Description
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Verbindungen, welche eine Peptidstruktur besitzen. Diese neuen Verbindungen weisen eine schmerzstillende Wirkung auf.
65 Aus dem Gehirn oder cerebraler Rückenmarksflüssigkeit von Säugetieren sind bereits endogene Substanzen mit morphinartigen Eigenschaften extrahiert worden. Diese als En-kephaline bezeichneten Substanzen sind von Hughes et al.,

Nature, Bd. 258, S. 577 (1975) als Pèntapeptide mit dem folgenden Aufbau
H-Tyr-Gly-Gly-Phe-Met-OH H-Tyr-Gly-Gly-Phe-Leu-OH
identifiziert worden. Diese Verbindungen werden als Me-thionin-enkephalin bzw. Leucin-enkephalin bezeichnet.
Es wurde zwar nachgewiesen, dass diese Verbindungen bei Mäusen nach intracerebroventrikularer Verabreichung analgetische Wirksamkeit zeigen [Buscher et al., Nature, Bd. 261, S. 423 (1976)], doch ermangeln sie einer nutzbaren analgetischen Wirksamkeit, wenn sie parenteral verabreicht werden.
5 Ziel der vorliegenden Erfindung war es, ein Verfahren zur Herstellung neuer Verbindungen zu entwickeln, welche eine analgetische Wirksamkeit besitzen.

April 1976 beansprucht wird, sind Verbindungen bekanntgeworden, die der obigen allgemeinen 6o Formel I entsprechen, wobei in diesen Verbindungen sämtliche Reste R1; R2, R3, R5, R6, R7 und Y Wasserstoffatome sind, R4 für einen Methylrest steht, Z eine Gruppierung der Formel

Diese in der deutschen Offenlegungsschrift beschriebenen Verbindungen wurden durch die Definition der Sub-stituenten aus dem Schutzbereich der beim erfindungsgemäs-sen Verfahren herzustellenden Verbindungen der Formel I ausgeschlossen.
Bevorzugte, nach dem erfindungsgemässen Verfahren hergestellte Verbindungen der Formel I sind diejenigen, in denen W die Gruppe -CH2-X-CH3 bedeutet, insbesondere solche mit einer derartigen Gruppe, worin X Schwefel bedeutet.
Zu den pharmazeutisch annehmbaren nichttoxischen Säureadditionssalzen der Verbindungen der Formel I gehören sowohl organische als auch anorganische Säureadditionssalze, zum Beispiel die mit Salzsäure, Schwefelsäure, Sulfonsäure, Weinsäure, Fumarsäure, Bromwasserstoffsäure, Glykolsäure, Citronensäure, Maleinsäure, Phosphorsäure, Bernsteinsäure, Essigsäure, Salpetersäure, Benzoesäure, Ascorbinsäure, p-Toluolsulfonsäure, Benzolsulfonsäure, Naphthalinsulfonsäure oder Propionsäure hergestellten. Die bevorzugten Säureadditionssalze sind die mit Salzsäure, Essigsäure oder Bernsteinsäure hergestellten. Alle diese Salze können nach üblichen bekannten Methoden hergestellt werden.
Wie sich aus der Definition der verschiedenen in Formel I angegebenen Substituenten ergibt, handelt es sich bei den Verbindungen der Formel I um unsubstituierte oder N-substituierte Amide von Pentapeptiden. Die Stereokonfiguration der Verbindungen der Formel I ist ein wesentliches Merkmal dieser Verbindungen. Aus Zweckmässigkeitsgründen werden die Aminosäurereste der Pentapeptide der Formel I, beginnend mit dem Rest mit der die Kette abschliessenden Aminofunktion numeriert. Die Chiralität (Händig-keit) der Aminosäurereste in der Reihenfolge von Stellung 1 bis Stellung 5 ist demnach L, D, L, L und L. Ferner sei daraufhingewiesen, dass der Rest in Stellung 3 u.a. ein Glycin-rest sein kann. In solchen Fällen ist hinsichtlich dieses Restes Chiralität natürlich nicht möglich. Wenn jedoch in Stellung 3 ein chiraler Aminosäurerest steht, dann muss die Chiralität dieses Restes L sein.
Die primären Alkylgruppen mit 1 bis 3 Kohlenstoffatomen, die zu den Bedeutungen von Rl5 R2, R3, R6 und R7 gehören, sind bevorzugt Methyl-, Ethyl- und n-Propylgruppen.
Die primären oder sekundären Alkylgruppen mit 1 bis 4 Kohlenstoffatomen, die die Gruppen R4 und R5 bedeuten können, sind insbesondere Methyl, Ethyl, n-Propyl, Iso-propyl, n-Butyl, Isobutyl und sek.-Butyl.
Die Alkylgruppen mit 1 bis 4 Kohlenstoffatomen, die R9 bedeuten kann, sind Methyl, Ethyl, n-Propyl, Isopropyl, n-Butyl, Isobutyl, sec.-Butyl oder tert.-Butyl, vorzugsweise die Ethylgruppe. Die Aralkylgruppe, die R9 bedeuten kann, ist insbesondere eine unsubstituierte oder substituierte Aralkylgruppe, vorzugsweise mit 7 bis 10 Kohlenstoffatomen. Besonders bevorzugte Aralkylgruppen sind die Benzylgruppen oder substituierte Benzylgruppen. Zu in Betracht kommenden Substituenten gehören u.a. Halogen, wie Fluor, Chlor oder Brom, C!-C3-Alkoxy, wie Methoxy, Ethoxy oder Propoxy, Trifluormethyl, Cj-Cj-Alkyl und Cj-Q-Alkyl-thio, wie Methylthio oder Ethylthio. Ein etwa vorhandener Substituent steht vorzugsweise in der para-Stellung. Ein in hohem Masse bevorzugter Substituent ist Methoxy, und eine in hohem Masse bevorzugte Aralkylgruppe ist die p-Meth-oxybenzylgruppe.
Hinsichtlich der Reste in den einzelnen Stellungen des Pentapeptids der Formel I ist folgendes auszuführen:
(A) Stellung 1
Diese Stellung stellt das Aminoendteil des Peptids dar. Der Rest ist der sich aus L-Tyrosin oder L-(0-acetyl)-tyrosin ergebende. In beiden Fällen kann der Rest am Stickstoff un-substituiert sein, d.h. Rj und R2 sind Wasserstoff. Er kann durch eine Allylgruppe substituiert sein, so dass dann R2 eine Allylgruppe bedeutet. Darüber hinaus kann der Rest durch eine oder zwei primäre Alkylgruppen mit 1 bis 3 Kohlenstoffatomen substituiert sein, d.h. Rx und/oder R2 ist eine primäre Alkylgruppe mit 1 bis 3 Kohlenstoffatomen. Zu Beispielen für substituierende primäre Alkylgruppen mit 1 bis 3 Kohlenstoffatomen gehören N-Methyl-, N-Ethyl-, N-n-Pro-pyl-, N,N-Di-methyl, N,N-Diethyl, N,N-Di-n-propyl, N-Methyl-N-ethyl, N-Methyl-N-n-propyl und N-Ethyl-N-n-propyl. Vorzugsweise ist der in Stellung 1 des Peptids der Formel I befindliche Tyrosyl- oder O-Acetyltyrosyl-Rest am Stickstoff nicht substituiert. Ferner ist bevorzugt, dass der Rest der Tyrosylrest ist.
(B) Stellung 2
Der in der zweiten Stellung des Peptids der Formel I befindliche Aminosäurerest muss das D-Stereoisomere eines Aminosäurerests sein. In Betracht kommen hierbei die Reste von D-Alanin (Ala) (R4 = Methyl), D-alpha-Aminobutter-säure (Abu) (R4 = Ethyl), D-Norvalin (Nva) (R4 = n-Pro-pyl), D-Vaün (Val) (R4 = Isopropyl), D-Norleucin (Nie) (R4 = n-Butyl), D-Leucin (Leu) (R4 = Isobutyl) und D-Isoleucin (Ile) (R4. = sek.-Butyl). Vorzugsweise ist der Rest der von D-Alanin. Bei allen diesen Aminosäureresten bedeutet die Gruppe R3 an dem Stickstoffatom, das der Ami-nogruppe der ursprünglichen Aminosäure entspricht, entweder ein Wasserstoffatom oder einen primären Alkylrest mit 1 bis 3 Kohlenstoff atomen. Im letztgenannten Fall ist der Aminosäurerest N-substituiert. Solche N-Substituenten sind N-Methyl, N-Ethyl und N-n-Propyl. Vorzugsweise ist die Aminosäure in Stellung 2 N-unsubstituiert, d.h. R3 bedeutet ein Wasserstoffatom.
(C) Stellung 3
Der in dieser Stellung vorliegende Aminosäurerest ist der des Glycins (Gly) oder einiger anderer Aminosäuren, wie L-Alanin, L-(alpha-Amino)-buttersäure, L-Norvalin, L-Valin, L-Norleucin, L-Leucin und L-Isoleucin. Vorzugsweise leitet sich der Rest in dieser Stellung des Peptids von Glycin ab.
(D) Stellung 4
Der Aminosäurerest in dieser Stellung ist der des L-Phe-nylalanins (Phe). Er kann am Aminostickstoffatom unsub-stituiert oder substituiert (R6) sein. Wenn der Rest N-substituiert ist, handelt es sich um N-Methyl, N-Ethyl oder N-n-Propyl. Vorzugsweise ist der Rest am Stickstoff unsub-stituiert (Re = Wasserstoff).
(E) Stellung 5
Der Aminosäurerest in Stellung 5 des Pentapeptids ist der Rest eines Amids von L-Methionin (Met) (W = CH2SCH3), L-Norleucin (Nie) (W = -CH2CH2CH3), L-(0-Methyl)-homoserin [Hse(Me)] (W = -CH2OCH3), L-Leucin (Leu) [W = -CH(CH3)2], L-(0-Alkyl- oder -Aral-kyl)-serin [Ser(Alk) oder Ser(Aralk)] (W = OR9), oder L-(S-Alkyl- oder -Aralkyl)-cystein [Cys(Alk) oder Cys(Aralk)] {W = SR9). Vorzugsweise ist der Aminosäurerest in Stellung 5 der Rest des Amids von L-Methionin oder L-Leucin. In den Fällen, in denen der Rest in Stellung 5 O-substituiertes Serin oder S-substituiertes Cystein ist, ist der bevorzugte Alkyl-substituent die Ethylgruppe und der bevorzugte Aralkylsub-stituent die p-Methoxybenzylgruppe.
Wenn es sich bei dem Rest dieser endständigen Aminosäure um einen anderen als den des L-Leucins handelt, kann er an seinem Aminostickstoff unsubstituiert oder substituiert sein. Handelt es sich dabei um den Rest des L-Leucins, dann

ist er am Aminostickstoff substituiert. Als Substituenten kommen primäre Alkylgruppen mit 1 bis 3 Kohlenstoffatomen in Betracht, nämlich N-Methyl, N-Ethyl und N-n-Pro-pyl. Vorzugsweise ist der Aminostickstoff substituiert, d.h. R7 ist eine primäre Alkylgruppe mit 1 bis 3 Kohlenstoffatomen und insbesondere eine Methylgruppe.
Da die Aminosäure in Stellung 5 des Pentapeptids die das Carboxylende liefernde Aminosäure darstellt, liegt sie ausserdem als Amid vor. Vorzugsweise ist das Amid N-un-substituiert, d.h. R8 bedeutet Wasserstoff. Die Amidgruppe kann jedoch auch N-monosubstituiert sein, wobei der Substituent eine Alkylgruppe mit 1 bis 3 Kohlenstoffatomen ist, zum Beispiel N-Methyl, N-Ethyl, N-n-Propyl oder N-Iso-propyl.
Ein bevorzugt anwendbares Verfahren zur Herstellung der Verbindungen der Formel II beruht auf der Festphasen-peptidsynthese. Dabei wird der Rest mit dem C-Ende an einen polymeren Träger gebunden und das Peptid mit jeweils einem Rest verlängert, bis das gewünschte Peptid, das immer noch an das Polymere gebunden ist, erhalten wird. Dann kann das Peptid durch ein dafür geeignetes Spaltungsmittel von dem Polymeren abgelöst werden. Beispielsweise kann die am NaIpha-Stickstoff mit tert.-Butyloxycarbonyl geschützte N-Methylaminosäure mit C-Ende durch Aktivierung mit Dicyclohexylcarbodiimid mit einem Benzylhydryl-aminopolymeren verknüpft werden. Die N-tert.-Butoxy-carbonylgruppe wird bevorzugt durch Umsetzung des an das 5 Polymere gebundenen Restes mit Trifluoressigsäure in Methylenchlorid entfernt. Neutralisation des Polymersalzes mit einer tertiären Base und Anfügen eines zweiten Restes kann auf die gleiche Weise erfolgen. Das blockierte Peptid wird von dem Polymeren erfindungsgemäss durch Behandlung io mit flüssigem HF bei 0 °C entfernt und kann dann durch Chromatographie gereinigt werden. Die genauen Bedingungen dieser Synthese sind dem Durchschnittsfachmann auf dem Gebiet der Festphasenpeptidsynthese bekannt und geläufig.

Wie vorstehend angegeben, wird die Aminosäure gewöhnlich zuerst mit Kaliumhydrid in Gegenwart eines dafür geeigneten Ethers zur Ausbildung des Dianions behandelt, das dann mit dem entsprechenden Alkyl- oder Allyliodid in die gewünschte N-substituierte Aminosäure übergeführt wird.
Wie für den Durchschnittsfachmann auf dem Gebiet der Peptidchemie ohne weiteres ersichtlich, kann unter stark alkalischen Bedingungen, wie sie bei der obigen Alkylierung angewandt werden, Racemisierung am alpha-Kohlenstoff-atom erfolgen. Das Ausmass der Racemisierung kann in Abhängigkeit von der jeweils betroffenen Aminosäure schwanken. Die Racemisierung kann durch Verwendung von überschüssigem Alkylierungsmittel und dadurch, dass die Reak-45 tionszeit so kurz wie möglich gehalten wird, auf ein Minimum gesenkt werden. Wenn aber tatsächlich eine stärkere Racemisierung erfolgt, dann kann das Produkt durch Umkristallisieren als Salz eines dafür geeigneten chiralen Amins, zum Beispiel als das Salz von d(+)-alpha-Phenylethylamin, so gereinigt werden.
In Fällen, in denen Ri und R2 gleiche primäre Alkylgruppen mit 1 bis 3 Kohlenstoffatomen bedeuten, kann das gewünschte N,N-disubstituierte Tyrosin nach folgender Gleichung hergestellt werden.

Der Aldehyd RxCHO kann Formaldehyd, Acetaldehyd oder Propionaldehyd sein.
In den Fällen, in denen Rt und R2 für verschiedene primäre Alkylgruppen mit 1 bis 3 Kohlenstoffatomen stehen, kann das N,N-disubstituierte Tyrosin durch Behandlung des entsprechenden N-monosubstituierten Tyrosins, das wie oben angegeben hergestellt wurde, mit Formaldehyd oder Acetaldehyd, wie vorstehend beschrieben, erhalten werden.
Die Verbindungen der Formel II, für die Y Acetyl bedeutet, können aus dem entsprechenden Peptid, das als Rest Y Wasserstoff enthält, hergestellt werden, und die Amino-gruppe am Ende der Kette wird geschützt. Die letztgenannte

Verbindung wird zum Beispiel mit Essigsäureanhydrid in Gegenwart von Pyridin behandelt, wodurch das entsprechende N-blockierte O-Acetylpeptid erhalten wird. Nach Entblockierung mit einer Mischung aus Salzsäure und Essigsäure wird die gewünschte Verbindung erhalten.

Die Verbindungen der Formel I sind wertvolle pharmazeutische Mittel. Sie zeigen analgetische Wirkung und sind insbesondere wirksam, wenn sie Säugetieren oder Menschen parenteral verabreicht werden.
Die Verbindungen der Formel I können als solche verabreicht werden, oder sie können zu pharmazeutischen Zubereitungen in Form von Dosierungseinheiten für die parenterale Verabreichung verarbeitet werden. Für solche Zubereitungen können organische oder anorganische Feststoffe und/oder Flüssigkeiten, die pharmazeutisch annehmbare Träger darstellen, verwendet werden. Hierfür in Betracht kommende Träger sind allgemein bekannt. Die Zubereitungen können Tabletten, Pulvergranulate, Kapseln, Suspensionen, Lösungen oder dergleichen sein.
Wenn sie in einer wirksamen Menge verabreicht werden, rufen die Verbindungen der Formel I eine analgetische Wirkung hervor. Die Dosierungen liegen im Bereich von etwa 0,1 bis 100 mg/kg Körpergewicht des Empfängers. Der bevorzugte Dosierungsbereich liegt zwischen 1,0 und 20 mg/kg Körpergewicht des Empfängers.
5 Die Erfindung wird durch das folgende Beispiel weiter erläutert.
Beispiel io Herstellung von L-Tyrosyl-D-alanyl-glycyl-L-
phenylalanyl-Na-methyI-L-methionylamid-acetat Das hier verwendete Ausgangsprodukt wurde nach der Festphasenpeptidsynthese hergestellt, indem man das C-Ende der Peptidstruktur, also das an seiner Aminogruppe i5 geschützte Na-Methyl-L-methionin, an ein Benzhydrylamin-harz kuppelte, die andere Aminosäuren ankuppelte und in der letzten Stufe des Verfahrens das als Aminoschutzgruppe wirkende Harz durch Behandlung mit einer trockenen Säure abspaltete, wobei das entsprechende Peptid mit seiner Na-20 methyl-L-methionylamid-Endgruppierung freigesetzt wurde.
A) Herstellung des Trifluoressigsäuresalzes des L-Tyrosyl-D-alanyl-glycyl-L-phenylalanyl-Na-methyl-L-methionyl-benzhydrylamin-harzes 25 Das im Titel genannte Peptidharz wurde nach der automatischen Festphasensynthese in der Vorrichtung zur Pep-tidsynthese «Beckman 990» hergestellt, indem man 5,0 g an dem Benzhydrylaminharz einsetzte.
Das verwendete Benzhydrylaminharz wurde unter Ver-30 wendung einer 4%igen Lösung von Diisopropyläthylamin, abgekürzt als DIEA, in Methylenchlorid neutralisiert, und dann kuppelte man das an seiner freien Aminogruppe mit der tert.-Butyloxycarbonylgruppe geschützte Na-Methyl-L-methionin, also die Verbindung der Formel BOC-(N-Me)-35 Met-OH, unter Verwendung von N,N-Dicyclohexylcarbodi-imid, abgekürzt als DCC, in Methylenchlorid an das Harz an. Man erhielt dabei ein mit der Gruppierung BOC-(N-Me)-Met-NH substituiertes Harz. Dann wurden die nachfolgenden Aminosäuren, nämlich L-Phenylalanin bzw. Glycin 40 bzw. D-Alanin bzw. L-Tyrosin, nacheinander in ihrer an der Aminogruppe mit der tert.-Butyloxycarbonylgruppe geschützten Form, also als BOC-Phe-OH bzw. BOC-Gly-OH bzw. BOC-Ala-OH bzw. BOC-Tyr-OH angekuppelt, wobei jeweils vor der Ankuppelung der nächsten Aminosäure die 45 Aminoschutzgruppe der vorhergegangenen bereits angekuppelten Aminosäure entfernt wurde.
Die Aminosäuren BOC-Phe-OH, BOC-Gly-OH, BOC-Ala-OH und BOC-Tyr-OH wurden nacheinander angekuppelt, indem man sie zuerst nach dem weiter unten kommen-50 den Programm Nr. 1 ankuppelte und anschliessend die gleiche Aminosäure nach dem unten erläuterten Programm Nr. 2 ankuppelte. Das Programm Nr. 2 wurde für jede der genannten Aminosäuren einmal durchgeführt, mit Ausnahme der Ankuppelung der Aminosäure L-Phenylalanin, für 55 welche das Kupplungsprogramm Nr. 2 dreimal durchgeführt wurde. Nach der Ankuppelung der letzten Aminosäure erhielt man das an das Harz gekuppelte Pentapeptid, bei dem die Aminogruppe des L-Tyrosins noch mit der tert.-Butyloxycarbonylgruppe geschützt war. Von diesem BOC-6o Pentapeptid-Harz wurde die Aminoschutzgruppe nach den Verfahrensschritten 1 bis 8 des Programms Nr. 1 abgespaltet, also unter Verwendung von Trifluoressigsäure als praktisch trockenes Säuremedium zur Abspaltung der Aminoschutzgruppe, und man erhielt so 6,13 g der im Titel genann-65 ten Verbindung.
In den Programmen Nr. 1 und 2 wurden die Waschvorgänge durchgeführt, indem man 10 ml der jeweiligen Waschflüssigkeit pro Gramm des Harzes einsetzte.

1. Dreimalige Waschung mit Methylenchlorid.
2. 5 Minuten Behandlung mit einer Mischung aus 30 Volumen-Teilen Trifluoressigsäure + 5 Volumen-Teilen Aet3SiH und 65 Volumen-Teilen Methylenchlorid,

B) Herstellung des Fluorwasserstoffsalzes des L-Tyrosyl-D-alanyl-glycyl-L-phenylalanyl-Na-methyl-L-methionyl-amides
Das aus dem Verfahrensschritt A) erhaltene Peptidharz . wurde mit flüssigem wasserfreiem Fluorwasserstoff im Vakuum während 60 Minuten bei 0 °C unter Verwendung von Anisol als Spülmittel behandelt. Die flüchtigen Komponenten wurden aus dem Reaktionsmedium unter Vakuum entfernt, und das Peptidharz wurde mit Äther trituriert, und man filtrierte, um restliche Mengen an der Fluorwasserstoffsäure und dem Anisol zu entfernen. Das Peptid wurde von dem Harz entfernt, indem man mit 10%iger Essigsäure tritu-rierte, filtrierte, dreimal mit 50 ml 10%iger Essigsäure wusch und lyophilisierte, wobei man 940 mg der rohen, im Titel genannten Verbindung erhielt.
C) Chromatographische Reinigung zur Herstellung des
Endproduktes
470 mg des rohen Peptid-diastereomeren, das aus dem Verfahrensschritt B) erhalten wurde, wurden auf einer Säule der Grösse 3,8 cm x 58,5 cm auf einem Reversephasen-(Ci8)-silicagel bei einem Niederdruck (7,17 kg/cm2) mit 25% Essigsäurenitril in 0,lnormalem Ammoniumacetat Chromatographien. Das austretende Material wurde in 1-Minuten-Fraktionen aufgesammelt, die Fraktionen der Nr. 98 bis 150 wurden miteinander vereinigt, und man lyophilisierte, wobei man 360 mg des Produktes erhielt.
Der Rest des rohen Peptides aus dem Verfahrensschritt B) wurde chromatographiert, wie dies im obigen Abschnitt erläutert wird, und man erhielt dabei 320 mg des Produktes.
Die nach den beiden obigen Abschnitten erhaltenen lyophilisierten Produkte wurden miteinander vereinigt, und man chromatographierte erneut über einer mit Sephadex G-10 gefüllten Säule einer Grösse von 2,5 cm x 10,0 cm, wobei 0,2normale Essigsäure als Laufmittel verwendet wurde, um das Ammoniumacetat zu entfernen.
Die Fraktionen wurden lyophilisiert und man erhielt 636,7 mg des im Titel genannten Pentapeptides.
Die optische Drehung des Produktes war wie folgt: (a)D25 + 19,7° (c = 0,5, in In HCl).
(a)3ds25 + 71,70 (c = 0,5, in In HCl).
Bei der Elementaranalyse ergab das Produkt der Summenformel C31H43N608S die folgenden Werte:
berechnet: C 56,35 H 6,71 N 12,72 S 4,85 gefunden: C 56,60 H 6,43 N 12,97 S 4,92

Die nach dem erfindungsgemässen Verfahren hergestellten neuen Peptidderivate der Formel I und ihre pharmazeutisch annehmbaren nichttoxischen Säureadditionssalze besitzen eine analgetische Wirksamkeit.
In der Folge werden nunmehr Tests beschrieben, mit denen die analgetische Wirksamkeit dieser Verbindungen überprüft wurde.
Die analgetische Wirksamkeit der Verbindungen der Formel I wird durch den Maus-Heizplattentest nachgewiesen. Bei diesem Test wird eine Maus in einen aufrechtstehenden Acrylzylinder eingebracht, dessen Boden eine an ihrer Oberfläche bei 52 °C gehaltene Heizplatte bildet. Der Maus wird durch subkutane Injektion eine vorbestimmte Menge der Testverbindung, die in einem Träger gelöst oder suspendiert ist, verabreicht. Nach Verstreichenlassen einer vorbestimmten Zeit nach der Verabreichung der Testverbindung wird die Maus auf die Heizplattenoberfläche gebracht. Die Zeit in Sekunden bis zum jeweiligen Eintritt zweier gesonderter Phänomene wird aufgezeichnet. Erstens wird die Zeit, bis die Maus ihre Hinterpfote leckt, gemessen und zweitens wird die Zeit gemessen, bis die Maus von der Heizplattenoberfläche hochspringt. Ein Mittel mit analgetischer Wirksamkeit führt zu einer Erhöhung dieser Zeiten im Vergleich zu Kontrollmäusen, die lediglich Injektionen des Trägers erhalten. Dies muss in einem Dosisbereich erfolgen, der nicht zu einer motorischen Inkoordination oder Inkapazität führt. In den folgenden Tabellen sind die bei diesem Test erhaltenen Ergebnisse zusammengestellt, wobei mit einer Kontrolle mit natürlichem Enkephalin und mit dem Amid von natürlichem Enkephalin verglichen wird. In der Tabelle I sind die Zeiten bis zum Lecken der Hinterpfote, in Tabelle II die Zeiten bis zum Fluchtsprung und in Tabelle III die Pro8

zentsätze der Tiere jeder Testgruppe angegeben, die eine analgetische Wirkung zeigen. Das Kriterium für einen positiven analgetischen Effekt ist folgendes: Die Zeit bis zum Lek-ken der Hinterpfote oder bis zum Fluchtsprung muss bei einem behandelten Tier gleich der oder grösser als die mittlere
Kontrollzeit plus zwei Standardabweichungen vom Mittel sein. Jedes Ergebnis in den folgenden Tabellen I und II stellt den Mittelwert plus oder minus Standardfehler dar, und Tabelle III zeigt die Prozentsätze, die mit wenigstens 9 Mäusen s und bis zu 40 Mäusen erhalten werden.

I « w(ii)worinRio Wasserstoff, Methyl, Äthyl, n-Propyl oder eine Aminoschutzgruppe darstellt,Z die durch ein Polymerharz geschützte Carboxamido-gruppe bedeutet undR2, R3, R4, Rs, R6, R7, Y und W die weiter oben angegebenen Bedeutungen haben, oder Säureadditionssalze davon,einer Behandlung mit flüssiger Fluorwasserstoffsäure bei 0 °C zur Abspaltung der Schutzgruppe unterwirft und erhaltene Verbindungen gegebenenfalls in die entsprechenden Salze überführt.
- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, so dass Z die durch ein Benzhydrylaminharz geschützteCarboxamidogruppe ist.
- 3. Verfahren nach Anspruch 1 zur Herstellung von L-Ty-rosyl-D-alanyl-glycyl-L-phenylalanyl-Na-methyl-L-methionylamid, dadurch gekennzeichnet, dass man das Tri-55 fluoressigsäuresalz von L-Tyrosyl-D-alanyl-glycyl-L-phenyl-alanyl-Na-methyl-L-methionyl-benzhydrylaminharzmit flüssigem HF bei 0 °C behandelt.
