PATENTANSPRÜCHE
1. Verfahren zur Herstellung von Verbindungen der Formel:

worin bedeuten R?B geschütztes Amino, R2 eine aliphatische Kohlenwasserstoffgruppe, welche einen oder mehrere Substituenten aufweisen kann, und Za Carboxy oder geschütztes Carboxy bedeuten, und ihrer Salze, dadurch gekennzeichnet, dass man eine Verbindung der Formel:

worin R2 und Za die oben angegebenen Bedeutungen haben, oder ein reaktionsfähiges Derivat an deren Aminogruppe oder ein Salz derselben mit einem Schutzagens der Aminogruppe umsetzt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass in der Formel I bzw. II R2 Niedrigalkyl oder Niedrigalkenyl bedeutet, welche einen oder zwei Substituenten aus der Gruppe Carboxy, geschütztes Carboxy, Arylthio, Niedrigalkylthio, Aryl, Acyloxy, Niedrigalkoxy, Aryloxy und Heterocyclen tragen können.
3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass in der Formel I bzw. II Za eine veresterte Carboxygruppe darstellt
4. Verfahren nach Anspruch 3, dadurch gekennzeichnet, dass die veresterte Carboxygruppe ein niedriger Alkylester, ein Niedrigalkenylester, ein Niedrigalkinylester, ein Mono-, Die oder Tri-halogen(niedrig)-alkylester, ein Niedrigalkanoyloxy(niedrig)alkylester, ein Niedrigalkansulfonyl(niedrig)alkyl- ester, ein Aryl(niedrig)alkylester oder ein Arylester der Carboxylgruppe ist.
5. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass in der Formel I geschütztes Amino für Ria Acylamino oder Benzylamino bedeutet.
6. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass als Schutzagens der Aminogruppe ein Acylierungsmittel, ein Halogenameisensäureester, ein Isocyansäureester, die Carbamidsäure, die entsprechende Thiosäure und die reaktionsfähigen Derivate der oben genannten Säuren verwendet werden.
Die Erfindung betrifft ein Verfahren zur Herstellung neuer syn-Isomeren von substituierten 2-Oxyiminoessigsäuren und ihrer Salze. Diese Verbindungen stellen wertvolle Ausgangsprodukte zur Herstellung von 3, 7-disubstituierten 3-Cephem-4-carbonsäuren dar, welche antibakterielle Aktivität, insbesondere gegen pathogene Bakterien, aufweisen.
Die neuen Verbindungen entsprechen der Formel:

Diese Tautomerie-Typen zwischen 2-Amino- und 2-Hydroxythiazol-Verbindungen und 2-Imino- oder 2-Oxothiazolinverbindungen, wie oben angegeben, sind in der Literatur bekannt und es ist für den Fachmann ohne weiteres ersichtlich, dass beide tautomeren Isomeren im Gleichgewicht miteinander vorliegen und leicht ineinander umwandelbar sind und demzufolge umfassen die erfindungsgemäss erhältlichen Verbindungen auch diese Isomeren. Dementsprechend fallen beide tautomeren Formen der Endprodukte und der Ausgangsverbindungen in den Rahmen der vorliegenden Erfindung.
Nachfolgend werden sowohl in der Beschreibung als auch in den Beispielen und in den Ansprüchen die erfindungsgemäss erhältlichen Verbindungen und die Ausgangsverbindungen, welche die Gruppe dieser tautomeren Isomeren umfassen (enthalten), der Einfachheit halber durch Verwendung eines der Ausdrücke dafür, d.h. durch die Formel:

Beispiele für Salze der Verbindungen der Formel I sind ein anorganisches Salz, wie z.B. ein Metallsalz, wie ein Al kalimetallsalz (z.B. ein Natrium-, Kaliumsalz und dgl.), und ein Erdalkalimetallsalz (z.B. ein Calcium-, Magnesiumsalz und dgl.), ein Ammoniumsalz und dgl., ein organisches Salz (z.B. ein organisches Aminsalz (wie ein Trimethylamin-, Tri äthylamin-, Äthanolamin-, Diäthanolamin-, Pyridin-, Picolin-, Dicyclohexylamin-, N,N'-Dibenzyläthylendiaminsalz und dgl.) und dgl., ein olganisches Säuresalz (z.B. ein Acetat, Maleat, Tartrat, Methansulfonat, Benzolsulfonat, Toluol sulfonat und dgl.), ein anorganisches Säuresalz (z.B. ein Hydrochlorid, Hydrobromid, Sulfat,
Phosphat und dgl.) oder ein Salz mit einer Aminosäure (wie z.B. Arginin, Asparaginsäure, Glutaminsäure und dgl.) und dgl.
Nachfolgend werden einige geeignete Beispiele und Erläuterungen der verschiedenen Definitionen, die in der Beschreibung angewendet werden und in den Rahmen der Erfindung fallen, näher erläutert.
Unter dem hier verwendeten Ausdruck niedrig ist, wenn nichts anderes angegeben ist, zu verstehen, dass eine damit bezeichnete Gruppe 1 bis 6 Kohlenstoffatome enthält.
Unter dem hier verwendeten Ausdruck aliphatische Kohlenwasserstoffgruppe ist eine unverzweigte (gerade) oder verzweigte aliphatische Kohlenwasserstoffgruppe mit 1 bis 6 Kohlenstoffatomen zu verstehen und dieser Ausdruck kann umfassen Niedrigalkyl, Niedrigalkenyl und dgl. Diese aliphatische Kohlenwasserstoffgruppe kann 1 bis 2 geeignete Substituenten, wie z.B. Carboxy-, geschützte Carboxy-, Arylthio-, Niedrigalkylthio-, Aryl-, Acyloxy-, Niedrigalkoxy-, Aryloxy-, heterocyclische Gruppen oder dgl. aufweisen.
Ein geeignetes Halogen ist z.B. Chlor, Brom, Fluor und Jod.
Bei einem geeigneten Niedrigalkoxy und einem geeigneten Niedrigalkoxyrest in dem Ausdruck Ar(niedrig)alkoxy kann es sich um ein solches handeln, das verzweigt sein kann, wie z.B. Methoxy, Äthoxy, Propoxy, Isopropoxy, Butoxy, sobutoxy, tert.-Butoxy, Pentyloxy, Hexyloxy und dgl., und vorzugsweise handelt es sich dabei um ein solches mit 1 bis 4 Kohlenstoffatomen, insbesondere mit 1 bis 2 Kohlenstoffatomen.
Ein geeignetes geschütztes Amino kann umfassen ein Acylamino und eine Aminogruppe, die durch eine andere konventionelle Schutzgruppe als die Acylgruppe, wie z.B.
Benzyl oder dgl., substituiert ist.
Ein geeignetes Niedrigalkyl und ein geeigneter Niedrigalkylrest in den Ausdrücken Niedrigalkylthio , Carboxy -(niedrig)alkyl , geschütztes Carboxy(niedrig)alkyl , Ar (niedrig)alkyl und Di(niedrig)alkylamino kann ein solches umfassen, das verzweigt sein kann, wie z. B. Methyl, Äthyl, Propyl, Isopropyl, Butyl, Isobutyl, tert.-Butyl, Pentyl, Hexyl und dgl., vorzugsweise ein solches mit 1 bis 4 Kohlenstoffatomen, insbesondere ein solches mit 1 bis 2 Kohlenstoffatomen.
Ein geeignetes geschütztes Carboxy und ein geeigneter geschützter Carboxyrest in dem Ausdruck geschütztes Carboxy(niedrig)alky1 kann umfassen verestertes Carboxy, worin der Ester einer der folgenden sein kann: z.B. ein niedriger Alkylester (wie Methylester, Äthylester, Propylester, Isopropylester, Butylester, Isobutylester, t-Butylester, Pentylester, t-Pentylester, Hexylester, l-Cyclopropyläthylester und dgl.), wobei der Niedrigalkylrest vorzugsweise 1 bis 4 Kohlenstoffatome enthalten kann; ein Niedrigalkenylester (z.
B. ein Vinylester, Allylester und dgl.), ein Niedrigalkinylester (z.B. ein Äthinylester, Propinylester und dgl.); ein Mono (oder Di-oder Di- oder Tri-)halogen(niedrig)alkylester (z.B. ein 2-Jod- äthylester, 2,2,2-Trichloräthylester und dgl.); ein Niedrig alkanoyloxy (niedrig) alkylester (z.
B. ein Acetoxymethylester, Propionyloxymethylester, Butyryloxymethylester, Valeryloxymethylester, Pivaloyloxymethylester, Hexanoyloxymethylester, 2-Acetoxyäthylester, 2-Propionyloxyäthylester und dgl.); ein Niedrigalkansulfonyl(niedrig)alkylester (z.B. ein Mesylmethylester, 2-Mesyläthylester und dgl.); ein Ar (niedrig)alkylester, z.B. ein Phenyl (niedrig) alkylester, der einen oder mehrere geeignete Substituenten aufweisen kann (wie z.B. ein Benzylester, 4-Methoxybenzylester, 4-Nitrobenzylester, Phenäthylester, Tritylester, Diphenylmethylester, Bis-(methoxyphenyl)methylester, 3,4-Dimethoxybenzyl- ester, 4-Hydroxy-3,5-di-tert.-butylbenzylester und dgl.);
ein Arylester, der einen oder mehrere geeignete Substituenten aufweisen kann (wie z.B. ein Phenylester, Tolylester, tert. Butylphenylester, Xylylester, Mesitylester, Cumenylester und dgl.) und gl.
Ein bevorzugtes Beispiel für geschütztes Carboxy kann sein Niedrigalkoxycarbonyl (z.B. Methoxycarbonyl, Äthoxycarbonyl, Propoxycarbonyl, Butoxycarbonyl, t-Butoxycarbonyl, t-Pentyloxycarbonyl, Hexyloxycarbonyl und dgl.) mit 2 bis 7 Kohlenstoffatomen, vorzugsweise mit 2 bis 5 Kohlenstoffatomen.
Ein geeignetes Aryl und ein geeigneter Arylrest in den Ausdrücken Ar(niedrig)alkyl , Ar(niedrig)alkoxy , Arylthio und Aryloxv kann z.B. sein Phenyl, Tolyl, Xylyl, Mesityl, Cumenyl, Naphthyl und dgl., wobei die Arylgruppe 1 bis 3 geeignete Substituenten aufweisen kann, wie z. B.
Halogen (wie Chlor, Brom, Jod oder Fluor), Hydroxy und dgl.
Zu einem geeigneten Niedrigalkenyl gehört ein solches mit 2 bis 6 Kohlenstoffatomen und dabei kann es sich beispielsweise handeln umVinyl, Allyl, Isopropenyl, l-Propenyl, 2-Butenyl, 3-Pentenyl und dgl., vorzugsweise um ein solches mit 2 bis 4 Kohlenstoffatomen.
Zu geeigneten Acylresten in den Ausdrücken Acylamino , Acylimino , Acyloxy und Acyloxymethyl , wie sie oben erwähnt worden sind, gehören z.B. Carbamoyl, eine aliphatische Acylgruppe und eine Acylgruppe, die einen aromatischen oder heterocyclischen Ring enthält. Geeignete Beispiele für das Acyl sind Niedrigalkanoyl (wie Formyl, Acetyl, Propionyl, Butyryl, Isobutyryl, Valeryl, Isovaleryl, Oxalyl, Succinyl, Pivaloyl und dgl.), vorzugsweise ein solches mit 1 bis 4 Kohlenstoffatomen, insbesondere ein solches mit
1 bis 2 Kohlenstoffatomen; Niedrigalkoxycarbonyl mit 2 bis 7 Kohlenstoffatomen (wie Methoxycarbonyl, Äthoxycarbonyl, Propoxycarbonyl, l-Cyclopropyläthoxycarbonyl, Isopropoxycarbonyl, Butoxycarbonyl, t-Butoxycarbonyl, Pentyloxycarbonyl, t-Pentyloxycarbonyl, Hexyloxycarbonyl und dgl.), vorzugsweise ein solches mit 3 bis 6 Kohlenstoffatomen;
Niedrigalkansulfonyl (wie Mesyl, Äthansulfonyl, Propansulfonyl, Isopropansulfonyl, Butansulfonyl und dgl.), vorzugsweise ein solches mit 1 bis 4 Kohlenstoffatomen, insbesondere ein solches mit 1 bis 2 Kohlenstoffatomen; Arensulfonyl (wie Benzolsulfonyl, Tosyl und dgl.); Aroyl (wie Benzoyl, Toluoyl, Naphthoyl, Phthaloyl, Indancarbonyl und dgl.); Ar(niedrig)alkanoyl (wie Phenylalcetyl, Phenylpopionyl und dgl.); Ar (niedrig) alkoxycarbonyl (wie Benzyloxycarbonyl, Phenäthyloxycarbonyl und dgl.) und dgl.
Der oben angegebene Aecylrest kann 1 bis 3 geeignete Substituenten aufweisen, wie z.B. Halogen (wie Chlor, Brom, Jod oder Fluor), Hydroxy, Cyano, Nitro, Niedrigalkoxy (wie Methoxy, Äthoxy, Propoxy, Isopropoxy und dgl.), Niedrigalkyl (wie Methyl, Äthyl, Propyl, Isopropyl, Butyl und dgl.), Niedrigalkenyl (wie Vinyl, Allyl und dgl.), Acyl, wie z.B. Halogen-(niedrig)alkanoyl (wie Chloracetyl, Dichloracetyl, Trichloracetyl, Trifluoracetyl und dgl.), Aryl (wie Phenyl, Tolyl und dgl.) und dgl. Geeignete Beispiele für das Acyl mit dem (den) Substituenten können sein Mono(oder Di- oder Tri)halogen(niedrig)alkanoyl (z.B. Trifluoracetyl, Trichloracetyl und dgl.), vorzugsweise ein solches mit 2 bis 4 Kohlenstoffatomen; Mono(oder Di-oder Tri)halogen(nied rig)alkanoylcarbamoyl (wie z.B.
Trichloracetylcarbamoyl und dgl.), vorzugsweise ein solches mit 3 bis 4 Kohlenstoffatomen oder dgl.
Zu geeigneten Aminoschutzgruppen für R7" gehören z.B.
Acyl, wie Halogen (niedrig) alkanoyl (z.B. Chloracetyl, Dichloracetyl, Trichloracetyl, Trifluoracetyl und dgl.), vorzugsweise ein solches mit 2 bis 3 Kohlenstoffatomen oder dgl.

alkyl [besonders bevorzugt Phenoxy(niedrig)- alkyl], das Hydroxy aufweisen kann, worin Alkenyl und der Alkenylrest 2 bis 6, vorzugsweise 2 bis 4 Kohlenstoffatome, und der Alkylrest vorzugsweise 1 bis 4, insbesondere 1 bis 2 Kohlenstoffatome, enthält.
Nachfolgend werden die Verfahren zur Herstellung der Ausgangsverbindung (II), d.h. des Syn-Isomeren und des Anti-Isomeren davon, die als Bezugsbeispiele verwendet werden, näher erläutert.
F) Verfahren (XXV)) e (XXVII) [Reaktionsschema (5) 1
Die Verbindungen (XXVII) können hergestellt werden durch Alkylierung der Verbindungen (XXVI).
Beispiele für Alkylierungsmittel, die in der Alkylierungsreaktion verwendet werden können, sind Di(niedrig) alkylsulfat (wie Dimethylsulfat( Diäthylsulfat und dgl.), Diazo (niedrig)alkan (wie Diazomethan, Diazoäthan und dgl.), ein Niedrigalkylhalogenid (wie Methyljodid, Äthyljodid und dgl.), ein Niedrigalkylsulfonat (wie Methyl-p-toluolsulfonat und dgl.), und dgl.
Die Umsetzung, die unter Verwendung eines Di(niedrig)- alkylsulfats, eines Niedrigalkylhalogenids oder eines Niedrigalkylsulfonats durchgeführt wird, wird in der Regel in einem Lösungsmittel, wie Wasser, Aceton, Äthanol, Äther, Dimethylformamid oder in irgendeinem anderen Lösungsmittel, welches die Reaktion nicht nachteilig beeinflusst, durchgeführt.
Die Reaktion wird vorzugsweise in Gegenwart einer Base, wie z.B. eines Alkalimetallhydroxids, eines Alkalimetallbicarbonats, eines Alkalimetallcarbonats, eines Alkalimetalacetats, von Tri(niedrig)alkylamin, Pyridin, N-(Niedrig)alkylmorpholin, N,N-Di(niedrig)alkylbenzylamin, N,N-Di(niedrig)alkylanilin, wie weiter unten beispielhaft angegeben, oder dgl. durchgeführt. Die Reaktionstemperatur ist nicht kritisch und die Umsetzung wird in der Regel unter Kühlen bis unter Erhitzen bis etwa zum Siedepunkt des Lösungsmittels durchgeführt.
Die Umsetzung, die unter Verwendung von Diazoalkan durchgeführt wird, wird in der Regel in einem Lösungsmittel, wie Äther, Tetrahydrofuran oder dgl., durchgeführt.
Die Reaktionstemperatur ist nicht kritisch und die Umsetzung wird in der Regel unter Kühlen oder bei Umgebungstemperatur durchgeführt.
J) Verfahren (XXVIII) + (XXII) < (XXIX) [Reaktionsschema (5)]
Die Verbindungen (XXIX) können hergestellt werden durch Umsetzung der Verbindung (XXVIII) mit der Verbindung (XXII).
Die Reaktion wird in der Regel in einem Lösungsmittel, wie Wasser, in einem Alkohol (wie Methanol, Äthanol und dgl.), in Benzol, Dimethylacetamid, Dimethylformamid, Tetrahydrofuran, einer Mischung davon oder in irgendeinem anderen Lösungsmittel, welches die Reaktion nicht nachteilig beeinflusst, durchgeführt. Die Reaktionstemperatur ist nicht kritisch und die Umsetzung wird in der Regel bei Umgebungstemperatur bis unter Erwärmen etwa bis zum Siedepunkt des Lösungsmittels durchgeführt.
Zur selektiven Herstellung des syn-Isomeren der Verbindung (XXIX) in hoher Ausbeute muss das syn-Isomere der Ausgangsverbindung (XXVIII) verwendet werden und die Reaktion wird vorzugsweise unter neutralen Bedingungen in Gegenwart einer Base, wie oben erwähnt, durchgeführt. Bevorzugte Beispiele für verwendbare Basen sind eine schwache Base, wie Alkalimetallacetat (z.B. Natriumacetat, Kaliumacetat und dgl.), ein Alkalimetallbicarbonat (wie Natriumbicarbonat, Kaliumbicarbonat und dgl.), ein Alkalimetallcarbonat (wie Natriumcarbonat, Kaliumcarbonat und dgl.) oder dgl.
K) Verfahren (XXIX)-3 (rille) [Reaktionsschema (5)]
Die Verbindungen Ille können hergestellt werden, indem man die Verbindung (XXIX) einer Eliminierungsreaktion zur Entfernung der Carboxyschutzgruppe unterwirft.
Bei der Eliminierungsreaktion können konventionelle Verfahren, wie sie für die Eliminierung der Schutzgruppe angewendet werden, beispielsweise die Hydrolyse und dgl., angewendet werden. Wenn die Schutzgruppe ein Ester ist, so kann sie durch Hydrolyse eliminiert werden.
Die Hydrolyse wird vorzugsweise in Gegenwart einer Base oder einer Säure durchgeführt. Zu geeigneten Basen können gehören eine anorganische Base und eine organische Base, wie z.B. ein Alkalimetall (wie Natrium, Kalium und dgl.), ein Erdalkalimetall (wie Magnesium, Calcium und dgl.), das Hydroxid oder Carbonat oder Bicarbonat davon, ein Trialkylamin (wie Trimethylamin, Triäthylamin und dgl.), Picolin, 1,5-Diazabicyclo [4,3,Oj non-5-en, 1 ,4-Diazabicyclo- [2,2,2]octan, 1,5-Diazabicyclo [5,4,0]undecen-5- oder dgl.
Zu geeigneten Säuren können gehören eine organische Säure (z.B. Ameisensäure, Essigsäure, Propionsäure, Trifluoressigsäure und dgl.) und eine anorganische Säure (wie z.B.
Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure und dgl.).
Die Umsetzung wird in der Regel in einem Lösungsmittel, wie Wasser, einem Alkohol (wie Methanol, Äthanol und dgl.), einer Mischung davon oder in irgendeinem anderen Lösungsmittel, welches die Reaktion nicht nachteilig beeinflusst, durchgeführt. Als Lösungsmittel kann auch eine flüssige Base oder eine flüssige Säure verwendet werden.
Die Reaktionstemperatur ist nicht kritisch und die Umsetzung wird in der Regel unter Kühlen bis unter Erwärmen durchgeführt.
L) Verfahren (XXVII) (XXVIII) [Reaktionsschema (5)]
Die Verbindung (XXVIII) kann hergestellt werden durch Halogenierung der Verbindung (XXVII).
Beispiele für in der Reaktion verwendbare Halogenierungsmittel sind ein konventionelles Halogenierungsmittel, wie es bei der Halogenierung der sogenannten aktivierten Methylengruppe verwendet wird, wie z.B. Halogen (wie Brom, Chlor und dgl.), Sulfurylhalogenid (wie Sulfurylchlorid und dgl.), ein Hypohalogenit (wie Unterchlorige Säure, Unterbromige Säure, Natriumhypochlorit und dgl.), ein N-halogeniertes Imid (wie N-Bromsuccinimid, N-Bromphthalimid, N-Chlorsucinimid und dgl.) und dgl.
Die Reaktion wird in der Regel in einem Lösungsmittel, wie z.B. in einem organischen Lösungsmittel (wie Ameisensäure, Essigsäure, Propionsäure und dgl.), in Tetrachlorkohlenstoff oder in irgendeinem anderen Lösungsmittel, welches die Reaktion nicht nachteilig beeinflusst, durchgeführt. Die Reaktionstemperatur ist nicht kritisch und die Umsetzung wird in der Regel unter Kühlen, bei Umgebungstemperatur, unter Erwärmen oder unter Erhitzen durchgeführt.
M) Verfahren (XXIX) -, (Illh) [Reaktionsschema (7)]
Die Verbindung (XXXh) kann hergestellt werden durch Umsetzung der Verbindung (XXXIX) oder eines reaktionsfähigen Derivats davon an der Aminogruppe oder eines Salzes davon mit einem Aminoschutzagens.
Ein geeignetes reaktionsfähiges Derivat an der Aminogruppe der Verbindung (XXXIX) kann z.B. sein ein konven tlonelles reaktionsfähiges Derivat, wie es bei der Amidierung verwendet wird, z.B. ein Silylderivat, das bei der Umsetzung der Verbindung (XXXIX) mit einer Silylverbindung gebildet wird, wie z.B. Bis(trimethylsilyl)acetamid, Trimethylsilylacetamid oder dgl.
Ein geeignetes Salz für die Verbindung (XXXIX) kann z.B. sein ein Säureadditionssalz, z.B. ein organisches Säuresalz (wie ein Acetat, Maleat, Tartrat, Benzolsulfonat, Toluolsulfonat und dgl.) oder ein anorganisches Säuresalz (wie ein Hydrochlorid, Hydrobromid, Sulfat, Phosphat und dgl.); ein Salz mit einer anorganischen Base, wie z.B. ein Alkalimetallsalz (wie ein Natriumsalz, Kaliumsalz und dgl.), ein Erdalkalimetallsalz (wie ein Calciumsalz, Magnesiumsalz und dgl.) oder ein Ammoniumsalz; ein Salz mit einer'organischen Base (z.B. ein Triäthylaminsalz, Pyridinsalz und dgl.) und und dgl.
Zu geeigneten Aminoschutzagentiert gehören ein Acylierungsmittel wie z.B. eine aliphatische, aromatische und heterocyclische Carbonsäure und die entsprechende Sulfonsäure, ein Halogenameisensäureester, ein Isocyansäureester und die Carbamidsäure sowie die entsprechende Thiosäure davon und die reaktionsfähigen Derivate der oben angegebenen Säuren.
Zu geeigneten reaktionsfähigen Derivaten der oben angegebenen Säuren gehören ein Säurehalogenid, ein Säureanhydrid, ein aktiviertes Amid, ein aktivierter Ester und dgl.
Geeignete Beispiele sind ein Säurechlorid, ein Säureazid, ein gemischtes Säureanhydrid mit einer Säure, wie substituierter Phosphorsäure (z.B. Dialkylphosphorsäure, Phenylphosphorsäure, Diphenylphosphorsäure, Dibenzylphosphorsäure, halogenierter Phosphorsäure und dgl.), Dialkylphosphoriger Säure, Schwefliger Säure, Thioschwefelsäure, Schwefelsäure, Alkylkohlensäure, einer aliphatischen Carbonsäure (wie Pivalinsäure, Pentansäure, Ihopentansäure, 2-Äthylbuttersäure oder Trichloressigsäure und dgl.) oder einer aromatischen Carbonsäure (wie Benzoesäure und dgl.); ein symmetrisches Säureanhydrid; ein aktiviertes Amid mit Imidazol, 4-substituiertem Imidazol, Dimethylpyrazol, Triazol oder Tetrazol;
oder ein aktivierter Ester (wie Cyanomethylester, Methoxymethylester, Dimethyliminomethyl (CH3)2N+ = CH-)-ester, Vinylester, Propargylester, p-Nitrophenylester, 2,4-Dinitrophenylester, Trichlorphenylester, Pentachlorphenylester, Mesylphenylester, Phenylazophenylester, Phenylthioester, p-Nitrophenylthioester, p-Kresilthioester, Carboxymethylthioester, Pyranylester, Pyridylester, Piperidylester, 8-Chinolylthioester] oder ein Ester mit N,N Dimethylhydroxylamin, l-Hydroxy-2-(1H)-pyridon, N-Hydroxysuccinimid, N-Hydroxyphthalimid oder l-Hydroxy-6- -chlor-1H-benzotriazol und dgl. Diese reaktionsfähigen Derivate können gewünschtenfalls je nach Art des zu verwendenden Acylierungsmittels ausgewählt werden.
Zu geeigneten Acylierungsmitteln können ferner gehören ein aliphatisches, aromatisches oder heterocyclisches Isocyanat oder Isothiocyanat (z.B. Methylisocyanat, Phenylisocyanat, Trichloracetylisocyanat, Methylisothiocyanat und dgl.) und ein Halogenformiat (wie Äthylchlorformiat, Benzylchlorformiat und dgl.). In diesem Falle wird beispielsweise dann, wenn das Trichloracetylisocyanat als Acylierungsmittel verwendet wird, die Trichloracetylcarbamoylgruppe als Acylgruppe eingeführt und diese Gruppe kann durch Behandlung mit einer Base in die Carbamoylgruppe umgewandelt werden, und wenn Äthylchlorformiat als Acylierungsmittel verwendet wird, kann als Acylgruppe die Äthoxycarbonylgruppe eingeführt werden.
Ein Beispiel für die Schutzgruppe (z. B. eine Acylgruppe), die in die Aminogruppe der Verbindung (XXXIX) durch das oben angegebene Aminoschutzagens (z.B. das Acylierungsmitel) eingeführt werden soll, kann die gleiche Schutzgruppe (z.B. Acylgruppe) sein, wie sie bei der Erläuterung des Schutzgruppenreste (z.B. des Acylrestes) in dem Ausdruck Acylamino angegeben worden ist.
Die Umsetzung wird in der Regel in einem konventionellem Lösungsmittel, wie Wasser, Aceton, Dioxan, Acetonitril, Chloroform, Methylenchlorid, Äthylenchlorid, Tetrahydrofuran, Äthylacetat, N,N-Dimethylformamid, Pyridin oder irgendeinem anderen organischen Lösungsmittel, welches die Reaktion nicht nachteilig beeinflusst, durchgeführt.
Unter diesen Lösungsmitteln können die hydrophilen Lösungsmittel in Form einer Mischung mit Wasser verwendet werden.
Die Reaktion wird vorzugsweise in Gegenwart eines Kondensationsmittels, wie z.B. eines sogenannten Vilsmeier-Reagens, wie z.B. (Chlormethylen)dimethyl-ammoniumchlorid, das bei der Umsetzung von Dimethylformamid mit Thionylchlorid oder Phosgen gebildet wird, einer Verbindung, die durch Umsetzung von Dimethylformamid mit Phosphoroxychlorid gebildet wird, und dgl., oder dgl. durchgeführt.
Die Reaktion kann auch in Anwesenheit einer anorganischen oder einer organischen Base, wie z.B. eines Alkalimetallhydroxids, eines Alkalimetallbicarbonats, eines Alkalimetallcarbonats, eines Alkalimetallacetats, von Tri(niedrig)alkylamin, Pyridin, N-(Niedrig)alkylmorpholin, N,N-Di (niedrig)alkylbenzylamin, N,N-Di-(niedrig)alkylanilin, wie weiter unten beispielhaft angegeben, oder dgl. durchgeführt werden. Wenn die Base oder das Kondensationsmittel flüssig ist, kann sie (es) auch als Lösungsmittel verwendet werden. Die Reaktionstemperatur ist nicht kritisch und die Reaktion wird in der Regel unter Kühlen oder bei Umgebungstemperatur durchgeführt.
Beispiel 1
1.) Eine Lösung von 17,4 g Äthyl-2-methoxyimino-4- -bromacetoacetat (ein Gemisch der sind und anti-Isomeren) und 5,4 g Thioharnstoff in 100 ml Äthanol wurde 4 Stunden lang unter Rückfluss erhitzt. Die Reaktionsmischung wurde stehen gelassen und in einem Kühlschrank abgekühlt, wobei Kristalle ausfielen. Die Kristalle wurden durch Filtrieren gesammelt, mit Äthanol gewaschen und getrocknet, wobei man 9,5 g Äthyl-2-methoxyimino-2-(2-amino-1,3-thiazol- -4-yl)acetathydrobromid (Anti-Isomeres) erhielt. Das Filtrat und die Waschwässer wurden vereinigt und unter vermindertem Druck eingeengt. Zu dem Rückstand wurden 100 ml Wasser zugegeben und die Mischung wurde mit Äther gewaschen.
Die wässrige Schicht wurde mit einer 20% eigen wässrigen Ammoniaklösung alk gesättigten wässrigen Natriumbicarbonatlösung gewaschen, bis das Waschwasser schwach alkalisch war. Der Extrakt wurde mit einer gesättigten wässrigen Natriumchloridlösung weiter gewaschen und über Magnesiumsulfat getrocknet.
Das Lösungsmittel wurde abdestilliert und der Rückstand wurde mit Diisopropyläther gewaschen, durch Filtrieren gesammelt und getrocknet, wobei man 4,22 g Äthyl-2-methoxyimino-2-(2-formamido-1,3-thiazol-4-yl)acetat (syn-Isomeres) F. 122 bis 124 C (Zers.), erhielt.

Beispiel 2
1.) 50 g Äthyl-2-methoxyimino-4-chloracetoacetat (syn Isomeres) wurden über einen Zeitraum von 3 Minuten unter Rühren bei Umgebungstemperatur zu einer Lösung von 18,4 g Thioharnstoff und 19,8 g Natriumacetat in einem Gemisch aus 250 ml Methanol und 250 ml Wasser zugegeben. Nach 35minütigem Rühren bei 40 bis 45 C wurde die Reaktionsmischung mit Eis gekühlt und mit einer gesättigten wässrigen Natriumbicarbonatlösung auf pH 6,3 eingestellt.
Nach 30minütigem Rühren bei der gleichen Temperatur wurden die Niederschläge durch Filtrieren gesammelt, mit 200 ml Wasser und dann mit 100 ml Diisopropyläther gewaschen und getrocknet, wobei man 37,8 g Äthyl-2-meth oxyimino-2-(2-amino- 1 ,3-thiazol-4-yl)acetat (syn-Isomeres), F. 161 bis 162 C, in Form von farblosen Kristallen erhielt.

2.) 3 g Pyridin wurden zu einer Lösung von 6,5 g Äthyl -2-methoxyimino-2-(2-amino- 1 ,3-thiazol-4-yl)acetat (syn-Isomeres) in einem Gemisch aus 60 ml Äthylacetat und 20 ml Dimethylformamid zugegeben. Zu der Lösung wurden unter Rühren bei 40C 8 g Äthylchlorformiat zugetropft. Nach der Zugabe von 50 ml Wasser zu der Reaktionsmischung wurde die organische Schicht abgetrennt, mit Wasser und dann mit einer gesättigten wässrigen Natriumchloridlösung gewaschen und über Magnesiumsulfat getrocknet. Das Lösungsmittel wurde unter vermindertem Druck abdestilliert.
Der Rückstand wurde unter Verwendung eines Äther/Petroläther (5/2)-Gemisches als Eluierungsmittel einer Säulenchromatographie an Silicagel (120 g) unterworfen, wobei man 5,4 g Äthyl-2-methoxyimino-2-(2-äthoxycarbonylamino- 1 ,3-thiazol -4-yl)acetat (syn-Isomeres) erhielt.

Beispiel 3
1.) 10 ml Äthanol wurden zu einer Suspension von 2,2 g Äthyl-2-methoxyimino-2-(2-amino- 1,3 ,3-thiazol-4-yl)acetat (syn-Isomeres) in 12 ml einer wässrigen 1 n Natriumhydroxidlösung zugegeben und die Mischung wurde 15 Stunden lang bei Umgebungstemperatur gerührt. Die Reaktionsmischung wurde mit 10% Der Chlorwasserstoffsäure auf pH 7,0 eingestellt und das Äthanol wurde unter vermindertem Druck abdestilliert.
Die zurückbleibende wässrige Lösung wurde mit Äthylacetat gewaschen, mit 10% Der Chlorwasserstoffsäure auf pH 2,8 eingestellt und unter Eiskühlung gerührt, wobei Kristalle ausfielen. Die Kristalle wurden durch Filtrieren gesammelt, mit Aceton gewaschen und aus Äthanol umkristallisiert, wobei man 1,1 g 2-Methoxyimino-2-(2 -amino-1,3-thiazol-4-yl)essigsäure (syn-Isomeres) in Form von farblosen Nadeln erhielt.

2.) 15,5 g Äthyl-2-methoxyimino-2-(2-amino-1,3-thiazol- 4-yl)acetathydrobromid (anti-Isomeres) wurden in einer Lösung von 4,4 g Natriumhydroxid in 150 ml Wasser gelöst und die dabei erhaltene Lösung wurde 1 Stunde lang bei Umgebungstemperatur gerührt. Ein unlösliches Material wurde abfiltriert und das Filtrat wurde auf pH 5,0 eingestellt, wobei Kristalle ausfielen. Die Kristalle wurden durch Filtrieren gesammelt und getrocknet, wobei man 8,0 g 2-Methoxyimino-2-(2-amino-1,3-thiazol-4-yl)essigsäure (anti-Isomeres) erhielt.

3.) 5 ml Pyridin wurden zu einer Suspension von 2,0 g 2-Methoxyimino-2-(2-amino- 1 ,3-thiazol-4-yl)essigsäure (syn- Isomeres) in 20 ml Äthylacetat zugegeben. Unter Rühren wurde bei 5 bis 70C eine Lösung von 2,5 g Bis-(2,2,2-tri- fluoressigsäure)anhydrid in 3 ml Äthylacetat zugetropft und die Mischung wurde 30 Minuten lang bei 3 bis 5 C gerührt.
Zu der Reaktionsmischung wurden 30 ml Wasser zugegeben und die Äthylacetatschicht wurde abgetrennt. Die wässrige Schicht wurde mit Äthylacetat weiter extrahiert und zwei Äthylacetatschichten wurden miteinander vereinigt, mit Wasser und einer gesättigten wässrigen Natriumchloridlösung gewaschen und über Magnesiumsulfat getrocknet. Das Lösungsmittel wurde unter vermindertem Druck abdestilliert, wobei man 0,72 g 2-Methoxyimino-2-[2-(2,2,2-trifluoracet- amido)- 1,3-thiazol-4-yl] essigsäure (syn-Isomeres) erhielt.

Beispiel 5
1.) 35,2 g Sulfurylchlorid wurden auf einmal zu einer gerührten Lösung von 48,9 g Äthyl-2-äthoxyimino-3-oxobu- tyrat (syn-Isomeres) in 49 ml Essigsäure bei Raumtemperatur zugegeben und das Rühren wurde bei der gleichen Temperatur 1 Stunde lang fortgesetzt. Nach der Zugabe der dabei erhaltenen Lösung zu 200 ml Wasser wurde die Lösung mit Methylenchlorid extrahiert. Der Extakt wurde mit einer gesättigten wässrigen Natriumchloridlösung gewaschen, mit einer wässrigen Natriumbicarbonatlösung neutralisiert und mit Wasser gewaschen.
Die Lösung wurde über Magnesiumsulfat getrocknet und unter vermindertem Druck eingeengt, wobei man 53,8 g Äthyl-2-äthoxyimino-3-oxo-4-chlorbutyrat (syn-Isomeres) in Form eines blassgelben Öls erhielt.
2.) Eine Mischung von 38,7 g Äthyl-2-äthoxyimino-3 -oxo-4-chlorbutyrat, 13,2 g Thioharnstoff, 14,3 g Natriumacetat, 95 ml Methanol und 95 ml Wasser wurde 40 Minuten lang bei 480C gerührt. Nachdem die dabei erhaltene Lösung mit einer wässrigen Natriumbicarbonatlösung auf pH 6,5 eingestellt worden war, wurden die auftretenden Niederschläge durch Filtrieren gesammelt und mit Diisopropyl äther gewaschen, wobei man 14,7 g Äthyl-2-äthoxyimino-2 -(2-aminothiazol-4-yl)acetat (syn-Isomeres), F. 130 bis 131"C, erhielt.

3.) 5 g Äthyl-2-äthoxyimino-2-(2-aminothiazol-4-yl)acetat (syn-Isomeres) wurden zu einer Mischung von 45,9 ml 1 n Natriumhydroxid und 30 ml Äthanol zugegeben und es wurde 5 Stunden lang bei Raumtemperatur gerührt. Nach der Entfernung des Äthanols aus der dabei erhaltenen Lösung unter vermindertem Druck wurde der Rückstand in 60 ml Wasser gelöst und mit 10% iger Chlorwasserstoffsäure auf pH 2,0 eingestellt. Die Lösung wurde einer Aussalzung unterworfen und die Niederschläge wurden durch Filtrieren gesammelt und getrocknet, wobei man 2,9 g 2-Athoxyimino-2- -(2-aminothiazol-4-yl)essigsäure (syn-Isomeres) erhielt.

Die folgenden Verbindungen liessen sich herstellen, wenn man in ähnlicher Weise arbeitete, wie dies in den vorangehen den Beispielen beschrieben worden ist.

Beispiel 8 (1) 40,0 g 2-Hydroxyimino-3-oxobuttersäureäthylester (syn-Isomer), 43,6 g 4-Fluorbenzylchlorid, 60,0 g N,N-Di methylformamid, 52,0 g Kaliumcarbonat und 60,0 ml Äthyl acetat wurden in üblicher Weise behandelt, wobei man
64,4 g 2-(4-Fluorbenzyloxyimino)-3-oxobuttersäureäthylester (syn-Isomer) erhielt. l. R. (Film):

(2) 64,0 g Äthyl-2-(4-fluorbenzyloxyimino)-3-oxobutyrat (syn-Isomer), 35,6 g Sulfurylchlorid und 70,0 ml Essigsäure wurden in ähnlicher Weise wie in Beispiel 5 (1) behandelt, wobei man 29,55 g 2-(4-Fluorbenzyloxyimino)-3-oxo-4 -chlorbuttersäureäthylester (syn-Isomer) erhielt.

(3) 29,0 g 2-(4-Fluorbenzyloxyimino)-3-oxo-4-chlorbut- tersäureäthylester (syn-Isomer), 8,8 g Thioharnstoff, 7,9 g Natriumacetat, 72,5 ml Wasser, 60 ml Tetrahydrofuran und 72,5 ml Äthanol wurden in ähnlicher Weise wie in Beispiel 5 (2) behandelt, wobei man 28,0 g 2-(4-Fluorbenzyloxyimino)-2-(2-aminothiazol-4-yl)-essigsäureäthylester (syn-Isomer) erhielt.

(4) 25,5 g Äthy.l-2-(4-fluorbenzyloxyimino)-2-(2-amino- thiazol-4-yl)-acetat (syn-Isomer), 1.3 g l-Methylimidazol,
118,3 ml einer ln-Natriumhydroxydlösung, 52 ml Methanol und 200 ml Tetrahydrofuran wurden in ähnlicher Weise wie in Beispiel 5(3) behandelt, wobei man 22,11 g 2-(4-Fluorbenzyloxyimino)-2-(2-aminothiazol-4-yl)-essigsäure (syn-Isomer) erhielt.

(5) 23,4 g 2-Fluorbenzyloxyimino)-2-(2-aminothiazol-4 -yl)-essigsäure (syn-Isomer), 32,2 g Bis-(trimethylsilyl)-acet amid, 49,9 g 2,2,2-Trifluoressigsäureanhydrid und 234 ml trockenes Äthylacetat wurden in ähnlicher Weise wie in Bei spiel 3(3) behandelt, wobei man 18,9 g 2-(4-Fluorbenzyloxy- imino)-2- [2-(2,2,2-trifluoracetamido)-thiazol-4-y -essigsäure (syn-Isomer), Schmelzpunkt 180 bis 182 C, erhielt.

Beispiel 9 (1) Durch Umsetzung von 2-Hydroxyimino-3-oxobutter säureäthylester (syn-Isomer) mit 3,4-Dichlorbenzylchlorid in an sich üblicher Weise erhielt man das Äthyl-2-(3,4-dichlor- benzyloxyimino)-3-oxobutyrat (syn-Isomer), Öl

Claims (6)
- PATENTANSPRÜCHE 1. Verfahren zur Herstellung von Verbindungen der Formel: EMI1.1 worin bedeuten R?B geschütztes Amino, R2 eine aliphatische Kohlenwasserstoffgruppe, welche einen oder mehrere Substituenten aufweisen kann, und Za Carboxy oder geschütztes Carboxy bedeuten, und ihrer Salze, dadurch gekennzeichnet, dass man eine Verbindung der Formel: EMI1.2 worin R2 und Za die oben angegebenen Bedeutungen haben, oder ein reaktionsfähiges Derivat an deren Aminogruppe oder ein Salz derselben mit einem Schutzagens der Aminogruppe umsetzt.
- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass in der Formel I bzw. II R2 Niedrigalkyl oder Niedrigalkenyl bedeutet, welche einen oder zwei Substituenten aus der Gruppe Carboxy, geschütztes Carboxy, Arylthio, Niedrigalkylthio, Aryl, Acyloxy, Niedrigalkoxy, Aryloxy und Heterocyclen tragen können.
- 3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass in der Formel I bzw. II Za eine veresterte Carboxygruppe darstellt
- 4. Verfahren nach Anspruch 3, dadurch gekennzeichnet, dass die veresterte Carboxygruppe ein niedriger Alkylester, ein Niedrigalkenylester, ein Niedrigalkinylester, ein Mono-, Die oder Tri-halogen(niedrig)-alkylester, ein Niedrigalkanoyloxy(niedrig)alkylester, ein Niedrigalkansulfonyl(niedrig)alkyl- ester, ein Aryl(niedrig)alkylester oder ein Arylester der Carboxylgruppe ist.
- 5. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass in der Formel I geschütztes Amino für Ria Acylamino oder Benzylamino bedeutet.
- 6. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass als Schutzagens der Aminogruppe ein Acylierungsmittel, ein Halogenameisensäureester, ein Isocyansäureester, die Carbamidsäure, die entsprechende Thiosäure und die reaktionsfähigen Derivate der oben genannten Säuren verwendet werden.Die Erfindung betrifft ein Verfahren zur Herstellung neuer syn-Isomeren von substituierten 2-Oxyiminoessigsäuren und ihrer Salze. Diese Verbindungen stellen wertvolle Ausgangsprodukte zur Herstellung von 3, 7-disubstituierten 3-Cephem-4-carbonsäuren dar, welche antibakterielle Aktivität, insbesondere gegen pathogene Bakterien, aufweisen.Die neuen Verbindungen entsprechen der Formel: EMI1.3 worin bedeuten R7a geschütztes Araiho, R2 eine aliphatische Kohlenwasserstoffgruppe, welche einen oder mehrere Substituenten aufweisen kann, und Za Carboxy oder geschütztes Carboxy bedeuten.Das erfindungsgemässe Verfahren ist also auf die Herstellung des syn-Isomeren von Oximen der obigen Formel I gerichtet während das entsprechende anti-Isomere durch die Formel: EMI1.4 dargestellt werden kann. Wenn es für die Erläuterung der Erfindung zweckmässig ist, sowohl das syn-Isomere als auch das anti-Isomere durch eine allgemeine Formel auszudrükken, werden sie durch die Formel: EMI1.5 wiedergegeben.Das erfindungsgemässe Verfahren ist im Anspruch 1 definiert. Das Verfahren und die Herstellung der Ausgangsprodukte können durch die folgende Reaktionsschemata veranschaulicht werden.
