Wesentliche Vorteile des erfindungsgemässen Verfahrens io sind der schnelle Reaktionsablauf, die erhöhte Ausbeute und die leichte Zugänglichkeit sowie der niedrige Preis der Oxi-dationsmittel.
Als Ausgangsverbindungen der Formel I kommen Cyclohexenderivate, die allyl-ständige Methylen- oder Hy-15 droxylgruppen und als Substituenten Y einen Polyenylrest tragen, sowie ß-Jonon in Betracht.

Die Erfindung betrifft ein Verfahren zur Einführung einer allyl-ständigen Carbonylgruppe in einen Cyclohexenring durch Oxidation einer allyl-ständigen Methylen- oder Hydroxylgruppe mit Halogen(V)-sauerstoffsäuren oder Salzen dieser Säuren.
Es ist bekannt, dass Retinol, Retinal und der Vitamin-A-säuremethylester mit Mangan(IV)-dioxid in 4-Stellung im Cyclohexenring oxidiert werden können (Tetrahedron Letters, 1972, S. 1823-1825; J. Chem. Soc. 1957, S. 4909-4912). Diese Verfahrensweise erfordert einen bis zu 20fachen Über-schuss an Oxidationsmittel, bezogen auf Substrat, und führt zu stark schwankenden Ausbeuten, die auf die unterschiedliche Aktivität des Mangan(IV)-dioxids, die wiederum eine Funktion seiner Herstellung ist, zurückzuführen sind.
Ausserdem ist bekannt, dass die Oxidation einer allyl-ständigen Methylengruppe im Cyclohexenring mit Metaper-jodat möglich ist. So können beispielsweise Retinal (DE-OS 2 064 495), ß-Carotin oder Retro-dehydro-carotin (DE-PS 1 793 308) mit Natriummetaperjodat in Gegenwart eines Halogens oder eines Oxids eines Metalls aus den Gruppen Va, Via, Vlla und VIII des Periodensystems in 4-Oxo-reti-nal bzw. Canthaxanthin überführt werden. Wesentliche Nachteile dieses Verfahrens sind der hohe Preis des Oxydationsmittels sowie die geringe Ausbeute bei der Herstellung von Canthaxanthin, die zwischen 17 und 38% schwankt.
Es wurde gefunden, dass man eine allyl-ständige Carbonylgruppe in einen Cyclohexenring einführen kann, wenn man ein Cyclohexenderivat der Formel I

Die Ausgangsstoffe werden in Form von Lösungen der Oxidation unterzogen. Bevorzugt verwendet man Lösungen in leicht flüchtigen Lösungsmitteln, die nicht mit Wasser mischbar sind.
Als Verdünnungs- oder Lösungsmittel kommen u.a. chlorierte aliphatische Kohlenwasserstoffe, beispielsweise Chloroform, Methylenchlorid, 1,1-Dichloräthan, 1,2-Di-chloräthan, 1,1,2-Trichloräthan, 1,1,2,2-Tetrachloräthan, 1,2-Dichloräthylen, 1,1,2-Trichloräthylen, aromatische Kohlenwasserstoffe, beispielsweise Benzol, Toluol, Nitro-benzol oder Chlorbenzol, Dialkyläther, beispielsweise Di-äthyläther, Di-n-propyläther, oder Schwefelkohlenstoff in Betracht. Besonders geeignete Lösungsmittel sind Chloroform, Methylenchlorid und 1,2-Dichloräthan. Auch Gemische dieser Verdünnungs- oder Lösungsmittel können verwendet werden.
Als Oxidationsmittel kommen Halogen(V)-sauerstoff-säuren oder Salze dieser Halogen(V)-sauerstoffsäuren in Betracht. Geeignet sind Chlor(V)-säure, Brom(V)-säure, Jod(V)-säure sowie die Alkali-, Erdalkali- oder Ammoniumsalze dieser Säuren, insbesondere Chlor(V)-säure und Brom(V)-säure sowie ihre Alkalisalze. Die Oxidationsmittel werden zweckmässigerweise in Form von wässrigen Lösungen mit einer Konzentration von 5 bis 50 Gew.-% zur Reaktionsmischung gegeben. Das Molverhältnis Oxidationsmittel zu Ausgangssubstanz liegt im allgemeinen im Bereich von 1:1 bis 20:1, vorzugsweise im Bereich von 1:1 bis 10:1 Mol. Ein mehr als 20facher molarer Überschuss an Oxidationsmittel bleibt im allgemeinen ohne Einfluss auf die Umsetzung.
Katalysiert wird die Oxidation durch die Halogene Brom oder Jod oder durch Oxide der Elemente der Gruppen Va, Via, Vlla oder VIII des Periodensystems. Geeignete Katalysatoren sind beispielsweise Vanadiumpentoxid, Molybdän-trioxid, Wolframtrioxid, Mangandioxid, Nickeloxid, Osmiumtetroxid. Bevorzugte Katalysatoren sind Brom, Jod und Osmiumtetroxid. Insbesondere eignet sich Jod als Katalysator.
Der Katalysator wird in Substanz oder in Lösung zugegeben, z.B. in dem Lösungsmittel, das zur Lösung des Ausgangsstoffes verwendet wurde, oder in Wasser. Zweckmässigerweise wird der Katalysator in situ gebildet, so z. B. Jod, das unter den Reaktionsbedingungen aus Natriumjodid gebildet wird. Die Menge an Katalysator beträgt zweckmässigerweise 0,1 bis 25 Gew.-%, bezogen auf Ausgangssubstanz. Grössere Katalysatormengen beeinflussen im allgemeinen die Umsetzung nicht.
Die Oxidation wird bei Temperaturen im Bereich zwischen 0 und 100 °C, vorzugsweise zwischen 10 und 70 °C durchgeführt. Die Umsetzungstemperatur kann in weiten Grenzen variiert werden, wobei die obere Grenze durch die Thermostabilität des Ausgangs- bzw. Endstoffes vorgegeben ist.
Die Umsetzung verläuft in saurem Medium, beginnend in stark saurem Medium bis zum pH-Wert 7. Vorzugsweise arbeitet man bei einem pH-Wert im Bereich von 1 bis 3. Zur Einstellung des gewünschten pH-Wertes werden Säuren, beispielsweise Schwefelsäure, Salzsäure, Essigsäure oder Puffergemische verwendet.
Die Reaktionsdauer beträgt im allgemeinen, je nach den gewählten Bedingungen, 1 bis 24 Stunden.
Um eine Weiteroxidation der Endprodukte durch Einwirkung von Luftsauerstoff zu verhindern, kann die Umsetzung unter Inertgasatmosphäre durchgeführt werden. Unter den Reaktionsbedingungen geeignete Inertgase sind z. B. Argon, Neon, Helium, Kohlendioxid, insbesondere Stickstoff.
Nach einer bevorzugten Ausführungsform des Verfahrens gibt man zu einer Lösung oder einer Aufschlämmung der Ausgangssubstanz in einem unter den Reaktionsbedingungen inerten Verdünnungs- oder Lösungsmittel, das nicht mit Wasser mischbar ist, unter Inertgasatmosphäre eine wässrige Lösung des Oxidationsmittels. Anschliessend stellt man die wässrige Phase mit einer Säure oder einem Puffergemisch auf den gewünschten pH-Wert ein. Man gibt den Katalysator in fester Form oder in Lösung zu und durchmischt die beiden Phasen durch Rühren, bis der vollständige Umsatz durch eine geeignete Analysenmethode, z. B. Dünnschichtchromatographie, angezeigt wird.
Dann trennt man die organische Phase ab und isoliert aus ihr entweder durch Abziehen des Lösungsmittels oder durch Ausfällen rohes Endprodukt. Normalerweise reicht es aus, das Rohmaterial in einem Lösungsmittel, in dem sich das Endprodukt nicht löst, z. B. in Alkoholen, aufzukochen, um ein kristallines Endprodukt zu erhalten. Gegebenenfalls kann das Reaktionsprodukt chromatographisch gereinigt werden.
Die folgenden Beispiele erläutern die Durchführung des erfindungsgemässen Verfahrens.
Beispiel 1
9,6 g (0,05 Mol) ß-Jonon werden in 250 ml Chloroform gelöst. Dazu gibt man 26,5 g (0,25 Mol) Natriumchlorat und 1,125 g Natriumjodid in 100 ml Wasser. Man säuert mit 0,11 ml konz. Schwefelsäure an und rührt 24 Stunden lang bei 45 °C. Dann lässt man abkühlen, trennt die Phasen und wäscht die Chloroformlösung zunächst mit gesättigter Na-triumbicarbonat-Lösung und anschliessend mit Wasser. Nach dem Abziehen des Lösungsmittels bleiben 9,1 g eines dunklen Öls zurück, das 64 Gew.-% 4-Oxo-ß-Jonon enthält. Fraktionierende Destillation im Hochvakuum ergibt 4,1 g eines Öls, das langsam erstarrt. Das aus Methanol/Wasser 1:1 umkristallisierte Produkt vom Schmelzpunkt 51 bis 52 °C erweist sich in allen Eigenschaften identisch mit dem durch Umsetzung von ß-Jonon mit N-Bromsuccinimid und nachfolgender Oxidation mit Mangan(IV)-dioxid (J. Chem. Soc. 1951,1074) erhaltenen Produkt.
Beispiel 2
1 g 4-Hydroxy-ß-apo-8'-carotinsäuremethylester, hergestellt nach US-PS 3 068 257, wird in 30 ml Chloroform gelöst, Man gibt dazu eine Lösung von 1 g Natriumchlorat und 0,1 g Osmiumtetroxid in 10 ml Wasser und rührt bei 10 °C. Man säuert mit 1 Tropfen Essigsäure an und hält die Phasen 24 Stunden lang in Bewegung. Danach trennt man die Phasen, wäscht die organische mit Wasser, trocknet und destilliert das Lösungsmittel unter vermindertem Druck ab. Der teilweise kristalline Rückstand wird mit 50 ml Heptan 60 Minuten lang auf Rückflusstemperatur erwärmt. Nach dem Abkühlen filtriert man nach 24 Stunden 0,61 g 4-Oxo-ß-apo-8'-carotinsäuremethylester.
Beispiel 3
0,1 Mol Vitamin-A-acetat wird in 850 ml Methylenchlorid eingetragen. Man gibt eine Lösung von 79,5 g Natriumchlorat (0,75 Mol) und 1,125 g (7,51 mMoI entsprechend 7,5 Mol.-%, bezogen auf Vitamin-A-acetat) Natriumjodid in 300 ml Wasser zu. Nach Kühlung auf 10 °C säuert man mit 0,015 g konz. Schwefelsäure an und rührt bei dieser Temperatur 18 Stunden lang. Dann trennt man die Phasen, wäscht die organische Lösung mit 400 ml gesättigter Na-triumbicarbonat-Lösung und dann mit Wasser. Nach dem Trocknen und Einengen erhält man ein öliges Rohmaterial, das 57 Gew.-% aus 4-Oxo-Vitamin-A-acetat enthält. Das Keton kann durch Säulenchromatographie rein gewonnen werden.

5 g ß-Apo-12'-carotinal werden in 100 ml 1,2-Dichlor-äthan gelöst. Dazu gibt man eine Lösung von 4 g Natriumchlorat und 0,3 g Natriumjodid in 80 ml Wasser. Man erhitzt auf Rückflusstemperatur, rührt und säuert mit 1 ml wässriger Salzsäure, die 3 mg Chlorwasserstoff gelöst enthält, an. Nach 4 Stunden lässt man auf Raumtemepratur abkühlen. Nach Trennung der Phasen wäscht man die organische Lösung des Carotinoids mit Wasser, dann mit einer verdünnten Natriumcarbonat-Lösung und wieder mit Wasser. Nach dem Trocknen über Magnesiumsulfat wird das Lösungsmittel unter vermindertem Druck abgedampft. Zurück Bleibt ein viskoses rotes Öl, aus dem man durch Aufkochen in 10 ml Isobutanol, Abkühlen und Absaugen nach 24 Stunden 3,45 g kristallines 4-Oxo-ß-apo-12'-carotinal gewinnt. 220 MHz - »H - NMR-Spektrum (CDC13); 5-Werte: 1,25 s (6 H); 1,9 s (6 H); 2,1 d (6 H); 2,551 (2 H); 6,2-7,3 m (9 H); 9,5 s (1 H)

Beispiel 5
10 g ß-Apo-8'-carotinsäureäthylester werden in 250 ml Methylenchlorid gelöst und auf 10 °C gekühlt. Man gibt 8 g Natriumchlorat und 0,2 g Natriumjodid, gelöst in 100 ml Wasser, zu. Nachdem beide Phasen die Temperatur von 10 C erreicht haben, tropft man während 60 Minuten 0,12 ml Essigsäure, gelöst in 20 ml Wasser, zu. Nach lOstündigem Rühren bei 10 °C hat sich die Hälfte des Ausgangsmaterials umgesetzt. Man gibt noch einmal 0,2 g Natriumjodid und 0,12 ml Essigsäure, gelöst in 20 ml Wasser, zu und rührt weitere 12 Stunden bei Raumtemperatur. Man arbeitet wie in Beispiel 2 beschrieben auf und kocht das Rohmaterial 10 h in 30 ml Äthanol unter Rückfiuss. Nach dem Abkühlen filtriert man nach 24 Stunden 6,15 g des 4-Oxo-ß-apo-8'-carotinsäureäthylesters vom Fp = 142-143 °C ab. 220 MHz -1H - NMR-Spektrum (CDC13); 5-Werte: s 1,15 s (6 H); 1,3 s (3 H); 1,95 s (12 H); 2,51 (2 H); 4,2 q (2 H); 6-6,7 m (11 H); 7,25 d (1 H)

Beispiel 6
10 g ß-Apo-4'-carotinsäureäthylester werden wie in Beispiel 4 oxidiert; Man rührt 10 Stunden bei 10 °C und arbeitet auf, ohne Katalysator und Säure nachzusetzen. Nach dem Aufkochen in Äthanol und Abkühlen werden 7,96 g des 4-15 Oxo-ß-apo-4'-carotinsäureäthylesters isoliert.

Beispiel 7
10 g ß-Apo-8'-carotinal werden in 200 ml 1,2-Dichlor-25 äthan gelöst. Dazu gibt man 12 g Natriumchlorat und 0,5 g Natriumbromid in 100 ml Wasser und erhitzt auf Rückflusstemperatur. Während 60 Minuten werden unter Rühren 10 ml wässriger Salzsäure, die 3 mg Chlorwasserstoff gelöst enthält, zugetropft. Man kocht weitere 60 Minuten unter 30 Rückfiuss und kühlt ab. Nach Phasentrennung wird die organische Phase mehrfach mit Wasser gewaschen und dann eingeengt. Man gibt zum öligen Rohprodukt 30 ml Iso-propylalkohol und kocht 2 Stunden lang unter Rückfiuss. Nach 24 Stunden filtriert man 7,14 g kristallines 4-Oxo-ß-35 apo-8'-carotinal vom Fp = 152°C ab.
