Description
Die Erfindung betrifft ein Verfahren zur Herstellung neuer Benzo-(b)-bicyclo-[3,3,l]-nonene der allgemeinen Formel I

Rj ein Wasserstoffatom, eine (Q-QO-Alkyl- oder eine gegebenenfalls substituierte Aryl-(C1-C6)-alkylgruppe ist, R2 ein Wasserstoffatom bedeutet,
R3 eine freie, verätherte oder veresterte Hydroxygruppe ist und
X und Y jeweils ein Wasserstoff- oder Halogenatom, eine Hydroxygruppe, (C1-C6)-Alkyl-, (Q-Cg^Alkoxy-, Nitro-, Trifluormethyl- oder Acyloxygruppe bedeuten und deren Salze, das dadurch gekennzeichnet ist, dass man die Imino-gruppe einer Verbindung der Formel IV
in welcher X, Y und R3 die oben angegebene Bedeutung haben und R4 ein Wasserstoffatom, eine Hydroxy-, eine Alk-oxy-, eine (Ci-C6)-Alkyl- oder eine gegebenenfalls substituierte Aryl-(C1-C6)-alkylgruppe bedeutet oder ein Salz davon, reduziert und anschliessend die so erhaltene Verbindung gegebenenfalls in ein pharmazeutisch geeignetes Salz umwandelt.
Die Verbindungen der allgemeinen Formel I besitzen wertvolle biologische Aktivitäten, insbesondere anorektische (appetitzügelnde) Aktivität. Ausserdem ist die Toxizität der Verbindungen sehr gering.
Die Verbindungen der Formel IV können hergestellt werden durch Umsetzung eines Ketons der Formel II
in welcher X, Y und R3 die oben angegebene Bedeutung haben, mit einem primären Amin oder mit Hydroxylamin oder einem Hydroxylaminalkyläther unter alkalischen Bedingungen auf eine für die Herstellung von Iminen oder Oximen übliche Weise.
Die Reduktion des Amins der Formel IV kann unter milden Reaktionsbedingungen mit Hilfe von Standardverfahren zur Reduktion einer Iminogruppe durchgeführt werden. Zum Beispiel kann die Reduktion durchgeführt werden durch Hydrierung in Gegenwart eines geeigneten Katalysators, wie Raney-Nickel, Pt oder PtOz, mit einem Metallhydrid, besonders komplexen Metallhydriden, die abgeleitet sind von Aluminium oder Bor, wie Lithiumaluminiumhydrid, gelöst oder suspendiert in einer geeigneten Flüssigkeit, wie Äther, mit einem Alkalimetall, vorzugsweise Natrium, in einer geeigneten Flüssigkeit, wie Äther, Benzol oder Alkohol oder mit Natriumamalgam oder Zinkstaub in beispielsweise Natriumhydroxid. Die Reaktion kann gleichzeitig zu einer Abspaltung der 4-Äther- oder der 4-Ester-gruppe unter Bildung des entsprechenden 4-Alkohols führende nach dem angewandten Reduktionsmittel.
Nach der Reaktion zur Herstellung der Verbindungen der Formel I können weitere Reaktionen durchgeführt werden zur Umwandlung einer Verbindung der Formel I in ein Salz oder Stickstoffoxid oder zur Umwandlung einer Verbindung der Formel I in eine andere Verbindung der Formel I.
So ist es möglich, einen an dem Phenylkern vorhandenen Substituenten in einen anderen Substituenten, der unter die Definition von X und/oder Y fällt, umzuwandeln. Zum Beispiel kann eine Methoxygruppe umgewandelt werden in eine Hydroxylgruppe, z.B. durch Behandlung mit geschmolzenem Pyridinhydrochlorid in Abwesenheit eines Lösungsmittels oder durch Hydrolyse mit HBr: eine Hydroxygruppe kann umgewandelt werden in eine Alkoxygruppe, ein Halogenatom oder eine Acyloxygruppe auf übliche Weise.
Die am Stickstoffatom unsubstituierten oder monosub-stituierten erfindungsgemässen Amine (Rx und/oder R2 = Wasserstoff) können (ar)alkyliert werden auf übliche Weise, z.B. durch Umsetzung der Verbindung mit einem (Ar)-alkylhalogenid oder durch Acylierung der Verbindung mit anschliessender Reduktion der Carbonylgruppe.
Zur Einführung von Methylgruppen an dem Stickstoffatom können das bekannte Verfahren nach Eschweiler-Clar-ke (Umsetzung mit Formaldehyd und Ameisensäure) oder die Umsetzung mit Formaldehyd oder Halogenameisensäureestern und anschliessende Reduktion mit z. B. Natriumcyanoborhydrid vorzugsweise angewandt werden.
Ein N-Acylderivat der erfindungsgemäss erhaltenen Verbindungen kann vorzugsweise erhalten werden durch Acylierung einer Verbindung der Formel I, bei der zumindest eine der Gruppen Rj und/oder R2 ein Wasserstoffatom ist, auf übliche Weise unter Anwendung eines Anhydrides oder Säurehalogenides.
Derartige Acylierungen von 4-Alkoholderivaten I (R3 = OH) führen zur gleichzeitgen Veresterung der 4-Hydroxygruppe.
Ein O-Acylderivat (R3 = Acyloxy) der neuen Verbindungen, bei denen die primäre oder sekundäre Amingruppe unverändert bleibt, kann hergestellt werden durch Behandlung des Amins I, bei dem R3 = OH ist, mit einem Acylierungs-mittel, wie einem Säurehalogenid oder Anhydrid in Gegenwart einer starken Säure, wie Trifluoressigsäure oder Perchlorsäure.
Alle diese zusätzlichen Umwandlungen sind bekannte Standardverfahren. Soweit spezielle Reagenzien für diese zusätzlichen Umwandlungen erwähnt worden sind, ist es selbstverständlich, dass diese Reagenzien durch andere in der organischen Chemie bekannte Reagenzien, die eine ähnliche Wirkung wie die angegebenen besitzen, ersetzt werden können.
Die Herstellung des als Ausgangsverbindung für die er-findungsgemässe Synthese einer Verbindung der Formel I verwendeten Ketone II ergibt ein Gemisch, bestehend aus zwei Enantiomeren der Formel II, bei denen die 4-Alkohol-oder Estergruppe sich in exo-Stellung befindet, und zwei Enantiomeren der Formel II, bei denen sich die 4-Alkohol-oder Estergruppe in der endo-Stellung befindet. Die Exo-Enantiomeren können von den endo-Enantiomeren durch Kristallisation oder Chromatographie getrennt werden. Jedes der so erhaltenen racemischen Gemische II kann als solches weiter verwendet werden und dann gegebenenfalls nach Umwandlung in die racemische Verbindung I optisch aufge5

spalten werden, oder es kann zunächst aufgespalten werden und dann in ein optisch aktives Endprodukt der Formel I umgewandelt werden.
Die Reduktion des Imins der Formel IV führt ein weiteres Asymmetriezentrum ein und führt zu einem Gemisch von zwei Diastereomeren I, bei denen die Aminogruppe in synoder anti-Stellung, bezogen auf den Benzolring, vorliegt. Diese einzelnen diastereomeren Formen können von dem Gemisch auf übliche Weise isoliert werden, z.B. durch Säulenchromatographie, präparative Dünnschichtchromatographie und/oder fraktionierte Kristallisation.
Die Herstellung der Diastereomeren, Enantiomeren und deren Gemische fallt ebenfalls unter die Erfindung.
Die neuen Verbindungen der Formel I können von dem Reaktionsgemisch in Form eines pharmazeutisch verträglichen Salzes isoliert werden, je nach den Bedingungen, unter denen die Reaktion durchgeführt wird. Die pharmazeutisch geeigneten Salze können auch erhalten werden durch Behandlung der freien Base der Formel I mit einer organischen oder anorganischen Säure, wie HCl, HBr, HJ, H2S04, H3PO4, Essigsäure, Propionsäure, Glykolsäure, Maleinsäure, Malonsäure, Bernsteinsäure, Weinsäure, Zitronensäure, Benzoesäure, Ascorbinsäure usw.
Der Ausdruck «Alkylgruppe», wie er für die Definition von X, Y, Rj und R2 der Formel I angewandt wird, bedeutet eine gesättigte, verzweigte oder nicht-verzweigte Kohlenwasserstoffkette mit 1 bis 6 Kohlenstoffatomen, wie eine Methyl-, Äthyl-, n-Propyl-, n-Butyl-, Cyclopropyl-, Cyclopropylmethyl-, Isopropyl-, Isobutyl-, tert.-Butyl-, n-Pentyl-, Isopentyl-, Cyclopentyl- oder Hexylgruppe. Das Gleiche trifft für die Alkylgruppen zu in dem Ausdruck «Alkoxygruppe», wie er zur Definition von X und Y angewandt wird.
Der Ausdruck «Alkenylgruppe», wie er zur Definition von Rj und R2 bei der Formel I angewandt wird, bedeutet eine ungesättigte verzweigte oder unverzweigte Kohlenwasserstoffgruppe mit 2 bis 6 Kohlenstoffatomen, wie eine Vi-nyl-, Allyl- und 3-Methyl-but-2-enylgruppe.
Unter «Halogenatom» ist vorzugsweise ein Chlor- oder Bromatom zu verstehen.
Unter einer «Aralkylgruppe», wie sie zur Definition von Ri und R2 angegeben ist, ist eine Alkylgruppe mit 1 bis 6 Kohlenstoffatomen zu verstehen, die durch mindestens eine aromatische Gruppe substituiert ist, vorzugsweise eine Phe-nylalkylgruppe, bei der die Alkylgruppe 1 bis 4 Kohlenstoffatome besitzt und die Phenylgruppe durch ein oder mehrere Halogenatome, niedere Alkylgruppen oder niedere Alkoxy-gruppen (1 bis 4C) substituiert sein kann, wie eine Benzyl-, Phenyläthyl-, Phenylpropyl-, Phenylbutyl-, 1-Methyl-l-phenyläthyl-, o-, m- oder p-Anisyl-, o-, m- oder p-Chlor-benzyl-, Veratryl-, o-, m- oder p-Methylphenäthyl- oder o-, m- oder p-Hydroxyphenäthylgruppe.
Eine «Acylgruppe», wie sie zur Definition von Rx und R2 angegeben ist, bedeutet eine Gruppe, die abgeleitet ist von einer aliphatischen, araliphatischen oder aromatischen Carbonsäure. Die aliphatischen Carbonsäuren umfassen vorzugsweise Säuren mit 1 bis 18 Kohlenstoffatomen einschliesslich solche mit einem carbocyclischen Ring und besonders solche mit 1 bis 8 Kohlenstoffatomen, wie Essigsäure, Propionsäure, Buttersäure, Isobuttersäure, Valerian-säure, Hexansäure, Heptansäure, Trimethylessigsäure, Cy-clopentan- oder Cyclohexancarbonsäure. Die araliphatischen oder aromatischen Carbonsäuren umfassen nicht-sub-stituierte sowie substituierte araliphatische oder aromatische Carbonsäuren mit vorzugsweise 1 bis 18 Kohlenstoffatomen, besonders die gegebenenfalls substituierten Phenyl-carbonsäuren und gegebenenfalls substituierte Phenylalkyl-carbonsäuren, bei denen die Alkylgruppe 1 bis 4 Kohlenstoffatome enthält und gesättigt oder ungesättigt sein kann, wie Benzoesäure, o-, m- oder p-Toluolsäure, o- oder p-Chlorbenzoesäure, p-Methoxybenzoesäure, Phenylessig-säure, Phenylpropionsäure, Zimtsäure, Phenylbuttersäure, p-Methylphenylessigsäure, p-Nitrobenzoesäure usw.
Die Acylgruppe bei den Ausdrücken «Acyloxygruppe» und «veresterte Hydroxygruppe» bei den Definitionen von X, Y und R3 besitzt eine ähnliche Bedeutung.
Die für die Definition von R3 erwähnte verätherte Hydroxygruppe ist vorzugsweise eine aliphatische (1 bis 6C)-Äthergruppe, eine cycloaliphatische (5 bis 10C)-Äthergrup-pe, eine aromatische Äthergruppe, vorzugsweise ein Phenyl-äther, eine araliphatische Äthergruppe, vorzugsweise ein Phenylalkyläther, oder eine heterocyclische Äthergruppe, wie ein Tetrahydropyranyläther. Die Phenylgruppe bei den Phenyl- oder Phenylalkyläthern kann substituiert sein durch Ci-C4-Alkylgrappen, C1-C4-Alkoxygruppen, Halogenatome und/oder Nitrogruppen.
Die heterocyclischen 5- oder 6-gliedrigen Ringe, wie sie bei der Definition von Rj und R2 (zusammen mit dem Stickstoffatom) erwähnt sind, sind abgeleitet von 5- oder 6-gliedrigen cyclischen Aminen der Formel III, wie Pyrrol, Pyrrolidin, Pyrrolin, Piperidin, Piperazin, Imidazol oder Morpholin.
Die Stickstoffoxide der erfindungsgemäss erhaltenen Verbindungen werden hergestellt durch Oxidation mit einer Verbindung der Formel I mit H202 oder einer Persäure.
Die erfindungsgemäss hergestellten Verbindungen sowie deren pharmazeutisch geeignete Salze besitzen, wie erwähnt, wertvolle appetithemmende Eigenschaften. Eine lang andauernde orale Verabreichung von Verbindungen der Formel I führt nicht zu einer Gewöhnung bezüglich der anorek-tischen Wirkung. Die Verbindungen der Formel I, besonders die 11-syn-Isomeren I, zeigen darüber hinaus eine massige entzündungshemmende Aktivität. Ausserdem zeigt das gesamte pharmakologische Profil der erfindungsgemässen Verbindungen eine antidepressive Wirksamkeit.
Die erfindungsgemäss erhaltenen Verbindungen können oral, parenteral oder lokal (das letztere im wesentlichen für entzündungshemmende Zwecke) in einer täglichen Dosis von 0,005 bis 50 mg, vorzugsweise 0,01 bis 10 mg/kg Körpergewicht verabreicht werden.
Im Gemisch mit geeigneten Hilfsstoffen können die Verbindungen der Formel I zu festen Dosiseinheiten verpresst werden, wie Pillen, Tabletten und Dragées, oder in Kapseln abgefüllt werden.
Mit Hilfe geeigneter Flüssigkeiten können die Verbindungen der Formel I auch als Injektionszubereitungen in Form von Lösungen, Suspensionen oder Emulsionen zubereitet werden.
Zur topischen bzw. lokalen Verabreichung können die Verbindungen auch in eine Crème oder Salbe eingebaut werden.
Bevorzugte erfindungsgemäss hergestellte Verbindungen sind solche der Formel I, bei denen Rx und R2 jeweils ein Wasserstoffatom oder eine Methylgruppe bedeuten und/ oder der Benzolkern durch ein oder zwei Halogenatome substituiert ist und/oder R3 ein Wasserstoffatom, eine niedere aliphatische Acylgruppe (1 bis 8C) oder eine niedere araliphatische Acylgruppe, wie eine Benzoylgruppe, bedeutet.

Verschiedene 4-Hydroxy-benzo-(b)-bicyclo-[3,3, l]-no-nen-11-oximderivate wurden reduziert mit Natrium in Iso-

propanol unter Bildung der entsprechenden primären Amine I. Die Oxime wurden erhalten durch Umsetzung von 4-(endo- oder -exo)-Benzoyloxy-benzo-(b)-bicyclo-[3,3,l]-no-nen-11-on-derivaten mit Hydroxylamin-HCl in Natriumhydroxid und Äthanol. Auf diese Weise wurden die folgenden 5 Verbindungen hergestellt:

4-exo-Hydroxy-8-methoxy-11 -anti-amino-benzo-(b)-bicy-clo-[3,3,l]-nonen-HCl, Schmelzpunkt 256 bis 258 °C;
4-exo-Hydroxy-8,9-dichlor-11 -anti-amino-benzo-(b)-bicy-clo-[3,3,l]-nonen-HCl, Schmelzpunkt 286 bis 300 °C;
4-exo-Hydroxy-ll-anti-amino-benzo-(b)-bicyclo-[3,3,l]-no-nen-HCl, Sublimation > 265 °C.

- 2. Verfahren nach Patentanspruch 1 zur Herstellung von Verbindungen der Formel I, in welcher X und Y Chlor oderBrom bedeuten oder einer dieser Substituenten Chlor oder Brom, der andere Wasserstoff bedeuten.20
- 3. Verfahren nach Patentanspruch 1, dadurch gekennzeichnet, dass man die erhaltene Verbindung in die einzelnen Diasteroisomeren und/oder Enantiomeren auftrennt.2[I]Y 7 6in welcherRt ein Wasserstoffatom, eine (Cj-CJ-Alkyl- oder eine gegebenenfalls substituierte Aryl-(Ci-C6)-alkylgruppe ist, R2 ein Wasserstoffatom bedeutet,R3 eine freie, verätherte oder veresterte Hydroxygruppe ist undX und Y jeweils ein Wasserstoff- oder Halogenatom, eine Hydroxygruppe, (Cj-C^-Alkyl-, (C1-C6)-Alkoxy-, Nitro-, Trifluormethyl- oder Acyloxygruppe bedeuten und deren Salze, dadurch gekennzeichnet, dass man die Iminogruppe einer Verbindung der allgemeinen Formel IV[IV]in welcher X, Y und R3 die oben angegebene Bedeutung haben und R4 ein Wasserstoffatom, eine Hydroxy-, eine Alk-oxy-, eine (C1-Cs)-Alkyl- oder gegebenenfalls eine substituierte Aryl-(C!-C6)-alkylgruppe bedeutet oder ein Salz davon, reduziert und anschliessend die so erhaltene Verbindung gegebenenfalls in ein pharmazeutisch geeignetes Salz umwandelt.2PATENTANSPRÜCHE 1. - 4. Verfahren nach Patentanspruch 1, dadurch gekennzeichnet, dass man eine erhaltene Verbindung der Formel I,25 in welcher X und/oder Y eine Methoxygruppe ist, hydroly-siert zu der entsprechenden Verbindung, in welcher X und/ oder Y eine Hydroxygruppe ist.
- 5. Verfahren nach Patentanspruch 1, dadurch gekennzeichnet, dass man eine erhaltene Verbindung der Formel I,so in welcher R3 eine freie Hydroxygruppe ist, acyliert, gegebenenfalls in Gegenwart einer starken Säure.
- 6. Verfahren zur Herstellung einer Verbindung der Formel I, in welcher R2 in eine Acylgruppe übergeführt ist, und deren Salze, dadurch gekennzeichnet, dass man eine Verbin-35 dung der Formel I nach dem Verfahren gemäss Patentanspruch 1 herstellt und die erhaltene Verbindung acyliert und gegebenenfalls in ein Salz umwandelt.
- 7. Verfahren zur Herstellung einer Verbindung der Formel I, in welcher R2 in eine (Cx-C6)-Alkyl- oder eine gegebe-40 nenfalls substituierte Aryl-CCj-C^-alkylgruppe übergeführt ist, und deren Salze, dadurch gekennzeichnet, dass man eine entsprechende Verbindung der Formel I nach dem Verfahren gemäss Patentanspruch 1 herstellt und die erhaltene Verbindung alkyliert oder aralkyliert und gegebenenfalls in ein45 Salz umwandelt.
- 8. Verfahren zur Herstellung eines N-Oxides der Verbindungen der Formel I, dadurch gekennzeichnet, dass man nach dem Verfahren gemäss Patentanspruch 1 eine Verbindung der Formel I herstellt und diese mit H202 oder einer so Persäure umsetzt.
