In den in vivo-Versuchen bei der experimentellen Pankreatitis der Ratte, hervorgerufen durch Applikation von Natri-umdesoxycholat in das Organ, wurde bei gleichzeitiger Applikation von Glt-(Ala)3-NH-Et (20 mg intraperitoneal) ein niedriger Spiegel der Serum-Amylase nachgewiesen. Die Inhibitoren Glt-(Ala)3-NH-Et, Glt-Ala-Ala-Pro-NH-Et und insbesondere Glt-Ala-Ala-Pro-NH-Pr (in einer Dosis von 20 mg/Ratte) vermindern die Entwicklung eines Ödems, hervorgerufen durch eine subcutane Injektion der Elastase in die Rattenpfote, um 20 bis 40%.
Im Vergleich mit den bekannten Alkanoylderivaten (Dipeptiden), (Biorgan. Chem. 8,299, 1979) ist eine signifikante (zahlenordnungsgemäss höhere) Steigerung der Inhibition der elastolytischen Hydrolyse bemerkbar, was auf die Bedeutung des Charakters des N-terminalen Substituenten hindeutet.
Die erfindungsgemässen Inhibitoren enthalten keine unnatürliche Gruppe, so dass sich bei dem pharmacotherapeuti-schen Einsatz, insbesondere bei der akuten Pankreatitis, bei der chronischen Obstruktionslungenkrankheit und einigen Gelenkentzündungen kein Auftreten von unerwünschten Reaktionen erwarten lässt.
Gemäss der Erfindung lassen sich die Carboxyalkanoyl-peptid-alkylamide der allgemeinen Formel I so herstellen, dass man zunächst eine Verbindung der allgemeinen Formel II

worin X dieselbe Bedeutung hat wie in Formel I, insbesondere mit einem Anhydrid, Monohalogenid, Monoester oder Monoamid in Reaktion bringt.
Die Synthese dieser biologisch aktiven Peptide lässt sich im wesentlichen nach dem Verfahren der Fragmentkondensie-rung in Lösung oder durch den schrittweisen Aminosäureaufbau, d.h. durch die sogenannte «ste-wise»-Methode oder nach dem Feststoffphasenverfahren durchführen.
Als Schutzgruppe für die Zwischenprodukte wurde eine Gruppe vom Urethan-Typ (die Benzyloxycarbonylgruppe) benutzt. Man kann allerdings auch die in schwach saurem Medium labilen Gruppen (z.B. die tert.-Butyloxycarbonyl-oder o-Nitrobenzolsulfenylgruppe und ähnliche) oder die mit einem Metall, gegebenenfalls elektrolytisch reduzierten Gruppen (die 2-Halogenäthodycarbonylgruppe), verwenden.
Die Kondensierungsreaktionen wurden mit der Azid-, bzw. Carbodiimid-Methode und mit der Methode der Mischanhydride durchgeführt, aber ebenso gut lässt sich eine andere in der Peptidsynthese übliche Methode anwenden.
Nähere Einzelheiten des Verfahrens sind aus den nachfolgenden Beispielen ersichtlich.
Beispiel 1
Benzyloxycarbonylalanin-äthylamid
Zu einer Lösung von Benzyloxycarbonylalanin (23,3 g) in Tetrahydrofuran (100 ml) wurde N-Äthylpiperidin (14 ml) und nach dem Abkühlen auf -10°C sec.-Butylchlorformiat (14 ml) zugegeben. Nach 8 Minuten Rühren und Kühlen (-6°C) wurde eine 4,1 M Äthylaminlösung in Tetrahydrofuran (30 ml) zugesetzt. Nach 0,5 Stunden Rühren bei 0°C und 2 Stunden Rühren bei Zimmertemperatur wurde die Reaktionslösung abgedampft, der Rückstand in Äthylacetat (250 ml) gelöst, die Lösung nacheinander mit einer 5%igen Natriumhydrogencarbonatlösung und mit Wasser durchgeschüttelt, mit wasserfreiem Natriumcarbonat getrocknet und wiederum abgedampft. Der Rückstand wurde aus Äthylacetat (30 ml) und Petroläther umkristallisiert. Es wurden 18,1 g eines Produktes mit einem Schmelzpunkt von 116 bis 117°C erhalten.
Alaninäthylamid-hydrobromid
Benzyloxycarbonylalanin-äthylamid (10 g) wurde mit einer 36%igen Bromwasserstofflösung in Eisessig (40 ml) Übergossen. Nach 1 Stunde wurde zu der entstandenen

Lösung Äther (150 ml) zugegeben und durch Kratzen schied sich das kristalline Produkt aus, das abfiltriert mit Äther gründlich gewaschen und im Exzikator über Phosphorpent-oxid und Natriumyhdroxid getrocknet wurde. Es wurden 7,7 g Hydrobromid mit einem Schmelzpunkt von 224 bis 226°C erhalten. Rr 0,26/Si, 0,58/S2.
Si: n-Butanol-Essigsäure-Wasser (4:1:1), S2: n-Butanol-Essigsäure-Pyridin-Wasser (15:3:10:6).
Benzyloxycarbonylalanyl-alanyl-alanin-äthylamid
Zu einer Lösung von Benzyloxycarbonylalanyl-alaninhy-drazid (3,08 g) in Tetrahydrofuran (100 ml) und azeotroper Chlorwasserstoffsäure (4 ml) wurde bei einer Temperatur von -12°C eine Lösung von Natriumnitrit (690 mg) in Wasser (2,8 ml) zugegeben. Nach 8 Minuten Rühren und Kühlen (-10°C) wurde die Reaktionslösung mit vorgekühltem Äthyl-acetat (200 ml) verdünnt, die organische Phase wurde bei -15°C mit einer 3%igen Natriumhydrogencarbonatlösung in Salzsole ausgeschüttelt und zu einer vorgekühlten (~10°C) Lösung des Alaninäthylamides in Dimethylformamid (40 ml) zugegeben, das aus dem entsprechenden Hydrobromid (1,97 g) durch Neutralisieren mit N-Äthylpiperidin (1,4 ml) erhalten wurde. Nach 12 Stunden Stehen bei einer Temperatur von 0°C wurde die Lösung abgedampft und der feste Rückstand wurde aus einem Gemisch aus Isopropylalkohol (50 ml) und Dimethylformamid (50 ml) unter Zusatz von Wasser (250 ml) umkristallisiert. Es wurden 2,71 g eines Produktes mit einem Schmelzpunkt von 271 bis 273°C erhalten. [a]°0 - 8,3° (c = 0,3; Dimethylformamid).

Alanyl-alanyl-alanin-äthylamid
Benzyloxycarbonylalanyl-alanyl-alanin-äthylamid (1 g) wurde mit einer 36%igen Bromwasserstofflösung in Essigsäure (3 ml) übergössen und nach 1 Stunde wurde die entstandene Lösung mit Äther (30 ml) übergössen. Das ausgeschiedene Hydrobromid wurde abfiltriert, 2 Stunden im Exzikator über Phosphorpentoxid und Natriumhydroxid getrocknet, danach wurde es in 90%igem wässerigem Methanol (20 ml) gelöst und mit Zerolit FF im OH-Zyklus in Methanol dejoni-siert. Das methanolische Eluat wurde abgedampft, der Rückstand wurde durch azeotrope Destillation mit einem Methanol-Benzol-Gemisch getrocknet und schliesslich aus Methanol (10 ml) und Äther (30 ml) auskristallisiert. Es wurden 495 mg eines Produktes mit einem Schmelzpunkt von 259 bis 260°C erhalten.

3-Carboxypropionyl-alanyl-alanyl-alanin-äthylamid Zu einer Lösung von Alanyl-alanyl-alanin-äthylamid
(260 mg) in Dimethylformamid (5 ml) und Tetrahydrofuran (30 ml) wurde Bernsteinsäureanhydrid (30-mg) zugegeben und die Reaktionslösung wurde 2 Stunden unter Rückfluss zum Sieden erhitzt. Danach wurde das ausgeschiedene kri-s stalline Produkt abfiltriert und mit Tetrahydrofuran und Äther gewaschen. Durch Umkristallisieren aus Wasser (18 ml) wurden 180 mg eines Produktes mit einem Schmelzpunkt von 285 bis 287°C erhalten.

Benzyloxycarbonyl-alanyl-alanin-phenyläthylamid Zu einer Lösung von Alaninphenyläthylamid (4,9 g), das aus dem entsprechenden Benzyloxycarbonylderivat mit einer 36%igen Bromwasserstofflösung in Eisessig in Freiheit 45 gesetzt wurde (Rr 0,44/Si; 0,58/S2) und Benzyloxycarbonyl-alanin (5,6 g) in Dimethylformamid (200 ml), vorgekühlt auf -5°C, wurde N,N'-Dicyclohexylcarbodiimid (5,5 g) zugegeben. Nach 1 Stunde Rühren bei 0°C und 3 Stunden bei Zimmertemperatur wurde der ausgeschiedene N,N'-Dicyclo-50 hexyl-Harnstoff abfiltriert, mit Dimethylformamid gewaschen und das Filtrat wurde im Vakuum abgedampft. Der kristalline Rückstand wurde nacheinander mit 1 M Chlorwasserstoffsäure, Wasser, 5%iger Natriumhydrogen-carbo-natlösung und Wasser verrieben und aus einem Äthanol-55 Wasser-Gemisch (150 und 100 ml) kristallisiert. Es wurden 7,75 g eines Produktes mit einem Schmelzpunkt von 211 bis 212°C erhalten, [aß, -1,95° (c = 0,3; Dimethylformamid).

Beispiel 3
Prolinäthylamid
Zu einer Lösung von Benzyloxycarbonylprolin (11 g) in Eisessig (10 ml) wurde eine 36%ige Bromwasserstofflösung in Eisessig (40 ml) zugegeben. Nach 1 Stunde wurde zu der Reaktionslösung Äther (300 ml) zugegeben, das ausgeschiedene (nicht-kristalline) Hydrobromid wurde 2 Stunden im Exzikator über Phosphorpentoxid und Natriumhydroxid getrocknet, danach in einer gesättigten Ammoniaklösung in Chloroform (0°C), (60 ml), suspendiert, die Reaktions-Suspension wurde nach 20 Minuten Stehen bei 3°C filtriert und das Filtrat wurde abgedampft. Es wurden 5,7 g eines nicht-kristallinen Produktes erhalten.

Benzyloxycarbonyl-alanyl-prolin-äthylamid Zu einer Lösung von Prolinäthylamid (5,6 g) und Benzyl-oxycarbonylalanin (9,0 g) in einem auf -5°C abgekühlten Dimethylformamid-Tetrahydrofuran-Gemisch (5 bzw. 60 ml) wurde N,N'-Dicyclohexylcarbodiimid (9,0 g) zugegeben. Nach 1 Stunde Rühren bei 0°C wurde das Reaktionsgemisch 12 Stunden bei Zimmertemperatur stehengelassen. Der ausgeschiedene N,N'-Dicyclohexylharnstoff wurde abfiltriert, das Filtrat wurde abgedampft, der Rückstand wurde in einem Äthylacetat-Wasser-Gemisch gelöst, die organische Phase wurde nacheinander mit 1 M Chlorwasserstoffsäure, Wasser, einer 5%igen Natriumhydrogencarbonatlösung und Wasser gewaschen, mit wasserfreiem Natriumsulfat getrocknet und abgedampft. Der Rückstand wurde aus einem Äther-Petrol-äther-Gemisch (30 bzw. 30 ml) umkristallisiert. Es wurden 5,4 g eines Produktes mit einem Schmelzpunkt von 99 bis 101°C erhalten.

Es wurde in analoger Weise wie das Prolinäthylamid aus dem entsprechenden Benzyloxycarbonylderivat in einer quantitativen Ausbeute erhalten.

3-Carboxypropionyl-alanyl-prolin-äthylamid Zu einer Lösung von Alanyl-prolin-äthylamid (500 mg) in Dioxan (10 ml) und Dimethylformamid (0,25 ml) wurde Bernsteinsäureanhydrid (500 mg) zugegeben, die Reaktionslösung wurde 30 Minuten unter Rückfluss zum Sieden erhitzt, danach abgedampft und der Rückstand wurde aus Aceton (10 ml) und Äther (30 ml) kristallisiert. Es wurden 510 mg eines Produktes mit einem Schmelzpunkt von 139 bis 141 °C erhalten.

Beispiel 4
Benzyloxycarbonyl-alanyl-alanyl-prolin-äthylamid Zu einer auf -5°C abgekühlten Lösung von Alanyl-prolin-äthylamid (hergestellt aus 2,45 g des Benzyloxycarbonylderi-vates durch Einwirkung eines 36%igen Bromwasserstofflösung in Eisessig und durch anschliessende Freisetzung mit Zerolit FF im OH-Zyklus) in Dimethylformamid (20 ml) und Benzyloxycarbonyl-alanin (1,55 g) wurde N,N'-Dicyclohe-xylcarbodiimid (1,6 g) zugegeben. Nach 1 Stunde Rühren bei 0°C und 12 Stunden Stehen bei 3°C wurde der ausgeschiedene N,N'-Dicyclohexylharnstoff abfiltriert und das Filtrat abgedampft. Der nicht-kristalline Rückstand wurde in Äthylacetat gelöst (50 ml) und nach 12 Stunden Stehen bei 3°C wurde das ausgeschiedene Produkt abfiltriert. Es wurden 1,75 g eines Produktes mit einem Schmelzpunkt von 138 bis 139°C erhalten.

Es wurden 10,6 g eines Produktes mit einem Schmelzpunkt von 139 bis 140°C erhalten.

4-Carboxybutyryl-alanyl-alanyl-prolin-äthylamid Zu einer Lösung von Alanyl-alanyl-prolin-äthylamid (4,0 g) in Dimethylformamid (25 g) wurde Glutarsäureanhy-drid (1,7 g) zugegeben und die Reaktionslösung wurde 1 Stunde auf 60°C erwärmt. Danach wurde die Reaktionslösung abgedampft und der nicht-kristalline Rückstand wurde in Äthylacetat (30 ml) gelöst. Nach 48 Stunden Stehen bei 3°C wurde das ausgeschiedene Produkt (3,6 g) abfiltriert. Schmelzpunkt: 154 bis 156°C.

Beispiel 5
Benzyloxycarbonyl-alanin-isobutylamid Zu einer Lösung von Benzyloxycarbonyl-alanin (23 g) in Methylenchlorid (200 ml) und N-Äthylpiperidin (14 ml) wurde bei -10°C sec.-Butylchlorformiat (14 ml) zugegeben. Nach 10 Minuten Rühren bei -5°C wurde Isobutylamid (8,5 g) in Methylenchlorid (40 ml) zugegeben. Nach analoger Aufarbeitung wie in Beispiel 1, wurden nach dem Kristallisieren aus Äthylacetat (60 ml) und Petroläther (200 ml) 20,7 g eines Produktes mit einem Schmelzpunkt von 109 bis 111 °C erhalten.

Benzyloxycarbonyl-alanyl-alanyl-alanin-isobutylamid Es wurde in analoger Weise wie das entsprechende Benzyl-oxycarbonyl-alanyl-alanyl-alanin-äthylamid in einer Ausbeute von 58% der Theorie und mit einem Schmelzpunkt von 260 bis 262°C hergestellt.
M20 -10.9° (c = 0,3; Dimethylformamid).

Beispiel 8
Benzyloxycarbonyl-prolyl-alanin-methylester Zu einer Lösung von Benzyloxycarbonyl-prolin (12,45 g), freigesetzt aus dem entsprechenden Hydrochlorid (7,0 g) mit N-Äthylpiperidin (7 ml) in Methylenchlorid (40 ml), abgekühlt auf -5°C, wurde N,N'-Dicyclohexylcarbodiimid (11,0 g) zugegeben, und die Reaktionssuspension wurde 1 Stunde bei 0°C gerührt. Nach 12 Stunden Stehen bei 3°C wurde der ausgeschiedene N,N'-Dicyclohexylharnstoff abfiltriert, das Fitlrat wurde nacheinander mit 1 M Chlorwasserstoffsäure, Wasser, einer 5%igen Natriumhydrogen-carbo-natlösung und Wasser gewaschen, mit wasserfreiem Natriumsulfat getrocknet und abgedampft. Der feste Rückstand wurde aus Äthylacetat (100 ml) und Petroläther (500 ml) kristallisiert. Es wurden 12,2 g eines Produktes mit einem Schmelzpunkt von 45 bis 49°C erhalten.
Benzyloxycarbonyl-prolyl-alanin-hydrazid Zu einer Lösung von Benzyloxycarbonyl-prolyl-alanin-methylester (10,0 g) in Methanol (100 ml) wurde 80%iges Hydrazinhydrat (7,5 ml) zugegeben und die Reaktionslösung wurde 2 Stunden zum Sieden unter Rückfluss erwärmt. Danach wurde die Lösung abgedampft und der feste Rückstand wurde aus Wasser (40 ml) kristallisiert. Es wurden 6,8 g eines Produktes mit einem Schmelzpunkt von 142 bis 143°C erhalten.

Beispiel 9
3-Carboxyacryloyl-alanyI-alanyl-aIanin-äthylamid Zu einer Lösung von Alanyl-alanyl-alanin-äthylamid 15 (260 ml) in Dimethylformamid (20 ml), erwärmt auf 60°C, wurde Maleinsäureanhydrid (200 mg) zugegeben und die Reaktionslösung wurde 30 Minuten bei 80°C gehalten. Danach wurde das Reaktionsgemisch abgedampft und der Rückstand wurde aus Dimethylformamid und Isopropylal-20 kohol umkristallisiert. Es wurden 185 mg eines Produktes mit einem Schmelzpunkt von 260 bis 266°C (unter Zersetzung) erhalten.

Claims (5)
- 653 0382PATENTANSPRÜCHE 1. Carboxyalkanoylpeptid-alkylamide der allgemeinen Formel IX<✓COOHsCO-B-Ala-A-NH-R(I)in welcherR einen geraden oder verzweigten Alkylrest mit 1 bis 3 Kohlenstoffatomen oder einen Aralkylrest,A einen peptidisch gebundenen Prolin- oder Alaninrest, B einen peptidisch gebundenen Prolin- oder Alaninrest oder nichts, undX eine -CH2-, -CH2CH2-, -CH2CH2CH2- oder -CH=CH-Gruppe'bedeutet.
- 2. Verbindungen der Formel I gemäss Patentanspruch 1, dadurch gekennzeichnet, dass R als Aralkylrest ein Phenyl-äthylrest ist.
- 3. Verfahren zur Herstellung von Carboxyalkanoylpeptid-alkylamiden der allgemeinen Formel I nach Patentanspruch 1, dadurch gekennzeichnet, dass man zunächst eine Verbindung der allgemeinen Formel IIA-NH-R(II)in welcher R und A dieselbe Bedeutung wie in der Formel I haben, mit einer Verbindung der allgemeinen Formel IIIY-B-Ala(HI)in welcher B dieselbe Bedeutung wie in der Formel I hat, und Y eine Schutzgruppe bedeutet, umsetzt, wonach man das entstandene Zwischenprodukt nach Abspaltung der Schutzgruppe mit einem reaktionsfähigen Dicarbonsäurederivat der allgemeinen Formel IV✓COOHX<NZOOH(IV)in welcher X dasselbe wie in der Formel I bedeutet, zur Reaktion bringt.
- 4. Verfahren nach Patentanspruch 3, dadurch gekennzeichnet, dass Y in Formel III eine Benzyloxycarbonyl-gruppe bedeutet.
- 5. Verfahren nach Patentanspruch 3 oder 4, dadurch gekennzeichnet, dass das Dicarbonsäurederivat der Formel IV ein Anhydrid, Monohalogenid, Monoester oder Monoamid ist.lenstoffatomen oder einen Aralkylrest, insbesondere einen Phenyläthylrest,A einen peptidisch gebundenen Prolin- oder Alaninrest, B einen peptidisch gebundenen Prolin- oder Alaninrest oder 5 nichts, undX eine -CH2-, -CH2CH2-, -CH2CH2CH2- oder -CH=CH-Gruppe bedeutet.Dipeptidalkylamide sind wirksame Elastaseinhibitoren 10 (belgische Patente No. 855 851 und No. 856 064). Hierbei liegt eine Dipeptidsequenz vor, welche eine für das elastoly-tische Substrat optimale Aminosäurezusammensetzung enthält, d.h. beispielsweise Alanyl-Alanin oder Alanyl-Proplin. In den erwähnten Patentschriften werden jedoch einige neue 15 Erkenntnisse über die Wechselwirkung zwischen Substrat und Elastase nicht gänzlich berücksichtigt, welche gemäss den Erfahrungen für die Funktion eines wirksamen Inhibitors entscheidend sind. In früheren Arbeiten, welche sich mit der Optimierung des Substrates für die pankreatische Ela-20 stase befassen, wurde festgestellt, dass für die Wechselwirkung zwischen dem Enzym und dem Substrat die elektrostatische Bindung zwischen P4-S4 und Ps-Ss von entscheidender Bedeutung ist (J. Schechter, A. Berger: Biochem. Biophys. Res. Commun. 32,898, 1968), welche vor der primären 2s Wechselwirkung bevorzugt wird (Eur. J. Biochem. 69,1, 1976; FEBS Lett. 40,353,1974).Es wurde nun überraschenderweise festgestellt, dass die Carboxygruppe im N-terminalen Teil des Inhibitors im Vergleich mit einem hydrophoben Rest (z.B. Acetylrest) dessen 30 Inhibitionskapazität wesentlich erhöht, ähnlich, wie sie die Konstanten der elastolytischen Hydrolyse bei Substraten dieses Typs erhöht. Die Eigenschaften eines elastolytischen Inhibitors sind auch von der Länge der Peptidkette, gleich wie bei den Substraten abhängig (Eur. J. Biochem. 69,1, 35 1976); ein Tripeptid hat eine höhere Inhibitionskapazität als ein Dipeptid. Bei Versuchen in vitro wurden die Inhibitionskonstanten (Ki) der 3-Carboxypropionyl- und 4-Carboxybu-tyryl-di- und -tripeptide bestimmt, und zwar mit Hilfe der pankreatischen Elastase und der chromogenen Substrate, 40 d.h. der Succinyl- oder Glutaryltetraalanin-p-nitroanilide. Die Ergebnisse sind aus der Tabelle I ersichtlich. Auf ähnliche Weise wurden die Inhibitionskonstanten (Ki) der neuen Inhibitoren mit Hilfe der humanen Elastase, d.h. der humanen Duodenalelastase nach der Stimulierung von 4s Cholecystochinen (Ei) bzw. mit Hilfe des gefriergetrockneten humanen, durch Kanülierung des Pankreaseauslasses erhaltenen Pankreasesaftes (E2) bestimmt und mit der pankreatischen Schweineelastase verglichen (Es).
