65 Die Erfindung betrifft ein neues Verfahren zur Herstellung von 7-(subst.)-Amino-4-subst.-thiomethyl-A3-cephem-4-car-bonsäuren.

1 795 484,2 018 600 und 2 065 621, US-PS 3 516 997 und JA-OS 154 287/75, berichtet über die Reaktion eines Thiols oder dessen Salz mit der Acetoxygruppe in 3-Stellung einer 7-Aminocephalosporansäure bzw. den an der Carboxylgruppe derivatisierten Abkömmlingen oder Salzen dieser Säure, wobei die Acetoxygruppe in der 3-Stellung verändert wird. Nach den erwähnten Publikationen ist es nicht wünschenswert, die besagte Reaktion in einem wasserfreien organischen Lösungsmittel durchzuführen, und es wird bevorzugt, die Reaktion bei pH 6 bis 7 in Wasser oder einer Mischung von Wasser und einem organischen Lösungsmittel ablaufen zu lassen. Das erhaltene Produkt ist jedoch selbst unter den genannten, bevorzugten Reaktionsbedingungen extrem unrein, und die Ausbeuten liegen bei 30 bis 50%. Die Nacharbeitung der genannten Reaktion durch die Erfinder hat bestätigt, dass die Ausbeute höchstens 30 bis 50% beträgt und dass das Reaktionsprodukt mit dem Ausgangsmaterial, 7-Aminocephalosporansäure, vermischt ist. Andererseits wird in der US-PS 3 840 531, den JA-OSen 295/74 und 10 077/73, der DE-OS 2 332 045, der JA-PS 13 023/71 u.a. ein Verfahren zur schonenenden Umwandlung in der 3-Stellung beschrieben, das darin besteht, dass man als Ausgangsmaterial entweder Cephalosporin C bzw. eines seiner Derivate oder eine 7-Aminocephalosporansäure bzw. deren Salz verwendet, wobei die Aminogruppe in der 7-Stellung mit einer Acylgruppe, wie Formyl, niedrig-Alkanoyl oder dergl., geschützt ist. Selbst bei Anwendung dieser Methode wird gemäss den obengenannten Publikationen die Reaktion vorzugsweise in Wasser oder einer Mischung von Wasser und einem organischen Lösungsmittel in der Nähe des Neutralpunkts durchgeführt.
Bezüglich eines Verfahrens, bei dem ein Cephalosporin C-Derivat als Ausgangsmaterial verwendet wird, beschreiben z.B. GB-PS 1 400 804 und JA-OS 95 088/76, dass die Veränderung in der 3-Stellung in Wasser oder einer Mischung von Wasser und einem organischen Lösungsmittel in Gegenwart eines Halogenids oder anorganischen Salzes eines Metalls der Gruppe I oder II des Periodensystems, wie KJ, NaJ-CaJ2, BaJ2, NaCl, NH-tCl, BaCh, MgCk o.ä., durchgeführt werden kann. Das Verfahren, bei dem eine acylierte Cephalosporan-säure, Cephalosporin C oder eines seiner Derivate als Ausgangsmaterial verwendet wird, führt bei der Reaktion zu Schwierigkeiten, weil die Aminogruppe in der 7-Stellung acy-liert werden muss, oder weil ein acyliertes Ausgangsmaterial verwendet werden muss und die Acylgruppe durch Iminoha-logenierung, Iminoätherbildung, Hydrolyse o.ä. nach der Veränderung in der 3-Stellung wieder entfernt werden muss. Bei der genannten Reaktionsfolge wird die Veränderung in der 3-Stellung mit einem Thiol oder dessen Salz in einer Mischung von Wasser und einem organischen Lösungsmittel unter den oben erwähnten Bedingungen durchgeführt, und es werden im allgemeinen Ausbeuten von 60 bis 80% erhalten.
Vor dem oben erwähnten Hintergrund haben die Erfinder intensive Untersuchungen mit dem Ziel angestellt, ein Verfahren zur Veränderung der Gruppe in der 3-Stellung durch ein Thiol oder dessen Salz zu entwickeln, das mit hoher Ausbeute arbeitet und leicht industriell anwendbar ist. Es wurde überraschenderweise gefunden, dass man ein in dieser Hinsicht befriedigendes Resultat erreicht, wenn man die Reaktion in einem organischen Lösungsmittel in Gegenwart einer Protonensäure oder Lewissäure oder einer Komplexverbindung von Lewissäure, mit Ausnahme von Bortrifluorid oder seiner Komplexverbindungen, durchführt.
Aufgabe der vorliegenden Erfindung ist es, ein Verfahren zur Herstellung einer 7-(subst.)-Amino-3-subst.-thiomethyl-/ J-cephem-4-carbonsäure bzw. eines an der Carboxylgruppe derivatisierten Abkömmlings oder eines Salzes dieser Säure zu schaffen, das bei einfacher industrieller Durchführung die
Produkte in hoher Ausbeute und grosser Reinheit liefert. Diese Verbindungen sind wichtig als Zwischenprodukte bei der Synthese von Cephalosporinen.
Die erfindungsgemässe Aufgabe wird dadurch gelöst, dass s man ein Verfahren zur Herstellung einer 7-(subst.)-Amino-3-subst.-thiomethyl-A3-cephem-4-carbonsäure bzw. eines an der Carboxylgruppe derivatisierten Abkömmlings oder eines Salzes dieser Säure schafft;

Als organische Reste, für die R3, R4, R5, R6 und R7 stehen, die nicht an der Reaktion teilnehmen, können solche verwendet werden, die auf diesem Gebiet herkömmlich angewendet werden. Beispiele dafür sind unsubstituierte oder substituierte aliphatische Reste, alicyclische Reste, aromatische Reste, araliphatische Reste, heterocyclische Reste, Acyl-gruppen u.ä.; insbesondere werden beispielsweise die folgenden Gruppen verwendet:
1. Aliphatische Reste: Alkylgruppen, z.B. Methyl, Äthyl, Propyl, Butyl, Isobutyl, Pentyl u.ä.; und Alkenylgruppen, z.B. Vinyl, Propenyl, Butenyl u.ä.
2. Alicyclische Reste: Cycloalkylgruppen, z.B. Cyclo-pentyl, Cyclohexyl, Cycloheptyl u.ä.; und Cycloalkenyl-gruppen, z.B. Cyclopentenyl, Cyclohexenyl u.ä.
3. Aromatische Reste: Arylgruppen, z.B. Phenyl, Naph-
thyl u.ä.
4. Araliphatische Reste: Aralkylgruppen, z.B. Benzyl, Phenäthyl u.ä.
5. Heterocyclische Reste: heterocyclische Gruppen mit einem oder mehreren Heteroatomen (Sauerstoff, Stickstoff und Schwefel) in jeglicher Kombination und Position im Molekül, z.B. Pyrrolidyl, Piperazinyl, Furyl, Thienyl, Pyr-rolyl, Pyrazolyl, Oxazolyl, Thiazolyl, Pyridyl, Imidazolyl, Chinolyl, Benzothiazolyl, Oxadiazolyl, Thiadiazolyl, Tria-zolyl, Tetrazolyl u.ä.
6. Acylgruppen: Acylgruppen, die von organischen Carbonsäuren abgeleitet sind. Solche organischen Carbonsäuren sind z.B. aliphatische Carbonsäuren; alicyclische Carbonsäuren; alicycloaliphatische Carbonsäuren; araliphatische Carbonsäuren, aromatisch-oxyaliphatische Carbonsäuren, aromatisch-thio-aliphatische Carbonsäuren, heterocyclisch-ringsubstituiert-aliphatische Carbonsäuren, heterocyclisch-oxy-aliphatische Carbonsäuren und heterocyclisch-thio-ali-phatische Carbonsäuren, bei denen ein aromatischer Rest oder eine heterocyclische Gruppe über ein Sauerstoff- oder Schwefelatom oder direkt an eine aliphatische Carbonsäure gebunden ist; organische Carbonsäuren, in denen ein aromatischer Ring, eine aliphatische Gruppe oder eine alicyclische Gruppe durch ein Sauerstoff-, Stickstoff- oder Schwefelatom direkt an die Carbonylgruppe gebunden ist; aromatische Carbonsäuren; und heterocyclische Carbonsäuren.
Beispiele für die oben erwähnten aliphatischen Carbonsäuren sind Ameisensäure, Essigsäure, Propionsäure, Buttersäure, Isobuttersäure, Pentansäure, Methoxyessigsäure, Me-thylthioessigsäure, Acrylsäure, Crotonsäure usw. Die oben erwähnten alicyclischen Carbonsäuren sind z.B. Cyclohexan-säuren usw., und die alicycloaliphatischen Carbonsäuren sind z.B. Cyclopentan-essigsäure, Cyclohexan-essigsäure, Cyclohexan-propionsäure, Cyclohexadien-essigsäure usw. Aromatische Reste in den obengenannten organischen Carbonsäuren sind u.a. Phenyl, Naphthyl usw.; und heterocyclische Reste sind z.B. heterocyclische Verbindungen, die wenigstens ein Heteroatom im Ring enthalten, wie Furan, Thiophen, Pyrrol, Pyrazol, Imidazol, Triazol, Thiazol, Iso-thiazol, Oxazol, Isooxazol, Thiadiazol, Oxadiazol, Thia-triazol, Oxatriazol, Tetrazol, Benzoxazol, Benzofuran usw.
Jede der Gruppen, aus denen die obengenannten organischen Carbonsäuren aufgebaut sind, kann weitere Substituenten, wie z.B. ein Halogenatom, eine Hydroxylgruppe, eine geschützte Hydroxylgruppe, eine Ci-s-Alkylgruppe, eine C1-5-Alkoxygruppe, eine C1-4-Acylgruppe, eine Nitrogruppe, eine Aminogruppe, eine geschützte Aminogruppe, eine Mercap-togruppe, eine geschützte Mercaptogruppe, eine Carboxylgruppe, eine geschützte Carboxylgruppe usw., tragen.
Als Schutzgruppen für die oben erwähnten geschützten Hydroxyl-, geschützten Amino-, geschützten Mercapto- und geschützten Carboxylgruppen können diejenigen verwendet werden, die weiter unten im Zusammenhang mit den Substituenten in der R8-Gruppe genannt werden.
R3, R4 und Rä können gleich oder verschieden sein, und bedeuten vorzugsweise Wasserstoff, Ci-s-Alkyl, C2-4-Alkenyl, C5-7-Cycloalkyl, Cs-7-Cycloalkenyl, Aryl, Aralkyl, heterocyclische Verbindungen, die entweder nur O, N oder S enthalten oder diese Elemente in jeder möglichen Kombination an jeder möglichen Position enthalten, oder Acyl; und R6 und R7, die gleich oder verschieden sein können, bedeuten vorzugsweise Wasserstoff, Ci-s-Alkyl, Cj-4-Alkenyl, Cs-7-CycIo-alkyl, C5-7-Cycloalkenyl, Aryl, Aralkyl, heterocyclische Verbindungen, die entweder nur O, N oder S enthalten oder diese Elemente in jeder möglichen Kombination in irgendeiner der möglichen Positionen enthalten, oder Acylgruppen.

Der Begriff «Salz» in dem Ausdruck «eine Verbindung der allgemeinen Formel (I) oder (II) bzw. ein an der Carboxylgruppe derivatisierter Abkömmling oder ein Salz dieser Verbindung», wie er in der Beschreibung und in den Ansprüchen verwendet wird, schliesst sowohl Salze an der sauren Gruppe 60 (z.B. Carboxylgruppe) als auch Salze der basischen Gruppe (z.B. Aminogruppe) ein. Salze der sauren Gruppe sind z.B.
Salze mit Alkalimetallen, wie Natrium, Kalium usw.; Salze mit Erdalkalimetallen, wie Calcium, Magnesium usw.; Ammoniumsalze; Salze mit stickstoffhaltigen organischen 65 Basen, wieTriäthylamin, Diäthylamin, Pyridin, N-Methyl-piperidin, N-Methylmorpholin, N,N-Dimethylanilin usw.
Salze der basischen Gruppe sind z.B. Salze mit Mineralsäuren, wie Chlorwasserstoffsäure, Schwefelsäure usw., Salze mit organischen Säuren, wie Oxalsäure, Ameisensäure, Trichloressigsäure, Trifluoressigsäure usw.; und Salze mit Sulfonsäuren, wie Methansulfonsäure, Toluolsulfonsäure, Naphthalinsulfonsäure usw. Die Salze der Thiolverbindungen gemäss der allgemeinen Formel (III) können, abhängig von dem Typ von R8, in der Form basischer Salze oder saurer Salze vorliegen, und es sind beide Arten eingeschlossen. Bezüglich der Beispiele für das Salz wird auf die bei den allgemeinen Formeln (I) und (II) oben gegebene Erläuterung verwiesen. Als Material zur Bildung der Salze der Thiolverbindung können diejenigen Materialien verwendet werden, die zur Bildung der Salze der Verbindungen gemäss den allgemeinen Formeln (I) und (II) verwendet werden.
Als Protonensäuren kommen z.B. Pyrophosphorsäure, Pyroschwefelsäure, Schwefelsäuren, Sulfonsäuren und Supersäuren in Frage. Der hier verwendete Begriff «Super-säure» bedeutet Säuren, die stärker als 100%ige Schwefelsäure sind, und schliesst einen Teil der Schwefelsäuren und
Sulfonsäuren ein. Bei dem Begriff «Schwefelsäuren» sind im einzelnen eingeschlossen Schwefelsäure, Chlorschwefelsäure, Fluorschwefelsäure usw., und der Begriff «Sulfonsäuren» schliesst Alkyl-(mono- oder di)-sulfonsäuren, wie Methansulfonsäure, Trifluormethansulfonsäure usw., und Aryl-(mono-, di- oder tri)-sulfonsäuren, wie Benzolsulfon-säure, Naphthalinsulfonsäure, p-Toluolsulfonsäureusw., ein. Der Begriff «Supersäuren» schliesst Perchlorsäure, FSO^H-SbFs (magic acid), FSOjH-AsFs, CFaSOjH-SbFs, H2SO4-SO3 usw. ein. Bei Lewissäuren, mit Ausnahme von Bortrifluorid, sind z.B. Zinkhalogenide und Zinnhalogenide und insbesondere Zinkchlorid, Zinkbromid, Zinn(IV)-chlorid und Zinn(IV)-bromid usw. eingeschlossen. Die Komplexverbindungen von Lewissäuren, mit Ausnahme der Komplexverbindungen von Bortrifluorid, umfassen Komplexsalze der oben erwähnten Lewissäuren, mit Ausnahme von Bortrifluorid, mit Dialkyläthern, wie Diäthyläther, Di-n-propyläther, Di-n-butyläther usw.; Komplexsalze der oben erwähnten Lewissäuren mit Aminen, wie Äthylamin, n-Pro-pylamin, n-Butylamin, Triäthanolamin, Dimethylformamid usw.; Komplexsalze der oben erwähnten Lewissäuren mit Fettsäuren, wie Essigsäure, Propionsäure usw.; Komplexsalze der oben erwähnten Lewissäuren mit Nitrilen, wie Ace-tonitril, Propionitril usw.; Komplexsalze der oben erwähnten Lewissäuren mit Carbonsäureestern, wie Methylformiat, Äthylformiat, Äthylacetat usw.; und Komplexsalze der oben erwähnten Lewissäuren mit Phenolen, wie Phenol, (1- oder 2-)Naphthol usw. Die obengenannten Sulfonsäuren können durch Halogenatome, wie Fluor, Chlor, Brom usw., substituiert sein sowie durch Carboxylgruppen, Sulfogruppen, Nitro-gruppen, niedrig-Alkylgruppen, wie Methyl, Äthyl usw.,

Als organische Lösungsmittel können bei dem erfindungs-gemässen Verfahren alle organischen Lösungsmittel verwendet werden, die die Reaktion nicht nachteilig beeinflussen, vorzugsweise Nitrile, Nitroalkane, organische Carbonsäuren, Ketone, Äther und Sulfolane. Diese Lösungsmittel können als Mischung von zwei oder mehr Komponenten eingesetzt werden. Die oben erwähnten Nitrile umfassen z.B. aliphatische Nitrile, aliphatische Dinitrile, aromatische Nitrile und heterocyclische Nitrile, wie Acetonitril, Propionitril, Butyronitril, Isobutyronitril, Valeronitril, Iso-valeronitril, Capronitril, Enanthonitril, Caprylonitril, Pelar-gononitril, Caprinitril, Crotononitril, Lauronitril, Palmitoni-tril, Stearonitril, Acrylonitril, Malononitril, Succinonitril, Glutaronitril, Adiponitril, Benzonitril, Tolunitril, cyaniertes Benzyl, Cinnamonitril, Naphthonitril, Cyanothiophen usw.; die Nitroalkane umfassen Nitromethan, Nitroäthan, Nitro-propan, Nitrobutan, Nitropentan, Nitrohexan, Nitroheptan,

Die bei der Erfindung verwendeten organischen Lösungsmittel können mit der Lewissäure, mit Ausnahme von Bortrifluorid, einen Komplex bilden, und dieser Komplex des organischen Lösungsmittels mit einer Lewissäure, ausgenommen Bortrifluorid, wird ebenfalls als organisches Lösungsmittel erfin-dungsgemäss verwendet.
Die Protonensäure oder die Lewissäure oder die Komplexverbindung einer Lewissäure, ausgenommen Bortrifluorid und dessen Komplexverbindungen, können in einer Menge von wenigsten 1 Mol/Mol der Verbindung gemäss der allgemeinen Formel (II) bzw. eines an der Carboxylgruppe derivatisierten Abkömmlings oder eines Salzes dieser Verbindung verwendet werden, und bevorzugt sind wenigstens 2 Mol, insbesondere 2 bis 10 Mol, pro Mol der letztgenannten Verbindung. Falls die Komplexverbindung eingesetzt wird, kann sie ebenfalls als Lösungsmittel verwendet werden, und eine Mischung von zwei oder mehr Komplexverbindungen kann ebenfalls eingesetzt werden. Es ist im allgemeinen zur Kontrolle der Reaktionsgeschwindigkeit wünschenswert, die Menge der Protonensäure oder der Lewissäure oder der Komplexverbindung einer Lewissäure, mit Ausnahme von Bortrifluorid und dessen Komplexverbindungen, in Abhängigkeit von der Art des Lösungsmittels und der verwendeten Thiolverbindung oder dessen Salzes zu variieren. Die Thiol-verbindung gemäss der allgemeinen Formel (III) oder deren Salz wird im allgemeinen in einer Menge von 1 Mol/Mol der Verbindung gemäss der allgemeinen Formel (II) bzw. des an der Carboxylgruppe derivatisierten Abkömmlings oder des Salzes dieser Verbindung eingesetzt, vorzugsweise in einer Menge von 1 bis 1,5 Mol/Mol der letzteren. Falls als Ausgangsmaterial die Verbindung mit >Y = >S — O verwendet wird, wird die Thiolverbindung oder deren Salz vorzugsweise in einer Menge von 2 bis 3 Mol/Mol des Ausgangsmaterials eingesetzt.
Obwohl die Reaktionstemperatur keiner ausdrücklichen Beschränkung unterworfen ist, wird die Reaktion im allgemeinen bei einer Temperatur von -20° bis 80°C durchgeführt, und die Reaktionszeit beträgt im allgemeinen einige Minuten bis zu mehreren Stunden.
Bei dem erfindungsgemässen Verfahren können die folgenden Dehydrationsmittel dem Reaktionssystem zugesetzt sein: Phosphorverbindungen, wie Phosphorpentachlorid, Polyphosphorsäure, Phosphorpentoxid, Phosphortrichlorid, Phosphoroxylchlorid usw.; organische Silylverbindungen,
wie N,0-Bis-(trimethylsilyl)-acetamid, Trimethylsilylacet-amid, Trimethylchlorsilan, Dimethyldichlorsilan usw.; organische Säurechloride, wie Acetylchlorid, p-Toluolsulfonyl-chlorid usw.; Säureanhydride, wie Essigsäureanhydrid, Tri-fluoressigsäureanhydrid usw.; und anorganische Trocknungsmittel, wie wasserfreies Magnesiumsulfat, wasserfreies Calciumchlorid, wasserfreies Calciumsulfat, Molekularsiebe, Calciumcarbid usw.
Die oben erwähnten Reaktionsbedingungen haben keinen limitierenden Charakter und können, abhängig von der Art der Reaktanten und Lösungsmittel, entsprechend variiert werden, um die erfindungsgemässen Verbindungen zu erhalten.
Die Schutzgruppe

in der allgemeinen Formel (I) und die Schutzgruppe des an der Carboxylgruppe derivatisierten Abkömmlings der Verbindung gemäss der allgemeinen Formel (I) kann im allgemeinen durch Hydrolyse oder eine andere herkömmliche Methode entfernt werden. Dabei werden die geschützten Gruppen in eine Aminogruppe bzw. eine Carboxylgruppe umgewandelt. Bei der Verwendung von einigen Gruppen der Formeln

oder falls eine gewisse Nachbehandlung durchgeführt wird, kann indessen die Schutzgruppe der Aminogruppe leicht während der Reaktion entfernt werden, und man erhält eine Verbindung der allgemeinen Formel (I), bei der R2 für eine Aminogruppe steht. Falls die Carboxylgruppe der Verbindung gemäss der allgemeinen Formel (I) durch gewisse Schutzgruppen geschützt ist, oder falls eine bestimmte Nachbehandlung durchgeführt wird, kann die Schutzgruppe leicht bei der Behandlung entfernt werden, die zur Überführung der geschützten Carboxylgruppe in eine freie Carboxylgruppe durchgeführt wird. Man erhält dabei eine Verbindung gemäss der allgemeinen Formel (I). Falls R3, R4, R5, R6 und R7 organische Reste sind, die nicht an der Reaktion teilnehmen und die eine geschützte Hydroxyl-, Amino-, Mercapto- oder Carboxylgruppe als Substituenten tragen, können diese Gruppen in die gewünschten Substituenten dadurch umgewandelt werden, dass man die resultierende Verbindung auf herkömmliche Weise einer Abspaltungsreaktion unterwirft. Die oben erwähnte Schutzgruppen-Abspaltungsreaktion kann durchgeführt werden, ohne dass man das resultierende Produkt isoliert. Die erfindungsgemäss angestrebten Verbindungen mit der allgemeinen Formel (I), die auf diese Art erhalten werden, können nach herkömmlichen Verfahren isoliert werden.
Die angestrebten Verbindungen gemäss der allgemeinen Formel (I) können direkt als Ausgangsmaterial für Acylie-rungsreaktionen verwendet werden, man kann sie jedoch,
falls notwendig, auf eine herkömmliche Art reinigen und erhält dabei in hohen Ausbeuten hochreine 7-(subst.)-Amino-3-subst.-thiomethyl-A3-cephem-4-carbonsäure.
Das erfindungsgemässe Verfahren wird durch die folgenden Beispiele näher erläutert.
Beispiel 1
1. In 27 ml Essigsäure werden 2,72 g 7-Aminocephalospo-

ransäure suspendiert, und 1,16 g 5-Mercapto-l-methyl-1H-tetrazol und 5,76 g Methansulfonsäure werden zu dieser Suspension zugegeben, um eine Lösung zu erhalten. Diese Lösung lässt man bei 50°C während 2,5 h reagieren. Nach Beendigung der Reaktion wird die Reaktionsmischung abgekühlt und unter Eiskühlung allmählich zu 27 ml Wasser gegeben. Danach wird die Mischung mit wässrigem Ammoniak (28 Gew.%) auf pH 4,0 eingestellt. Die dabei ausgefallenen Kristalle werden durch Filtration gesammelt, mit 5 ml Wasser und 5 ml Aceton in dieser Reihenfolge gewaschen und danach getrocknet. Man erhält 2,70 g (Ausbeute 82,3%) 7-Amino-3-[5-(l-methyl-l,2,3,4-tetrazolyl)-thiomethyl]-A3-cephem-4-carbonsäure, Fp 224 bis 226°C (Zers.).

Bei dem obigen Verfahren (1) wird nach Beendigung der Reaktion eine wässrige Ammoniumacetatlösung (0,77 g Ammoniumacetat/4 ml Wasser) und 3,3 ml 12N Chlorwasserstoffsäure zu der Reaktionsmischung gegeben. Die resultierende Mischung wird 2 h bei 15°C gerührt, danach werden die auf diese Weise abgeschiedenen Kristalle durch Filtration gesammelt, mit zwei 5 ml-Portionen Aceton gewaschen und dann getrocknet. Man erhält 2,44 g (Ausbeute 67,0%) 7-Amino-3-[5-(l-methyl-l,2,3,4-tetrazolyl)-thiomethyl]-A3-cephem-4-carbonsäure-hydrochlorid, Fp. 184 bis 186°C (Zers.).

Beispiel 2
In 80 ml einer 0,1N Perchlorsäurelösung in Essigsäure werden 0,54 g 7-Aminocephalosporansäure und 0,25 g 5-Mercapto-l-methyl-lH-tetrazol gelöst, und diese Lösung wird 2,5 h bei 50 bis 55°C umgesetzt. Nach Beendigung der Reaktion wird das Lösungsmittel durch Destillation unter verringertem Druck entfernt. Der Rückstand wird in 10 ml Wasser aufgelöst.
Zu der so erhaltenen, wässrigen Lösung wird unter Eiskühlung tropfenweise konz. wässriger Ammoniak zugegeben und danach der pH der Lösung auf 3,5 eingestellt, woraufhin die Lösung 15 min gerührt wird. Die so präzipitierten Kristalle werden durch Filtration gesammelt, mit 2 ml Wasser und 3 ml Methanol in dieser Reihenfolge gewaschen und dann getrocknet. Man erhält 0,53 g (Ausbeute 80,8%) 7-Amino-3-[5-(l-methyl-l,2,3,4-tetrazolyl)-thiomethyl]-A3-cephem-4-carbonsäure, Fp. 224 bis 226°C (Zers.). Die IR- und NMR-Spektren des Produktes sind mit denen einer Standardprobe identisch.
Beispiel 3
In 13,5 ml Essigsäure werden 1,36 g 7-Aminocephalospo-ransäure und 0,58 g 5-Mercapto-l -methyl-1 H-tetrazol suspendiert und 3,9 g wasserfreies Zinn(IV)-chlorid werden zu der Suspension zur Bildung einer Lösung zugegeben.
Diese Lösung wird 1,5 h bei 50°C umgesetzt, und anschliessend wird das Lösungsmittel durch Destillation bei vermindertem Druck entfernt. Zu dem Rückstand gibt man 10 ml Wasser und weiterhin wässriges Ammoniak (28 Gew.%) unter Eiskühlung zur Einstellung des pH der Lösung auf 7,5. Die so präzipitierten Kristalle werden durch Filtration gesammelt, mit 5 ml Wasser und 5 ml Aceton in dieser Reihenfolge gewaschen und dann getrocknet. Man erhält 1,28 g (Ausbeute 78,0%) 7-Amino-3-[5-(l-methyl-1,2,3,4-tetrazolyl)-thio-methyl]-A3-cephem-4-carbonsäure.
Beispiel 4
in 3 ml Essigsäure werden 0,27 g 7-Aminocephalosporan-säure und 0,12 g 5-Mercapto-1 -methyl-1 H-tetrazol suspendiert und 1,36 g wasserfreies Zinkchlorid werden zur Bildung einer Lösung zu der Suspension zugegeben. Diese Lösung wird 4 h bei 50°C umgesetzt und dann mit 3 ml Wasser verdünnt. Der pH der Lösung wird auf 3,8 eingestellt, und zwar mit wässrigem Ammoniak (28 Gew.%) unter Eiskühlung. Die dabei präzipitierten Kristalle werden durch Filtration gesammelt, mit 2 ml 0,1N Chlorwasserstoffsäure, 2 ml Wasser und 1 ml Aceton in dieser Reihenfolge gewaschen und dann getrocknet. Man erhält 0,26 g (Ausbeute 79,2%) 7-Amino-3-[5-(l-methyl-l,2,3,4-tetrazolyl)-thiomethyl]-A3-cephem-4-carbonsäure. IR, NMR und Schmelzpunkt des Produktes sind mit denen einer Standardprobe identisch.
Falls 3,2 g Zinkbromid anstelle des Zinkchlorids verwendet werden, ist die Ausbeute 77,3%.
Beispiel 5
In 15 ml Essigsäure werden 1,36 g 7-Aminocephalosporan-säure und 0,58 g 5-Mercapto-l-methyl-l H-tetrazol suspendiert, und zu der Suspension werden 0,48 g Methansulfonsäure zugefügt, um eine Lösung zu bilden. Zu dieser Lösung gibt man 6,80 g wasserfreies Zinkchlorid und setzt die erhaltene Mischung 4 h bei 50°C um. Nach Beendigung der Reaktion wird die Reaktionsmischung mit 15 ml Wasser verdünnt, woraufhin der pH unter Eiskühlung mit wässrigem Ammoniak (28 Gew.%) auf 3,8 eingestellt wird. Die dabei präzipitierten Kristalle werden durch Filtration abgetrennt, mit 10 ml 0,1 N Chlorwasserstoffsäure, 10 ml Wasser und 5 ml Aceton in dieser Reihenfolge gewaschen und dann getrocknet. Man erhält 1,35 g (Ausbeute 82,3%) 7-Amino-3-[5-( 1 -methyl-1,2,3,4-tetrazolyl)-thiomethyl]- A3-cephem-4-carbonsäure. IR, NMR und Schmelzpunkt des Produktes sind mit denen einer Standardprobe identisch.

In 5 ml Essigsäure werden 0,305 g p-Toluolsulfonsäuresalz von DiphenyImethyl-7-aminocephalosporanat und 0,058 g 5-Mercapto-l-methyl-1 H-tetrazol suspendiert. Man gibt 0,45 g Trifluormethansulfonsäure zu der Suspension, um eine Lösung zu bilden. Diese Lösung wird 1,5 h bei 50°C umgesetzt, danach wird das Lösungsmittel durch Destillation unter vermindertem Druck entfernt. Zu dem erhaltenen Rückstand gibt man 2,5 ml Wasser und 2,5 ml Aceton und rührt die resultierende Mischung 30 min unter Eiskühlung. Daraufhin wird durch Zugabe von wässrigem Ammoniak (28 Gew.%) der pH der Mischung auf 4,0 eingestellt. Die dabei präzipitierten Kristalle werden durch Filtration abgetrennt, mit 3 ml Wasser und 3 ml Aceton in dieser Reihenfolge gewaschen und getrocknet. Man erhält 0,127 g (Ausbeute 77,2%) 7-Amino-3-[5-( 1 -methyl-1,2,3,4-tetrazolyl)-thiomethyl]-A3-cephem-4-carbonsäure. IR, NMR und Schmelzpunkt des Produktes sind mit denen einer Standardprobe identisch.
Beispiel 7
In 8 ml Essigsäure werden 0,796 g Natrium-7-(2-hydroxy-benzylidenamino)-cephalosporanat und 0,232 g 5-Mercapto-1-methyl-1 H-tetrazol suspendiert. 1,80 g Trifluormethansulfonsäure werden zu der Suspension zugegeben, um eine Lösung zu bilden. Diese Lösung wird 3 h bei Zimmertemperatur umgesetzt. Unter Eiskühlung werden 1 ml Wasser und 1,5 ml 12N Chlorwasserstoffsäure zu der Reaktionsmischung zugesetzt und die resultierende Mischung wird 2 h bei Zimmertemperatur gerührt. Die dabei präzipitierten Kristalle werden durch Filtration gesammelt, mit zwei 1-ml-Portionen Essigsäure und zwei 3-ml-Portionen Aceton in dieser Reihenfolge gewaschen und dann getrocknet. Man erhält 0,503 g (Ausbeute 69,0%) 7-Amino-3-[5-(l-methyl-l,2,3,4-tetrazolyl)-thiomethyl]-A3-cephem-4-carbonsäure-hydrochlorid. IR, NMR und Schmelzpunkt des Produktes sind mit denen einer Standardprobe identisch.
Beispiel 8
1. In 6,8 ml Acetonitril werden allmählich 0,58 g Trifluormethansulfonsäure eingetropft, zu der Lösung gibt man 0,58 g 5-Mercapto-l-methyl-1 H-tetrazol und 1,36 g 7-Aminocephalosporansäure in dieser Reihenfolge, wobei man eine Lösung erhält. Die Lösung wird auf 30°C erhitzt und bei dieser Temperatur während 60 min umgesetzt. Nach dieser Zeit kühlt man die Reaktionsmischung mit Eis ab und fügt allmählich 5,7 ml Wasser zu. Der pH der Lösung wird mit wässrigem Ammoniak (28 Gew.%) auf 3,9 eingestellt, und die Lösung wird bei der gleichen Temperatur 2 h gerührt. Die dabei ausgefallenen Kristalle werden durch Filtration abgetrennt, mit zwei 1 ml-Portionen Wasser und zwei 1 ml-Portionen Aceton in dieser Reihenfolge gewaschen und dann getrocknet. Man erhält 1,50 g (Ausbeute 91,5%) 7-Amino-3-[5-( 1 -methyl-1,2,3,4-tetrazolyI)-thiomethyl[-A3-cephem-4-carbonsäure. IR, NMR und Schmelzpunkt des Produktes sind mit denen einer Standardprobe identisch.
2. Die gleiche Reaktion, wie oben, wird durchgeführt und unter Eiskühlung zu der Reaktionsmischung tropfenweise 0,84 ml einer 12N Chlorwasserstoffsäure und 0,68 ml Wasser in dieser Reihenfolge gegeben, woraufhin die resultierende Mischung 3 h gerührt wird. Die dabei ausgefallenen Kristalle werden durch Filtration abgetrennt, mit zwei 2-ml-Portionen Acetonitril und zwei 3-ml-Portionen Aceton in dieser Reihenfolge gewaschen und dann getrocknet. Man erhält 1,64 g (Ausbeute 90,0%) 7-Amino-3-[5-(l-methyl-l,2,3,4-tetrazolyl)-thiomethyl]-A3-cephem-4-carbonsäure-hydrochlorid. IR, NMR und Schmelzpunkt des Produktes sind mit denen einer Standardprobe identisch.
3. Wenn Propionitril anstelle von Acetonitril in der obigen Reaktion (1) eingesetzt wird, ist die Ausbeute 88,4%.
4. Wenn Sulfolan anstelle von Acetonitril in der obigen Reaktion (1) eingesetzt wird, ist die Ausbeute 89,6%.
5. Wenn Nitromethan anstelle von Acetonitril in der obigen Reaktion (1) verwendet wird, ist die Ausbeute 84,3%
Beispiel 9
In 27 ml Acetonitril werden 2,72 g 7-Aminocephalospo-ransäure und 1,16 g 5-Mercapto-l-methyl-1 H-tetrazol suspendiert. 9,75 g konz. Schwefelsäure werden allmählich zugegeben, und zwar unter Eiskühlung, und es bildet sich eine Lösung. Diese Lösung wird 1 h bei 30°C umgesetzt, und die Reaktionsmischung wird auf 5°C abgekühlt und dann allmählich unter Eiskühlung mit 60 ml Wasser versetzt. Der pH der Lösung wird mit wässrigem Ammoniak (28 Gew.%) auf 3,7 eingestellt und weitere 30 ml Wasser werden zugegeben. Die resultierende Lösung wird 1 h bei der gleichen Temperatur gerührt. Die dabei ausgefallenen Kristalle werden durch Filtration entfernt, mit zwei 15-ml-Portionen Wasser und drei 10-ml-Portionen Aceton in dieser Reihenfolge gewaschen und dann getrocknet. Man erhält 2,93 g (Ausbeute 89,3%) 7-Amino-3-[5-(l-methyl-l,2,3,4-tetrazolyl)-thio-methyl]-A3-cephem-4-carbonsäure. IR, NMR und Schmelzpunkt des Produktes sind mit denen der Standardprobe identisch.
Beispiel 10
In 6,8 ml Acetonitril werden 1,36 g 7-Aminocephalosporansäure und 0,60 g 5-Mercapto-1 -methyl-1 H-tetrazol suspendiert, und 3,50 g konz. Schwefelsäure werden allmählich unter Eiskühlung zugesetzt, um eine Lösung zu bilden. Diese Lösung wird 75 min bei 30°C umgesetzt.
Danach werden 1,7 ml 12N Chlorwasserstoffsäure und 2,0 ml Wasser in dieser Reihenfolge eingetropft, während die Temperatur auf dem gleichen Wert gehalten wird. Die erhaltene Mischung wird auf 15°C abgekühlt und 2,5 h bei 10 bis 15°C zur Ausfällung der Kristalle stehengelassen. Die ausgefallenen Kristalle werden durch Filtration abgetrennt, mit zwei 5-ml-Portionen Acetonitril und zwei 5-ml-Portionen Aceton in dieser Reihenfolge gewaschen und anschliessend getrocknet. Man erhält 1,51 g (Ausbeute 83,0%) 7-Amino-3-[5-( 1 -methyl-1,2,3,4-tetrazolyl)-thiomethyl]-A3-cephem-4-carbonsäure-hydrochlorid. IR, NMR und Schmelzpunkt des Produktes sind mit denen einer Standardprobe identisch.
Beispiel 11
In 10 ml Essigsäure werden 1,36 g 7-Aminocephalosporansäure und 0,58 g 5-Mercapto-l-methyl-1 H-tetrazol suspendiert und 6,6 g Zinn(IV)-bromid wird zugesetzt, um eine Lösung zu bilden. Diese Lösung wird 2 h bei 50°C umgesetzt. Die Reaktionsmischung wird mit 10 ml Wasser verdünnt und wässriges Ammonium (28 Gew.%) werden unter Eiskühlung zugegeben, um den pH auf 3,8 einzustellen. Die dabei ausgefallenen Kristalle werden durch Filtration abgetrennt und in 10 ml einer wässrigen Methanollösung (50 Gew.%) unter Zugabe von wässrigem Ammoniak (28 Gew.%) gelöst. Eine geringe Menge unlöslicher Stoffe wird durch Filtration entfernt und dann wird der pH der Lösung mit 6N Chlorwasserstoffsäure auf 3,8 eingestellt. Die dabei ausgefallenen Kristalle werden durch Filtration gesammelt, mit zwei 5-ml-Portionen Wasser und zwei 5-ml-Portionen Aceton in dieser Reihenfolge gewaschen und anschliessend getrocknet; man erhält 1,28 g (Ausbeute 78,2%) 7-Amino-[5-(l-methyl-1,2,3,4-tetrazolyl)-thiomethyl]-A3-cephem-4-carbonsäure.

In 10 ml Essigsäure werden 1,36 g 7-Aminocephalosporan-

säure und 0,60 g 5-Mercapto-l-methyl-1 H-tetrazol suspendiert und 8,9 g Pyrophosphorsäure werden zu der Suspension zugegeben. Danach wird die entstehende Mischung 10 h bei 45 bis 50°C umgesetzt. Nach Beendigung der Reaktion wird die Reaktionsmischung in 10 ml Eiswasser gegossen und ihr pH mit wässrigem Ammoniak (28 Gew.%) auf 3,8 eingestellt. Die dabei ausgefallenen Kristalle werden durch Filtration abgetrennt, mit zwei 5-ml-Portionen Wasser und zwei 5-ml-Portionen Aceton in dieser Reihenfolge gewaschen und anschliessend getrocknet. Man erhält 1,17 g (Ausbeute 70,9%) 7-Amino-3-[5-( 1 -methyl-1,2,3,4-tetrazolyl)-thiomethyl]-A3-cephem-4-carbonsäure.
Beispiel 13
In 7,0 ml Essigsäure werden 1,35 g 7-Aminocephalospo-ransäure und 0,60 g 5-Mercapto-l-methyl-1 H-tetrazol suspendiert. Man gibt 0,93 ml einer 100%igen Magic Acid (äquimolare Mischung aus Fluorschwefelsäure und Anti-monpentafluorid) unter Eiskühlung zu der Suspension und setzt daraufhin die Mischung 3 h bei 30°C um. Nach Beendigung der Reaktion wird die Reaktionsmischung in 35 ml Eis-Wasser gegossen, und der pH der resultierenden Lösung wird mit konz. wässrigem Ammoniak auf 3,7 eingestellt. Anschliessend wird die Lösung unter Eiskühlung 1 h gerührt. Die dabei ausgefallenen Kristalle werden durch Filtration entfernt, mit 10 ml Wasser und 10 ml Aceton in dieser Reihenfolge gewaschen und anschliessend getrocknet. Man erhält 1,34 g (Ausbeute 82,0%) 7-Amino-3-[5-(l-methyl-1,2,3,4-tetrazolyl)-thiomethyl]-A3-cephem-4-carbonsäure.
Beispiel 14
Die folgenden erfindungsgemässen Verbindungen werden in Ausbeuten von 65 bis 90% bei im wesentlichen den gleichen Bedingungen wie in den Beispielen 1 bis 13 erhalten, wenn man 7-Aminocephalosporansäure, eine geeignete Thiolverbindung der Formel (III) und eine Säure einsetzt, die aus den folgenden ausgewählt wird: Schwefelsäure, Pyrophosphorsäure, Perchlorsäure, Chlorschwefelsäure, Pyroschwe-felsäure, Fluorschwefelsäure, Magic Acid (FSCbH-SbFs), FSOaH-AsFs, CFsSChH-SbFs, H2SO4-SO3, Methansulfonsäure, Trifluormethansulfonsäure, p-Toluolsulfonsäure, Zinkchlorid, Zinn(IV)-chlorid und Zinn(IV)-bromid, und bei der Reaktion als Lösungsmittel Acetonitril oder Essigsäure verwendet.

oxyd und 0.38 g 5-Mercapto-l-methyl-lH-tetrazol suspendiert und 0.98 g (0.53 ml) konz. Schwefelsäure der resultierenden Suspension unter Eiskühlung beigegeben, um die Suspension in eine Lösung umzuwandeln. Die Reaktion wurde bei 30°C während einer Stunde durchgeführt und anschliessend wurde eisgekühlt und zu 6 ml Wasser bei 5-10°C beigegeben, woraufhin diese Mischung mit 28%igem wässrigem Ammoniak auf einen pH-Wert von 3,5 eingestellt wurde. Die gefällten Kristalle wurden durch Filtration gesammelt und mit zwei Portionen von 1,5 ml Wasser und zwei Portionen von 1 ml Azeton in dieser Reihenfolge gewaschen und getrocknet, um 0,25 g (Ausbeute 76,2%) von 7-Amino-3-[5-( 1 -methyl-1,2,3,4-tetrazolyl)thiomethyl]-A3-cephem-4-carbocylsäure zu erhalten.

Verfahren zur Herstellung einer 7-(subst.)-Amino-3-subst.-thiomethyl-A3-cephem-4-carbonsäure der folgenden allgemeinen FormelCH2SR8(I)10COOHwobei R1 ein Wasserstoffatom oder eine Ci-4-Alkoxygruppe bedeutet; R2 eine Aminogruppe oder eine Gruppe mit der FormelR3RVC=C-NH-,Räbei der R3, R4 und R5, die gleich oder verschieden sein können, für Wasserstoffatome oder organische Reste stehen, die nicht an der Reaktion teilhaben, oder der FormelR6-R7/"C=N-,15202530bei der R6 und R7, die gleich oder verschieden sein können, für Wasserstoffatome oder organische Reste stehen, die nicht an der Reaktion teilnehmen, bedeutet; und R8 den Rest einer Thiolverbindung bedeutet, oder eines an der Carboxylgruppe derivatisierten Abkömmlings aus der Gruppe bestehend aus einem Ester, einem Anhydrid und ein Amid davon oder eines Salzes dieser Säure, dadurch gekennzeichnet, dass man eine Cephalosporansäure der folgenden allgemeinen Formel(II)COOHwobei R1 und R2 die oben definierte Bedeutung haben; X eine unsubstituierte oder substituierte Acyloxy- oder Carbamoyl-oxygruppe bedeutet; >Y für^S oder >S —■ O steht; oder einen an der Carboxylgruppe derivatisierten Abkömmling aus der Gruppe bestehend aus einem Ester, einem Anhydrid und ein Amid davon oder ein Salz der Säure mit einer Thiolverbindung der allgemeinen FormelR8-SH(III)wobei R8 die oben angegebene Bedeutung hat, oder einem Salz dieser Verbindung zur Reaktion bringt, und zwar in einem organischen Lösungsmittel in Gegenwart einer Protonensäure oder einer Lewissäure oder einer Komplexverbindung der Lewissäure, mit Ausnahme von Bortrifluorid und seinen Komplexverbindungen, wobei für den Fall, dass Y als Sulfoxyd vorliegt, dieses während der Reaktion zum Sulfid reduziert wird.
- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man als Protonensäure Pyrophosphorsäure, eine Schwefelsäure, eine Sulfonsäure oder eine Supersäure verwendet.
- 3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man als Lewissäure oder ihre Komplexverbindung (ausgenommen Bortrifluorid und dessen Komplexverbindungen), ein Zinkhalogenid, ein Zinnhalogenid oder eine deren Komplexverbindungen verwendet.
- 4. Verfahren nach einem der Ansprüche 1 oder 2, dadurch gekennzeichnet, dass man als Protonensäure oder Lewissäure (ausgenommen Bortrifluorid), Pyrophosphorsäure, Schwefelsäure, Chlorschwefelsäure, Fluorschwefelsäure, Methan-sulfonsäure, Trifluormethansulfonsäure, p-Toluolsulfon-säure, Magic Acid (FSCbH-SbFs), Perchlorsäure, Zinkchlorid, Zinkbromid, Zinn(IV)-chlorid oderZinn(IV)-bromid verwendet.
- 5. Verfahren nach einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass man als organisches Lösungsmittel eine organische Carbonsäure, ein Nitrii, ein Nitroalkan oder ein Sulfolan verwendet.
- 6. Verfahren nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass man eine Verbindung einsetzt, bei der R2 für eine Aminogruppe steht.
- 7. Verfahren nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass man eine Verbindung einsetzt, bei der R2 eine Gruppe der folgenden FormelnR3 R6,C=C-NH- oder ~~^,C=N-bedeutet, R4^ | R7^R5in denen R3, R4, R5, R6 und R7 die in Anspruch 1 angegebenen Bedeutungen haben.35 8. Verfahren nach einem der Ansprüche 1 bis 7, dadurch gekennzeichnet, dass man eine Verbindung einsetzt, bei der für 2>S steht.
- 9. Verfahren nach einem der Ansprüche 1 bis 7, dadurch gekennzeichnet, dass man eine Verbindung einsetzt, bei der40 >Y für ;>S —■ O steht.
- 10. Verfahren nach einem der Ansprüche 1 bis 9, dadurch gekennzeichnet, dass man eine Verbindung einsetzt, bei der R1 ein Wasserstoffatom ist.
- 11. Verfahren nach einem der Ansprüche 1 bis 9, dadurch45 gekennzeichnet, dass man eine Verbindung einsetzt, bei derR1 eine Methoxygruppe ist.
- 12. Verfahren nach einem der Ansprüche 1 bis 11, dadurch gekennzeichnet, dass man eine Verbindung einsetzt, bei der R8 eine unsubstituierte oder substituierte heterocyclische so Gruppe bedeutet.
- 13. Verfahren nach Anspruch 12, dadurch gekennzeichnet, dass man eine Verbindung einsetzt, bei der R8 eine 5-(l -Methyl-1,2,3,4-tetrazolyl)-, 2-(l ,3,4-Thiadiazolyl)-oder 5-(2-methyl-1,3,4-thiadiazolyl)-Gruppe bedeutet.55 14. Verfahrennach Anspruch 13, dadurch gekennzeichnet, dass man eine Verbindung einsetzt, bei der R8 eine 5-(l -Methyl-1,2,3,4-tetrazolyl)-Gruppe bedeutet.
- 15. Verfahren nach einem der Ansprüche 1 bis 14, dadurch gekennzeichnet, dass man die Reaktion bei einer Temperatur60 von -20° bis 80°C durchführt.
