bedeutung der Methyl-, Äthyl-, Propyl- oder Isopropylgruppe und für die bei der Definition der Reste R6 und R7 erwähnten Alkoxyreste insbesondere die der Methoxy-, Äthoxy-, Propoxy- oder Iso-propoxygruppe in Betracht.
Die Verbindungen der Formel I und deren Säureadditionssalze besitzen wertvolle pharmakologische Eigenschaften, insbesondere eine herzfrequenzsenkende Wirkung.
Das erfindungsgemässe Verfahren zur ihrer Herstellung ist dadurch gekennzeichnet, dass man ein 1H-Phthalimidin der Formel II

in der Ra, R2, R3 und X wie eingangs definiert sind, mit einem Alkylamin der Formel III

in der R4, Rs, R6, R7 und n wie eingangs definiert sind und Z eine nukleophil austauschbare Gruppe, wie ein Chlor-, Bromoder Jodatom, eine Alkylsulfonyl- oder Arylsulfonyloxy-gruppe darstellt, umsetzt und gewünschtenfalls eine erhaltene Verbindung der Formel I, in der R4 ein Wasserstoffatom darstellt, niederalkyliert und/oder eine erhaltene Verbindung der Formel I in ein physiologisch verträgliches Säureadditionssalz mit einer anorganischen oder organischen Säure überführt.
Die Umsetzung wird gegebenenfalls in einem Lösungsmittel, z.B. in Aceton, Dimethylformamid, Dimethylsulfoxid oder Methanol, und zweckmässigerweise je nach der Reaktionsfähigkeit des Restes Z bei Temperaturen zwischen 0 und 150 °C durchgeführt. Vorteilhaft ist die Gegenwart eines säurebindenden Mittels, wie z.B. eines Alkoholats, eines Alkalihydroxyds, eines Alkaliamids, Natriumhydrids oder einer tertiären organischen Base, wie Triäthylamin oder Pyridin und/oder eines Reaktionsbeschleunigers, wie beispielsweise Kaliumjodid.
Erhält man eine Verbindung der Formel I, in der R4 ein Wasserstoffatom darstellt, so kann diese mittels Alkylierung, z.B. durch Umsetzung mit einem entsprechenden Alkylhalo-genid oder Dialkylsulfat, niederalkyliert oder durch Umsetzung mit Formaldehyd/Ameisensäure methyliert werden.
Die erhaltenen Verbindungen der Formel I können mit anorganischen oder organischen Säuren in ihre physiologisch verträglichen Salze übergeführt werden. Als Säuren haben sich beispielsweise Salzsäure, Phosphorsäure, Bromwasserstoffsäure, Schwefelsäure, Milchsäure, Weinsäure oder Maleinsäure als geeignet erwiesen.
Die als Ausgangsstoffe verwendeten Verbindungen der Formeln II und III lassen sich nach an sich bekannten Verfahren herstellen. So erhält man beispielsweise ein 1H-Phthalimi-din der Formel II durch Reduktion eines entsprechenden Phthalimids mit Zinkstaub in Eisessig und gegebenenfalls anschliessender Alkylierung.
Wie bereits eingangs erwähnt, weisen die neuen Verbindungen der Formel I und deren Säureadditionssalze wertvolle pharmakologische Eigenschaften auf. Sie besitzen neben einer milden blutdrucksenkenden Wirkung insbesondere eine selektive herzfrequenzsenkende Wirkung.
Beispielsweise wurden folgende Verbindungen auf ihre biologischen Eigenschaften untersucht: A = 5,6-Dimethoxy-2N-(3-[3,4-dimethoxy)-phenyl-äthyl-methylamino]-propyl)-phthalimidin-hydro-chlorid;

1. Wirkung auf die Herzfrequenz an narkotisierten Meerschweinchen
An Meerschweinchen in Urethan-Narkose wurde die Herzfrequenz per Elektrokardiogramm registriert. Die zu untersuchenden Substanzen wurden in steigenden Dosen zwischen 0,5 und 20 mg/kg i. v. gegeben.
Die nachfolgende Tabelle enthält die Veränderungen der Herzfrequenz:

2. Wirkung auf die Herzfrequenz an Meerschweinchen-Vorhöfen Isolierte spontan schlagende Vorhöfe von Meerschweinchen beider Geschlechter mit einem Körpergewicht von 300 bis 400 g wurden in einem Organbad in Tyrode-Lösung untersucht. Die Nährlösung wurde mit Carbogen (95% 02 + 5% C02) versorgt und konstant bei 30 °C gehalten. Die Kontraktionen wurden isometrisch mittels eines Dehnungsmessstreifens auf einem Grass-Polygraphen registriert. Die zu untersuchenden Substanzen wurden den Organbädern zugesetzt, so dass Endverdünnungen von 10"s g/ml entstanden. 5 Vorhöfe pro Substanz.
In der folgenden Tabelle wird die prozentuale Verminderung der Herzfrequenz im Mittel von 5 Vorhöfen bei einer Substanzkonzentration von 10~5 g/ml angegeben.

3. Akute Toxizität Die akute Toxizität der zu untersuchenden Substanz wurde an Mäusen (Beobachtungszeit: 14 Tage) nach oraler bzw. intravenöser Gabe bestimmt. Es wurde die LDS0 aus dem Prozentsatz der Tiere, die nach verschiedenen Dosen innerhalb der Beobachtungszeit verstarben, berechnet (siehe J. Pharma-col. exp. Therap. 96 [1949] 99).:

Zur pharmazeutischen Anwendung lassen sich die Verbindungen der Formel I sowie deren physiologisch verträgliche Säureadditionssalze gegebenenfalls in Kombination mit anderen Wirksubstanzen in die üblichen galenischen Zubereitungsformen, wie Tabletten, Dragees, Pulver, Suspensionen, Lösungen oder Suppositorien, einarbeiten. Die Einzeldosis beträgt hierbei 20 bis 300 mg, vorzugsweise 25 bis 200 mg.
Die nachfolgenden Beispiele sollen die Erfindung näher erläutern.
Beispiel 1
5,6-Dimethoxy-2N-(3-[2-(3,4-dimethoxy)-phenyläthyl-
methylamino]-propyl-phthalimidin-hydrochlorid In eine Lösung von 3,0 g (15 mMol) 5,6-Dimetho-xy-phthalimidin in 100 ml Dimethylformamid trägt und man 0,5 g Natriumhydrid ein und erwärmt anschliessend für 30 Minuten auf 80 °C. Danach tropft man 8,5 g (30 mMol) l-[2-(3,4-Dimethoxy-phenyl)-äthyl-methylamino]-3-chlor-propan gelöst in 100 ml Dimethylformamid zu und erhitzt 7 Stunden auf 140 °C. Nach dem Erkalten wird mit Wasser versetzt und mehrmals mit Chloroform ausgeschüttelt. Die organischen Phasen werden getrocknet und im Vakuum zur Trockne eingeengt. Das Rohprodukt wird säulenchromatographisch über
Kieselgel gereinigt. Durch Fällung mit ätherischer Salzsäure erhält man das Hydrochlorid.
Ausbeute: 4,2 g (60 % der Theorie),
Schmelzpunkt: 170-172 DC.
Beispiel 2
5,6-Dimethoxy-2N-(3-[2-(3,4-dimethoxy)-phenyläthyl-
methylamino]-propyI)-phthalimidin-hydrochlorid 5 g (12,1 Mol) 5,6-Dimethoxy-2N-(3-[2-(3,4-dimethoxy)-phenyläthyI-
amino]propyl-phthalimidin-hydrochlorid werden in einer Mischung von 1,38 g (30 mMol) Ameisensäure und 1,5 g (20 mMol) Formalin eine Stunde lang auf 100 °C erhitzt. Nach dem Abkühlen wird die Reaktionslösung durch Zugabe von 2 n Natronlauge alkalisch gestellt, mit Chloroform extrahiert und die Chloroformphase mit Wasser gewaschen, getrocknet und im Vakuum eingeengt. Man chromato-graphiert den Rückstand an Kieselgel (Chloroform/Methanol = 45/1), engt die Hauptfraktion ein und fällt die Base aus ätherischer Salzsäure als Hydrochlorid.
Ausbeute: 3,2 g (57% der Theorie),
Schmelzpunkt: 170-172 °C.
Beispiel 3
5,6-Dimethoxy-2N- (3 -[2- (3,4-dimethoxy)-phenyläthyl-
n-propylamino]-propyl)-phthalimidin-hydrochlorid Eine Lösung von 2,5 g (5,5 mMol) 5,6-Dimethoxy-2N-(3-[2-(3,4-dimethoxy)-phenyläthyl-
amino]propyl)-phthalimidin in 100 ml Aceton wird nach Zugabe von 20 ml 1-Brompropan und 5 g Kaliumcarbonat 6 Stunden unter Rückfluss erhitzt. Nach dem Abkühlen wird von der Festsubstanz abfiltriert und das Filtrat eingeengt. Man nimmt in Äther auf, filtriert erneut vom Unlöslichen ab und fällt nach dem Einengen das Hydrochlorid aus ätherischer Salzsäure.
Ausbeute: 0,8 g (29% der Theorie),
Schmelzpunkt: 120-122 °C (Aceton/Methanol).
Analog den Beispielen 1 bis 3 wurden noch folgende Verbindungen hergestellt:
2N-(3-[2-(3,4-Dimethoxy)-phenyläthyl-methylamino]-propyl)-phthalimidin-hydrochlorid.
Schmelzpunkt: 146—148 °C. 5,6-Dimethoxy-2N-(3-[2-(3,4-dimethoxy)-phenyläthylamino]-propyl)-phthalimidin-hydrochlorid.
Schmelzpunkt: 207-209 °C. 5,6-Dimethoxy-2N-(3-[2-(3,4-dimethoxy)-phenyläthyl-n-propylamino]propyl)-phthalimidin-hydrochlorid. Schmelzpunkt: 120—122 °C (Aceton/Methanol). 5,6-Dimethoxy-2N-(3-[2-(3,4-dimethoxy)-phenyläthyl-methylamino]äthyl)phthalimidin-hydrochlorid. Schmelzpunkt: 149-151 °C. 2N-(3-[2-(3,4-Dimethoxy)-phenyläthyl-methylamino]-propyl)-3-phenyl-phthalimidin.
Rf-Wert (Chloroform/Methanol = 19/1): 0,4.
3 -Phenyl-5 -chlor-2N- (3 -[2- (3,4-dimethoxy) -phenyläthyl-methylamino]propyl) -1,2-benzisothiazolin-1,1 -dioxyd-hydrochlorid.
RrWert (Chloroform/Methanol = 19/1): 0,5. 5,6-Dimethoxy-2N-(3-[2-(3,4-dimethoxy)-phenyläthyl-methylamino]propyl- 1,2-benzisothiazolin-1,1-dioxyd-hydrochlorid.
Schmelzpunkt: 196—198 °C (Aceton). 5,6-Methylendioxy-2N-(3-[2-(3,4-dimethoxy)-phenyläthyl-methylamino]-phthalimidin-hydrochlorid.
Schmelzpunkt: 237—239 °C. 5,6-Äthylendioxy-2N-(3-[2-(3,4-dimethoxy)-phenyläthyl-methylamino]propyl)-phthalimidin-hydrochlorid. Schmelzpunkt: 208—210 °C.

Schmelzpunkt: 135-136 °C. 5,6-Dimethoxy-2N-(3-[2-(3,4-dimethoxy)-phenylisopropyl-methylamino]propyl)-phthalimidin-hydrochlorid. Schmelzpunkt: 183-185 °C.
Claims (5)
- 626 6072PATENTANSPRÜCHE 1. Verfahren zur Herstellung neuer substituierter Arylalkylamine der Formel I
- R.•N~(CH2)n~N"CH~CH2R„(I),worinR.! ein Wasserstoffatom, eine niedere Alkylgruppe oder die Phenylgruppe;R2 ein Wasserstoffatom, ein Chloratom oder die Methoxy-gruppe;R3 ein Wasserstoffatom oder die Methoxygruppe oder zusammen mit R2 die Methylendioxy- oder Äthylendioxygrappe ;R4 und Rä die gleich oder verschieden sein können, Wasserstoffatome oder niedere Alkylgruppen;R6 ein Wasserstoffatom oder eine niedere Alkoxygruppe;R7 eine niedere Alkoxygruppe oder zusammen mit R6 die Methylendioxy- oder Äthylendioxy grappe;X die Carbonyl- oder Sulfonylgruppe; und n die Zahl 2 oder 3 bedeuten, sowie von deren physiologisch verträglichen Säuren, dadurch gekennzeichnet, dass man eine Verbindung der Formel IIH R<N - H (II),IZ-(CH2)n-N-CH-CH2 R*(III),in der Rt, R2, R3 und X wie eingangs definiert sind, mit einem Alkylamin der Formel III25in der R4, Rs, R6, R7 und n wie eingangs definiert sind und Z eine nukleophil austauschbare Gruppe darstellt, umsetzt und gewünschtenfalls eine erhaltene Verbindung der Formel I in ein physiologisch verträgliches Säureadditionssalz mit einer an-30 organischen oder organischen Säure überführt.
- 2. Verfahren nach Patentanspruch 1, dadurch gekennzeichnet, dass man eine Verbindung der Formel III verwendet, in der Z ein Chlor-, Brom- oder Jodatom, eine Alkylsulfonyl-oxy- oder Arylsulfonyloxygruppe bedeutet.35 3. Verfahren nach den Patentansprüchen 1 und 2, dadurch gekennzeichnet, dass man die Umsetzung in einem Lösungsmittel und bei Temperaturen zwischen 0 und 150 °C durchführt.
- 4. Verfahren nach den Patentansprüchen 1 bis 3, dadurch 40 gekennzeichnet, dass man die Umsetzung in Gegenwart eines säurebindenden Mittels und/oder eines Reaktionsbeschleunigers durchführt.
- 5. Verwendung einer Verbindung der Formel I, hergestellt nach dem Verfahren gemäss Patentanspruch 1, in der R4 Was-45 serstoff bedeutet, zur Herstellung der entsprechenden Nieder-alkylderivate, dadurch gekennzeichnet, dass man sie durch Al-kylieren in eine Verbindung der Formel I überführt, in der R4 eine niedere Alkylgruppe bedeutet.
