Description
Der Erfindung liegt die Aufgabe zugrunde, ein Verfahren zur 35 Herstellung von neuen w-Aminoalkoxybenzolderivaten mit hoher antidepressiver Wirkung zu schaffen. Diese Aufgabe wird durch die Erfindung gelöst.
Die Erfindung betrifft somit die in den Patentansprüchen gekennzeichneten Gegenstände.
Spezielle Beispiele für Cj_5-Alkylreste sind die Methyl-, Ähyl-, Propyl-, Isopropyl-, Butyl- und Isobutylgruppe. Spezielle Beispiele für C1_5-Hydroxyalkylreste sind die Hydroxymethyl-, 1-Hydroxy-äthyl-, 2-Hydroxyäthyl-, 1-Hydroxypropyl-, 2-Hydroxypropyl- und 45 3-Hydroxypropylgruppe. Spezielle Beispiele für die C3_7-N,N-Polymethyleniminylgruppe sind die 1-Azetidinyl-, 1-Pyrrolidinyl-und Piperidino- bzw. 1-Hexamethylniminylgruppe. Als Halogenatome Y und Y' kommen vorzugsweise Fluor-, Chlor- und Bromatome in Frage. Spezielle Beispiele für C^-Alkoxyreste sind die Me-50 thoxy-, Äthoxy-, Propoxy- und Isopropoxygruppe, n und n' haben vorzugsweise den Wert 1 oder 2. Spezielle Beispiele für den Rest R sind die Aminogruppe, C^-Alkylaminogruppen, wie die Methyl-amino- und Äthylaminogruppe, C2.6-Dialkylaminogruppen, wie die Dimethylamino-, und Diäthylaminogruppe, C^-Hydroxyalkyl-55 aminogruppen, C2.6-Bis-(hydroxyalkyl)aminogruppen, wie die Bis-(2-hydroxyäthyl)aminogruppe, C2.6-N-Alkyl-N-hydroxyalkylamino-gruppen und die 1-Pyrrolidinylgruppe.
Bevorzugte Reste Y und Y' sind Wasserstoff- oder Chloratome, Hydroxy- oder Trifluormethylgruppen, C^-Alkylreste, wie die 60 Methylgruppe, und Cj.j-Alkoxyreste, wie die Methoxy- und Äthoxy-gruppe. Der Rest Y kann in der 2'-, 3'-, 4'-, 5'- oder 6'-Stellung stehen. In gleicher Weise kann der Rest Y' in der 3-, 4-, 5- und 6-Stel-lung stehen. Spezielle Beispiele für den Rest Z sind die Oxy-, Thio-, Methylen- und Methylmethylengruppe. Der bevorzugte Rest Rist 65 die Methylamino-, Äthylamino-, Dimethylamino-, Diäthylamino-, Bis-(2-hydroxyäthyl)amino- und 1-Pyrrolidinylgruppe. Die bevorzugten Reste Y und Y' sind Wasserstoff- oder Chloratome, Hydroxy-, Methyl-, Methoxy- und Äthoxygruppen. Die bevorzugten

Reste Z sind die Oxy- und Methylengruppe. Vorzugsweise hat r den Wert 3 oder 4.
Die Salze der Verbindungen der Formel I können sich von anorganischen oder organischen Säuren ableiten. Spezielle Beispiele für die zur Salzbildung verwendbaren Säuren sind Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Salpetersäure, Essigsäure, Bernsteinsäure, Adipinsäure, Propionsäure, Weinsäure, Äpfelsäure, Citronensäure, Benzoesäure, Toluolsulfonsäure und Methansulfonsäure.
Bevorzugt sind folgende Verbindungen der Formel I: 2-(4-Methylaminobutoxy)-3-methyldiphenylmethan, 2-(4-Dimethylaminobutoxy)-3-methyldiphenylmethan, 2-(4-Dimethylaminobutoxy)-5-methyldiphenylmethan, 2-(4-Methylaminobutoxy)-4'-methyldiphenylmethan, 2-(4-Methylaminobutoxy)-4'-chlordiphenylmethan, 2-[4-Bis-(2-hydroxyäthyl)-aminobtuoxy]-4'-methoxydiphenyl-methan,
2-(4-Methylaminobutoxy)-2'-methoxydiphenylmethan, 2-(4-Methylaminobutoxy)-2'-hydroxydiphenylmethan, 2-(4-Dimethylaminobutoxy)-2'-hydroxydiphenylmethan, 2-(4-Methylaminobutoy)-2'-äthoxydiphenyläther, 2-(4-Methylaminobutoxy)-2'-methyldiphenyläther, 2-(3-Methylaminopropoxy)-2'-methyldiphenyläther, 2-(3-Dimethylaminopropoxy)-2'-methyldiphenyläther, 2-(3-Äthylaminopropoxy)-2'-methyldiphenyläther, 2-(3-Diäthylaminopropoxy)-2'-methyldiphenyläther und 2-[3-( 1 -Pyrrolidinyl)-propoxy]-2'-methyldiphenyläther.
Aufgrund ihrer hohen antidepressive Wirkung und ihrer geringen Toxizität sind folgende Verbindungen der Formel I besonders bevorzugt:
2-(4-Methylaminobutoxy)-3-methyldiphenylmethan, 2-(4-Dimethylaminobutoxy)-3-methyldiphenylmethan, 2-(4-Dimethylaminobutoxy)-5-methyldiphenylmethan, 2-(4-Methylaminobutoxy)-2'-methoxydiphenylmethan, 2-(4-Methylaminobutoxy)-2'-hydroxydiphenylmethan, 2-(4-Dimethylaminobutoxy)-2'-hydroxydiphenylmethan, 2-(4-Methylaminobutoxy)-2'-äthoxydiphenyläther, 2-(4-Methylaminobutoxy)-2'-methyldiphenyläther, 2-(3-Methylaminopropoxy)-2'-methyldiphenylätherund 2-(3-Dimethylaminopropoxy)-2'-methyldiphenyläther.
Die im erfindungsgemässen Verfahren eingesetzten «-Halogenalkoxybenzolderivate der Formel II können durch Umsetzung eines 2-substituierten Phenols der Formel:

n in der Y, Y', Z, n, n' und m die in Anspruch 1 angegebene Bedeutung haben, mit einem 1,3-Dihalogenpropan, 1,4-Dihalogenbutan oder 1,5-Dihalogenpentan in Gegenwart einer Base hergestellt werden.
Spezielle Beispiele für verfahrensgemäss eingesetzte 2-substituier-te Phenole der Formel IV sind 2-Hydroxy-3-methyldiphenylmethan, 2-Hydroxy-5-methyldiphenylmethan, 2-Hydroxy-4'-chlordiphenyl-methan, 2-Hydroxy-4'-methoxydiphenylmethan, 2-Hydroxy-2'-methoxydiphenylmethan, 2,2'-Dihydroxydiphenylmethan, 2,2'-Dihydroxybiphenyl, 2,2'-Dihydroxydiphenyläther, 2-Hydroxy-2'-methoxydiphenyläther, 2-Hydroxy-2'-äthoxydiphenyläther, 2-Hy-droxy-2'-methyldiphenyläther, 2-Hydroxy-3,5-dimethyl-2',3'-dimethoxydiphenylmethan, 2-Hydroxy-3-methyl-5,4'-dichlor-diphenylmethan, 2-Hydroxy-5-chlor-3-isopropyl-6-methyldiphenyl-methan, 2-Hydroxy-4,6-dimethyl-4'-bromdiphenylmethan, 2-hy-droxy-4'-chlordiphenyläther, 2-Hydroxy-3,4'-dimethyldiphenyl-
äther, 2-Hydroxy-3,5-dibrombiphenyl, 2-Hydroxy-3,5,4'-tribrombi-phenyl, 2-Hydroxy-3,5-dimethylbiphenyl und 2-Hydroxy-3-äthyl-biphenyl.
Im erfindungsgemässen Verfahren setzt sich das Amin beispielsweise mit der äquimolaren Menge des w-Halogenalkoxybenzol-derivats um. Bei Verwendung überschüssigen Amins wird die Reaktion beschleunigt. Im allgemeinen wird das Amin in einer Menge von 1 bis 100 mol, vorzugsweise 2 bis 40 mol pro mol u-Halogenalkoxy-benzolderivat eingesetzt. Bei Verwendung einer grossen Menge des Amins dient dieses gleichzeitig als Lösungsmittel. Die Umsetzung kann in Gegenwart eines Lösungsmittels durchgeführt werden. Spezielle Beispiele für verwendbare Lösungsmittel sind Wasser, Dioxan, Tetrahydrofuran, Dimethylsulfoxid, niedere aliphatische Alkohole und deren Gemische. Vorzugsweise wird als Lösungsmittel ein Gemisch von Wasser mit einem niederen aliphatischen Alkohol eingesetzt. Die Reaktionstemperatur ist nicht kritisch. Im allgemeinen liegt sie im Bereich von Zimmertemperatur bis 150°C, vorzugsweise von Zimmertemperatur bis 100°C. Die Reaktionsdauer hängt von der Reaktionstemperatur und der Reaktionsfähigkeit der Ausgangsverbindungen ab. Normalerweise liegt sie im Bereich von 10 min bis 40 h.
Die Gegenwart einer Base, die den während der Umsetzung gebildeten Halogenwasserstoff neutralisiert, beschleunigt die Reaktion. Beispiele für derartige Basen bzw. Halogenwasserstoffacceptoren sind anorganische Basen, wie Kaliumhydroxid, Natriumhydroxid, Kaliumcarbonat und Natriumcarbonat, sowie tertiäre Amine, wie Pyridin und Triäthylamin. Die Base wird im allgemeinen in einer Menge von 1 bis 5 mol pro mol co-Halogenalkoxybenzolderivat eingesetzt. Beim Arbeiten in Abwesenheit einer Base setzt sich das entstandene (O-Aminoalkoxybenzolderivat der allgemeinen Formel I mit dem bei der Umsetzung entstandenen Halogenwasserstoff unter Bildung des entsprechenden Salzes um.
Die Herstellung der Salze durch Umsetzung der freien Base mit einer anorganischen oder organischen Säure erfolgt in an sich bekannter Weise.
Die erfindungsgemäss hergestellten co-Aminoalkoxybenzol-derivate der Formel I und ihre Salze können durch Umkristallisation aus einem Lösungsmittel, wie einem Gemisch von Äthanol und Diäthyläther, gereinigt werden.
Die Verbindungen der Formel I sind wertvolle Arzneistoffe mit antidepressiver Wirkung. Dies haben pharmakologische Versuche ergeben, bei denen die Verbindungen mit Amitriptylin auf ihre Antireserpinaktivität an Mäusen untersucht wurden. Die Verbindungen wurden intraperitoneal gegeben. Die antidepressive Wirkung wird anhand des Antagonismus der durch Reserpin (5 mg/kg i.p.) induzierten Hypothermie ausgedrückt; vgl. P.S.J. Spencer, « Antidep-ressant Drugs», S. Garattini und M.N.G. Duhes, «Excerpta Medica Foundation», Amsterdam, S. 194 bis 204,1967. Die Antireserpinaktivität wird als relative Aktivität, bezogen auf Amitriptylin = 1, ausgedrückt.
Die akute Toxizität (LD50) wird nach der Methode von Litch-field-Wilcoxon berechnet.
Die ZNS-dämpfende Wirkung ergibt sich aus der Fähigkeit der untersuchten Verbindungen, im Zugtest und in der spontanen motorischen Aktivität ein neurologisches Defizit hervorzurufen; vgl. S. Courvoisier, R. Ducrot, L. Julou, «Psychotoropic Drugs», S. Garattini, V. Ghetti, 1957, S. 373. Die spontane motorische Aktivität wird in einem Animex-Apparat bestimmt. Die antiepileptische Wirkung wird im Elektroschocktest bestimmt: vgl. L.S. Goodman, M. Singh Grewak, W.C. Brown und E.A. Swinyard, « J. Pharmacol. Exptl. Therap.», Bd. 108 (1953), S. 168.
Die zentrale anticholinergische Wirkung wird durch Untersuchung des durch Tremorin induzierten Tremors bei Mäusen bestimmt; vgl. G.M. Everett, L.E. Bloucus und J.M. Sheppard, «Science», Bd. 124 (1956), S. 79.

Aus Tabellen I und II ist ersichtlich, dass die untersuchten Verbindungen der Formel I eine dem Amitriptylin vergleichbare Anti-reserpinwirkung entfalten, jedoch eine niedrige Toxizität, schwache ZNS-dämpfende und anticholinergische Wirkung aufweisen.
Die Verbindungen können oral, parenteral, subcutan, intravenös, intramuskulär oder intraperitoneal in üblichen Darreichungsformen, gegeben werden.
Zur Herstellung von Arzneipräparaten können die üblichen Trä-gerstoife und/oder Verdünnungsmittel und/oder Hilfsstoffe verwendet werden.

methan, 20 ml einer 40prozentigen wässrigen Lösung von Methylamin und 100 ml Äthanol wird 8 h lang bei Zimmertemperatur stehen gelassen. Sodann werden Äthanol und überschüssiges Methylamin unter vermindertem Druck abdestilliert. Der Rückstand wird mit 2 n Natronlauge versetzt und das Gemisch mit Diäthyläther extrahiert. Der Ätherextrakt wird destilliert, der Rückstand mit 2 n Salzsäure versetzt und zur Trockene eingedampft. Der erhaltene
Rückstand wird aus einem Gemisch von Äthanol und Diäthyläther umkristallisiert. Es werden 4,1 g (85% d. Th.) 2-(4-Methylamino-butoxy)-3-methyldiphenylmethanhydrochlorid vom F. 94 bis 98° C 65 erhalten.

methan in 10 g Diäthanolamin wird 10 h lang bei Zimmertemperatur stehen gelassen. Sodann wird das Diäthanolamin unter vermindertem Druck abdestilliert, der Rückstand mit 2 n Natronlauge versetzt und mit Diäthyläther extrahiert. Der Ätherextrakt wird eingedampft, der Rückstand mit 2 n Salzsäure versetzt und das Gemisch zur Trok-kene eingedampft. Der erhaltene Rückstand wird aus einem Gemisch von Äthanol und Diäthyläther umkristallisiert. Es werden 4,0 g (69%
d. Th.) 2-[4-Bis-(2-hydroxyäthyl)aminobutoxy]-4'-methoxydiphenyl-methanhydrochlorid vom F. 102 bis 104°C erhalten.
Die in der nachstehenden Tabelle III aufgeführten Verbindungen werden gemäss Beispiel 1 oder 2 aus den entsprechenden Ausgangsverbindungen hergestellt.

2. co-Aminoalkoxybenzolderivate nach Anspruch 1 der allgemeinen Formel I, in der R eine Amino-, C1.3-Alkylamino-, C2.6-Dialkylamino-, C^-Hydroxyalkylamino-, C2_6-Bis-(hydroxyalkyl)-65 amino-, C2.6-N-Alkyl-N-hydroxyalkylamino- oder 1-Pyrrolidinyl-gruppe, Y und Y' Wasserstoff- oder Halogenatome, Hydroxy- oder Trifluormethylgruppen, C1_3-Alkyl- oder C1_3-Alkoxyreste und Z eine Oxy-, Thio-, Methylen- oder Äthyldiengruppe bedeuten, n den

3. co-Aminoalkoxybenzolderivate nach Anspruch 1 und 2 der allgemeinen Formel I, in der R eine Methylamino-, Äthylamino-, Dimethylamino-, Diäthylamino-, Bis-(2-hydroxyäthyl)amino- oder 1-Pyrrolidinylgruppe, Y und Y' Wasserstoff- oder Chloratome, Hydroxy-, Methyl-, Methoxy- oder Äthoxygruppen und Z eine Oxy-oder Methylengruppe bedeuten, r den Wert 3 oder 4, m den Wert 1, n den Wert 1 und n' den Wert 1 hat, und ihre Salze mit Säuren.
4. co-Aminoalkoxybenzolderivate nach Anspruch 1 bis 3 der allgemeinen Formel I, in der R eine Methylamino- oder Dimethyl-aminogruppe, Y und Y' Methyl-, Methoxy-, Äthoxy- oder Hydroxy-gruppen bedeuten, und ihre Salze mit Säuren.
5. 2-(4-Methylaminobutoxy)-3-methyldiphenylmethan.
6. 2-(4-Dimethylaminobutoxy)-3-methyldiphenylmethan.
7. 2-(4-Dimethylaminobutoxy)-5-methyldiphenyimethan.
8. 2-(4-Methylaminobutoxy)-2'-methoxydiphenylmethan.
9. 2-(4-Methylaminobutoxy)-2'-hydroxydiphenylmethan.
10. 2-(4-Dimethylaminobutoxy)-2'-hydroxydiphenylmethan.
11. 2-(4-Methylaminobutoxy)-2'-äthoxydiphenylmethan.
12. 2-(4-Methylaminobutoxy)-2'-methyldiphenylmethan.
13. 2-(3-Methylaminopropoxy)-2'-methyldiphenyläther.
14. Verfahren zur Herstellung der Verbindungen nach Anspruch
1, dadurch gekennzeichnet, dass man ein w-Halogenalkoxybenzol-derivat der allgemeinen Formel II:

io in der X ein Halogenatom darstellt und Y, Y', Z, r, m, n und n' die in Anspruch 1 angegebene Bedeutung haben, mit einem Amin der allgemeinen Formel III :

in der R die in Anspruch 1 angegebene Bedeutung hat, zur Umsetzung bringt und gegebenenfalls die erhaltene Base mit einer Säure in ein Salz überführt.
15. Verfahren nach Anspruch 14, dadurch gekennzeichnet, dass 20 man 1 bis 100 mol Amin pro Mol co-Halogenalkoxybenzolderivat bei
Temperaturen von Raumtemperatur bis 150° C zur Umsetzung bringt.
16. Arzneimittel mit antidepressiver Wirkung, enthaltend eine Verbindung nach Anspruch 1 bis 13.
Claims (3)
1. Verfahren zur Herstellung von w-Aminoalkoxybenzol-derivaten der Formel:

in der R! und R2 gleich oder verschieden Wasserstoff, C1_5-Alkyl- oder C^-Hydroxyalkyl darstellen, oder
(b) eine C3.7-N,N-Polymethyleniminyl-, Morpholino-, Thiomor-pholino-, 4-Oxothiomorpholino-, 1-Piperazinyl-, 4-Methyl-l-pipera-zinyl-, 4-Äthyl-l-piperazinyl- oder 4-Acetyl-l-piperazinylgruppe bedeutet,
Y und Y' Wasserstoff oder Halogen, Hydroxy oder Trifluormethyl, C].5-Alkyl oder C[.6-Alkoxy darstellen, Y und Y'jedoch nicht gleichzeitig Wasserstoff sind, Z Oxy, Thio oder Methylen, eine Gruppe der Formel:

zur Umsetzung bringt und gegebenenfalls die erhaltene Base mit einer Säure in ein Salz überführt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man 1 bis 100 mol Amin pro mol w-Halogenalkoxybenzolderivat bei Zimmertemperatur bis 150°C zur Umsetzung bringt.

PATENTANSPRÜCHE
3. Nach dem Verfahren nach Anspruch 1 hergestellte w-Amino-alkoxybenzolderivate der Formel I und deren Salze mit Säuren.
L.C. Cheney u. Mitarb., «J. Am. Chem. Soc.», Bd. 71 (1949), S. 60 bis 64, haben mehrere Diphenylmethane mit einem Substituen-ten in der 2-Stellung, beispielsweise einer 2-Dimethylaminoäthoxy-, 2-Diäthylaminoäthoxy-, 2-Morpholinoäthoxy-, 2-(l-Piperidyl)-5 äthoxy, 2-Isopropylaminoäthoxy-, 3-(l-Piperidyl)propoxy-, 3-Dimethylaminopropoxy- und 3-Dibutylaminopropoxygruppe, beschrieben. Es ist ferner beschrieben, dass 2-(2-Aminoäthoxy)-diphenylmethane und 2-(3-Aminopropoxy)diphenylmethane Antihi-staminica und Lokalanästhetica sind. Ferner sind in den US-PS io Nr. 2703324,2966518,2534236 und 2768207 2-co-Aminoalkoxy-diphenylmethane, wie 2-(3-Diäthylaminopropyl)-diphenylmethan und 2-(5-Dimethylaminopentyl)diphenylmethan, beschrieben. In der Zeitschrift «J. Med. Chem.», Bd. 9 (1966), S. 806, ist 2-(4-Dimethyl-aminobutoxy)diphenylmethan beschrieben. Ferner sind in der Zeit-15 schrift «Journal Pharm. Soc. Japan», Bd. 89 (1969), S. 1078 bis 1084, und Bd. 89 (1970), S. 1417 bis 1425, sowie den JP-AS 9937/1970 und 9938/1970 2-(o)-Dialkylaminoäthoxy)-diphenyläther, 2-(o>-Cyclo-iminoäthoxy)diphenyläther, 2-(co-Dialkylaminopropoxy)diphenyl-äther und 2-(w-Cycloiminopropoxy)diphenyläther beschrieben, bei 20 denen jeder Benzolring gegebenenfalls durch ein Chloratom oder eine Methylgruppe substituiert ist. In «Chemical Abstracts», Bd. 72, 3152 y und Bd. 73, P 76852, ist der 2-(3-Diäthylaminopropoxy)-diphenyläther beschrieben. In «Chemical Abstracts», Bd. 45 (1951), S. 577, ist das 2-(Diäthylaminoäthoxy)diphenylsulfid beschrieben. In 25 der US-PS Nr. 2703324 ist das 2-(2-Diäthylaminoäthoxy)diphenyl-methylmethan beschrieben. In den US-PS Nr. 2187723 und 3213140 sind 2-co-Aminoalkoxybiphenylverbindungen, wie 2-(2-Pentylamino-äthoxy)biphenyl, beschrieben. Schliesslich sind in der GB-PS Nr. 521575 2-w-Aminoalkoxybiphenylverbindungen, wie 2-(3-30 Dimethylaminopropoxy)biphenyl, 2-(2-Aminoäthoxy)biphenyl und 2-(2-Äthylaminoäthoxy)biphenyl, beschrieben. Es findet sich jedoch in den vorstehend aufgeführten Druckschriften kein Hinweis, dass diese Verbindungen eine antidepressive Wirkung aufweisen.
